This work was undertaken to explore the potential of proteomics to dissect parallel and consecutive events of cadmium stress response in the lichen Physcia adscendens (Fr.) H. Olivier. Thalli were exposed to 0 (control) and 36 μM Cd for 6, 18, 24 and 48 h. Two-dimensional electrophoresis and mass spectrometry analyses showed an 80-85% spot identity between 6 and 18 h vs. 24 and 48 h of Cd exposure. Putative heat-shock proteins and glutathione S-transferase generally increased their expression all over the Cd treatments. By contrast, ABC transporters were underexpressed after 6-18 h, but in some cases induced after 24-48 h of Cd exposure. The cytochrome P450 appeared to have a variable expression pattern over time. Overall these data suggest that a considerable importance in the response of P. adscendens thalli to Cd stress can be assumed by differential expression of various protein families.  相似文献

Some algae inhabit Cu-polluted soils. Intracellular Cu-accumulation and production of non-protein thiols in response to copper stress were compared in Stichococcus minor and Geminella terricola isolated from Cu-polluted and unpolluted soils, respectively. Cu-exposed (0.5 μM) S. minor accumulated lower amounts of copper (0.38 mM) than G. terricola (4.20 mM) and maintained 8.5-fold higher level of glutathione (GSH) than G. terricola. The ratio GSH/0.5 GSSG in the Cu-treated S. minor (7.21) was 7-times higher than in G. terricola. Reduced and oxidized forms of phytochelatins were found in both algae. Under copper stress (5 μM) the ratio -SHtotal/Cuintracellular in S. minor ranged from 2.3 to 6.2, while it was lower than 1.0 in G. terricola. Low intracellular Cu-accumulation and maintenance of high GSH level concomitant with PCs production seem to be responsible for a higher Cu-resistance of S. minor than G. terricola.  相似文献

通过修复培养实验和BCR连续提取实验研究牛粪生物炭(DM)和水稻秸秆生物炭(RS)对2种镉污染土壤的修复效果、影响因素及修复后的Cd的形态分布,探讨可能存在的修复机理。经56 d修复后,与CK相比,5%添加量的牛粪生物炭(DM5%)和水稻秸秆生物炭(RS5%)使TCLP提取态Cd在S1土壤中分别降低了15%和18%,在S2土壤中分别降低了5%和6%。但生物炭添加量为1%时对S2土壤中Cd无显著修复效果。DM5%和RS5%处理使Cd的酸可溶态在S1土壤中降低8.66%和9.25%,在S2土壤中降低7.86%和13.4%,相应的残渣态在S1土壤中升高8.30%和10.54%,在S2土壤中升高8.67%和14.92%。同时,DM5%和RS5%处理使土壤pH提高了7.69%~13.13%,TCLP提取态P增高了0.046~0.39 mg·g-1。结果表明,添加量为5%的牛粪生物炭和秸秆生物炭可有效修复Cd污染土壤。  相似文献

The uptake and metabolism of arsenate, As(V), as a function of time and concentration were examined in the lichen Hypogymnia physodes (L.) Nyl. Lichen thalli were exposed to As(V) in the form of a solution. Exponential uptake of As(V) from 4 microg mL(-1) As(V) solution was accompanied by constant arsenite, As(III), excretion back into the solution. Arsenate taken up into the lichens from 0, 0.1, 1, 10 microg mL(-1) As(V) solutions was partially transformed into As(III), dimethylarsinic acid (DMA) and (mono)methylarsonic acid (MA). 48 h after exposure, the main arsenic compound in the lichens was DMA in 0.1, As(III) in 1 and As(V) in 10 microg mL(-1) treatment. The proportion of methylated arsenic compounds decreased with increasing arsenate concentration in the exposure solution. These results suggest that at least two types of As(V) detoxification exist in lichens; arsenite excretion and methylation.  相似文献

An acute toxicity study of three metals to Hydra species carried out using two different assessment methods, (i) determination of the LC50 and (ii) measurement of progressive morphological changes, demonstrated that relative toxicity decreased from copper to cadmium with zinc the least toxic for all species. The latter method revealed more details of the effect on Hydra in terms of physical damage to the polyp but both methods indicated that H. viridissima was more sensitive to copper and cadmium than H. vulgaris1 (Zurich strain, male clone), H. vulgaris2 (a dioecious strain reproducing sexually and asexually) and H. oligactis (dioecious, reproducing sexually and asexually). The responses to zinc were similar for all Hydra. The possible role of metabolic interactions between H. viridissima and its symbiotic green algae in contributing to the greater sensitivity of this polyp is discussed.  相似文献

