The striped mullet, Mugil cephalus Linnaeus, 1758, is one of the few species of marine shore fish with a worldwide circumtropical distribution. Because of this distribution and the dependency of M. cephalus on coastal waters during various phases of its life cycle, as well as nearshore living habits, questions have been raised regarding levels of genetic divergence and gene flow among transoceanic populations. To cast more light on this, allozyme variation at 27 presumptive gene loci was investigated in ten globally diverse populations. The observed heterozygosity ranged from 0.018 (Hawaii) to 0.081 (Florida), averaging 0.050. The proportion of polymorphic loci showed a similar trend. Several populations were characterised by fixed allelic differences. Estimated gene diversities were very high, the allele frequency variation among populations was found to be 68%; genetic distances reached 0.242, with an average of 0.117. Estimated rates of gene flow were high among Mediterranean populations (Nm = 7.26), and between Mediterranean and East Atlantic populations (Nm= 2.86), but extremely low between non-contiguous populations within the Indian, Pacific and Atlantic Oceans, where Nm ranged from 0.03 to 0.05. Acute single exposures to the water-soluble fraction (WSF) of a No. 2 fuel oil influences several biochemical parameters in juveniles of Mugil cephalus Linnaeus. Plasma cortisol and glucose concentrations were measured at 1 and 3 h after exposure to 1, 5, 10 or 20% WSF. No elevation of plasma cortisol or glucose levels occurred in fish exposed to the lowest concentration of oil, whereas a dose-response relationship was observed at higher doses. The dynamics of plasma corticosteroid, glucose and cholesterol concentrations and osmolality as well as accumulation of naphthalenes in the fish tissues were monitored during exposure to 20% WSF. Circulating cortisol concentrations rose rapidly to 5 times normal values 1h after exposure to oil and subsequently declined to control levels 6 h after oil addition. A smaller secondary rise occurred at 12 h, but cortisol had returned to basal levels 12 h later. In contrast, plasma glucose, cholesterol and osmolality rose more slowly to reach maximum values between 3 and 4 h after oil addition. By 24 h plasma cholesterol and osmolality had returned to normal values whereas the hyperglycemia persisted. However, 72 h after the addition of WSF all biochemical parameters had returned to control levels. At this time considerable accumulation of total naphthalenes had occurred in several fish tissues, whilst the concentration of total naphthalenes in the exposure tanks had declined to background levels. When freshly prepared 20% WSF was added to the exposure tanks during this period, all biochemical parameters were again elevated. The results suggest that the volatile components of fuel oil in the water trigger the biochemical changes described in a dosedependent manner. The possible ecological significance of these changes and the potential use of these parameters as sublethal indicators of environmental contamination are discussed.  相似文献

The effect of body size and salinity on the rate of digestion of young grey mullet, Mugil cephalus L., was studied using the sacrifice method. The rate of digestion was found to be salinity-dependent, being slower at lower salinities than at higher salinities. This is correlated to higher food intake at the lower salinities. Rate of digestion was also found to be dependent on body size, increasing with increasing body weight.  相似文献

Starch-gel electrophoresis of allozymes was used to differentiate the two red mullet species (Mullus barbatus L. and M. surmuletus L.) in the Mediterranean Sea and, further, to investigate the genetic stock structure of M. barbatus in the eastern Mediterranean area. Twenty putative enzyme-coding loci were examined in eight M. barbatus samples caught in the Aegean and Ionian Seas (Greece) and in the Gulf of Lion (France), and two M. surmuletus samples caught in the Aegean and Gulf of Lion. A high degree of genetic polymorphism was found in both species. Species-specific electrophoretic patterns were found in PGI* and PGM*. Estimates of variance of allele frequencies among samples (F ST) and 2 analyses both revealed significant differences (P < 0.05) among the M. barbatus samples. Most of the genetic variation was among samples regardless of region. The mean value of Nei's genetic distance between the two species was 0.329. Genetic distance among M. barbatus samples was low (maximum Nei's D = 0.012), with the sample from Platania differing most from other M. barbatus samples. This is probably be due to founder effects existing at this area. These results suggest that allozyme analysis may provide important information on the genetic structure of the red mullet to ensure sustainable management of this species. The marine fish Mugil cephalus was caught in the Bay of Marseilles (France) in 1985 and the liver microsomal cytochrome P450 was identified at a concentration of about 0.230 nmol mg-1 protein. Optimal assay conditions were established to measure two mixed-function cytochrome P450-dependent oxidases: aniline hydroxylase and p-nitroanisole O-demethylase. Induction of cytochrome P450 occurred after exposure of mullet to an aromatic hydrocarbon and a water-soluble petroleum fraction. The liver cytochrome P450 was solubilized and purified. The monomer molecular weight of partially purified cytochrome P450, determined by sodium dodecyl-sulfate-gel electrophoresis, was 51 000.  相似文献

