To study the kinetics of mercury uptake in oysters, adult Crassostrea virginica (Gmelin) were held in seawater containing 10 g mercury/l (ppb) or 100 g mercury/l (ppb), added in the form of mercuric acetate, for 60 days. Mercury concentration in tissues was determined by analysis of individually homogenized oyster meats, using wet digestion and flameless absorption spectrophotometry. After 45 days, average mercury tissue concentration was 140,000 g mercury/kg tissue (ppb) and 28,000 g mercury/kg tissue (ppb) in the 100 ppb and 10 ppb experimental groups, respectively. After this time, concentrations dropped sharply, probably due to spawning. Clearance of mercury from tissue was studied by exposing treated adults to estuarine water (with no additions) for 30 days (100 ppb group) and 160 days (10 ppb group). Tissue concentrations in the 100 ppb mercury environment group declined from 115,000 to 65,000 ppb, and those of the 10 ppb group declined from 18,000 to 15,000 ppb, in 18 days; there-after, no further decline occurred in either group. Oysters accumulated mercury 1,400 times and 2,800 times above the environmental concentrations of 100 and 10 ppb mercury, respectively. Total self-purification was not achieved over a 6 month cleansing period.  相似文献

Oysters (Crassostrea virginica) were exposed for 3 days to mercury-203 labeled HgCl2 or CH3HgCl added directly to artificial seawater or added preconcentrated on the marine diatom Phaeodactylum tricornutum. The concentration of mercury in 5 tissues was measured for 45 days after mercury was removed from the ambient water. At the beginning of the depuration period, the highest concentrations of mercury in tissues were attained in: gill>digestive system>mantle>gonad>muscle in oysters exposed to water containing mercury; and in digestive system>gill>mantle> gonad>muscle in oysters fed labeled algae. This same distribution pattern is seen for both chemical forms of mercury. Although the initial pattern of accumulation was identical for both mercury compounds within each exposure group, the fate of the accumulated mercury was very different after 45-days depuration. In oysters accumulating mercury directly from seawater, inorganic mercury residues rapidly declined in gill and digestive tissue, but were slowly reduced in mantle, gonadal, and muscle tissue. This pattern was duplicated by oysters exposed to methyl mercuric chloride in seawater except that gonadal and muscle residues greatly increased during depuration. In oysters ingesting labeled P. tricornutum cells, mercuric chloride and methyl mercuric chloride residues rapidly declined in gill and digestive tissue, remained constant in the mantle, but sharply increased in gonadal and muscle tissue during depuration.  相似文献

To test the hypothesis that oysters, Crassostrea virginica, from the northernmost part of the species range in the Gulf of St. Lawrence (48°N) open their valves at lower temperatures than those reported for more southern oysters, Hall element sensors were used to monitor their gaping behaviour. These observations were made in a flow-through system and the temperature, salinity and relative fluorescence of unfiltered seawater were monitored. Photoperiod was controlled (15?h dark:9?h light) and light levels were measured but not closely controlled. Gaping behaviour was followed from February to June 2010 (113?days) and from April to May 2011 (34?days) and was classified into three successive phases: quiescent, awakening and active. Although valves were either closed or slightly open during the quiescent phase (maximum gape angle?=?0.49°, SE?=?0.04), they abruptly opened to maximum angles of about 5.88° (SE?=?0.29) during the awakening phase. Moreover, there was noticeable synchrony amongst individuals, since approximately one-half of the monitored population awoke within a 6.6-h period in both study years. Correlative analyses identified temperature as a factor influencing valve movement, and oysters awakened when temperatures were 0.2–4.0?°C (mean?=?2.2, SE?=?0.2). Oysters exerted their maximal gape angle as soon as temperatures reached 2.8–6.6?°C (mean?=?4.8, SE?=?0.2). During the active phase, valves remained open 68.6?% (2010) and 79.7?% (2011) of the time. An unexpected result was the observation of a diurnal rhythm in valve openness whereby the openness was greatest near the end of the afternoon and least in the early morning.  相似文献

