Five polymorphic microsatellite loci were developed and then used to assess the population genetic structure of a commercially harvested merobenthic octopus species (Octopus maorum) in south-east Australian and New Zealand (NZ) waters. Beak and stylet morphometrics were also used to assess population differentiation in conjunction with the genetic data. Genetic variation across all loci and all sampled populations was very high (mean number alleles = 15, mean expected heterozygosity = 0.85). Microsatellites revealed significant genetic structuring (overall F ST = 0.024, p < 0.001), which did not fit an isolation-by-distance model of population differentiation. Divergence was observed between Australian and NZ populations, between South Australia and north-east Tasmania, and between two relatively proximate Tasmanian sites. South Australian and southern Tasmanian populations were genetically homogeneous, indicating a level of connectivity on a scale of 1,500 km. Morphometric data also indicated significant differences between Australian and NZ populations. The patterns of population structuring identified can be explained largely in relation to regional oceanographic features.  相似文献

The genetic population structure of the recently introduced Asian clam, Potamocorbula amurensis, in San Francisco Bay was described using starch gel electrophoresis at eight presumptive loci. Specimens were taken from five environmentally distinct sites located throughout the bay. The population maintains a high degree of genetic variation, with a mean heterozygosity of 0.295, a mean polymorphism of 0.75, and an average of 3.70 alleles per locus. The population is genetically homogeneous, as evidenced from genetic distance values and F-statistics. However, heterogeneity of populations was indicated from a contingency chi-square test. Significant deviations from Hardy-Weinberg equilibrium and heterozygote deficiencies were found at the Lap-1 locus for all populations and at the Lap-2 locus for a single population. High levels of variability could represent a universal characteristic of invading species, the levels of variability in the source population(s), and/or the dynamics of the introduction. Lack of differentiation between subpopulations may be due to the immaturity of the San Francisco Bay population, the general purpose phenotype genetic strategy of the species, high rates of gene flow in the population, and/or the selective neutrality of the loci investigated.  相似文献

Comparative restriction-fragment analysis was used to analyze the nuclear ribosomal DNA, and alcohol dehydrogenase-1 loci of Zostera marina L., for variation within and among populations. Eelgrass is a perennial marine flowering plant that is widespread and ecologically significant throughout the temperate northern hemisphere. A chemical method was developed to obtain restriction-quality DNA without CsCl fractionation from experimentally relevant quantities of seagrass tissues (0.5 to 1.0 g). The yield was 25 g g-1 fresh weight. The three morphologically distinct forms of Z. marina from disjunct populations examined in this study were found to be genetically distinct; morphologically similar populations were indistinguishable genetically. Genetic distinction also correlated with habitat depth, as subtidal and intertidal populations were clearly divergent. Homologous probes for the 17S and 28S ribosomal DNA genes were used to map 24 restriction sites on the rDNA repeat of Z. marina, which was determined to be about 14 kb in length. At least 1 length mutation and 5 restriction-site changes were identified that distinguished Z. marina populations from San Diego and Monterey Bay (Del Monte Beach) from Z. marina populations from Elkhorn Slough and Tomales Bay. Estimated sequence variation (100×p) between eelgrass populations ranged from 0.00 to 0.69. Individual plants were observed to contain as many as four different rDNA-repeat length variants. The mean number of rDNA-repeat length variants per individual in Z. marina was about two. Intrapopulation variation in rDNA-repeat type was observed in only one individual from the Tomales Bay population.  相似文献

The complete larval development of Euphausia pacifica in the Yellow Sea is described and the stages are compared with larvae of E. nana. Euphausiid larvae examined in the present study were collected at 30 stations in the Yellow Sea in 1989. During the stages of Calyptopis III to Furcilia II, E. pacifica larvae are smaller than E. nana larvae but they are larger from Furcilia III onward. There are geographical variations in body size of Calyptopis III among the southern California waters, the eastern Japan waters, the Japan Sea, and the Yellow Sea; body size is smallest in the Yellow Sea and Japan Sea, while it is largest in southern California waters and intermediate in eastern Japan waters. Segmentation of antennal endopods was observed in the 56 to 51 furcilia forms in the Yellow Sea population, suggesting that this characteristic is not as reliable for identification of furcilia stages as the pleopods and terminal telson spines. Thus, we propose here that there are six furcilia stages of E. pacifica, instead of the seven previously reported by Boden (1950).  相似文献