This study aimed at investigating the cellular distribution of Hg in the lichens Cladonia arbuscula subsp. mitis and Peltigera rufescens treated with Hg2+ and at testing if Hg treatment affects selected physiological parameters. In both species, increasing Hg accumulation under increasing Hg supply in the treatment solutions was found. P. rufescens showed a higher intracellular accumulation. Photosynthetic parameters were negatively affected in both species, as indicated by the decrease in photosynthetic pigments content, photosynthetic efficiency and chlorophyll integrity. Cell membranes of both species endured damage as indicated by the increase in the concentration of products of lipid peroxidation and decrease in ergosterol content. Nevertheless, differences between the two species were found, suggesting a differential sensitivity to Hg.  相似文献

As mercury and lead, cadmium (Cd) is one of the highly toxic metals in both the ocean and land environments, but its toxicological mechanism in organisms including human is still unclear because of the complex toxicological pathways in vivo. Here, the alga Chlorella vulgaris were cultivated at room temperature under the stress of cadmium (1 mg L−1) to obtain a toxic food, and then the contaminated food were directly supplied to oyster (Saccostrea cucullata) in seawater. After feeding with C. vulgaris contaminated with Cd (C. vulgaris-Cd), the differential proteins in the oyster gonad (OG) were effectively separated and identified with proteomic approaches. Eleven protein spots were observed to be significantly changed in the OG feeding with C. vulgaris-Cd, which seven spots of these differential proteins were down-regulated while four spots were up-regulated. These altered spots were further excised in gels and identified by a combined technique of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF/TOF MS) and database searching. A portion of these differential proteins were further proofed by real-time PCR and Western blotting. The results indicate that the major functions of these differential proteins were described as follows: binding, protein translocation, catalysis, regulation of energy metabolism, reproductive function and skeleton structure. These differential proteins in part may effectively provide a few novel biomarkers for the evaluation of Cd pollution level via a food pathway for harming halobios, mammal and human health, and for understanding the complex mechanisms of Cd toxicity in vivo.  相似文献

在镉(Cd)污染区域筛选富集能力强的本土野生植物是Cd污染土壤植物修复的路径之一。在云南松华坝流域土壤Cd含量超风险筛选值地块生长的杂草中,初步筛查表明牛膝菊、艾蒿和灰藜具有Cd富集潜力。为进一步探明其修复能力,通过盆栽和田间实验比较了这3种野生植物的Cd富集特性和修复能力。结果表明,牛膝菊和艾蒿Cd富集系数与转运系数均大于1,灰藜小于1。施肥措施不仅能显著提高3种本土野生植物的株高和地上部生物量,而且能显著提高牛膝菊地上部和根系的Cd含量、Cd转运系数和Cd富集系数。牛膝菊地上部和根系的Cd含量以及艾蒿的根系Cd含量均随土壤Cd含量的增加而增加。与艾蒿相比,牛膝菊具有更强的Cd转运和富集能力。在100 mg·kg−1外源Cd添加的土壤中,牛膝菊的地上部和根系Cd含量分别为165.5 mg·kg−1和147.5 mg·kg−1。田间实验表明,每茬牛膝菊和艾蒿的鲜重分别为26.0 t·hm−2和32.0 t·hm−2,可分别带走Cd 3.29 g·hm−2和3.35 g·hm−2,均可用作当地土壤修复的备选植物。该结果可为相似环境下的土壤镉污染植物修复材料的选择提供参考。  相似文献

Cetaceans hunted coastally in Japan include several species of odontocete (dolphins, porpoises and beaked whales), and fresh and frozen red meat and blubber, as well as boiled internal organs, such as liver, lung, kidney and small intestine, are still sold for human consumption. Furthermore, red meat and blubber products originating from mysticete minke whales caught in the Antarctic and Northern Pacific are also sold for human consumption. We surveyed mercury and cadmium contamination levels in boiled liver, lung, kidney and red meat products being marketed in Japanese retail outlets. We also analyzed the DNA of these products to obtain information concerning gender and species. Total mercury (T-Hg) and methyl mercury (M-Hg) contamination levels in all the cetacean products were markedly higher in odontocete species than in mysticete species, and slightly higher in females than in males. T-Hg contamination in the organs was seen in the following order: boiled liver > boiled KIDNEY=boiled lung > red meat. In particular, T-Hg concentrations in the boiled liver were high enough to cause acute intoxication even from a single ingestion: the mean ±SD (range) of T-Hg was 388 ± 543 (0.12–1980) μg/wet g. In contrast, although M-Hg contamination in the liver was not markedly higher than that in other organs, M-Hg contamination was in the following order: boiled liver > odontocete red meat > boiled kidney > boiled lung. The contamination levels of T-Hg and M-Hg in odontocete red meat, the most popular whale product, were 8.94 ± 13.3 and 5.44 ± 5.72 μg/wet g, respectively. These averages exceeded the provisional permitted levels of T-Hg (0.4 μg/wet g) and M-Hg (0.3 μg/wet g) in marine foods set by the Japanese Ministry of Health, Labor and Welfare by 22 and 18 times, respectively, suggesting the possibility of chronic intoxication by T-Hg and M-Hg with frequent consumption of odontocete red meat. Cadmium contamination levels in boiled liver, kidney and lung were 8.59 ± 12.0, 10.4 ± 8.6 and 1.66 ± 1.27 (μg/wet g), respectively.  相似文献

