Free amino acid patterns and the classification of the Demospongiae   总被引:1,自引:0,他引:1  
An investigation of the free amino acid patterns of 67 species representing 50 genera of West Indian Demospongiae has been undertaken to examine the value of this biochemical approach in adding another parameter to phylogenetic inquiries. Two-dimensional thin-layer electrophoresis and chromatography were used to separate mixtures of naturally occurring ninhydrin-positive compounds. Individual components were identified as far as possible, and the fingerprints or patterns of the species studied were used as a basis for evaluating relationships. The subclass Ceractinomorpha is a relatively homogeneous group, with free amino acid patterns characterized by a consistent representation of protein amino acids and by the absence of compounds other than taurine and hypotaurine in the neutral region of the chromatogram. Two variants of this general pattern can be recognized, one found in the orders Dictyoceratida and Poecilosclerida, the other occurring in the order Haplosclerida and the family Halichondriidae of the order Halichondrida. Unexpected results within this subclass are the wide separation of the dictyoceratid families Spongiidae and Verongiidae, the anomolous patterns of the genera Agelas and Iotrochota and of the family Callyspongiidae, and the suggestive affinities of the family Hymeniacidonidae with the order Hadromerida. The subclass Tetractinomorpha appears to represent a polyphyletic assemblage in which 5 recognizable patterns can be distinguished. These represent the orders Homosclerophorida, Choristida, Spirophorida, Hadromerida and Axinellida. Of interest are the suggestions that the order Choristida is in itself a heterogeneous group, and that certain genera now placed in the order Axinellida may in fact be more closely related to the Poecilosclerida.  相似文献

The phylogenetic relationships among local populations of undescribed vestimentiferan species that belong to the genera Lamellibrachia and Escarpia and had been collected at six sites around Japan were analyzed on the basis of the partial (1023 bp) nucleotide sequences of the mitochondrial gene for cytochrome oxidase I (COI), using a pogonophoran and a polychaete as outgroups. The identical amino acid sequence was deduced from the nucleotide sequence obtained from each of the vestimentiferans analyzed. The strong similarity among deduced amino acid sequences of COI suggested a close relationship between vestimentiferans and pogonophorans. On the basis of the phylogenetic relationships, the analyzed vestimentiferan populations were classified tentatively as five species. The genetic differentiation of vestimentiferans was suggested to occur bathymetrically as well as being a consequence of horizontal segregation. Two of these tentatively identified species inhabit both a hydrothermal area and a cold seep area, as is the case for some species of bivalves that belong to the genera Bathymodiolus and Calyptogena. Glycine was the most abundant free amino acid (FAA) in abdominal muscle of grass shrimp Palaemonetes pugio, accounting for over 50% of the total pool. Arginine, alanine, proline, taurine and serine were also present at relatively high concentrations. Following transfer from 17%. to 2 and 32%. S, new steady-state levels of total FAA were observed at 72 h. Total FAA and the sum of glycine, alanine and proline exhibited a positive correlation with salinity. Exposure to Aroclor 1254 did not have appreciable effects on total FAA levels, indicating that disruption of intracellular osmoregulation was not a major consequence of PCB toxicity. However, changes in individual amino acid concentrations in exposed shrimp were reflective of an altered metabolic state. Glycine, which did not show changes immediately following exposure, underwent large decreases after transfer to PCB-free water and indicated a delayed effect of PCB exposure. A correlation between salinity and the sum of glycine, alanine and proline did not exist in exposed shrimp.  相似文献

