二苯并噻吩脱硫微生物菌种的筛选与活性   总被引:5,自引:0,他引:5       下载免费PDF全文
从被石油污染的土壤中筛选出1株能对二苯并噻吩(DBT)进行高效脱硫的微生物菌种,初步鉴定为红串红球菌USTB-03(RhodococcuserythropolisUSTB-03).该菌种可以按特异性脱硫途径(简称4S途径)将DBT转化为2-羟基联苯(2HBP)和亚硫酸作为最终脱硫产物.在葡萄糖、甘油和乙醇分别作为微生物生长的唯一碳源时,葡萄糖是支持该菌生长和提高其脱硫比活性的较好碳源,使培养出的微生物对DBT的脱硫比活性达到了68.63mmol2HBP/(kg·h).该菌株还可以对4,6-二甲基二苯并噻吩(4,6-DMDBT)进行脱硫.  相似文献

Harmful cyanobacterial blooms are a growing environmental problem worldwide in natural waters, the biodegradation is found to be the most efficient method for removing microcystins (MCs) produced by harmful cyanobacteria. Based on the isolation of a promising bacterial strain of Sphingopyxis sp. USTB-05 for biodegrading MCs, we for the first time cloned and expressed a gene USTB-05-A (HM245411) that is responsible for the first step in the biodegradation of microcystin LR (MC-LR) in E. coli DH5αup, with a cloning vector of pGEM-T easy and an expression vector of pGEX-4T-1, respectively. The cell-free extracts (CE) of recombinant E. coli DH5αup containing USTB-05-A had high activity for biodegrading MC-LR. The initial MC-LR concentration of 40 mg/L was completely biodegraded within 1 hr in the presence of CE with a protein concentration of 0.35 mg/mL. Based on an analysis of the liquid chromatogram-mass spectrum (LC-MS), the enzyme encoded by gene USTB-05-A was found to be active in cleaving the target peptide bond between 3-amino-9-methoxy-2,6, 8-trimethyl-10-phenyl-deca-4,6-dienoic acid (Adda) and arginine of MC-LR, and converting cyclic MC-LR to linear MC-LR as a first product that is much less toxic than parent MC-LR, which offered direct evidence for the first step on the pathway of MC-LR biodegradation by Sphingopyxis sp. 利用定向筛选驯化的二苯并噻吩(DBT)降解菌对石油污染土壤进行为期40 d的土壤模拟培养试验,研究了功能菌(红球菌,Rhodococcus sp. ZYL-1)接种对石油污染土壤中DBT的降解效果,结合16S rDNA高通量测序及生物信息学分析,解析了功能菌接种对土壤细菌群落演替的影响。结果表明:在25 ℃的暗箱培养过程中,红球菌添加显著提高了DBT污染土壤的降解率(P<0.001),生物接种处理组(BIOEIF)的DBT降解主要发生在培养前10 d,培养结束后DBT降解率接近60%,比依赖土壤土著微生物的自然衰减组(NAT)降解率提升10%以上;对比BIOEIF组和NAT组土壤培养过程中细菌群落组成,BIOEIF组香农多样性指数和系统发育指数显著低于NAT组,但接种红球菌未对DBT污染土壤的细菌群落组成造成显著影响,Micromonospora、Bacillusunclassified_f_Planococcaceae为污染土壤培养过程中的优势菌属,而接种的红球菌并未成为优势菌属;网络分析表明,功能菌接种显著提升污染土壤微生物的DBT关键降解菌属类群,通过强化土壤土著微生物Shimazuella协同降解DBT,进而提升了石油烃污染土壤的修复效果,Shimazuella可能是参与DBT的代谢的关键微生物。所接种的功能菌(红球菌)可协同提高土著微生物对于石油污染土壤DBT的生物降解,具有较高的土壤修复应用潜力。

