Following uptake of technetium-95m and americium-241 by lobsters (Homarus gammarus) from food, the subcellular distribution and binding in the major organ of accumulation, the digestive gland, were analysed.95mTc was predominantly bound to soluble proteins; approximately 40% was bound to two proteins displaying the same elution characteristics as Cdthioneins as indicated by gel permeation and ion-exchange chromatography, indicating reduction and sequestration by metallothionein in vivo. The identity of a third anionic low molecular weight Tc-binding component was not established. No evidence was found for binding of Tc to haemocyanin, ferritin or transferrin. Particulate Tc [probably TcO(OH)2] was bound to plasma membranes and was also present in lysosomes, indicating reduction of pertechnetate and pinocytosis in the digestive gland. In contrast,241Am was mainly sequestered by the lysosomal/vacuolar system, indicative of uptake by pinocytosis. Approximately 2 to 3% of the Am was bound to ferritin and 20% was bound to unidentified ligands of 10 and 20 kdaltons mol. wt. There was no evidence for sequestration of Am by metallothionein. These studies prove that waste radionuclides discharged by the nuclear industry are metabolically active and enter the normal metal metabolism and detoxication pathways of a marine shellfish.  相似文献

The biochemical composition of the pyloric caeca of female seastars (Asterias rubens) was studied throughout the annual reproductive cycle. The pyloric caeca index is high during the pseudo-rest stage of the ovaries and decreases gradually during ovarian growth. The pyloric caeca dry weight varies between 20 and 38% of the fresh weight. Lipids, and less prominently carbohydrates (both glycogen and other reducing sugars), seem to constitute the primary nutrient reserves for gonad growth. Proteins are also available to meet ovarian requirements during vitellogenesis. Glycogen levels are high in late autumn and winter and low at spawning time and in summer, whereas levels of the other reducing sugars are high in post-spawning months and subsequently gradually decrease during gonad development. The total lipid level is extremely variable (averaging 75 mg g-1 organ fresh weight), peak levels of over 200 mg g-1 being found in some individuals in summer and at spawning time. The free amino acid level is constant at about 20 mg g-1. The protein level is rather high throughout the annual cycle, and displays only minor changes. The observed patterns of decrease in major constituents suggest a rapid release of nutrient reserves from the pyloric caeca (deposited during the summer) at the onset of vitellogenesis. Moreover, the needs of the ovaries during further development seem to be so tremendous that during the winter months the digested food may be transported to the ovaries without prior deposition in the pyloric caeca. The changes observed in the pyloric caeca are discussed and compared with those occurring simultaneously in the ovaries.  相似文献

Interactions between mercury and selenium accumulation and subcellular binding inAsterias rubens (L.), collected in 1987 from Lille Bælt at Middelfart, Funen, Denmark, were investigated in laboratory experiments. Sea stars exposed to 10µg Hg l–1 for 30 d accumulated mercury in body wall, tube feet and stomach linearly with time at 1.2, 1.2 and 0.5µg Hg g–1 dry wt d–1, respectively. Mercury was accumulated in pyloric caeca and coelomic fluid initially at 1.4µg Hg g–1 dry wt d–1 and 9.4 ng Hg ml–1 d–1, respectively; after 10 d uptake rates decreased. Sea stars exposed to 75µg Se-SeO 3 - - l–1 accumulated selenium linearly with time over 30 d in the stomach, pyloric caeca, tube feet and body wall at 2.0, 1.2, 1.2 and 0.6µg Se g–1 dry wt d–1. Sea stars exposed to 75µg Se-SeO 4 - - l–1 maintained selenium levels in the coelomic fluid at 75µg Se l–1 over 30 d. Exposure to selenate did not alter the selenium concentrations in the tissues. Sea stars exposed concurrently to 75µg Se-SeO 3 - - and 10µg Hg l–1 accumulated more mercury and selenium in tube feet and body wall than did sea stars exposed to the two elements alone. In pyloric caeca and stomach concurrent exposure reduced accumulation of both elements. Mercury was bound predominantly in the insoluble fraction of the tissues, and soluble mercury was bound in proteins of high (> 70 kilodaltons) or very low (< 6000 daltons) molecular weight. Ca. half of the selenium recovered was bound in the insoluble fraction, and soluble selenium was bound in proteins of high (> 70 kilodaltons) or very low (< 6000 daltons) molecular weight. Interaction between the two elements was exerted predominantly in the insoluble fraction of the tissues.  相似文献

