The content of heavy metals in mineralised dental tissues is an indicator of the exposure of their mineral phase to heavy metals during the time of tooth development and dental tissue formation. Therefore, teeth have been used as bio-indicators which accurately reflect the environmental or dietary exposure of animals and humans to heavy metals. This study follows from the earlier work in which it was demonstrated that the teeth from bank voles inhabiting various environmentally polluted and non-polluted forests in Southern Poland were reliable indicators of exposure to heavy metals. Using analytical techniques employed in the earlier study heavy metal concentrations were obtained in the teeth of bank voles trapped in 1998-2000 to determine if efforts to clean up the environment could be detected in changes in heavy metal concentrations in the teeth. The results show that these efforts are reflected in lower concentrations of heavy metals in the teeth but that cross border contamination remains a problem.  相似文献

Stable lead isotopes are used to illustrate the relation source–receptor and to differentiate between sources of pollution in Greece. Air filters collected in the Kozani–Ptolemais lignite mining area, West Macedonia, point to an impact from gasoline lead as well as lead from the combustion of lignite. This is supported by lead isotope data of wheat grown on reclaimed land. Lead isotope analyses of contemporary teeth from the Lavrio sulphide mining area, southeast of Greece, show the imprint of previous mining activities as well as traffic emissions. Moreover, the Lavrio teeth can be distinguished from one tooth from Athens; the Athens tooth show a stronger impact of gasoline lead. Lead data also imply that the Greek top soil is contaminated by air pollution from earlier sulphide mining and smelting since Hellenic and Roman times.  相似文献

This investigation is based on the premise that heavy metals are sequestered by the mineral phase of teeth, hydroxyapatite, during their formation. Once formed these tissues are not subject to significant turnover and it is suggested, therefore, that they provide a permanent and cumulative record of heavy metal exposure during the development of the teeth. The teeth of the bank vole (Clethrionomys glareolus) were analysed by inductively coupled plasma mass spectrometry to monitor the animals' exposure to lead, cadmium, zinc, copper and strontium. Animals were trapped in clean and polluted areas in Poland in and around Krakow in 1996 and in Olkusz and Legnica in 1996 and 1997. The control area in 1996 was Bieszcady National Park and in 1997 Borecka Forest. The results showed that there were statistically different concentrations of heavy metals in the teeth of animals from different sites. The high levels of pollution from industrial emissions were related to higher concentrations of heavy metals in the teeth. Unexpectedly high levels of heavy metals in the teeth of animals from Bieszczady National Park, thought to be environmentally clean, suggested cross-border pollution from Slovakia and the Ukraine. It was concluded that the teeth of small mammals, such as the bank vole, are a sensitive and reliable marker of environmental pollution by heavy metals.  相似文献

Although fluctuating asymmetry (FA) increases with exposure to certain types of environmental stressors such as temperature extremes, relatively little is known about the effects of interaction (e.g., synergism) between known sources of environmental stress on FA. Knowledge of such interaction effects, and of the magnitude of genotype-by-environment interaction, are of fundamental importance toward predicting the usefulness of FA as a bioindicator of environmental pollution. We tested for synergistic effects on FA between elevated temperature and exposure to lead, and examined FA responses simultaneously in four genetic strains of Drosophila melanogaster known to differ in their degree of developmental instability, and presumably in their buffering capacity. In the absence of heavy metal, bristle FA increased with temperature, but in the presence of lead, FA at high temperature (30 (degrees)C) was reduced to levels similar, or below, that at lower temperature (25 (degrees)C). This temperature by lead interaction was statistically significant, but paradoxical in that the disruptive effects of temperature appeared to be attenuated in the presence of the heavy metal. In no case was there a significant effect of lead on bristle FAs, despite documented assimilation of heavy metal by flies, and in no case was the genotype by environment interaction significant. Whereas lead treatment did not influence survival, survival was reduced at the high temperature, but significantly so only in one genetic strain (Oregon-R). There was no relationship between survival and FA across stress treatments within lines. Thus, any disproportionate stress-induced mortality in developmentally unstable classes (developmental selection) was unlikely to bias the FA results. Our results underscore the need for independent replication of significant findings before FA-based biomonitoring can be responsibly and effectively implemented. The results call for caution in using FA as a biomarker of stress, because stress factors may interact in complex and unpredictable ways, which could result in erroneous conclusions about real levels of stress present in field populations, under the unduly simplistic assumption that stress factors will act additively to increase FA.  相似文献