Cephalopod mollusks exhibit highly plastic life cycle traits influenced primarily by the interactive effects of food availability, light cycle and temperature, with the latter perhaps the most influential. Hatchlings of the tropical reef squid Sepioteuthis lessoniana were hatched from field-collected eggs in the laboratory and cultured at different temperatures to evaluate the effect of temperature on growth rates. All groups showed rapid, sustained growth rates from hatching to a size of 10–25 g. Beyond this size range, growth was slower and not clearly exponential in form. Growth rate was closely linked to temperature. Squids grown at approximately 27 °C attained a size of 10 g in as little as 45 days at sustained growth rates of 12.2% body weight day−1 (%bw day−1), while squids cultured at 20 °C required almost 100 days to attain the same size at rates of 5.7%bw day−1. At an age of 55 days and approximately 1 g body weight, juvenile squids cultured at 20 °C were able to accelerate growth rates from 5.7%bw day−1 to over 12%bw day−1 when temperature was raised to 27 °C. They maintained this growth rate to a size of about 10 g and an age of at least 75 days post-hatching, indicating that body size and not age is the limiting factor for this rapid post-hatching growth. By comparison, conspecifics cultured near 27 °C from hatching had shifted out of the rapid post-hatching growth phase by day 50 at sizes between 10 and 50 g. The hatchlings from temperate to subtropical Japan had consistently higher growth rates at comparable temperatures than hatchlings from tropical Okinawa. When plotted as growth rate versus temperature, the Japanese group had a clearly higher slope to the relationship than the tropical populations, equivalent to a 2%bw day−1 difference in growth rate at 25 °C. The results of toxicity experiments have shown that a level of 17 ppb ionic mercury is acutely toxic (96-h LC50) to postlarval white shrimp (Penaeus setiferus). In addition, the size of the shrimp, within the range tested (7 to 35 mm) did not significantly alter the toxicity values of mercury to this species. Fifty-seven days pre-exposure to low levels of mercury (0.5 and 1.0 ppb) had no effect on the acute toxicity (96-h LC50) of mercury to the shrimp. The effect of chronic exposure to low levels of mercury on the respiratory rate, growth, and molting rate of postlarval P. setiferus was studied, and the results showed that 60 days exposure to 0.5 and 1.0 ppb mercury did not significantly affect any of these parameters for the postlarvae.  相似文献

Marine polychaetes Nereis virens (Sars) were exposed to 9 ppb Hg as mercuric chloride in the aquarium water. Concentrations of mercury in the water and in the worms were monitored using radiolabelled mercury. After 11 d, the average mercury concentration in the nereids was 8.41 ppm Hg and the bioconcentration factor was calculated to be 930. Accumulations of mercury in the tissues were made visible for light and electron microscopy by autometallographic silver enhancement. Mercury was demonstrated in the intestine, nephridia, epidermis and cuticula. In the intestine, mercury was found to be located predominantly in the apical part of the epithelial cells. In the nephridia, mercury deposits were apparent in the peritoneal cells of the nephridial tubules. Silver-enhanced mercury was also observed at the epicuticula, in the collagen fibres of the endocuticula and in the epidermal basal cells. Intracellularly, the observed mercury accumulations were localised predominantly in lysosomes. Extracellularly, mercury was observed in the basal laminae of the intestine and the epidermis, and also in the intestinal peritrophic membrane. How N. virens copes with mercury toxicity is discussed.  相似文献

Oxygen consumption of individual larvae of the Antarctic sea-star Odontaster validus was measured during the 50-day period following fertilisation. Values ranged from 0.76 pmol O2 h-1 for one specimen at the coeloblastula stage to 77.6 pmol O2 h-1 for one bipinnaria larva. At 0°C the mean oxygen consumption rate of an individual larva increased from 10.9 pmol O2 h-1 (standard error of the mean, SEM, 0.13) for a gastrula larva, 13 days post-fertilisation, to 25.4 pmol O2 h-1 (SEM 3.5) at the bipinnaria stage (50 days post-fertilisation). Gastrulae reared at -0.5°C did not have significantly different oxygen consumption rates between days 13 and 45 post-fertilisation (mean=11.4 pmol O2 h-1). Individual metabolic rates were highly variable, covering more than a 40-fold range. At 2°C gastrula oxygen consumption was on average 45% higher (17.35 pmol O2 h-1), giving a Q10 temperature effect of 4.4. For bipinnaria, mean oxygen consumption in 2°C larvae (31.4 pmol O2 h-1) was not significantly different from that in larvae at -0.5°C, suggesting bipinnaria metabolism may be less sensitive to temperature change than earlier stages. At 2°C the bipinnaria stage was reached at 30-35 days compared with 45-50 days at 0°C, giving a Q10 of 4.5 for temperature effects on development. The method here used a new, highly sensitive micro-respirometry method that is inexpensive and straightforward in design. Individual larvae of O. validus were held in 35- to 50-µl respirometers. These larvae have very low metabolic rates, and published work on such organisms have utilised at least 25 individuals per chamber. The oxygen content of the respirometers was measured using a 25-µl sample injected into a couloximeter. Oxygen consumption rates down to -1 pmol h-1 can be detected. Under optimum conditions oxygen consumption of a single larva of -4 pmol O2 h-1 was measured with an accuracy of ᆨ%. Values of ~15 pmol h-1 could routinely be measured with this accuracy. This method would allow oxygen consumption to be evaluated in individual field-caught larvae of most marine ectotherms.  相似文献

