Detailed multispecies studies on the patterns of genetic variability and differentiation in marine environments are still rare. Using mitochondrial and nuclear genetic markers, we compared genetic variability and population structuring of threespine (Gasterosteus aculeatus) and ninespine (Pungitius pungitius) sticklebacks from the same eleven marine and six freshwater locations within the Baltic Sea basin. Analyses of both marker types revealed a significantly lower degree of genetic structuring in both marine and freshwater populations of threespine than those ninespine sticklebacks. Isolation-by-distance (IBD) was detected across the marine populations in both species, suggesting spatially limited gene flow. However, the levels of genetic diversity and differentiation across the localities were uncorrelated between the two species in both marine and freshwater environments. Accordingly, estimates of effective population sizes were larger and migration rates were higher for three- than for ninespine sticklebacks. Hence, ninespine stickleback populations from the Baltic Sea basin appear to be subject to stronger genetic drift than sympatric threespine sticklebacks, and the proximate reason for this difference is likely to be found from autecological differences between the two species. In accordance with the earlier studies, genetic variability was higher and the degree of genetic differentiation was lower in marine than in freshwater populations in both species.  相似文献

Extracts of 39 species of Caribbean gorgonians were tested for antimicrobial activity against 15 strains of marine bacteria. The bacteria consisted of three opportunistic pathogens, Vibrio parahaemolyticus, Leucothrix mucor, and Aerococcus viridans, and 12 strains isolated from either healthy or decayed gorgonians. Overall, only 15% (79 out of 544) of the tests resulted in antibacterial activity with 33% (13 out of 39) of the gorgonians inhibiting only one bacterial strain and 23% (9 out of 39) showing no activity. The extracts of four Pseudopterogorgia species showed relatively high levels of activity, inhibiting 43 to 86% of the bacterial strains. The potency of the active Pseudopterogorgia species was variable, however, and three additional Pseudopterogorgia species were inactive against all bacterial strains. With the exception of one sensitive strain, Vibrio species were resistant to gorgonian metabolites. Our results indicate that organic extracts of most Caribbean gorgonians do not possess potent, broad-spectrum antibacterial activity inhibitory to the growth of opportunistic marine pathogens and bacteria associated with healthy and decayed gorgonian surfaces. These findings suggest that the inhibition of bacterial growth is not the primary ecological function of gorgonian secondary metabolites and that bacteria may not be important selective agents in the evolution of gorgonian secondary chemistry.  相似文献

Allozyme electrophoresis of two corals was used to assess whether populations at Ningaloo Reef, Western Australia are primarily self-seeding or whether recruitment is from a broader geographic pool. Significant genetic subdivision across a range of spatial scales (between 6.5 km and 155 km) was found for both Acropora digitifera and A. aspera, with mean F ST values of 0.010 and 0.067 respectively. Large departures from Hardy-Weinberg expectations were found for both species. Without exception these were due to deficits of heterozygotes; mean D values were –0.341 for A. digitifera and –0.455 for A. aspera. The magnitude of the deficits was consistent both across loci for all sites and across all sites for each locus. Some loci were found to be in linkage disequilibrium but no consistent pattern was observed. Also, multi-locus genotypic diversity values were generally high (between 0.83 and 1.00) and so departures from equilibria cannot be attributed to asexual reproduction. The most plausible explanation for the patterns observed is restricted gene flow at both the planktonic and gametic stages, with mating between close relatives.Communicated by G.F. An on-going, Caribbean-wide epizootic affecting sea fan corals (Gorgonia spp.) is caused by the fungus Aspergillus sydowii (Thom et Church). We examined the role of crude extracts in resistance of two species of sea fans, Gorgonia ventalina (L.) and G. flabellum (L.), against A. sydowii and a bacterial pathogen of fish, Listonella anguillarum (MacDonell et Colwell). Sea fans were collected in January 1997 from San Salvador, Bahamas, and in June 1997 and January 1998 from Alligator Reef, Florida Keys, USA. Crude extracts from both species were tested to determine concentrations inhibiting germination of A. sydowii spores. Crude extracts from both species inhibited spore germination at concentrations as low as 1.5 mg ml−1; most samples were active at 5 to 10 mg ml−1. These concentrations are within the range estimated in living tissue and were higher in healthy colonies suggesting their role in mediating disease susceptibility. We also detected within-colony gradients in antifungal activity, which varied with the disease state of the colony. In healthy sea fans, resistance was highest at colony edges and lowest in medial and central regions of the colony. Among sea fans with lesions in the colony center, resistance in tissue from proximal and medial regions was as high as tissue from the colony edge (i.e. distal region). The increase in antifungal activity suggests an inducible response by the coral host to the fungal pathogen. This response is most evident among sea fans with lesions in the colony center and not among colonies with lesions at the edge. Antibacterial activity of crude extracts against L. anguillarum was highest at the colony edge but did not vary with disease state or tissue location. Three procedures for the extraction of coral lipids were compared and a rapid and effective method for future use is suggested. This method was used to measure the lipid content of the branching coral Porites porites (Pallas) and the massive corals Montastrea annularis (Ellis and Solander) and Siderastrea siderea (Ellis and Solander) during July and August 1991. P. porites and M. annularis were sampled on two fringing reefs, each characterised by different water quality affecting light transmission, and at depths of down to 30 m on a barrier reef on the west coast of the island. m. annularis contained, on average, 29% of dry weight as lipid, and there were no significant differences in lipid levels between corals sampled on either fringing reef at 3 and 6 m, or between fringing reefs and the barrier reef at 13, 20 or 30 m depth. Five samples were also taken from a single massive colony of S. siderea at 3 m on a fringing reef and at 13, 20 and 30 m depth on the barrier reef. Values for lipid ranged from 26 to 35% of dry tissue weight. P. porites from 3 and 6 m depth on both fringing reefs contained the same amount of lipid (11% of dry tissue weight). However, at 13 m depth on the barrier reef this coral contained significantly less lipid (8.5% of dry tissue weight). This difference may be attributable to a higher nutritional intake by P. porites on the fringing reefs.  相似文献

