The histopathological effects of malathion, an organophosphate pesticide, on the gill tissues in mosquitofish, Gambusia affinis, were determined by light microscope. The fish were exposed to sublethal concentrations (0.01 and 0.02 mg/L) of malathion for 10, 20 and 30 days. Histological examination of the gills treated with malathion showed a variety of histopathological effects. The gill lesions included necrosis and desquamation of secondary lamellar epithelium, lifting up of epithelium, intraepithelial oedema, fusion of adjacent secondary lamellae, haemorrhage at primary lamellae, disorganization and rupture in seconder lamellae, hypertrophy and hyperplasia of epithelial cells. These alterations were time- and dose-dependent.  相似文献

Histopathological alterations in gill, liver and kidney of common carp, Cyprinus carpio, intoxicated with sub-lethal concentrations of chlorpyrifos (O,O,-diethyl-O-3,5,6-trichloro-2-pyridyl phosphorothioate) pesticide (1 and 100 μg/L) for a period of 14 days were analyzed under light microscope. Gill exhibited hyperplasia and hypertrophy of gill epithelium, blood congestion, dilation of marginal channel, epithelial lifting, lamellar fusion, lamellar disorganization, lamellar aneurysm, rupture of the lamellar epithelium, rupture of pillar cells and necrosis. Alterations in hepatocytes were more pronounced, including nuclear and cellular hypertrophy, cellular atrophy, irregular contour of cells and nucleus, cytoplasmic vacuolation, cytoplasmic and nuclear degeneration, cellular rupture, pyknotic nucleus, necrosis and melanomacrophages aggregations. Histopathological lesions in kidney were cellular and nuclear hypertrophy, narrowing of tubular lumen, cytoplasmic vacuolation, hyaline droplet degeneration, nuclear degeneration, occlusion of tubular lumen, tubular regeneration, dilation of glomerular capillaries, degeneration of glomerulus and hemorrhage in Bowman's space. The most significant conclusion drawn from this study was that with the increased concentration and duration the toxicosis of chlorpyrifos would be enhanced as shown through the analysis of mean assessment value (MAV) and degree of tissue changes (DTC) also.  相似文献

The effect of 'safe application rate (SAR)' concentrations of the insecticides fenitrothion 50% EC (an organophosphate) and carbofuran 3% G (a carbamate) on histopathological changes in the thyroid gland have been studied during chronic exposure for 120 days (mid-April to mid-August). The studies reveal significant declines in the diameters of the follicle and the colloid of the thyroid, but a significant increase in the height of the epithelium, following exposure to both fenitrothion or carbofuran. In the fenitrothion treatment, invasion by blood corpuscles into the follicular lumen, following breakdown of the epithelium, was also observed. The degeneration of the epithelium and the loss of colloid in the follicles suggest that the fenitrothion treatment is more severe than the carbofuran treatment.  相似文献

In this study the great ramshorn snail (Planorbarius corneus), one of the most abundant gastropod of Turkish limnic systems, was investigated to determine the histopathological effects of endosulfan on the digestive gland, foot and mantle under laboratory conditions. Samples were collected from small artificial pools in Karaot at Gelendost-Isparta (southwest of Turkey), where agricultural activities are widespread. The snails were exposed to two sublethal concentrations of endosulfan (0.4 and 0.8 mg/l) for periods of 10, 20 and 30 days. Fifteen snails were kept in 2.5 l glass jars containing dechlorinated tap water and exposed under semi-static test (daily exchange of test water). All the testing was carried out on adult specimens, and snails were maintained on a photothermal period with 16 light hours at 22 +/- 2 degrees C. The histopathological examinations revealed the following changes: amoebocytes infiltration, dilatation in hemolymphatic spaces between the tubules, degeneration of cells, abnormal lumen, necrosis of cells and atrophy in the connective tissue of digestive gland; desquamation of the epithelium cells, changes in the number of mucocytes and protein gland cells, lipid vacuolus and atrophy of the columnar muscle fibers of the foot and mantle tissues. Pycnotic state of cells was also seen in the mantle tissues. Endosulfan caused significant histopathological alterations in the digestive gland, foot and mantle tissues of the snail, irrespective of concentrations of the pesticide and its exposure periods. The results are discussed, particularly in comparison to those of other aquatic organisms.  相似文献

