Metal uptake and its effect on foliar metallothionein 2b (MT2b) mRNA levels were studied in hybrid aspen (Populus tremula × tremuloides) in field conditions. The trees were planted on a site contaminated with several metals, including cadmium (mean 5.1 mg kg−1), chromium (80 mg kg−1), copper (180 mg kg−1), nickel (81 mg kg−1), vanadium (240 mg kg−1) and zinc (520 mg kg−1). Of the ten trace elements analyzed, only Cd and Zn accumulated in the leaves with maximal foliar concentrations of 35 and 2400 mg kg−1 (dry weight), respectively. There was a strong correlation between Cd and Zn concentrations and bioaccumulation factors (concentration in plant/soil) in the leaves, branches and roots, suggesting similar transport mechanisms for these two metals. The levels of MT2b correlated with Cd and Zn concentrations in the leaves, demonstrating that increased MT2b expression is one of the responses of hybrid aspen to chronic metal exposure.  相似文献

The chronic toxicity of zinc oxide nanoparticles (ZnO-NP) to Folsomia candida was determined in natural soil. To unravel the contribution of particle size and free zinc to NP toxicity, non-nano ZnO and ZnCl2 were also tested. Zinc concentrations in pore water increased with increasing soil concentrations, with Freundlich sorption constants Kf of 61.7, 106 and 96.4 l/kg (n = 1.50, 1.34 and 0.42) for ZnO-NP, non-nano ZnO and ZnCl2 respectively. Survival of F. candida was not affected by ZnO-NP and non-nano ZnO at concentrations up to 6400 mg Zn/kg d.w. Reproduction was dose-dependently reduced with 28-d EC50s of 1964, 1591 and 298 mg Zn/kg d.w. for ZnO-NP, non-nano ZnO and ZnCl2, respectively. The difference in EC50s based on measured pore water concentrations was small (7.94-16.8 mg Zn/l). We conclude that zinc ions released from NP determine the observed toxic effects rather than ZnO particle size.  相似文献

Tire wear particles filed from the treads of end-of-life vehicle tires have been added to sea water to examine the release of Zn and the toxicity of the resulting leachate and dilutions thereof to the marine macroalga, Ulva lactuca. Zinc release appeared to be diffusion-controlled, with a conditional rate constant of 5.4 μg[L(h)1/2]−1, and about 1.6% of total Zn was released after 120 h incubation. Exposure to increasing concentrations of leachate resulted in a non-linear reduction in the efficiency of photochemical energy conversion of U. lactuca and, with the exception of the undiluted leachate, increasing accumulation of Zn. Phototoxicity was significantly lower on exposure to equivalent concentrations of Zn added as Zn(NO3)2, suggesting that organic components of leachate are largely responsible for the overall toxicity to the alga. Given the ubiquity and abundance of TWP in urban coastal sediments, the generation, biogeochemistry and toxicity of tire leachate in the marine setting merit further attention.  相似文献

The aim of this study was to investigate the effects of metal mobilizing plant-growth beneficial bacterium Phyllobacterium myrsinacearum RC6b on plant growth and Cd, Zn and Pb uptake by Sedum plumbizincicola under laboratory conditions. Among a collection of metal-resistant bacteria, P. myrsinacearum RC6b was specifically chosen as a most favorable metal mobilizer based on its capability of mobilizing high concentrations of Cd, Zn and Pb in soils. P. myrsinacearum RC6b exhibited a high degree of resistance to Cd (350 mg L−1), Zn (1000 mg L−1) and Pb (1200 mg L−1). Furthermore, P. myrsinacearum RC6b showed multiple plant growth beneficial features including the production of 1-aminocyclopropane-1-carboxylic acid deaminase, indole-3-acetic acid, siderophore and solubilization of insoluble phosphate. Inoculation of P. myrsinacearum RC6b significantly increased S. plumbizincicola growth and organ metal concentrations except Pb, which concentration was lower in root and stem of inoculated plants. The results suggest that the metal mobilizing P. myrsinacearum RC6b could be used as an effective inoculant for the improvement of phytoremediation in multi-metal polluted soils.  相似文献

Although anthropogenic pollutants are thought to threaten reptilian species, there are few toxicity studies on reptiles. We evaluated the toxicity of Pb as lead acetate to the Western fence lizard (Sceloporus occidentalis). The acute lethal dose and sub-acute (14-day) toxicity studies were used to narrow exposure concentrations for a sub-chronic (60-day) study. In the sub-chronic study, adult and juvenile male lizards were dosed via gavage with 0, 1, 10 and 20 mg Pb/kg-bw/day. Mortality was limited and occurred only at the highest dose (20 mg Pb/kg-bw/d). There were statistically significant sub-lethal effects of 10 and 20 mg Pb/kg-bw/d on body weight, cricket consumption, organ weight, hematological parameters and post-dose behaviors. Of these, Pb-induced changes in body weight are most useful for ecological risk assessment because it is linked to fitness in wild lizard populations. The Western fence lizard is a useful model for reptilian toxicity studies.  相似文献

