Polychlorinated biphenyls and methylmercury are two of the most ubiquitous environmental contaminants in Guizhou province. Rice is eaten with almost every meal and provides more calories than any single food in Guizhou province. The estimated tolerable daily intake of total mercury, MeHg, Se and PCBs from Guizhou contaminated rice by Chinese people showed that MeHg and/or PCBs exceeded the corresponding limits. The aim of this study was to characterize the effects of exposure to environmental contaminated rice on neurobehavioral development and neurobiological disruptions in mice. Animals were treated from postnatal day (PND) 22 to 91. At PND 26–91 days of age, mice were tested for neurobehavioural development and neurochemical level changes. We showed that dietary exposure to environmentally contaminated rice gave rise to different changes in antioxidants. Reduced superoxide dismutase (SOD) activity and excess increased nitric oxide (NO) indicated aggravation of oxidative status after long-term dietary intake of Hg and PCBs. Neurobehavioral derangement in the central nervous system and significant delay in the Morris water maze test response on PND 91 are correlated with the increased of c-fos/c-jun expression levels in the cerebral cortex. These results suggest that MeHg neurotoxicity might be a greater hazard than that associated with PCB, but PCB may augment the neurobehavioral deficits caused by increased levels of mercury exposure. The simultaneous intake of selenium might have a protective effect on Hg accumulation in the body, and vitamin C might protect mice against the toxic effects of PCBs. However, the protective role of Se and vitamin C is very limited for multiple-agent pollution. Immediately early genes in the brain response to contaminated rice might be dependent on interaction among NO, NO synthase (NOS), SOD and reduced glutathione (GSH). We should be alert to mental health problems in human beings when any kind of Hg- and PCB-polluted food is consumed.  相似文献

The aims of the current study were to assess plasma concentrations and predictors of halogenated phenolic compounds (HPCs), polychlorinated biphenyls (PCBs) and chlorinated pesticides in a representative group of postmenopausal Norwegian women (n = 311, mean age: 56 years). In addition to delivering a blood sample, the participants had also filled out a detailed questionnaire regarding their current health status, use of medication and dietary habits, etc. The association between a large number of dietary variables, lifestyle factors and plasma concentration of organic pollutants were investigated using multivariate statistics.Within the current study group, p,p′-DDE (median: 903 ng/L w.w.) and pentachlorophenol (PCP, median: 711 ng/L w.w.) were the dominating compounds on a wet weight basis and were present in considerable higher concentrations than the PCBs, the hydroxylated PCBs (OH-PCBs) and the remaining chlorinated pesticides. Of the PCBs, PCB 153 was detected in highest concentrations, whereas 4-OH-PCB187 was present in highest concentration of the selected OH-PCBs. The ratio of ΣOH-PCB to ΣPCB varied between 0.06 and 0.77 ng/L and were significantly lower in individuals with higher ΣPCB concentration. PCP did not co-vary with the other contaminants, indicating a different route of exposure. No clear food cluster was positively associated with PCP exposure; however intake of marine food was negatively linked to PCP concentration and age and being a non-smoker were positive predictors of PCP. The other investigated contaminants co-varied and were positively associated with age, plasma concentrations of marine derived omega-3 fatty acids and a traditional Norwegian fish and bread based diet. The difference in contaminant concentration between high, medium and low consumers of total marine food were however marginal for the dominating PCBs and chlorinated pesticides. The current results indicate however that marine food in itself could be a source of some of the OH-PCBs, regardless of their PCB precursors.As PCP is one of the dominating organic contaminants within the general female Norwegian population, future research on human concentrations, exposure routes and potential health effects of PCP is encouraged. Continued monitoring of human OH-PCB levels should also be performed as they could be present at levels almost as high as the PCBs and they are expected to be more toxic than their mother substances.  相似文献

We determined the contamination status and accumulation profiles of polychlorinated biphenyls (PCBs), hydroxylated PCB congeners (OH-PCBs), polybrominated diphenyl ethers (PBDEs), hydroxylated PBDEs (OH-PBDEs), methoxylated PBDEs (MeO-PBDEs), and bromophenols (BPhs) in serum from e-waste recycling workers and residents near a coastal area in India. Residue levels of penta- to octa-chlorinated PCBs, penta- to octa-chlorinated OH-PCBs, 6MeO-BDE47, 6OH-BDE47, and 2,4,6-tri-BPh in serum from residents living near the coastal area were significantly higher than those in serum from e-waste recycling workers. Residue levels of tri- to tetra-chlorinated PCBs, tri- to tetra-chlorinated OH-PCBs, PBDEs, octa-brominated OH-PBDEs, and tetra-BPhs in serum from e-waste recycling workers were higher than those in serum from residents living near the coastal area. Principal component analysis revealed that residents living near the coastal area and e-waste recycling workers had different serum profiles of chlorinated and brominated compounds.  相似文献

