This investigation was undertaken to determine the effect of culture filtrates of different isolates of Fusarium oxysporum f. sp. lycopersici on mortality of Meloidogyne incognita juveniles and egg hatching. It was observed that different concentrations including standard extract (S.E), 1:10 and 1:100 dilutions of all fungal filtrates inhibited egg hatch when compared with control. Minimum mortality and maximum hatching was observed in BRT (showing least mortality) isolate of F. oxysporum, while maximum mortality and minimum egg hatching was recorded in BGT (showing maximum mortality) isolate. Larval mortality was decreased with a decrease in concentration and the least mortality was observed in 1:100 when compared with SE and 1:10. The potentiality of both the isolates (BRT and BGT) against root-knot nematode M. incognita was confirmed by the pathogenicity test on tomato. These observations confirmed that F. oxysporumisolates possesses variability in pathogenicity ranging from pathogenic to bio-control agent. The plants inoculated with BRT isolate failed to show wilt symptoms while plants inoculated with BGT isolate showed wilt indices.  相似文献

Fusarium wilt of cucumber caused by the fungus, Fusarium oxysporum, is one of the most destructive soilborne diseases and can result in serious economic loss. No efficient fungicide is currently available to control the disease. The aim of this study was to examine the disease suppression ability of pig manure and sludge composts in peat-based container media and explore the possible disease suppression mechanisms. Pig manure and sewage sludge compost were made in laboratory-scale tanks. Plant growth media were formulated with peat mixture and compost (or 60?°C heated compost) in a 4:1 ratio (v/v). Cucumber seedlings were artificially inoculated with F. oxysporum conidia (5?×?105 conidia mL?1) by the root-dip method. Cucumber Fusarium wilt was effectively suppressed in sludge compost-amended media, while the disease suppression effect of pig manure compost was limited. The ammonia levels in the manure compost-amended media were significantly higher than those of sludge compost-amended media, which could explain its lower disease suppression ability. Heated composts behaved similarly with respect to disease suppression. Adding composts increased microbial biomass, microbial activity, and the microbial diversity of the growth media. PCR-DGGE results indicated that the fungal community had a significant correlation to the disease severity. The artificially inoculated pathogen was retrieved in all treatments and one possible biocontrol agent was identified as a strain of F. oxysporum by phylogenetic analyses. The results indicated that the sludge compost used in this study could be applied as a method for biocontrol of cucumber Fusarium wilt.  相似文献

This study investigated bioamplification of polychlorinated biphenyls (PCBs) in yellow perch (Perca flavescens) eggs resulting from nutrient utilization by developing embryos during incubation. Newly fertilized eggs containing trace levels of PCBs via maternal deposition were collected from an aquaculture pond in which adult broodstock had been reared over their natural lives. The eggs were incubated using a flow through system that received the same pond water at in-situ temperatures from which they were spawned. Replicate samples of eggs were collected at six time points throughout incubation, ranging from day 0 (newly fertilized eggs) to post-hatch larvae (2-d old). Congener specific PCB fugacities in pooled egg samples showed increases over the incubation period. Just prior to hatching, incubated eggs averaged 2.7-fold higher PCB fugacities compared to fresh eggs. The increase in PCB fugacity with egg incubation time was independent of chemical KOW. After hatching, PCB residues were lost from the larvae, attenuating the maximum chemical fugacity achieved in late-incubated eggs. However, the rate of PCB elimination in the early larvae stages was KOW dependent such that a significant larvae/egg fugacity ratio was still evident for intermediate and highly hydrophobic compounds 2 d post-hatching. This study provides the first evidence of in-ovo PCB bioamplification in eggs of an aquatic species and suggests that incubating fish embryos are exposed to higher chemical fugacities in-ovo than would be predicted by maternal deposition alone.  相似文献