The present study was to isolate and identify a potent algicidal compound from extract of Salvia miltiorrhiza and study the potential inhibition mechanism on Microcystis aeruginosa. Column chromatography and bioassay-guided fractionation methods were carried out to yield neo-przewaquinone A, which was identified by spectral analysis. The EC50 of neo-przewaquinone A on M. aeruginosa were 4.68 mg L−1. In addition, neo-przewaquinone A showed relatively higher security on Chlorella pyrenoidosa and Scenedesmus obliquus, with the EC50 values of 14.78 and 10.37 mg L−1, respectively. For the potential inhibition mechanisms, neo-przewaquinone A caused M. aeruginosa cells morphologic damage or lysis, increased malondialdehyde content and decreased the soluble protein content, total antioxidant and superoxide dismutase activity, and significantly inhibited three photosynthesis-related genes (psaB, psbD, and rbcL). The results demonstrated the algicidal effect of neo-przewaquinone A on M. aeruginosa and provided the possible inhibition mechanisms.  相似文献

Root and shoot lead concentrations and the impact of chelating agents on these were investigated in two populations of the novel metallophyte Matthiola flavida. Plants were exposed in hydroponics to Pb(NO3)2, supplied alone, or in combination with citric acid, or EDDS. When supplied at concentrations expected to bind about 95% of the Pb in a solution containing 1-μM Pb (1000 μM citrate or 3.1 μM EDDS, respectively), the root and shoot Pb concentrations were dramatically lowered, in comparison with a 1-μM free ionic Pb control exposure. A 1-mM EDDS + 1-μM Pb treatment decreased the plants’ Pb concentrations further, even to undetectable levels in one population. At 100 μM Pb in a 1-mM EDDS-amended solution the Pb concentration increased strongly in shoots, but barely in roots, in comparison with the 1-μM Pb + 1-mM EDDS treatment, without causing toxicity symptoms. Further increments of the Pb concentration in the 1-mM EDDS-amended solution, i.e. to 800 and 990 μM, caused Pb hyperaccumulation, both in roots and in shoots, associated with a complete arrest of root growth and foliar necrosis. M. flavida seemed to be devoid of constitutive mechanisms for uptake of Pb-citrate or Pb-EDDS complexes. Hyperaccumulation of Pb-EDDS occurred only at high exposure levels. Pb-EDDS was toxic, but is much less so than free Pb. Free EDDS did not seem to be toxic at the concentrations tested.  相似文献

This experiment examined the effects of sulfate (S) and reduced glutathione (GSH) on arsenic uptake by arsenic hyperaccumulator Pteris vittata after exposing to arsenate (0, 15 or 30 mg As L−1) with sulfate (6.4, 12.8 or 25.6 mg S L−1) or GSH (0, 0.4 or 0.8 mM) for 2-wk. Total arsenic, S and GSH concentrations in plant biomass and arsenic speciation in the growth media and plant biomass were determined. While both S (18-85%) and GSH (77-89%) significantly increased arsenic uptake in P. vittata, GSH also increased arsenic translocation by 61-85% at 0.4 mM (p < 0.05). Sulfate and GSH did not impact plant biomass or arsenic speciation in the media and biomass. The S-induced arsenic accumulation by P. vittata was partially attributed to increased plant GSH (21-31%), an important non-enzymatic antioxidant countering oxidative stress. This experiment demonstrated that S and GSH can effectively enhance arsenic uptake and translocation by P. vittata.  相似文献

We measured heavy metal concentrations in yellow-legged gulls (n = 196) and European shags (n = 189) in order to assess the temporal pattern of contaminant exposure following the Prestige oil spill in November 2002. We analysed Pb, Cu, Zn, Cr, Ni and V levels in chick feathers sampled at four colonies during seven post-spill years (2003-2009), and compared results with pre-spill levels obtained from feathers of juvenile shag corpses (grown in spring/summer 2002). Following the Prestige wreck, Cu (4.3-10 μg g−1) and Pb concentrations (1.0-1.4 μg g−1) were, respectively, between two and five times higher than pre-spill levels (1.5-3.6 and 0.1-0.4 μg g−1), but returned to previous background concentrations after three years. Our study highlights the suitability of chick feathers of seabirds for assessing the impact of oil spills on heavy metal contamination, and provides the best evidence to date on the persistence of oil pollution after the Prestige incident.  相似文献

