针对雅鲁藏布江中游河谷黑颈鹤国家级自然保护区黑颈鹤生境适宜性的制约因素及其面临的主要威胁,利用生境适宜性指数、保护比例和保护效率指数等对雅鲁藏布江中游河谷黑颈鹤国家级自然保护区日喀则片区黑颈鹤生境适宜性及保护现况进行评价并分析其变化。结果表明,1994、2005和2015年黑颈鹤适宜生境与次适宜生境面积总和占比依次为31.05%、32.00%和41.00%,黑颈鹤可生存空间逐渐增长;对比1994和2015年保护比例和保护效率指数发现,适宜觅食生境和夜栖生境分布趋向核心区和缓冲区,次适宜觅食生境同样向核心区移动,保护区规划对适宜觅食生境和夜栖生境保护的合理性降低,对次适宜觅食生境的合理性升高,从外延区对各生境的保护比例来看,保护区周边仍存在大量黑颈鹤可利用生境没有纳入保护范围。  相似文献

利用1次SephadexG50凝胶过滤和2次QAESephadexA25离子交换层析的方法,从最后6d用CuCl2处理(ρ=20mg/L)、20d龄的紫羊茅(Festucarubracv.Merlin)根中分离纯化了1个铜结合肽(CuBP2a),该肽在SuperdexPeptideHR10/30预装柱的FPLC凝胶过滤中在λ254nm处只出现一个峰,经分子量标准曲线估计,其Mr约为1400,在DeltaPakC18的反相HPLC中,在λ254nm处也只有1个紫外吸收峰值,这都说明该肽已经达到了较高的的纯度.通过光谱分析,发现该肽的最大紫外吸收值在248.1nm处.对Cu结合物在植物体内的存在状态进行了讨论.  相似文献

从粗壮女贞(Ligustrum robustum)叶子中分离鉴定了4 个新的单萜配糖体, 命名为粗壮女贞甙D、G、H和L. 通过波谱及化学方法,它们分别鉴定为香叶醇3′OαL吡喃鼠李糖基4′顺式香豆酰基βD吡喃葡萄糖甙(1) ,6羟基3,7二甲基2E,7葵二烯醇3′OαL吡喃鼠李糖基4′反式香豆酰基βD吡喃葡萄糖甙(2) ,6羟基3,7二甲基2E,7葵二烯醇3′OαL吡喃鼠李糖基4′顺式香豆酰基βD吡喃葡萄糖甙(3) 以及6,7二羟基3,7二甲基2E葵烯醇3′OαL吡喃鼠李糖基4′顺式香豆酰基βD吡喃葡萄糖甙(4) . 此外,还从该植物得到了7 个已知化合物,芹菜素(5)、cosmosiin(6) 、rhoifolin(7) 、山梨糖醇(8)、阿克苷(9) 、β谷甾醇(10) 和齐墩果酸(11) .  相似文献

黑颈鹤(Grus nigricollis)是国家I级重点保护野生动物,易危物种,是全球唯一生活于高原、最晚被人类识别和记录的鹤种.黑颈鹤迁徙途中需选择适宜的生境作短暂停歇,以补充体能或夜栖,但目前关于其中途停歇地点选择的信息缺乏. 2014年12月10日和2023年3月22日,在四川省九龙县洪坝乡(102°3′14″E,29°13′23″N,海拔2 189 m)、泸定县二郎山森林公园(102°13′44″E,29°49′30″N,海拔1 824 m)各拍摄到一只黑颈鹤.其中洪坝飞往南方越冬地的成体在灌木林中空地停留约50 min,二郎山飞往繁殖地的亚成体在临近318国道和居民点的针阔混交林中停留约41 min.本研究补充了黑颈鹤东部种群迁徙停歇地点的信息,可为当地林草部门开展有效保护提供支撑.(图3参18)  相似文献

用8种限制性内切酶酶解茶尺蠖Ectropisobliqua核型多角体病毒(EoNPV)基因组DNA,获得大于0.88kb的片段数分别为7、38、19、17、16、9、32和14.由此统计,基因组平均大小大于118.5kb,即Mr约大于78.6×106.用BmNPV的多角体蛋白(ph)基因和AcMNPV的egt基因为探针,应用Southern杂交方法,将EoNPV的ph基因定位于BamHIA、ClaID、EcoRIM、EcoRVL、HindⅢF、PstIA、SalIH和XhoIB片段上,将egt基因定位于BamHIA、ClaIl、EcoRIK、EcoRVA、HindⅢH、PstIA、SalIB、XhoII片段上.通过克隆和酶切鉴定EcoRIM和EcoRVL片段,测定ph基因部分序列,并以EcoRIM为探针对基因组酶切印迹进行杂交,制作了EoNPVph基因和egt基因区的酶切图谱,推定egt基因保守区位于ph基因上游约4.55kb处.  相似文献