Damage to animals by atmospheric fluoride pollution involves only those animals who subsequently graze on contaminated forages. The chronic disease which results has been most extensively studied in cattle, where the general symptoms are: A n interference with normal dentition, development of osseous lesions, and a diminished appetite which may lead to weight loss or decreased milk production. Studies completed to date would indicate that depending on the conditions of exposure the tolerance level for dairy cattle is about 1.3 mg F/kg (40 ppm F in diet). Because the route of exposure is via the forage, it would seem unreasonable to base any control standard on atmospheric fluoride levels. The forage fluoride levels, analytical evidence from the animal, or even some early sign of fluoride ingestion on the animal might form a more logical basis for a standard.  相似文献

Developmental disorders of teeth are among the most sensitive targets of polychlorinated dibenzo-p-dioxin and -furan (PCDD/F) exposure. In rats, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) reduces dose-dependently the size of molars, most severely the third lower molars. Dioxins also have effects on developing bone, including altered bone mineral density as well as reduced bending breaking force and stiffness. The aim of this study was to evaluate the use of the third lower molar and long bones as biomarkers of PCDD/F exposure in two wild vole species, the bank vole (Clethrionomys glareolus) and the field vole (Microtus agrestis) collected from a PCDD/F contaminated former sawmill area. Survey of soil and biota of the sawmill area indicated a PCDD/F contamination with a congener profile characteristic for the chlorophenol wood preservative Ky-5. The PCDD/F concentration in the bank vole was notably higher than in the field vole. The third molar of the bank vole was significantly smaller in dioxin-exposed animals compared to control group, while there was no difference between these two groups in the field vole. No significant alterations were observed in bone density and strength in either species except for reduced bending strength of the femur neck in bank vole males exposed to dioxins. Even though the bone changes are among the sensitive endpoints of dioxin-exposure, high variability due to age, size and gender limits their use as biomarkers of wildlife exposure. In conclusion, the size of molar teeth seems to be a sensitive and robust biomarker for PCDD/F exposure in wild bank vole populations and thus worth of further studies.  相似文献

The study of lead exposure among workers in Selangor and the Federal Territory was carried out based on the delta-aminolevulinic acid (ALA) level in urine. Occupations which are expected to have higher lead exposure were chosen in this research. The ALA level in the workers' urine was linked to a few variables which may contribute to the lead level in the body. The result of this study showed that the ALA level of the urine of university students (0.352 +/- 0.038 mg/100 ml) < clerical staff (0.560 +/- 0.043 mg/100 ml) < traffic police (0.612 +/- 0.064 mg/100 ml) < vehicle workshop workers (0.673 +/- 0.099 mg/100 ml) < petrol kiosk workers (0.717 +/- 0.069 mg/100 ml) < bus drivers/conductors (0.850 +/- 0.055 mg/100 ml) which was similar to workers in the printing industry (0.852 +/- 0.110 mg/100 ml). The ALA levels in the urine of the exposed workers were significantly different from the control group (university students). However, results obtained from clerical staff revealed that they were also in the exposed group category. Analysis of variance showed that the exposed groups are in a population which is different from the control population. Correlation tests suggest that there is no significant connection between the ALA level in the urine and the variables tested. Furthermore, Duncan's Multiple Range Test showed no significant differences between the smoking/non smoking group, alcoholic/non-alcoholic group, race and sex (p > 0.05).  相似文献

