Phosphate depleted Pyrocystis noctiluca (Murray) Schuett 1895 has at least one phosphomonoesterase (EC 3:1:3:1) which is triphasic between 0.1 and 222 mol P. The enzyme has a broad temperature range with maximum activity at 50 °C and a Q10 of 1.4 to 1.5. A break in the Arrhenius plot at 35 °C implies the enzyme is membrane-bound. Cytological staining of whole cells and cell fractionation studies (showing 26 times higher specific activity in the particulate compared with the cytoplasmic fraction) suggest the enzyme is plasmalemma-bound. The enzyme has an absolute metal requirement which would be satisfied by Mg++ but not Mn++, Zn++, Fe++, or Co++ at seawater concentrations. Alkaline phosphatase is a stable enzyme whose activity is not altered by inhibitors of protein synthesis. Orthophosphate inhibition of enzyme activity was largely eliminated in the presence of these inhibitors. Apparently, a protein induced by PO4 3-, rather than PO4 3- itself, inhibits alkaline phosphatase. Cell-free alkaline phosphatase can hydrolyze a variety of phosphate esters and linear polymers of inorganic phosphorus as well as disolved organic phosphorus from tropical oceanic waters. These same hydrolysable organic and inorganic phosphorus compounds support the axenic culture growth of P. noctiluca, suggesting that naturally occurring hydrolysable organic phosphorus compounds may also support the growth of this alga.  相似文献

g -glucosidase and alkaline phosphatase, two ectoenzymes involved in the microbial decomposition of polysaccharides and organic phosphates, were evaluated in water samples collected from two areas of the Northern Adriatic Sea during the multidisciplinary Prisma II research project. The distribution of g -glucosidase and alkaline phosphatase is reported together with that of leucine aminopeptidase, an enzyme involved in the degradation of protein compounds. The data obtained showed the prevalence of g -glucosidase in summer months in the northern area, located in front of the Po delta, while in winter it is higher in the southern area (in front of Ancona). Phosphatase activity during February, 1998 had a maximum of 107 r nml m 1 h m 1 at a coastal station in the northern area; this enzyme appeared to be mainly associated with phytoplankton because the increase in alkaline phosphatase was associated with the increase of phytoplankton biomass, measured by chlorophyll a content. The relations between the microbial activities and environmental parameters are discussed.  相似文献

In late summer (Antiprod I cruise, March 1977), antarctic waters of the southern part of the Indian Ocean contain low amounts of dissolved Vitamin B12: 1 ng l-1 on average. However, higher concentrations were recorded in two areas: in the subantarctic convergence zone and between 50° and 57°S, with values greater than 3,5 ng l-1. Despite nutrient-rich waters, primary production was low and the phytoplankton sparse. Abundance of diatoms was related, to a certain extent, to the concentration of Vitamin B12. An experimental approach (Antiprod II cruise, February and March 1980) was applied to (1) artificially enriched natural sea water, and (2) cultures of two diatom species isolated from antarctic waters, Nitzschia turgiduloides and Chaetoceros sp. The experiments demonstrated that antarctic diatoms in general do not require any of the three investigated vitamins for growth. However, Vitamin B12 has a slightly stimulating effect on growth. This may explain the observed relation between phytoplankton density and vitamin concentration in the Antarctic Ocean. This experimental approach elucidates the minor role of vitamins in the primary production of antarctic waters and provides indirect evidence of the major importance of turbulence as a limiting factor.  相似文献

Seawater containing natural phytoplankton populations from Vineyard Sound, USA was enriched in the laboratory with three levels each of ammonium and phosphate and with a combination of ammonium and phosphate which provided three different N:P ratios. The addition of ammonium produced more cells and chlorophyll a than the control or the phosphate enrichments. However, enrichment with ammonium and phosphate, regardless of the N:P ratio, yielded the most cells and chlorophyll a. Thus, nitrogen seems to be the primary limiting nutrient, with phosphate showing secondary limiting effects. The ratios of photosynthetic pigments decreased with the increased chlorophyll a production in the enriched cultures. There were no significant changes in the species composition within the cultures, so that the observed changes in pigment ratio and chlorophyll a content were due to physiological responses.  相似文献