Inshore and offshore populations ofSclerasterias mollis (Hutton, 1872) were sampled on the outer continental shelf off the Otago Peninsula, New Zealand, and their nutritional and reproductive cycles are described from 1985–1986. Histological changes in the gonads are generally typical of other asteroids. The gametogenic cycle takes 12 mo. The gonad and pyloric caeca indices in both females and males had an inverse relationship.S. mollis accumulates nutrients in the pyloric caeca during summer and early autumn. The gonads develop in the autumn and winter. Offshore seastars were much larger and had significantly higher gonad and pyloric caeca indices than inshore individuals. These differences in body sizes and organ indices arise from differences in food availability and/or population density.  相似文献

Seasonal changes in biochemical composition of tissues are compared in a shallow inshore and deeper offshore population of the seastarSclerasterias mollis (Hutton, 1872) on the Otago shelf, New Zealand, from 1985 to 1986. The biochemical composition of the body wall, gonads and pyloric caeca of the two populations did not differ greatly, even though reproductive output and nutrient storage of reserves were consistently higher in the offshore population. The biochemical composition of the ovaries and testes differed. High levels of carbohydrate and lipid were maintained in the ovaries from the commencement of vitellogenesis until spawning occurred. The testes had higher ash levels during spermatogenesis. During the annual reproductive cycle, an inverse relationship in the carbohydrate levels between the ovary and the pyloric caeca suggests nutrient transfer from the pyloric caeca to the gonads during gametogenesis. Annual changes in biochemical composition indicate that the body wall is also used for nutrient storage.  相似文献

The starfish Cosmasterias lurida was sampled from the pier piles of Almirante Storni in the Golfo Nuevo, northern Patagonia, from November 2001 to November 2003. In this anthropogenically modified site there is an unusually abundant food supply of mussels and large aggregations of starfishes. The reproductive cycle was studied by organ indices and histological examination of gonads. The highest gonad index (GI) values were found in the summer (February–March), followed by a drop in GI values, indicating spawning. Histological analyses indicated that gametogenesis takes place from June to January, sexual maturity occurred during the summer and spawning occurred in April. The GI and pyloric caeca index (PCI) tended to have an inverse relationship, but this was only statistically significant in males. However, biochemical analyses of the starfish indicated no transfer of nutrients between pyloric caeca and gonads in either sex. In fact, simultaneous accumulation of carbohydrates was found in the pyloric caeca and gonads of both sexes.  相似文献

The protease activity of crude extracts from various organs of the digestive tract of two groups of milkfish was determined. One group (Sample A) derived their food from ponds that had predominantly unicellular algae while the other group (Sample B) were reared on ponds dominated by the filamentous green algae Chaetomorpha brachygona. In general, crude extracts from Sample A fish had a higher protease activity than Sample B fish. In both samples, high protease activity was observed in crude extracts from the pyloric caeca, intestines and pancreas. Chymotryptic activity was observed in crude extracts of pancreas, intestines and pyloric caeca of both fish samples. Tryptic activity was, however, observed only in fish grown on unicellular algae. Experimental evidence suggests that a powerful trypsin inhibitor in Chaetomorpha brachygona may account for the absence of tryptic activity in all crude extracts of Sample B fish. The subcellular distribution of 237Pu in the livers of the plaice Pleuronectes platessa has been studied by differential and isopycnic gradient centrifugation. It has been shown that the 237Pu is particularly accumulated in pellets sedimenting out at 3000 and 6000 g which, by deduction from marker enzyme tests, contain a mixture of mitochondria and lysosomes. Separation of lysosomes from mitochondria was achieved by using a non-lytic detergent injected into the fish 3 d prior to removal of the livers; it was found that 237Pu was associated with the lysosomal fraction. In this respect, the metabolism of plutonium of the plaice resembles that of the rat.  相似文献