Using the starvation technique, changes in protein and free amino acids were examined in Penaeus esculentus Haswell collected from Moreton Bay, Australia, by trawling in 1985. Prawns of 17.7±0.26 g wet weight were held at 25°C until 2 d after moulting. Groups of seven or eight were then starved fro 5, 10, or 15 d, with appropriate control groups. At the end of each period, ecreted amino acids were collected for 24 h and whole-muscle amino acids and free amino acids (FAA) g-1 in each prawn were analysed. Concentrations of whole-muscle amino acids showed only minor changes with starvation, but concentrations of many of the FAA changed significantly. Total FAA averaged 1 182±45 mol g-1 dry weight. Individual FAA, in order of abundance, were glycine, arginine, proline, taurine, threonine, hydroxyproline, alanine, glutamic acid, valine, aspartic acid and lysine; the remaining FAA each contributed <0.2% of the total. Only taurine and alanine did not show significant changes with starvation. Concentrations of glycine, arginine, hydroxyproline, glutamic and aspartic acid increased, while those of proline, threonine, valine and lysine decreased with starvation, that of proline approaching zero after 15 d starvation. Excreted amino acid-nitrogen represented <2% of excreted ammonianitrogen ornithine being the most abundant (35%), followed by leucine (22%) and lysine (17%). The relative abundance of excreted amino acids did not correspond with those of the FAA. It is suggested that, as starvation progresses, the muscle protein is progressively hydrolysed, but with the remaining muscle maintaining its amino acid composition. The liberated amino acids enter the FAA pool and become available for energy production. Proline may have an important role as an energy source, but the ability to synthesise proline may be limited, and thus the artificial food of penaeid prawns may be improved by its addition.  相似文献

The present paper studied the influence of different food regimes on the free amino acid (FAA) pool, the rate of protein turnover, the flux of amino acids, and their relation to growth of larval turbot (Scophthalmus maximus L.) from first feeding until metamorphosis. The amino acid profile of protein was stable during the larval period although some small, but significant, differences were found. Turbot larvae had proteins which were rich in leucine and aspartate, and poor in glutamate, suggesting a high leucine requirement. The profile of the FAA pool was highly variable and quite different from the amino acid profile in protein. The proportion of essential FAA decreased with development. High contents of free tyrosine and phenylalanine were found on Day 3, while free taurine was present at high levels throughout the experimental period. Larval growth rates were positively correlated with taurine levels, suggesting a dietary dependency for taurine and/or sulphur amino acids. Reduced growth rates in Artemia-fed larvae were associated with lower levels of free methionine, indicating that this diet is deficient in methionine for turbot larvae. Leucine might also be limiting turbot growth as the different diet organisms had lower levels of this amino acid in the free pool than was found in the larval protein. A previously presented model was used to describe the flux of amino acids in growing turbot larvae. The FAA pool was found to be small and variable. It was estimated that the daily dietary amino acid intake might be up to ten times the larval FAA pool. In addition, protein synthesis and protein degradation might daily remove and return, respectively, the equivalent of up to 20 and 10 times the size of the FAA pool. In an early phase (Day 11) high growth rates were associated with a relatively low protein turnover, while at a later stage (Day 17), a much higher turnover was observed. Effects of the predatory naticid snail Polinices duplicatus (Say) on species composition, diversity and density, of the infauna of intertidal sand-flats at Barnstable Harbor, Massachusetts, USA, were studied using field experiments. Responses of molluscs (prey of P. duplicatus) and nonmolluscs (nonprey) were considered separately to distinguish between the effects of feeding and sediment disturbance during foraging. The fauna of 0.25 m2 predator-exclusion cages, coarse-mesh cages, sieved areas, and controls was followed for 1 yr. Species associations within cages from which predators were excluded were denser, more diverse, and richer in molluscs than those in other treatments. Larger areas (3×3 m) of natural bottom were fenced and maintained as snail exclosures and enclosures during two feeding seasons. For both molluscs and nonmolluscs, diversity (H'), number of species, evenness (SD), and density all decreased with increasing snail density. Intense predation pressure on molluscs and selective feeding on thin-shelled bivalves, which were rare, removed individuals and species of molluscs from the community. Comparison of samples taken inside and outside trails made by snails showed that disturbance of the surface sediment layers by snails decreased the abundance of spionid polychaetes and total nonmolluscs. The sipunculan Phascolopsis gouldi and the bivalve Gemma gemma dominated the community in all experimental treatments and were little affected by the activity of Polinices duplicatus. Predation and disturbance by snails lowered community diversity by removing individuals of the less abundant species, and generally maintained population densities below the level where strong competition would occur.  相似文献