利用同位素示踪法对红球菌BAP-1跨膜运输14C-荧蒽的过程进行了研究.结果表明在有ATP抑制剂NaN3的存在下,红球菌BAP-1细胞膜内所结合的14C-荧蒽含量几乎无变化.结合对微生物体内包涵体的观察,说明当有ATP抑制剂存在的情况下,荧蒽无法通过细胞膜进入到微生物的体内.这表明若是荧蒽无法通过跨膜运输进入到红球菌细胞内,就不能得到有效的生物降解.在不同的底物浓度条件下,微生物对14C-荧蒽的跨膜运输过程是主动运输过程;在一定底物浓度条件下,菌体膜结合污染物的量会在一定的条件下达到饱和.结合米氏方程分析了红球菌BAP-1对14C-荧蒽的跨膜运输动力学过程,结果表明底物14C-荧蒽与微生物之间一直保持较高的亲和力,较高的亲和体系有助于跨膜运输过程的顺利进行.  相似文献

采用温室盆栽试验方法,研究了丛枝菌根真菌(AMF)对菲、芘复合污染土壤中微生物数量和酶活性的影响.结果表明,接种AMF显著增加了三叶草根际和菌丝际土壤中细菌、真菌和放线菌的数量,并对微生物区系有选择性.在供试菲、芘污染浓度范围内,低浓度菲、芘对土壤多酚氧化酶、酸性磷酸酶和过氧化氢酶活性有激活作用;但当菲、芘浓度升高时,3种酶活性受到抑制.与无植物对照土壤相比,接种AMF后三叶草菌根际酸性磷酸酶活性降低了2.4%~23.1%,过氧化氢酶活性增加了12.6%~20.3%,除高浓度处理外多酚氧化酶活性均增加;菌丝际多酚氧化酶活性比菌根际低12.9%~62.9%,酸性磷酸酶活性比菌根际高3.3%~24.0%,过氧化氢酶活性则高于菌根际.  相似文献

通过研究不同浓度(30、40、50、60、70 mg·L-1)和不同粒径(0~75μm、75 ~ 96μm、96 ~150 μm、150~ 250 μm)水体悬浮颗粒物对斜生栅藻(Scenedesmus obliquus)叶绿素a含量、抗氧化系酶活性、脂质过氧化物丙二醛(MDA)含量、可溶性蛋白含量及光合参数变化的影响,探讨了水体悬浮颗粒物对斜生栅藻生理生化及光合活性影响的机制.结果表明,预处理与未处理悬浮颗粒物在一定浓度范围内会促进藻类生长,超过各自临界浓度(预处理为40 mg·L-1、未处理为50 mg·L-1)后,随着颗粒物浓度的增加会对藻类生长产生抑制现象未经处理的相同浓度不同粒径颗粒物对斜生栅藻都表现出抑制作用,0~75μm粒径组颗粒物作用下藻体内超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、MDA和可溶性蛋白质含量等均显著区别于其他粒径组的作用(p<0.05).经过预处理的相同浓度不同粒径颗粒物中,96~150μm粒径组对藻类的生长抑制较其他粒径组明显,且小粒径组颗粒物对藻类体内SOD、CAT、MDA和可溶性蛋白质含量等影响较大.受不同浓度悬浮颗粒物胁迫的斜生栅藻相对电子传递速率随着时间延长在一定程度上降低,但这种抑制作用在藻细胞的耐受范围,藻细胞可以通过自身调节将电子传递速率恢复到正常水平甚至更高以抵抗逆境环境.  相似文献

目前,有关三氟羧草醚(Acifluorfen,AF)的神经毒性未见报道且亟待确定.为探讨AF经口暴露对小鼠学习记忆能力的影响及其可能机制,将30只雄性昆明小鼠随机分成生理盐水对照组、0.13、1.3、13和130 mg·kg~(-1)·d~(-1) AF染毒组共5组,灌胃染毒14 d,进行Morris水迷宫实验,观察脑海马病理切片,并检测脑组织中活性氧(ROS)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、磷酸化cAMP反应元件结合蛋白(pCREB)和脑源性神经营养因子(BDNF)含量.结果显示,与对照组相比,AF染毒剂量分别为13和130 mg·kg~(-1)·d~(-1)时,小鼠行为学上学习记忆能力下降;海马细胞排列松散;出现氧化损伤,其中,ROS含量显著升高(p0.05),GSH含量显著减少(p0.05);神经保护能力减弱,其中,pCREB和BDNF水平显著(p0.05)或极显著(p0.01)下降.结果表明,AF能够导致小鼠学习记忆能力下降,氧化应激和CREB-BDNF通路级联下调可能是AF造成神经毒性的机制之一.  相似文献

