Methionine is the first limiting amino acid in poultry feed. Currently, methionine supplement is synthesized from an expensive chemical process requiring hazardous chemicals. Therefore, the objectives of this study were isolation of methionine producing bacteria from environmental samples and quantification of methionine production in these isolated bacteria. MCGC medium was selected as the isolation medium for methionine-producing bacteria by using Corynebacterium glutamicum ATCC13032 and Escherichia coli ATCC23798 as the positive and negative controls, respectively. Thirty-nine bacterial strains were obtained from environmental samples. Only strains A121, A122, A151 and A181 were able to tolerate up to 0.1% (w/v) of ethionine or norleucine. These isolated strains were identified by sequencing small subunit rRNA genes. The results revealed that bacterial strains A121, A122, A151and A181 were Klebsiella species, Acinetobacter baumannii, A. baumannii and Pseudomonas aeruginosa, respectively. When methionine production in strains A121 and A181 was quantitated, strains A121 and A181 generated methionine up to 31.1 and 124.6 μg/ml, respectively.  相似文献

The effect of nitrogen on biomass production, shoot elongation and relative density of the mosses Pleurozium schreberi, Hylocomium splendens and Dicranum polysetum was studied in a chamber experiment. Monocultures were exposed to 10 N levels ranging from 0.02 to 7.35 g N m−2 during a 90-day period. All the growth responses were unimodal, but the species showed differences in the shape parameters of the curves. Hylocomium and Pleurozium achieved optimum biomass production at a lower N level than Dicranum. Pleurozium had the highest biomass production per tissue N concentration. Tolerance to N was the widest in Dicranum, whereas Hylocomium had the narrowest tolerance. Dicranum retained N less efficiently from precipitation than the other two species, which explained its deviating response. All species translocated some N from parent to new shoots. The results emphasize that the individual responses of bryophytes to N should be known when species are used as bioindicators.  相似文献

主要对面包酵母菌在Zn2+和Zn2+-Pb2+两种离子体系中Zn2+的吸附行为进行了探讨。选择灭活面包酵母菌为吸附剂,对pH、菌体加入量等吸附影响因素进行了探讨并进行了吸附动力学过程和等温吸附效果的研究。结果表明,不同离子体系中相同初始浓度情况下,动力学研究显示实验所用面包酵母菌对溶液中Zn2+而言是一种快速高效的生物吸附剂,且在整个吸附过程中存在:qt(Pb2+)qt(Zn2+-Pb2+)qt(Zn2+)。实验条件下,以单离子体系中的Zn2+、Pb2+和双离子体系中的总离子为研究对象时,都能很好符合Langmuir等温吸附模型,计算获得面包酵母菌对不同离子体系的最大吸附量大小顺序为:qmax(Zn2+-Pb2+)qmax(Zn2+)qmax(Pb2+);然而通过对动力学吸附过程、实际平衡吸附量qeqex、吸附能力参数KL和EDS测试结果的综合分析,认为面包酵母菌对各体系中离子的吸附能力大小顺序为:Pb2(单+)(Zn2+-Pb2+)Zn(2单+)。同时认为Pb2+的存在对面包酵母菌吸附Zn2+的行为过程具有强烈抑制作用。  相似文献

应用酵母菌对木薯酒精废液进行了小型批量的处理试验研究.结果表明,该菌种对于高浓度有机废水具有很强的适应能力,能迅速形成优势菌群,分解有机物.在进水pH 5.0~5.5、温度30℃、曝气时间72 h、接种量1‰(v/v,活菌数为4.1×106个/mL)、溶解氧2~3 mg/L、BOD5∶N∶P为400∶2∶1条件下,COD和BOD5去除率可分别达85%和92%.该技术操作简单,可作为木薯酒精废液的一级生物处理,并可回收利用酵母蛋白.  相似文献

In certain environments nutrient and energy sources available to microorganisms can be limited. Foodborne pathogens must efficiently adapt in order to be successfully transmitted through the food chain to their hosts. For the intracellular foodborne pathogen Listeria monocytogenes, little is known regarding its response to nutrient/energy-limiting conditions. The alternative stress responsive sigma factor σB has been reported to contribute to survival under specific stresses. Therefore, the effects of several metabolic inhibitors on growth of L. monocytogenes wild-type and a ΔsigB mutant were examined. In the absence of inhibitors, both strains reached stationary phase after 18 h at 23°C and 10 h at 37°C. All of the metabolic inhibitors slowed growth of either strain, with few differences observed among the different inhibitors.  相似文献

利用筛选得到的10株酵母菌组成复合酵母菌系统,并将该复合菌系统接种到泥浆反应器中对模拟油泥样品进行了处理。在对反应器进行优化的基础上,比较了复合酵母菌体系和经驯化的活性污泥体系对模拟风化油泥的处理效果,发现复合酵母菌在反应速度和油去除率上都优于活性污泥。利用GC—MS对复合酵母菌处理体亲中主要脂肪烃组分的变化进行了分析,结果表明原油组分中脂肪烃部分在处理8d后基本被完全降解。  相似文献