Abstract

Sensitivity of 24 isolates of Colletotrichum destructivum O’Gara, collected from alfalfa plants in Serbia, to eight selected fungicides, was investigated in this study. Molecular identification and pathogenicity test of isolates tested were also performed. Fungicide sensitivity was evaluated in vitro, using mycelial growth assay method. All isolates exhibited significant pathogenicity, causing necrosis at the alfalfa seedling root tips two days after inoculation. Using the primer pair GSF1-SR1 and by comparing the amplified fragments of the tested isolates with the marker (M), the presence of the amplicon of the expected size of about 900?bp was determined for all isolates. The isolates tested in this study showed different sensitivity towards fungicides in vitro. Mycelial growth was highly inhibited by QoI (quinone outside inhibitors) fungicide pyraclostrobin (mean EC50=0.39?µg mL?1) and by DMI (demethylation-inhibiting) fungicide tebuconazole (mean EC50=0.61?µg mL?1), followed by azoxystrobin (mean EC50=2.83?µg mL?1) and flutriafol (mean EC50=2.11?µg mL?1). Multi-site fungicide chlorothalonil and MBC (methyl benzimidazole carbamate) fungicide thiophanate-methyl evinced moderate inhibition with mean EC50=35.31 and 62.83?µg mL?1, respectively. Thirteen isolates were sensitive to SDHI (succinate dehydrogenase inhibitors) fungicide boscalid and fluxapyroxad, (mean EC50=0.49 and 0.19?µg mL?1, respectively), while the rest of isolates were highly resistant.  相似文献

We evaluated whether life-time exposure to PFOS affects egg development, hatching, larval development, survival, metamorphosis and body mass of Enallagma cyathigerum (Insecta: Odonata). Eggs and larvae were exposed to five concentrations ranging from 0 to 10 000 μg/L. Our results show reduced egg hatching success, slower larval development, greater larval mortality, and decreased metamorphosis success with increasing PFOS concentration. PFOS had no effect on egg developmental time and hatching or on mass of adults. Eggs were the least sensitive stage (NOEC = 10 000 μg/L). Larval NOEC values were 1000 times smaller (10 μg/L). Successful metamorphosis was the most sensitive response trait studied (NOEC < 10 μg/L). The NOEC value suggests that E. cyathigerum is amongst the most sensitive freshwater organisms tested. NOEC for metamorphosis is less than 10-times greater than the ordinary reported environmental concentrations in freshwater, but is more than 200-times smaller than the greatest concentrations measured after accidental releases.  相似文献

The inhibitory activity of amphotericin B, clotrimazole, econazole, miconazole and nystatin was compared against Fusarium oxysporum f.sp. radiciscucumerinum. The most efficient antifungal agent against the growth of Fusarium oxysporum was econazole, followed by clotrimazole, miconazole, amphotericin and nystatin. The ED5 0 and ED9 0 values were 0.053 and 1.002 ppm for econazole, 0.088 and 1.100 ppm for clotrimazole, 0.173 and 3.210 ppm for miconazole, 0.713 and greater than 48 ppm for amphotericin and 3.860 and 16.702 ppm for nystatin. The ED5 0 values of nystatin and amphotericin against spore germination of Fusarium oxysporum were determined at 3.1427 ppm and 8.3990 ppm respectively, nystatin was 2.76 times more effective than amphotericin, while no effect was observed after the addition of econazole, clotrimazole and miconazole. The tested azoles were more effective than amphotericin and nystatin on growth inhibition of Fusarium oxysporum but amphotericin and nystatin acted significantly better on spore germination of Fusarium.  相似文献

