The threshold of toxicological concern (TTC) of a compound represents an exposure value below which the associated human health risk is considered negligible. As such, this approach offers assessing the risk of potential toxicants when little or no toxicological information is available. For the inhalation repeated-dose TTC, the goal was to derive structural alerts that discriminate between high- and low-toxic compounds. A further aim was to identify physicochemical parameters related to the inhalation-specific bioavailability of the compounds, and to explore their use as predictors of high vs low toxicity. 296 compounds with subacute, subchronic and chronic inhalation toxicity NOEC (no-observed effect concentration) values were subdivided into three almost equal-sized high-, medium- and low-toxic (HTox, MTox, LTox) potency classes. Whereas the derived 14 HTox and 7 LTox structural alerts yield an only moderate discrimination between these three groups, the high-toxic vs low-toxic mis-classification is very low: LTox-predicted compounds are not HTox to 97.5%, and HTox-predicted compounds not LTox to 88.6%. The probability of a compound being HTox vs LTox is triggered further by physicochemical properties encoding the tendency to evaporate from blood. The new structural alerts may aid in the predictive inhalation toxicity assessment of compounds as well as in designing low-toxicity chemicals, and provide a rationale for the chemistry underlying the toxicological outcome that can also be used for scoping targeted experimental studies.  相似文献

REACH (registration, evaluation, authorisation and restriction of chemicals) regulation requires that all the chemicals produced or imported in Europe above 1 tonne/year are registered. To register a chemical, physicochemical, toxicological and ecotoxicological information needs to be reported in a dossier. REACH promotes the use of alternative methods to replace, refine and reduce the use of animal (eco)toxicity testing. Within the EU OSIRIS project, integrated testing strategies (ITSs) have been developed for the rational use of non-animal testing approaches in chemical hazard assessment. Here we present an ITS for evaluating the bioaccumulation potential of organic chemicals. The scheme includes the use of all available data (also the non-optimal ones), waiving schemes, analysis of physicochemical properties related to the end point and alternative methods (both in silico and in vitro). In vivo methods are used only as last resort. Using the ITS, in vivo testing could be waived for about 67% of the examined compounds, but bioaccumulation potential could be estimated on the basis of non-animal methods. Ecological risk of chemicals is measured by the quotient of predicted no-effect concentrations and predicted exposure concentrations, which are hard to assess for manufactured nanomaterials (NMs). This paper proposes modifications to currently used models, in order to make them suitable for estimating exposure concentrations of NMs in the aquatic environment. We have evaluated the adequacy of the current guidance documents for use with NMs and conclude that nano-specific fate processes, such as sedimentation and dissolution need to be incorporated. We have reviewed the literature on sedimentation and dissolution of NMs in environmentally relevant systems. We deduce that the overall kinetics of water-sediment transport of NMs should be close to first order. The lack of data on dissolution of NMs under environmentally realistic conditions calls for a pragmatic decision on which rates to be used in modeling. We find that first order removal kinetics for dissolution seems adequate. Based on limited data from literature, probable removal rates range from 0 to 10(-4)s(-1) for sedimentation, and from 0 to 10(-5)s(-1) for dissolution. Further experimental data at environmentally relevant conditions for sedimentation and dissolution of NMs is needed.  相似文献

Understanding the public health implications of chemical contamination of drinking water is important for societies and their decision-makers. The possible population health impacts associated with exposure to disinfection by-products (DBPs) are of particular interest due to their potential carcinogenicity and their widespread occurrence as a result of treatments employed to control waterborne infectious disease.We searched the literature for studies that have attempted quantitatively to assess population health impacts and health risks associated with exposure to DBPs in drinking water. We summarised and evaluated these assessments in terms of their objectives, methods, treatment of uncertainties, and interpretation and communication of results.In total we identified 40 studies matching our search criteria. The vast majority of studies presented estimates of generic cancer and non-cancer risks based on toxicological data and methods that were designed with regulatory, health-protective purposes in mind, and therefore presented imprecise and biased estimates of health impacts. Many studies insufficiently addressed the numerous challenges to DBP risk assessment, failing to evaluate the evidence for a causal relationship, not appropriately addressing the complex nature of DBP occurrence as a mixture of chemicals, not adequately characterising exposure in space and time, not defining specific health outcomes, not accounting for characteristics of target populations, and not balancing potential risks of DBPs against the health benefits related with drinking water disinfection. Uncertainties were often poorly explained or insufficiently accounted for, and important limitations of data and methods frequently not discussed. Grave conceptual and methodological limitations in study design, as well as erroneous use of available dose–response data, seriously impede the extent to which many of these assessments contribute to understanding the public health implications of exposure to DBPs. In some cases, assessment results may cause unwarranted alarm among the public and potentially lead to poor decisions being made in sourcing, treatment, and provision of drinking water. We recommend that the assessment of public health impacts of DBPs should be viewed as a means of answering real world policy questions relating to drinking water quality, including microbial contaminants; that they should be conducted using the most appropriate and up-to-date data and methods, and that associated uncertainties and limitations should be accounted for using quantitative methods where appropriate.  相似文献