Polychlorinated biphenyls (PCBs) are present in the environment as complex mixtures, which make it challenging to identify PCB congeners that may be subject to active transport processes. Here we employ a transgenic mouse model in combination with multivariate analyses to investigate if chiral PCBs 91, 95, 132, 136, 149, 174, 176 and 183 are subject to active (enantioselective) transport by multidrug resistance (MDR) transporters. A synthetic PCB mixture containing these congeners was administered orally to female FVB or mdr1a/1b knockout mice. Due to the short half-life of chiral PCB congeners, mice were euthanized after 24 h and PCB concentrations and enantiomeric fractions were determined in selected tissues and excreta. Principal component analysis did not reveal differences between wild-type and mdr1a/1b knockout mice. However, Hotelling T2-test revealed significantly lower PCB concentrations and a more pronounced enantiomeric enrichment in the adipose tissue of mdr1a/1b knockout mice. These differences are due to higher body weights and higher fecal fat contents of mdr1a/1b knockout mice. Analysis of the enantiomeric fractions of PCBs 91, 95, 136, 149 and 174 showed a significant enantiomeric enrichment for all five congeners in wild-type and mdr1a/1b knockout mice. Overall, by studying a PCB mixture in a transgenic mouse model in combination with a multivariate data reduction approach, PCBs 91, 95, 136, 149 and 174 could be excluded as substrates of multidrug resistance transporters 1a/b.  相似文献

It was recently hypothesized that pets may serve as sentinels to explore human exposure to organohalogenated chemicals (OHCs) via indoor environments and adverse health effects. The current study investigates OHCs contamination in domestic cats suffering from diabetes mellitus (DM), particularly DM induced by acromegaly and a form of DM akin to human type 2 DM (T2DM). Plasma from three groups of domestic cats was analyzed: acromegaly induced DM, T2DM and age matched control cats without DM. Analytes targeted included organochlorine pesticides, polychlorinated biphenyls (PCBs), and polybrominated diphenyl ethers (PBDEs), together with their hydroxylated (HO-) metabolites. Similar PCB profiles were measured in cat plasma compared to humans, while the PBDE profile (dominated by BDE-99 (48%–55%) and BDE-47 (19%–25%)), the PCB and PBDE metabolite profiles were different in cat plasma than found in humans. Significantly higher OHC concentrations were recorded in plasma of acromegalic cats compared to the other two groups. Group differences in the PCBs/HO-PCBs ratios suggest that acromegalic cats have a lower capacity to metabolize persistent OHCs, like PCBs, than diabetic cats or cats without an endocrinopathy. As pituitary tumorigenesis in animals can be induced by estrogens, and PCBs may act as xenoestrogens, further investigation into whether there could be a causative link with the induction of feline acromegaly is warranted. Interestingly, BDE-47/BDE-99 ratios in cats were similar to the ratios in house dust. The results of this study suggest that domestic cats may represent a good model to assess human exposure to chemicals present in indoor dust.  相似文献