采用聚乙烯醇(PVA)加少量海藻酸钠及活性碳的方法对一株能高效降解甲胺磷的细菌假单胞菌B82菌株进行固定化.正交试验结果表明,三者合适配比为聚乙烯醇100(ρ/gL-1),海藻酸钠5(ρ/gL-1),活性碳50(ρ/gL-1).此方法制得的凝胶颗粒机械强度好、经久耐用.固定化B82凝胶小球能在pH6~11的范围内发挥其降解甲胺磷的作用,其最适作用温度为35℃.B82菌经固定化后,降解甲胺磷的速度明显加快.  相似文献

从贵州产雷公藤(TripterygiumwilfordiHokF.)中分离得到8个化合物,其中一个为新的降倍半萜化合物雷公藤酮(1),根据光谱(IR,1HNMR,13CNMR,HRMS,CD等)数据分析并通过X射线晶体分析确定了结构;另外7个化合物分别鉴定为雷公藤素(2),(±)6氧2(4′羟基3′,5′二甲氧苯基)3,7二氧杂二环[3.3.0]辛烷(3),tripterifordin(4),雷公藤内酯酮(5),雷公藤内酯醇(6),雷公藤内酯甲(7)和对乙氧基乙酰替苯胺(8).其中,3和8均为首次从该植物中分离得到.用二维核磁共振谱对3的氢谱和碳谱进行了指定  相似文献

由甘薯叶片诱导出愈伤组织,并由此建立起悬浮培养细胞系.分析其生长表明:南薯88和81294两种细胞系生长曲线均呈“S”型,其中南薯88生长较快,是81294的3倍以上,10d培养物的鲜重是初始鲜重的10倍.培养条件研究结果表明:南薯88细胞悬浮培养的最佳2,4D质量浓度为1.0mgL-1,最佳蔗糖质量浓度为30gL-1,椰乳和酵母汁能明显促进其生长;对于81294,2,4D最佳质量浓度为2.5mgL-1、蔗糖最佳质量浓度15gL-1,酵母汁可促进其生长,但椰乳却无效;谷氨酰胺、色氨酸和脯氨酸不能促进甘薯悬浮细胞的生长,水解酪蛋白对其生长的促进作用亦不大;葡萄糖则抑制其生长  相似文献

研究了苏云金芽孢杆菌无质粒突变株BMB171的转化性能和表达cry1Ac和cry1Ca基因的性能.用pHT3101、pBMB305、pBMB1736、pBMB671、pBTL1和pHV1249等6种外源质粒电转化无质粒突变株BMB171,其转化频率分别是Bt4Q7、Bt4D10和Bti.IPS.78/11等3种常用受体菌相应最高转化频率的1000、6.7、12.5、66.7、3500和2倍,其每μgDNA的最高转化子数达107.导入BMB171的外源质粒的稳定性与其复制子类型和质粒大小有关.无质粒突变株BMB171表达cry1Ac基因的表达量高于对照受体菌Bt4Q7,略低于Bt4D10和Bti.IPS.78/11,而BMB171表达cry1Ca基因的表达量高于这3种受体菌;对小菜蛾3龄幼虫和甜菜夜蛾初孵幼虫的毒力测定结果表明,BMB171表达cry1Ac基因的杀虫毒力高于Bt4Q7和Bt4D10,略低于Bti.IPS.78/1;而表达cry1Ca基因的毒力高于这3种受体菌  相似文献

In the conservation of endangered species, suppression of a population of one native species to benefit another poses challenges. Examples include predator control and nest parasite reduction. Less obvious is the control of blood-feeding arthropods. We conducted a case study of the effect of native black flies (Simulium spp.) on reintroduced Whooping Cranes (Grus americana). Our intent was to provide a science-driven approach for determining the effects of blood-feeding arthropods on endangered vertebrates and identifying optimal management actions for managers faced with competing objectives. A multiyear experiment demonstrated that black flies reduce nest success in cranes by driving incubating birds off their nests. We used a decision-analytic approach to develop creative management alternatives and evaluate trade-offs among competing objectives. We identified 4 management objectives: establish a self-sustaining crane population, improve crane well-being, maintain native black flies as functional components of the ecosystem, and minimize costs. We next identified potential management alternatives: do nothing, suppress black flies, force crane renesting to occur after the activity period of black flies, relocate releases of cranes, suppress black flies and relocate releases, or force crane renesting and relocate releases. We then developed predictions on constructed scales of 0 (worst-performing alternative) to 1 (best-performing alternative) to indicate how alternative actions performed in terms of management objectives. The optimal action depended on the relative importance of each objective to a decision maker. Only relocating releases was a dominated alternative, indicating that it was not optimal regardless of the relative importance of objectives. A rational decision maker could choose any other management alternative we considered. Recognizing that decisions involve trade-offs that must be weighed by decision makers is crucial to identifying alternatives that best balance multiple management objectives. Given uncertainty about the population dynamics of blood-feeding arthropods, an adaptive management approach could offer substantial benefits.  相似文献

