This experiment examined the effects of sulfate (S) and reduced glutathione (GSH) on arsenic uptake by arsenic hyperaccumulator Pteris vittata after exposing to arsenate (0, 15 or 30 mg As L−1) with sulfate (6.4, 12.8 or 25.6 mg S L−1) or GSH (0, 0.4 or 0.8 mM) for 2-wk. Total arsenic, S and GSH concentrations in plant biomass and arsenic speciation in the growth media and plant biomass were determined. While both S (18-85%) and GSH (77-89%) significantly increased arsenic uptake in P. vittata, GSH also increased arsenic translocation by 61-85% at 0.4 mM (p < 0.05). Sulfate and GSH did not impact plant biomass or arsenic speciation in the media and biomass. The S-induced arsenic accumulation by P. vittata was partially attributed to increased plant GSH (21-31%), an important non-enzymatic antioxidant countering oxidative stress. This experiment demonstrated that S and GSH can effectively enhance arsenic uptake and translocation by P. vittata.  相似文献

Pteris vittata plants were grown on twenty-one UK soils contaminated with arsenic (As) from a wide range of natural and anthropogenic sources. Arsenic concentration was measured in fern fronds, soil and soil pore water collected with Rhizon samplers. Isotopically exchangeable soil arsenate was determined by equilibration with 73AsV. Removal of As from the 21 soils by three sequential crops of P. vittata ranged between 0.1 and 13% of total soil As. Ferns grown on a soil subjected to long-term sewage sludge application showed reduced uptake of As because of high available phosphate concentrations. A combined solubility-uptake model was parameterised to enable prediction of phytoremediation success from estimates of soil As, ‘As-lability’ and soil pH. The model was used to demonstrate the remediation potential of P. vittata under different soil conditions and with contrasting assumptions regarding re-supply of the labile As pool from unavailable forms.  相似文献

Increasing availability of soil arsenic is of significance for accelerating phytoremediation efficiency of As-polluted sites. The effects of seven amendments, i.e., citrate, oxalate, EDTA, sodium polyacrylate (SPA), phosphate rock (PR), single superphosphate (SSP), and compost on fractionation and phytoavailability of soil As were investigated in lab culture experiment. The results showed that the addition of PR, SPA, EDTA or compost to soils significantly increased the concentration of NaHCO3-extractable As over a 120 d incubation period compared with the control (amendment-free) soil. Then, the four amendments were selected to add to As-contaminated soil growing Pteris vittata. It was concluded that As accumulation by the fern increased significantly under the treatments of PR and SPA by 25% and 31%, respectively. For As fractionation in soil, SPA increased Fe-As significantly by 51% and PR increased Ca-As significantly by 18%, while both the two amendments reduced occluded-As by 16% and 19%, respectively. Adding PR and SPA in soil increased the activities of urease and neutral phosphatase resulting from the improvement the fertility and physical structure of the soil, which benefits plant growth and As absorption of P. vittata. The results of the research revealed that both PR and SPA were effective amendments for improving phytoremediation of As-contaminated sites by P. vittata.  相似文献

The frequent co-existence of arsenic (As) and lead (Pb) necessitates the investigation of clean-up technologies for multi-metal(loid)s. Field survey and hydroponic experiments were conducted to elucidate the co-accumulation of As and Pb in Pteris vittata L. The P. vittata population isolated from a Pb–Zn mine in Yunnan province, China is a potential extractor of As and Pb co-contamination. Hydroponic experiment found that the highest frond As and Pb concentrations in mining population of P. vittata reached 12.2 and 0.99 g kg?1, respectively. The interaction between As and Pb in P. vittata was further more disclosed. Pb (2 mg L?1) improved the frond As concentration by 60 to 150 % in mining populations of P. vittata. Micro-X-ray absorption spectroscopy indicated that under the combined exposure of As and Pb, the As content in the rhizoid epidermis increased by about 10-fold, and the As(V) percentage increased in each rhizoid tissue, as compared with that under As exposure alone. The co-absorption of As and Pb on the epidermis and the enhanced transportation of As(V) from epidermis into the rhizoid were suggested to contribute to the increased As accumulation.  相似文献

