The object of this study was to evaluate the acute toxicity of cadmium in different post-embryonic stages of the penaeid shrimp Penaeus japonicus (Bate, 1888) and to determine the effect of sublethal cadmium on the osmoregulatory capacity used as an indicator of physiological condition. Tolerance to cadmium increases with the developmental stage. The least tolerant stages are the nauplii (48 h LC50: 124 g Cd l-1) and the zoeae (96 h LC50: 10 to 30 g Cd l-1). The most tolerant stages are the postlarvae (96 h LC50: 200 to 3500 Cd l-1) and juveniles (96 h LC50: 5500 g Cd l-1). In juvenile shrimp, 2000 g Cd l-1 significantly reduce hypo- and hyper-OC. The effect of cadmium on hypo- and hyper-osmoregulatory capacity illustrates a dose- and time-dependent response. Surviving shrimp recover their hypo-osmoregulatory capacity after 6 d of readaptation in cadmium-free seawater.  相似文献

The toxicity of fenitrothion was determined in larvae (nauplii, Zoeae 1 to 3, Mysis 1 to 3), postlarvae (PL stages) and juvenile shrimp (Penaeus japonicus Bate), in two media, seawater (SW) and diluted seawater (DSW) (1100 and 550 mosM kg−1, ≃ 37 and 19‰ S). The effects of fenitrothion on the osmoregulatory capacities (OC) of juveniles were recorded. A gill and epipodite histopathological study was also conducted. For larvae in seawater, 24 and 48 h LC50s ranged from 32.9 μg l−1 (Zoeae 2) to 10.7 μg l−1 (Mysis 3), and from 3.9 μg l−1 (Zoeae 3) to 2.0 μg l−1 (Mysis 3), respectively; 48 and 96 h  LC50s in postlarvae (PL) at the same salinity ranged from 1.8 μg l−1 (PL1) to 0.6 μg l−1 (PL5), and from 0.3 μg l−1 (PL7) to 0.4 μg l−1 (PL15). In juveniles, 96 h LC50s were 0.8 μg l−1 in seawater and 1.5 μg l−1 in diluted seawater. From hatching to juvenile stages, the overall trend was a rapid decrease (from nauplii to PL5–PL7) followed by a slight increase (from PL7 to PL15 and juveniles) in the shrimp's ability to tolerate the insecticide. In juveniles kept in seawater and in diluted seawater, fenitrothion decreased the osmoregulatory capacity (OC = difference between the hemolymph osmotic pressure and the osmotic pressure of the medium) at both lethal and sublethal concentrations. This effect was time- and dose-dependent. In SW, the decrease in hypo-OC was ˜ 25% at sublethal concentrations and ˜ 35% at the 96 h LC50. In DSW, the decrease in hyper-OC was ˜ 10 to 15% at sublethal concentrations. In SW, shrimp were able to recover their OC in less than 48 h when transferred to water free of pesticide. In DSW, recovery at 48 h was only possible after exposure to the lowest tested sublethal concentration. Haemocytic congestions (thrombosis) of the gills, lamellae necrosis and other alterations of gills and epipodites (breakage of the cuticle, reduction of the hemolymph lacunae) were noted in juveniles exposed to lethal and sublethal concentrations of fenitrothion. Acute toxicity of ammonia was determined for cultured larval, postlarval, and wild adult lobsters (Homarus americanus) in 1988. Ammonia tolerance was found to increase with ontogenetic development. Based on 96-h LC50 values of 58 mg l–1 NH4 + + NH3 l–1 seawater (0.72 mg NH3 l–1) for Stage I larvae, 87 mg NH4 + + NH3 l–1 (1.7 mg NH3 l–1) for Stage II larvae, 125 mg NH4 + + NH3 l–1 (2.13 mg NH3) for Stage III larvae, 144 mg NH4 + + NH3 l–1 (2.36 mg NH3 l–1) for Stage IV postlarvae, 377 mg NH4 + + NH3 l–1 (5.12 mg NH3 l–1) for adult lobsters at 5°C and 219 mg NH4 + + NH3 l–1 (3.25 mg NH3 l–1) for adult lobsters at 20°C, recommendations for safe levels of total ammonia and un-ionized ammonia were calculated using an application factor of 0.1. Effects of ammonia on osmoregulatory capacity were studied on postlarvae and adults. Ability of postlarvae and adults to hyper-regulate in low-salinity media decreased after exposure to ammonia. In postlarval lobsters, osmoregulatory capacity was significantly affected in ammonia concentrations exceeding 32 mg l–1. Osmoregulatory capacity in adult lobsters (5 and 20°C) was affected at 150 mg l–1. In postlarval lobsters, a minimum exposure time of 12 h was required to impair osmoregulatory capacity. The decrease in hemolymph osmotic pressure was caused by lower hemolymph sodium concentrations. The presence of ammonia in the external medium could markedly affect the Na+/NH4 + transport mechanism by permanently, temporarily, or partially impairing the transport sites for sodium.  相似文献