We studied the behavior of 13 radiotagged cranes dispersing from a communal roost over days when they changed their main daily foraging area between consecutive days during two winter seasons. Individuals went to a new foraging zone when on the previous day their morning food intake had fallen below their mean morning food intake measured over the whole winter. Food intake on the day before a change in foraging area was positively correlated with dominance rank. Dominant cranes changed to new zones with higher numbers of birds and food density, while subordinate cranes went to new zones with lower numbers of birds. As a result, all birds increased their food intake over that of the previous day. Dominant cranes remained more faithful to their most preferred foraging zone, where they spent 69% of the mornings, while subordinate birds were more mobile, switching among zones frequently. Dominant birds left the roost later than subordinate birds on the days they changed to a new zone, which could be used to track the main departing flows. The results suggest that the dynamics that led to a truncated phenotype-limited distribution were determined by social dominance and food abundance, with dominant cranes shifting to a new zone to maintain their high intake levels and subordinates changing more frequently whenever their daily intake did not reach the minimum metabolic requirements. Cell cultures have been established from 33 individual black rhinoceroses. These were from wild populations from various localities in southern Africa and include representatives from three geographical regions (southwestern, south-central, and eastern) corresponding to currently accepted conservation units, and include individuals previously attributed to one of the four subspecies, Diceros b. minor, D. b. bicornis, D. b. michaeli , and D. b. chobiensis (du Toit et al. 1987). Comparative mitochondrial DNA restriction maps were constructed using 16 restriction enzymes. These showed in each case two site differences between representative individuals from any two of the above geographical regions. Maps were monomorphic within geographical regions and, therefore, have the potential to provide diagnostic markers. The map from a single individual attributed to the D. b. chobiensis subspecies was identical to other individuals (attributed to D. b. minor ) in the south-central geographical region. The low amount of genetic diversity implied by these few differences renders it unlikely that problems with outbreeding depression will arise if, given the continuing decline in numbers of black rhinoceroses, it becomes necessary to supplement wild or captive populations with individuals from a different conservation unit in order to avoid inbreeding depression.  相似文献

The Japanese National Biodiversity Strategy 2010 calls for the creation of ecological networks as a biodiversity conservation policy. However, there is an obvious lack of information on the spatial distribution of many species and a lack of scientific methods for examining habitat requirements to establish the need for constructing these networks for target species. This study presents a quantitative method for assessing the need for ecological networks through modeling the potential geographic distributions of species based on a case study of local populations of Asiatic black bear (Ursus thibetanus japonicus) in Fuji and Tanzawa, Japan. A total of 1541 point records of occurrences of Asiatic black bears and 11 potential predictors were analyzed in a GIS environment. After a predictive distributional map was obtained using the Maximum Entropy (MaxEnt) algorithm, a gap analysis was carried out and population size was estimated. Approximately 24% of the bear's predicted habitat area fell within a wildlife protection area, 2% within a nature reserve, and 37% within natural parks. Conservation forest comprised 54% of the total area of predicted habitat; of this, national forest comprised 2%, and private and communal forest comprised 37%. The total estimated Asiatic black bear population in this region was 242, with 179 individuals in the Fuji local population, 26 in the Tanzawa local population, and 37 in the corridor patch between the two local populations. Our study also found a potential corridor connecting the Fuji and Tanzawa local populations, as well as potential habitat corridors in the Fuji region containing subpopulations on Mt. Fuji (119 individuals) and Mt. Kenashi (53 individuals). An additional subpopulation on Mt. Ashitaka (7 individuals) is isolated and not fully protected by a zoning plan. Mt. Furo's subpopulation is considered to be almost extinct, although black bears were observed here until 2002 based on the report by Mochizuki et al. (2005). The total black bear population of the Fuji-Tanzawa region is considered to be “endangered”; thus, an adequate population size might be difficult to maintain even if this region were to be internally connected by means of an ecological network.  相似文献