The roles of low-molecular-weight thiols (LMWTs), such as glutathione and phytochelatins, in arsenic (As) tolerance and hyperaccumulation in Pteris vittata an As-hyperaccumulator fern remain to be better understood. This study aimed to thoroughly characterize LMWT synthesis in P. vittata to understand the roles played by LMWTs in As tolerance and hyperaccumulation. LMWT synthesis in P. vittata was induced directly by As, and not by As-mediated oxidative stress. Expression of PvECS2, one of the putative genes of γ-glutamylcysteine synthetase (γECS), increases in P. vittata shoots at 48 h after the onset of As exposure, almost corresponding to the increase in the concentrations of γ-glutamylcysteine and glutathione. Furthermore, localization of As showed similar trends to those of LMWTs in fronds at both whole-frond and cellular levels. This study thus indicates the specific contribution of LMWTs to As tolerance in P. vittata. γECS may be responsible for the As-induced enhancement of LMWT synthesis.  相似文献

Arsenic, Pb and Zn tolerance and accumulation were investigated in six populations of Pteris vittata collected from As-contaminated and uncontaminated sites in southeast China compared with Pteris semipinnata (a non-As hyperaccumulator) in hydroponics and on As-contaminated soils. The results showed that both metallicolous and nonmetallicolous population of P. vittata possessed high-level As tolerance, and that the former exhibited higher As tolerance (but not Pb and Zn tolerance) than the latter. In hydroponic culture, nonmetallicolous population clearly showed significantly higher As concentrations in fronds than those in metallicolous populations. In pot trials, As concentrations in fronds of nonmetallicolous population ranged from 1060 to 1639 mg kg?1, about 2.6- to 5.4-folds as those in metallicolous populations. It was concluded that As tolerance in P. vittata resulted from both constitutive and adaptive traits, Pb and Zn tolerances were constitutive properties, and that nonmetallicolous population possesses more effective As hyperaccumulation than metallicolous populations.  相似文献

Impacts of diazinon (O,O-diethyl O-2-isopropyl-6-methylpyrimidin-4-yl phosphorothioate), imidacloprid [1-(6-chloro-3-pyridylmethyl)-N-nitroimidazolidin-2-ylideneamine] and lindane (1,2,3,4,5.6-hexachlorocyclohexane) treatments on ammonium, nitrate, and nitrite nitrogen and nitrate reductase enzyme activities were determined in groundnut (Arachis hypogaea L.) field for three consecutive years (1997 to 1999). Diazinon was applied for both seed- and soil-treatments but imidacloprid and lindane were used for seed treatments only at recommended rates. Diazinon residues persisted for 60 days in both the cases. Average half-lives (t1/2) of diazinon were found 29.3 and 34.8 days respectively in seed and soil treatments. In diazinon seed treatment, NH4 +, NO3 ?, and NO2 ? nitrogen and nitrate reductase activity were not affected. Whereas, diazinon soil treatment indicated significant increase in NH4 +-N in a 1-day sample, which continued until 90 days. Some declines in NO3 ?N were found from 15 to 60 days. Along with this decline, significant increases in NO2 ?N and nitrate reductase activity were found between 1 and 30 days. Imidacloprid and lindane persisted for 90 and 120 days with average half-lives (t1/2) of 40.9 and 53.3 days, respectively. Within 90 days, imidacloprid residues lost by 73.17% to 82.49% while such losses for lindane residues were found 78.19% to 79.86 % within 120 days. In imidacloprid seed-treated field, stimulation of NO3 ?N and the decline in NH4 +NO2 ?-N and nitrate reductase enzyme activity were observed between 15 to 90 days. However, lindane seed treatment indicated significant increases in NH4 +-N, NO2 ?-N and nitrate reductase activity and some adverse effects on NO3 ?N between 15 and 90 days.  相似文献

AsIII uptake in living cells is through aquaglyceroporin transporters, but it is unknown in arsenic-hyperaccumulator Pteris vittata. We investigated the effects of AsIII analogs glycerol and antimonite (SbIII) at 0-100 mM and aquaporin inhibitor AgNO3 at 0-0.1 mM on the uptake of 0.1 mM AsIII or AsV by P. vittata over 1-2 h. Glycerol or SbIII didn’t impact AsIII or AsV uptake by P. vittata (p < 0.05), with As concentrations in the fronds and roots being 4.4-6.3 and 3.9-6.2 mg/kg. However, 0.01 mM AgNO3 reduced As concentrations in the fronds and roots by 64% and 58%. Hence, AsIII uptake in P. vittata might be via an aquaporin transporter different from glycerol and SbIII transporters. Further as AsIII analogs and aquaporin inhibitor had no impact on AsV uptake, AsIII and AsV were likely taken up by different transporters in P. vittata. Our results imply a different AsIII transporter in P. vittata from other plants.  相似文献