Young oysters (Crassostrea virginica) were continuously exposed to Aroclor® 1254, a polychlorinated biphenyl (PCB), in flowing, unfiltered seawater. Growth rate (height and in-water weight) was significantly reduced (=0.05) in oysters exposed to 5 g/l (ppb) for 24 weeks. Growth rate was not affected in oysters exposed to 1 ppb for 30 weeks. Mortality was not significant in exposed and control groups. In oysters exposed to 5 ppb, greatest PCB residue (whole body) was 425 mg/kg (ppm), 85,000x the concentration in the water, and less than 0.3 ppm was retained after 28 weeks depuration in PCB-free water. In oysters exposed to 1 ppb, greatest residue was 101 ppm, 101,000x the concentration in the water, and less than 0.2 ppm was retained after 12 weeks depuration. Examination of oysters exposed to 5 ppb of this PCB for pathogenesis revealed atrophy of digestive diverticular epithelium and degeneration of vesicular connective tissues concomitant with leukocytic infiltration, but tissue recovery seemed excellent after 12 weeks depuration.Registered trademark, Monsanto Company, St. Louis, Missouri, USA. The possible interactive effect of the chlorides of copper and mercury on the euryhaline amphipod Gammarus duebeni in 100% sea water was examined using the following indices: (i) 96 h LC50 values, (ii) urine production rates and (iii) degree of mercury accumulation. Both (a) the interaction of the chlorides of mercury and copper together in solution and (b) the influence of cupric chloride pre-treatment of individuals prior to exposure to mercuric chloride were investigated. Presence of a sublethal level of cupric chloride protected G. duebeni against the toxic action of mercuric chloride. Cupric chloride pretreatment was not so effective. The nature of the interaction between mercury and copper is discussed.  相似文献

The accumulation of cadmium was investigated in two species of oysters [Crassostrea gigas (L.) and Ostrea edulis (L.)] from the same environment and in oysters of the same species (O. edulis) from two different environments (contaminated and uncontaminated), under controlled laboratory conditions (33‰ salinity, 10°C, 100 μg Cd l-1) for up to 111 d in 1982. C. gigas accumulated cadmium twice as fast as O. edulis (1.07 vs 0.52 μg Cd g-1 wet wt d-1). Furthermore, O. edulis from an uncontaminated environment accumulated cadmium faster than O. edulis from a metal-contaminated environment (0.52 vs 0.34 μg Cd g-1 wet wt d-1). There was no effect of cadmium exposure on total soft-tissue copper and zinc concentrations. Investigation of cytosolic metal-binding using Sephadex G-75 gel-permeation chromatography indicated that binding to very low molecular weight ligands (MW<1000) accounted for>70% of the cytosolic zinc in all oysters and>40% of the cytosolic cadmium in all oysters except O. edulis from Conwy at 83 d. In copper-contaminated oysters, excess copper was also associated with very low molecular weight ligands. Intermediate molecular weight cadmium/copper-binding proteins (similar to metallothionein in molecular weight) were observed in the cytosol and were shown to differ between species in terms of their behavior on Sephadex G-75. Finally, the distribution of accumulated cytosolic cadmium in O. edulis from the contaminated environment was shown to have a unique distribution, i.e., there was no cadmium associated with high molecular weight cytosolic macromolecules. The data indicate that both genetic and environmental factors influence cadmium accumulation in oysters.  相似文献

