A comparison of biodegradation efficiencies was done for volatile benzene, toluene, ethylbenzene, m-xylene, p-xylene, and o-xylene elimination in a compost biofilter. The column was first exposed to a synthetic mixture and then a free phase product mixture containing these compounds at increasing pollutant loads. The optimal moisture content of the system was determined, and this was used in the biodegradation experiments. An acclimated culture was used as an inoculum for the biofilter, the matrix of which consisted of composted forestry products, composted sewage sludge, lime, and perlite. Inlet and outlet concentrations were measured, and pollutant loads, elimination capacities, and removal efficiencies were determined for each of the compounds. Optimal moisture content for this system was found to be 40%, and the short lag times (one to five days) in acclimating to the compounds was ascribed to the presence of the well-acclimated inoculum. The compounds in the synthetic mixture had higher removal efficiencies (80-99%) even at the higher pollutant loads experienced, with the exception of o-xylene. Dynamic removal efficiencies and acclimation periods were seen in the free phase product mixture, with a removal efficiency range from 70 to 95%. 针对降解实验中接种物采集困难、活性差异大、实验可重复性及结果可比性差的问题展开接种物标准化研究。对多来源的接种物在实验室可控条件下进行驯养,驯养后生长状态良好的活性污泥分别固定于灭菌麸皮和藻酸钙凝胶等2种微生物载体上,形成2种固定化接种物(分别简称为Bran及Ball)。对上述固定化接种物进行微生物活性、适应性、降解能力以及稳定性研究:通过2种固定化接种物与商品化接种物Polyseed对葡萄糖-谷氨酸(GGA)五日生化需氧量(BOD5)的结果与活菌细胞数的关系,确认3种接种物在降解实验中的用量可控制在相同数量级;比较不同批次及保存时间的2种固定化接种物、相同批次不同保存时间的Polyseed以及污水处理厂曝气池污泥为接种物对苯胺及二甘醇的快速生物降解实验(OECD301F)结果,发现Ball及Bran较Polyseed及新鲜采集活性污泥的微生物适应性及降解能力更佳。此外,在较长保存期内2种固定化接种物活性相对稳定。2种固定化接种物初具成为标准化接种物的必要条件。  相似文献

Addition of pesticide-primed soil containing adapted pesticide degrading bacteria to the biofilter matrix of on farm biopurification systems (BPS) which treat pesticide contaminated wastewater, has been recommended, in order to ensure rapid establishment of a pesticide degrading microbial community in BPS. However, uncertainties exist about the minimal soil inoculum density needed for successful bioaugmentation of BPS. Therefore, in this study, BPS microcosm experiments were initiated with different linuron primed soil inoculum densities ranging from 0.5 to 50 vol.% and the evolution of the linuron mineralization capacity in the microcosms was monitored during feeding with linuron. Successful establishment of a linuron mineralization community in the BPS microcosms was achieved with all inoculum densities including the 0.5 vol.% density with only minor differences in the time needed to acquire maximum degradation capacity. Moreover, once established, the robustness of the linuron degrading microbial community towards expected stress situations proved to be independent of the initial inoculum density. This study shows that pesticide-primed soil inoculum densities as low as 0.5 vol.% can be used for bioaugmentation of a BPS matrix and further supports the use of BPS for treatment of pesticide-contaminated wastewater at farmyards.  相似文献

The distribution between sea water and sediment of a polio virus has been studied under an experimental system. The possibility to improve atrazine degradation in soils by bioaugmentation was studied. The atrazine-mineralizing strain, Chelatobacter heintzii Cit1, was inoculated in four sterile and four non-sterile soils, at varying inoculum densities. Two soils, which had shown enhanced atrazine mineralization, were used to determine which inoculum density was capable of restoring their original mineralizing capacity after sterilization. The two other soils, with intermediate and low capacity to mineralize atrazine, were used in order to demonstrate that atrazine mineralization in such soils could be improved by inoculation. Mineralization kinetics were fitted using the Gompertz model. In the case of soils adapted to atrazine mineralization, inoculation of C. heintzii did not accelerate the rate of atrazine mineralization, which was essentially performed by the indigenous microflora. However, with soils that did not mineralize atrazine, the introduction of 10(4) cfug(-1) resulted in a 3-fold increase of atrazine mineralization capacity.  相似文献

