A Zn- and pH-buffered medium was used to study the accumulation of Zn in batch-cultured algae (Selenastrum capricornutum Printz.). All experiments were carried out using (65)Zn radiotracers, which were measured by gamma-ray spectrometry. Zinc was applied in the presence of nitrilotriacetic acid (NTA), leading to free Zn(2+) concentrations ranging from 4.2 x 10(-11) to 1.6 x 10(-7) mol litre(-1). During the 75-h experiments, the rates of Zn influx and efflux were determined, the latter with full consideration for algal growth rates. Algal growth stopped completely at a free Zn(2+) concentration of 1.6 x 10(-8) mol litre(-1). Initial Zn binding was determined and, based on free Zn(2+) concentrations, presented by the apparent dissociation constant K(diss) and capacity C as 4.6 x 10(-9) mol litre(-1) and 19.8 x 10(6) mol gDW(-1), respectively. The rate constant of Zn efflux could be calculated as a constant 0.021 +/- 0.003 h(-1), irrespective of the Zn concentrations applied. The rates of Zn influx were expressed by the V(max) and K(M) values as 3.3 x 10(-10) mol gDW(-1) s(-1) and 6.3 x 10(-9) mol litre(-1), respectively. These values, which are based on applied free Zn(2+) concentrations, are compatible with the operation of a high-affinity low-concentration carrier mechanism. The results suggest that, under the conditions applied, free Zn(2+) may be the most relevant Zn species for Zn uptake; furthermore, relative growth rate may be regarded as a relatively sensitive signal for Zn stress circumstances.  相似文献

In order to study the short-term ecotoxicity of metals to the freshwater mussel Dreissena polymorpha, the effects of Cu, Zn and Cd on the filtration rate of this mussel were determined in laboratory experiments. Filtration rate was chosen as the endpoint, because it is a sensitive sublethal parameter compared to mortality and it is an important parameter given the ecological role D. polymorpha fulfills. The filtration rate was calculated from the decrease in algal concentration, fed to mussels in aquaria, containing different metal concentrations. The EC50 for Cu (41 microg litre(-1)) was lower than for Cd (388 microg litre(-1)) and Zn (1350 microg litre(-1)). The NOEC(accumulation) for the essential metal Zn was higher than for the essential metal Cu. Cadmium, a non-essential metal, was accumulated at all elevated water concentrations, so the NOEC(accumulation) was the concentration in the control water (<0.2 microg litre(-1)). All (no) effect concentrations found in this study were above the quality criteria set for metal concentrations in Dutch surface water, suggesting that the zebra mussel is sufficiently protected by these quality criteria.  相似文献

An evaluation of water quality and phytoplankton composition position was carried out in order to determine the trophic conditions of Lake Patzcuaro (2035 m above sea level), a high altitude tropical lake. Temperatures ranged from 15 to 23 degrees C. Total phosphorus and inorganic nitrogen showed a seasonal variation; highest values coincided with the rainy season (0.48 and 2.1 mg litre(-1), respectively). Dissolved oxygen ranged from 2 to 7.9 mg litre(-1) at the surface and from 0.6 to 7.3 mg litre(-1) on the bottom, the lowest values being found in shallow zones. Average transparency varied from 0.62 to 1.4 m Secchi depth. Rainfall was a primary factor in seasonal variability as it influenced both physical and biological conditions by contributing to the transport and deposition of silt, which mixed with sinking algal cells. The composition of the surface phytoplankton segregated along five major divisions comprising a total of 49 species. General seasonal patterns of dominance alternated in a sequence beginning with Bacillariophyta, through Chlorophyta to Cyanophyta. Diatoms, the dominant group from February to early June, included Melosira granulata, Stephanodiscus sp., Synedra sp. and Fragilaria sp. During the rainy season (late June to September), Microcystis aeruginosa, Oscillatoria sp., Anabaena sp., Merismopedia sp., Crucigenia cuadrata, Oocystis lacustris, Selenastrum gracile, Mallomonas sp. and Tetraediella sp. were important. Melosira granulata was present throughout the period of study. Spatial and temporal variability in both physical and biological conditions make it difficult to assign a specific trophic state to Lake Patzcuaro. Nevertheless, analysis of the algal community indicates a generally mesotrophic condition.  相似文献