Petals of red, yellow and white roses (Rosa damascene Mill.) of the family Rosaceae were extracted with (1:1) methylene chloride/methanol and tested for their antimicrobial activities against four species of Gram-positive bacteria (Bacillus cereus, Bacillus subtilis, Micrococcus luteus and Staphylococcus aureus), five species of Gram-negative bacteria (Enterobacter aerogenes, Escherichia coli, Klebsiella pneumonia, Pseudomonas aeruginosa and Serratia marcescens) and five species of fungi (Penicillium notatum, Aspergillus niger, Rhizopus stolonifer, Saccharomyces cerevisiae and Fusarium oxysporum). All of the crude extracts showed a wide range of antimicrobial activities according to the tested organism and rose's type. Micrococcus luteus was found to be the most susceptible bacteria to all crude extracts. Red and yellow petal extracts showed much higher antibacterial activity than the white petals extract. Bacillus subtilis was found to be the least susceptible to all extracts. The fungus, Penicillium notatum was found to be the most susceptible with white petal extract being the most effective. Saccharomyces cerevisiae and Fusarium oxysporum were the least susceptible to all extracts. White roses extract showed much higher antifungal activities against Penicillium notatum than red or yellow roses, therefore, it was subjected to several bioassay guided chromatographic fractionations and purification to isolate the active chemical(s) responsible for the antifungal activity. Chemical structure of the isolated antifungal compounds were identified by spectroscopy techniques and found to be a γ-sitosterol and (Z,Z)-9,12-octadecadienoic acid. Antibacterial activity of the various types of rose extracts were due to complex mixtures of organic compounds which are still under chemical investigation and will be published later.  相似文献

An invertebrate embryo toxicity test with the ampullariid snail, Marisa cornuarietis, to assess the toxicity of pesticides and heavy metals recently was established. Snail embryos were treated with atrazine (100, 1000, 10 000, and 30 000 μg/L), imidacloprid (10 000, 25 000, and 50 000 μg/L), Ni2+ (0.1, 1, 10, and 100 μg/L) or Zn2+ (100, 200, 500, 1000, 2000, and 5000 μg/L). The effect of these substances was examined by monitoring the following endpoints: mortality, formation of tentacles and eyes, heart rate, hatching, and weight after hatching. Effects in term of a significant delay on the formation of both tentacles and eyes were found after treatment with 100 μg/L Ni2+ or 200 μg/L Zn2+. The heart rate was shown to significantly decrease at 25 000 μg/L imidacloprid or 1000 μg/L Zn2+. At 100 μg/L atrazine, 10 μg/L Ni2+, or 1000 μg/L Zn2+ a significant delay in hatching became visible. No significant mortality was observed for the tested concentrations of atrazine, imidacloprid, or Ni2+, while 5000 μg/L Zn2+ resulted in 100% mortality after 10 d. The weight of freshly hatched individuals remained unaffected in all treatments. On the basis of the lowest observed effect concentrations (LOECs) recorded, we could show the M. cornuarietis embryo toxicity test (MariETT) to react up to three orders of magnitude more sensitive (for metals) and at least one order of magnitude more sensitive (for the tested organics) than the established Danio rerio embryo test.  相似文献

Twenty microfungal isolates were collected from diseased fruiting bodies of Agaricus bisporus sampled from Serbian mushroom farms during 2003–2007. Based on morphological characteristics and pathogenicity tests, the isolates were identified as Cladobotryum dendroides. The isolates of C. dendroides and A. bisporusF56 and U3 were tested for sensitivity to several selected fungicides in vitro. C. dendroides isolates were found to be more sensitive to prochloraz manganese and flusilazole + carbendazim than to the other fungicides tested (EC50 values were 0.09 and 0.11 mg L? 1, respectively) and weakly resistant to thiophanate-methyl (EC50 values ranged between 6.53 and 12.09 mg L? 1). Selectivity indexes of the tested fungicides on both C. dendroidesand A. bisporusindicated that thiophanate-methyl, cyproconazole + carbendazim and flusilazole + carbendazim had much less selective fungitoxicity than benomyl, carbendazim and prochloraz manganese.  相似文献