Site selection is a typical strategic decision for many industries that deals with specifying the most appropriate location for a facility. In the context of sustainable development, site selection decisions need to be improved by adopting environmental, economic and social requirements. This study aims to frame sustainable location decisions by conducting a case study of siting a new cement plant in the state of Florida. As a part of the study, a wide range of technical and sustainability criteria has developed. These criteria can help decision-makers in the cement industry evaluate the selection of a location with the requirements of sustainable development. The sustainability characteristics of alternative sites in the state of Florida are evaluated based on the criteria to identify the most appropriate ones. The analytical hierarchy processand geographic information system techniques are utilized to weight the criteria and evaluate the characteristics of site candidates.  相似文献

Organic chlorine compounds can be persistent environmental contaminants and may be accumulated through the food chain to the aquatic organisms, to fish and humans, depending basically on their hydrophobic properties. Consequently, there is an interest to measure these organic compounds from both the scientific and regulatory communities. The analytical essays have been improved for measuring specific organic chlorine compounds that present the most toxicological potential (polychlorinated biphenyls [PCBs], certain pesticides and dioxins), although they are tedious and time-consuming procedures. The existing tests to measure adsorbable organic halogens (AOX) or extractable organic halogens (EOX) do not distinguish the more hydrophobic organic chlorine matter. The intention of this paper is to make a review of the existing methods to measure the potentially bioaccumulating organochlorine compounds (OCs) from wastewater and propose a methodology to a standardisation procedure for complex mixtures of OCs in wastewater, such as pulp mill effluents. A new method has been proposed for determining the most hydrophobic part of the extractable organic halogens (EOX(fob)), the lowest reported value is 0.6 microg/l, expressed as chloride, and the relative standard deviation at 20 microg/l is 7% on laboratory samples and 30% on real effluents. This new procedure could be a valuable tool to complement environmental risk assessment studies of wastewater discharges.  相似文献

Production volumes and the use of engineered nanomaterials in many innovative products are continuously increasing, however little is known about their potential risk for the environment and human health. We have reviewed publicly available hazard and exposure data for both, the environment and human health and attempted to carry out a basic risk assessment appraisal for four types of nanomaterials: fullerenes, carbon nanotubes, metals, and metal oxides (ENRHES project 2009(1)). This paper presents a summary of the results of the basic environmental and human health risk assessments of these case studies, highlighting the cross cutting issues and conclusions about fate and behaviour, exposure, hazard and methodological considerations. The risk assessment methodology being the basis for our case studies was that of a regulatory risk assessment under REACH (ECHA, 2008(2)), with modifications to adapt to the limited available data. If possible, environmental no-effect concentrations and human no-effect levels were established from relevant studies by applying assessment factors in line with the REACH guidance and compared to available exposure data to discuss possible risks. When the data did not allow a quantitative assessment, the risk was assessed qualitatively, e.g. for the environment by evaluating the information in the literature to describe the potential to enter the environment and to reach the potential ecological targets. Results indicate that the main risk for the environment is expected from metals and metal oxides, especially for algae and Daphnia, due to exposure to both, particles and ions. The main risks for human health may arise from chronic occupational inhalation exposure, especially during the activities of high particle release and uncontrolled exposure. The information on consumer and environmental exposure of humans is too scarce to attempt a quantitative risk characterisation. It is recognised that the currently available database for both, hazard and exposure is limited and there are high uncertainties in any conclusion on a possible risk. The results should therefore not be used for any regulatory decision making. Likewise, it is recognised that the REACH guidance was developed without considering the specific behaviour and the mode of action of nanomaterials and further work in the generation of data but also in the development of methodologies is required.  相似文献