Female kuruma prawns (Penaeus japonicus Bate) with undeveloped, early developing, developing, nearly ripe and ripe ovaries, were collected from Ise Bay, Japan, in 1984. Oocyte development of the kuruma prawn was classified into ten stages according to morphological characters, namely: (1) synapsis stage, (2) chromatin nucleolus stage, (3) early perinucleolus stage, (4) late perinucleolus stage, (5) oil globule Stage I, (6) oil globule Stage II, (7) yolkless stage, (8) yolk granule stage, (9) prematuration stage, and (10) maturation stage. The synapsis stage is a multiplication stage. The chromatin nucleolus stage, early and late perinucleolus stages are previtellogenesis and primary growth stages. Oil globule Stage I is an initial stage of primary vitellogenesis and secondary growth. Follicle cells on the oil globule Stage I oocytes expand rapidly and reach maximum size during oogenesis. Yolk granule stage oocytes are in the initial stages of secondary vitellogenesis. Strongly acidophilic yolk granules accumulate within basophilic vesicles of the cytoplasm. The yolk granules are first concentrated in the inner part of the cytoplasm, then gradually spread to the periphery. Cortical crypts, which are separated from the oocyte cytoplasm by the cytoplasmic membrane, are situated outside of oocyte cytoplasm. Germinal vesicle breakdown (GVBD) is initiated in the late phase of prematuration and continues until the late phase of maturation immediately prior to spawning. At the beginning of the maturation stage, the oocytes are ovulated, after which the nuclei further shrink and migrate out-wards. After ovulation, meiotic division of the ovarian oocyte progressed up to the metaphase of primary maturation division. Finally, the meiotic metaphase is visible just beneath the cytoplasmic membrane in the mature oocyte. Though ovulation is synchronous within the same ovary, GVBD is not completely synchronous. Ovulated mature oocytes have many club-shaped cortical crypts in the peripheral part of the cytoplasm and contain extensive accumulations of yolk granules dispersed throughout the cytoplasm. The apical end of the club-shaped cortical crypts and cytoplasmic membrane are coated by the vitellin envelope in the mature oocyte.  相似文献

Individuals of the oriental brown shrimp Penaeus japonicus Bate, were raised separately (1 shrimp per rearing cage) with surplus food, in almost dark, non-sediment conditions. Results were obtained regarding growth of each part of the body in conjunction with exuviation and molting cycle. Increases in carapace, body length and body weight conformed to the general pattern discribed by Hiatt (1948), with no difference in growth resulting from sex, or inflexion point in juvenile stage. The molting cycle of shrimp weighing 1.5 to 15 g was 6 to 17 days at a water temperature of 20° to 28°C. The molting cycle was prolonged in proportion to the size of the shrimp; shortened as the water temperature increased.  相似文献

Fatty acid biosynthesis in the larval stages of Penaeus japonicus Bate was examined by feeding microencapsulated diets containing (1-14C) palmitic acid, and fat-free diets supplemented with defined fatty acids. Highest larval growth rates were achieved on diets containing Tapes philippinarum lipid and, when defined fatty acids were substituted, on diets containing 20:63 fatty acid. The radioactive tracer experiments indicate that 16:17, 18:0 and 18:19 fatty acids may be synthesised from palmitic acid and that P. japonicus larvae may possess the ability to elongate 18:33 to 20:53 and 22:63, and 18:26 to 20:46. However, the rates of these reactions appear to be too slow to meet the larval requirements for essential fatty acids and the 3 series of polyunsaturated fatty acids must be provided in the diet.  相似文献

The results of toxicity experiments have shown that a level of 17 ppb ionic mercury is acutely toxic (96-h LC50) to postlarval white shrimp (Penaeus setiferus). In addition, the size of the shrimp, within the range tested (7 to 35 mm) did not significantly alter the toxicity values of mercury to this species. Fifty-seven days pre-exposure to low levels of mercury (0.5 and 1.0 ppb) had no effect on the acute toxicity (96-h LC50) of mercury to the shrimp. The effect of chronic exposure to low levels of mercury on the respiratory rate, growth, and molting rate of postlarval P. setiferus was studied, and the results showed that 60 days exposure to 0.5 and 1.0 ppb mercury did not significantly affect any of these parameters for the postlarvae.  相似文献

