More knowledge on the composition of aqueous extracts of broiler litter amended for Water Soluble P (WSP) reduction would help to understand how amendments work. We measured pH, concentrations of Ca, Mg, Fe, Al, Cu, Mn, Zn, Molybdate Reactive P (MRP), and Dissolved Unreactive P (DUP) in water extracts of broiler litter treated with aluminum sulfate (ALS), ferrous sulfate (FES), ferric chloride (FEC), and gypsum (GYP) at 0, 5, 15, and 25% w/w. In order to study the effects of acidification, the same properties were measured in aqueous extracts of broiler litter suspensions that were titrated to end-points 3, 4, or 6 with 0.5N HCl. Concentrations of MRP, DUP, Ca and Mg, were 61%, 53%, 3.8 times, and 2.6 times greater in extracts from suspensions acidified to pH 6 than at the original pH of 8.9. ALS, FES, and FEC reduced pH, and showed similar effects on WSP concentrations, which were greater than with GYP. The magnitude of the reductions in WSP by ALS, FES, and FEC is uncertain because the actual amount of WSP immobilized cannot be determined. This is because of two opposite effects: 1) Through adsorption, soluble aluminum and iron remove phosphates from solution, and 2) Through acidification, iron and aluminum compounds release phosphates to solution.  相似文献

The potential applications of immobilized bitter gourd peroxidase in the treatment of model wastewater contaminated with phenols have been investigated. The synthetic water was treated with soluble and immobilized enzyme preparations under various experimental conditions. Maximum removal of phenols was found in the buffers of pH values 5.0-6.0 and at 40 degrees C in the presence of 0.75 mM H(2)O(2). Fourteen different phenols were independently treated with soluble and immobilized bitter gourd peroxidase in the buffer of pH 5.6 at 37 degrees C. Chlorinated phenols and native phenol were significantly removed while other substituted phenols were marginally removed by the treatment. Phloroglucinol and pyrogallol were recalcitrant to the action of bitter gourd peroxidase. Immobilized bitter gourd peroxidase preparation was capable of removing remarkably high percentage of phenols from the phenolic mixtures. Significantly higher level of total organic carbon was removed from the model wastewater containing individual phenol or complex mixture of phenols by immobilized bitter gourd peroxidase as compared to the soluble enzyme. 2,4-dichlorophenol and a phenolic mixture were also treated in a stirred batch reactor with fixed quantity of enzyme for longer duration. The soluble bitter gourd peroxidase ceased to function after 3h while the immobilized enzyme was active even after 6h of incubation with phenolic solutions.  相似文献

Contaminant desorption constrains the long-term effectiveness of remediation technologies, and is strongly influenced by dynamic non-equilibrium states of environmental and biological media. Information is currently lacking in the influence of biochar and activated carbon amendments on desorption of heavy metal contaminants from soil components. In this study, copper sorption-desorption isotherms were obtained for clay-rich, alkaline San Joaquin soil with significant heavy metal sorption capacity, and eroded, acidic Norfolk sandy loam soil having low capacity to retain copper. Acidic pecan shell-derived activated carbon and basic broiler litter biochar were employed in desorption experiments designed to address both leaching by rainfall and toxicity characteristics. For desorption in synthetic rain water, broiler litter biochar amendment diminished sorption-desorption hysteresis. In acetate buffer (pH 4.9), significant copper leaching was observed, unless acidic activated carbon (pHpzc = 3.07) was present. Trends observed in soluble phosphorus and zinc concentrations for sorption and desorption equilibria suggested acid dissolution of particulate phases that can result in a concurrent release of copper and other sorbed elements. In contrast, sulfur and potassium became depleted as a result of supernatant replacements only when amended carbon (broiler litter biochar) or soil (San Joaquin) contained appreciable amounts. A positive correlation was observed between the equilibrium aluminum concentration and initial copper concentration in soils amended with acidic activated carbon but not basic biochar, suggesting the importance of cation exchange mechanism, while dissolution of aluminum oxides cannot be ruled out.  相似文献

The article presents the results of research into the content of metalloid fractions in the tsunami deposits from southern Thailand. The following fractions, which are potentially most easily released from deposits to the environment, have been distinguished: the water soluble fraction, the exchangeable fraction extracted with the phosphate buffer and the fraction eluted with the solution of hydrochloric acid. The analytical technique atomic absorption spectrometry with hydride generation was applied. Spatial variability of the metalloid fractions in deposits and changes occurring in deposits over a period of several years of observation were determined. Based on the statistical analysis of the results, an attempt was made to determine the post-depositional release of deposits components to the environment. Based on the conducted research, the 4 years forming process of the arsenic, antimony and selenium occurrence after the deposition of sediments on land were described, as well as the balance in the amount of deposit components released to the environment.  相似文献