Ecological remediation is an important measure for the protection of lake water quality in removing nutrients, such as nitrate (NO3 ?). In this study, four bioremediation processes (bare sediment, immobilized nitrogen cycling bacteria (INCB) added, Elodea nuttallii added, E. nuttallii-INCB assemblage) were operated at a lab to elucidate the effect of macrophyte appearance and INCB addition on NO3 ? removal and achieve the optimal processes for biomediation. 15?N-NO3 solution was added to microcosms to identify the key nitrogen transformation processes responsible for NO3 ? removal. Results showed that nitrate removal was significantly enhanced after the addition of INCB and E. nuttallii. In the treatments with INCB added, E. nuttallii added, and INCB and E. nuttallii-INCB assemblage, nitrate removal ratio achieved 94.74, 98.76, and 99.15 %, respectively. In contrast, only 23.47 % added nitrate was removed in the control. Plant uptake and denitrification played an important role in nitrogen removal. The water quality was substantially improved by the addition of INCB and macrophyte that can accelerate denitrification and promote nitrogen assimilation of plants. The results indicated that plant uptake and microbial denitrification were key processes for nitrate removal.  相似文献

Arsenic (As) accumulation in food crops such as rice is of major concern. To investigate whether phytoremediation can reduce As uptake by rice, the As hyperaccumulator Pteris vittata was grown in five contaminated paddy soils in a pot experiment. Over a 9-month period P. vittata removed 3.5-11.4% of the total soil As, and decreased phosphate-extractable As and soil pore water As by 11-38% and 18-77%, respectively. Rice grown following P. vittata had significantly lower As concentrations in straw and grain, being 17-82% and 22-58% of those in the control, respectively. Phytoremediation also resulted in significant changes in As speciation in rice grain by greatly decreasing the concentration of dimethylarsinic acid (DMA). In two soils the concentration of inorganic As in rice grain was decreased by 50-58%. The results demonstrate an effective stripping of bioavailable As from contaminated paddy soils thus reducing As uptake by rice.  相似文献

The aims of this study are to investigate whether and how the nitrogen form (nitrate (NO3 ) versus ammonium (NH4 +)) influences cadmium (Cd) uptake and translocation and subsequent Cd phytoextraction by the hyperaccumulator species Sedum plumbizincicola. Plants were grown hydroponically with N supplied as either NO3 or NH4 +. Short-term (36 h) Cd uptake and translocation were determined innovatively and quantitatively using a positron-emitting 107Cd tracer and positron-emitting tracer imaging system. The results show that the rates of Cd uptake by roots and transport to the shoots in the NO3 treatment were more rapid than in the NH4 + treatment. After uptake for 36 h, 5.6 (0.056 μM) and 29.0 % (0.290 μM) of total Cd in the solution was non-absorbable in the NO3 and NH4 + treatments, respectively. The local velocity of Cd transport was approximately 1.5-fold higher in roots (3.30 cm h?1) and 3.7-fold higher in shoots (10.10 cm h?1) of NO3 - than NH4 +-fed plants. Autoradiographic analysis of 109Cd reveals that NO3 nutrition enhanced Cd transportation from the main stem to branches and young leaves. Moreover, NO3 treatment increased Cd, Ca and K concentrations but inhibited Fe and P in the xylem sap. In a 21-day hydroponic culture, shoot biomass and Cd concentration were 1.51 and 2.63 times higher in NO3 - than in NH4 +-fed plants. We conclude that compared with NH4 +, NO3 promoted the major steps in the transport route followed by Cd from solution to shoots in S. plumbizincicola, namely its uptake by roots, xylem loading, root-to-shoot translocation in the xylem and uploading to the leaves. S. plumbizincicola prefers NO3 nutrition to NH4 + for Cd phytoextraction.  相似文献

A field survey was conducted to evaluate soil metal pollution and endogenous trans-zeatin content in the leaves of plants growing at six sites in a metal-polluted area located in Gejiu, Yunnan, China. Five plant species were collected, and the physicochemical properties and concentrations of five metals in the soil were analyzed. The trans-zeatin content in plant leaves was measured by high-performance liquid chromatography. Based on the Nemerow pollution index, the six sites were classified into four levels of pollution (i.e., low, medium, high, and severely high). The degree of soil metal pollution was cadmium (Cd) > arsenic (As) > lead (Pb) > zinc (Zn) > copper (Cu). The leaf trans-zeatin content in Pteris vittata (an arsenic hyperaccumulator) increased significantly by 98.6 % in soil with a severely high level of pollution compared with soil at a low level of pollution. However, in non-hyperaccumulators Bidens pilosa var. radiata and Ageratina adenophora, a significant decrease in leaf trans-zeatin content of 35.6 and 87.6 %, respectively, was observed. The leaf trans-zeatin content in Artemisia argyi also decreased significantly by 73.6 % in high metal-polluted soil compared with that in medium metal-polluted soil. Furthermore, significant correlations were observed between leaf trans-zeatin content in Pteris vittata and As, Pb, and Cd concentrations in the soil; however, either no correlation or a negative one was observed in the other plant species. Therefore, a high content of trans-zeatin in the leaves of Pteris vittata may play an important role in its normal growth and tolerance to metals.  相似文献