The halophytic shrub Halimione portulacoides is known to be capable of growth in soils containing extremely high concentrations of Zn. This study evaluated in detail the tolerance and accumulation potential of H. portulacoides under moderate and high external Zn levels. A greenhouse experiment was conducted in order to investigate the effects of a range of Zn concentrations (0-130 mmol L−1) on growth and photosynthetic performance by measuring relative growth rate, total leaf area, specific leaf area, gas exchange, chlorophyll fluorescence parameters and photosynthetic pigment concentrations. We also determined the total zinc, nitrogen, phosphorus, calcium, magnesium, sodium, potassium, iron and copper concentrations in the plant tissues. H. portulacoides demonstrated hypertolerance to Zn stress, since it survived with leaf concentrations of up to 2300 mg Zn kg−1 dry mass, when treated with 130 mmol Zn L−1. Zinc concentrations greater than 70 mmol L−1 in the nutrient solution negatively affected plant growth, in all probability due to the recorded decline in net photosynthesis rate. Our results indicate that the Zn-induced decline in the photosynthetic function of H. portulacoides may be attributed to the adverse effect of the high concentration of the metal on photosynthetic electron transport. Growth parameters were virtually unaffected by leaf tissue concentrations as high as 1500 mg Zn kg−1 dry mass, demonstrating the strong capability of H. portulacoides to protect itself against toxic Zn concentrations. The results of our study indicate that this salt-marsh shrub may represent a valuable tool in the restoration of Zn-polluted areas.  相似文献

Leaching of Cu and Zn from a composite of discarded antifouling paint residues ([Cu] = 288 mg g−1; [Zn] = 96 mg g−1) into natural sea water has been studied over a period of 75 h. Total Cu and Zn were released according to a pseudo first-order reaction, with rate constants on the order of 0.3 and 2.5 (mg L−1)−1 h−1, respectively, and final concentrations equivalent to the dissolution of about 8 and 2% of respective concentrations in the composite. Time-distributions of hydrophobic metals, determined by solid phase extraction-methanol elution, were more complex. Net release of hydrophobic Cu was greater in the absence of light than under a sequence of light-dark cycles; however, hydrophobic Zn release was not detected under the former conditions but contributed up to 50% of total aqueous Zn when light was present. These observations are interpreted in terms of the relative thermodynamic and photolytic stabilities of biocidal pyrithione complexes.  相似文献

UV filters are increasingly used in sunscreens and other personal care products. Although their residues have been widely identified in aquatic environment, little is known about the influences of UV filters to protozoan. The growth inhibition effects, cell viability and oxidative stress responses of four commonly used UV filters, 2-ethylhexyl 4-methoxycinnamate (EHMC), benzophenone-3 (BP-3), 4-methyl-benzylidene camphor (4-MBC) and octocrylene (OC), to protozoan Tetrahymena thermophila were investigated in this study. The 24-h EC50 values with 95% confidence intervals for BP-3 and 4-MBC were 7.544 (6.561–8.675) mg L−1 and 5.125 (4.874–5.388) mg L−1, respectively. EHMC and OC did not inhibit the growth of T. thermophila after 24 h exposure at the tested concentrations. The results of cell viability assays with propidium iodide (PI) staining were consistent with that of the growth inhibition tests. As for BP-3 and 4-MBC, the relatively higher concentrations, i.e. of 10.0 and 15.0 mg L−1, could lead to the cell membranes impairment after 4 h exposure. With the increase of the exposure time to 6 h, their adverse effects on cell viability of T. thermophila were observed at the relatively lower concentration groups (1.0 mg L−1 and 5.0 mg L−1). In addition, it is noticeable that at environmentally relevant concentration (1.0 μg L−1), BP-3 and 4-MBC could lead to the significant increase of catalase (CAT) activities of the T. thermophila cells. Especially for the BP-3, the oxidative injuries were further confirmed by the reduction of glutathione (GSH) content. It is imperative to further investigate the additive action of UV filters and seek other sensitive endpoint, especially at environmentally relevant concentration.  相似文献

A Cd and Zn contaminated soil was mixed and equilibrated with an uncontaminated, but otherwise similar soil to establish a gradient in soil contamination levels. Growth of Thlaspi caerulescens (Ganges ecotype) significantly decreased the metal concentrations in soil solution. Plant uptake of Cd and Zn exceeded the decrease of the soluble metal concentrations by several orders of magnitude. Hence, desorption of metals must have occurred to maintain the soil solution concentrations. A coupled regression model was developed to describe the transfer of metals from soil to solution and plant shoots. This model was applied to estimate the phytoextraction duration required to decrease the soil Cd concentration from 10 to 0.5 mg kg−1. A biomass production of 1 and 5 t dm ha−1 yr−1 yields a duration of 42 and 11 yr, respectively. Successful phytoextraction operations based on T. caerulescens require an increased biomass production.  相似文献