The histopathological changes of fenvalerate on the gill, kidney, liver and intestine tissues of the Cirrhinus mrigala were determined by light microscopy. The fish were exposed to two sub-lethal concentrations of fenvalerate (1.5-3.0 ppb). The most common gill changes at all concentrations of fenvalerate were epithelial hyperplasia, epithelial necrosis, desquamation and lamellar fusion. Besides, epithelial lifting, oedema, swelling at the tips of secondary lamellae and curling of secondary lamellae were other histopathological changes. Necrosis of tubular epithelium, pycnotic nuclei in the hematopoietic tissue, hypertrophied epithelial cells of renal tubules, narrowing of the tubular lumen, expansion of space inside the Bowman's capsule and contraction of the glomerulus were observed in kidney tissues of fish. Hepatic lesions in the liver tissues of fish exposed to fenvalerate were characterized by congestion, cloudy swelling of hepatocytes and focal necrosis. Atrophy of epithelial cells, necrosis of epithelial cells, desquamation of mucosal epithelium and infiltration of lymphocytes into the lamina propria were detected in intestine tissues of fish after exposure to fenvalerate.  相似文献

Fish live in direct contact with their immediate external environment and, therefore, are highly vulnerable to aquatic pollutants. In this study, Oreochromis niloticus were caught at three different sites in Al-Hassa irrigation channels, namely Al-Jawhariya, Um-Sabah and Al-Khadoud. The histological changes in gills and liver were detected microscopically and evaluated with semi-quantitative analyses. Also, heavy metals have been determined in the water samples in these sites. Results showed that all sites were polluted by different kinds of heavy metals. Cd and Pb were mostly detected at concentrations above the WHO reference values. Meanwhile, various histopathological abnormalities were observed in gills and liver of fish specimens. In the gill filaments, cell proliferation, lamellar cell hyperplasia, lamellar fusion, lifting of the respiratory epithelium, and the presence of aneurysmal areas were observed. In the liver, there was vacuolization of the hepatocytes, sinusoidal congestion, necrosis of the parenchyma tissue, nuclear pyknosis, eosinophilic hepatocellular degeneration, pigment accumulation, an increase in the number and size of melanomacrophage centers. Liver tumors with severe chronic inflammation were occasionally found in fish at Al-Khadoud area (first-time report). The histological lesions were comparatively most severe in the liver. Despite heavy metals assessment did not show marked differences among sites, histopathological biomarkers indicated that the surveyed fish are living under stressful environmental conditions. So, we suggest use those biomarkers in future monitoring of aquatic systems.  相似文献

The chronic toxic effects of a prolonged exposure of 6 months, from January to June, to a 'safe dose' (0.20 mg litre(-1)) of a commercial organomercurial fungicide, Emisan (methoxy ethyl mercuric chloride: MeEHgCl), on the histophysiology of liver in adults and young (yearlings) of Channa punctatus, are described in this investigation. In both of the experimental groups, liver histology exhibited various abnormalities, including hyperplasia, nuclear pyknosis, fatty necrosis, and degeneration of hepatocytes leading to tumour and syncytium formation, which are indicative of carcinogenesis. Apart from this, blood vessel congestion and oedema were also observed in young treated fish. In the livers of both exposed groups, corresponding with this cellular damage, a marked reduction in hepatosomatic index, levels of total protein and lipid, and an elevation in cholesterol and acid and alkaline phosphatase contents were recorded. However, these alterations were more pronounced in young than in adult fish. These Emisan-induced, histopathological and biochemical changes are suggestive of severe hepatic dysfunction, which may result in the impairment of the physio-metabolic process, in this species.  相似文献