Abstract

The production of broiler chickens has become one of the largest sectors in U.S. agriculture, and the growing demand for poultry has led to an annual production growth rate of 5%. With increased demand for poultry, litter management has become a major challenge in the agriculture industry. Although the catalytic steam gasification has been accepted as a possible and feasible method for litter management, concern has been expressed about the presence of nitrogen and phosphorus containing species in the fuel gas and/or in the final solid residue. The possible release of phosphorus as phosphine gas in the fuel gas can have an adverse impact on the environment. Similarly, possible release of ammonia from the nitrogen containing species is also not acceptable. Hence, under partial U.S. Department of Agriculture support, a study was conducted to examine the fate and the environmental impact of the nitrogen- and phosphorus-containing species released during catalytic steam gasification of poultry litter. From various preliminary tests, it was concluded that most (~100%) of the phosphorus would remain in the residue, and some (20–70%) of the nitrogen would end up as ammonia in the fuel gas. The effects of temperature, catalyst loading, and type of catalyst on ammonia liberation were studied in a muffled furnace setup at atmospheric pressure. The fraction of nitrogen released as ammonia was found to decrease with an increase in temperature during pyrolysis and steam gasification. The bioavailability of phosphorus (P) in sediments from West Lake and Lake Tai (China) and Lough Erne (Northern Ireland) was evaluated using total P (TP), water soluble P (WSP), readily desorbable P (RDP), algal available P (AAP) and Olsen-P. The results indicated that the TP content, the amounts of phosphorus extracted by each method and their proportions to TP varied widely between sediments of the three lakes. TP and chemical extractable phosphorus in the sediments of Lough Erne were considerably higher than those of the two lakes in China. Although TP in the West Lake sediments was similar to that of Lake Tai, the extractable phosphorus data suggested that the bioavailability of P in West Lake sediments was higher than that in sediments from Lake Tai. Nevertheless, the rank order of extraction efficiency was the same in all lakes and the amount extracted was in the sequence AAP > Olsen-P > WSP > RDP. There was a good correlation between extracted and TP in all the methods examined.  相似文献

Plant litter and organic sediments are a main sink for metals and metalloids in aquatic ecosystems. The effect of invertebrate shredder (a key species in litter decay) on metal/metalloid fixation by organic matter is described only under alkaline water conditions whereas for slightly acidic waters nothing can be found. Furthermore, less is known about the effect of invertebrate shredders on the quality of dissolved organic carbon (DOC) and nitrogen (DON) released during litter decay. We conducted an experiment to investigate the impact of invertebrate shredder (Gammarus pulex) on metal/metalloid fixation/remobilization and on the quality of DOC/DON released under slightly acidic water conditions. During decomposition of leaf litter, invertebrate shredder facilitated significantly the emergence of smaller particle sizes of organic matter. The capacity of metal fixation was significantly higher in smaller particles (POM 2,000?C63???m) compared to original leaf litter and litter residues. Thus, G. pulex enhanced metal fixation by organic partition of sediments by increasing the amount of smaller particle of organic matter in aquatic ecosystems. In contrast, the capacity of metal/metalloid fixation in the smallest fraction of POM (<63???m) was lower compared with leaf residues in treatment without invertebrates. Remobilization of metals and metalloids was very low for all measured elements. A significant effect of invertebrates on quantitative formation of DOC/DON was confirmed. The quality of released DOC/DON, which may affect metal/metalloid remobilization, was also significantly affected by invertebrate shredders (e.g., more carboxylates). Hence, invertebrate shredder enhanced significantly the fixation of metals/metalloids into POM in slightly acidic environments.  相似文献