The sensitivity of 56 algal strains, representing 7 taxonomic groups to the triazine herbicide, simetryne, was examined using EC50 values for growth. There was a wide range of values from 6.5 to 1500 microg litre(-1). The Volvocales (Chlorophyceae, Chlorophyta) and Cyanophyceae (Cyanophyta) as a whole were the most sensitive, whereas the Desmidiales (Charophyceae, Chlorophyta) and Bacillariophyceae (Chromophyta) were the most tolerant, although sensitivity differed among strains of a single species. Sensitive and tolerant species were both isolated from samples collected at the same site. The results suggest that changes in species composition and relative abundance will occur when herbicides are applied in natural habitats.  相似文献

The purpose of this study was to assess certain physiological responses of Lemna minor L. (duckweed) and Allium cepa L. (onion) to aquatic mercury at low concentrations. Following a 96-h exposure of plants to nutrient medium contaminated with known levels of mercuric chloride (HgCl(2)), 0.001 to 4 mg litre(-1) (0.0007 to 2.95 mg Hg litre(-1)) or methyl mercuric chloride (MeHgCl(2)), 0.0001 to 0.1 mg litre(-1) (0.0007 to 0.07 mg Hg litre(-1)), the physiological endpoints measured were the growth of fronds (Lemna minor) or roots (Allium cepa), and catalase and peroxidase activities in both plant assays. The EC(50) for HgCl(2) on the basis of the growth curve of Lemna minor was found to be 2.1 mg litre(-1). HgCl(2) and MeHgCl(2) were lethal to L. minor at concentrations of 4 and 0.01 mg litre(-1), respectively. The range of low concentrations that accelerated growth as well as enzymic activities in L. minor was 0.004 to 0.04 mg litre(-1) for HgCl(2) and 0.001 mg litre(-1) for MeHgCl(2). HgCl(2) and MeHgCl(2) induced maximum enzymic activity in Lemna fronds at concentrations of 0.008 and 0.0005 mg litre(-1), respectively. In Allium roots, catalase activity was accelerated at all the concentrations of HgCl(2) (0.001-2 mg litre(-1)) and MeHgCl(2) (0.0001-0.1 mg litre(-1)) tested. The activity of peroxidase was, however, accelerated by HgCl(2) at concentration range 0.01-1.0 mg litre(-1), or by MeHgCl(2) at 0.001 mg litre(-1). The concentrations of HgCl(2) and MeHgCl(2) that induced the highest enzymic activity in Allium roots were 0.05 mg litre(-1) and 0.001 mg litre(-1), respectively.  相似文献

The toxicity of nonylphenol to the common mussel (Mytilus edulis L.) has been determined in both semistatic and continuous flow test systems. The LC50 values obtained were for 96 h, 30 mg litre(-1); 360 h, 0.5 mg litre(-1); and 850 h, 0.14 mg litre(-1). Sublethal effects, manifested as decreased byssus strength and change of scope for growth, were obtained at a concentration as low as 0.056 mg litre(-1). Fertilization and early developmental success were not affected at the highest concentration tested (0.2 mg litre(-1)).  相似文献