Technetium-99 (99Tc) is a radioactive isotope with a half-life of 2.13 × 105 year. 99Tc is a major radionuclide contaminant of concern for the world. Therefore, detailed understanding of 99Tc environmental chemistry is important for protecting human and ecological health. Here, we review the aqueous geochemistry of 99Tc, focusing on chemical properties and behavior of 99Tc in the hydrosphere and pedosphere. We describe the general chemistry of Tc including coordination chemistry, isotope chemistry and solid-state chemistry. Then we present anthropogenic and indigenous sources of 99Tc and their environmental distribution. We also discuss the radiotoxicity of 99Tc and the use of 99Tc for medical purpose. Tc biogeochemistry is described through sorption and desorption reactions at the mineral–water interface, thermodynamic of surface complexation, abiotic and biotic redox reaction of Tc(VII). Finally, we summarize the 99Tc remediation technologies including cement waste forms, natural remediation, vitrification, nuclear waste transmutation.  相似文献

Solution culture was conducted in order to understand accumulation characteristics and chemical forms of Pb in Arenaria orbiculata (A. orbiculata) and the response of root exudates to Pb addition. The results showed that: 1) Pb contents in the shoot and root of A. orbiculata increased with increasing in Pb concentrations in solution. 2) The contents of Pb chemical forms under Pb addition followed as: HAc extractable fraction (FHAC)>HCl extractable fraction (FHCl)>NaCl extractable fraction (FNaCl)>ethanol-extractable fraction (FE)>water extractable fraction (FW). 3) Increased Pb level in the medium caused increases in Pb contents in the four subcellular fractions of shoots and roots, with most accumulation in FIV (Fraction IV, soluble fraction) in shoots and FI (Fraction I, cell wall fraction) in roots. 4) Contents of soluble sugar and free amino acid of root exudates increased with increasing Pb concentration in solution. Significantly positive correlations between Pb and contents of soluble sugar and free amino acid were observed. 5) With Pb concentrations in solution, low molecular weight organic acids (LMWOAs) contents followed the tendency: tartaric acid>acetic acid>malic acid>citric acid. Significantly positive correlation was observed between Pb and citric acid contents. The results indicate that soluble sugars, free amino acid and citric acid in root exudates of A.orbiculata facilitate the absorption and accumulation of Pb, which exist in NaCl-, HCl- and HAc- extractable Pb forms, FI and FIV fractions, resulting in tolerance of A.orbiculata to Pb.  相似文献

A comparative study on mixed-function oxygenase (MFO) systems was carried out on four echinoderm species: the asteroidsAsterias rubens andMarthasterias glacialis, a holothurianHolothuria forskali and an echinoidEchinus esculentus. Cytochromes P-450 andb 5 and the MFO system-associated NADH-ferricyanide reductases NADH-cytochromec reductase, NADPH-cytochromec reductase and benzo(a)pyrene (BP) hydroxylase activities were present in microsomal fractions of pyloric caeca ofA rubens andM. glacialis and in the haemal plexus ofH. forskali. In contrast, cytochrome P-450 and BP hydroxylase activity were not detectable in the gonads ofE. esculentus. The tissue and subcellular distribution of the MFO system was studied inA. rubens. MFO system components were found in the stomachs and gonads, although detection of cytochrome P-450 in the latter tissue was difficult. Sex-related differences were not significant. The contents of the MFO system components and BP hydroxylase activities were highest in the microsomal (100 000 ×g) fractions, but MFO system components were also found in the mitochondrial (12 000 ×g) and cytosolic fractions. The BP hydroxylase activity in pyloric caeca microsomes ofA. rubens was NADPH-dependent and was inhibited by several agents known to be inhibitors of vertebrate cytochromes P-450. In the former respect, the characteristics of the MFO system were more like those of vertebrates and crustaceans than that of molluscs.  相似文献

