Identification of a sex pheromone from a rotifer   总被引:7,自引:0,他引:7  
The first isolation and characterization of a sex pheromone from a zooplankter is reported. A 29 kdalton glycoprotein on the surface of females of the marine rotifer Brachionus plicatilis acts as a contact-mating pheromone. This glycoprotein (gp29) is glycosylated with oligosaccharides containing Nacetylglucosamine, mannose, and fucose residues, and these oligosaccharides are necessary for male recognition of females. Males detect this signal by contact chemoreception with receptors located in their corona. Binding of purified glycoprotein to male receptors reduces mating attempts by 93%. An antibody to the glycoprotein binds to females, reducing male mating attempts by 86%. When purified gp29 is bound to sepharose beads, it is sufficient to elicit male mating behavior. This glycoprotein is likely to play a key role in the evolution and maintenance of reproductive isolation in rotifers.  相似文献

Eutrophication caused by the enrichment of nutrients from diffusing sources is degrading surface water quality throughout the world. Assessing the potential contributions of different land areas in diffuse nutrient export has become an important task in non-point source (NPS) pollution control. Existing methods were often limited by the availability of local data and the complexity of model formulation. This study developed a spatial multicriteria method to evaluate the nitrogen loss potential at the basin level. Four criteria were formulated to characterize the source capacity of nitrogen export, the flow path to water body, the efficiency of runoff generation and the climatic driving force. The proposed method is a low-effort approach since the required data is either already available in a global context or easily produced with limited inputs. Being implemented in GIS environment, this method generates maps that can be easily interpreted to provide decision support. The method was applied to the Huai River Basin, China. The results were validated based on the correlation between the nitrogen loss potential of sub-basin and the water quality class of river. The maps of nitrogen loss potential were helpful for examining the regional pattern of diffuse nitrogen loss, and could facilitate the decisions of NPS pollution management at the provincial or basin level.  相似文献

Olfactory chemical cues have been described to play important roles in the control of mate recognition in many decapod crustaceans. However, we still know very little about the chemical characteristics of the cues that coordinate pre-copulative behaviour. In this study, we partially characterized a waterborne sex pheromone of a marine shrimp, Lysmata wurdemanni. Female moulting water was collected and ultrafiltered using 1,000 and 500-Da membranes, respectively, and analysed using HPLC with a Lichrosphere™ RP18 (C18) column. The sex pheromone is likely to be a molecule between 500 and 1,000 Da in size because behavioural bioassays showed that males responded to the supernatant of 500-Da and to the 1,000-Da filtrate, but did not respond to the supernatant of 1,000-Da or to the 500-Da filtrate. There was only one dominant peak (2.86 min) detectable in HPLC chromatograms of the supernatant of the 500-Da filtration. This peak showed a UV absorbance maximum at 274 nm, similar to the recently identified shore crab sex pheromone Uridine-di-phosphate (UDP). Behavioural bioassays confirmed that this peak is a bioactive component of a potential pheromone bouquet, but is different from UDP, which showed no bioactivity in Lysmata wurdemanni. Our results lay the foundation for future studies to purify and eventually identify this sex pheromone.  相似文献

There is a demand for the development of environmental friendly methods for the synthesis of graphene composites. Reduced graphene oxide/silver (RGO/Ag) nanocomposites are very good catalysts. Here, we propose a simple, green method for the synthesis of RGO/Ag nanocomposite using the amino acid tyrosine as bioreductant and stabilizing agent. RGO/Ag nanocomposite was characterized by using various analytical techniques and studied for its catalytic degradation of 4-nitrophenol. Results of attenuated total reflectance Fourier transform infrared spectroscopy and Zeta potential at ?55 mV reveal the surface capping of tyrosine onto the reduced graphene oxide nanosheets. RGO/Ag nanocomposites show excellent catalytic reduction of 4-nitrophenol with NaBH4, when compared to actual individual silver nanoparticles.  相似文献