The bioaccumulation and toxicity of zinc in Cladophora glomerata from two populations in the River Roding, Essex, UK, were examined in experimental laboratory flowing-water channels. Plants were subjected to zinc concentrations ranging from 0 to 4.0 mg litre(-1) at current velocities of 20-33 cm s(-1) for up to 3 h. Zinc in algal tissue was then quantified and toxicity was assessed by the ability of the alga to grow in a recovery medium after the experimental treatment. There was little difference in zinc bioaccumulation between Cladophora from the site showing mild organic pollution and that from the site subjected to considerable inputs from urban and motorway runoff. Uptake of zinc increased with increasing concentration in the test solution and was linear and proportional up to 0.4 mg litre(-1). Three stages of uptake were identified with the most dramatic accumulation occurring in the first 10 min. Experimental concentration factors ranged from 1.9-5.2 x 10(3), which were in agreement with those previously obtained in the field. Cellular damage was evident in Cladophora subjected to 0.4 mg litre(-1) zinc, and this increased with increasing zinc concentration, thus leading to the conclusion that, at times, the levels of zinc found in the river could be potentially damaging.  相似文献

The influence of zinc, manganese, and nickel on the degradation of MTBE (methyl tert-butyl ether), by an aerobic MTBE-degrading strain, Ochrobactrum cytisi, were investigated. The result showed that unlike previous findings, O. cytisi was able to degrade MTBE through direct metabolism when MTBE was present as the only carbon source. The degradation rate of MTBE was rapid, completed within 80 h. MTBE biodegradation by this strain was stimulated at low concentrations of Zn(2+) (1-5 mg l(-1)) and Mn(2+) (1-5 mg l(-1)) but inhibited at high concentrations of Zn(2+) (20 mg l(-1)) and Mn(2+) (20 mg l(-1)), and at low concentration of Ni(2+) (1-4 mg l(-1)). Kinetic parameters for MTBE degradation in the presence or absence of metals were obtained through nonlinear regression and a least-square minimization procedure. In all cases, a good agreement was achieved between kinetic simulations and experimental results.  相似文献

The effects of exposure to 40 nl litre(-1) SO2 + 40 nl litre(-1) NO2 on the reproductive biology of Polypodium interjectum (Shivas), Dryopteris affinis (Lowe) Fraser-Jenkins and Phyllitis scolopendrium (L.) Newman were investigated after 14 weeks exposure in a closed chamber fumigation system. The numbers of sori per pinna were reduced in response to SO2 and NO2 for D. affinis but were unaffected for the other species. Numbers of sporangia in sori and spore viability were reduced in P. interjectum and P. scolopendrium but not in D. affinis in response to the SO2 and NO2 treatment. Spore size was not affected by the pollution treatment. A separate experiment tested viabilities of spores collected from the three species in response to daily spraying with simulated mists at pHs of 2.5, 3.5, 4.5 and 5.6. For all three species, there was little or no spore germination in the pH 2.5 treatment and significantly reduced germination in response to the pH 3.5 as compared to the pH 4.5 and pH 5.6 treatments.  相似文献

The microscopic green alga, Chlorella pyrenoidosa was grown in settled and activated sewage under two different culture systems, batch and semi-continuous. Good growth was obtained in both types of wastewater and the algal production was comparable to and even higher than that found in commercial Bristol medium. The semi-continuous culture supported more growth than the batch system. There was a close relationship between algal growth and the amount of nutrient removed from both settled and activated sewage. A more rapid drop in NH(4)(+)-N was found in settle rather than activated sewage. The NH(4)(+)-N of settled sewage dropped from its initial 27 to 5 mg litre(-1) in both culture systems. On the other hand, the NO(3)(-)-N of activated sewage started to decrease from Day 2 onwards and the final NO(3)(-)-N concentration was less than 1 mg litre(-1) (over 90% removal efficiency). The amount of total inorganic nitrogen being reduced due to algal culture was similar in both types of sewage. The changes of phosphate content followed the same trend in both sewage, the P concentration increased slightly in the first two days then decreased, especially in the semi-continuous cultures. The final ortho-P in the sewage treated by Chlorella in semi-continuous culture was less than 5 mg litre(-1) (about 62% reduction). Such removal efficiency was slightly lower than those reported in previous studies. In general, the semi-continuous algal culture appeared to be a more suitable and efficient way for wastewater treatment than the batch system. With respect to the total reduction of wastewater inorganic N and P by means of Chlorella cells, there was no significant difference between settled and activated sewage.  相似文献

