A study was under taken, under controlled laboratory conditions, to investigate the influence of non-ionizing radiation (UV-B) and an organochlorine pesticide on the growth, photosynthetic pigments, protein content and DCPIP photoreduction of a cyanobacterium Nostoc carneum. Test algae was isolated from rice field soils of Sambalpur, Western Orissa, India and grown in nitrogen free BG 11 culture medium. Culture of algae from log phase of growth was treated with 5 ppm of the insecticide, Endodhan and UV-B (20 mW m −2) for 2 h daily, separately and in combination of insecticide and UV-B radiation. Algal samples treated with UV-B and pesticide separately showed distinct inhibitory effects on growth, pigments, protein content and DCPIP reduction of the test samples. However, when pesticide treated samples were subjected to UV-B exposure, the effect showed additive as well as synergetic effect. Experiment conducted to check the ability of the organism to recover from the stress, exposed for various time periods, suggest their ability to partially recover from the stress. 相似文献

A two stage anaerobic/aerobic bacterial process was used to decolorize and partially mineralize a reactive vinyl sulfone diazo dye C.I. Reactive Black 5 (RB5) in a synthetic wastewater. Since the anchor group of reactive dyes reacts during the dyeing process, the effect the degree of hydrolysis of the vinyl sulfone dye had on decolorization, mineralization and toxicity in each stage was investigated. An overall color removal of approximately 65% was found for both the fully and partially hydrolyzed dye. Partial mineralization of the fully hydrolyzed RB5 was achieved in the two stage rotating disc reactors. While the anchor group metabolite p-aminobenzene-2-hydroxyethylsulfonic acid (p-ABHES) was mineralized, an oxidized form of the center metabolite (1,2-ketimino-7-amino-8-hydroxynaphthalene-3,6-disulfonic acid) remained in the aerobic stage effluent, causing the effluent to be colored although no RB5 was present. Partially hydrolyzed dye in the influent with vinyl forms of the anchor group caused cessation of biogas production and a reduction in decolorization efficiency in the anaerobic stage. No evidence for mineralization of the partially hydrolyzed dye or its metabolites was found. A method for evaluating dye mineralization using lumped parameters is presented. 相似文献

An advanced oxidation treatment, UV/H2O2, was applied to an azo dye, Hispamin Black CA, widely used in the Peruvian textile industry. Rates of color removal and degradation of the dye have been evaluated. A strongly absorbing solution was completely decolorized after 35 min of treatment, and after 60 min an 82% reduction of the total organic carbon (TOC) was obtained. It has been found that the degradation rate increased until an optimum value, beyond which the reagent exerted an inhibitory effect. The degradation rate was also function of pH. 相似文献

Effects of thiobencarb on growth, photosynthesis and photoinhibition of Nostoc sphaeroides colonies were examined. Thiobencarb concentrations higher than 2 mg/l led to a significant decrease in phycoerythrin, phycocyanin and allophycocyanin content, but no significant effect on chlorophyll a (chl a) content. Biomass yield, protein content and photosynthetic rate were reduced only in the highest thiobencarb concentration (10 mg/l). Colonies exposed to high thiobencarb dose (10 mg/l) were more sensitive to high light than those exposed to the control and 4 mg/l thiobencarb. Dim light induced recovery from photoinhibition to more great extent in colonies grown in the control and 4 mg/l thiobencarb than 10 mg/l thiobencarb. Darkness resulted in a insignificant recovery regardless of thiobencarb treatments. 相似文献

This study provides preliminary evidence that NO production could be a general attribute of algae. Anabaena doliolum was found to be a better NO producer than Scenedesmus and Synechoccocus. Experiments conducted with inhibitors of photosynthesis (DCMU), ATP synthesis (DCCD), and the uncoupler (2,4-DNP) and its analog arsenate clearly revealed that inhibition of nitrite assimilation through the blockage of nitrite reductase (NiR) is primarily responsible for NO emission. A linear relationship between nitrite concentration in the culture medium and NO in the exhaust gas supports the view that accumulation of nitrite is responsible for NO formation. A failure of Scenedesmus, grown in the medium substituted with W for Mo, to produce either NO/NO-2 in light or a 'light-off' peak, and a resumption of these activities upon the addition of Mo proved beyond doubt that a functional nitrate reductase (NR) is necessary for the production of nitrite and NO by algae grown on nitrate as the nitrogen source. Moreover, the appearance of a NO peak immediately after nitrite supplementation under dark conditions in W-substituted cultures with or without glucose ruled out an enzymatic role of NR in NO emission. 相似文献