The actinomycete strain LA-29 isolated from the gut contents of the fish, Mugil cephalus of the Vellar estuary showed excellent L-asparaginase activity The enzyme was purified 18-fold and the final recovery of protein was 1.9%, which exhibited an activity of 13.57 IU/mg protein. The partially purified L-asparaginase inhibited the growth of leukemia cells in male wistar rats. Average survival period of the rats was more in an optimum enzyme dose of 100 units and the survival period was less when the dosages were increased and at the same time the enzyme became less effective when the dosages were decreased. Higher survival of 17.2 days was recorded when 100 units of the enzyme was given in three intermittent doses (50/25/25 units) at the interval of 24 hr. Analysis of cell components of the strain LA-29 has revealed the wall type-I which is the characteristic of the genus Streptomyces. Further the morphological, physiological and biochemical features along with the micromorphological results obtained for the strain LA-29 were compared with that of the Streptomyces species found in Bergey's Manual of Determinative Bacteriology and the strain LA-29 has been tentatively identified as Streptomyces canus.  相似文献

We measured the vertical and horizontal movements of striped marlin (Tetrapturus audax) off the leeward coast of the Island of Hawaii between 20 November and 18 December 1992 while simultaneously gathering data on water temperature and oceanic currents. Fish movements were monitored by ultrasonic depth-sensitive transmitters, depth-temperature profiles by an expendable bathythermograph system, and oceanic current patterns by an acoustic Doppler current profiler. Like Indo-Pacific blue marlin (Makaira mazara), striped marlin near Hawaii spend >85% of their time in the mixed layer (i.e., above 90 m depth). The maximum depth for striped marlin appears to be limited by water temperatures 8 C° colder than the mixed layer, rather than by an absolute lower temperature. We also found that the horizontal displacements of some striped marlin can be strongly influenced by currents.  相似文献

To evaluate the hypothesis that a general correlation exists between species range size and dispersal ability, we surveyed mitochondrial cytochrome b sequence variation in three surgeonfish species with vastly different ranges: Ctenochaetus strigosus, Hawaiian endemic, N = 531; Zebrasoma flavescens, North Pacific, N = 560; Acanthurus nigrofuscus, Indo-Pacific, N = 305. Collections were made throughout the 2,500 km expanse of the Hawaiian Archipelago and adjacent Johnston Atoll. Analyses of molecular variance demonstrate that all three species are capable of maintaining population connectivity on a scale of thousands of km (all species global ΦST = NS). However, rank order comparison of pairwise ΦST results and Exact test P-values revealed modest but significantly different patterns of gene flow among the three species surveyed, with the degree of genetic structure increasing as range size decreases (P = 0.001). These results are consistent with mtDNA surveys of four additional Hawaiian reef fauna in which a wide-spread Indo-Pacific species exhibited genetic homogeneity across the archipelago, while three endemics had significant population subdivision over the same range. Taken together, these seven cases invoke the hypothesis that Hawaii’s endemic reef fishes evolved from species with reduced dispersal ability that, after initial colonization, could not maintain contact with parent populations. The RNA and DNA contents of the male and female portions of the gonad of the hermaphroditic marine bivalve molluscPecten maximus L. have been monitored throughout an annual reproductive cycle, from October 1985–September 1986, for a population from the Baie de Seine (Normandy, France). The DNA content of the male gonad was consistently 5 to 10 times greater than that of the female gonad, whilst the RNA content was lower. This underlines the basic premise that spermatogenesis involves the production of large numbers of small gametes, whereas oogenesis involves the production of few, large, synthetically active gametes. The RNA:DNA ratio is a good indicator of sexual maturation in the male gonad. Two peaks of RNA:DNA ratio were observed for the female gonad, possibly corresponding to gonad restoration and vitellogenesis.  相似文献

Soluble matrices of coleoid cephalopod shells, namely the organic pen of Loligo sp. and mincralized calcified shells of Spirula spirula and Sepia sp., were extracted and studied by HPLC (high-performance liquid chromatography) gel filtration and isoelectrofocusing (IEF). Molecular weights of Spirula spirula and Sepia sp. extracts are not well separated, whereas Loligo sp. shows several well-defined peaks. In Loligo sp. and Sepia sp., molecular weights of > 100 kdaltons were found. Spirula spirula yielded a profile devoid of high molecular material. The main part of the extracted matrix of the organic pen of Loligo sp. is basic. In mineralized shells of both Spirula spirula and Sepia sp., the soluble organic matrices are acidic. These results support the view that the acidity of the organic matrix is related to mineralization processes.  相似文献