Changes in size, morphology, and biochemical composition in adults and embryos of a brooding sea star, Neosmilaster georgianus (Sladen), were studied in a population adjacent to Anvers Island, Antarctic Peninsula (64°46′S; 64°04′W) during the austral spring, 1991. Five morphological stages of development were designated in 24 broods, and for each the weight and biochemical composition of the brooding adults and their embryos were determined. Between Stage 1 and 2, the dry weight (dw) and organic weight (ow) of the embryo did not change. From Stage 2 to 3, the dw and ow increased significantly by 10%. Stage 2 and 3 embryos were in clusters of a few (2 to 10) to as many as 40 individuals. In the smaller clusters, individual embryos were attached by tissue cords to another, sometimes atrophied, brood member. In the larger clusters, they were attached to a central mass of tissue containing remnants of embryos. We interpret these interactions as a form of cannibalism which may account for the weight gains between Stage 2 and 3. During Stages 4 and 5, as juvenile form was approached, the dw and ash weight of the young increased significantly and the ow decreased significantly. The calculated energy content for the juvenile (Stage 5) was not significantly different from the energy content of the earliest undifferentiated stage (Stage 1), an indication that most of the organic matter in the egg is the primary contribution to the large juvenile. In brooding females, pyloric caeca indices declined by 52% from Stage 1 to Stage 5 and pyloric energy stores declined by 63% due to proportionately equivalent declines in protein and lipid. The ovary index was low and increased only slightly during brood protection, while the size of the largest oocytes remained approximately 23% that of ova. Energy stores in the pyloric caeca of brooding N. georgianus thus become depleted over a long period of incubation during which the adults apparently starve. In the carnivorous isopod Eurydice pulchra Leach the mid-gut caeca have a pH of 5.6 and produce a strong acid proteinase, a lipase and a carbohydrase. Electron microscope studies of the digestive caeca of starved and fed animals show that secretion is merocrine and may originate from a complex involving the basal cell membrane and associated mitochondria, producing lysosomes. Absorbed materials, in fed animals, appear to take the form of neutral lipid bodies, glycogen, and proteinaceous crystals, the latter forming within the golgi complex. Digestion of both lipid and protein inclusions was observed to occur intra-cellularly by fusion with free lysosomes. Secretion and absorption may occur in the same cell, and the scheme for the sequence of epithelial cell stages is proposed.  相似文献

The purple seastar Pisaster ochraceus contains clearly measurable protease and amylase activity. Centrifuged supernatants of pyloric caeca homogenates undergo a spontaneous threefold increase in protease activity when incubated under toluene for 50 h at 25°C. Amylase activity remains nearly constant over this period. Bovine trypsin at a 1 to 100 ratio (trypsin to supernatant protein) induces a twofold increase in protease activity over that of the control supernatant while having virtually no effect on amylase activity over the control. The data indicate a specific interaction of trypsin with a protease zymogen rather than a conspecific hydrolysis of membrane components or vesicles by trypsin. Two percent Triton X-100 used as a diluent in place of distilled, deionized water causes a sevenfold increase in protease activity and a twofold increase in amylase activity in pyloric caeca supernatants. The use of Triton as a diluent in the preparation of a stomach-tissue supernatant allows quantitative measurement of both protease and amylase activity in that tissue.  相似文献