针对免水生物降解型生态厕所专用的进口菌种成本偏高、不能长期高效运行等现状,从土壤中分离筛选得到6株降解菌并将其培养成1组复合菌,然后分别进行降解实验。结果表明,7组实验菌对人体排泄物降解效果良好,其中复合菌种更好,COD降解率达到59%。对实验菌种进行初步鉴定,显示6株菌分别是粪肠球菌、木糖葡萄球菌、蜡状芽孢杆菌、产氨棒杆菌、地衣芽孢杆菌和活动肉杆菌。优选的专用菌剂的制备及保存方法效果良好,保存半年后菌剂仍有较高活性。  相似文献

A statistical correlative study (simple and multiple) between 14 herbicides and insecticides and composition of 51 soil samples has been performed by considering top- and sub-soil horizons, separately. The effects of Al on Panicum virgatum (switchgrass), a widespread perennial grass, were determined in relation to factors which might interact with Al in the soil. Plants were grown for 8 weeks in sand culture and were treated with 3 Al levels (0.5, 2.0, 5.0 mM), 2 P levels (0.065, 0.161 mM), 2 inoculum types (vesicular-arbuscular mycorrhizal (VAM) inoculum or VAM-free soil inoculum) and 2 inoculum sources (a high Al forest in NY or a low Al forest in Ohio) in a factorial design. Plant growth decreased with increasing Al and increased with increasing P, but the Al effect was less at high P than low P. VAM-inoculated plants outgrew non-VAM plants, especially at low and medium Al levels. Total P and Ca uptake decreased with increasing Al concentration, especially at low P levels. VAM inoculation did not result in increased P uptake at any Al level though VAM plants took up significantly more Ca than non-VAM plants at any Al level. VAM plants had lower tissue Al concentrations and took up less Al than non-VAM plants; Al uptake increased with increasing soil Al in non-VAM plants but not in VAM plants. Plants given inoculum from the high Al site had significantly lower tissue Al than plants given the low Al site inoculum, regardless of VAM status. We conclude that the presence of a VAM infection, moderate levels of soil P, and the source of the inoculum can reduce the effects of soluble Al. We discuss potential physiological and edaphic mechanisms by which Al may be immobilized and Ca availability increased in the presence of VAM fungi and other soil microflora.  相似文献

Trichosporon mycotoxinivorans XPY-10是一株分离自抗生素制药厂的高效四环素降解酵母菌。为了建立该菌株降解四环素的适宜条件,分别研究了碳源、有机氮源、金属离子等营养物质及初始底物浓度、接种量、pH、温度、装液量、摇床转速等理化因素对菌体生长及四环素降解效率的影响。结果表明,菌株XPY-10生长的最适碳源和氮源分别为蔗糖和蛋白胨。在含有0.05%FeSO4的培养基中,菌株XPY—10降解四环素的适宜条件为:接种量2%,pH8,温度3422,装液量100mL(250mL三角瓶),摇床转速180r/min。在此条件下,菌株XPY—10在7d内对初始浓度为600mg/L的四环素降解率为83.63%。本菌株对养殖废水及制药废水中四环素的污染治理有一定的应用前景。  相似文献

阿特拉津是长残留除草剂,其环境行为和生物修复技术已成为有机污染控制领域的研究热点。以廉价的高岭土、凹凸棒土和腐殖酸为载体材料,采用正交实验,把功能菌存活率作为目标性状,参考材料成球率,筛选出性能较好的高岭土、凹凸棒土和腐殖酸质量配比3种,分别为1∶0.5∶0.5(A3B2C1)、0.5∶0∶0.5(A2B3C1)和1∶0∶1(A3B1C2);在温度和紫外线耐受力实验中,A3B2C1材料配比更能够有效提升功能菌在高温和紫外线作用下的存活率,即载体中高岭土、凹凸棒土和腐殖酸质量比为1∶0.5∶0.5时效果最佳;利用A3B2C1载体材料配比制备菌剂,进行室内土壤修复实验,35 d时0.1%和0.5%载体菌剂添加量修复土壤中阿特拉津完全降解,而2个游离菌修复土壤中残留率均16%,0.1%载体菌剂添加量修复过程中土壤微生物Shannon多样性指数和均匀度变化幅度较其他修复方式小,有利于土壤微生物生态系统的平衡,因此0.1%载体菌剂添加量修复效果为最优。  相似文献