Measurement of chlorophyll fluorescence has been shown to be a rapid, non-invasive, and reliable method to assess photosynthetic performance in a changing environment. In this study, acute toxicity of excess Hg on the photosynthetic performance of the cyanobacterium S. platensis, was investigated by use of chlorophyll fluorescence analysis after cells were exposed to excess Hg (up to 20 microM) for 2 h. The results determined from the fast fluorescence kinetics showed that Hg induced a significant increase in the proportion of the Q(B)-non-reducing PSII reaction centers. The fluorescence parameters measured under the steady state of photosynthesis demonstrated that the increase of Hg concentration led to a decrease in the maximal efficiency of PSII photochemistry, the efficiency of excitation energy capture by the open PSII reaction centers, and the quantum yield of PSII electron transport. Mercury also resulted in a decrease in the coefficients of photochemical and non-photochemical quenching. Mercury may have an acute toxicity on cyanobacteria by inhibiting the quantum yield of photosynthesis sensitively and rapidly. Such changes occurred before any other visible damages that may be evaluated by other conventional measurements. Our results also demonstrated that chlorophyll fluorescence analysis can be used as a useful physiological tool to assess early stages of change in photosynthetic performance of algae in response to heavy metal pollution. 相似文献

Phytoremediation provides an ecofriendly alternative for the treatment of pollutants like textile dyes. The purpose of this study was to explore phytoremediation potential of Petunia grandiflora Juss. by using its wild as well as tissue-cultured plantlets to decolorize Brilliant Blue G (BBG) dye, a sample of dye mixture and a real textile effluent. In vitro cultures of P. grandiflora were obtained by seed culture method. The decolorization experiments were carried out using wild as well as tissue-cultured plants independently. The enzymatic analysis of the plant roots was performed before and after decolorization of BBG. Metabolites formed after dye degradation were analyzed using UV–vis spectroscopy, high-performance liquid chromatography, Fourier transform infrared spectroscopy, and gas chromatography–mass spectrometry. Phytotoxicity studies were performed. Characterization of dye mixture and textile effluent was also studied. The wild and tissue-cultured plants of P. grandiflora showed the decolorized BBG up to 86 %. Significant increase in the activities of lignin peroxidase, laccase, NADH-2,6-dichlorophenol-indophenol reductase, and tyrosinase was found in the roots of the plants. Three metabolites of BBG were identified as 3-{[ethyl(phenyl)amino]methyl}benzenesulfonic acid, 3-{[methyl (phenyl)amino]methyl}benzenesulfonic amino acid, and sodium-3-[(cyclohexa-2,5-dien-1-ylideneamino)methyl]benzenesulfonate. Textile effluent sample and a synthetic mixture of dyes were also decolorized by P. grandiflora. Phytotoxicity test revealed the nontoxic nature of metabolites. P. grandiflora showed the potential to decolorize and degrade BBG to nontoxic metabolites. The plant has efficiently treated a sample of dye mixture and textile effluent. 相似文献

Pre-ozonation of 14 different reactive dyestuff hydrolysates at alkaline pH was investigated to assess possible relationships between ozone transfer efficiency, first order decolourization kinetics, release of initially complexed heavy metals and relative changes in the biodegradability of the partially oxidized dye waste samples. Biocompatibility of the raw (untreated) and ozonated dye hydrolysates was comparatively tracked through specific oxygen uptake rate measurements from which the respirometric inhibition of biological activated sludge imparted by raw and ozonated reactive dye wastewater with respect to synthetic domestic wastewater was determined. It could be demonstrated that preliminary ozonation of reactive azo dyes increases their biological compatibility more significantly than formazan copper complex, copper complex azo and phythalocyanine dyes as a consequence of heavy metal release associated with the cleavage of associated chromophoric groupings right at the initial stages of pre-ozonation. 相似文献

Biofilms harboring simultaneously anoxygenic and oxygenic phototrophic bacteria, diazotrophic bacteria, and hydrocarbon-utilizing bacteria were established on glass slides suspended in pristine and oily seawater. Via denaturing gradient gel electrophoresis analysis on PCR-amplified rRNA gene sequence fragments from the extracted DNA from biofilms, followed by band amplification, biofilm composition was determined. The biofilms contained anoxygenic phototrophs belonging to alphaproteobacteria; pico- and filamentous cyanobacteria (oxygenic phototrophs); two species of the diazotroph Azospirillum; and two hydrocarbon-utilizing gammaproteobacterial genera, Cycloclasticus and Oleibacter. The coexistence of all these microbial taxa with different physiologies in the biofilm makes the whole community nutritionally self-sufficient and adequately aerated, a condition quite suitable for the microbial biodegradation of aquatic pollutant hydrocarbons. 相似文献