Seasonal changes in pyloric caecum and gonad indices were studied in a population of the seastar Asterias forbesi (Desor) from East Rockaway Inlet (Long Island, New York) during two annual cycles (1980/81, 1982/83). Pyloric caeca indices increased during fall, reached a maximum about April, and declined sharply to a minimum in mid-summer. Gonad indices increased during fall and winter and reached a maximum about May. Judging from gonad size analysis, spawning occurred in late June to early July, at bottom temperatures of 16° to 18°C. There was no long-term inverse relationship between pyloric caecum and gonad indices. Since nutrients and energy ingested during the important fall feeding period are utilized simultaneously for body growth and gonad development, it is unnecessary for the pyloric caeca to store nutrients over long periods as occurs in A. rubens and many other species. The specific caeca-gonad relationship is probably an adaptation to the extreme seasonal thermal fluctuations of the NW Atlantic, which constrain the feeding activity of A. forbesi and limit the possibility of long-term storage. In this population, most individuals grow rapidly in their first year (including first two summers) and spawn in their third summer. Few seastars survive to spawn again in their fourth summer. However, gametogenesis may take place in some individuals in their second summer. Year-to-year variability in mean size at spawning may reflect temporal variation in environmental conditions (weather, food availability) at this site.  相似文献

通过溶液培养实验,研究了镉胁迫下2个箭舌豌豆(Vicia sativa)品种(镉耐性品种L3,镉敏感品种ZM)中镉的积累、镉的亚细胞分布和化学形态以及非蛋白巯基化合物(NPTs)的响应。结果表明2个箭舌豌豆品种根较地上部积累更多的镉。镉在箭舌豌豆根、茎和叶中主要分布于可溶性组分,敏感品种ZM根中可溶性组分的镉含量和占总镉的比例显著高于耐性品种L3。细胞壁结合镉占总镉的比例在2个品种中均为叶>茎>根。镉在箭舌豌豆根和茎内主要以去离子水(d-H2O)提取态和1 mol·L-1氯化钠(Na Cl)提取态存在,ZM根中d-H2O提取态镉的含量和比例显著大于L3。箭舌豌豆叶中镉主要以2%醋酸(HAc)提取态存在。ZM叶中80%乙醇提取态镉的比例大于L3,0.6 mol·L-1盐酸(HCl)提取态镉和残渣态镉的比例小于L3。2个品种根和茎中NPTs的含量在镉胁迫下显著升高,ZM根中NPTs的含量显著大于L3。研究结果表明,箭舌豌豆镉解毒的主要机制包括限制镉由根部向地上部转运和液泡隔离。此外,根和茎中镉与有机酸结合和NPTs螯合,叶中镉以移动性低、毒性低的形态存在也是箭舌豌豆镉解毒的重要机制。ZM较L3根中更多镉以移动性高的形态存在,L3较ZM叶中更多镉以难移动的形态存在,是L3较ZM具有更高镉耐性的重要原因。  相似文献

Sexual and asexual reproduction of the fissiparous starfish Allostichaster capensis were examined for 24 months at Bahía Kaiser, Golfo Nuevo, Chubut, Argentina. Gonad indices revealed an annual reproductive cycle, with peak development occurring in late winter (August). Spawning occurred in spring (September). Pyloric caeca indices displayed a clear annual cycle and a reciprocal relationship with gonad indices, showing the storage function of pyloric caeca. The starfish population appears to be largely maintained by fission, as only two females were found. A. capensis also showed an annual cycle of fission, with the highest frequency in spring and summer (from November to January), just after spawning. Detailed information is presented on the changes in lipid-class profiles (polar lipids, total neutral lipids, free sterols, steryl esters, triacylglycerols, neutral ether lipids and free fatty acids), which are manifest during the annual reproductive cycle of the seastarAsterias rubens for both the storage organs (pyloric caeca) and utilizing organs (ovaries). The ovaries appear to accumulate all individual lipid components proportional to progression of vitellogenesis. Pyloric caeca, in contrast, show variable lipid-class levels, probably due to more complex processes in these organs, operating simultaneously: digestion of food, accumulation of storage lipids, mobilization of stored lipids and their release. The results are discussed in terms of current knowledge on the role of lipids in echinoderm reproduction.  相似文献