The insecticide chlordecone is a contaminant found in most of the banana plantations in the French West Indies. This study aims to search for fungal populations able to grow on it. An Andosol heavily contaminated with chlordecone, perfused for 1 year in a soil–charcoal system, was used to conduct enrichment cultures. A total of 103 fungal strains able to grow on chlordecone-mineral salt medium were isolated, purified, and deposited in the MIAE collection (Microorganismes d'Intérêt Agro-Environnemental, UMR Agroécologie, Institut National de la Recherche Agronomique, Dijon, France). Internal transcribed spacer sequencing revealed that all isolated strains belonged to the Ascomycota phylum and gathered in 11 genera: Metacordyceps, Cordyceps, Pochonia, Acremonium, Fusarium, Paecilomyces, Ophiocordyceps, Purpureocillium, Bionectria, Penicillium, and Aspergillus. Among predominant species, only one isolate, Fusarium oxysporum MIAE01197, was able to grow in a liquid culture medium that contained chlordecone as sole carbon source. Chlordecone increased F. oxysporum MIAE01197 growth rate, attesting for its tolerance to this organochlorine. Moreover, F. oxysporum MIAE01197 exhibited a higher EC50 value than the reference strain F. oxysporum MIAE00047. This further suggests its adaptation to chlordecone tolerance up to 29.2 mg l?1. Gas chromatography–mass spectrometry (GC-MS) analysis revealed that 40 % of chlordecone was dissipated in F. oxysporum MIAE01197 suspension culture. No chlordecone metabolite was detected by GC-MS. However, weak amount of 14CO2 evolved from 14C10-chlordecone and 14C10-metabolites were observed. Sorption of 14C10-chlordecone onto fungal biomass followed a linear relationship (r 2?=?0.99) suggesting that it may also account for chlordecone dissipation in F. oxysporum MIAE01197 culture.  相似文献

We examined the effects of tributyltin (TBT) on embryonic development, hatching success and sexual differentiation in Japanese medaka (Oryzias latipes). Embryos (within 8h after fertilization) were exposed to TBT in ovo via nanoinjection at concentrations of 0 (control), 0.16, 0.80, 3.96, 19.2 and 82.1 ng/egg. Embryonic survival, development and hatching were observed. Hatched fry were reared until 60 days when they sexually matured, and sexual differentiation was also examined by accordance of genetic and phenotypic sex, based on existence of DMY (a male determining gene in medaka) and secondary sex characteristics. As results, TBT caused a concentration-dependent mortality and impaired the embryonic development. However, no masculinization was detected at 60 dph medaka adults. Lowest observed effective concentration for inducing abnormal embryonic development was estimated to 0.16 ng/egg (ca. 160 ng/g egg).  相似文献

In the present investigation, the bioefficacy of developed carbofuran formulations, with PEG-600 (7a, CP1) & PEG-900 (7b, CP2) @ 5, 10 and 20 ppm, along with commercial formulation of carbofuran 3G (CP0) were evaluated against the root-knot nematode, Meloidogyne incognita infecting tomato (cv. Pusa Ruby) in pot and field conditions. The bioefficacy data indicated that the formulations developed by utilizing polymers having PEG – 900 (7b) as hydrophilic segment were effective even at 14 days post inoculation (dpi) as evident from shoot and root length. Also, the reduction in penetration was found to be maximum with CP2 (3.6 – 4.6 J2s) at all concentrations compared to CP1 (6.6 – 16.4 J2s) and CP0 (29.3 – 32.6 J2s). Overall, CP2 was more effective in reducing the number of nematodes up to 14 days, compared to CP1 and CP0. Both the CR formulations (CP1 and CP2) in general significantly reduced the number of galls, when compared to CP0. However, under field conditions, lower concentrations (5, and 10 ppm) of CP2, were less effective in controlling the gall formation whereas, CP2 at 20 ppm, was most effective than other treatments. The study revealed that the developed CR formulations of carbofuran have the potential for effective management of M. incognita in tomato under field conditions.  相似文献