A method for the isolation of intact coupled mitochondria from the hepatopancreas of Mytilus edulis has been developed. These mitochondria prefer lipid substrates and accept a range of fatty acid chain lengths. Acetyl carnitine is also oxidized at high rates. Evidence for the presence of the tricarboxylate and the dicarboxylate transporters is presented. The absence of the pyruvate transporter or inhibition of pyruvate dehydrogenase is indicated by the absence of pyruvate oxidation. Glutamate is oxidized at high rates with a pH optimum of 7.5 based on the RCR. Cysteine oxidation may indicate the presence of the enzymes involved in the synthesis of taurine. Ornithine transport is also indicated, implying the existence of the urea cycle. Alanine, glycine, proline, asparagine, arginine, aspartate and citrulline were not oxidized at detectable levels. The osmotic optimum for the oxidation of glutamate extends from 600 to 1000 mOsm l-1. The state 3 rate is more profoundly affected by the osmolarity than the state 4 rate. The role of osmolarity in the control of oxidation of amino acids during volume regulation of osmotically stressed M. edulis is suggested. Mitochondrial volume changes correlate with the optimal range of osmolarity for glutamate oxidation based on the RCR.  相似文献

Benthic organisms revealed enhanced uptake of mercury from polluted surface sediments (in the vicinity of a chlor‐alkali plant) which were enriched in mercury by up to 148 times relative to sediments from an unpolluted area. There is a strong correlation between the concentrations of mercury in the benthic organisms and sediments. No correlation was found between cadmium, lead, copper and zinc levels in the benthic fauna populations and the concentrations of these metals in the sediments. Ranges of concentrations (μg/g wet wt.) in the whole soft tissues varied: Hg, <0.005–3.31; Cd <0.018–1.99; Pb, <0.025–1.56; Cu, 0.242–28.8; and Zn, 9.75–310.2. The three gastropod molluscs show higher metal concentrations than do the bivalves.  相似文献

Spectroscopic determinations of Fe, Zn, Mn, Cu, Co and Ni were carried out in 23 organisms including seaweeds, zooplankters, molluscs and fishes from Hooghly estuary at the confluence of Bay of Bengal. Seaweeds and gastropods were more efficient in accumulating levels while zooplankters bivalves and fishes showed intermediate levels. Analyses were performed on the sediment, water and on the individual dissected organs of a mussel, oyster and teleost fish. Bioaccumulation of Fe, Zn, Mn and Cu in the soft body parts (gills, mantle, liver etc.) of the three species showed a high degree of organ specificity in some cases. Major sources of the micropollutants include natural weathering, catchment runoff, urban and industrial discharges that may pose an ecological risk to the local estuarine ecosystem.  相似文献