Four species of microalgae (Chaetoceros muelleri, Tetraselmis suecica, Tahitian Isochrysis sp. (T-iso) and Dunaliella tertiolecta) with distinctly different fatty acid profiles were grown in continuous culture and fed to prawn larvae (Penaeus japonicus, P. semisulcatus and P. monodon) as monospecific diets. The best two diets (C. muelleri and T. suecica) were also fed as a mixed diet. Experiments were run until the larvae fed the control diet of C. muelleri metamorphosed to Mysis 1. The survival and development (i.e. performance) of the larvae were affected by algal diet, and the diets were ranked in the order of decreasing nutritional value: C. muelleri ≥ T. suecica > T-iso > D. tertiolecta. Larvae fed a mixed diet of C. muelleri and T. suecica (2:3 by dry weight) performed as well or better than those fed C. muelleri, and the performance of both these groups of larvae was better than those fed T. suecica. The lipid and carbohydrate compositions of the algae had little or no effect on the lipid and carbohydrate compositions of the larvae or their performance. However, the larvae that performed best (i.e. those fed C. muelleri) had significantly more lipid and carbohydrate than those that performed worst (i.e. those fed D. tertiolecta). Larvae fed C. muelleri or the mixed-algae diet had higher proportions of the essential fatty acids eicosapentaenoic acid [EPA, 20:5(n-3)] and arachidonic acid [ARA, 20:4(n-6)] than the larvae fed on other diets. Furthermore, the larvae fed T. suecica, which showed intermediate performance between larvae fed C. muelleri and T-iso or D. tertiolecta, also had higher proportions of EPA and ARA. Both C. muelleri and T. suecica contained EPA and ARA, but T-iso and D. tertiolecta did not, except for trace amounts of EPA in T-iso. The fatty acid ARA appears to be much more important in the diet of larval prawns than has so far been considered. The level of the essential fatty acid docosahexaenoic acid [DHA, 22:6(n-3)] in the algal diet and the larvae was not related to the performance of the larvae; only C. muelleri and T-iso contained DHA. However, the nauplii contained large proportions of DHA, suggesting that these were sufficient to meet the larval requirements for DHA during their development to Mysis 1. Growth increments of the penaeid shrimp Penaeus vannamel (Boone) and P. schmitti (Burkenroad) grown in commercial production ponds in Ecuador and Colombia were measured over a 4 mo period between October 1987 and January 1988. P. vannamel grown from wild caught seed in Ecuadorian ponds exhibited a growth rhythm which significantly coincided with lunar phases. Growth increments of at least 1 gwk-1 occurred around a new and full moon, while growth increments of less than 1 gwk-1 occurred during first and last quarters. P. vannamel transported from Ecuador as nauplii, raised in Colombian hatcheries and on grown in Colombian ponds exhibited a cyclic pattern in wet weight increments but not correlation to lunar cycles was observed. P. schmitti showed no cyclic pattern in growth. The two species under consideration are indigenous to two different, and quite unique areas displaying different environmental characteristics. The link between the growth pattern, biological and lunar cycles is discussed.  相似文献

Levels of calcium, magnesium, zinc, copper, iron, and manganese were examined in the muscle, hepatopancreas, and gonad of white shrimp, Penaeus vannamei, females at different stages of ovary maturation. Eyestalk-ablated shrimp were sampled during the intermolt stage and classified using the following maturation stages evaluated histologically: cortical stage, vitellogenesis, previtellogenesis, and without gonadic development. In muscle, the copper concentration was significantly lower in the vitellogenic stage than in other stages of ovary development. In the hepatopancreas, manganese levels were higher in the cortical and vitellogenic stages than in animals in previtellogenesis and without gonadic development. No other influence of ovary maturation on muscle and hepatopancreas mineral content was found. In contrast, in the ovary, significant differences of some mineral concentrations were detected. Zinc, iron, and magnesium concentrations were significantly increased in the cortical stage compared to the vitellogenic and previtellogenic stages. Our results indicate that these elements are selectively accumulated during ovary development, especially when the ovaries reach the cortical stage. The polychlorinated biphenyl Aroclor® 1254 was released in an accidental leakage of heat-exchange fluid from an industrial plant, into the Escambia River, near Pensacola, Florida, USA. This material was carried downstream, and is now found in the fauna of Escambia Bay and its contiguous waters, prime nursery areas for fishes and invertebrates such as penaeid shrimp. The significance of pollution by this chemical was assessed by establishing toxicity levels, determining routes of entry, and investigating its movement and distribution in various tissues of shrimp under controlled conditions in the laboratory. The variation in the concentration and fatty acid composition of lipid classes during the molting cycle of the prawn Penaeus japonicus was investigated. The lipid concentration of the whole body reached a maximum at mid-premolt (Stage D2) and then decreased to low level at late premolt (Stage D3–4). The accumulation of lipids during the premolt period seemed to be attributable to the increase of both polar and neutral lipids. The increase of neutral lipids at Stage D2 was derived from not only triglycerides but also free sterols and free fatty acids. Regarding the fatty acid composition of every lipid class, a marked variation occurred mainly at the intermolt (Stage C). In this stage, the polar lipids were rich in monoenoic acids such as 18:1 and poor in polyenoic acids such as 20:53 and 22:63. The triglycerides were rich in polyenoic acids at Stage C, but poor in monoenoic acids such as 16:1 and 18:1. The steryl esters contained large amounts of saturated acids such as 16:0 and 18:0 throughout the molting cycle, however the level of polyenoic acids increased at Stage C.  相似文献