Sexual reproduction in Posidonia oceanica (L.) Delile is likely to be highly variable both in space and time. However, there has been little effort to identify at what spatial scale variability exists in the distribution and abundance of reproductive shoots or the magnitude of these variations. Such information is important for elucidating several aspects of the reproductive strategy of this species. Here, by using a reconstructing technique, I estimated the rate at which flowering events occurred over the last decades in three P. oceanica meadows along the coastline of the north-western Mediterranean (Italy). Then, variation in inflorescence frequency (i.e. the percentage of shoots that flowered in a particular year of the total number of shoots present in the sample in that year) was examined on a hierarchy of spatial scales, ranging from a few metres to meadows separated by tens of kilometres over a 3-year period. Separate analyses were also conducted to examine the patterns of spatial variability of several plant characteristics (i.e. shoot age and density of vegetative shoots) that may influence the timing of flowering. In total, ten reconstructed flowering events were recorded during the period 1983–1999. Despite the similar climatic conditions experienced in the three meadows surveyed, no inflorescence scars were found in one meadow. In the other two meadows, the flowering rate fluctuated considerably from year to year (0–26%), and simultaneous flowerings were detected only in 3 years (1993, 1994 and 1995). The pattern of spatial variability in the inflorescence frequency varied among years. An investigation of the flowering events recorded along the north-western Mediterranean coast reveals that in the studied period flowering also occurred across a broad range of meadows. Therefore, flowering could be a far more widespread phenomenon in this sector of the Mediterranean basin than previously thought. Analysis of variance showed significant variation in 1993 at the mean inflorescence frequency on very small scales (metres to tens of metres), whereas variation was not found at any of the scales investigated in 1994 and 1995. Overall, variation in the inflorescence frequency within individual meadows (65–93%) was at least an order of magnitude greater than that among separate meadows (0.5–19%). Patterns of spatial variation in the plant characteristics examined did not reflect those of flowering. It may be hypothesised that internal rhythms of populations, in conjunction with changes in environmental conditions, which are consistent over relatively large spatial (kilometres), are involved in the control of flowering induction. Environmental heterogeneity (in terms of nutrient availability) on a very local scale (metres to tens of metres) and intrinsic factors might affect the distribution of reproductive shoots within P. oceanica meadows.Communicated by R. In order to better formulate an artificial shrimp diet, the protein composition of shrimp was assessed and the essential amino acids determined. Penaeus aztecus were treated to remove lipids, carbohydrates and other interfering substances, and the protein was hydrolyzed to its constituent amino acids. The amino acids were then quantitatively analyzed by gasliquid chromatography as their trimethylsilyl derivatives. The essential amino acids were then determined. Shrimp were injected with 14C-labeled glucose and analyses performed to determine which amino acids then exhibited radioactivity, i.e., were synthesized from the labeled glucose. Those amino acids which were not manufactured from the glucose were categorized as essential.  相似文献

Atomic-force microscopy of the major class of soluble matrix protein from the oyster Crassostrea virginica revealed that this protein forms a ring structure, perhaps rendering the NH2-terminus unavailable to Edman degradation. Cleavage of these proteins using mild acid hydrolysis and hydroxylamine produced linear peptides that were able to be sequenced. Peptides consisted of a domain of polyserine-phosphoserine and runs of aspartic acid of 4 to 7 residues, with several other amino acids present. Hydrophobic domains were also isolated. Although phylogenetically distant, oyster shell-matrix protein is similar to the phosphophoryn isolated from dentin and, to a lesser extent, proteins isolated from verteorate bone.  相似文献

Seasonal variations in the microphytobenthic diatom community were investigated in an intertidal sand flat of a tropical marine environment influenced by monsoons. Cores of sediments were collected along the beach gradient: low tide, mid tide and high tide zone up to a depth of 15 cm.. Diatom abundance was lowest during the monsoons and highest during the post-monsoons and the early pre-monsoon season throughout the intertidal transect. Diatom diversity was highest at the mid tide, followed by the high and low tide zones. Diatoms were viable up to a depth of 15 cm throughout the intertidal transect. The diatom community included the pennates, the permanent residents of this area, centric genera, which lead an attached mode of life and also some planktonic genera, brought in from ambient waters. Among the pennates, Navicula and Amphora were the dominant genera whereas in the case of centrics, Thalassiosira dominated the community throughout the intertidal transect down to a 15 cm depth. . Grain size fractions, which served as predictors of some diatom genera changed with tidal zones. The effect of winds on the resuspension of the pennate diatoms was evident only at the low tide zone down to a depth of 5 cm . Chlorophyll a concentration proved to be a good predictor of both pennate and centric diatom abundance at the low tide zone down to a depth of 10 cm and at the mid tide zone down to a depth of 5 cm.. However, even though chlorophyll a concentrations failed to reveal any positive correlation with the diatom abundance at both the deeper sediment layers and the high tide zone, the fact that viable cells were present at these areas reveal that the diatoms adopt survival strategies, contributing significantly to the carbon budgets of such unstable habitats.  相似文献