Studies of environmental and toxic effects of polychlorinated biphenyls (PCBs) are ideally performed with PCB mixtures reflecting the composition of environmental PCB profiles to mimic actual effects and to account for complex interactions among individual PCB congeners. Unfortunately, only a few laboratory studies employing synthetic PCB mixtures have been reported, in part because of the challenges associated with the preparation of complex PCB mixtures containing many individual PCB congeners. The objective of this study was to develop a PCB mixture that resembles the average PCB profile recorded from 1996 to 2002 at a satellite station of the Integrated Atmospheric Deposition Network located at the Illinois Institute of Technology (IIT) in Chicago, Illinois, using commercial PCB mixtures. Initial simulations, using published Aroclor profiles, showed that a mixture containing 65% Aroclor 1242 and 35% Aroclor 1254 was a good approximation of the target profile. A synthetic Chicago air mixture (CAM) was prepared by mixing the respective Aroclors in this ratio, followed by GC/MS/MS analysis. Comparison of the PCB profile of the synthetic mixture with the target profile suggests that the synthetic PCB mixture is a good approximation of the average IIT Chicago air profiles (similarity coefficient cos θ = 0.82; average relative percent difference = 84%). The synthetic CAM was also a reasonable approximation of the average of 184 PCB profiles analyzed in 2007 at 37 sites throughout Chicago as part of the University of Iowa Superfund Basic Research Program (isbrp), with a cos θ of 0.70 and an average relative percent difference of 118%. While the CAM and the two Chicago air profiles contained primarily di- to pentachlorobiphenyls, higher chlorinated congeners, including congeners with seven or eight chlorine atoms, were underrepresented in the synthetic CAM. The calculated TCDD toxic equivalency quotients of the synthetic CAM (2.7 ng/mg PCB) and the IIT Chicago air profile (1.6 ng/mg PCB) were comparable, but lower by two orders of magnitude than the isbrp Chicago air profile (865 ng/mg PCB) due to surprisingly high PCB 126 levels in Chicago air. In contrast, the calculated neurotoxic equivalency quotients of the CAM (0.33 mg/mg PCB) and the two Chicago air profiles (0.44 and 0.30 mg/mg PCB, respectively) were similar. This study demonstrates the challenges and methods of creating and characterizing synthetic, environmental mixtures of PCBs.  相似文献

A cohort of low income, city dwelling, pregnant African American Women (delivered from 1994–1999) was assembled to identify factors related to organochlorine exposure through consumption of Great Lakes resources. The cohort is known as the Great Lakes Cohort of Pregnant African American Women (GLCPAAW). Pregnant women from metropolitan Chicago, IL area clinics were administered a questionnaire on diet, demographics, and health history. Weight, height, and serum lipids were measured at delivery along with serum organochlorines such as PCBs and DDE. Congener specific concentrations of PCBs and p,p'-DDE found in the maternal serum are reported. Dominant PCB congeners found in the serum of the pregnant women at delivery included PCB 101, 118, 138, 153, and 180. The high prevalence and magnitude of PCB 101 (greater than the limit of detection in > 80% of the women in the cohort) are unique characteristics of this cohort.Great Lakes fish has been identified as a source of exposure to organochlorines in several studies. Spearman correlations and robust regression models were utilized to identify the impact of Great Lakes fish ingestion on cohort serum organochlorine concentrations. Several potential confounders of the relationship between serum organochlorines and Great Lakes fish consumption were identified. Covariates related to organochlorines in correlations as well as regression models included age, body surface area, fish ingestion, lipids, parity, race and smoking. Lower chlorinated PCB congeners do not follow the same trends as the higher chlorinated congeners and DDE. The higher chlorinated PCB congeners (PCB 138, 153, and 180) and DDE were correlated with age while the lower chlorinated congeners were not. PCB 153 and 180 regression models included age as a significant covariate. None of the higher chlorinated congeners correlated to race, while both lower chlorinated congeners were correlated to race. Race was also significant in both lower chlorinated congeners' regression models. PCB 101, a lower chlorinated congener seldom found in human serum, is readily found in the cohort. Airborne PCB exposure as well as diminished metabolism of PCB 101 in African Americans may explain the increased presence of PCB 101 and it's correlation with race.High end sport fish consumers (≥ 1meal per week) carried elevated levels of DDE and higher chlorinated PCB congeners (138, 153, and 180) compared to non-sport fish eaters. Unexpectedly, DDE was correlated more consistently with fish ingestion and age (a marker of bioaccumulation) in comparison to PCBs. Small correlations were found between serum PCBs and fish ingestion (Spearman correlation = 0.19 for total PCBs and fish meals per year). Additionally, Serum PCBs in low end Great Lakes sport fish consumers were not higher than non-consumers. These findings suggest the women of the cohort are being exposed to PCBs through other routes in addition to Great lakes sport fish. One major route of exposure may be Chicago air. The observed trends amongst individual PCB congeners has important ramifications because lower chlorinated congeners or their metabolites may be mediators of toxicity. Organochlorine exposure through Great Lakes fish ingestion was clearly identified in high end fish consumers while associations with race, metabolism, and possible airborne exposures pose new questions.  相似文献