Cost-effective and environmentally acceptable methods are needed to remediate munitions-contaminated soil. Some perennial grass species are tolerant of soil contaminants and may promote remediation because of their high water use and extensive fibrous root systems. The effects of 2,4,6-trinitrotoluene (TNT) and its reduction product, 4-amino-2,6-dinitrotoluene (4ADNT), on germination and early seedling development of tall fescue (Festuca arundinacea Schreb.) were determined. Tall fescue seeds were germinated in nutrient-free agar containing 0-60 mg TNT litre(-1) or 0-15 mg 4ADNT litre(-1). Germination decreased linearly as TNT concentration increased but was not significantly affected by 4ADNT at these concentrations. Concentrations less than 30 mg TNT litre(-1) or 7.5 mg 4ADNT litre(-1) had little effect on seedling growth and development. Higher TNT or 4ADNT concentrations substantially delayed seedling development, caused abnormal radicle tissue development, and reduced secondary root and shoot growth. Seedling respiration rates decreased linearly with increasing TNT concentration. Experiments indicate that tall fescue may be grown in soils that maintain soil solution concentrations of 30 mg TNT litre(-1) or less.  相似文献

The effect of copper on the activity of Balbiani rings (BR1 and BR2) and nucleolar organizing region (NOR) in chromosome IV of the salivary gland of the 4th instar larvae of Chironomus ninevah has been investigated. Sublethal concentrations, i.e. 0.02, 0.1, 0.15 and 0.2 mg litre(-1) suppress the activity of BR1, and are statistically significant. The same concentrations reduced the activity of BR2 and NOR, but without significant differences, except 0.1 mg litre(-1) which was significant. The purpose of this work was to evaluate the effect of chronic toxicity of Cu(2+) on the expression of gene activity of midge salivary gland chromosomes Although there may be many more aquatic organisms that are more sensitive to copper, this midge was selected as a biological indicator species because of its association with benthic sediments.  相似文献

There is some concern that the antiozonant ethylenediurea (EDU), used for crop loss assessment due to ambient ozone (O3) may per se affect plant growth and yield. In view of this, and to provide knowledge for later field experiments, dose-response studies with EDU and O3 were carried out in greenhouses in winter and spring 1989, using radish (Raphanus sativus L.) cv. 'Cherry Belle' and 'Red Prince', grown in two different substrates. EDU was applied as a single or repeated soil drench in concentrations ranging from 300 to 800 mg litre(-1) in the first, and from 100 to 400 mg litre(-1) in the second trial. In the second experiment, plants were exposed to a chronic level of O3, mimicking ambient patterns, or to filtered air after the EDU-treatment. When applied in concentrations above 300 mg litre(-1), EDU reduced growth, thereby affecting the development of the thickened hypocotyl far more than the shoot growth that was partially stimulated by lower doses of EDU. Phytotoxic symptoms on the leaves, attributable to EDU, were observed at concentrations above 200 mg litre(-1), but complete protection from visible O3-injury was provided by a single application of EDU at a concentration as low as 100 mg litre(-1). Significant interactions on growth characters measured between O3-exposure and EDU application were observed only in one of the substrates. While these results demonstrate the need for careful dose-response studies prior to field assessments, they also provide evidence of a dosage that is effective in protecting radish from O3 damage without interfering with plant growth itself.  相似文献