Allozyme and mitochondrial DNA (mtDNA) genetic variation was compared in orange roughy (Hoplostethus atlanticus Collett) collected from waters off southern Australia and from waters about 22 000 km away in the North Atlantic west of Scotland. Samples were screened for 11 polymorphic allozyme loci and with 9 restriction enzymes. Significant heterogeneity between the two areas was detected for three allozyme loci (ADA *, CK * and GPI-1 *), and the overall G ST (gene-diversity statistic) value of 1% was small but significant. Significant mtDNA haplotype heterogeneity was observed after 2- of haplotype frequencies but not after a G ST analysis. Nucleotide sequence-diversity analysis showed very low net divergence (0.0023%) between the two samples. The Australian orange roughy had a lower allozyme heterozygosity and a lower mitochondrial DNA nucleon diversity than the North Atlantic sample. The very limited, although significant, allozyme and mitochondrial DNA heterogeneity between these areas suggests that there is some gene flow between these two populations. The species appears to be widespread, with its presence reported from the southern Pacific, southern Indian, and northern and southern Atlantic Oceans, and it is likely that gene flow between the antipodes is mediated by stepping-stone exchange between adjacent populations rather than by direct migration.  相似文献

From July 1982 to December 1986, a study of the age and growth of the bonnethead shark Sphyrna tiburo was conducted in Tampa and Florida Bays, Florida, USA. Tetracycline-injected sharks held in captivity and, to a lesser extent, tagged, released and recaptured, were utilized for validating the annual nature of the rings (herein defined as the narrow, translucent regions) appearing on vertebral centra. The technique was validated for all age groups (0 to 6+yr) included in the study. Marginal increment analysis likewise suggested annual ring formation. The rings formed during the winter, when water temperatures were lowest. Using the vertebral ring aging-technique, von Bertalanffy growth curves for males and females from both tampa and Florida Bays were constructed. Growth of sharks born and held in captivity demonstrated that the male and female growth curves diverge after 1 yr and that mean sizes at age are statistically distinct after 2 yr. In both populations, females grew to larger sizes tha males and apparently are longer-lived. Reproductively mature females from Tampa Bay were significantly larger than those from Florida Bay.  相似文献

Crassostrea ariakensis is an important aquacultured oyster species in Asia, its native region. During the past decade, consideration was given to introducing C. ariakensis into Chesapeake Bay, in the United States, to help revive the declining native oyster industry and bolster the local ecosystem. Little is known about the ecology and biology of this species in Asia due to confusion with nomenclature and difficulty in accurately identifying the species of wild populations in their natural environment. Even less research has been done on the population genetics of native populations of C. ariakensis in Asia. We examined the magnitude and pattern of genetic differentiation among 10 wild populations of C. ariakensis from its confirmed distribution range using eight polymorphic microsatellite markers. Results showed a small but significant global θ ST (0.018), indicating genetic heterogeneity among populations. Eight genetically distinct populations were further distinguished based on population pairwise θ ST comparisons, including one in Japan, four in China, and three populations along the coast of South Korea. A significant positive association was detected between genetic and geographic distances among populations, suggesting a genetic pattern of isolation by distance. This research represents a novel observation on wild genetic population structuring in a coastal bivalve species along the coast of the northwest Pacific.  相似文献