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) accumulates and remains stable in the fatty tissues and liver of rodents for a long time. Considering the pronounced difference between species, long-term, low dose hepatic effects of TCDD were investigated after subcutaneous administration of TCDD into rhesus monkeys during pregnancy. Macroscopic and histopathological examination of the liver carried out 4 y after TCDD administration demonstrated intrahepatic focal fatty changes, infarction, hemorrhage, microthrombi-formation, sinusoidal ectasia, small hepatocyte hyperplasia, and increased number of alpha-smooth muscle actin (alpha-SMA)-positive cells. An electron microscopic study disclosed sinusoidal endothelial cell degeneration and injury in the liver of TCDD-treated monkeys. Western blot analysis showed downregulation of aryl hydrocarbon receptor (AhR) protein expression and decreased level of vascular endothelial (VE) cadherin but increased expression levels of CYP1A1 and transforming growth factor beta (TGF-beta) protein in the liver tissues. These changes observed in TCDD-exposed monkeys indicated sinusoidal endothelial cell injury and impairment in intrasinusoidal microcirculation. Infarction, focal fatty change, and microthrombi-formation are considered to be closely associated with intrahepatic circulatory impairment. Increased number of alpha-SMA-positive cells and decreased level of VE cadherin expression in the liver tissues might also be associated with sinusoidal endothelial cell injury. In addition, downregulation of AhR expression and increased CYP1A1 protein levels in the liver were consistent with persistent effects of TCDD. Although it has been reported that TCDD induced endothelial cell injury, this is the first report to describe vascular disorders and protein expression in the liver after injection with TCDD in a primate model.  相似文献

Thiodan (33.7% endosulfan), a polychlorinated cyclodiene insecticide, was evaluated for its histopathological effects on mosquitofish, Gambusia affinis, by light microscopy. Fish were exposed to doses of 0.00 (control), 1.00, 2.50, and 5.00 microg/L on days 7, 14, 21, and 30. No histopathological effects were apparent at control group. The histopathological alterations were characterized as oedema, degeneration, accumulation of lymphocytes in the lamina propria, disintegration of villuses, pycnotic state of nuclei, and necrosis in gut; degeneration, hypertrophy, sinusoids enlargement, hemorrhage, pycnosis position of nuclei, vacuolization of cell cytoplasm, infiltration of mononuclear lymphocyte, and congestion in liver. The effects of the insecticide, diazinon (an organophosphorous compound), on the ovaries of bluegill (Lepomis macrohirus) were studied. Histological preparations of bluegill ovarian tissue was examined at 24, 48, 72 and 96 h and 1, 2 and 3 weeks following exposure to sublethal doses of diazinon (60 microg/l). The control contained primary follicles with an intact ovigerous lamellae and tunica albuginea. The control also revealed well developed stage IV oocytes with properly distributed provitelline nucleoli. After 24 h of exposure to diazinon, primary follicles began to show adhesion and cytoplasmic retraction in oocyte II occured. Cytoplasmic degeneration and additional adhesion and more retraction were visible at 48 h in oocyte II; 72 h brought forth adhesion, retraction and cytoplamic expulsion from oocyte IV. The number of atretic oocytes increased. Damages to the oocyte IV started to occur after 72 h of exposure. Cytoplasmic retraction and clumping was more visible at 96 h in oocyte IV. Partial destruction of the ovigerous lamellae and vitellogenic membrane occurred after 1 week. Two weeks continued to reveal destruction of follicles. Severe damage of the ovigerous lamellae, increased intrafollicular spaces, vacuolated cytoplasm, extrusion of karyoplasm and necrosis in the cytoplasm were most evident following 3 weeks of exposure. The ovarian wall became frayed and broken. Additionally, a marked increase of atretic follicles, shrinkage, and embedded nucleoli into the surrounding cytoplasm in oocyte II, III and IV were observed at week 3. This study revealed that oocytes at their different stages of maturation get affected differently at various exposure. Based on observations of the ovarian tissue compositional and structural changes following given exposure times, it becomes evident that consistent sublethal doses of diazinon can and will alter microscopic anatomy of the fish ovary.  相似文献