Fragile coastal areas suffer from human activities. Environmental quality is one of the most important aspects in a tourist destination of sun and sand. The quality of bathing water and sand became indicators in the worldwide competition of beach destinations. We studied the water and sand quality along the beach of Puerto Velero, in northern Colombia. Water and sand beach quality were monitored during thirteen months. This allowed identifying the most significant sources of pollution along the beach, and understanding the interrelationship between tourism and the effects on the environment. Linear correlations allow assessing the association between the number of visitors and the physicochemical and microbiological parameters. The number of visitors was directly correlated with the presence of grease and oils, both in water and sand, as well as with fecal coliforms in water. A relation between the suspended solids and the presence of fecal coliforms in water and sand was observed. This statistical approach allows understanding the origin of beach sand and swimming water pollution at tourist beaches. Overall, sand quality has an important influence and is directly related to the water quality of the beach. Both the number of tourists and their activities are main factors explaining the contamination of the water and sand. This shows the importance of management of waste and wastewater in combination with awareness raising initiatives. More research should include monitoring and evaluating of both water and sand beach and provide data to revise the existing legal framework.  相似文献

Addressing uncertainties in human health risk assessment is a critical issue when evaluating the effects of contaminants on public health. A range of uncertainties exist through the source-to-outcome continuum, including exposure assessment, hazard and risk characterisation. While various strategies have been applied to characterising uncertainty, classical approaches largely rely on how to maximise the available resources. Expert judgement, defaults and tools for characterising quantitative uncertainty attempt to fill the gap between data and regulation requirements. The experiences of researching 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) illustrated uncertainty sources and how to maximise available information to determine uncertainties, and thereby provide an ‘adequate’ protection to contaminant exposure. As regulatory requirements and recurring issues increase, the assessment of complex scenarios involving a large number of chemicals requires more sophisticated tools. Recent advances in exposure and toxicology science provide a large data set for environmental contaminants and public health. In particular, biomonitoring information, in vitro data streams and computational toxicology are the crucial factors in the NexGen risk assessment, as well as uncertainties minimisation. Although in this review we cannot yet predict how the exposure science and modern toxicology will develop in the long-term, current techniques from emerging science can be integrated to improve decision-making.  相似文献

The strengthened environmental regulatory requirements and general awareness of the need for environmental conservation worldwide, design for environment has become a key design criterion in new product development processes. Due to their resources-consuming attribute and the prerequisite expert knowledge in environmental sciences for the impacts interpretations, the existing forms of product-oriented environmental impact assessment methods are impractical to be adopted by product designers in small-and-medium enterprises. Life Cycle Assessment (LCA) is a generally accepted quantitative approach to assess a product’s environmental impact. However, a full LCA study often requires a considerable amount of data and therefore, it is regarded as not a handy tool for product design evaluations particularly at the initial design stage. This leads to our intention to develop an immediately applicable approach for decision-makers to evaluate the design options. The proposed approach integrates Analytic Hierarchy Process and Fuzzy Theory, with Evidential Reasoning (ER) to support the environmental impact evaluations of design options. A case study is carried out to demonstrate its applicability to prioritize the environmental impact of various design options. A symmetrical triangular distribution is introduced for calculating the expected utility and for testing the sensitivity of results.  相似文献

Sediments act as a net sink for anthropogenic contaminants in marine ecosystems and contaminated sediments may have a range of toxicological effects on benthic fauna and associated species. When resuspended, however, particulate-bound contaminants may be remobilised into the water column and become bioavailable to an additional assemblage of species. Such resuspension occurs through a range of natural and anthropogenic processes each of which may be thought of as pulsed disturbances resulting in pulsed exposures to contaminants. Thus, it is important to understand not only the toxicological responses of organisms to resuspended contaminated sediments (RCS), but also the frequency, magnitude and duration of sediment disturbance events. Such information is rarely collected together with toxicological data. Rather, the majority of published studies (> 50% of the articles captured in this review) have taken the form of fixed-duration laboratory-based exposures with individual species. While this research has clearly demonstrated that resuspension of contaminated sediments can liberate sediment-bound contaminants leading to toxicity and bioaccumulation under controlled conditions, the potential for ecological effects in the field is often unclear. Monitoring studies suggest that recurrent natural disturbances such as tides and waves may cause the majority of contaminant release in many environments. However, various processes also act to limit the spatial and temporal scales across which contaminants are remobilised to the most toxic dissolved state. Various natural and anthropogenic disturbances of contaminated sediments have been linked to both community-level and sub-lethal responses in exposed populations of invertebrates and fish in the field. Together these findings suggest that resuspension of contaminated sediments is a frequently recurring ecological threat in contaminated marine habitats. Further consideration of how marine communities respond to temporally variable exposures to RCS is required, as well as research into the relative importance of various disturbances under field conditions.  相似文献