细胞色素P450酶(CYP450)和谷胱甘肽转移酶(GST)是动物体内主要的解毒酶,在外源毒物的转化和代谢过程中具有重要作用.本文在实验条件下, 将健康性成熟黑斑蛙(Rana nigromaculata)暴露于浓度分别为0.005、0.01、0.05、0.1、0.5和1.0 mg·L-1的Cd溶液中30 d, 采用荧光分光光度法和紫外分光光度法测定精巢组织乙氧基异酚恶唑脱乙基酶(EROD)、红霉素N-脱甲基酶 (ERND)、氨基比林-N-脱甲基醇酶(APND)和谷胱甘肽转移酶(GST)活性,探讨Cd在精巢的可能代谢过程和毒性机理.结果表明, 与对照组比较, EROD和GST酶活性在0.5和1.0 mg·L-1 Cd处理组被显著抑制; ERND酶活性在0.01~1.0 mg·L-1 Cd处理组均被显著抑制;而APND酶活性在各处理组响应变化不明显.结果显示, 精巢EROD、ERND和GST对Cd胁迫的敏感性高于APND, 黑斑蛙精巢中EROD、ERND和GST酶活性的响应可用来评价环境中低水平Cd的污染效应.  相似文献

在实验室条件下,观察质量浓度分别为高、中、低(0·1,1,10ng·L-1(以Sn计))3组的三丁基锡(TBT)暴露2、8、20d以及恢复7d和20d后对文蛤鳃的谷胱甘肽硫转移酶(GST)、过氧化氢酶(CAT)活性以及丙二醛(MDA)含量、还原型谷胱甘肽(GSH)含量的影响.结果显示,除了高浓度TBT(10ng·L-1)暴露在早期(2d)对GSH含量、GST和CAT活性产生抑制之外,TBT暴露对GSH含量、GST和CAT活性的作用主要为诱导效应.在暴露8d后,MDA的含量表现出显著的诱导.恢复7d后除了高浓度组对MDA还有影响外,其它各指标均恢复到与对照组相当的水平.结果表明,环境水平的TBT暴露对文蛤产生明显的氧化胁迫,抗氧化防御系统可以作为海洋环境有机锡污染监测的潜在的生物标志物.  相似文献

This study focuses on the effects of pH and fluoride at different molar ratios of fluoride to Al (RF:Al) on the removal of cadmium (Cd2+) and phosphate by Al coagulation. Fluoride at RF:Al ≥ 3:1 inhibits the removal of Cd over wide Al dose ranges from 5 to 10 mg/L as Al. The removal of phosphate decreases significantly at high RF:Al of 10:1 whereas at lowered RF:Al (i.e., ≤ 6:1), an adverse effect is observed only at insufficient Al doses below 2 mg/L. Fluoride shows inhibitive effects towards the removal of Cd at pH 7 and 8 and that of phosphate at pH 6. Fluoride decreases the ζ-potential in both systems, and the decreasing extent is positively correlated to the elevated RF:Al. The Al fluoride interactions include the formation of Al–F complexes and the adsorption of fluoride onto Al(OH)3 precipitates, i.e., the formation of Al(OH)nFm. Al–F complex formation inhibits Al hydrolysis and increases residual Al levels, and a more significant increase was observed at lower pH. Al–F complexes at high RF:Al complicate the coagulation behavior of Al towards both negative and positive ionic species. Moreover, fluoride at low RF:Al shows little effect on Al coagulation behavior towards Cd2 + and phosphate, and the spent defluoridation adsorbent, i.e., aluminum (Al) hydro(oxide) with adsorbed fluoride at RF:Al of below 0.1:1, may be reclaimed as a coagulant after being dissolved.  相似文献