The rational for the study was to review the literature on the toxicity and corresponding mechanisms associated with lead (Pb), mercury (Hg), cadmium (Cd), and arsenic (As), individually and as mixtures, in the environment. Heavy metals are ubiquitous and generally persist in the environment, enabling them to biomagnify in the food chain. Living systems most often interact with a cocktail of heavy metals in the environment. Heavy metal exposure to biological systems may lead to oxidation stress which may induce DNA damage, protein modification, lipid peroxidation, and others. In this review, the major mechanism associated with toxicities of individual metals was the generation of reactive oxygen species (ROS). Additionally, toxicities were expressed through depletion of glutathione and bonding to sulfhydryl groups of proteins. Interestingly, a metal like Pb becomes toxic to organisms through the depletion of antioxidants while Cd indirectly generates ROS by its ability to replace iron and copper. ROS generated through exposure to arsenic were associated with many modes of action, and heavy metal mixtures were found to have varied effects on organisms. Many models based on concentration addition (CA) and independent action (IA) have been introduced to help predict toxicities and mechanisms associated with metal mixtures. An integrated model which combines CA and IA was further proposed for evaluating toxicities of non-interactive mixtures. In cases where there are molecular interactions, the toxicogenomic approach was used to predict toxicities. The high-throughput toxicogenomics combines studies in genetics, genome-scale expression, cell and tissue expression, metabolite profiling, and bioinformatics.  相似文献

Lead from historical mining and mineral processing activities may pose potential human health risks if materials with high concentrations of bioavailable lead minerals are released to the environment. Since the Joint Expert Committee on Food Additives of Food and Agriculture Organization/World Health Organization withdrew the Provisional Tolerable Weekly Intake of lead in 2011, an alternative method was required for lead exposure assessment. This study evaluated the potential lead hazard to young children (0–7 years) from a historical mining location at a semi-arid area using the U.S. EPA Integrated Exposure Uptake Biokinetic (IEUBK) Model, with selected site-specific input data. This study assessed lead exposure via the inhalation pathway for children living in a location affected by lead mining activities and with specific reference to semi-arid conditions and made comparison with the ingestion pathway by using the physiologically based extraction test for gastro-intestinal simulation. Sensitivity analysis for major IEUBK input parameters was conducted. Three groups of input parameters were classified according to the results of predicted blood concentrations. The modelled lead absorption attributed to the inhalation route was lower than 2 % (mean?±?SE, 0.9 %?±?0.1 %) of all lead intake routes and was demonstrated as a less significant exposure pathway to children’s blood, compared with ingestion. Whilst dermal exposure was negligible, diet and ingestion of soil and dust were the dominant parameters in terms of children’s blood lead prediction. The exposure assessment identified the changing role of dietary intake when house lead loadings varied. Recommendations were also made to conduct comprehensive site-specific human health risk assessment in future studies of lead exposure under a semi-arid climate.  相似文献

Complex mixtures of toxic substances occurring in surface waters are difficult to characterise by chemical analyses because each compound occurs at a very low concentration and requires a specific analytical method to be identified. Ecotoxicological tests on water extracts can be used as a screening tool to evaluate quickly and simply the overall quality of a water body with regard to micropollutant contamination. In this work, a pre-concentration procedure based on solid-phase extraction (SPE), suitable for both biological testing and analytical determination, is proposed. The extraction procedure is an improved version of a methodology used to evaluate the toxicity of organic micropollutants occurring in surface waters. It offers the advantage of using disposable commercial cartridges, which are easier to manage than the columns prepared with macromolecular resins. Water extracts from two representative Italian rivers, characterised by a different gradient of potential contamination and prepared according to the new concentration techniques, are used. The acute toxicity of the water extracts is tested on Daphnia magna and the bioluminescence inhibition in Vibrio fischeri. Criteria based on the concentration factor (CF) are proposed for assessing the hazard to aquatic life due to the exposure to toxic substances in surface waters. The aim of hazard ranking is to focus analytical efforts towards those samples that show the highest toxic potential.  相似文献

The lead acetate paper tape reagent method for the estimation of hydrogen sulfide in air has been reviewed with the conclusions that the colored spots on which the quantitative estimation is based are not stable and do not lend by themselves to accurate results. Although it may be retained for grab sampling work, the lead acetate paper tape reagent is not acceptable as a stoichiometric reagent in any case where long duration sampling periods are concerned like in air pollution studies. A new mercuric chloride paper tape reagent has been developed as a substitute. Due to the fact that the colored spots resulting from the action of hydrogen sulfide on mercuric chloride are sensitive and very stablet, this new paper tape reagent is proposed as an adequate and reliable analytical too for the estimation of hydrogen sulfide in air. Also, a more comprehensive application of the paper tape analytical system is proposed throughout a suitable control of the factor tape retention capacity vs sampling rate which imposes some restrictions in the selection of the practical working range of concentrations. Nevertheless this working range is still wide enough to encompass all the hydrogen sulfide concentrations normally encountered in the city atmosphere.  相似文献