Abstract

The subchronic toxicity of acridine was investigated in rats following dietary exposure at 0, 1, 10, 100 and 500 ppm for 13 weeks. The growth rate and food consumption were not affected by treatment and no clinical signs of toxicity were observed. There was a slight but significant decrease in spleen weight, both in absolute terms and as a percent of body weight, in the 500 ppm males and a slight increase in absolute thymus weight in the females of the same dose group. Both hepatic ethoxyresorufin O‐deethylase (EROD) and pentoxyresorufin O‐dealkylase (PROD) activities were slightly, but significantly, elevated in females in the 500 ppm dose group. No haematological or other biochemical changes were observed. Females also displayed dose‐related increases in inorganic phosphate and uric acid levels. Treatment‐related histopathological changes were seen in the thyroid, liver and kidney and included hepatic anisokaryosis and vesiculation of nuclei and glomerular adhesions, reticulin sclerosis and nuclear pyknosis in the kidney. Residue data showed a dose‐dependent accumulation of acridine in liver, kidney and adipose with the highest concentration being found in the fat of the 500 ppm dose group. Based on these data, the no observable adverse effect level of acridine was judged to be 100 ppm or 12 mg/kg bw/day.  相似文献

Vitamin B17 (VB17), also known as amygdalin and laetrile, is a type of carbohydrate occurring naturally in many plants, such as apricot kernels which have obtained a great interest in cancer therapy. This study aimed to investigate the hepatic protective potential of VB17 against Ehrlich ascites carcinoma (EAC)–bearing mice-induced liver injury, DNA damage, apoptotic P53, and PCNA alterations. A total of 100 female mice were divided into 5 groups (1st group, control group; 2nd group, VB17 group; 3rd group, EAC group; 4th group, pre-treated EAC with VB17; 5th group, co-treated EAC with VB17). Results showed that the presence of VB17 in pre-treated and co-treated groups lead to decreased DNA damage, microsomal protein, NADPH cytochrome c reductase, alpha-fetoprotein (AFP), AST, ALT, and ALP while showed increased cytochrome b5, cytochrome P450 amidopyrine N-demethylase, and aniline 4-hydroxylase compared with the EAC group. Many histopathological changes were observed in liver sections in EAC as moderate fibrosis and marked diffuse necrosis of hepatic tissue, marked inflammatory cells, and congested blood sinusoids. On the other hand, there was a moderate degree of improvement in hepatocytes in liver sections in pre-treated VB17+EAC, while a mild degree of improvement in hepatocytes, moderate cellular infiltrations, and moderate cytoplasmic vacuolization of hepatocytes in liver sections in co-treated EAC+VB17. In addition, there was a depletion in hepatic P53 and PCNA protein expression compared with the EAC group. It could be concluded that VB17 has a potential hepatoprotective effect against EAC cell–induced liver toxicity.

Haematological (WBC, RBC, Hgb and Hct) and genotoxicity (MNT) parameters, hepatic enzymatic activities (GST, GPx and GR), and a histopathological evaluation of liver, kidneys and gonads were assessed as general biomarkers of metal pollution in the shrew Crocidura russula inhabiting a pyrite mining area. Specimens exposed to metals presented a few significant alterations when compared with reference animals: GST activity decreased; micronuclei increased; and evident liver alterations related to metal exposure were observed. On the basis of all the parameters studied, age was an important factor that partly explained the observed variation, whereas sex was the least important factor. Significant correlations were also found between heavy metal concentrations and biomarkers evaluated, demonstrating the great influence of these metals in the metabolic alterations. To the best of our knowledge, these data constitute the first measurements of a battery of biomarkers in shrews from a mine site and are among the few available for insectivorous mammals.  相似文献