A carbon-14 assimilation method was used to determine action spectra and photosynthesis versus irradiance (P versus I) curves of natural populations of phytoplankton and zooxanthellae from a coral reef fringing Lizard Island in the Australian Barrier Reef. The action spectra were related to the phytoplankton species composition. The curves showed shade adaptation in phytoplankton from deeper waters and in the zooxanthellae. Rates of photosynthesis of zooxanthellae were shown to be highly but variably dependent on their host organisms. Photosynthetic production by zooxanthellae was about 0.9 gC m-2 day-1, which is about three times higher than phytoplankton production in the waters close to the reef.  相似文献

Investigations on the influence of pressure on the activity of alkaline phosphatase of some marine invertebrates from the Baltic Sea were carried out at temperatures between 5° and 55°C. The results obtained indicate that the influence of pressure on alkaline phosphatase activity is modified by temperature. At low temperatures, pressure has a decreasing effect on enzyme activity, while it has an increasing effect at higher temperatures. The gill homogenates of the relatively pressure-resistent bivalves Cyprina islandica and Mytilus edulis show a lower decrease of activity under pressure at low temperatures than the gill-homogenate of Carcinus maenas and a purified enzyme preparation of a homoiothermal animal.  相似文献

Alkaline phosphatase activity in seawater samples taken from Tokyo Bay was measured by both spectrophotometric and fluorometric methods. A stratified distribution pattern of the enzyme was observed in August, a vertically mixed pattern in December. The distribution of phosphatase activity in the eutrophic seawater was paralleled by variations in other parameters, such as viable counts of bacteria, chlorophyll a content, inorganic and total phosphorus concentrations, amounts of seston, particulate deoxyribonucleic acids (DNA) and protein. A significant correlation between phosphatase activity and these measurements indicated that the enzyme was a good indicator of the degree of eutrophication. The positive relationship between phosphatase and inorganic phosphorus indicates that enzyme activity was not inhibited at inorganic phosphorus levels present in the bay and that production of phosphatase by microorganisms inhabiting the bay was not repressed at the inorganic phosphorus levels in the bay. Culture experiments revealed that the formation of repressible phosphatase by bacteria isolated from the bay was not affected by the inorganic phosphorus levels in the bay.  相似文献

In the period from 1980 to 1984 organic phosphorus, nutrients, primary production rates (14C), chlorophyll a (chl a) standing crops, and basic oceanographic parameters were measured during 23 cruises at six stations in the open waters of the northern Adriatic Sea. These waters are significantly influenced by polluted Po River discharge. Organic phosphorus was correlated with several parameters which characterize phytoplankton activity and organic matter decomposition processes. In the late winter-spring period, organic phosphorus is produced during phytoplankton blooms. It is hypothesized that microzooplankton grazing is the main factor increasing the organic phosphorus concentrations in summer (up to 1.1 mol 1-1). Fall and winter had much lower values (below 0.3 mol 1-1) due to remineralization processes and an increased water mass exchange between the northern and central Adriatic regions. The direct contribution of organic phosphorus by freshwater discharge was not found to be significant. The higher organic phosphorus concentrations that can occur in low salinity waters are most likely due to their increased capability to support primary production.  相似文献

In high-latitude waters such as the Southern Ocean, the primary production of phytoplankton supports the ecosystem. To understand the photo-acclimation strategy of such phytoplankton within cold environments, the vertical distribution profile of photosynthetic pigments was analyzed in the Southern Ocean. Samples were taken along 110°E during the austral summer, and along 150°E and around the edge of the seasonal sea ice of the Antarctic Continent during the austral autumn. Pigment extraction methods were optimized for these samples. The standing crop of chlorophyll a was larger in the region along the edge of the seasonal sea ice than at sampling stations in open ocean areas. Chlorophyll concentration seemed to be dependent on the formation of thermo- and haloclines along the edge of the seasonal sea ice, but not in the open ocean where such clines are less pronounced. The marker pigments fucoxanthin and/or 19′-hexanoyloxyfucoxanthin were dominant at most sampling stations throughout the water column, while other marker pigments such as alloxanthin were quite low. This indicated that diatoms and/or haptophytes were the major phytoplankton in this area. Comparison of the relative ratio of fucoxanthin with that of 19′-hexanoyloxyfucoxanthin allowed some stations to be characterized as either diatom-dominant or haptophyte-dominant. The relative ratio of xanthophyll-cycle pigments (diadinoxanthin plus diatoxanthin) to chlorophyll a was high in surface waters and decreased gradually with depth. This suggests that near the ice edge during summer in the Southern Ocean, both diatoms and haptophytes acclimate to their light environments to protect their photosystems under high-light conditions.  相似文献