In spring 2004 and 2005 we performed two sets of experiments with Baltic sprat (Sprattus sprattus balticus Schneider) eggs and larvae from the Bornholm Basin simulating ten different temperature scenarios. The goal of the present study was to analyse and parameterise temperature effects on the duration of developmental stages, on the timing of important ontogenetic transitions, growth during the yolk sac phase as well as on the survival success of eggs and early larval stages. Egg development and hatching showed exponential temperature dependence. No hatching was observed above 14.7°C and hatching success was significantly reduced below 3.4°C. Time to eye pigmentation, as a proxy for mouth gape opening, decreased with increasing temperatures from 17 days post hatch at 3.4°C to 7 days at 13°C whereas the larval yolk sac phase was shortened from 20 to 10 days at 3.8 and 10°C respectively. Maximum survival duration of non-fed larvae was 25 days at 6.8°C. Comparing the experimental results of Baltic sprat with existing information on sprat from the English Channel and North Sea differences were detected in egg development rate, thermal adaptation and in yolk sac depletion rate (YSDR). Sprat eggs from the English Channel showed significantly faster development and the potential to develop at temperatures higher than 14.7°C. North Sea sprat larvae were found to have a lower YSDR compared to larvae from the Baltic Sea. In light of the predictions for global warming, Baltic sprat stocks could experience improved conditions for egg development and survival.  相似文献

为探讨镉、汞、锌3种重金属的雌激素样作用,采用双杂交酵母法测定了醋酸镉、氯化汞、醋酸锌单独作用时对重组基因酵母β-半乳糖苷酶诱导活性的EC50值.实验结果表明,醋酸镉浓度为1.0×10-5mol·L-(1实验浓度范围10-7~10-2mol·L-1),氯化汞浓度为5.0×10-7mol·L-(1实验浓度范围10-9~10-5mol·L-1),醋酸锌浓度为1.0×10-4mol·L-(1实验浓度范围10-9~10-2mol·L-1)时对酵母β-半乳糖苷酶诱导活性最大,分别达到1.3、0.95和1.1U.不能求出镉、汞、锌单独作用时对β-半乳糖苷酶诱导活性的EC50值.一味计算EC50值不仅难以评价重金属的雌激素活性,而且限制重组受体基因酵母法的普遍运用.一些重金属通过MCF-7细胞法(E-screen)呈阳性,通过重组hERα酵母法YES检测呈阴性,有可能是重金属通过与ERβ作用而产生效应.若构建出含ERβ基因的双杂交酵母菌株,与现有的方法互补将能完善环境雌激素的体外测评系统.  相似文献

Acute toxicity bioassays conducted at various salinities demonstrated that mercury (as mercuric chloride) at low concentrations was lethal to Petrolisthes armatus. Ninety-six hour LC50 values varied from 50 to 64 parts per billion (ppb) of mercury, depending on test salinities. Lower salinities. decreased the time to death of mercuryexposed crabs. Differences in survival after 96 h due to salinity were not statistically significant. Blood chloride concentrations were regulated hyperchloride to the medium at low salinities and hypochloride at high salinities by acclimated crabs. The salinity isochloride to blood was 20 S. Transfer of crabs from 15 S to salinities ranging from 7 to 35 S resulted in new steadystate chloride levels within 12 h. Exposure to 50 ppb mercury did not alter chloride ion regulation of either acclimated crabs or crabs adjusting to new salinities.  相似文献

The filtration rates of Mytilus edilis (=galloprovincialis; 40 mm) were determined in relation to food concentration and temperature, using pure suspensions of the unicellular alga Platymonas suecica in concentrations ranging from 3x105 cells/l to 1.5x108 cells/l. The rate of filtration (ml/h/mussel) generally decreased as cell concentrations increased, and dropped to low values when concentrations above 5x107 cells/l were supplied. The amount of water swept clear varied continuously, and noticeable differences in the filtration activity of M. edulis were observed over short time intervals (5 min). Fluctuations of filtered volumes per unit time were greater with lower than with higher concentrations of algae. The influence of temperature on filtration activity was highest between 5°–15°C and 25°–30°C. A temperature increase from 15° to 25°C resulted in only a slight increase in filtration rate. At 5° and 30°C, filtration dropped to very low values, namely 350 and 100 ml/h, respectively. The temperature coefficients for the filtration rates of M. edulis were determined as: Q10 (5° to 15°C)=4.96; Q10 (10° to 20°C)=1.22. The amount of algae cells ingested per mussel per hour is directly related to food concentration. The maximum number of cells filtered/mussel/h in an algal suspension of 70x106 cells/l was 21.5x105 cells/h. Cell concentrations of up to 40x106 cells/l were swept clear without producing pseudofaeces. The critical cell density for M. edulis was reached at algal concentrations of 70 to 80x106 cells/l. Above these concentrations no normal filtration activity was observed.  相似文献