以鲫鱼(Carassius auratus)作为实验对象,经过40 d Pb2+、Cd2+不同浓度的暴露后,运用SDS-PAGE和Western Blotting方法检测鱼肝脏组织内应激蛋白HSP70的诱导表达情况.结果表明,在实验浓度下,与对照组相比,Pb2+、Cd2+对鱼肝脏内HSP70有显著的诱导(P<0.05),但在Cd2+浓度为0.2 mg/L有停止表达HSP70的现象,可能是由于浓度过高造成了组织病理损伤而破坏了诱导表达机制.实验还发现,在实验浓度低于国家渔业用水标准时,HSP70仍然表现为明显诱导(P<0.05),充分说明运用分子生物学指标要比传统的环境检测指标敏感,具有对污染物早期预警的作用.  相似文献

Abstract

Toxicity of heavy metals to living organisms is a worldwide research topic. Although, much has been discovered about cadmium and nickel impact on biological systems, a lot still remains unclear. We used inductively coupled plasma – optical emission spectroscopy to address the question of the effect of two different heavy metals nickel, and cadmium on intracellular ion balance. Increase or decrease of the content of several essential cations including Ca2+, Na+, K+, Mg2+, Cu2+, Fe3+ in the yeast Schizosaccharomyces pombe was determined. Our results revealed that the cell exposure to high nickel and cadmium concentrations led to significant elevation of Ca2+, Na+, Mg2+, Cu2+, Fe3+ levels in the yeast cell, while the content of K+ decreased. Correlation analyses showing in the presence of nickel and cadmium strong positive correlation among each tested element (Ca2+, Na+, Cu2+, Mg2+ and Fe3+) except for K+, demonstrate the significant impact of heavy metal treatment to ion homeostasis of the cell. Our data indicate that acute nickel and cadmium contamination leads to substantial ionome misbalance in yeast.  相似文献

In this study, phosphate-solubilizing bacteria (PSB), Bacillus megaterium, were used to enhance Cd bioavailability and phytoextractability of Cd from contaminated soils. This strain showed a potential for directly solubilizing phosphorous from soils more than 10 folds greater than the control without inoculation. The results of pot experiments revealed that inoculation with B. megaterium significantly increased the extent of Cd accumulation in Brassica juncea and Abutilon theophrasti by two folds relative to the uninoculated control. The maximum Cd concentrations due to inoculation were 1.6 and 1.8 mg Cd g−1 plant for B. juncea and A. theophrasti after 10 wk, respectively. The total biomass of A. theophrasti was not significantly promoted by the inoculation treatment, yet the total biomass of B. juncea increased from 0.087 to 0.448 g. It is also worth to mention that B. juncea predominantly accumulates Cd in its stems (39%) whereas A. theophrasti accumulates it in its leaves (68%) after 10 wk. The change of the Cd speciation indicated that inoculation of B. megaterium as PSB increased the bioavailabilty of Cd and consequently enhanced its uptake by plants. The present study may provide a new insight for improving phytoremediation using PSB in the Cd-contaminated soils.  相似文献

The combined effects of titanium dioxide (TiO2) nanoparticles and humic acid (HA) on the bioaccumulation of cadmium (Cd) in Zebrafish were investigated. Experimental data on the equilibrium Cd bioaccumulation suggest that only the dissolved Cd effectively contributed to Cd bioaccumulation in HA solutions whereas both the dissolved and TiO2 associated Cd were accumulated in TiO2 or the mixture of HA and TiO2 solutions, due likely to the additional intestine uptake of the TiO2-bound Cd. The equilibrium Cd bioaccumulation in the mixed system was comparable to that in the corresponding HA solutions, and significantly lower than that in the corresponding TiO2 solutions (n = 3, p < 0.05). The presence of either HA or TiO2 (5-20 mg L−1) in water slightly increased the uptake rate constants of Cd bioaccumulation whereas combining HA and TiO2 reduced the uptake rate constants.  相似文献