The Patagonian squid, Loligo gahi DOrbigny, has been described as having temporally and spatially identifiable spawning aggregations. Variation at six microsatellite loci was used to assess whether seasonal and geographical spawning groups around the Falkland Islands represent distinct sub-populations. Genetic variation at these loci is high in this species (mean expected heterozygosity=0.87; mean number of alleles=14.7). No evidence of significantly different allele frequencies was found, either between samples from putative spawning cohorts or geographical areas, indicating that L.gahi around the Falkland Islands comprise a single genetically homogeneous population. Age structure analysis of samples (from statolith growth increments) indicated substantial spread in hatching dates among individuals of similar size and maturity status, suggesting the potential for extensive interbreeding between seasonal cohorts. A sample of L.gahi from the SE Pacific (Peru) displayed distinctly different gene frequencies (and allele size distribution at one locus) from SW Atlantic samples, supporting the suggestion that SE Pacific and SW Atlantic populations may represent distinct subspecies.Contributed by J.P. The tellinid bivalve Macoma balthica (L.) has an extensive geographic range that reaches from temperate to arctic coastal waters in the North Atlantic and North Pacific oceans. Recent studies have indicated that eastern and western North Atlantic populations are morphologically and genetically different from one another, and that they may have diverged as sibling species. To determine the genetic relationship between M. balthica from the Pacific and Atlantic coasts of North America, populations from each coast were examined at 11 enzyme loci using standard starch gel electrophoresis. Allele frequency data indicate that M. balthica populations from San Francisco Bay, California appear more closely related to western North Atlantic populations than to populations from Oregon. We suggest that San Francisco Bay populations were introduced relatively recently from western North Atlantic populations. The Oregon populations are probably a natural extension of northern populations that occur along Northern Asia and in the eastern North Atlantic.  相似文献

Microheterotrophic dissolved free amino acid (DFAA) utilization, and microbial community and bacterial community carbon production and growth were studied using 3H-labeled organics as tracers in marine surface-film and subsurface (10 cm) waters off Baja California in November 1983. DFAA utilization was generally more rapid during the day (0.14 to 0.38 nM h-1) than at night (0.04 to 0.14 nM h-1) in surface-film and subsurface waters, but the percent of utilized amino acid which was respired was always greater during the night (22 to 57%) compared to the day (14 to 18%). Utilization of DFAA-carbon was estimated to range from 0.3 to 5.3 g C l-1 d-1 for all stations studied. In six of the 8 samples examined, the percentage of microbial carbon accounted for by the bacterial component of the population (1.4 to 5.9%) was strikingly similar to the percentage of microbial carbon production accounted for by bacterial carbon production (1.9 to 5.1%). In all of these six samples, total microbial specific-growth rates and bacterial specific-growth rates were approximately equivalent (0.9 to 2.2 d-1 for the microbial community; 0.7 to 1.9 d-1 for bacteria). The two exceptions were samples apparently influenced by transient flagellate populations migrating into the surface or subsurface waters at night. These observations support the conclusion that surface films contain unique and highly active microbial populations.  相似文献

Mitochondrial DNA (mtDNA) genotypes were examined in 1989 in threeCrassostrea virginica (Gmelin) populations native to Chesapeake Bay, USA, and in one population selectively inbred for rapid growth for ten generations. We wished to determine whether this character would be useful as a genetic marker for distinguishing between the inbred line and the native oysters and to determine whether detectable genetic differences exist among present-day native populations ofC. virginica. Thirty mtDNA haplotypes were identified. The average percentage nucleotide difference between native haplotypes was 1.8%. Inbred oysters were characterized by mtDNA haplotypes distinctly different from ancestral native oysters, indicating a high degree of genetic differentiation between the two groups (average percentage mtDNA nucleotide sequence divergence,=21.8%). The common native haplotype was not present in the selectively inbred sample, and six of the seven haplotypes detected in the inbred oysters were not found in the survey of native oysters. Chi-square tests on haplotype frequencies indicated that the native populations were not significantly different from one another. However, the distribution and relatedness of haplotypes suggest that significant change in the oyster gene pool may have occurred over the past few decades.  相似文献