The degree of population subdivision among collections of young-of-the-year (YOY) Spanish mackerel Scomberomorus maculatus sampled from along the Atlantic coast of the United States (Atlantic) and Gulf of Mexico (Gulf) was examined. Young-of-the-year were collected from spawning areas to minimize possible mixing among putative spawning stocks that may occur at later life history stages along migratory pathways or at over-wintering grounds. Restriction fragment length polymorphism (RFLP) analysis of the mitochondrial DNA (mtDNA) ND4 region and a nuclear actin intron was conducted. Collections in each of two years were analyzed to minimize bias associated with sampling at a single point in space or time. Substantial variation was detected at ND4 (haplotype diversity=0.81) and at the actin intron locus (gene diversity=0.21). Significant differences in gene diversity or allele frequencies were not detected among temporal samples at any of three locations. A homogeneous distribution of genetic variance among samples from widely spaced geographic regions was consistent with the hypothesis that Spanish mackerel comprise a single intermingling genetic stock. Power analysis showed that the mitochondrial analysis from this study was much more likely to detect population subdivision than analysis of the nuclear actin locus, despite lower mtDNA sample size. Summary Frontal gland secretions of termite soldiers of the speciesNasutitermes costalis, N. ephratae, N. nigriceps, N. rippertii, andN. hubbardii were analyzed. The chiral compositions of the monoterpene hydrocarbons were investigated by means of two-dimensional gas chromatography. Two of them, 3-carene and-pinene were present in the form of (–)-enantiomers of high purity in the majority of the samples. The other monoterpenes,-pinene, camphene, sabinene, limonene, and-phellandrene, exhibited a great variation in the enantiopurity between different species.-Pinene and limonene varied significantly even between different colonies within the species.N. costalis was the only species with an invariable composition of the secretion, independent of the locality or origin.  相似文献

We measured the horizontal and vertical movements of five adult yellowfin tuna (Thunnus albacares, estimated body mass 64 to 93 kg) near the main Hawaiian Islands, while simultaneously gathering data on oceanographic conditions and currents. Fish movements were recorded by means of ultrasonic depth-sensitive transmitters. Depth–temperature and depth–oxygen profiles were measured with vertical conductivity–temperature–depth (CTD) casts, and the current-velocity field was surveyed using an acoustic Doppler current profiler (ADCP). Large adult yellowfin tuna spent ≃60 to 80% of their time in or immediately below the relatively uniform-temperature surface-layer (i.e. above 100 m), a behavior pattern similar to that previously reported for juvenile yellowfin tuna, blue marlin (Makaira nigricans), and striped marlin (Tetrapturus audax) tracked in the same area. In all three species, maximum swimming depths appear to be limited by water temperatures 8 C° colder than the surface-layer water temperature. Therefore, neither large body mass, nor the ability to maintain elevated swimming-muscle temperatures due to the presence of vascular counter-current heat exchangers in tunas, appears to permit greater vertical mobility or the ability to remain for extended periods below the thermocline. In those areas where the decrease in oxygen with depth is not limiting, the vertical movements of yellowfin tuna, blue marlin and striped marlin all appear to be restricted by the effects of water temperature on cardiac muscle function. Like juvenile yellowfin tuna, but unlike blue marlin and striped marlin, adult yellowfin tuna remained within 18.5 km of the coast and became associated with floating objects, including anchored fish-aggregating devices (FADs) and the tracking vessel. Like juvenile yellowfin tuna, large adult yellowfin repeatedly re-visit the same FAD, and appear able to navigate precisely between FADs that are up to 18 km apart. Geographical variations in the numbers, biomass and production of euphausiids and the contribution of common species to the total are described from samples taken during 1966 and 1967 in the North Atlantic Ocean and the North Sea by the Continuous Plankton Recorder at 10 m depth. Euphausiids were most abundant in the central and western North Atlantic Ocean and the Norwegian Sea. Thysanoessa longicaudata (Krøyer) was numerically dominant. Biomass was greatest in the Norwegian Sea and the north-eastern North Sea where Meganyctiphanes norvegica (M. Sars) accounted for 81 and 59%, respectively, of the total biomass. Production was highest off Nova Scotia and in Iberian coastal waters; the dominant species were T. raschi (M. Sars) in the former area and Nyctiphanes couchi (Bell) in the latter.