Soil amendments previously shown to be effective in reducing metal bioavailability and/or mobility in calcareous metal-polluted soils were tested on a calcareous dredged sediment-derived soil with 26 mg Cd/kg dry soil, 2200 mg Cr/kg dry soil, 220 mg Pb/kg dry soil, and 3000 mg Zn/kg dry soil. The amendments were 5% modified aluminosilicate (AS), 10% w/w lignin, 1% w/w diammonium phosphate (DAP, (NH4)2HPO4), 1% w/w MnO, and 5% w/w CaSO4. In an additional treatment, the contaminated soil was submerged. Endpoints were metal uptake in Salix cinerea and Lumbricus terrestris, and effect on oxidation-reduction potential (ORP) in submerged soils. Results illustrated that the selected soil amendments were not effective in reducing ecological risk to vegetation or soil inhabiting invertebrates, as metal uptake in willows and earthworms did not significantly decrease following their application. Flooding the polluted soil resulted in metal uptake in S. cinerea comparable with concentrations for an uncontaminated soil.  相似文献

Potential exposures from ground-level pyrotechnics were assessed by air monitoring and developing emission factors. Total particulate matter, copper and SO2 exposures exceeded occupational health guidelines at two outdoor performances using consumer pyrotechnics. Al, Ba, B, Bi, Mg, Sr, Zn, and aldehyde levels were elevated, but did not pose a health hazard based on occupational standards. Emission factors for total particulate matter, metals, inorganic ions, aldehydes, and polyaromatic hydrocarbons (PAHs) were determined for seven ground-supported pyrotechnics through air sampling in an airtight room after combustion. Particle generation ranged from 5 to 13% of the combusted mass. Emission factors (g Kg?1) for metals common to pyrotechnics were also high: K, 23–45; Mg, 1–7; Cu, 0.05–7; and Ba, 0.03–6. Pb emission rates of 1.6 and 2.7% of the combusted mass for two devices were noteworthy. A high correlation (r2 ≥ 0.89) between metal concentrations in pyrotechnic compositions and emission factors were noted for Pb, Cr, Mg, Sb, and Bi, whereas low correlations (r2 ≤ 0.1) were observed for Ba, Sr, Fe, and Zn. This may be due to the inherent heterogeneity of multi-effect pyrotechnics. The generation of inorganic nitrogen in both the particulate (NO2?, NO3?) and gaseous (NO, NO2) forms varied widely (<0.1–1000 mg Kg?1). Aldehyde emission factors varied by two orders of magnitude even though the carbon source was carbohydrates and charcoal for all devices: formaldehyde (<7.0–82 mg Kg?1), acetaldehyde (43–210 mg Kg?1), and acrolein (1.9–12 mg Kg?1). Formation of lower molecular weight PAHs such as naphthalene and acenaphthylene were favored, with their emission factors being comparable to that from the combustion of household refuse and agricultural debris. Ba, Sr, Cu, and Pb had emission factors that could produce exposures exceeding occupational exposure guidelines. Sb and unalloyed Mg, which are banned from consumer fireworks in the US, were present in significant amounts.  相似文献

The consumption of paddy rice (Oryza sativa L.) is a major inorganic arsenic exposure pathway in S.E. Asia. A multi-location survey was undertaken in Guangdong Province, South China to assess arsenic accumulation and speciation in 2 rice cultivars, one an Indica and the other a hybrid Indica. The results showed that arsenic concentrations in rice tissue increased in the order grain < husk < straw < root. Rice grain arsenic content of 2 rice cultivars was significant different and correlated with phosphorus concentration and molar ratio of P/As in shoot, being higher for the Indica cultivar than for the hybrid Indica, which suggests altering shoot phosphorus status as a promising route for breeding rice cultivars with reduced grain arsenic. Speciation of grain arsenic, performed using HPLC-ICP-MS, identified inorganic arsenic as the dominant arsenic species present in the rice grain.  相似文献