The recent intensification of human disturbances in the Caribbean has increased the prevalence of partial mortality on coral colonies. Partial mortality can change colony size by directly shrinking colonies or by splitting colonies into fragments. A reduction in colony size can also adversely affect fecundity and fitness as internal resources shift away from reproduction toward colony maintenance. This study aimed to determine whether three Caribbean coral species, Siderastrea siderea, Montastraea faveolata, and Diploria strigosa, along the reef tract in Puerto Morelos, Mexico (20o52′N, 86o51′W), continued to dedicate resources to reproduction when colonies were fragmented to pre-maturation size. Contrary to expectations, eggs were found in colonies that were smaller than the maturation size and had been subjected to partial mortality. The continued dedication of resources toward reproduction, even in the smallest colonies, suggests that resource trade-offs away from reproduction are not as rigid as previously suggested in stressed corals.  相似文献

The Caribbean reef-building corals Porites porites (Pallas) and Montastrea annularis (Ellis and Solander) and the Red Sea corals Pocillopora verrucosa (Ellis and Solander), Stylophora pistillata (Esper) and Goniastrea retiformis (Lamark) were analysed for total lipid, wax ester and triglyceride content, and fatty acid composition. M. annularis contained about 32% of dry weight as total lipid, whereas much lower values of between 11 and 17% were recorded for the other four species. It is concluded that there is greater variation in coral total lipid than hitherto thought. The total lipid contained a substantial proportion of wax ester (22 to 49%) and triglyceride (18 to 37%). The storage lipids (wax esters and triglycerides) accounted for between 6 and 20% of the dry weight and between 46 and 73% of the total lipid. Variation in lipid content between species could not be attributed to geographical location, but the low values for total lipid in Red Sea corals may in part be due to environmental factors as these samples were collected in winter. All corals analysed contained high levels of saturated fatty acids, the most abundant fatty acids being 16:0, 18:0 and 18:1n-9. Marked differences were observed in polyunsaturated fatty acid (PUFA) content between species, with comparatively low levels of 10 and 11% of fatty acids being recorded in M. annularis and G. retiformis, respectively. The values for the other species ranged between 21 and 37%. Fatty acid composition may vary according to the proportions of fatty acids obtained from diet, algal photosynthesis and synthesis by the animal tissues.  相似文献