Following the introduction of automobile catalysts in the middle of the 1980s in Germany there is an increasing emission of the platinum-group-metals platinum, palladium (Pd) and rhodium. Still, it remains unclear if these metals are bioavailable for aquatic animals and to which extent they become accumulated by the aquatic biosphere. Because of analytical problems in detecting Pd in small biological samples the present investigation concentrates on the bioavailability of this metal. To answer the question of a Pd uptake by aquatic organisms experimental studies were conducted with European eels maintained in water containing road dust at a concentration of 10 kg/100 l. Following an exposure period of four weeks, samples of liver and kidney were analysed by total-reflection X-ray fluorescence analysis after co-precipitation of Pd with mercury. These experiments revealed an uptake of traffic related Pd by European eels which showed a mean liver Pd concentration of 0.18 +/- 0.05 ng/g (wet wt.), whereas the Pd concentration in the kidney ranged below the detection limit. Thus, in this study we can demonstrate for the first time that automobile catalyst emitted Pd is bioavailable for aquatic animals.  相似文献

Little is known about exposure to lead during childhood in Albania. An analytical survey was carried out in order to determine the levels of lead in blood (PbB) and associated risk factors for 107 inner-city, first-grade schoolchildren living in the town of Berat in Albania, where a plant producing lead-acid batteries exists. The analysis showed that 68% of the children examined had blood levels equal to or greater than 10 micrograms/dL. The mean concentration of lead in the blood of the children was 11.20 micrograms/dL. The influence of the sex and the age of the children, as well as the distance of their homes from the lead-acid battery plant, on the blood lead content was examined.  相似文献

It is known that solid phase microextraction (SPME) fibers can be equilibrated directly within environmental matrices such as water, sediment and soil slurries. Here it is shown that this method can also be applied to biological tissue. SPME extraction of biological matrices reportedly causes lipophilic fouling of the fiber. However, we found no significant measurement bias when combining equilibrium sampling with fiber surface cleaning. The uptake of lipophilic organic pollutants from the tissue and into the SPME fiber coating was characterized by fast equilibrium partitioning without sample depletion and without impacting the sorptive properties of the fiber. The precision of the method when applied to hexachlorobenzene and several PCB congeners in harbor porpoise blubber was 15%, which includes the variation between SPME samplings, manual injections and the instrumental analysis. A good correlation (r(2)=0.95) was obtained between SPME measurements of PCB 153 in blubber and concentrations obtained via a traditional analytical approach. These results indicate that SPME is a promising technique for measuring chemical activity in biological tissue, which would make it a useful tool for studying chemical distribution in organisms as well as biodilution and biomagnification phenomena.  相似文献

The extinction probability is one of the most useful endpoints that are utilized in conservation biology. A parallel approach is advocated for the ecological risk assessment of chemical pollutants. The presented framework estimates extinction probability induced by pollutant chemicals in order to evaluate ecological hazards of pollution, and is applicable to any biological community (aquatic or terrestrial). The analytical framework, which is based on stochastic population dynamics theory, is briefly explained. The extinction risk estimation is feasible if ecotoxicological data concerning pollutant effects on population growth rate of organisms (the intrinsic rate of natural increase), and if environmental exposure concentration is provided. Tentative risk estimation was made for some agrochemicals and surfactants on zooplankton populations (Daphnia) as target organisms.  相似文献

The neurotoxicity of lead is well known, but the exact mechanisms of its toxicity are not solved yet. Disturbances in glutamate homeostasis of neural tissue and interactions of lead with calcium metabolism have been considered as a potential mechanism. In the present study, we evaluate the acute effect of lead exposure by studying glutamate uptake, as well as calcium influx and intracellular lead in cultured rat cerebellar astrocytes. Glutamate uptake and calcium influx were evaluated with radioactive probes by liquid scintillation counting, and intracellular lead was measured by atomic absorption method. Lead increased calcium influx dose dependently, but in an acute 15-min exposure it had no statistically significant effect on glutamate uptake into astrocytes. Calcium decreased at high concentrations but did not hamper lead uptake. Lead uptake rate into astrocytes was high compared to calcium.  相似文献