Few studies have studied the food supply to larval fish in the Irish Sea; thus, we have assessed the full prey-field available to larval fish, ranging from protozoa to copepods. Specifically we assessed if fish larvae feed on protozoa, as suggested by others, and if densities of the protozoa and the appropriate size of metazoan prey were previously underestimated. By examining the gutcontents of fish larvae, the prey available to them, and the potential accessibility of prey to fish, we develop a simple food web, presented as a box-model. By doing so, we indicate that the lack of focus on small metazoa and protozoa has underestimated the food available to fish larvae; without these, we might have concluded that prey levels were too low to support the growth of the larval fish assemblage. Our methods were as follows. Sampling was at two sites, off the Isle of Man, with distinct physical and biological structures, soon after fish spawning: the southwest coast, where many species occur in spring-summer (23 April; 6, 19 May; 1, 22 June; 12 July) and the east coast, where only herring larvae occur in September–November (12, 28 October). Microplankton (15–200 μm), mesozooplankton, and larval fish were collected at 1, 15, and 25 m: microplankton with 1.5 L bottles and a 64 μm-mesh net; mesozooplankton and larval fish with a Gulf VII high-speed sampler (280 μm mesh). The 64 μm mesh net, mounted on the Gulf VII, provided simultaneous hauls. Fixed samples were evaluated to determine species composition, abundance, and biomass. Larval fish diet was determined from fish collected by short net hauls: fixed guts were examined and prey, including protozoa, analysed. Using physical data as a guide, plankton data were integrated through the water column to determine standing stocks. Size-based food availability to larval fish was estimated from the gut contents. The role of protozoa was examined, assuming that they are digested at the same rate as metazoan and if they are digested 2.5–10 times faster; increased digestion rates indicated that they contributed substantially to the larval fish diet.  相似文献

In recent years, Conogethes pluto (Lepidoptera: Crambidae) has become a major pest of Alpinia and other ornamental gingers in the Northern Territory and Queensland, Australia. This pest damages the flowers and bores into the stems, causing substantial losses to production. Currently, no synthetic sex pheromone is available to monitor or control this pest. This work aims at the identification of the sex pheromone of this pest. Analysis of the sex pheromone gland of female C. pluto by gas chromatography/electroantennogram detector revealed the presence of seven candidate pheromone compounds that elicited electroantennogram responses. Using gas chromatography/mass spectrometry analysis and micro-derivatization reactions, six compounds were identified as (E)-10-hexadecenal, as the main pheromone compound, (Z)-10-hexadecenal, hexadecanal, (E)-10-hexadecen-1-ol, (10E,12E)-hexadeca-10,12-dienal and (3Z,6Z,9Z)-tricosa-3,6,9-triene as minor pheromone compounds. In two-field trapping experiments, C. pluto responded to the six-component blend, and three of six compounds, i.e., (E)-10-hexadecenal, (3Z,6Z,9Z)-tricosa-3,6,9-triene, and (10E,12E)-hexadeca-10,12-dienal were shown to be necessary for attraction. In a subsequent experiment testing various doses (i.e., 0.01, 0.1, and 1 mg) of the six-component blend, the largest number of males was captured in traps baited with a lure loading of 1 mg. The availability of the sex pheromone of C. pluto will enable monitoring and provides the basis for additional control options for this pest.  相似文献

The stable isotope ratio 12C/13C was used to investigate the source of carbon in free-living barnacles and in coral-inhabiting barnacles from the Red Sea. The δ13C of most of the barnacles collected on the open shore ranges between −17.5 and −19.7‰, indicating relative enrichment of light carbon originating from the open-sea phytoplankton. Those collected in closed habitats showed heavier isotopic composition. The δ13C of the coral-inhabiting barnacles ranges from −14.1 to −16.7‰, suggesting that the carbon contribution of open-sea plankton to these barnacles is less important than it is to free-living barnacles. We hypothesize that coral organic matter and zooxanthellae expelled by the host coral contribute carbon to the barnacle, and that a mixture of this relatively heavy carbon with carbon from other sources is responsible for the high values of δ13C in coral barnacles. Sexual selection by competition for mates is a formidable force that has led to extraordinary adaptations in males. Here we present results suggesting a novel case of pheromone mimicry in males of Lariophagus distinguendus, a parasitic wasp of beetle larvae that develop in stored grain. Females of L. distinguendus produce a pheromone even before they emerge from a grain. Males are attracted to the parasitised grain and wait for females to emerge. Males emerging later than others are under enormous selection pressure since females mate only once. We show evidence that developing males fool their earlier emerging competitors by mimicking the female pheromone. Males exposed to pupae of either sex exhibit typical courtship behaviour. Searching males are not only arrested by grains containing developing females but spend as much time on grains containing developing males. Hence, by distracting their competitors away from receptive females late males may increase their own chance to mate with these females. After emergence, males decompose the active compounds within 32 h probably to decrease molestation during their own search for mates. Chemical analyses of active pheromone extracts and bioassays using fractions demonstrate that the active compounds are among the cuticular hydrocarbons.  相似文献