Assessment of human exposure to environmental persistent organic pollutants such as organochlorine pesticides (OCPs), polychlorinated biphenyls (PCBs), polychlorinated dibenzo-p-dioxins/furans (PCDD/Fs) and polycyclic aromatic hydrocarbons (PAHs) based on the levels in human breast milk provides a reasonable tool not only to assess the contaminant burden in mothers but also to assess potential exposure of breast-fed neonates. There are limited data on levels of PCBs and OCPs in humans but no previous reports from Turkey on chemically determined levels PAHs in human milk. The aim of this study was to report the levels and accumulation profiles of OCPs, PCBs and PAHs in 47 breast milk samples obtained from a Mediterranean city, Mersin. High resolution analyses were performed by a gas chromatography coupled with mass spectrometer (GC–MS). Dichlorodiphenyldichloroethane (4.4′-DDE) was the dominant pollutant. Beta-hexachlorocyclohexane (ß-HCH), dichlorodiphenyltrichloroethane (4.4′-DDT), dieldrin, hexachlorobenzene, oxy-chlordane, cis-heptachlorepoxide were the other main OCPs detected. Mean levels of ΣPCB congeners and WHOPCB-TEQ were 9.94 and 0.001 ng/g lipid, respectively. PCB 153 showed the highest concentration (3.37 ng/g lipid), followed by PCB 138 and 180. For the dioxin-like PCBs, PCB 118 was the dominant (0.97 ng/g lipid). Naphthalene, phenanthrene, pyrene and fluoranthene were the major PAHs among the 16 PAHs detected. The estimated daily intakes of DDTs, PCBs, HCHs and HCB were not exceeded the tolerable daily intake (TDI) proposed by the Health Canada Guideline. These results indicate that the neonates of Mersin city are exposed to persistent organic pollutants analyzed in this study. However, neonates born in Mersin province are less exposed than the ones born in other regions, considering OCP and PCB levels in breast milk.  相似文献

分析了太湖底泥中6种PCB同系物(PCB28、52、101、138、153、180),其中含量最高的为PCB52,平均为0.983 ng/g,最低为PCB138,平均为0.104 ng/g。表层底泥中,检出率最高的为PCB101,达100%,最低为PCB180,只有58.3%。太湖底泥的多氯联苯含量与底泥中有机质和营养元素几乎不相关,表明大气沉降可能是多氯联苯的重要来源。根据沉积物有关风险评价标准,太湖底泥测定的PCB总量尚未达到毒性评价的低值。尽管底泥中PCBs含量不高,但在底栖生物中可以富集,并通过食物链逐级传递,因而其潜在的危害性仍不容忽视。  相似文献

Dioxin exposure has experimentally been associated with changes in DNA methylation, an epigenetic change that is associated with disease. The present study aims to investigate if serum levels of dioxin and other persistent environmental pollutants are related to global DNA methylation in a human sample. In the population-based Prospective Investigation of the Vasculature in Uppsala Seniors (PIVUS) study (all aged 70), global DNA methylation was measured by the Luminometric Methylation Assay in 524 subjects. Twenty-three different POPs, including 16 PCBs, five pesticides, one dioxin (OCDD) and one brominated flame retardant (BDE47) were analysed by HRGC/HRMS. Ten single nucleotide polymorphisms (SNPs) in the Aryl hydrocarbon (Ah)-receptor were analysed by mini-sequencing. High levels of toxic equivalency (TEQ) for PCBs and dioxin were associated with DNA hypermethylation (p = 0.030). This was mainly attributed to coplanar non-ortho PCBs. While no significant associations were found between DNA methylation and SNPs in the Ah-receptor, an interaction was found between the SNP rs2237297 and TEQ so that TEQ was associated with hypermethylation (p = 0.009) only in subjects with one G-allele (n = 103). Also high levels of the PCB126 congener, the OCDD, and the pesticide metabolite p,p′-DDE were related to DNA hypermethylation (p = 0.01, 0.03 and 0.003, respectively). In conclusion, in a sample of elderly subjects, high TEQ including PCBs and the dioxin OCDD and high serum levels of PCB126, OCDD, and p,p′-DDE were related to global DNA hypermethylation in a cross-sectional analysis.  相似文献