A phosphate uptake system responding positively to light would offer the algae a competitive advantage over bacteria, particularly when phosphorus is limiting. Effects of light on phosphate uptake in phytoplankton were investigated in view of conflicting literature reports, to test the hypothesis that both the P status and the light history of the population controlled the response. Maximum stimulation, to about 2.7x dark uptake, was observed at a Strait of Georgia, British Columbia, Canada station where phytoplankton was phosphorus sufficient but light limited. Stimulation was similar, about 1.8 x the dark rate, in the Northern Sargasso Sea, where phytoplankton was probably limited by phosphate but not by light. Minimal stimulation was observed in the Sheepscot Estuary, Maine, USA, where the population appeared to be neither phosphorus nor light limited.  相似文献

Sediment samples were collected from different stations of the Vellar estuary for isolation of total actinomycetes and phosphate solubilizing actinomycetes. Phosphatase activity in the sediments was also investigated Consistently a higher number of actinomycetes, phosphate solubilizing actinomycetes and phosphatase activity were recorded from the clay sediments than the sandy sediments at all the stations. In all, 7 strains showed positive phosphatase activity. Among them, one strain PS-3 exhibited good activity and was further investigated for optimum phosphorus solubilization at different pH (6, 6.5, 7, 7.5 and 8) and incubation (1st day to 20th day) periods. The solubilizing activity was maximum at the pH 7 and an incubation period of 13 days was required for an appreciable quantity of phosphorus to be leached into the medium. Based on the chemotaxonomical and conventional methods of identification, the strain PS-3 has been tentatively identified as Streptomyces galbus. The present study indicates that phosphatase enzyme and S. galbus along with other actinomycetes species would play a major role in solubilizing the phosphate in the estuarine ecosystem and increasing the soluble phosphate concentration thereby enhancing the productivity  相似文献

Microzooplankton grazing and selectivity of phytoplankton in coastal waters   总被引:16,自引:0,他引:16  
Microzooplankton grazing activity in the Celtic Sea and Carmarthen Bay in summer 1983 and autumn 1984 was investigated by applying a dilution technique to high-performance liquid chromatographic (HPLC) analysis of photosynthetic pigments in phytoplankton present within natural microplankton communities. Specific grazing rates on phytoplankton, as measured by the utilisation of chlorophyll a, were high and varied seasonally. In surface waters during the autumn, grazing varied between 0.4 d-1 in the bay and 1.0 d-1 in the Celtic Sea, indicating that 30 and 65% of the algal standing stocks, respectively, were grazed daily. Grazing rates by microzooplankton within the thermocline in summer suggest that 13 to 42% of the crop was grazed each day. Microzooplankton showed selection for algae containing chlorophyll b, in spite of a predominance of chlorophyll c within the phytoplankton community. Changes in taxon-specific carotenoids indicated strong selection for peridinin, lutein and alloxanthin and selection against fucoxanthin and diadinoxanthin. This indicates a trophic preference by microzooplankton for dinoflagellates, cryptophytes, chlorophytes and prasinophytes and selection against diatoms, even when the latter group forms the largest crop within the phytoplankton. Interestingly, those algal taxa preferentially grazed also showed the highest specific growth-rates, suggesting a dynamic feed-back between microzooplankton and phytoplankton. Conversion of grazing rates on each pigment into chlorophyll a equivalents suggests firstly, that in only one experiment could all the grazed chlorophyll a be accounted for by the attrition of other chlorophylls and carotenoids, and secondly that in spite of negative selection, a greater mass of diatoms could be grazed by microzooplankton than any other algal taxon. The former may be due either to a fundamental difference in the break-down rates of chlorophyll a compared to other pigments, or to cyanobacteria forming a significant food source for microzooplankton. In either case, chlorophyll a is considered to be a good measure of grazing activity by microzooplankton.  相似文献