Enhanced biodegradation of carbofuran (2, 3-dihydro-2, 2 dimethyl-7-benzofuranyl methyl carbamate) is an economically significant, but poorly understood, microbial phenomenon in soil. A series of experiments was conducted to examine short term changes in soil bacterial populations stimulated by carbofuran application at field rates. In the field experiment, commercially formulated carbofuran and butylate (S-ethyl diisobutyl carbamothioate) were applied at 5.6 kg ai ha-1 and 8.4 kg ai ha-1, respectively, on a soil (Putnam silt loam) exhibiting enhanced degradation of carbofuran. In laboratory studies, technical grade carbofuran (20 mg kg-1 soil) was applied to samples of the field soil. Bacterial populations were estimated using non-selective (tryptic soy agar) and selective media containing carbofuran or butylate. Largest population increases in pesticide-treated soil were observed between 7 and 15 days after treatment (DAT) compared to populations in non-treated soil. Significant increases (P less than 0.05) in total bacterial populations and presumed carbofuran-degraders due to carbofuran application were associated with increased populations of Pseudomonas spp. and Flavobacterium spp. Application of carbofuran appeared to provide a competitive advantage to these species over actinomycetes persisting beyond 20 DAT. Growth responses of bacteria to carbofuran in the Putnam soil were compared to those in a native prairie soil (Mexico silt loam), which exhibited a much slower rate of carbofuran degradation. Bacterial population response to carbofuran was measurable, but small and short-lived. Perpetuation of the enhanced degradation phenomenon may lie in a persistent pesticide-induced competitive advantage given to a very small segment of the microbial population. This advantage may not be detectable after 20 days using conventional plating techniques.  相似文献

在使用混合酵母菌菌株处理高浓度含油废水中,研究了序批式反应器(SBR)内酵母菌在不同起始pH条件下对废水的处理效果。通过与pH中性或碱性条件比较发现,pH4~5的条件在酵母菌的发泡抑制、菌体生长以及COD/油去除等方面均显示出更好的效果,pH5是系统运行的最佳条件。长达100d的SBR连续运行试验表明,系统可以在pH=5条件下长期、稳定运行。  相似文献

采用直接接触式膜蒸馏工艺(DCMD)浓缩处理高浓度发酵废液,在热侧温度65℃、冷侧温度30℃条件下,连续运行21 h,考察了浓缩过程中产水通量、产水侧COD和TOC浓度变化及蛋白质浓缩效果.结果表明,产水通量随运行时间延长而下降且降速较快,15 h内由17.1 kg/(m2·h)降至8.9 kg/(m2·h);经去离子水冲洗,产水通量可恢复至56.1%.产水中COD和TOC浓度随处理时间延长而不断增大.此外,原水与膜蒸馏出水中GC-MS分析结果也表明,随运行时间延长,原水中有更多种类的有机物进入产水侧,主要为挥发性有机物.对膜内表面进行扫描电镜-能谱(SEM-EDS)分析发现,膜内表面有块状沉积物出现,主要成分为有机物,含有少量的无机物.DCMD运行21 h后,废液体积浓缩8.3倍,有用组分蛋白质浓缩6.9倍,COD和TOC的去除率均达95%以上.  相似文献

将一株产絮酵母菌(编号B-02号)发酵后的废菌体制成生物吸附剂,研究该生物吸附剂对废水中Cd2+的生物吸附特性。结果表明:(1)pH值对Cd2+会产生较大的影响,偏酸性(pH=4~6)条件利于吸附;该吸附剂对Cd2+吸附速率较快,8~10 min就可达到吸附平衡;(2)吸附剂的吸附动力学符合二级动力学模型,吸附Cd2+的实验数据对Langmuir等温式的拟合情况良好,吸附剂吸附Cd2+的最大吸附量为70.752 mg/g。用0.5 mol/L HNO3对吸附Cd2+的酵母菌进行解吸,解吸率可达89.7%。  相似文献

对采用放电等离子体与饲养酵母联合处理味精废水进行了初步研究。结果表明,放电等离子体预处理后废水的CODcr值普遍升高,但经假丝酵母联合处理后CODCr去除率增加了38.3%,效果明显优于仅采用饲养酵母法处理的味精废水,说明放电等离子体预处理可使味精废水中的有机大分子破坏成小分子,有利于生物降解,为味精废水治理提供了一条新的途径。实验初步研究了联合处理后的CODcr去除率与预处理电压、预处理电流、预处理次数的关系,结果表明存在相应的最优值。  相似文献