This investigation was undertaken to compare the percentage response of colonization and development of VA-Mycorrhiza (Glomus fasciculatum) on a number of pulse crops viz. cowpea, chickpea, soybean, pigeonpea and lentil under glasshouse conditions. Among the above-mentioned crops, pigeonpea exhibited the best performance and was selected for further studies. In this host the development and colonization percentage of G. fasciculatum was investigated under two separate substrates i. e. soil amended with FYM and karanj oilseed cake keeping a control treatment of field soil. A third treatment amended with karanj oilseed cake and farm yard manure (FYM) was also kept which responded best in terms of colonization percentage. This treatment showing improved plant health as well as integration with G. fasciculatum was selected as an ideal treatment for the management of disease complex caused by root knot nematode, Meloidogyne incognita and root wilt fungus, Fusarium udum on pigeonpea. Thus the treatment constituting FYM, karanj oilseed cake and VA-Mycorrhiza reduced the disease incidence caused by both maladies to a great extent with the most promising improvement in plant growth parameters as compared to all others. The present investigation, in addition to proposing an ideal eco-friendly treatment for the management of this disease complex also proposed an excellent medium for the proliferation of the obligate bio-protectant, G. fasciculatum.  相似文献

Plant-parasitic nematodes, such as Meloidogyne incognita, cause serious damage to various agricultural crops worldwide, and their control necessitates environmentally safe measures. We have studied the effects of plant secondary metabolites on M. incognita locomotion, as it is an important factor affecting host inoculation inside the soil. We compared the effects to the respective behavioral responses of the model saprophytic nematode Caenorhabditis elegans. The tested botanical nematicides, all reported to be active against Meloidogyne sp. in our previous works, are small molecular weight molecules (acids, alcohols, aldehydes, and ketones). Here, we specifically report on the attractant or repellent properties of trans-anethole, (E,E)-2,4-decadienal, (E)-2-decenal, fosthiazate, and 2-undecanone. The treatments for both nematode species were made at sublethal concentration levels, namely, 1 mM (<EC50), and the chemical controls used for the experiments were the commercial nematicides fosthiazate and oxamyl. According to our results, trans-anethole, decenal, and oxamyl attract C. elegans, while 2-undecanone strongly attracts M. incognita. These findings can be of use in the development of nematicidal formulations, contributing to the disruption of nematode chemotaxis to root systems.  相似文献

This study aimed to isolate and characterize bacteria able to use sulfentrazone in the commercial formulation as their sole carbon source. The isolation of the potential sulfentrazone-degrading bacteria was made from soil samples with a recent history of herbicide application and from isolates identified through rDNA sequencing. Subsequently, we assessed the growth of the isolates and their sulfentrazone degradation ability using high-performance liquid chromatography. Twenty-six potential sulfentrazone-degrading bacterial isolates were obtained in pure culture. Through analysis of the rDNA sequences, the predominance of bacterial species of the genus Pseudomonas was found. The isolates presented a differentiated ability of sulfentrazone degradation. The presence of herbicide in the culture medium reduced the log phase of four isolates. Pseudomonas putida, Pseudomonas lutea, Pseudomonas plecoglossicida and three isolates of Pseudomonas sp. showed higher sulfentrazone degradation capacity, which varied from 4 to 15%. This is the first report of the Pseudomonas genre capable of sulfentrazone degradation. The isolates obtained present potential use in bioremediation programs for soil contaminated with sulfentrazone.  相似文献

The aim of this paper is to examine the statistical relevance of bird species on the endpoints of avian long–term toxicity studies (eggs laid, eggs set, eggs hatching, embryo survivor, 14-day old survivors and eggshell thickness). Data from 561 animals of three different species (Colinus virginianus, Anas platyrynchos and Coturnix coturnix japonica) tested with five different pesticides were analyzed in this study. The substances considered were: Thiamethoxam (EZ-3-(2-chloro-1,3-thiazol-5-ylmethyl)-5-methyl-1,3,5-oxadiazinan-4-ylidene(nitro)amine), Thiacloprid ((Z)-3-(6-chloro-3-pyridylmethyl)-1,3-thiazolidin-2-ylidenecyanamide), Acetamiprid ((E)?N 1-[(6-chloro-3-pyridyl)methyl]-N 2-cyano-N 1-methylacetamidine), Phosmet (O,O-dimethyl S-phthalimidomethyl phosphorodithioate) and Dicofol (2,2,2-trichloro-1,1-bis(4-chlorophenyl)ethanol). Several general lineal mixed models were conducted to evaluate the factors affecting variables used in long-term reproductive toxicity tests. Test significance was p < 0.01 in all models tested. Model R2 value was high (0.80) for all variables except for eggs laid (R2 = 0.42) for the three species studied. Tukey studentized range test showed significant differences among species and pesticides. For pre-hatching period the differences were significant for eggs laid and eggs set among species. C. japonica showed statistical differences for egg hatching. With respect to embryo survivor and 14 days old survival, significant different were found for C.virginianus and A. platyrynchos, respectively. These results indicate that the selected species have an influence in the endpoints to be used for risk assessment.  相似文献