Mediterranean sandhoppers of the species Talitrus saltator (Montagu) from the Tyrrhenian population of Burano (Grosseto, Italy) were tested by rearing under different conditions in order to enquire into the innateness and modifiability of orientation with respect to substrate slope and landscape features. Tests were conducted from November 1990 to November 1991. The results were as follows: the sandhoppers responsed to slope only by orienting themselves downslope on dry substrate and upward on a wet one. These responses appeared to be innate but modifiable by experience. Visual dishomogeneity (half horizon covered with a black strip), with a horizontal substrate, elicited an innate orientation towards the black shape, again modifiable by experience. When a black strip was positioned around the upper half of the inclined arena, the sandhoppers responded differently, dependent upon their prior experience: wild individuals responded to the visual stimulus, amphipods reared in optical homogeneity did not make a choice and those reared in conditions identical to those of the test arena responded to slope by orienting downwards and away from the balck strip. With a black strip around the lower half of the inclined arena and with a dry substrate identical responses were shown by wild-caught and laboratory-reared sandhoppers, i.e., downslope and towards the black strip.  相似文献

Satellite-tracking of 15 White-naped Cranes ( Grus vipio ) from their Japanese wintering grounds through the Korean Peninsula shows that there are four important regions for conserving migrant cranes: the Three Rivers Plain, the People's Republic of China; Lake Khanka, Russia; Kumya, North Korea; and—most importantly—the demilitarized zone of the Korean Peninsula. Two sites along the Korean demilitarized zone, Panmunch'om and Ch'olwon, were the most heavily used stopover sites, and they present complex international conservation challenges. Cranes stopped at these sites for up to 87.1% of their total migration time; cranes migrating to Zhalong Nature Reserve, China, made it their only lengthy stop. We report the migration routes and the importance of the identified stopover sites, and we outline conservation issues at those sites.  相似文献

In field tests of the information-center hypothesis (ICH) in south Texas with black vultures (Coragyps atratus) and turkey vultures (Cathartes aura), large carcasses were provided and kept under continuous observation. The use vultures made of these bait sites and their patterns of arrival were recorded to evaluate predictions derived from the ICH. Turkey vultures discovered most bait sites (30 of 31) first, but frequently were displaced from the food by later-arriving black vultures. This competitive exclusion by black vultures limited subsequent feeding opportunities for turkey vultures sufficiently that few (27%) returned on subsequent days to bait sites they had previously visited. I found no evidence that those turkey vultures that did return to bait sites acted as leaders for groups of naive birds and led them to bait sites – knowledgeable and naive turkey vultures did not arrive at bait sites together, and groups arriving at bait sites were not larger on subsequent days than on the first days carcasses were available. In contrast, a significantly larger percentage (47%) of knowledgeable black vultures returned to bait sites they had visited on previous days, and the first groups of black vultures arriving at bait sites on subsequent days were significantly larger than the equivalent groups on first days. Nine flocks of black vultures that arrived on subsequent days at bait sites before sunrise (which suggests the birds had commuted directly from a roost) contained knowledgeable birds, and two of these flocks contained both knowledgeable and naive individuals. Overall, 10 of 54 naive tagged black vultures (18.5%) arrived at bait sites under circumstances that suggested they had followed conspecifics to the food from a roost. However, most black vultures apparently found carcasses through independent search or by using local enhancement. Therefore, I conclude that while following from roosts to food sites is a strategy used by black vultures, at this study site it is one they use relatively infrequently. 本文研究了植物生长调节剂多效唑(MET)在土壤中的环境引为,MET在土壤中降解较缓慢并随土壤环境条件而异,淹水厌氧条件比旱地好氧条件降解更缓慢,灭菌土壤中未见明显降解,微生物培养试验表明,MET对真菌有明显抑止作用,以MET为唯一营养源的培养基上能生长出几种放线菌,在三种供试土壤中,MET在高有机质含量的黑土中吸附常数远大于黄棕壤和潮土;MET的淋溶则在砂粒含量较高和有机质含量较低的潮土中最快,黑土最慢,黄棕壤居间.  相似文献

Abstract: The Whooping Crane ( Grus americana ) is an endangered bird that suffered a severe population bottleneck; only 14 adults survived in 1938. We assessed the genetic effect of this human-caused bottleneck by sequencing 314 base pairs ( bp) of the mitochondrial DNA control region from cranes that lived before, during, and after this bottleneck. The maximum length of DNA amplifiable from museum specimens was negatively correlated with age, and only 10 of 153 specimens yielded the entire 314 bp sequence. Six haplotypes were present among the prebottleneck individuals sequenced, and only one of these persists in the modern population. The most common modern haplotype was in low frequency in the prebottleneck population, which demonstrates the powerful effect of genetic drift in changing allele frequencies in very small populations. By combining all available data, we show that no more than one-third of the prebottleneck haplotypes survived the human-caused population bottleneck. High levels of variation of substitution rates among nucleotide sites prevented us from estimating the prebottleneck population size. Our data will be incorporated into the captive breeding program to allow better management decisions regarding the preservation of current genetic diversity. These data offer the first glimpse into the genetic toll this species has paid for human activities.  相似文献