In the period from 1980 to 1984 organic phosphorus, nutrients, primary production rates (14C), chlorophyll a (chl a) standing crops, and basic oceanographic parameters were measured during 23 cruises at six stations in the open waters of the northern Adriatic Sea. These waters are significantly influenced by polluted Po River discharge. Organic phosphorus was correlated with several parameters which characterize phytoplankton activity and organic matter decomposition processes. In the late winter-spring period, organic phosphorus is produced during phytoplankton blooms. It is hypothesized that microzooplankton grazing is the main factor increasing the organic phosphorus concentrations in summer (up to 1.1 mol 1-1). Fall and winter had much lower values (below 0.3 mol 1-1) due to remineralization processes and an increased water mass exchange between the northern and central Adriatic regions. The direct contribution of organic phosphorus by freshwater discharge was not found to be significant. The higher organic phosphorus concentrations that can occur in low salinity waters are most likely due to their increased capability to support primary production.  相似文献

The genetic population structure of the precominant zooplankter, the copepod Calanus finmarchicus (Gunnerus), was examined to determine whether genetically distinct populations exist in the Gulf of Maine. C. finmarchicus was sampled in three regions of the Gulf of Maine (Great South Channel, spring 1989; northern Gulf of Maine, winter 1990; Great South Channel and Georges Bank, spring 1990). Copepods from seven locations in the Great South Channel, five in the northern Gulf of Maine and four on or near Georges Bank were assayed for allozyme variation and mitochondrial DNA variation of amplified 16S rRNA and cytochrome b genes. Restriction fragment length polymorphism (RFLP) analyses of both mitochondrial DNA genes revealed no variation among any of the individuals assayed. Analysis of five polymorphic allozyme loci revealed that genetic variation among the three geographic regions was low, and genetic identities were high between all locations (I>0.97). Most of the genetic variation was among locations regardless of region. Chi-square tests were used to examine genetic similarity between specific pairs of locations within and between regions. In the northern Gulf of Maine, genetic homogeneity occurred over larger spatial scales (hundreds of km) than in either the Great South Channel or Georges Bank (tens of km). Only copepods from the Bay of Fundy and Nova Scotian Shelf locations were genetically distinct from Wilkinson Basin copepods at two loci. Copepod populations from the northern locations may have been partially isolated or they may represent immigrant populations (e.g., from the Gulf of St. Lawrence). Several pairs of locations were genetically distinct at one or more loci in the two southern regions. Differences between locations in these regions may represent distinct populations advected into the areas at different times or from different sources (e.g., genetic variation may represent a mixture of genetically distinct northern and southern copepod populations). These results suggest extensive gene flow among populations of C. finmarchicus in the Gulf of Maine with some evidence of genetic population subdivision near the Gulf's northeastern and southern boundaries.  相似文献

The in situ decomposition rate of urea was measured using 14C-labelled urea at 3 areas in the North Pacific Ocean: Sagami Bay on the southern coast of central Japan, the northwestern Pacific central waters and the subarctic Pacific waters. The mean values of the decomposition rates of urea in surface waters of these areas were 44.5, 1.51 and 1.32 mol urea m-3 d-1, respectively. These rates decreased with depth. High rates of urea carbon incorporation into particulate matter and the CO2 liberation from urea carbon into seawater were obtained in light bottles in the euphotic zone, while low rates were found in dark bottles. The turnover rates of urea in the 3 areas were calculated respectively as 12, 113 and 110 d at the surface, and the values increased with depth.  相似文献

Because of the abundance and size of Oikopleura vanhoeffeni its quantitative role as a suspension feeder in cold ocean waters needs to be defined. To minimize the effect of manipulation and containment, and to assess the effect of naturally occurring factors on clearance rate, I used an in situ latex microbead technique in Logy Bay, Newfoundland, from February 1985 to June 1986. Individual clearance rates ranged from 8–944 ml h-1, increasing exponentially with increasing trunk length. Partial correlation and principal components analysis indicated that trunk length and the concentration of ingestible chlorophyll a accounted for a majority of the variation in clearance rate. At densities of 4–110 m-3, O. vanhoeffeni populations removed from >1 to 13% of the standing stock of ingestible food particles each day. Grazing by near-surface populations was lowest during the spring diatom bloom (>1.4% of daily particle production removed per day), and was highest in June during the post-bloom crash (4 to 10% of daily production removed). Some populations in mid-depth waters had much higher population clearance rates (ca. 50% of daily production removed) because of a greater proportion of large animals. The median percentage daily ration (g Cxg C-1xd-1x100%) of 64% accounted for observed house production rates (1 to 2 d-1, with each house=23% of body carbon).  相似文献