The current study was conducted to assess the bactericidal effectiveness of several nitrocompounds against pathogens in layer hen manure and litter. Evidence from an initial study indicated that treatment of layer hen manure with 12 mM nitroethane decreased populations of generic E. coli and total coliforms by 0.7 and 2.2 log10 colony forming units (CFU) g?1, respectively, after 24 h aerobic incubation at ambient temperature when compared to untreated populations. Salmonella concentrations were unaffected by nitroethane in this study. In a follow-up experiment, treatment of 6-month-old layer hen litter (mixed with 0.4 mL water g?1) with 44 mM 2-nitroethanol, 2-nitropropanol or ethyl nitroacetate decreased an inoculated Salmonella typhimurium strain from its initial concentration (3 log10 CFU g?1) by 0.7 to 1.7 log10 CFU g?1 after 6 h incubation at 37°C in covered containers. After 24 h incubation, populations of the inoculated S. Typhmiurium in litter treated with 44 mM 2-nitroethanol, 2-nitropropanol, ethyl nitroacetate or nitroethane were decreased more than 3.2 log10 CFU g?1 compared to populations in untreated control litter. Treatment of litter with 44 mM 2-nitroethanol, 2-nitropropanol, ethyl nitroacetate decreased rates of ammonia accumulation more than 70% compared to untreated controls (0.167 µmol mL?1 h?1) and loses of uric acid (< 1 µmol mL?1) were observed only in litter treated with 44 mM 2-nitropropanol, indicating that some of these nitrocompounds may help prevent loss of nitrogen in treated litter. Results warrant further research to determine if these nitrocompounds can be developed into an environmentally sustainable and safe strategy to eliminate pathogens from poultry litter, while preserving its nitrogen content as a nutritionally valuable crude protein source for ruminants.  相似文献

In this study, we attempted to establish a new indicator for fecal pollution of river sediment using a sensitive detection method for urobilin. Urobilin contained in sediment was extracted with an alkaline buffer solution. The suitable buffer solution for extraction of urobilin consisted of 0.1 M Tris-HCl buffer (pH 10.0) containing 0.1 m sodium chloride. The ratio of sediment to buffer was 1:10. The distribution of urobilin in river sediment was studied by this method. The amount of urobilin in the river sediment of Tokyo (urban) and the Miura peninsula (rural) area was surveyed. Large quantities of urobilin were detected in the sediments of the urban rivers, but it was scarce in the rural rivers. Urobilin showed a higher content in sediment than in water. Urobilin in river sediment is distributed in large amounts in the lower reaches, where the water is polluted with domestic sewage and industrial waste, but is scarce in the upper streams. The amount of urobilin decreases to the lower layers.  相似文献

An enzyme-linked immunosorbent assay (ELISA) for pyrithiobac-sodium (Staple) produced by DuPont was validated in Australian soils. This pyrithiobac-sodium ELISA was shown to be highly sensitive with the limit of detection of 4-5 ppt. Soil samples were extracted either in PBS buffer by shaking or by accelerated solvent extraction (ASE). While pyrithiobac sodium can be analyzed directly by ELISA after ASE extraction with 1/10 or more dilutions, the analysis of PBS extract required filtration and dilution 1/20 or more depending on the concentration. Immunoassay results compared favorably with GC-MS results for both ASE and PBS extract of incurred residue of pyrithiobac sodium in soil samples, indicating that this ELISA can be an inexpensive and reliable alternative to conventional residue analysis methods for quantification of pyrithiobac-sodium. This validation provided the basis for applying the ELISA to a field study of pyrithiobac-sodium.  相似文献

Selective supercritical fluid extraction (SFE) at 40 degrees C, 120 bar and 60 min was utilised as a means to estimate the bioavailable fraction of PCBs to chironomid larvae in a naturally contaminated limnic sediment. This extraction methodology removed about 50% of the PCBs from the sediment. According to the equilibrium partitioning theory, organisms in that sediment should decrease their uptake from the sediment to the same extent, biota-to-sediment accumulation factors (BSAFs) thus remaining constant. Surprisingly, the BSAFs for 11 PCB congeners decreased some 40% for the selectively extracted sediment as compared to BSAFs for organisms dwelling in untreated sediment. The results were statistically significant at the 0.001 level using a paired t-test. This can only be interpreted so that selective SFE removed easily available PCBs preferentially, leaving more tightly bound PCBs behind. Hence, by fine-tuning extraction conditions, this methodology might be used to estimate bioavailable fractions by chemical means.  相似文献

A naturally contaminated sediment was partially extracted with selective supercritical fluid extraction (SFE) to remove a fraction of supposedly bioavailable PCBs from the matrix. Eels (Anguilla anguilla) were cultured in systems with untreated and pre-extracted sediment, respectively, and it was shown that the SFE treatment selectively removed bioavailable PCBs from the sediment, since relative biota-to-sediment accumulation factors (BSAFs) for the eight studied PCB congeners were much lower in the pre-extracted than in the untreated system at the end of the study. Relative BSAF values decreased with about the same relative amount for all eight congeners, independent of the degree of chlorination and the initial concentration in the sediment. The results demonstrate the ability of SFE to selectively remove sediment-bound PCBs that are available for uptake by the eels, thus demonstrating the feasibility of using selective SFE to estimate bioavailability of PCBs in sediments.  相似文献