We have observed the effect of copper and zinc on the biology of Euglena gracilis. The cells displayed different sensitivities to these metals, as the apparent LC50 for Cu2+ was 0.22 mM, and for Zn2+ it was 0.88 mM. While Zn2+ was able to increase cell proliferation even at 0.1 mM, the minimal CuCl2 concentration tested (0.02 mM) was sufficient to impair cell division. Higher concentrations of these metals not only inhibited cell division in a concentration-dependent manner, but also interfered with the metabolism of E. gracilis. A higher accumulation of proteins and lipids per cell was observed at the DI50 concentration for metal-treated cells. These results suggest that the test concentration of both metals leads to a failure in completing cell division. Ultrastructural analysis indicated a chloroplast disorganization in copper-treated cells, as well as the presence of electron dense granules with different shapes and sizes inside vacuoles. Microanalysis of these granules indicated an accumulation of copper, thus suggesting a detoxification role played by the vacuoles. These results indicate that E. gracilis is an efficient biological model for the study of metal poisoning in eukaryotic cells. They also indicate that copper and zinc (copper being more poisonous) had an overall toxic effect on E. gracilis and that part of the effect can be ascribed to defects in the structure of chloroplast membranes.  相似文献

To better understand the relationships between pulp manufacturing processes and mixed function oxidase (MFO) enzyme induction in fish, a practical and standardized exposure procedure is required. This study was undertaken to develop a laboratory-based exposure procedure to quantify the relative MFO induction potencies of different types of pulp and paper mill effluents. One major consideration in developing the procedure was to ensure that the protocol was practical so that tests could be performed in a short time, with small volumes of effluents and using simple experimental conditions. A series of concentration-response and time-course experiments were conducted to find the minimum time and effluent concentration which could distinguish the ability of different effluents to cause significant MFO induction in rainbow trout in the laboratory. Experiments were also conducted to determine the effects of biotic and abiotic factors such as loading density, fish size and feeding regime. This study showed that the exposure of rainbow trout in the laboratory to 10% concentration of secondary-treated effluent for 96 h caused significant increases in hepatic MFO activity. The magnitude of MFO induction was comparable to other field and laboratory observations. While fish size, loading density and feeding regime were found to affect the test results, consistent responses within a laboratory using this protocol are possible, provided that these factors are standardized. Therefore, the short-term exposure approach described in this paper could be a relevant tool for assessing the ability of different types of pulp and paper mill effluents to cause MFO induction in fish.  相似文献

The purpose of the present study was to research the sublethal and/or lethal effects produced by the exposure of fish and shellfish to the γ isomer of lindane, γ-hexachlorocyclohexane (γ-HCH). The teleostean fish Sparus aurata and the shellfish Crassostrea angulata and Scrobicularia plana, were exposed to 16 μ g/L of lindane for 15 days. Samples of different fish (liver, kidney and gills) and shellfish (gills, gut, digestive gland and mantle) tissues were extracted and processed for histopathological observations. Although mortality was not detected during the bioassay, sublethal effects (histopathological alterations) were observed. Vacuolization in the liver cells and lamellar fusion in gills from exposed fish were observed. Disorganization of normal gill structure, epithelial desquamation with the disappearance of apical ciliature in intestine, and inflammatory response in mantle from exposed shellfish were also observed. Thus, it can be concluded that the lindane concentration employed in the present research did not produce lethal effects in the exposed organisms but it caused sublethal effects. Lindane has time-dependent multiple toxic effects in S. aurata, C. angulata and S. plana, which were more severe at the end of the experimental time. The toxicological implications arising from these results are subjects for further multiconcentration tests dealing with lethal responses (mortality) or with sublethal responses (cellular/molecular biomarkers) of the aforementioned species.  相似文献