Individuals and populations of Odontaster validus Koehler differed markedly among different habitats, as revealed in a study from October 1984 through January 1986 in McMurdo Sound, Antarctica. At McMurdo Station, individual sizes (wet weight) and population biomass (g wet wt m-2 and kJ m-2) decreased significantly with increasing depth. Individuals from shallow (10 to 20 m) habitats were in superior nutritional condition to those from deeper water (30 and 165 m), as shown by higher gonad and pyloric cecum indexes, and by higher lipid and energetic levels in the pyloric ceca. Moreover, gonadal output (reproductive output) was higher in shallow-water individuals. Higher levels of chlorophyll in the pyloric ceca and richer yellow to red coloration of the body wall in the shallow-water individuals indicate that they utilize the higher levels of primary production at shallow depths. At East Cape Armitage, where nearly permanent, thick, snow-covered ice most of the year resulted in very low levels of benthic primary production, the lowdensity sea stars were all very small and nutritionally similar to the deep-water individuals at McMurdo Station. At Cape Evans, where the generally snow-free sea-ice that broke up in mid-summer resulted in a luxurient benthic cover of diatoms and macroalgae, the sea stars were smaller than at McMurdo Station at comparable depths, but population densities were higher, resulting in 4 to 9 times greater biomass. Growth rates of sea stars fed in the laboratory were very low, especially compared to laboratory-reared temperate and tropical species; well-fed individuals need about 9 yr to reach 30 g wet weight, near the mean size of shallowwater individuals at McMurdo Station. No growth was detected in individuals caged at McMurdo Station for one year, suggesting even lower growth rates in the field. The stable size-frequency distributions at the different sites and depths throughout the year-long study suggest highly stable populations with low temporal variability in recruitment, migration and mortality. These data indicate that individuals and populations of O. validus quantitatively and qualitatively reflect the general level of productivity of a habitat. Differences noted in size, coloration, nutrition, and reproductive effort may be the result of long-term integration of local levels of primary production. These ubiquitous sea stars may serve as a biotic indicator of productivity in localized habitats around the continental shelf of Antarctica.  相似文献

The biochemical composition of the sediment organic matter, and bacterial and meiofaunal dynamics, were monitored over an annual cycle in aPosidonia oceanica bed of the NW Mediterranean to test the response of the meiofauna assemblage to fluctuations in food availability. Primary production cycles of the seagrass and its epiphytes were responsible for relatively high (compared to other Mediterranean systems) standing stocks of organic carbon in sediments (from 1.98 to 6.16 mg Cg–1 sediment dry weight). The biopolymeric fraction of the organic matter (measured as lipids, carbohydrates, and proteins) accounted for only a small fraction (18%) of the total sedimentary organic carbon. About 25% of the biopolymeric fraction was of microphytobenthic origin. Sedimentary organic carbon was mostly refractory (56 to 84%) and probably largely not utilizable for benthic consumers. The biopolymeric fraction of the organic matter was characterized by high carbohydrate concentrations (from 0.27 to 5.31 mg g–1 sediment dry weight in the top 2 cm) and a very low protein content (from 0.07 to 0.80 mg g–1 sediment dry weight), which may be a limiting factor for heterotrophic metabolism in seagrass sediments. RNA and DNA concentrations of the Sediments varied significantly during the year. High RNA and DNA values occurred during the microphytobenthic bloom and in correspondence with peaks of bacterial abundance. Bacteria accounted for a small fraction of the total organic carbon (0.65%) and of the biopolymeric organic carbon (4.64%), whilst microphytobenthos accounted for 3.79% of total organic carbon and for 25.08% of the biopolymeric carbon. Bacterial abundance (from 0.8 to 5.8 × 108 g–1 sediment dry weight) responded significantly to seasonal changes of organic matter content and composition and was significantly correlated with carbohydrate concentrations. Bacteria might be, in the seagrass system, an important N storage for higher trophic levels as il accounted for 25% of the easily soluble protein. pool and contributed significantly to the total DNA pool (on average 12%). Total meiofaunal density ranged from 236 to 1858 ind. 10 cm–2 and was significantly related, with a time lag, to changes in bacterial standing stocks indicating that microbes might represent an important resource. Bacterial abundance and biomass were also significantly related to nematode abundance. These results indicate that bacteria may play a key role in the benthic trophic  相似文献