The Channel of Santa Cruz is a mangrove area in northeastern Brasil 40 km north of Recife. Until 1991 a chlor-alkali factory discharged large amounts of mercury into the main tributary of the channel, the Rio Botafogo. The objective of this study was to assess the potential of the mangrove oyster Crassostrea rhizophorae as a bioindicator for mercury in this area and to examine the influence of condition on mercury concentrations in the oysters. The investigation was carried out in the late rainy season (July to September) 1993, in the rainy season (April to June) 1994 and in the dry season (November/December) 1994. At 11 stations in the channel, mangrove oysters, surface sediments and suspended matter were sampled, representing different compartments of the system. The distribution of mercury in the Channel of Santa Cruz exhibited for all compartments the same distinct spatial pattern with maxima in the Rio Botafogo. Seasonal variations were small and revealed no clear tendencies. In addition, mangrove oysters were transplanted from more contaminated stations to less contaminated stations and vice versa. These experiments were designed to study the oysters' capability to adjust their mercury concentrations to a changing bioavailability of mercury. The transplantation experiments suggest that the oysters are capable to adjust to changing ambient mercury availability within a few months. The transplantation experiments also revealed the importance of condition changes on the mercury concentration of the oysters. This factor should not be ignored during pollution studies. However, condition had apparently no strong influence on the spatial or seasonal mercury variations of the oysters. The developmental stages from megalopa to third crab of the blue crab Callinectes sapidus Rathbun were tested in 12 combinations of cadmium (0, 50, and 150 ppb) and salinity (10, 20, 30, and 40) at 25°C. A reduction in survival and a significant delay in development from megalopa to third crab occurred within each salinity regime in 50 ppb compared with the control. Comparison of the delay in development within each salinity regime revealed that the sublethal effect of cadmium was most pronounced in the salinities normally preferred by C. sapidus. A similar comparison within each cadmium concentration, however, showed that the developmental time from megalopa to third crab was approximately the same irrespective of salinity. The developmental stages from hatch to first crab of the mud-crab Rhithropanopeus harrisii (Gould) were examined in 63 combinations of cadmium (0, 50, and 150 ppb), salinity (10, 20, and 30), constant temperature (20°, 25°, 30°, and 35°C) and cycling temperature (20° to 25°C, 25° to 30°C, and 30° to 35°C). The results indicated that cycling temperatures may have a stimulating effect on survival of the larvae compared to constant temperatures, both in the presence and in the absence of cadmium. Effects of cadmium and salinity and their interaction on the survival of the larvae from zoeae to megalopa were documented at most of the temperatures by analyses of variance. The zoeal larvae were more susceptible to cadmium than the megalopa. Effects of different combinations of cadmium and salinity on the duration of larval development were assessed by a t-test.  相似文献

Rhodeus amarus acclimated to 10° or 20°C die at 31.7° or 36.4°C, respectively (cessation of operculum movements). After raising the adaptation temperature (AT) from 10° to 20°C, adaptation is complete after 8 days. The toxicity of actinomycin D, injected intraperitoneally, is AT-dependent (e.g. at 5°C, LD50–7 days=0.8 μg/g fish; at 25°C, LD50–7 days=0.35 μg/g fish). In the following experiments 0.5 μg actinomycin/g fish were used. After actinomycin-injection, RNA-values in white dorsal muscle decreased about 50% within 12 h and the values of control individuals were finally attained after 16 days. Following an AT-change from 10° to 20°C, DNA-values in the white dorsal muscle increased about 10%, RNA-values about 30%, within 12 h. However, in fish injected intraperitoneally with actinomycin, no increase in DNA or RNA was noticed. Actinomycin injection per se causes increased heat resistance. This process lasts 10 days. If AT is raised now from 10° to 20°C, the new resistance level is reached later. The control fish gain resistance at a rate of about 1 C°/day during the first days after the temperature change. Actinomycin-treated individuals, however, exhibit an increase of 0.25 C°/day. Inhibition of resistance adaptation due to proteinbiosynthesis-inhibitors supports the view that protein synthesis is of importance for adjustments in heat resistance of intact animals.