The complete aluminium speciation of surface waters from an acidified catchment, during dry weather and during storm episodes, is obtained. For the first time, estimates of adsorbed aluminium (Al(ad)) are reported together with other aluminium species. Al(ad) is calculated from the difference between monomeric aluminium content before and after separation of suspended solids by filtration (0.015 microm pore diameter). This fraction was very significant (Al(ad) >; 40 microg litre(-1)) for samples with high suspended solids content (ss >; 20 mg litre(-1)) collected during storm episodes. Plots of pAl(3+) versus pH for samples collected during dry weather are approximated by a straight line, close to the solubility line of synthetic gibbsite, and have a slope of 2.5, which suggests that these samples were saturated with respect to gibbsite. The cloud of pAl(3+) versus pH points for samples from storm episodes has a near zero slope, and shows a significant degree of undersaturation with respect to synthetic gibbsite. Various mechanisms of aluminium mobilisation are discussed.  相似文献

The sulphite and sulphate concentrations in weathering products of limestone and in wet and total deposition samples were measured by the modified West-Gaeke method and by ion chromatography. The sulphite content in the weathering crust and in the runoff rainwater of two historical buildings in Belgium, was always much lower than the sulphate content. The maximum sulphite to sulphate ratio was 2.5%. The sulphite concentration in wet and total deposition samples was in the low or sub ppm range except during two misty periods when the sulphite concentration was 6.8 and 21 mg litre(-1). In those samples a high sulphate concentration was also found (nl. 57 and 137 mg litre(-1), respectively).  相似文献

Trout farm effluents: characterization and impact on the receiving streams   总被引:1,自引:0,他引:1  
Effluents from three rainbow trout (Oncorhynchus mykiss) farms located in Northern Portugal were characterized and their impact on the receiving streams was evaluated. Mean fish productions in the studied fish farms were 15, 55 and 500 t of trout per year, respectively. The feeding water was abstracted from Fornelo, Inha and Coura Rivers, at flow rates ranging from 1.2 (15 t year(-1) fish farm) to 4.8 litre s(-1) per ton annual fish production (500 t year(-1) fish farm). As the water flows through the farms, net variations in the chemical characteristics were observed: a mean reduction in the dissolved oxygen (DO) concentration between 0.7 and 2.4 mg litre(-1); mean increases between 1.9 and 3.2 mg CaCO3 litre(-1) for total alkalinity, between 0.9 and 14 mg litre(-1) for BOD5, between 0.27 and 1.46 mg litre(-1) for ammonia nitrogen (NH4-N), between 0.060 and 0.579 mg litre(-1) for soluble phosphorus (PO4-P) and less than 16 mg litre(-1) for suspended solids; variations in the pH value and nitrate nitrogen concentration were not statistically significant (p<0.05). At the 500 t year(-1) fish farm it was also possible to detect net increases of total hardness (3.2 mg CaCO3 litre(-1)), electric conductivity (19 mS cm(-1)) and permanganate value (3.6 mgO2 litre(-1)). At the other farms net variations in these parameters were not significant. Net mass flow variations reported to the annual fish production are presented. The DO mass flow decreased, on average, between 255 and 549 g t(-1) of fish per day. The mean daily BOD5 increase ranged from 353 to 1510 g t(-1) of fish. The corresponding ranges for the other parameters were 105-157 g t(-1) for NH4-N, 24-62 g t(-1) for PO4-P, 348-1035 g CaCO3 t(-1) for total alkalinity and 224 x 10(6)-506 x 10(6) t(-1) for mesophilic bacteria. Daily net variations of suspended solids, total hardness, electric conductivity and permanganate value were below 1753 g t(-1), 342 g CaCO3 t(-1), 2081 mS cm t(-1) and 392 gO2 t(-1), respectively. Longitudinal concentration profiles for the most relevant parameters show the impact of the effluent discharges on the physico-chemical and bacteriological river water quality downstream from the trout farms. Analyzing the situations from a purely chemical point of view, the polluted stretches were 3, 5 and 12 km long downstream from the effluent discharges, respectively. Algae are exposed to elevated levels of heavy metals in water bodies generally for a long-term, and occasionally for a short-term duration. The present study deals with oxidative stress in Scenedesmus sp., commonly found in nutrient-rich freshwaters, during short- (6h) and long-term (7d) exposure to Cu(2+) and Zn(2+). The cells accumulated almost 2- and 4-times more Cu(2+) and Zn(2+) inside the cells during long-term than during short-term exposure to these metals. But the data on photosynthetic O(2) evolution and cell viability suggest that Scenedesmus sp. experienced lesser metal stress in long-term than in short-term experiment. Although malondialdehyde content was slightly higher in the long-term experiment, the amount produced by one unit intracellular metal was significantly lower than that in the short-term experiment. Superoxide dismutase activity of Scenedesmus sp. was >30% higher during long-term than during short-term exposure to Cu(2+) and Zn(2+). But, catalase and ascorbate peroxidase activities increased only at 2.5 microM Cu(2+) and 25 microM Zn(2+) when oxidative stress was mild, but were inhibited at 10 microM Cu(2+) under intense oxidative stress. Cu(2+) and Zn(2+) reduced glutathione reductase activity and total SH content of Scenedesmus sp. in both the experiments, with greater reduction occurring in the long-term experiment. The depletion of total thiol was positively related with the intracellular level of metals. Thiols might have helped Scenedesmus sp. in overcoming metal-induced oxidative stress, but depletion of thiol pool is known to make cells vulnerable to oxidative stress. The study suggests that antioxidant enzymes play a role only under mild oxidative stress. An increased accumulation of proline seems to be an important strategy for alleviating metal-induced oxidative stress in Scenedesmus sp. The study shows that Scenedesmus sp. could acclimatize during long-term exposure to toxic concentrations of the test metals.  相似文献