Acid and alkaline phosphatase activities have been partially characterized in Ruditapes philippinarum (Adams and Reeve, 1850). Two activity peaks at pH=4.5 and pH 10.5 were detected in the gill, digestive gland, mantle, siphon and foot. Acid phosphatase activity was higher than that of alkaline phosphatase. The highest activity for both enzymes was observed in the digestive gland and, in decreasing order, the gill, foot, siphon and mantle. Alkaline phosphatase activity was similar in the mantle, siphon and foot. K m values were determined for both enzymes in the gill and digestive gland. Hill coefficients were near 1, indicating no allosteric behaviour for either enzyme in the two organs. The optimum temperature was the same for acid phosphatase in both gill and digestive gland (50 °C), while for alkaline phosphatase it differed for these two organs (gill, 40 °C; digestive gland, 35 °C). The apparent activation energy was obtained from Arrhenius plots, and ranged from 8.61 kcal/mol for alkaline phosphatase in the gill, to 10.84 kcal/mol for acid phosphatase in the digestive gland. The effects of metals (1 mM conc) on both enzyme activities were assayed in vitro. Hg strongly inhibited the enzyme activities in the gill and digestive gland, probably because of its affinity for the sulphydryl group. Histochemically, acid phosphatase in the gill was located in a granular form throughout the gill cells, but was undetectable in the ciliate epithelium of the gill filaments. Alkaline phosphatase was located in the gill skeleton. Clam size and phosphatase activities were inversely related, probably reflecting a decrease in shell deposition with inereasing size. As a function of season, both enzymes were present in lowest amounts in winter, when undifferentiated sex cells were predominant in the germinative epithelium, and highest in summer, when ripe individuals of both sexes were more frequent.  相似文献

In order to provide a background picture of the water quality of the Egyptian Red Sea a number of hydrological and chemical parameters have been measured bimonthly in 2000. Few data are available on this area, which is apparently subjected to an increasing human impact due to recreational (swimming and diving), industrial (mainly phosphate shipping and industry) and fishing/harbor activities. The results of the present study indicate that changes in the salinity and pH were not significant with highly oxygenated seawaters. The levels of suspended solids (as total suspended matter, TSM) and chlorophyll-a (Chl-a) were generally low and showed an homogeneous distribution in the study region. The ratio of chlorophyll-a to total suspended matter concentrations increased between November and March and decreased from May to September. Chlorophyll-a was significantly correlated with transparency and total suspended matter concentrations in July, September and November. Nitrogen, phosphorus and reactive silicate concentrations were generally low, and allowed classifying the Egyptian Red Sea coastal water as oligotrophic to mesotrophic. The middle region of the study area, which was located between Safaga and Qusair displayed relatively high phosphate contents when compared with other coastal areas. The high values of N:P ratios indicate that PO 4 -P is the limiting factor for phytoplankton growth in the Red Sea coastal waters, with the possible exception of the middle region. Significant relationships were found between chlorophyll-a concentrations and nutrient levels in different sampling periods. Spatial distribution patterns of the studied variables revealed that productivity of the Red Sea coastal waters is mostly controlled by phosphate concentrations, salinity, temperature and dissolved oxygen.  相似文献

The magnitude and physiological characteristics of biological nitrogen fixation have been studied in the oligotrophic waters of the North pacific gyre. The filamentous blue-green algae Trichodesmium spp. and Richelia intracellularis were the important nitrogen-fixing phytoplankton. Most of the nitrogen fixation occurs in the upper 40 m of the water column, with detectable fixation as deep as 90 m, which corresponds to about the 1 % light depth. There was no evidence of photoinhibition of nitrogen fixation, although CO2 reduction was depressed slightly at the highest light levels. The rate of nitrogen fixation in the water column varied throughout the day, being highest in mid-morning and in late afternoon. Relatively high fixation rates were also found during periods of darkness. Elevated oxygen concentrations had a marked inhibitory effect on rates of nitrogen fixation, a pO2 of 0.4 atm causing a 75% inhibition. Data from studies of nitrogen fixation and assimilation rates of 15N-labelled nitrate, ammonium, and urea indicate that nitrogen fixation furnished about 3% of the total daily fixed nitrogen requirement for phytoplankton growth. Studies with isolated colonies of Trichodesmium spp. indicated that 100% of their nitrogen requirement was met by nitrogen fixation. Chemical composition of the Trichodesmium colonies showed that the C:N ratio was 4.1 and that their phosphorus content relative to carbon or nitrogen was much lower than that of the total particulate material in the water column. Elevated ratios of carbon: adenosine triphosphate (ATP) also suggest that phosphorus deficiency may be limiting the growth of Trichodesmium. The magnitude of nitrogen fixation in the gyre is seasonally dependent, with high rates in late summer and autumn. At these times the water column is stratified, with phosphate and nitrate barely detectable in the upper 100 m. Our data suggest that during these months of stratification, biological fixation of nitrogen amounts to about 33 g-at N/m2/day.  相似文献