Polychlorinated biphenyls (PCBs) are a mixture of 209 possible congeners with different chlorine substitutions and they are potentially hazardous compounds in the environment for human beings. The high lipophilicity and the resistance to biodegradation of PCBs allow the bioaccumulation of these chemicals in the fatty tissues of organisms. The objective of this study was to determine the levels of PCBs in the adipose tissue of women living in Ankara, Turkey. For this purpose, levels of seven major persistent PCB congeners (IUPAC Numbers: 28, 52, 101, 118, 138, 153, 180) were measured in 29 primiparous mothers' adipose tissue samples by gas chromatography with electron capture detection (GC-ECD). PCB IUPAC numbers 138, 153 and 180 were the most abundant congeners, each accounting for > 20% of the total PCB content of adipose tissue. PCB 153 was the dominant congener in all the samples (141.7 +/- 132.3 ng/g lipid wt. basis). The results are compared with the similar studies from other countries. The concentrations of PCB 28, PCB 52 and PCB 101 were found to be higher than those found in industrialized countries. This study is important in terms of being the first study of determination of PCB levels in adipose tissue samples in Turkey.  相似文献

We have previously reported on the effects of in utero exposure to polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) and polychlorinated biphenyls (PCBs) on thyroid function and growth hormone concentrations at birth and in two and five year-old children. Herein, we present our most recent follow-up examination findings for the same cohort of children at eight-years of age. A total of 56 children (23 boys, 33 girls) were examined. Bone age (BA), hormone concentrations, and indicators of reproductive development including Tanner, breast, genital, and armpit stages were assessed. Estradiol concentrations were significantly lower in children exposed to higher levels than median of PCDD/Fs + PCBs TEQ compared to the children exposed to levels lesser than median (P = 0.003). Girls exposed to higher levels than median of indicator PCBs had a significantly greater proportion in genital stage 1 and shorter fundi and uteri lengths, as compared to those exposed to low levels (P = 0.025 and P < 0.05, respectively). There was a significant negative relationship between estradiol concentrations and PCDD/Fs + PCB exposure level (P = 0.005). After adjusting for BA, there was a significant association between fundus length and indicator PCB exposure level (P = 0.034). Exposure to both high levels of ΣPCDD/Fs + PCBs TEQ and high levels of total PCBs was associated with decreased fundus length (P = 0.016) and uterus length (P = 0.016). In utero exposure to high levels of PCDD/Fs and PCBs may result in lower estradiol concentrations in eight year-old children and impaired reproductive development in girls.  相似文献

During a regional monitoring project of organic-farmed, free-range and cage-free eggs, high levels of dioxin-like compounds were detected in organic-farmed eggs, using the dioxin responsive chemical-activated luciferase gene expression (DR-CALUX®) bioassay. Further evaluations performed with GC–HRMS (gas chromatography in combination with high resolution mass spectrometry) revealed elevated amounts of non-dioxin-like (non-dl) polychlorinated biphenyls (PCBs) dominated by most lipophilic congeners like PCB 138, 153 and 180 and of dioxin-like (dl) PCBs, with a congener pattern in the descending order of PCB 118, 156, 167, 105, 189, 157, 105, 126 and PCB 77. Contaminations with polychlorinated dibenzo-p-dioxins (PCDDs) appeared of minor priority, with only hepta- and octa-substituted dioxins above their limits of quantification (LOQs). The pattern of polychlorinated dibenzofurans (PCDFs) was dominated by low amounts of tetra- and penta-chlorinated congeners. To identify the source of contamination, several samples of organic-farmed eggs, soil, laying hens, feedstuff, corrugated asbestos-cement cover plates (ACPs), stable dust and debris collected in the gutter of the stable, were analyzed. Comparing PCB congener-pattern of individual samples, the source was traced back to the coating of ACPs, which covered roof and sidewalls of the stable. Because coating materials probably have been used for roofing and cladding in many countries worldwide, there is a high probability that the presented case report is not a local incident but rather describes a new source of PCB contamination, yet widely unknown or underestimated.  相似文献