The use of the organic As compound, roxarsone, as an antibiotic additive to poultry feed continues to raise concern over potential negative environmental impacts. Total As concentration in poultry litter can reach >40 mg kg−1 and both roxarsone and its mineralization product As(V) have been identified in poultry litters (PL). To investigate the fate of these As species upon land application of PL we conducted two studies. In the first, an Orangeburg soil (Ultisol from the Atlantic Coastal Plain) was spiked with either 20 mg kg−1 As(V) or roxarsone and incubated at 10% moisture content for 4 months. Exchangeable As was determined periodically by extraction with 0.1 M PO4. Both As(V) and roxarsone displayed similar desorption; initially, approximately 70% of added As was ligand exchangeable and this decreased to 35% after 4 months incubation, presumably due to either slow sorption reactions or a change in solid phase speciation of As to less exchangeable forms. In the second study, various manipulations of two PL samples were applied to the Orangeburg soil at realistic field application rates. The treatments were wet to 10% moisture content and water soluble As, Cu and organic carbon (DOC) was measured over 30 days. Arsenic and Cu solubility were highest from the dried litter samples. Ashing of the PLs decreased soluble As and Cu, presumably because of the loss of organic matter from the ashed litter and subsequent decrease in DOC. Application of leachates from either PL resulted in higher concentrations of soluble As and Cu than when the soil was amended with equivalent concentrations of soluble As and Cu dissolved in DI H2O. We hypothesize that the increased levels of DOC from the PL treatments enhance As and Cu solubility through competitive sorption and complexation, respectively. In fact, As and Cu solubility was correlated to DOC levels in the amended soil extracts. Even though land application of PL introduced relatively low concentrations of As and Cu to soil it appeared that other soluble constituents of PL significantly enhanced As and Cu solubility.  相似文献

The feasibility of a mild-solvent extraction procedure to predict the bioavailability of individual polycyclic aromatic hydrocarbons (PAHs) in soil was assessed. The quantities that were degraded during the course of biodegradation of phenanthrene and pyrene in soil with or without plants correlated with the amounts extracted by n-butanol, with R2 values of 0.971 and 0.994, respectively. Six consecutive groups of earthworms removed ca. 70% of the pyrene remaining after extensive biodegradation, a value similar to the quantity extracted by n-butanol. The amount of chrysene aged in sterilized soil that was extracted by n-butanol was not statistically different from the quantities assimilated by earth-worms (Eisenia fetida) introduced into the soil. Such a mild extraction procedure may be useful as a means of predicting PAH bioavailability.  相似文献

If the release mechanisms during selective chemical extraction of persistent organic pollutants (POP) mimic release mechanisms in natural systems during biological uptake, then a selective non-exhaustive extraction could give a quantitative measure of the bioavailable POP fraction. Supercritical fluid extraction (SFE) is suggested as a possible technique to estimate the amount of bioavailable polychlorinated biphenyls (PCBs) at contaminated sites and hence serve as a new tool in risk assessment. The uptake of PCBs by earthworm (Eisenia foetida) was investigated. PCB contaminated soil was pre-extracted with selective non-exhaustive SFE (50 degrees C, 350 bar, 1h), which removed on average 70% of the individual PCBs. Earthworms were placed in this pre-extracted soil, as well as in untreated soil. After 10 days, the PCB uptake by earthworms in the two systems was compared. The bioaccumulation factor (BAF) was 83% lower in the pre-extracted system than in the untreated system, demonstrating that SFE extracts primarily bioavailable contaminants. From the data, the bioavailable fraction could also be calculated to be 75%, which is very close to the 70% removed by SFE under the applied conditions. This suggests that the chemical methodology is capable of measuring the bioavailable fraction very accurately in this system.  相似文献