The objectives of this work were to evaluate the effects of environmentally relevant chlorpyrifos and endosulfan, concentrations in the incubation period, effective hatching and survival of embryos and neonates of the freshwater burrowing crab, Zilchiopsis collastinensis (Decapoda, Trichodactylidae). Both pesticides were prepared from commercial and technical grade products. The exposure to about 100, 200, and 400 ng endosulfan L?1, and 48, 240, and 1,200 ng chlorpyrifos L?1 did not cause differences in the incubation period or in effective hatching but decreased survival of neonates, especially in the concentrations prepared from the technical grade product. Even if these concentrations are below the median lethal concentration (LC50) values for embryos, these caused a significant decrease in the survival of neonates, i.e. when crabs are outside the egg and not protected by chorion. The decrease in the neonate population caused by these concentrations, which could be found in the environment, might impact aquatic communities.  相似文献

A series of novel N-alkyl substituted amides, synthesized by enzyme catalysis, were evaluated against root-knot nematode, Meloidogyne incognita and found to have potential antinemic activity. The corresponding amides were prepared by the condensation of equimolar amounts of carboxylic acids with different alkyl amines in the presence of Candida antarctica lipase at 60–90°C in 16–20 h. The reactions were carried out in a non - solvent system without the use of any activating agents. All the products were obtained in appreciable amounts and the yields for different compounds varied between 77.4–82.3%. The synthesized compounds were characterized using spectroscopy techniques namely Infra Red (IR) and Nuclear Magnetic Resonance (NMR) (1H and 13C). Nematicidal activity of synthesized amides was evaluated against J2s of Meloidogyne incognita at 500, 250, 125 and 62.5 ppm concentrations after 24 h, 48 h and 72 h of exposure. Among all the tested compounds, N-propyl-butyramide, N-propyl-pentanamide and N-propyl-hexanamide were found to possess significant activity with LC50 values of 67.46, 83.49 and 96.53 respectively. N-propyl-butyramide with LC50 value of 67.46 ppm was found to be most active amide against J2s of Meloidogyne incognita. The bioactivity study showed that an increase in alkyl chain significantly decreased the activity of amides against root-knot nematode.  相似文献

In a recent study, the pH of commonly used Salmonella pre-enrichment media became acidic (pH 4.0 to 5.0) when feed or feed ingredients were incubated for 24 h. Acidic conditions have been reported to injure or kill Salmonella. In this study, cultures of four known feed isolates (S. montevideo, S. senftenberg, S. tennessee, and S. schwarzengrund) and four important processing plant isolates (S. typhimurium, S. enteritidis, S. infantis, and S. heidelberg) were grown on meat and bone meal and later subjected to desiccation and heat exposure to stress the microorganism. The impact of stress on the isolates ability to survive in acidic conditions ranging from pH 4.0 to 7.0 was compared to the non-stressed isolate. Cell injury was determined on xylose lysine tergitol 4 (XLT4) and cell death determined on nutrient agar (NA). When measured by cell death in non-stressed Salmonella, S. typhimurium was the most acid tolerant and S. heidelberg was the most acid sensitive whereas in stressed Salmonella, S. senftenberg was the most acid tolerant and S. tennessee was the most acid sensitive. The pH required to cause cell injury varied among isolates. With some isolates, the pH required for 50% cell death and 50% cell injury was similar. In other isolates, cell injury occurred at a more neutral pH. These findings suggest that the pH of pre-enrichment media may influence the recovery and bias the serotype of Salmonella recovered from feed during pre-enrichment.  相似文献