The effect of ammonium (5, 10 M N) and phosphate (2, 5, 10 M P) on the growth of the giant clam Tridacna gigas and its symbiotic dinoflagellate Symbiodinium sp. was examined. A 3 mo exposure to these nutritients significantly increased the N or P composition of the soft tissues, as reflected in a corresponding change in C:N:P ratio. Furthermore, exposure to N or N+P markedly increased the amount of soft tissue, but P alone did not, demonstrating that increased availability of inorganic nitrogen enhances tissue growth of the clam host. With addition of N, or N+P, there was a significant increase in the total number of zooxanthellae per clam, with a corresponding decrease in chlorophyll a (chl a) content per zooxanthella. However, only with N+P was there an increase in the zooxanthellae mitotic index. The inverse relationship between zooxanthellae number and chl a per zooxanthella is consistent with phytoplankton studies indicating conditions of nutrient-limitation. Furthermore, the unaffected C:N:P composition of the zooxanthellae and their relatively low specific-growth rates (4 to 10%) also suggest that they are nutrient-limited in vivo. In particular, their high mean C:N:P ratio of 303:52:1 indicates that, relative to C, they are much more depleted in P and less in N than are free-living phytoplankton. Furthermore, polyphosphates (phosphate reserves) were undetectable, and the activity levels of acid phosphatase in the zooxanthellae were relatively high and not influenced by the host's exposure to increased P concentrations in the sea water, implicating the clam host in active regulation of P availability to its symbiotic algae. This is strong evidence that N-limitation of clam zooxanthellae is a function of the availability of ammonium to the symbiosis while, irrespective of nutrient levels in sea water, clam zooxanthellae still show characteristics of P-limitation.  相似文献

Potential environmental effects of the discharge of industrial wastewater from manufacturing of bleached straw pulp have been studied. Bleached neutral sulphite straw pulping results in discharges of both black cooking and spent bleaching liquors. Time proportional samples were taken from the combined bleaching effluent and the combined mill effluent, (i) Mutagenic activity of the two samples in an Ames’ test, and (ii) acute toxicity of the combined mill effluent sample towards the photosynthetic activity of natural marine phytoplankton from the receiving waters were determined. Also, acute toxicity was determined of the slowly biodegradable or persistent organics remaining after aerobic stabilization of the sample towards (ii) photosynthetic activity of natural phytoplankton, (iii) mussels (Mytilus edulis), (iv) eels (Anguilla anguilla) and, (v) crustacean (Nitocra spinipes); and furthermore, (vi) reproduction test of the crustacean and (vii) growth inhibition test of the marine diatom (Phaeodactylum tricornutum) were performed. Mutagenicity emission factors (MEF's) and toxicity emission factors (TEF's) were calculated to classify the effluents. Combined mill effluent sample showed a mutagenic activity per t90 of one fourth the activity of a kraft pulp bleaching effluent, and the inhibition of the photosynthesis of natural phytoplankton was significant up to 2600 times dilution. After aerobic stabilization, inhibition was observed up to 400 times dilution. Other effects of the combined mill effluent sample were only significant below 40 times dilution. The bleaching effluent showed a mutagenic activity per t90 comparable to kraft bleaching effluents. Other effects were only significant below 20 times dilution.  相似文献

Fatty acid analyses were used to study the transition from endogenous to exogenous feeding in Arcto-Norwegian cod larvae Gadus morhua L. sampled from Lofoten waters, Northern Norway in April 1985. Fatty acids of total lipids were analysed from phytoplankton, eggs and nauplii of Calanus finmarchicus, and cod eggs and larvae. Gas chromatographic and gas chromatographic-mass spectrometric methods were used. On the basis of these analyses it is suggested that lipids of phytoplankton origin form an important part of the diet of cod larvae during the first feeding period.  相似文献

A new method is described for the determination of phytoplankton growth rates and carbon biomass. This procedure is easy to apply and utilizes the labeling of chlorophyll a (chl a) with 14C. Pure chl a is isolated using two-way thin-layer chromatography, and the specific activity of chl a carbon is determined. Data from laboratory cultures indicate that the specific activity of chl a carbon becomes nearly equal to that of total phytoplankton carbon in incubations lasting 6 to 12 h and can be used to calculate phytoplankton growth rates and carbon biomass. Application of the method to the phytoplankton community in an eutrophic estuary in Hawaii indicates that the cells are growing with a doubling time of about 2 d and that about 85% of the particulate carbon consists of phytoplankton carbon.  相似文献