Arsenic (As) is highly mobilized when paddy soil is flooded, causing increased uptake of As by rice. We investigated factors controlling soil-to-solution partitioning of As under anaerobic conditions. Changes in As and iron (Fe) speciation due to flooded incubation of two paddy soils (soils A and B) were investigated by HPLC/ICP-MS and XANES. The flooded incubation resulted in a decrease in Eh, a rise in pH, and an increase in the As(III) fraction in the soil solid phase up to 80% of the total As in the soils. The solution-to-soil ratio of As(III) and As(V) (RL/S) increased with pH due to the flooded incubation. The RL/S for As(III) was higher than that for As(V), indicating that As(III) was more readily released from soil to solution than was As(V). Despite the small differences in As concentrations between the two soils, the amount of As dissolved by anaerobic incubation was lower in soil A. With the development of anaerobic conditions, Fe(II) remained in the soil solid phase as the secondary mineral siderite, and a smaller amount of Fe was dissolved from soil A than from soil B. The dissolution of Fe minerals rather than redox reaction of As(V) to As(III) explained the different dissolution amounts of As in the two paddy soils. Anaerobic incubation for 30 d after the incomplete suppression of microbial activity caused a drop in Eh. However, this decline in Eh did not induce the transformation of As(V) to As(III) in either the soil solid or solution phases, and the dissolution of As was limited. Microbial activity was necessary for the reductive reaction of As(V) to As(III) even when Eh reached the condition necessary for the dominance of As(III). Ratios of released As to Fe from the soils were decreased with incubation time during both anaerobic incubation and abiotic dissolution by sodium ascorbate, suggesting that a larger amount of As was associated with an easily soluble fraction of Fe (hydr) oxide in amorphous phase and/or smaller particles.  相似文献

Mediterranean coastal ecosystems are known to be highly subject to natural and anthropic environmental stress. In this study, we examine the effects of anthracene as a common pollutant on the total microbial communities from a Pinus halepensis litter of a typical Mediterranean coastal site (Les Calanques, Marseille). The main objective was to identify the microbial factors leading the resilience of this ecosystem. Two questions were addressed: (i) how lignin-degrading enzymes (Laccase, Lignin-peroxidase and Mn-peroxidase) are affected by the presence of this molecule, (ii) whether the indigenous consortia are involved in its degradation in mesocosms under favorable incubation conditions (25 °C, 60% WHC) and after different time intervals (1 and 3 month(s)). We found a strong increase in laccase production in the presence of anthracene after 3 months, together with anthracene degradation (28% ± 5). Moreover 9,10-anthraquinone is detected as the product of anthracene oxidation after 3 months. However neither lignin-peroxidase activity nor Mn-peroxidase activity is detected. Laccase proteins directly extracted from litter were sequenced via Nano-LC-MS/MS and reveal twelve different peptide sequences induced by the presence of anthracene in the mesocoms. Our study confirms the major detoxification role of this enzymatic system and highlights the high degradation potential of fungal species inhabiting P. halepensis litter, a factor in the resilience of Mediterranean ecosystems.  相似文献

This study investigated the forms of soil P released to solution, accuracy of their determination, and influence of colloids on P sorption/desorption dynamics. A Hagerstown silt loam, amended with dairy and poultry manure or superphosphate at five rates (0, 25, 50, 100, and 200 kg P ha(-1)), was extracted at two soil:solution ratios (1:5 and 1:100) and filtered at three pore sizes (0.8, 0.45, and 0.22 microm). Results showed that relative to the proportion of dissolved organic P (DOP, determined as the difference between total dissolved P [TDP] and P detected by ion chromatography), DRP increased with amendment rate. Relative to Mehlich-3 extractable P, DRP exhibited a power relationship with a much greater potential for soil P release at concentrations in excess of ca. 50 mg Mehlich-3 P kg(-1). Concentrations of DRP, determined by the acid molybdate method, were on average 12.5% greater than P detected by ion chromatography indicating P was solubilised during colorimetric determination. A linear relationship was found between total Al and DRP, which could indicate acid mediated hydrolysis of A1-humic-P substances, although acid mediated desorption of P from colloids cannot be discounted. No difference in solubilised P was found between solutions filtered at 0.22 and 0.45 microm, but was found between 0.8 microm and smaller filter sizes. Organic P extracted from manured soils was more recalcitrant than that extracted from soils amended with superphosphate, the later attributed to its accumulation in more labile pools. The sorption/desorption of P by colloids in solution were greatly affected by the rate of amendment and the soil:solution extraction ratio. More P was sorbed by superphosphate solutions compared to dairy manure amended soil solutions and was attributed to the saturation of colloidal P sorption sites by organic matter. In order to minimise the effects of colloids on P dynamics and the potential for hydrolysis in solution, filtration to at least 0.45 microm is required.