Tagetes erecta has a high potential for cadmium (Cd) phytoremediation. Through several hydroponic experiments, characteristics of 108Cd distribution and accumulation were investigated in T. erecta with split -roots or removed xylem/phloem. The results showed that 108Cd transport from roots to aboveground tissues showed the homolateral transport phenomenon in split-root seedlings. 108Cd content of leaves on the +108Cd side and the −108Cd side was not significantly different, which implied that there was horizontal transport of 108Cd from the +108Cd side to the −108Cd side in cut-root seedlings. Like 108Cd transport, the transport of 70Zn was homolateral. Reduction of water consumption in the removed xylem treatment significantly decreased 108Cd accumulation; whereas, the removed phloem treatment had no significant effect on water consumption, but did decrease 108Cd accumulation in leaves of the seedlings. The removal of phloem significantly reduced distal leaf 108Cd content, which was significantly lower than that in the basal leaves in both the split-root and unsplit-root seedlings. Overall, the results presented in this study revealed that the root to aboveground cadmium translocation via phloem is as an important and common physiological process as xylem determination of the cadmium accumulation in stems and leaves of marigold seedlings.  相似文献

The competitive adsorption equilibrium isotherms of Cu2+ and Cd2+ on kaolin have been measured at 298 K, in the presence and the absence of humic acids (HAs). HAs were found to enhance the metal adsorption capacity of mineral surfaces, in particular kaolin. This enhancement was also observed in the competitive adsorption of copper and cadmium on kaolin and kaolin–HA complex. This competitive adsorption shows that the presence of Cd2+ has not an important effect on Cu2+ adsorption, whereas a dramatic decrease is observed on the adsorption of Cd2+ in the presence of Cu2+. The Freundlich isotherm equation was found to provide an excellent fit to the experimental data. These results were compared with the independent adsorption of both heavy metals.  相似文献

The Gironde fluvial estuarine system is impacted by historic metal pollution (e.g. Cd, Zn, Hg) and oysters (Crassostrea gigas) from the estuary mouth have shown extremely high Cd concentrations for decades. Based on recent work (Chiffoleau et al., 2005) revealing anomalously high Ag concentrations (up to 65 mg kg−1; dry weight) in Gironde oysters, we compared long-term (∼1955-2001) records of Ag and Cd concentrations in reservoir sediment with the respective concentrations in oysters collected between 1979 and 2010 to identify the origin and historical trend of the recently discovered Ag anomaly. Sediment cores from two reservoirs upstream and downstream from the main metal pollution source provided information on (i) geochemical background (upstream; Ag: ∼0.3 mg kg−1; Cd: ∼0.8 mg kg−1) and (ii) historical trends in Ag and Cd pollution. The results showed parallel concentration-depth profiles of Ag and Cd supporting a common source and transport. Decreasing concentrations since 1986 (Cd: from 300 to 11 mg kg−1; Ag: from 6.7 to 0.43 mg kg−1) reflected the termination of Zn ore treatment in the Decazeville basin followed by remediation actions. Accordingly, Cd concentrations in oysters decreased after 1988 (from 109 to 26 mg kg−1, dry weight (dw)), while Ag bioaccumulation increased from 38 up to 116 mg kg−1, dw after 1993. Based on the Cd/Ag ratio (Cd/Ag ∼ 2) in oysters sampled before the termination of zinc ore treatment (1981-1985) and assuming that nearly all Cd in oysters originated from the metal point source, we estimated the respective contribution of Ag from this source to Ag concentrations in oysters. The evolution over the past 30 years clearly suggested that the recent, unexplained Ag concentrations in oysters are due to increasing contributions (>70% after 1999) by other sources, such as photography, electronics and emerging Ag applications/materials.  相似文献

This paper analyzes the detoxification mechanisms adopted by amino- and organic acids to alleviate Cd toxicity. In addition, with our published data on Zn–Cd interactions, the influence of Zn (200 μM) supplements on the detoxification mechanisms of amino- and organic acids have also been studied. The experimental studies on metal uptake, lipid peroxidation levels, estimation of reduced and oxidized glutathione levels as well as γ-glutamylcysteine synthetase activity in amino acid supplemented Cd treatments indicated glutathione-mediated detoxification system, which was also enhanced by Zn (200 μM) supplements. However Zn did not aid in glutathione synthesis, but maintained the ratio of reduced and oxidized forms. The supplementation of organic acids to Cd treatments indicated detoxification through the mechanism of chelation. Zn seemed to be less influential on organic acids-mediated detoxification mechanism as compared to amino acid mediated detoxification system.  相似文献

Plants grown in contaminated areas may accumulate trace metals to a toxic level via their roots and/or leaves. In the present study, we investigated the distribution and sources of Pb and Cd in maize plants (Zea mays L.) grown in a typical zinc smelting impacted area of southwestern China. Results showed that the smelting activities caused significantly elevated concentrations of Pb and Cd in the surrounding soils and maize plants. Pb isotope data revealed that the foliar uptake of atmospheric Pb was the dominant pathway for Pb to the leaf and grain tissues of maize, while Pb in the stalk and root tissues was mainly derived from root uptake. The ratio of Pb to Cd concentrations in the plants indicated that Cd had a different behavior from Pb, with most Cd in the maize plants coming from the soil via root uptake.  相似文献