The effect of arsenic (25 and 50 μM As for 1 and 5 d) was analysed in wild type (WT) and Arabidopsis thaliana (L.) Heynh plants deficient in NADPH oxidase C (AtrbohC). The content of H2O2 and malondialdehyde (MDA) increased with the As concentration, while the opposite effect was found for NO in WT and AtrbohC plants. The As treatment reduced catalase and increased glutathione reductase activities to the same extent in WT and AtrbohC plants, although the induction of all SOD isoforms (mainly CuZn–SODs) was observed in WT plants, the opposite effects being found in AtrbohC plants. Glycolate oxidase (H2O2 producers) considerably increased with the concentration and time of treatment with As in WT and AtrbohC mutants. Arsenic induced the uptake and translocation of P, S, Cu, Zn, and Fe in WT plants, while in AtrbohC plants the opposite trend was noted and the uptake of As became considerably lower than in WT plants. These results suggest that As causes oxidative stress by inducing glycolate oxidase, while NADPH oxidase does not appear to participate in ROS overproduction but could be critical in regulating antioxidant defences as well as the transport and translocation of As and macro/micronutrients.  相似文献

Salmonella and Escherichia coli are two bacteria that are important causes of human and animal disease worldwide. Chlorate is converted in the cell to chlorite, which is lethal to these bacteria. An HPLC procedure was developed for the rapid analysis of chlorate (ClO3 ?), nitrate (NO3 ?), and nitrite (NO2 ?) ions in bovine ruminal fluid samples. Standard curves for chlorite, nitrite, nitrate, and chlorate were well defined linear curves with R2 values of 0.99846, 0.99106, 0.99854, and 0.99138, respectively. Concentrations of chlorite could not be accurately determined in bovine ruminal fluid because chlorite reacts with or binds a component(s) or is reduced to chloride in bovine ruminal fluid resulting in low chlorite measurements. A standard curve ranging from 25 to 150 ppm ClO3 ? ion was used to measure chlorate fortified into ruminal fluid. The concentration of chlorate was more rapidly lowered (P < 0.01) under anaerobic compared to aerobic incubation conditions. Chlorate alone or chlorate supplemented with the reductants sodium lactate or glycerol were bactericidal in anaerobic incubations. In anaerobic culture, the addition of sodium formate to chlorate-fortified ruminal fluid appeared to decrease chlorate concentrations; however, formate also appeared to moderate the bactericidal effect of chlorate against E. coli. Addition of the reductants, glycerol or lactate, to chlorate-fortified ruminal fluid did not increase the killing of E. coli at 24 h, but may be useful when the reducing equivalents are limiting as in waste holding reservoirs or composting systems required for intense animal production.  相似文献

Nitrate (NO3 ?) is present in aquatic ecosystems as a natural component of the nitrogen cycle. However, in the last decades, several human activities are the causes of the rising amounts of organic matter and inorganic nitrogen nutrients in aquatic ecosystems, causing notable increase of nitrate above background natural levels. In spite of the toxicity of nitrate to aquatic animals, there are relatively few studies on the chronic toxicity of this compound to invertebrates. The aim of our study is to assess the effect of chronic (35 days) exposure to nitrate on the behaviour (velocity of movement) and reproduction (number of newborns) of the aquatic snail Potamopyrgus antipodarum. Four actual concentrations of nitrate were used (21.4, 44.9, 81.8 and 156.1 mg?N-NO3 ?/L). In each treatment, 12 animals were individually monitored for velocity (weekly) and newborn production (every 3–4 days). Velocity was recorded using quantitative video monitoring. Our results showed that nitrate did not cause mortality, but it reduced the velocity of movement (at 44.9, 81.8 and 156.1 mg?N-NO3 ?/L) and number of live newborns (in all tested concentrations). Reproductive impairment was caused at realistic nitrate concentrations which is relevant to the risk assessment of this compound. Our study contributes to the knowledge of the chronic effects of nitrate on the behaviour and reproduction of an aquatic snail.  相似文献