Changes in biomarkers of fish captured from stressed environments may represent a reliable tool in revealing sublethal effects of the pollutants found in aquatic ecosystems. The response patterns of selected biochemical and morphological variables biomarkers of effect were assessed in adult females of an indigenous teleost Cnesterodon decemmaculatus, caught at a polluted site (San Francisco) of the Reconquista river (Buenos Aires, Argentina). Combined field-caging experiments with controlled laboratory exposure to clean media were performed. The biochemical parameters measured were specific activities of gill (Na+ + K+)-ATPase, liver AlaAT and AspAT, and brain AChE; LSI and CF were also calculated. The changes in gill (Na+ + K+)-ATPase and liver AlaAT activities of fish captured in the field in most cases were reversible after transfer to clean media. The results were interpreted in association with the physicochemical profile of the water samples taken simultaneously with the capture of the fish. Results suggested the suitability of the test species used as tools in environmental monitoring programs of risk assessment. 相似文献

The effects of crude oil and three oil spill dispersants (Corexit 9600, 9550 and 7664) on nitrogenase activity in the cyanobacteria Nostoc sp. were examined. The addition of oil to Nostoc sp. cultures resulted in a catastrophic decline in nitrogenase activity with activity ceasing 7 h after treatment. The addition of a dispersant with the oil did not ameliorate this effect. Cultures exposed to high concentrations of dispersants showed lower rates on nitrogenase activity than untreated cultures. However, it is unlikely that dispersant concentrations of this magnitude would occur in the field. At the lowest concentration tested, which approximates the manufacturer's recommended application rate, the effects of the dispersant appear to be negligible. 相似文献

The possible effects of a natural substance amygdalin and its combination with the mycotoxin deoxynivalenol (DON) on the steroid hormone secretion (progesterone and 17-β-estradiol) by porcine ovarian granulosa cells (GCs) were examined in this in vitro study. Ovarian GCs were incubated without (control group) and with amygdalin (1, 10, 100, 1,000 and 10,000 μg mL 1), or its combination with DON (1 μg mL 1) for 24 h. The release of steroid hormones was determined by ELISA. The progesterone secretion by porcine ovarian GCs was not affected by amygdalin in comparison to the control. However, the highest amygdalin dose (10,000 μg mL 1) caused a significant stimulation of the 17-β-estradiol release. A combination of amygdalin with DON significantly ( P < 0.05) increased the progesterone release at all concentrations. Similarly, a stimulatory effect of amygdalin co-administered with DON was detected with respect to the 17-β-estradiol secretion at the highest dose (10,000 μg mL 1) of amygdalin and 1 μg mL 1 of DON. Noticeable differences between the effects of amygdalin alone and its combination with DON on the progesterone release were detected. In contrast, no differences between the stimulatory effects of amygdalin and its combination with DON on the 17-β-estradiol synthesis by porcine GCs were observed. Findings from this in vitro study did not confirm the expected protective effect of amygdalin on mycotoxin induced reprotoxicity. Our results indicate that the stimulatory effect of amygdalin combined with DON on the progesterone release was clearly caused by the DON addition, not by the presence amygdalin per se. On the other hand, the stimulation of 17-β-estradiol production was solely caused by the presence of amygdalin addition. These findings suggest a possible involvement of both natural substances into the processes of steroidogenesis and appear to be endocrine modulators of porcine ovaries. 相似文献

In order to determine whether clinoptilolite, a naturally occurring zeolite, had any ameliorative effect on lead (Pb) toxicity to fungi, a series of growth experiments were performed. Three fungi, Aspergillus niger, Botrytis cinerea, and Fusarium culmorum, were grown on appropriately amended solid agar media, and their linear extension rates determined. B. cinerea was 25% inhibited, as compared to a control, at 100 mg dm(-3) Pb, and completely inhibited at 1000 mg dm(-3) Pb. F. culmorum was completely inhibited, and A. niger 97% inhibited at 1000 mg dm(-3) Pb. The addition of 3% clinoptilolite partially removed this inhibition in the case of A. niger and B. cinerea and almost completely removed it for F. culmorum. At a constant 500 mg dm(-3) Pb, increasing concentrations of clinoptilolite increased the linear extension rate of F. culmorum and B. cinerea, close to the rates achieved by the untreated controls. A. niger was not inhibited markedly at this Pb concentration. The evidence suggests that the Pb is adsorbed by the clinoptilolite which reduces the availability, and hence toxicity, of the metal to the fungi. 相似文献