Several aspects of the biology of Bathypolypus sponsalis were studied from 297 individuals (115 males, 180 females and 2 indeterminates) caught in a depth range of 200–800 m depth in the western Mediterranean Sea. The paper presents data on sizes (length-weight relationships, size-frequency distributions) and reproduction (sex ratio, maturation, condition), and also analyses of the diet of B. sponsalis from samples taken throughout the year. Length-weight relationships showed that females are heavier than males at the same mantle length. Although mature individuals were found all year round, the maximum number occurred in spring and summer. Sexual maturation data revealed that males mature at smaller sizes than females. The gonadosomatic index increased with maturity in both sexes; the increase was gradual in males, but abrupt in females. The digestive gland index was used as a condition index and showed a differential behaviour with maturity; it increased gradually in females, but decreased in males. Like other octopus species, B. sponsalis appears to be an opportunistic predator, feeding on a great variety of preys. Stomach content analysis yielded a total of 19 different prey items belonging to four major groups (Crustacea, Mollusca, Ophiuroidea and Osteichthya). The first three groups were the more frequent preys, since crustaceans, molluscs and ophiuroids appeared in 76%, 49% and 30% of the stomachs, respectively. Decapoda Reptantia (among crustaceans) and cephalopods and bivalves (among molluscs) constituted the more abundant prey items. While the Decapoda Reptantia group was significantly more abundant in stomach contents of females, gastropods were taken more frequently by males. Curves relating instantaneous growth rate to aerial exposure were determined for six species of bivalves in laboratory and shore experiments. A dimensionless index equatable with the relative intertidal growth performance of a species was calculated by integration of the growth curve after converting both growth rate and aerial exposure to decimal fractions. Intertidal growth performance of the bivalves tested corresponded well with their natural levels of occurrence on the shore, and improved in the following order: Modiolus modiolus < Argopecten irradians < Ostrea edulis < Mytilus edulis < Crassostrea virginica. Geukensia demissa, for which an index value could not be determined, grew faster intertidally than subtidally. The relative contributions made to intertidal growth performance by energy-conserving and energy-supplementing capacity adaptations were assessed by resolving the growth curves into energy-input and energy-loss components. The rate of energy loss due to intertidal exposure was lower in the high-shore species, and also less affected by harsher subaerial conditions, than in the low-shore species. Moreover, M. edulis and C. virginica showed abilities to supplement energy input such that growth per unit immersion time was better at certain intertidal levels than subtidally. Energy conservation and supplementation in these forms made roughly equal contributions to their improved intertidal growth relative to species occurring lower on the shore.  相似文献

Growth of marine planktonic diatoms on inorganic and organic nitrogen   总被引:2,自引:0,他引:2  
A study was conducted to determine if coastal diatoms from eutrophic waters (Werribee, Port Phillip Bay, Australia) are able to grow better than diatoms from oligotrophic waters (Bass Strait, Australia) on organic nitrogen compounds as their principal nitrogen sources. Eight clones of marine planktonic diatoms, belonging to 5 species (Skeletonema costatum, Asterionella japonica, Nitzschia closterium, Coscinodiscus sp., and Fragilaria sp.), were incubated with inorganic (either nitrate or ammonia) or organic (either urea, uric acid, alanine, aspartic acid, glutamic acid, glycine, serine, threonine, or valine) nitrogen sources and growth response was measured under high and low light intensities. All clones grew well on the organic as well as thorganic nitrogen compounds under both light regimes. Intraspecific differences were not great, as no appreciable difference was noted between clones from oligotrophic and eutrophic waters. The two negatively-charged amino acids, aspartic and glutamic acids, were somewhat less effective in supporting growth of some clones than were the other amino acids. Virtually all of the dissolved organic nitrogen (DON) compounds tested were utilizable for algal growth. Further, all clones appeared able to utilize at least some natural DON (uncharacterized) for cell division; in 1980, DON represented 97% of total nitrogen in Bass Strait and 83% of total nitrogen in Werribee waters. We report the results of a detailed investigation on the trophoecology of two dominant meiofaunal species at the Håkon Mosby Mud Volcano (HMMV), a deep-sea cold methane-venting seep. Analyses of fatty acids (FAs) and their stable carbon isotopes were used to determine the importance of chemosynthetic nutritional pathways for the dominant copepod species (morphologically very similar to Tisbe wilsoni) inhabiting the volcano’s centre and the abundant nematode Halomonhystera disjuncta from the surrounding microbial mats. The strong dominance of bacterial biomarkers (16:1ω7c, 18:1ω7c and 16:1ω8c) coupled with their individual light carbon isotopes signatures (δ13C ranging from ?52 to ?81‰) and the lack of symbiotic relationships with prokaryotes (as revealed by molecular analyses and fluorescent in situ hybridisation) indicated that chemosynthetically derived carbon constitutes the main diet of both species. However, the copepod showed a stronger reliance on the utilisation of methanotrophic bacteria and contained polyunsaturated FAs of bacterial origin (20:5ω3 and 22:6ω3 with isotope signatures δ13C < ?80‰). Instead, the FA profiles of H. disjuncta suggested that sulphide-oxidising bacteria constituted the main diet of this nematode. Therefore, HMMV can be regarded as a persistent deep-sea cold seep, allowing a chemosynthesis-based trophic specialisation by the dominant meiofaunal species inhabiting its sediments. The present investigation, through the determination of the fatty acid profiles, provides the first evidence for trophic specialisation of meiofauna associated with sub-habitats within a cold seep.  相似文献