Levels of polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs) and organochlorine pesticides (OCPs) were determined in the sediment and several species (European flounder, Platichthys flesus; common sole, Solea solea; Chinese mitten crab, Eriocheir sinensis; shore crab, Carcinus maenas; brown shrimp, Crangon crangon; blue mussel, Mytilus edulis and bristle worms, Polychaeta) from 7 locations in the Scheldt estuary (SE, the Netherlands–Belgium). Overall POP levels in the sediment were low. The average PCB and PBDE concentrations were respectively 31.5 and 115 ng/g dry weight (dw). Highest sediment loads were measured in the vicinity of Antwerp (368 ng PCBs/g dw), a location with intense harbor and industrial activities. Pollution concentrations in the tissues of biota were species-specific. Blue mussels contained the highest lipid concentrations (2.74 ± 0.55%) and reached the highest contamination levels (from 287 to 1688 ng PCBs/g ww, from 2.09 to 12.4 ng PBDEs/g ww). Lowest tissue loads were measured in brown shrimp (from 3.27 to 39.9 ng PCBs/g ww, from 0.05 to 0.47 ng PBDEs/g ww). The PCB congener profile in most of the species was similar with the pattern found in the sediment. PCB 153 was the most abundant congener (16.5–25.7% in biota, 10.4% in sediment). In the sediment, the total amount of PBDEs consisted for more than 99% of BDE 209. Congener BDE 47 had the highest concentrations in all sampled species (38.5–70.1%). Sediment POP loadings and tissue concentrations were poorly correlated, indicating that a simple linear or non-linear relationship is insufficient to describe this relationship, possible caused by the complexity of the bioaccumulation processes and the variability in exposure. Because of the high PCB levels, regular consumption of fish and seafood, especially mussels, from the Scheldt estuary should be avoided.  相似文献

We are studying participants selected from the Child Health and Development Studies (CHDS), a longitudinal birth cohort of over 20,000 California pregnancies between 1959 and 1967, for associations between maternal body burden of organochlorine contaminants and thyroid function. We designed a pilot study using 30 samples selected among samples with high and low PCB concentrations to evaluate the feasibility of measuring OH-PCBs in the larger study population. GC-ECD and GC-NCI/MS were used to determine PCBs and OH-PCBs as methyl derivatives, respectively. Maternal serum levels of Σ11PCBs and Σ8OH-PCB metabolites varied from 0.74 to 7.99 ng/mL wet wt. with a median of 3.05 ng/mL, and from 0.12 to 0.98 ng/mL wet wt. with a median of 0.39 ng/mL, respectively. Average concentrations of Σ8OH-PCB metabolites in the high PCB group were significantly higher than those in the low PCB group (p < 0.05). The levels of OH-PCB metabolites were dependent on PCB levels (r = 0.58, p < 0.05) but approximately an order of magnitude lower (p < 0.05). The average ratio of Σ8OH-PCBs to Σ11PCBs was 0.14 ± 0.08. The primary metabolite was 4-OH-CB187 followed by 4-OH-CB107. Both of these metabolites interfere with the thyroid system in in vitro, animal, and human studies. OH-PCBs were detectable in all archived sera analyzed, supporting the feasibility to measure OH-PCB metabolites in the entire cohort.  相似文献

BackgroundMajor food contaminants such as polychlorinated biphenyls (PCBs) are proposed to play a role in the etiology of cardiovascular disease (CVD), but to date the impact of PCBs on cardiovascular health need to be explored.Methods and resultsWe assessed the association between validated food frequency questionnaire-based estimates of dietary PCB exposure and risk of myocardial infarction, ascertained through register-linkage, among 36,759 men from the population-based Swedish Cohort of Men, free of cardiovascular disease, diabetes and cancer at baseline (1997). Relative risks were adjusted for known cardiovascular risk factors, long-chain omega-3 fatty acids (eicosapentaenoic and docosahexaenoic acids) and methyl mercury exposure. During 12 years of follow-up (433,243 person-years), we ascertained 3005 incident cases of myocardial infarction (654 fatal). Compared with the lowest quintile of dietary PCB exposure (median 113 ng/day), men in the highest quintile (median 436 ng/day) had multivariable-adjusted relative risks of 1.74 (95% confidence interval [CI], 1.30–2.33; p-trend < 0.001) for total and 1.97 (95% CI 1.42–2.75; p-trend < 0.001) for non-fatal myocardial infarction. In mutually adjusted models, dietary PCB exposure was associated with an increased risk of myocardial infarction, while the intake of long-chain omega-3 fish fatty acids was associated with a decreased risk. We also observed an effect modification by adiposity on the association between of dietary PCB exposure and myocardial infarction, with higher risk among lean men (p value for interaction = 0.03).ConclusionsExposure to PCBs via diet was associated with increased risk of myocardial infarction in men.  相似文献