This work represents the first step to set up a toxicity testing procedure and to evaluate the sensitivity of the test microorganism to several classes of environmental pollutants. First, three different techniques were employed to standardise the microbial inoculum, then two different toxicity assessment protocols have been compared: Microtox and a dehydrogenase (DHase) activity inhibition test. The main goal was the optimisation of a microbial bioassay based on the dehydrogenase activity (DHase) inhibition in Pseudomonas fluorescens bacterial strain ATCC 13525. Triphenyl tetrazolium chloride (TTC) was used as electron acceptor and its reduction produces Triphenyl formazane (TPF). The P. fluorescens DHase inhibition bioassay was investigated for being a reliable and rapid method for assessing toxicity. The optimisation of the operating conditions resulted in a repeatable bioassay. Then, P. fluorescens and Vibrio fischeri sensitivity were firstly compared by testing Zn++, one of the reference compounds for Microtox test. In addition, other compounds (Ni++, Cd++, Cu++, phenol) were also tested with both bioassays. A high statistical significance of data was obtained with the logistic curve. The present work has demonstrated that P. fluorescens is as sensitive as Microtox culture (V. fischeri), for some of the metal ions. With reference to organic compounds, the lower sensitivity of P. fluorescens to phenol makes its use difficult in organic polluted samples.  相似文献

Wastewater nutrient removal by Chlorella pyrenoidosa and Scenedesmus sp   总被引:4,自引:0,他引:4  
Two different species of photosynthetic microalgae, Chlorella pyrenoidosa and Scenedesmus sp., were grown in settled and activated sewage filtrates at two different inoculum sizes, aimed to reduce nutrient load from wastewater. Higher growth rates were recorded in cultures with higher inoculum size, and algal cells usually grew better in settled sewage than in activated sewage. As algae started to grow and multiply, both nitrogen and phosphorus content in wastewater decreased significantly. The removal rate was rapid during the first week of growth and more than 2/3 of wastewater N and P was reduced. After the initial period, the removal rate slowed down. At the end of this study, more than 80% of total-P and inorganic N present in settled sewage were reduced but such removal efficiency was lower in activated sewage. In general, high inoculum size of algal cells provided more N and P removal than low inoculum, and Chlorella cells performed better than Scenedesmus. These results suggest that cultivation of Chlorella seems to be one of the feasible methods to reduce the amount of nitrogen and phosphorus entering the nearby coastal water, thus preventing the eutrophication problem. It is also clear that algal ponds with high inoculum size might be more suitable to be installed as a secondary rather than a tertiary treatment process.  相似文献

To better construct a bioaugmented system for tobacco wastewater treatment, activated sludge was inoculated with different concentrations of the nicotine-degrading bacterium Pseudomonas sp. HF-1. The results showed that inoculum concentrations of 0.55?±?0.01 and 1.10?±?0.03 mg/g (dry weight of strain HF-1/dry weight of activated sludge) were best to ensure strain HF-1 survival and successful bioaugmentation. The release pattern of autoinducer (AI) for quorum sensing in the bioaugmented system was also investigated. During the period of HF-1 inoculation, compared with failed bioaugmented systems, AI-2 was significantly increased in the successful systems, suggesting that AI-2-mediated bacterial communication played an important role in the colonization of HF-1. When inoculation of strain HF-1 was stopped, the amount of AI-2 decreased and leveled out in all systems. Notably, there was a greater than threefold increase of short-chain AHLs in failed bioaugmented systems, but no increase in successful ones, implying that the fluctuation of short-chain AHLs could be an indicator of the failure of bioaugmentation. Thus, AI-2-mediated quorum sensing could be implemented to facilitate HF-1 colonization.  相似文献

分别以厌氧污泥、脱氮硫杆菌菌悬液和厌氧污泥并添加脱氮硫杆菌菌悬液为接种物,以硫化物和硝酸盐为进水基质,考察不同接种物条件下,各反应器的硫化物氧化特性、反硝化特性、生化反应机理及微生物特性。结果表明,在无菌条件下,硫化物不能被硝酸盐化学氧化。接种脱氮硫杆菌菌悬液的2#反应器的硫氧化速率为1.98 g S/(m3.h),停留24 h硫化物的去除率高达97%,脱硫能力最强,该接种条件下以硝酸盐氧化硫化物为主反应,优势菌为杆菌,进水的NO3--N/S应控制在0.4以下,可以实现高效生物脱硫。接种厌氧污泥的1#和3#反应器的脱氮效果比2#反应器好,停留时间为24 h时,硝酸盐的平均去除率为96%。单独接种厌氧污泥的1#反应器的硫氧化速率为1.78 g S/(m3.h),其优势菌为球菌,该接种条件下以硝酸盐氧化硫化物和硝酸盐氧化单质硫为主反应,进水的NO3--N/S应控制在0.8左右。以厌氧污泥联合脱氮硫杆菌为接种物时,硫氧化速率为1.71 g S/(m3.h),反应器以硝酸盐氧化硫化物、硝酸盐氧化单质硫以及异养反硝化为主反应,驯化后优势菌为球形、卵圆形和短杆状,应控制进水NO3--N/S为1.2,可以实现同步脱硫反硝化,该工艺既可以用于含硫废水的处理,也可以用于C/N低的硝酸盐废水的处理。  相似文献