Restriction enzyme analysis of mitochondrial DNA (mtDNA) was used to test for genetic homogeneity of orange roughy (Hoplostethus atlanticus) in the southern hemisphere. Two hundred and eighty-six orange roughy specimens were collected from seven general localities: the Great Australian Bight; South Australia (off southeastern Kangaroo Island); the west coast of Tasmania; the east coast of Tasmania; New South Wales; New Zealand and South Africa. Mitochondrial DNA was extracted from developing ovary tissue and analysed with 10 six-base enzymes and 3 four-base enzymes. Both forms of analysis revealed a low level of genetic diversity in this species. The six-base enzyme study found no evidence of reproductively isolated populations of orange roughy in southeastern Australian waters. However, an analysis of 107 fish with 3 four-base enzymes identified at least partial genetic separation of the New South Wales (NSW) sample of orange roughy from South Australian (off southeastern Kangaroo Island) and Tasmanian samples. This finding supports biological evidence for the presence of a distinct subpopulation of orange roughy in NSW waters. The four-base study also provided evidence of the presence of genetically distinct samples of orange roughy occurring in the same localities off southeastern Kangaroo Island from consecutive years. Additional sampling and the use of a greater number of four-base enzymes may be needed to determine if any genetic structuring exists among orange roughy south of New South Wales.  相似文献

The existence of three distinct populations is widely accepted for the finless porpoise (Neophocaena phocaenoides) in Chinese waters: the Yellow Sea, Yangtze River, and South China Sea populations. Here, we use nine species-specific microsatellite loci, the complete mitochondrial DNA control region (912 bp), and the complete mitochondrial cytochrome b gene (1,140 bp) to further investigate potential population stratification in the Yellow Sea using 147 finless porpoise samples from the Bohai Sea and adjacent northern Yellow Sea, two regions that were largely underrepresented in previous genetic studies. Our F-statistics analyses confirm the previously described three populations, but further demonstrate significant genetic differentiation between the [Bohai + northern Yellow] Sea and the southern Yellow Sea. On the other hand, median-joining network analyses do not exhibit well-differentiated haplotype groups among different geographic populations, suggesting the existence of shared ancestral haplotypes. Levels of microsatellite diversity are moderate to high (mean H E = 0.794) among the 147 [Bohai + northern Yellow] Sea finless porpoises and no recent bottleneck was detected, whereas mtDNA control region and cytochrome b gene diversity is low to moderate. The microsatellite genotypic and mtDNA haplotypic data also confirm the presence of mother-calf pairs in single-net bycatch cases. The results presented here highlight the necessity to treat the [Bohai + northern Yellow] Sea population (highly impacted by anthropogenic threats) as a separate Management Unit.  相似文献