This study investigated the status of contamination of organohalogen compounds (OCs) such as polychlorinated biphenyls (PCBs) and brominated flame retardant (BFRs), including polybrominated diphenyl ethers (PBDEs) and hexabromocyclododecanes (HBCDs) in human milk samples from several locations in India. The levels of OCs were significantly higher in the milk of mothers living in and near municipal dumping site than other locations indicating that the open dumping sites for municipal wastes act as potential sources of these contaminants in India. The PCB concentrations observed in this study tended to decrease compared to those in the matched locations reported previously, probably due to the restriction of technical PCB usage in India. PBDE levels in human milk were two to three folds lower than those of PCBs in all the sampling locations investigated. Congener profiles of PCBs and PBDEs were different between samples from the dumping site mothers and general populations in other areas suggesting the presence of region-specific sources and pathways. HBCDs were detected in human milk from only two sites, with much lower concentrations and detection frequencies compared to PCBs and PBDEs. When hazard quotients (HQs) of PCBs and PBDEs were estimated for infant health risk, the HQs in some milk samples from the dumping site exceeded the threshold value (HQ > 1) of PCBs, indicating the potential risk for infants in the specific site.  相似文献

Detailed polychlorinated biphenyl (PCB) signatures and chiral Enantiomer Fractions (EFs) of CB-95, CB-136 and CB-149 were measured for 30 workers at a transformer dismantling plant. This was undertaken to identify sources of exposure and investigate changes to the PCB signature and EFs over different exposure periods. Approximately 1.5 g of serum was extracted and PCB signatures were created through analysis by comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry (GC × GC–TOFMS) and EFs calculated following analysis by gas chromatography with high resolution mass spectrometry (GC–HRMS). A total of 84 PCBs were identified in the serum samples with concentrations of the 7 indicator PCBs ranging from 11–350 ng g 1 of serum (1.2–39 μg g 1 lipid). The PCB signatures were interpreted using principal component analysis (PCA) which was able to distinguish workers with background or recent minimal exposure from those with prolonged occupational exposure. Occupationally exposed individuals had a similar PCB profile to Aroclor A1260. However, individuals with prolonged exposure had depleted proportions of several PCB congeners that are susceptible to metabolism (CB-95, CB-101 and CB-151) and elevated proportions of PCBs that are resistant to metabolism (CB-74, CB-153, CB-138 and CB-180). The results also identified a third group of workers with elevated proportions of CB-28, CB-60, CB-66, CB-74, CB-105 and CB-118 who appeared to have been exposed to an additional source of PCBs. The results show near complete removal of the CB-95 E2 enantiomer in some samples, indicating that bioselective metabolism or preferential excretion of one enantiomer occurs in humans. By considering PCB concentrations along with detailed congener specific signatures it was possible to identify different exposure sources, and gain an insight into both the magnitude and duration of exposure.  相似文献

Isomer specific concentrations of individual polychlorinated biphenyls (PCBs) including toxic non-ortho (IUPAC 77, 126, 169) and mono-ortho (105, 118, 156) coplanar congeners were determined in the blubber of nine bottlenose dolphins (Tursiops truncatus) stranded along the Eastern Italian coast. The total PCB concentrations ranged from 3534 to 24375 ng/g wet wt. The PCB profile was dominated by congeners 138 and 153 collectively accounting for 55% of the total PCB concentrations. Among the most toxic congeners the order of abundance was 126>169>77. The mean total 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) equivalent of six coplanar PCBs in the blubber of bottlenose dolphins was 45596 pg/g. Non-ortho congeners contributed greater to the 2,3,7,8-TCDD toxic equivalents than mono-ortho members. Particularly, PCB 126 was the major contributor to the estimated toxic potency of PCBs in dolphins.  相似文献

PCB levels in fish (collected from local rivers), atmosphere and human milk samples have been studied to determine the exposure levels of PCBs for local residents and e-waste workers in Guiyu, a major electronic waste scrapping center in China. The source appointment and correlation analyses showed that homologue composition of PCBs in 7 species of fish were consistent and similar to commercial PCBs Aroclor 1248. PCB levels in air surrounding the open burning site were significantly higher than those in residential area. Inhalation exposure contributed 27% and 93% to the total body loadings (the sum of dietary and inhalation exposure) of the local residents, and e-waste workers engaged in open burning respectively. Total PCB concentrations in human milk ranged from N.D. to 57.6 ng/g lipid, with an average of 9.50 ng/g lipid. The present results indicated that commercial PCBs derived from e-waste recycling are major sources of PCBs accumulating in different environmental media, leading to the accumulation of high chlorinated biphenyls in human beings.  相似文献