Crassostrea virginica (Gmelin) collected in 1989 from several sites within the Chesapeake Bay have narrower salinity tolerances than conspecific oysters collected in 1989 from several Atlantic coast sites (Georgia to Cape Cod). The basis of this physiological difference appears to be the biochemical mechanisms that control cellular osmolality following salinity stress. When adapted to the same salinity, the amino acid pools of both gill and adductor muscles of Atlantic oysters are larger than those of Bay oysters and different in composition. The Atlantic oyster tissues rely primarily on taurine for salinity tolerance, while the Bay oyster tissues have relatively less taurine, depending instead upon alanine, glycine and proline to adapt to high salinity. In addition, Atlantic oyster gill and adductor have 10 to 25 times the glycine betaine concentrations of these tissues from Bay oysters, depending upor the salinity of acclimation. The betaine concentration varies with salinity in Atlantic oysters, but does not change in Bay oysters. The results suggest that these biochemical differences are the basis of the narrower salinity tolerance in Bay oysters. The biochemical differences may reflect genetic differences between Bay and Atlantic oysters.  相似文献

Patterns of nine intracellular free amino acids (FAA), which are utilized as organic osmolytes for salinity-induced cell volume regulation in marine osmoconformers, were compared in five Macoma balthica populations and seven Mytilus spp. populations along their European distribution. Three types of FAA patterns were classified within both taxa: a northern Baltic type, a southern Baltic type and an Atlantic/Mediterranean type which mainly differ regarding the share of alanine and taurine. Differences are discussed in relation to habitat salinity and population genetics. Along a salinity gradient, the total size of the intracellular FAA pool did not differ between sympatric M. balthica and Mytilus spp., and was significantly correlated with habitat osmolality in a range from 70 to 600 mmol kg−1 H2O (oligohaline to mesohaline) in both bivalves. In M. balthica, this correlation was mainly based on significant correlations of alanine (15–100 mmol kg−1 DW), glycine (30–100 mmol kg−1 DW) and taurine (0–70 mmol kg−1 DW) with habitat osmolality. In Mytilus spp., only glycine (25–100 mmol kg−1 DW) and taurine (4–180 mmol kg−1 DW) were significantly correlated with habitat osmolality. The concentration of alanine was three times lower in Mytilus spp. than in M. balthica and did not correlate with habitat osmolality. Within a habitat osmolality range from 600 to 1,100 mmol kg−1 H2O (mesohaline to marine) the concentration of FAA remained constant in both taxa. It is suggested that under marine conditions additional organic osmolytes must become more important for cell volume regulation in Macoma and Mytilus.  相似文献