In industrial effluents, the presence of an infinite number of possible mixtures of substances and the high variability of chemical conditions ask for an evaluation of biodegradability by a global and simple method. Biological oxygen demand after five days (BOD5) using synthetic wastewater was studied by two different ways: dilution and manometric methods. It can therefore be established that BOD5 obtained by adding manufactured inocula to the synthetic medium (effluent containing known and easily biodegradable substances) is close to the values obtained with inocula taken from the treated effluent of an urban and a rural purification plant. It was found that BOD5 measurement of effluents presenting factors affecting biodegradation, similar of those found in industrial effluents, is very questionable. The BOD is in this case influenced by the synergic and antagonist interactions between numerous and variable parameters like as pH, nature and concentration of inoculum, concentration of nutriments, amount and nature of assimilable substances, presence of toxicants, and presence of nitrification inhibitors, which are typical of real industrial wastewaters.  相似文献

The use of activated sludge as inoculum source in ready biodegradability tests (RBT) suffers from several drawbacks related to the heterogeneity of these communities. In this work, the ability of a 7-day aeration period in a mineral medium to homogenize the characteristics of various activated sludges, as suggested by some RBT, was studied. The biodegradation potential of three activated sludge supernatants obtained from different wastewater treatment plants was assessed in terms of cultivable cell density, dehydrogenasic activity and a profile of hydrolytic enzymes. After the preconditioning, the homogenization of these characteristics in the supernatants was observed, as well as a decrease. When preconditioned inocula were used in acetate RBT, the biodegradation kinetics were homogenized. However, some preconditioned supernatants lost their ability to degrade an easily-assimilable xenobiotic compound (aniline) during the observation period, showing the effect of inoculum preconditioning on the behavior of complex bacterial communities, specialist populations (e.g. aniline degraders) being more sensitive than generalist populations (e.g. acetate degraders). These results show that preconditioning cannot be an optional inoculum pretreatment in RBT, and emphasize the importance of further studies focusing on inoculum homogenization.  相似文献

The relative abundance of the denitrifier, Pseudomonas sp. JR12, was examined in an alginate-based entrapment complex under non-sterile, denitrifying conditions. Immuno-labeling of the Pseudomonas inoculant followed by flow cytometry (FCM) was used for determination of the relative abundance of this bacterium under the various incubation conditions. Additional information on the relative abundance of the inoculant was obtained by a quantitative enzyme-linked immunosorbent assay (ELISA) and results obtained by FCM and ELISA were compared. Ambient nitrate levels controlled the successful, long-term proliferation of the inoculant. At low ambient nitrate levels, Pseudomonas sp. remained the dominant microorganism during incubation. Higher ambient nitrate concentrations, attained by either decreasing the inoculum size of Pseudomonas sp. or raising inlet nitrate concentrations of the medium supplied to the incubation vessels, resulted in a gradual shift toward other, nitrite-accumulating denitrifiers. Thus far, most studies on the use of entrapped microorganisms for bioremediation purposes have been conducted under controlled laboratory conditions. Based on this study, conducted under non-sterile laboratory conditions, it is concluded that in-situ bioremediation using entrapped target microorganisms is bound to fail without a proper understanding of the factors that cause the target microorganism to out-compete undesired microbial invaders. Furthermore, based on the close agreement between the two detection methods used, it is concluded that flow cytometry provides a rapid and accurate tool for the detection of the relative abundance of immuno-labeled target organisms in heterogeneous microbial populations.  相似文献

Four aerobic species of mesophilic bacteria which catabolise phenol as the sole carbon source were isolated from landfill waste, and identified as Acinetobacter, Arthrobacter, Micrococcus and Nocardia spp. In vitro studies in defined medium revealed variations in abilities of the isolates to catabolise phenol, and in pH growth optima ranging from 6.8 to 7.6. The pH of culture media decreased with growth, and phenol catabolism was impeded below pH 5.4. Phenol catabolism in landfill leachate occurred after longer lag phases than in defined medium, and relative efficiencies of phenol catabolism did not correspond with the patterns in defined medium. Inoculum size affected phenol removal rates. The results describe factors which may be manipulated to prevent pollution problems in the codisposal of phenolic industrial waste.  相似文献