Diversity in guilds of primary producers enhances temporal stability in provision of organic matter to consumers. In the Antarctic ecosystem, where temporal variability in phytoplankton production is high, sea ice contains a diatom and microbial community (SIMCO) that represents a pool of organic matter that is seasonally more consistent, although of relatively small magnitude. The fate of organic material produced by SIMCO in Antarctica is largely unknown but may represent an important link between sea ice dynamics and secondary production in nearshore food webs. We used whole tissue and compound-specific stable isotope analysis of consumers to test whether the sea ice microbial community is an important source of organic matter supporting nearshore communities in the Ross Sea. We found distinct gradients in delta13C and delta15N of SIMCO corresponding to differences in inorganic carbon and nitrogen acquisition among sites with different sea ice extent and persistence. Mass balance analysis of a suite of consumers demonstrated large fluxes of SIMCO into the nearshore food web, ranging from 5% to 100% of organic matter supplied to benthic species, and 0-10% of organic matter to upper water column or pelagic inhabitants. A delta13C analysis of nine fatty acids including two key biomarkers for diatoms, eicosapentaenoic acid (EPA, 20:5omega3), and docosahexaenoic acid (DHA, 22:6omega3), confirmed these patterns. We observed clear patterns in delta13C of fatty acids that are enriched in 13C for species that acquire a large fraction of their nutrition from SIMCO. These data demonstrate the key role of SIMCO in ecosystem functioning in Antarctica and strong linkages between sea ice extent and nearshore secondary productivity. While SIMCO provides a stabilizing subsidy of organic matter, changes to sea ice coverage associated with climate change would directly affect secondary production and stability of benthic food webs in Antarctica.  相似文献

The spatial development of a passive scalar plume is studied within the inhomogeneous turbulence of a boundary layer flow in a recirculating laboratory flume with a smooth bed. The source of the scalar is located flush with the bed, and the low-momentum source design is intended to simulate a diffusive-type scalar release. A weakly diffusive fluorescent dye is used as the scalar. Planar laser-induced fluorescence (PLIF) techniques were used to record the structure of the plume at a spatial resolution of 150 μm. The measured structure of the mean concentration field is compared to an analytical solution for shear-free, homogeneous turbulence. The laboratory plume exhibits spatial development in the mean concentration field that deviates from the self-similar behavior predicted by the analytical solution; this deviation is due to the mean shear and inhomogeneity of the turbulence. In particular, the influence of the viscous sublayer on the plume development is seen to be significant. Nonetheless, the analytical solution replicates some of the features seen in the laboratory plume, and the solution suggests methods of reducing the laboratory data even for cases where the results deviate from the analysis. We also examine the spatial development of the root-mean-square (rms) fluctuating concentration field, and use scalar probability density functions to examine the relationship between the mean and fluctuating concentrations.  相似文献

Insect pheromone traps are becoming an increasingly important tool in biosecurity and pest surveillance, alerting managers to the presence of unwanted organisms. To expand the role of these traps beyond their present sentinel role, it is necessary to develop reliable operational models of local insect dispersal. Following the detection of an insect incursion using a pheromone trap, such models could simulate the dispersal of the insect from its emergence site to the point of detection, enabling biosecurity managers to estimate the most likely proximal source of the incursion. An individual-based moth movement model was developed to simulate observed patterns of moth movement in response to the presence or absence of a pheromone. Using parameters derived from a genetic algorithm, it was possible to fit a model based on the three behavioural components (upwind, upwind with zigzags and casting) described in insect anemotaxis theory to a subset of observed movement patterns (0-135° to the wind), but not to the whole spectrum of movement patterns. It appears that current insect anemotaxis theory is missing a downwind flight component. Whilst the frequency of downwind movements is small; their ground speed could lead to significant downwind displacement, having a disproportionately strong influence on a moth movement model, and hence projections of the likely source or target locations.  相似文献