Endocrine disrupting chemicals (EDCs) are exogenous agents that interfere or disrupt the normal synthesis, secretion, transportation, binding and metabolism of natural hormones; eventually dysregulating homeostatic mechanisms, reproduction and development. They are emitted into the atmosphere during anthropogenic activities and physicochemical reactions in nature. Inhalation of these EDCs as particulate and gaseous vapors triggers their interaction with endocrine glands and exerts agonist or antagonists actions at hormone receptors. The endocrine disruption at nanogram levels of EDC's has gained concern in the last decade, due to infertility among men and women, early puberty, obesity, diabetes and cancer. Thus, the review explores the literature that addresses the major occurring EDCs in the atmosphere including phthalates, polychlorinated biphenyls (PCBs), polycyclic aromatic hydrocarbons (PAHs), brominated flame retardants (BFRs), dioxins, alkylphenols (APs) and perfluorinated chemicals (PFCs). Sources, fate, half-life, mechanism, measured concentrations in air, bioaccumulation in tissues, laboratory exposures correlating to toxicological effects of these EDCs in humans and wildlife are discussed.  相似文献

本文构建了包括监管的立法、组织体系、监管权力配置、监管工具与程序、问责机制、监管能力等要素在内的环境监管体制分析框架,从污染物排放趋势、国家制度建设等角度分析了"十三五"时期中国环境监管体制改革面临的挑战、目标和思路。"十三五"时期是中国生态文明建设的重要时期,完善环境监管制度是推进生态文明制度体系建设的重要内容。面对严重环境污染所带来的挑战,在此期间加快环境监管体制改革、提高环境监管有效性是切实实现治污减排的根本保证。完善环境监管体系、改进监管方式、创新监管手段是建立治污减排长效机制的基础和前提,也是完善环境治理体系的重要内容。按照市场经济条件下政府依法行政的要求,中国环境监管体制改革的目标,就是要按照合法性、独立性、透明性、可问责性、专业性和执法能力、可信性等原则建立和完善现代环境监管体制,为此必须从专门立法、组织结构、权力分配、问责机制、监管程序与工具、监管能力建设等各方面推进改革。本文提出了"十三五"时期环境监管体制改革的思路及若干建议,主要包括:加快修订环保法律法规,提高法律的可操作性,推进环境司法常态化、规范化和专门化,进一步提升环境监管法治化水平;从纵向、横向上优化环境监管组织结构,强化区域层级的监管与督查功能,强化中央层级环境监管机构对环境信息的获取能力;优化环境保护主管部门机构设置以及相关职能,建立并完善内部机构的协调机制;完善环境监管的内外部问责机制,把做实对监管者的监管机制作为优化环境监管体制的抓手;强化对监管工具的顶层设计,做实监管程序中关键环节,建立环境监管影响评估制度;加强环境监管能力建设,推进环境监管制度队伍职业化发展,保障各级环境监管机构具有充分履职的能力。环境监管体制的改革和完善,是国家治理体系和治理能力现代化的重要内容。  相似文献

The biodegradation of 16 United States Environmental Protection Agency (USEPA)-listed polycyclic aromatic hydrocarbons (PAHs) present in contaminated soil from a manufactured gas plant site was investigated using laboratory-scale in-vessel composting-bioremediation reactors over 8 weeks. The influence of temperature (T, 38, 55, and 70 degrees C) and soil/green waste ratio (S:GW, 0.6:1, 0.7:1, 0.8:1, and 0.9:1) was investigated. A comparative study using a temperature profile during in-vessel composting-bioremediation to meet current regulatory requirements was also investigated. Temperature and amendment ratio were found to be important operating parameters for PAH removal for in-vessel composting-bioremediation of aged coal tar-contaminated soil. After 8 weeks of continuous treatment, the highest removal of 16 USEPA PAHs was observed at T=38 degrees C and S:GW=0.8:1 (75.2%). Lower removal of 16 USEPA PAHs was observed for temperature profile treatment (60.8%). We recommend that when conventional composting processes using temperature profiles to meet regulatory requirements for pathogen control need to be used, these should start with a prolonged mesophilic stage (6 weeks in this investigation) followed by thermophilic, cooling, and maturation stages.  相似文献