Larval dispersal is a critical component of marine species’ life histories because it controls their population dynamics. Dispersal distance can be inferred by the presence and scale of spatial genetic structure (SGS). The larvae of Antillogorgia elisabethae, a surface brooding Caribbean octocoral, have been observed to settle within meters of maternal colonies, although many disperse over greater distances. Using a spatially hierarchical sampling design, A. elisabethae recruits were collected from a site off the coast of Great Abaco, The Bahamas, in June of 2009–2011. Seven microsatellite loci were used to determine whether genetic structure was present among recruits on scales of <1 m to 10s of meters. Multilocus genotypes were autocorrelated within spatial distance classes using Moran’s I coefficient as an estimate of genetic similarity, and Rousset’s genetic distance (â). Genetic structure was present on a scale of <10 m in 2010, but not in the other 2 years, suggesting that larval dispersal was more localized in 2010. Moran’s I coefficients were positive and significant for 0–1, 1–2 and 2–5 m classes in 2010, and the average genetic distance (â) among recruits in the 0–1 m class was significantly lower than the other classes. Autocorrelograms suggested that genetic patch sizes were 7.5 m in 2010, and 20 m in 2009 and 2011. Differences in the SGS of recruits between years might reflect temporal variation in temperature, wind and/or current speeds affecting larval dispersal, or variation in reproduction at the site.  相似文献

Very little information exists on the effects of nitrate on corals, although this is the major form in which nitrogen is prescrit in tropical eutrophie coastal waters. In this study we incubated nubbins ofPorites porites and explants ofMontastrea annularis in laboratory photostats illuminated by halide lamps, with concentrations of nitrate of 0, 1, 5 and 20 M, for 40 and 30 d, respectively, At the end of this period it was found that the population density of the zooxanthellae had increased significantly with increased nitrate concentration, suggesting nitrogen limitation of the growth rate of zooxanthellae in the control group. There were also significant increases in the amount of chlorophylla ande 2 per algal cell, in the volume of the algal cells, and in the protein per cell. Overall, the protein per unit surface increased, but this was attributable solely to increased algal protein: there was no significant change in host protein. Maximum gross photosynthesis normalized to surface area was enhanced by nitrate addition, reflecting the increase in algal population density. There was no change when normalized on a per cell basis. Respiration rate normalized to protein content was decreased by nitrate. The most dramatic change was in the rate of skeletogenesis, which decreased by in both species when exposed to nitrate enrichment. A model is presented which suggests that the diffusion-limited supply of CO2 from surrounding seawater is used preferentially by the enlarged zooxanthellae population for Photosynthesis, thereby reducing the availability of inorganic carbon for calcification. It is concluded that enhanced nitrate levels in tropical coastal waters will have a hitherto unrecognized effect on the growth rate of tropical coral reefs.  相似文献

Reproduction and development of the ophiuroidOphionereis olivacea H. L. Clark, a species that broods its young, were investigated in Florida from January through December 1985 and in Belize in June 1985 and April 1986.O. olivacea has a maximum disc diameter of 5.2 mm and is a protandric hermaphrodite. Sex reversal occurs at disc diameters ranging between 2.2 and 4.0 mm; it has 400 µm diameter oocytes and is ovoviviparous. As expected from an obligate out-crosser, this ophiuroid has a similar allocation to male and female reproduction, in terms of gonad volume. Reproduction of the Florida population was monitored for one year.O. olivacea is a sequential brooder and incubates its young during the coldest months of the year. The females produce a single clutch of juveniles each year. Spawning starts in November and peaks in January and February. The ova are spawned synchronously from all the ovaries of each female into the bursae and are retained there. Fertilization is accomplished by intake of sperm into the female bursae.O. olivacea develops through a ciliated embryonic stage that appears to be a modified vitellaria larva lacking ciliary bands. These modified larvae swim in seawater in the bursae. Direct development in this species has resulted from heterochrony in the loss of ophiopluteus features. The number of brooding females in the population increases in November and reaches a maximum in March. JuvenileO. olivacea emerge from the bursae at a disc diameter of 480 µm and each arm has three segments. Recruitment was first detected in April, and juveniles were particularly abundant in June. Small size inO. olivacea is associated with paedomorphic heterochrony with precocious maturation of the males at a disc diameter of 1.5 mm, ~2 mo after taking up their free existence. Brooding as a life history mode inO. olivacea is examined and the adaptations of this species to its warm-water habitats are discussed.  相似文献