The interactions of arsenic and phenanthrene on plant uptake and antioxidative response of Pteris vitatta L. were studied hydroponically. The combination of arsenic and phenanthrene decreased arsenic contents in fronds by 30-51%, whereas increased arsenic concentrations 1.2-1.6 times in roots, demonstrating the suppression of arsenic translocation compared to the corresponding treatment without phenanthrene. Under the co-exposure, As(III) concentrations in fronds deceased by 12-73%, and at higher arsenic exposure level (≥10 mg/L), As(V) in fronds and As(III) in roots increased compared to the single arsenic treatment. Arsenic exposure elevated phenanthrene concentrations in root by 39-164%. The co-existence of arsenic and phenanthrene had little impact on plant arsenic accumulation, although synergistic effect on antioxidants was observed, suggesting the special physiological process of P. vitatta in the co-exposure and application potential of P. vitatta in phytoremediation of arsenic and PAHs co-contamination.  相似文献

We assessed the association between arsenic intake through water and diet, and arsenic levels in first morning-void urine under variable conditions of water contamination. This was done in a 2-year consecutive study in an endemic population. Exposure of arsenic through water and diet was assessed for participants using arsenic-contaminated water (≥50 μg L?1) in a first year (group I) and for participants using water lower in arsenic (<50 μg L?1) in the next year (group II). Participants with and without arsenical skin lesions were considered in the statistical analysis. Median dose of arsenic intake through drinking water in groups I and II males was 7.44 and 0.85 μg kg body wt.?1 day?1 (p <0.0001). In females, it was 5.3 and 0.63 μg kg body wt.?1 day?1 (p <0.0001) for groups I and II, respectively. Arsenic dose through diet was 3.3 and 2.6 μg kg body wt.?1 day?1 (p?=?0.088) in males and 2.6 and 1.9 μg kg body wt.?1 day?1 (p?=?0.0081) in females. Median arsenic levels in urine of groups I and II males were 124 and 61 μg L?1 (p?=?0.052) and in females 130 and 52 μg L?1 (p?=?0.0001), respectively. When arsenic levels in the water were reduced to below 50 μg L?1 (Indian permissible limit), total arsenic intake and arsenic intake through the water significantly decreased, but arsenic uptake through the diet was found to be not significantly affected. Moreover, it was found that drinking water mainly contributed to variations in urine arsenic concentrations. However, differences between male and female participants also indicate that not only arsenic uptake, but also many physiological factors affect arsenic behavior in the body and its excretion. As total median arsenic exposure still often exceeded 3.0 μg kg body wt.?1 day?1 (the permissible lower limit established by the Joint Expert Committee on Food Additives) after installation of the drinking water filters, it can be concluded that supplying the filtered water only may not be sufficient to minimize arsenic availability for an already endemic population.  相似文献

In a hydroponic culture, experiments were performed to study the influence of potassium (K) supplementation (0, 20, 40, 60, 80, and 100 mg L?1) on the arsenic (As; 0, 8, and 10 mg L?1)-accrued changes in growth traits (plant biomass, root–shoot length) and the contents of lepidine, As and K, in garden cress (Lepidium sativum Linn.) at 10 days after treatment. The changes in these traits were correlated with shoot proline content, protein profile, and the activities of antioxidant enzymes namely superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), glutathione reductase (GR, EC 1.8.1.7), and ascorbate peroxidase (APX, EC 1.11.1.11). In general, As-alone treatments significantly decreased the growth traits but lead to significant enhancements in shoot proline and enzyme activities. K-supplementation to As-treated L. sativum seedlings decreased shoot-As content, reduced As-induced decreases in growth traits but enhanced the content of shoot proline, and the activities of the studied enzymes maximally with K100 + As8 and As10 mg L?1. Both 8 and 10 mg L?1 of As drastically downregulated the shoot proteins ranging from 43–65 kDa. With As10 mg L?1, there was a total depletion of protein bands below 23 kDa; however, K80 mg L?1 maximally recovered and upregulated the protein bands. Additionally, protein bands were downregulated (at par with As-alone treatment) above K80 mg L?1 level. Interestingly, As-stress increased lepidine content in a dose-dependent manner which was further augmented with the K-supplementation. It is suggested that K protects L. sativum against As-toxicity by decreasing its accumulation and strengthening antioxidant defense system and protein stability.  相似文献