Due to human activities to a greater extent and natural processes to some extent, a large number of organic chemical substances such as petroleum hydrocarbons, halogenated and nitroaromatic compounds, phthalate esters, solvents and pesticides pollute the soil and aquatic environments. Remediation of these polluted sites following the conventional engineering approaches based on physicochemical methods is both technically and economically challenging. Bioremediation that involves the capabilities of microorganisms in the removal of pollutants is the most promising, relatively efficient and cost-effective technology. However, the current bioremediation approaches suffer from a number of limitations which include the poor capabilities of microbial communities in the field, lesser bioavailability of contaminants on spatial and temporal scales, and absence of bench-mark values for efficacy testing of bioremediation for their widespread application in the field. The restoration of all natural functions of some polluted soils remains impractical and, hence, the application of the principle of function-directed remediation may be sufficient to minimize the risks of persistence and spreading of pollutants. This review selectively examines and provides a critical view on the knowledge gaps and limitations in field application strategies, approaches such as composting, electrobioremediation and microbe-assisted phytoremediation, and the use of probes and assays for monitoring and testing the efficacy of bioremediation of polluted sites.  相似文献

The reasons for developing quantitative estimates of uncertainty in environmental risk assessments are discussed along with a method for developing them which involves scientific judgement. In the situation considered here the regulatory needs are ahead of the science, which makes the development of the estimates on uncertainty more difficult, but not impossible. Quantitative estimates for all uncertainties involved in the estimation of risk resulting from exposure to volatile organic compounds (VOCs) in drinking water are developed and tabulated. By far the largest contribution to the uncertainty in the risk estimates for VOCs in drinking water are due to uncertainty in the extrapolation of the dose-response curve to low levels. The uncertainty due to extrapolation is on the order of 104 and 106. Other components of the analysis may contribute uncertainties of a few orders of magnitude. In general the largest uncertainties are in the toxicological data base and the manipulation of it needed to estimate risk. The data base and manipulations needed to estimate exposure due to VOCs in drinking water were at the more an order to magnitudes in uncertainty.  相似文献

Economic appraisal and technical effectiveness of adaptation options are key criteria for judging climate change adaptation investment decisions in all sectors. Yet relatively little methodological guidance exists for determining the most appropriate appraisal techniques for different adaptation options. This paper provides adaptation options and scopes relevant appraisal methods in agriculture focussing on livestock production specifically. We find that for many adaptation options for livestock agriculture, standard (expected) cost-benefit analysis is an appropriate tool. For adaptation options requiring long lead times or those with long lifetimes, techniques incorporating uncertainty (‘robust’ methods) are more suitable, including real options analysis, portfolio analysis and robust decision-making. From a comprehensive list of adaptation options in the livestock sector, we identify the most appropriate appraisal technique for each option and describe how the robust appraisal tools could be applied to heat stress, flood risk and water management.

Zinc oxide nanoparticles (ZnO NPs) are one of the most abundantly used nanomaterials in consumer products and biomedical applications. As a result, human exposure to these NPs is highly frequent and they have become an issue of concern to public health. Although toxicity of ZnO NPs has been extensively studied and they have been shown to affect many different cell types and animal systems, there is a significant lack of toxicological data for ZnO NPs on the nervous system, especially for human neuronal cells and tissues. In this study, the cytotoxic and genotoxic effects of ZnO NPs on human SHSY5Y neuronal cells were investigated under different exposure conditions. Results obtained by flow cytometry showed that ZnO NPs do not enter the neuronal cells, but their presence in the medium induced cytotoxicity, including viability decrease, apoptosis and cell cycle alterations, and genotoxicity, including micronuclei production, H2AX phosphorylation and DNA damage, both primary and oxidative, on human neuronal cells in a dose- and time-dependent manner. Free Zn2 + ions released from the ZnO NPs were not responsible for the viability decrease, but their role on other types of cell damage cannot be ruled out. The results obtained in this work contribute to increase the knowledge on the genotoxic and cytotoxic potential of ZnO NPs in general, and specifically on human neuronal cells, but further investigations are required to understand the action mechanism underlying the cytotoxic and genotoxic effects observed.  相似文献

Global forest product value chains are increasingly subject to regulatory requirements, such as legality and sustainability certification. For African forest products, a shift in the export destinations of forest products towards China, the leading timber product manufacturer, has raised concerns that social and environmental product and process standards are declining. Aggregate drivers of this shift have been documented, yet there has been little enterprise-level analysis of Chinese actors in African timber imports based in China. This paper provides an initial analysis, highlighting implications for existing and emerging regulatory initiatives. Data show that although an increasing number of Chinese private enterprises is engaged in African timber imports, import volumes remain concentrated among a small number of geographically clustered private and state-owned firms. Government-led schemes are beginning to address timber legality concerns. But given China’s growing domestic market for finished wood products, sustainability certification requirements driven by the US and EU will not be sufficient to ensure improved sourcing by Chinese firms.  相似文献