Results are recorded from an integrated hydrodynamical numerical model of the Irish Sea using, as boundary conditions, first current meter data and secondly sea elevations from the 1971 BISOP exercise. These data are utilised at time intervals of less than a tidal cycle.When current meter data were used as boundary conditions the comparison between the calculated results and observed current meter vectors, averaged over a tidal period, showed discrepancies in both magnitude and direction. This comparison was much improved when the northern open boundary conditions were replaced by sea elevations.The significance of these results to the process of modelling marine ecosystems is discussed.  相似文献

We evaluated the duration of Copepodite Stages C1 to C6, the biological cycle and the number of annual generations of the planktonic copepod Acartia clausi in a meso-oligotrophic area of the eastern Mediterranean Sea (Saronikos Gulf, Greece). The results were based on 95 zooplankton samples collected during the period November 1988 through June 1990, at intervals of 1, 2, 7 and 15 d, the sampling intervals being dependent on the abundance of A. clausi. Time-series analysis (cross-correlation) of fluctuations in the comparative abundance (percentages) of the copepodite stages present was used to determine the duration of the development stages and generation length. This methodology could significantly contribute to the identification of cohorts, and hence to the estimation of stage duration, from field data for a given copepod species. The development of A. clausi stages was not isochronal; duration of the first copepodite stage was shorter than that of the last three stages. The mean generation length estimated (28.6 d) is among the highest recorded in the literature for A. clausi at the range of temperatures prevailing in the area (13 to 25°C). Throughout the year there were four or five generations. The possible limiting role of food availability on the duration of each stage and hence on generation length is also discussed.  相似文献

Respiration and ammonia excretion rates of 19–24 euphausiids from the epipelagic through bathypelagic zones of the world’s oceans were compiled. Body mass (expressed in terms of dry mass, carbon or nitrogen), habitat temperature and sampling depth were designated as parameters in multiple regression analysis. Results suggested that the three parameters were highly significant, contributing 71–89 % of the variance in respiration rates and 69–81 % of the variance in ammonia excretion rates. Atomic O:N ratios derived from simultaneous measurements of respiration and ammonia excretion rates ranged from 11 to 90 (median: 27), and no appreciable effects of the three parameters on O:N ratios were detected. If global-bathymetric models for the metabolism and chemical composition of copepods and chaetognaths are compared with those of euphausiids, it becomes evident that euphausiids are unique in that they maintain high metabolic rates and accumulate moderate amounts of energy reserves (lipids).  相似文献

Volatiles from male hindgut extracts of a newly described spruce bark beetle, Ips shangrila Cognato and Sun, from different attack phases were analyzed by GC–MS/FID with both polar and enantioselective columns. The GC–MS/FID analyses showed that unmated males (Phase 1) or males mated with <3 females (Phases 2–4) produced 2-methyl-3-buten-2-ol and 99%-(+)-ipsdienol as major components, and (−)-cis-verbenol, (−)-trans-verbenol, myrtenol and 2-phenyl ethanol as minor or trace components. The release of these male-produced compounds was confirmed by the analysis on aeration sample of an I. shangrila infested wind-thrown spruce trunk. The quantities of 2-methyl-3-buten-2-ol, cis-verbenol and trans-verbenol from male hindgut extracts were almost unchanged or even slightly increased during gallery development, while ipsdienol decreased dramatically after mating with three females. No obvious Ips-related aggregation pheromone components were detected in the female hindgut extract. A field trapping bioassay in Qinghai, China, showed that the ternary blends containing 2-methyl-3-buten-2-ol, (−)-cis-verbenol and 97%-(+)-ipsdienol or (±)-ipsdienol, caught significantly more I. shangrila (♂:♀ = 1:2.14) than did the unbaited control. Replacing 97%-(+)-ipsdienol (close to the naturally produced enantiomeric ratio) with (±)-ipsdienol in the ternary blend seemed to reduce trap catches by 50%, but the difference was not statistically significant. Surprisingly, addition of (−)-trans-verbenol (at 0.2 mg/day) to the active ternary blends significantly reduced traps catches to the level not different from the unbaited control. Our results suggest that the two major components, 2-methyl-3-buten-2-ol and 99%-(+)-ipsdienol, plus a minor component, (−)-cis-verbenol, produced by fed males, are likely the aggregation pheromone components of I. shangrila.  相似文献