Partitioning of the food resources by two coexisting pufferfishes (Sphoeroides spengleri and S. testudineus) from Biscayne Bay, Florida, USA, was investigated. Gut contents from 453 bandtail and 339 checkered puffers were analyzed. The diets of both species consisted of a variety of benthic prey, but only crustaceans and molluscs were important prey groups. While differences were found in the proportions of general prey categories eaten by these fishes, both species consumed substantial quantities of brachyuran crabs, bivalves, and gastropods. Specific identification of the prey items within these three food categories revealed additional differences in prey between the two puffer species. This partitioning of the food resources by bandtail and checkered puffers was found between both species overall, between overlapping size ranges, and between both species' most abundant size group. Despite the important roles played by parasites in local population dynamics and community structure of marine ecosystems, there is a lack of information on the geographical variation in infection levels displayed by particular host–parasite species combinations. This study examines geographical variation in infection levels by the metacercarial stages of trematode parasites in crustacean and bivalve second intermediate hosts. Analyses were based on a dataset compiled from the literature, consisting of 164 local samples representing 49 host–parasite species pairs for crustaceans, and 338 entries representing 36 host–parasite species pairs for bivalves. The analyses indicate that for all measures of infection levels [prevalence (percentage of individuals infected), intensity (mean no. of metacercariae per infected individual), abundance (mean no. of metacercariae across all individuals in a sample)], there was statistically significant repeatability of infection values within host–parasite species pairs. However, it is only for values of intensity and abundance of infection in crustacean hosts that the repeatability was strong; this suggests that infection levels are specific properties of crustacean–trematode species pairs, showing significant consistency across localities despite spatial variation in abiotic and biotic conditions. Although the magnitude of variation in infection levels within parasite species pairs (measured as coefficients of variation) was independent of scale in crustacean hosts, infection levels in bivalves increased in variability at large (>100 km) spatial scales. These results suggest that there is a considerable geographical consistency in parasite load, especially in crustacean hosts, which should lead to consistent ecological and ecosystem effects of marine trematodes. Ammonium uptake and assimilation by zooxanthellae (Symbiodinium sp.) cultured with an excess of nitrate was enhanced in light. Uptake was decreased by the same amount when zooxanthellae were incubated in darkness either after 6 h pretreatment in light, or at the end of the dark period of a 12 h light: 12 h dark cycle. This suggested that short-term incubations of zooxanthellae were valid tests for light enhancement of dissolved inorganic nitrogen (DIN) uptake. Assimilation of ammonium into glutamine (Gln) and glutamate (Glu) was also decreased in darkness. During a 12 h light: 12 h dark cycle, free pools of both Gln and Glu fell quickly at the start of the light period, followed by steady increases until the beginning of the next dark period. Of the four other major components of free amino acid pools tested, only the nonprotein amino acid taurine showed diel fluctuations. Gln and Glu pools in zooxanthellae freshly isolated from reef-forming corals also showed differences between day and night, suggesting changes in patterns of DIN assimilation over the diurnal cycle.  相似文献

Comparisons between invertebrate communities hosted by similar foundation species under different environmental conditions permit identification of patterns of species distributions that might be characteristic of the different ecosystems. Similarities and differences in community structure between two major types of chemosynthetic ecosystems were assessed by analyzing samples of invertebrates associated with Bathymodiolus heckerae Gustafson et al. mussel beds at the Florida Escarpment seep (Gulf of Mexico, 26°01.8N; 84°54.9W; October 2000) and B. puteoserpentis von Cosel et al. mussel beds at the Snake Pit vent (Mid-Atlantic Ridge, 23°22.1N; 44°56.9W; July 2001). Macrofaunal species richness was nearly twice as high in the seep mussel bed compared to the vent mussel bed, and only a single morphospecies, the ophiuroid Ophioctenella acies Tyler et al., was shared between the sites. Similarities between the two faunas at higher taxonomic levels (genus and family) were evident for only a small percentage of the total number of taxa, suggesting that evolutionary histories of many of these seep and vent macrofaunal taxa are not shared. The taxonomic distinctiveness of the seep and vent mussel-bed macrofaunal communities supports the hypothesis that environmental and oceanographic barriers prevent most taxa from occupying both types of habitats. Macrofaunal community heterogeneity among samples was similar in seep and vent mussel beds, indicating that spatial scales of processes regulating community variability may be similar in the two types of ecosystems. Suspension feeders were not represented in the macrofauna of seep or vent mussel beds. Primary consumers (deposit feeders and grazers) contributed more to the total abundance of macrofauna of seep mussel beds than vent mussel beds; secondary consumers (polychaetes and shrimp) were more abundant in the vent mussel beds.Electronic Supplementary Material Supplementary material is available in the online version of this article at http://dx.doi.org/10.1007/s00227-004-1304-z.Communicated by J.P.