A field survey was conducted to identify potential Zn accumulators from an artisanal Zn smelting area in southwest China’s Guizhou Province. Hydroponic and soil culture experiments were performed to investigate the accumulation ability of Zn in Corydalis davidii. Zn concentrations in roots, stems and leaves of C. davidii in the smelting site were 1.1-3.5, 1.2-11.2, and 3.3-14 mg g1, respectively, whereas Zn concentrations in roots, stems and leaves of C. davidii in the contaminated site impacted by the Zn smelting were 1.0-2.4, 1.9-6.5, and 3.0-1.1 mg g−1, respectively. Zn concentrations in leaves and stems of C. davidii were observed at above 10 mg g−1 that refers to the threshold of Zn hyperaccumulator. The concentration distribution of Zn in C. davidii was leaf > stem > root, and the Zn bioaccumulation factors of C. davidii were above 1. It is concluded that C. davidii has high tolerance to concentrate Zn stress, and that C. davidii is a newly discovered Zn-hyperaccumulator with high biomass in the aboveground parts. Based on the cultivation experiments, C. davidii could reduce Zn concentration by 26.6, 21.2, and 10.2 mg kg−1yr−1 by phytoextraction from the smelting slag, Zn-contaminated soil, and background soil, respectively.  相似文献

Separate 77-d fish feeding studies were conducted on the cyclic volatile methylsiloxane (cVMS) chemicals octamethylcyclotetrasiloxane and decamethylcyclopentasiloxane with the rainbow trout, Oncorhynchus mykiss, with the determination of biomagnification factor (BMF) and lipid-adjusted BMF (BMF(L)) values as the final experimental metrics. The studies used fish food concentrations of ∼500 μg g−1 for exposure periods of 35 d, followed by a depuration period of 42 d with clean food. The fish tissue concentrations of D4 and D5 achieved empirical steady-state by day 21 in each study. By day 7 of exposure, total 14C activity of both compounds had moved from the fish gastrointestinal (GI) tract into surrounding tissue. An absence of significant fish growth during the initial depuration phase allowed for measurement of empirical depuration rate constants (k2) independent of growth dilution for D4 and D5 of 0.035 and 0.040 d−1, respectively, corresponding to elimination half-lives of approximately 20 d. These rate constants indicated that ∼70–75% of steady-state was achieved during exposure in both studies, resulting in empirical steady-state BMF and BMF(L) values of 0.28 and 0.66 for D4, respectively, and 0.32 and 0.85 for D5, respectively. Kinetic modeling using simple first-order uptake and depuration dynamics produced good agreement with experimental data, with D4 and D5 assimilation efficiencies of 40% and 44%, respectively. Growth-corrected depuration rate constants modeled over the entire study data set indicated slower elimination kinetics for D4 (k2 of 0.007 d−1 or half-life of 100 d) compared to D5 (k2 of 0.010 d−1 or elimination half-life of 69 d). Kinetic BMFk values (i.e., k1/k2) for D4 and D5 were 1.7 and 1.3, respectively, with lipid-adjusted BMFk(L) values of 4.0 and 3.4, respectively.  相似文献

Evaluation of aqueous uranium (U) uptake and depuration in larvae of the midge Chironomus tentans were investigated in two separated experiments. First, a static-renewal experiment was performed with 10-d old C. tentans larvae exposed to 300 μg U/L. The animals steadily accumulated U (Ku = 20.3) approaching steady-state conditions (BAF = 56) in approximately 9-11 d. However, accumulated U was readily depurated (Kd = 0.36) with U tissue concentration decreasing rapidly within 3 d of the larvae being placed in clean water (t1/2 = 1.9 d). Also, the growth of C. tentans larvae appeared to decrease after 6-11 d of U exposure, probably due to the reallocation of resources into U detoxification mechanisms. However, growth significantly increased once C. tentans were transferred to clean water. A separate short-term experiment was performed to evaluate the possible mechanism of U uptake in this invertebrate. Results suggested a passive mechanism of U uptake coupled with an active mechanism of U depuration but no details related to the type of mechanisms or pathway was investigated.  相似文献

A field survey and greenhouse experiments were conducted using Physalis alkekengi L. to investigate strategies of phytoremediation. In addition, ZnO nanoparticles were synthesized using P. alkekengi. P. alkekengi plants grew healthily at Zn levels from 50 to 5000 mg kg−1 in soils. The plants incorporated Zn into their aerial parts (with mean dry weight values of 235-10,980 mg kg−1) and accumulated biomass (with a mean dry weight of 25.7 g plant−1) during 12 weeks. The synthesized ZnO nanoparticles showed a polydisperse behavior and had a mean size of 72.5 nm. The results indicate that P. alkekengi could be used for the remediation of zinc-contaminated soils. Moreover, the synthetic method of synthesizing ZnO nanoparticles from Zn hyperaccumulator plants constitutes a new insight into the recycling of metals in plant sources.  相似文献