The nutritional value of Artemia sp. as food for marine fish and crustacean larvae has been linked to the level of its polyunsaturated fatty acid (PUFA) content. Experiments in August 1984 were conducted to determine the effects of various artificial diets and algae on fatty acid composition of PUFA-deficient Artemia sp. (Utah GSL strain) and their resulting value as food for postlarvae of the prawn Penaeus monodon (Fabricius). Nauplii of the brine shrimp were grown on extracts of corn, copra, soybean and rice bran containing precursors (C18) to long-chain PUFA and also on algal species containing different levels of long-chain PUFA (C20). The nauplii were then used as food for P. monodon postlarvae. The results revealed that absence of C20 polyunsaturates from the feeds and their presence in the algae were reflected in the polyunsaturated fatty acid content of the tissues of Artemia sp. When fed with brine shrimp fed on algae, P. monodon displayed better postlarval survival and significantly higher growth; related to the content of polyunsaturated fatty acids in Artemia sp. A practical feeding approach in prawn hatcheries would be to grow Artemia sp. on a cheap diet such as rice bran, and then to enhance its nutritional value with a diet high in PUFA prior to harvesting, in order to improve hatchery production.  相似文献

The biological effects of a naturally occurring perturbation were compared with one which may be induced by man's activities in the coastal zone. The estuarine shrimp Palaemonetes pugio was exposed for 32 d to fluctuating (18°–22°C) temperatures (FT) and/or 2,6-dimethylnaphthalene (DMN)-contaminated food at a concentration of 0.24 g DMN g-1 wt food. After exposure, relative survival rates were evaluated in a matrix of environmental challenge tests. Low salinity (2 S) had little overall effect on survival. Survival at elevated temperatures (33°C) was significantly lower than in reduced salinity and was independent of the treatment effects. Resistance to the challenge of hypoxia+reduced salinity, however, in contrast to the other tests, was significantly reduced by exposure to FT and DMN-contaminated food. The influence of FT was much stronger and obscured the effects of DMN-contaminated food when shrimp were exposed to both perturbations at the same time. The persistent, residual effects of FT and DMN-contaminated food on the survival of P. pugio during exposure to hypoxia were evaluated after a 16-d recovery period with stable temperatures and uncontaminated food. The initial deleterious effects of FT appeared to be ameliorated by the stable temperatures. On the other hand, shrimp which had ingested DMN-contaminated food, prior to the clean food, exhibited enhanced survival during exposure to hypoxia. After ingesting contaminated food for 32 d, P. pugio contained concentrations of DMN approximately 1 order of magnitude greater than that in the food. Shrimp kept in FT generally had higher concentrations of DMN than shrimp held in stable temperatures. Soy cake* was homogenized and filtered through an 80 mesh net to produce small, detritus-like particles. The effect of soy-cake particles, an artificial detritus, on the survival and growth of Penaeus japonicus Bate zoea was then tested. The soy cake was considered to be effective for survival and growth of the zoea. The best survival rate, 85.9%, was obtained in a tank receiving soy cake in quantities of 0.16 mg/zoea/day. An experiment was also performed to compare three kinds of food: particles of soy cake alone, diatoms (Chaetoceros rigidus) alone, and a half and half mixture of the two. The best growth, 2.49 mm, and the highest survival rate, 79.1%, were found in larvae fed the mixture of soy-cake particles and diatoms. The results thus seem to demonstrate that particles of soy cake could be usefully applied to the mass-culture of the penaeid prawn.  相似文献