Dioxin and PCB monitoring programs for food and feeding stuff in most countries of the world, including many European Countries are currently inadequate. Better control of food production lines and food processing procedures is needed to minimize entry of dioxin to the food chain and will help to avoid dioxin contamination accidents. This would also improve the ability to trace back a possible contamination to its source. European guidelines for monitoring programs should be established to ensure comparable and meaningful results. These guidelines should define the minimum requirements for the design of monitoring programs, analytical methods, and quality assurance. Though data from Northern Europe shows that the general population exposure to dioxin and PCB has decreased during the last ten years these compounds continue to be a risk of accidental contamination of the food chain. The most prominent recent example is the Belgian dioxin contamination of feeding stuff in 1999. The Belgian dioxin contamination was not detected due to dioxin monitoring programs but by their direct biological effects seen in animals. Four other cases of dioxin contamination have been detected in Europe since 1997 due to local monitoring programs. One of them (citrus pulp pellets 1998) was in a much larger scale than the Belgian dioxin contamination. The general population's exposure to dioxins and PCBs is still in the same range (1-4 pg WHO-TEQ/kg body weight and day) as the recently revised WHO tolerable daily intake (TDI). There is concern that short-term high level exposure to dioxins, furans, and PCB may cause biological effects on the human fetal development and further research is required. Further actions to control sources building on considerable advances already made in many countries may need to be supplemented by measures to prevent direct contamination of feeding stuff or food to reduce general population exposure further.  相似文献

Sentinel species are biological monitors that accumulate a pollutant in their tissues without significant adverse effects. Primarily used to measure the amount of a pollutant that is biologically available, they may also increase the sensitivity of an analytical procedure or summarise a complex pollution signal. This paper examines the validation of sentinels, referring particularly to the use of invertebrates in monitoring toxic metal pollution in terrestrial ecosystems. Few studies measure the capacity of a sentinel to quantify changes in ambient levels. Definitions of 'bioavailability' also differ between workers and few note that this may be a highly species-specific measure. Validation requires that the temporal and spatial scales over which a sentinel integrates a pollution signal are known. The sentinel has to be calibrated against source concentrations and this relationship shown to be consistent over the normal range of exposure. This requires some consideration of the environmental and biological determinants of pollutant assimilation. Differences between populations can confound simple comparisons between sites based on native populations. Transplanted individuals, matched for age, sex and physiological state, might be used when the aim is not to assess bioavailability to the resident population. A simple measure of their capacity to detect differences in ambient pollutant levels is proposed to evaluate candidate species, to assess their consistency and capacity to equilibrate with their source. A small survey of earthworms from a well-defined gradient of Pb pollution is used to illustrate problems of interpreting tissue concentrations in sentinels.  相似文献

采用正极性高压直流供电和串齿线放电极--管式接地极构成的放电等离子体反应器,研究了苯系物(苯、甲苯和对二甲苯)去除效率与供电电压之间的关系,以及放电极齿轮数对苯系物去除效率、COx(CO2 CO)生成量和能量效率的影响.研究结果表明,苯系物的去除效率、COx生成量皆随电压升高而增大.随着电压升高,能量效率先升后降,当电压为11 kV左右时,能量效率最高.对应放电齿轮数为31的苯系物去除效率、COx生成量和能量效率皆高于放电齿轮数为55或7,这表明对应特定的等离子体反应器,有一最佳放电齿数匹配.  相似文献

Screening-level ecological risk assessments are commonly conducted to identify those contaminants and receptors on which to focus future phases or tiers of the ecological risk assessment process. Most screening assessments are performed using a suite of individual species subjected to intensive evaluation of exposure (endpoint species) and selected for their appropriateness for serving as representatives or 'indicators' for all other species. As site complexity and the number of contaminants of concern increase, it becomes more difficult to assure with confidence that the potential for significant effects has been adequately assessed through an appropriate choice of endpoint species. As an alternative, functional groups demonstrating biological similarity and similar potential for contaminant exposure were developed for INEEL screening-level ecological risk assessments using taxonomic, trophic and habitat parameters. Data for individual species within each group are then integrated to address the potential for risk of adverse effects from contaminant exposure for the group as a whole.  相似文献