We present a simple model of within-group leveling coalitions among male primates. The model assumes that the value of the coalition is the sum of the payoffs of its members, that the individuals payoff is monotonically decreasing with its rank and that coalitions do not cause rank changes. It predicts that mainly mid- to low rankers engage in leveling coalitions, and that most coalition partners are of adjacent ranks. These predictions agree reasonably well with observations in nature. The model also makes the novel predictions that leveling coalitions are found where male mating competition has only a moderate contest component, and that male dominance ranks will become poorly differentiated where leveling coalitions are frequent. Both these predictions are consistent with observations on groups of macaques and baboons. The model also may account for leveling coalitions among egalitarian human foragers, without making additional assumptions about special human capabilities.

This paper presents an investigation of the feasibility of recycling silicon carbide waste (SCW) as a source of mixture materials in the production of cement mortar. Mortars with SCW were prepared by replacing different amounts of cement with SCW, and the properties of the resulting mortars, such as the fluidity, strength and shrinkage, were studied in this work. Thermogravimetry-differential scanning calorimetry and scanning electron microscopy were employed to understand the reasons for the property changes of the mortars. The results indicate that SCW decreases the initial and 1-h fluidity of fresh mortar but improves the loss of fluidity. The mortar with SCWexhibits a lower strength at 3 d and 7 d but a higher strength at 28 d and 56 d compared to the control. The shrinkage rate of cement mortar with SCW shows an obvious decrease as the replacement ratio increases. In addition, the content of calcium hydroxide in hardened paste also shows that SCW has some impact on the hydration of the cement-SCW system. The microstructures of the hardened paste also show evidence for a later strength change of mortar containing SCW. This work provides a strategic reference for possibly applying SCW as a mixture material in the production of cement mortar.

Cell cultures of marine invertebrates can provide simple and adequate model systems for studying different aspects of complex biological processes, with the additional advantage of allowing study under controlled experimental conditions. In the present study an attempt was made to establish long-term cell cultures from regenerating intestine of the sea cucumber Apostichopus japonicus and showed that cells obtained at different stages of gut regeneration display different behaviors in culture. Cultured holothurian cells were viable for a long period of time, and displayed a high level of activity after 1 month in vitro. However, only cells of primary intestinal cultures, obtained 14–16 days after evisceration, were involved in active proliferation. Furthermore, the cells undergo mitosis, and a very intensive growth period was observed during the first week of culture. Some cells (conceivably, enterocytes) were able to grow in suspension only, whereas others (presumably, coelomic epithelial cells, which form the outer layer of the gut in intact animals) grew as a multilayered mass on the polylysine surface. In contrast, cultures of intestinal cells from both intact and regenerating animals at other stages of regeneration displayed no proliferative activity. This seems to be due to specific events that take place in the regenerating gut tissues 14–16 days after evisceration. In addition, under TEM, we found that an active morphogenetic reorganization of both the gut coelomic and digestive epithelia occurred on the 14th day of regeneration. This was accompanied by cell dedifferentiation, migration and proliferation. Excessive transport of fine sediment, and its associated pollutants, can cause detrimental impacts in aquatic environments. It is therefore important to perform accurate sediment source apportionment to identify hot spots of soil erosion. Various tracers have been adopted, often in combination, to identify sediment source type and its spatial origin; these include fallout radionuclides, geochemical tracers, mineral magnetic properties and bulk and compound-specific stable isotopes. In this review, the applicability of these techniques to particular settings and their advantages and limitations are reviewed. By synthesizing existing approaches, that make use of multiple tracers in combination with measured changes of channel geomorphological attributes, an integrated analysis of tracer profiles in deposited sediments in lakes and reservoirs can be made. Through a multi-scale approach for fine sediment tracking, temporal changes in soil erosion and sediment load can be reconstructed and the consequences of changing catchment practices evaluated. We recommend that long-term, as well as short-term, monitoring of riverine fine sediment and corresponding surface and subsurface sources at nested sites within a catchment are essential. Such monitoring will inform the development and validation of models for predicting dynamics of fine sediment transport as a function of hydro-climatic and geomorphological controls. We highlight that the need for monitoring is particularly important for hilly catchments with complex and changing land use. We recommend that research should be prioritized for sloping farmland-dominated catchments.  相似文献