研究了3株酵母菌(Candida lipolytica、Candida tropicalis和Candida halophila)产乳化剂能力与废水降解性能的关系。研究结果表明,3株酵母菌在含油废水中生长良好,且均可产生乳化剂。培养5 d后,菌株Candida lipolytica和Candida tropicalis的油脂去除率达到了94%以上;培养第3天时废水溶液中的乳化剂含量最高,分别为21 g/L和34 g/L;菌体细胞产乳化剂最高量出现在培养的第1天,分别为17(ng?mL-1)/100 cfu和13.5(ng?mL-1)/100 cfu;Candida halophila产乳化剂能力较弱,溶液中乳化剂量和菌体细胞产乳化剂量最高分别为12 g/L、0.45(ng?mL-1)/100 cfu,油脂去除率最大为66%。研究表明,酵母菌株产乳化剂量及乳化能力直接影响到酵母菌对废水中油脂的降解效果。3株酵母菌对废水COD去除率均达到80%以上,差异不明显,可能与不同菌株对不同种类基质的利用效率有关。  相似文献

为了缩短大型蚤急性毒性实验的周期,并且验证毒性实验的灵敏度和稳定性,对投加酵母提取物饲养的大型蚤的生长周期、2次产卵时间间隔、产卵数量、以及幼蚤灵敏度和稳定性进行研究,并将其应用于已知毒性物质和环境样品的毒性检测.结果表明,酵母提取物的适宜投加量为18.00~22.00 mg/L.在该浓度下饲养的大型蚤可保持较好的怀卵量,生长至成熟期和2次怀卵的时间间隔较短,幼蚤的灵敏度较好,毒性测试结果稳定.采用此方法测定的4种重金属和4种有机物的大型蚤急性毒性的24 h-EC50和48 h-LC50与已发表的实验结果比较相近,证明该方法准确、可靠.将该方法应用于城市生活污水急性毒性的测定发现,投加酵母提取物饲养的大型蚤所产幼蚤可以明显区分生活污水原水和处理后最终出水.因此,采用投加适量酵母提取物饲养大型蚤的方法可以缩短实验周期,为测定环境样品生物毒性提供方便.  相似文献

从餐饮废水集水井壁上的生物膜中筛选出酵母菌菌株,选择废水降解能力相对较高的3株酵母菌对餐饮废水进行处理。以COD去除率为主要指标,在单因素实验的基础上,运用响应面法对pH、TN和TP进行优化,得到酵母菌处理餐饮废水的最优条件。结果表明,酵母菌处理餐饮废水最优的工艺条件为pH=5.55、TN=27.63 mg/L、TP=4.40 mg/L,COD去除率达到90.3%;当进水COD为2 235 mg/L、油为660. 2 mg/L时,COD与油实际去除率分别达到了89.9%和89.8%。  相似文献

Empirical models of tree growth have been used for many years to predict timber yields and other properties of trees. However, such models rely on measured relationships between tree growth and historic environmental conditions. As anthropogenic actions alter the environment, especially atmospheric composition, empirical models become less reliable and process-based models become more useful. Process-based models are challenged to simulate growth of structurally and physiologically complex organisms using explicit mathematical expressions to capture growth response to environmental conditions. In this review we summarize the physiological requirements of process-based models and examine the capabilities of six published models (CARBON, ECOPHYS, PGSM, TREE-BGC, TREGRO, W91) for simulating tree response to changes in environmental conditions (elevated temperature, increased CO(2) concentration, and enhanced concentrations of tropospheric ozone). These analyses indicate that current models are reliable integrators of environmental effects on individual processes (e.g. photosynthesis), but may be less reliable where physiological acclimation occurs or when extrapolated to growth of specific tree compartments.  相似文献

In the present study, the yeast estrogen screen (YES) has been used to assess the estrogenic activity in surface waters of a coastal region in the German Baltic Sea. Solid-phase extraction using the copolymer Oasis HLB followed by a clean-up on silica was carried out on approximately 50-l water samples. From the final 400 μl extract volume, 100 μl aliquots were used for the measurement of estrogenic activity and for chemical analysis, which was performed by liquid chromatography coupled to tandem mass spectrometry (LC–MS/MS). From 29 samples taken during two campaigns (2003 and 2004) at five different stations 27 samples showed an estrogenic response higher than 10%. The response in the YES was expressed as measured estradiol equivalents (EEQs), which were in the range of 0.01 (Darss Peninsula) to 0.82 ng/l (Inner Wismar Bay). Samples from stations located in inner coastal waters showed higher estrogenic activities than those from outer located stations. A comparison of measured estrogenicity (YES) and calculated estrogenicity (chemical analysis) showed significant differences, probably due to the presence of anti-estrogenic compounds and/or the estrogenic activity of unknown, not identified contaminants. The main contributors to the overall estrogenic activity were synthetic and natural hormones.  相似文献