Gluconacetobacter diazotrophicus an endophytic diazotroph also encountered as rhizosphere bacterium is reported to possess different plant growth promoting characteristics. In this study, we assessed the zinc solubilizing potential of G. diazotrophicus under in vitro conditions with different Zn compounds using glucose or sucrose as carbon sources. G. diazotrophicus showed variations in their solubilization potential with the strains used and the Zn compounds tested. G. diazotrophicus PAl5 efficiently solubilized the Zn compounds tested and ZnO was effectively solubilized than ZnCO(3) or Zn(3)(PO(4))(2). The soluble Zn concentration was determined in the culture supernatant through Atomic Absorption Spectrophotometer. Gas chromatography coupled Mass Spectrometry analysis revealed 5-ketogluconic acid, a derivative of gluconic acid as the major organic acid produced by G. diazotrophicus PAl5 cultured with glucose as carbon source. This organic anion may be an important agent that helped in the solubilization of insoluble Zn compounds. 相似文献

This work is an investigation on the use of manganese oxides-modified diatomite (MOMD) for the removal of color from textile wastewaters. The modification of the diatomite was carried out by treatment with manganese oxides; delta-bimessite type resulted. The surface area, pH(ZPC), Fourier transform infrared (FTIR), and scanning electron microscopy of MOMD were studied. The influence of concentration, pH, particle size, and temperature on the adsorption capacities of methylene blue (MB) and hydrolyzed reactive black (RB) and reactive yellow (RY) was investigated. Key thermodynamic parameters such as deltaH degrees, deltaS degrees, and deltaG degrees were also determined. The deltaH degrees of MB and hydrolyzed RB and RY was +94.64, -143.1, and -38.78 kJ/mol, respectively, whereas the deltaG degrees values of -17.68, -27.93, and -23.26 kJ/mol were obtained, indicating a spontaneous process with low activation-energy requirements. The findings further indicated that coulomb interaction, molecular size, and orientation of the dye and its surface charge played an important role in the adsorption and attachment of the dyes to the bimessite layers of MOMD. 相似文献

A new Aeromonas bioassay is described to assess the potential harmful effects of the glyphosate-based herbicide, Roundup ®, in the Albufera lake, a protected area near Valencia. Viability markers as membrane integrity, culturability and β-galactosidase production of Aeromonas caviae were studied to determine the influence of the herbicide in the bacterial cells. Data from the multifactor analysis of variance test showed no significant differences ( P > 0.05) between A. caviae counts of viability markers at the studied concentrations (0, 50 and 100 mg l −1 of glyphosate). The effects of Roundup® on microbial biota present in the lake were assessed by measuring the number of indigenous mesophilic Aeromonas in presence of different amounts of the herbicide at 0, 50 and 100 mg l−1 of glyphosate. In samples containing 50 and 100 mg l−1 of glyphosate a significant (P < 0.05) increase in Aeromonas spp. counts and accompanying flora was observed. The acute toxicity of Roundup® and of Roundup® diluted with Albufera lake water to Microtox® luminescent bacterium (Vibrio fischeri) also was determined. The EC50 values obtained were 36.4 mg l−1 and 64.0 mg l−1 of glyphosate respectively. The acidity (pH 4.5) of the herbicide formulation was the responsible of the observed toxicity. 相似文献

Despite its extraordinary price, ceramic membrane can still be able to surpass polymeric membrane in the applications that require high temperature and pressure conditions, as well as harsh chemical environment. In order to alleviate the high cost of ceramic material that still becomes one of the major factors that contributes to the high production cost of ceramic membrane, various attempts have been made to use low cost ceramic materials as alternatives to well-known expensive ceramic materials such as alumina, silica, and zirconia in the fabrication of ceramic membrane. Thus, local Malaysian kaolin has been chosen as the ceramic material in this study for the preparation of kaolin hollow fibre membrane since it is inexpensive and naturally abundant in Malaysia. Due to the fact that the sintering process plays a prominent role in obtaining the desired morphology, properties, and performances of prepared ceramic membrane, the aim of this work was to study the effect of different sintering temperatures applied (ranging from 1200 to 1500 °C) in the preparation of kaolin hollow fibre membrane via dry/wet phase inversion-based spinning technique and sintering process. The morphology and properties of membrane were then characterised by SEM, AFM, FTIR, XRD, and three-point bending test, while the performances of membrane were investigated by conducting water permeation and Reactive Black 5 (RB5) dye rejection tests. From the experimental results obtained, the sintering temperature of 1400 °C could be selected as the optimum sintering temperature in preparing the kaolin hollow fibre membrane with the dense sponge-like structure of separation layer that resulted in the good mechanical strength of 70 MPa with the appreciable water permeation of 75 L/h m 2 bar and RB5 rejection of 68%. 相似文献