As part of a pollution monitoring study, oysters (Crassostrea gasar) were sampled in June 1996, in order to determine the concentrations and distribution of chlorinated hydrocarbons in two stations (near Boulay Island and Riviera Golf) of the Ebrie Lagoon, in the Abidjan area.

The chromatographic analysis of the oven dried samples revealed the presence of PCBs (2.13 to 86.22 ppb or ng/g dry weight), DDE (<0.48 to 169.55 ppb), DDD (<1.04 to 60.27 ppb), DDT (1.43 to 77.81 ppb, lindane (<0.22 to 91.64 ppb), aldrin (<0.30 to 154.1 ppb), endrin (<0.23 to 293.31 ppb) and dieldrin (<0.26 to 199.74 ppb). the results indicated high concentrations in oysters sampled near Boulay Island, due to industrial, agricultural and port activities going on in this area.

Compared to DDD/ΣDDT and DDT/ΣDDT, the DDE/ΣDDT ratios were high indicating old DDT inputs. the ΣDDT/PCBs ratios were higher than 1 illustrated the predominance of organochlorine inputs from agriculture activities. the presence of lindane in both stations illustrated the problem of fishing with toxic products.

Negative correlations found between dry tissue weight and organochlorine compounds concentrations showed that young oysters may concentrate more of these substances. More studies are needed in order to make any conclusions on their bioaccumulation trends.  相似文献

The effects of environmental factors on bioactivity, adsorption and persistence of pretilachlor were studied in the laboratory and greenhouse using cucumber (Cucumis sativus L.) as the bioassay species. The three soils studied viz. Bernam, Selangor and Sabrang series were chosen for their different characteristics. The half‐life of pretilachlor decreased from 10.24 to 4.90 days as temperature increased from 25°C to 35°C in the Selangor Series soil and from 10.86 to 7.63 days in the Bernam Series soil at 60% field capacity. At the same moisture level, an increase of temperature from 25°C to 35°C also reduced the half‐life of pretilachlor in Sabrang soil from 8.87 to 2.59 days. The half‐life of pretilachlor also decreased with increasing moisture levels in Selangor and Sabrang series but not in Bernam series soils. The greatest adsorption of pretilachlor was observed in Bernam series, followed by Selangor and Sabrang series. No phytotoxic residue of pretilachlor was detected in the supernatant after 10 h incubation. Since the residue was strongly adsorbed in Bernam series, its biological activity was less than in the other soils studied.  相似文献

The first incident of green oysters (Crassostrea gigas) was reported in Taiwan in the Charting coastal area in January, 1986 and mortality was reported three months later. the cause of the greened oysters was identified as copper pollution. the copper content of the green oysters was extremely high - 2100 ppm, 2225 ppm, and 4400 ppm dry weight, in January, 1986, February 1987, and January 1989, respectively. in this paper we summarize the seasonal and regional distributions of copper species (complexed by inorganic and organic ligands, labile and non-labile, polar and non-polar) and forms (dissolved and particulate) and the hydrographic and biomass parameters (mainly particulate organic carbon, chloropyll -α, adenosine triphosphate and primary production) in sea water in the Erhjin Chi coastal area. in general, high concentrations of particulate material (0.24 to 724 ppb) and non-labile organic copper (0.03 to 21.5 ppb) were observed. Low values of polar organic copper (<0.02 to 16.5 ppb) indicated that non-polar organic complexes (0.3 to 20.4 ppb) from man-made organic pollutants were the major complexes in the area studied. On the basis of this data, the cause of greening and mortality in oysters is evaluated in relation to the bioavailable copper (sum of particulate and labile copper) and copper assimilative capacity (detoxicant). Finally, the correlations between the species and forms of copper, hydrographic and biomass parameters are also discussed.  相似文献