Vermilion snapper (Rhomboplites aurorubens) were collected from four sites off the Atlantic coast of the USA and one site in the Gulf of Mexico to evaluate effective population size and genetic stock structure. Previous studies had suggested geographic variation in the ratio of males to females, so this population characteristic was explored in conjunction with the genetic analysis. Sex ratio varied greatly among the five sample sites; males comprised 57% of samples in the Gulf of Mexico, while within the South Atlantic Bight they comprised between 36% (Morehead City, North Carolina) and 53% (Carolina Beach, North Carolina) of samples. No clear geographic trends in the sex ratio emerged; instead, it was found to vary with fish length, the percentage of males decreasing with increasing size. Allelic variation assessed at seven dinucleotide microsatellite loci was large; gene diversities ranged from 0.43 to 0.95 and allelic counts from 7 to 39. Estimates of the effective population size ranged from 24 500 (based on the infinite-alleles model) to 150 500 (based on the stepwise-mutation model). There was evidence for excess homozygosity within samples: estimates of F IS (the correlation of alleles within individuals) ranged from 0.01 to 0.03 among the seven loci, and three estimates were significantly greater than zero. Differentiation among localities was very weak, as estimates of F ST (the correlation of alleles within populations) were on the order of 0.001 to 0.002 and genetic distance estimates between localities were not related to geographic distances. This suggested that vermilion snapper in the South Atlantic Bight (Cape Hatteras, North Carolina to Cape Canaveral, Florida) and Gulf of Mexico are likely to consist of one genetic stock. We developed quantitative PCR (qPCR) assays to distinguish each of the four clades (AD) of dinoflagellate endosymbionts (genus Symbiodinium) commonly found in Caribbean corals. We applied these primer sets, which target portions of the multi-copy ribosomal DNA (rDNA) gene family, to assess the presence/absence of symbionts in clade D (as indicated by the detection of clade D DNA). We detected these symbionts in five of six Caribbean host species/genera (21% of samples analyzed, N = 10 of 47 colonies), from which clade D had rarely or never been observed. This suggests that Symbiodinium in clade D are present in a higher diversity of coral species than previously thought. This qPCR-based approach can improve our understanding of the total microbial diversity associated with corals, particularly in hosts thought to be relatively specific, and has many other potential applications for studies of coral reef ecology and conservation.  相似文献

Individual specialisation is increasingly recognised to be an ecological and evolutionary process having important consequences for population dynamics of vertebrates. The South American fur seal (SAFS) and the South American sea lion (SASL) are two otariid species with similar ecology that coexist in sympatry in the Uruguayan coast. These two species have contrasting trends and widely different population sizes. The underlying reasons for these population trends, unique in their geographical ranges, remain unknown. We studied the foraging ecology of these otariid species over 2 years at the individual- and population levels using the isotopic ratios (δ13C–δ15N) in whiskers of both sexes. We compared the isotope ratios between species and sexes and used several metrics to characterise the degree of overlap and distinctiveness in the use of isotopic niche space at the individual- and population levels. Interspecific trophic niche overlap was minimal, thus ruling out interspecific competition as the cause for the contrasting population trends of both species. At the intraspecific level, both species had sexual segregation in their foraging areas, but each species had a large overlap in the isotopic niches between sexes. While SAFS had a wider niche and generalist individuals, SASL had the narrower niche with a higher degree of individual specialisation. Behavioural constraints during the breeding season, intraspecific competition and a major dependence on resources of the Uruguayan coastal shelf may explain why SASL had a higher trophic individual specialisation and a larger vulnerability in a heavily exploited habitat by fisheries and, by consequence, a locally declining population trend.  相似文献

Coral diseases have increased in frequency over the past few decades and have important influences on the structure and composition of coral reef communities. However, there is limited information on the etiologies of many coral diseases, and pathways through which coral diseases are acquired and transmitted are still in question. Furthermore, it is difficult to assess the impacts of disease on coral populations because outbreaks often co-occur with temperature-induced bleaching and anthropogenic stressors. We developed spatially explicit population models of coral disease and bleaching dynamics to quantify the impact of six common diseases on Florida Keys corals, including aspergillosis, dark spots, white band, white plague, white patch, and Caribbean yellow band. Models were fit to an 8-year data set of coral abundance, disease prevalence, and bleaching prevalence. Model selection was used to assess alternative pathways for disease transmission, and the influence of environmental stressors, including sea temperature and human population density, on disease prevalence and coral mortality. Classic disease transmission from contagious to susceptible colonies provided the best-fit model only for aspergillosis. For other diseases, external disease forcing, such as through a vector or directly from pathogens in the environment, provided the best fit to observed data. Estimates of disease reproductive ratio values (R0) were less than one for each disease, indicating coral colonies were below densities required for diseases to become established through contagious spread alone. Incidences of white band and white patch disease were associated with greater susceptibility or slower recovery of bleached colonies, and no disease outbreaks were associated with periods of elevated sea temperatures alone. Projections of best-fit models indicated that, atleast during the period of this study, disease and bleaching did not have substantial impacts on populations and impaired rates of population growth appeared to be attributable to other stressors. By applying epidemiological models to field data, our study gives qualitative insights into the dynamics of coral diseases, relative stressor impacts, and directions for future research.  相似文献