Perna viridis is an ideal animal for studying the impairment caused by the effects of heavy metals that are often detected in coastal areas. Preliminary bioassay tests revealed that the lethal (LC(100)), median lethal (LC(50)) and sublethal (LC(0)) concentration of silver and chromium to P. viridis were 6.5, 4.0, 2.0 mg l(-1) and 4.5, 2.5, 1.0 mg l(-1), respectively. Toxic effect of silver and chromium was evaluated in the green mussel P. viridis, with reference to oxygen consumption, filtration rate and ATPase system in laboratory experiments. These parameters were selected as the end point of sublethal stress. Oxygen consumption and filtration rates were calculated as a measure of decline in the dissolved oxygen level and algal concentration (feed) in the aquaria water, respectively. Silver and chromium affects both oxygen consumption and filtration rate significantly (P<0.01) at 96 h when compared to control. The activity of ATPases system in the gills, hepatopancreas, ovary and muscle of mussels were inhibited by silver and chromium indicating that metals exerted significant toxic effect. The inhibition of Na(+)K(+) ATPase, Ca(2+) ATPase and Mg(2+) ATPase in the mussels were significant (P<0.05) for silver and highly significant (P<0.01) for chromium, which indicates that chromium was more toxic to mussels when compared to silver. The assessment of oxygen consumption, filtration and ATPases system can thus be used as a valid biomarker in aquatic ecotoxicology studies.  相似文献

The acute toxicity of phenol and ammonia, singly and in combination, to larvae of the ephemeropteran, Baetis rhodani, was examined using a computerized continuous flow system in the laboratory. The 24 h LC50 values (with 95% confidence intervals) were calculated to be 29.9 (17.3-51.5) mg phenol litre(-1) and 8.2 (2.0-33.0) mg un-ionized ammonia litre(-1). When phenol and ammonia were together in low concentration (< 20 mg litre(-1) and < 3 mg litre(-1) [un-ionized], respectively), they expressed their toxicity additively, but at higher concentrations they behaved in a greater-than-additive manner. When the 24 h LC50 values were used to predict the toxicity of a coking plant effluent, containing principally ammonia and phenol, it was found that the B. rhodani larvae died quicker than expected indicating the presence of other toxic chemicals. This exemplifies the value of using direct toxicity assessments to detect the presence of unknown toxicants.  相似文献