Five species of unicellular algae of the same age, cultured bacteria-free under standard growth conditions, were analyzed for chemical composition and fed to different size classes of Artemia salina. The green algae Chlamydomonas sphagnicolo, Dunaliella viridis, Platymonas elliptica and Chlorella conductrix had significantly higher percentages of protein and lipid than did the diatom Nitzschia closterium. Total ash value was highest in populations of N. closterium. Shrimp fed Chlamydomonas sphagnicolo cells assimilated highest percentages of organic matter, while those fed Chlorella conductrix had lowest assimilation rate. Respiration rates were inversely proportional to animal size (weight) and algal cell volume. Growth, survival, rate of sexual maturtion, and sex ratio were dependent on the growth and assimilation efficiencies obtained from each respective algal food. Shrimp fed Chlamydomonas sphagnicolo, D. viridis, or P. elliptica cells displayed highest growth and assimilation efficiencies.  相似文献

This study compares water quality parameters, shrimp growth and mortality rates, and biomass at harvest in two ponds of equal size, seeded with the same density (7 m2) of White Spot Syndrome Virus (WSSV) and Monodon Baculo Virus (MBV) negative post-larvae (PL)-20 of shrimp, Penaeus monodon in the Vellar estuary of South India. The primary difference between the ponds was the water source; one was filled from the estuary and the second with water from bore wells with high alkalinity. Temperature in both ponds was similar and reached 320C after 185 days of culture. Dissolved oxygen (DO) levels were within the acceptable range although levels in the alkaline pond were near the lower limit for the last 90 days before harvest. Salinity levels were similar in both ponds, above optimal levels, and increased over the 185 days. Alkalinity in the estuarine water was typically <50 ppm and again 200-320 ppm in the alkaline pond. In the alkaline pond, beginning on the 75th day mineral deposits was observed covering all parts of the shrimp including the eye and the inner gill chambers, and by harvest, 42% of the shrimp showed this coating. Elemental analysis identified the major constituents as calcium, phosphorus and manganese. Survival rates in the estuarine-water-fed pond was 92% with a total pond biomass at harvest of 1.65 tons ha-1 compared to survival of 79% in the alkaline pond and a biomass at harvest of 1.020 tons ha-1. When well water must be used, its alkalinity should be monitored and diluted with water from other sources.  相似文献

 The ability to introduce and express foreign genes can greatly facilitate in vitro studies of the molecular biology of an organism. We report here the first use of modified retroviral vectors to achieve foreign gene expression in primary cultured cells from the Oka organ and ovary of the blue shrimp Penaeus stylirostris (Litopenaeus) stylirostris (Holthuis 1980). Of the heterologous promoters tested, the Moloney murine leukemia virus and Rous sarcoma virus promoters mediated the highest levels of reporter gene expression. Firefly luciferase and Escherichia coliβ-galactosidase were successfully used as reporter genes in cultured cells from both Oka organ and ovary. Veligers ofCrepidula fornicata (L.) were reared for 12 days at constant temperatures of 15°, 20°, 25°, 30° and 35°C, and at 5 C° daily cycles of equal periodicity (COEP) over the temperature ranges 15° to 20°C, 20° to 25°C, 25° to 30°C and 30° to 35°C. COEP consisted of equal periods (6 h) of maximum temperature, minimum temperature, and uniformly increasing and decreasing temperature each 24 h period. Survival was high and not influenced by cyclic or constant temperature from 15° to 30°C. At 35°C and COEP 30° to 35°C, all larvae died before Day 6. Shell growth rate increased markedly over the range 15° to 25°C, and growth rates at cyclic temperatures in this range were intermediate between growth rates at the corresponding constant temperatures. Larvae reared at COEP 15° to 20°C and COEP 30° to 35°C had discontinuities in their shells due to inhibition of shell secretion during the adverse part of each temperature cycle. Groups ofc. fornicata veligers were exposed for 2 days to daily temperature cycles of equal and unequal periodicity in the critical 30° to 35°C range. [Cycles of unequal periodicity (COUP) consisted of unequal periods (varying between 3 and 15 h) of maximum and minimum temperature and uniformly increasing and decreasing temperature each 24 h period.] These veligers showed shell growth although their body tissue declined, as indicated by decreasing carbon content per larva. Least shell growth and most body tissue loss occurred in those cycles with the longest exposure to higher temperature. Larvae exposed for arious days to the mildest 30° to 35°C COUP (15 h at 30°C, 3 h increasing temperature, 3 h at 35°C and 3 h decreasing temperature) recovered and resumed normal growth when transferred to constant 30°C, but their growth was retarded in proportion to the number of days in the temperature cycle. Rates of shell growth of veligers in temperature cycles show an immediate effect of environmental temperature, while changes in carbon content per larva better reflect the effects of temperature on general metabolism and survival.  相似文献