The northern Pacific asteroid Asterias amurensis (Lütken) has been recently introduced to Tasmania and is now well-established in east and southeast Tasmania. This sea star is conspicuous throughout the Derwent River estuary and is particularly abundant in the Hobart port area. Reproduction of two populations of A. amurensis at Sullivans Cove and at Sandy Bay was investigated from August 1993 to October 1994 by gonadal histology and measurement of the gonad index (GI). An intense period of vitellogenic and spermatogenic growth started in April, with the breeding condition reached by June. In 1994, peak GI was recorded at Sullivans Cove in July and at Sandy Bay in August. Although these data indicated that the Sullivans Cove population spawned before the Sandy Bay population, histological examination revealed that major spawning activity occurred in both populations from July onwards. In the early part of the breeding season, gametogenesis proceeded in parallel with spawning, with released gametes being replaced by continual gametogenesis. Maintenance of a higher GI during the early part of the breeding season at Sandy Bay was due to prolonged gametogenic replacement at this site. Spawning continued to October 1994, resulting in a sharp decline in the GI. The cyclic expansion and regression of the genital haemal sinus coincident with germinal proliferation and growth, respectively, support the contention that haemal fluid provides nutrients for gametogenesis. In Tasmania, A. amurensis experiences temperature and photoperiod regimes similar to those experienced by endemic populations of this species in the north Pacific. Comparison of reproduction of A. amurensis in Japan and Tasmania at similar latitudes shows that gametogenesis of the northern and southern populations is 6 mo out of phase. This phase shift provides evidence for photoperiodic regulation of gametogenesis in A. amurensis. The similar trend in sea-temperature regimes associated with the stages of gametogenesis in northern and southern populations suggests that temperature also plays a modulatory role. Based on the periodicity of reproduction and the ontogeny of A. amurensis, the planktotrophic larvae of this species would be expected to be in the plankton for several months from the end of winter through summer. Considering the highly fecund, dispersive life-history of A. amurensis, the southeast Tasmanian populations have considerable potential to serve as a seed source for establishment of new populations of this asteroid elsewhere in Tasmania and in mainland Australia. An investigation was conducted to examine the species composition, abundance and biomass of benthic macrofaunal invertebrates in Oppa Bay, Miyagi Prefecture, Japan, in August 1984. Among 212 species identified, nearly half (105) were Polychaetes. Usually five species, Ampharete sp. 1, Neosabellides sp. 1, Nephtys polybranchia, Chaetozone sp. 1, and Lumbrineris longifolia were numerically dominant. The faunal features of dominant species were clearly divided into two areas using two indicator species, Amphalete sp. 1 and Neosabellides sp. 1. The former species decreased in numbers in the area from the river to the bay mouth, while the latter increased. Sediments consisted of sand at the river mouth and in the northern bay; and of silt-clay in the central bay, bay mouth and southern part of the bay. Gradients of species diversity (H) were expressed as a function of distance by linear regression. The relationship was positive (r=0.812) from the river to the bay mouth accompanied by increasing depth; and negative (r=-0.906) from northern to southern parts of the bay in proportion to the increase of silt-clay in sediments.  相似文献

Spatial and seasonal distribution pattern, life history and production of three species of Neomysis (Mysidacea) which commonly occur in northwestern subarctic Pacific coastal waters, were investigated throughout the year in the Akkeshi-ko estuary, northern Japan. The most abundant species Neomysis awatschensis (annual mean density: 179.8 inds. m−2, biomass: 108.8 mg DW m−2) occurred at the inner part of the estuary including low salinity areas with no clear preference for the seagrass bed. The second most abundant Neomysis mirabilis (mean density: 95.8 inds. m−2, biomass: 90.1 mg DW m−2) occurred at relatively saline seagrass site throughout the year. Occurrence of Neomysis czerniawskii in the estuary was limited to the seagrass bed during summer when their population mainly consisted of juveniles, suggesting that this species is a seasonal migrant between the estuary and the marine environment. Both N. awatschensis and N. mirabilis populations were composed of two generation types, a larger sized overwintering and smaller sized spring/summer generations; however, each species had a different reproductive strategy. N. awatschensis was characterized by fast growth to maturity at a smaller size than N. mirabilis with a relatively high fecundity during warm season, suggesting that this species is an r-strategist which can utilize opportunistically a wide variety of habitats. In contrast, the seagrass bed resident N. mirabilis was a K-strategist which matures at a larger size producing fewer but larger offspring. The annual production of N. awatschensis (0.57–0.70 g DW m−2, mean of the whole estuary) and N. mirabilis (0.58–0.68 g DW m−2, mean of the seagrass bed) at their respective habitats was comparable. Consequently, species-specific life history and distribution pattern are concluded to allow Neomysis spp. to coexist in the estuary and the high carrying capacity of seagrass bed is suggested to contribute to maintain their high biomass level.  相似文献