The response of bacteria to various carbohydrates in the deep-sea sediments and the Antarctic soils was investigated using cellulose, chitin, and olive oil. It was found that the carbohydrates significantly increased the corresponding specific ectoenzyme activity (β- glucosidase, β-N-acetylglucosaminidase, lipase) in the samples from deep-sea sediments. In the case of Antarctic soil samples, the cellulose or olive oil amendments had minor or no effect on β-glucosidase or lipase activity, except the chitin which stimulated β- N-acetylglucosaminidase production. The responses of the bacteria in the deep-sea sediment sample WP02-3 and the Antarctic soil sample CC-TY2 towards the chitin amendment were further analyzed. Chitin amendments were shown to stimulate the ectoenzyme activity in all the tested sediments and the soils. The bacterial response before and after the carbohydrates amendments were compared by denaturing gradient gel electrophoresis and quantitative competitive polymerase chain reaction. Significant changes were found in the structure and density of the bacterial community in the deep sea sediments as compared to the Antarctic soil sample, where the effects were relatively lower. There was no change in the bacterial population in both studied samples in response to carbohydrates amendments. These data indicate that the bacterial communities in the oligotrophic deep-sea sediments are more dynamic than that in the Antarctic soils as they respond to the nutrient sources efficiently by regulation of ectoenzyme activity and/or changing community structure.  相似文献

This study focuses on the effects of p H and fluoride at different molar ratios of fluoride to Al(R F:Al) on the removal of cadmium(Cd~(2+)) and phosphate by Al coagulation. The removal of phosphate decreases significantly at high R F:Al of 10:1 whereas at lowered R F:Al(i.e., ≤ 6:1), an adverse effect is observed only at insufficient Al doses below 2 mg/L. Fluoride shows inhibitive effects towards the removal of Cd at p H 7 and 8 and that of phosphate at p H 6. Fluoride decreases the ζ-potential in both systems, and the decreasing extent is positively correlated to the elevated R F:Al. The Al fluoride interactions include the formation of Al–F complexes and the adsorption of fluoride onto Al(OH)_3 precipitates, i.e., the formation of Al(OH)n F m. Al–F complex formation inhibits Al hydrolysis and increases residual Al levels, and a more significant increase was observed at lower p H. Al–F complexes at high R F:Al complicate the coagulation behavior of Al towards both negative and positive ionic species. 以斜生栅藻(Scenedesmus obliquus JNU49)为研究对象,研究了重金属Pb2+、Cr3+、Cr6+对斜生栅藻生物量积累、光合性能及抗氧化系统的影响.结果表明,Pb2+、Cr3+、Cr6+对斜生栅藻的半数抑制浓度(IC50)分别为82.94、121.15、23.65μmol·L-1.半数抑制浓度胁迫下,斜生栅藻的放氧速率分别在第2~8 d(Pb2+胁迫)、1~3 d(Cr3+、Cr6+胁迫)受到明显抑制,而呼吸速率无明显变化;Pb2+胁迫下,斜生栅藻光系统Ⅱ(PSⅡ)的原初光能转化效率(Fv/Fm)、实际光能转化效率(Yield)、相对电子传递速率(rETR)均明显低于对照,光化学淬灭(qP)从第1~5 d高于对照,而第6~8 d则显著低于对照;Cr3+和Cr6+胁迫对斜生栅藻光合性能的影响不如Pb2+胁迫明显.斜生栅藻丙二醛(MDA)含量在重金属胁迫下变高,以第3、4 d尤为明显;超氧化物歧化酶(SOD)活性升高,于第3、4 d较为明显;过氧化氢酶(CAT)活性、总抗氧化活性(T-AOC)在重金属胁迫下均小幅度提升.通过本研究发现,斜生栅藻的生物量、光合速率、叶绿素荧光参数和SOD活性等指标对重金属胁迫的反应均较为灵敏,可作为评估重金属胁迫对微藻危害的指标.  相似文献

使用大庆油田石油污染土壤中分离出的优势菌种(蜡状芽孢杆菌DQ01和芽孢杆菌DQ02),在实验室可控条件下,研究了生物表面活性剂鼠李糖脂对微生物降解正十六烷以及降解菌生长、菌体表面疏水性和外表形态的影响.结果表明,鼠李糖脂可以提高正十六烷的降解率,48 h时蜡状芽孢杆菌DQ01和芽孢杆菌DQ02对正十六烷的降解率比未加鼠李糖脂的体系分别增加了8.1%和11.6%. 正十六烷培养基中的鼠李糖脂能引起菌体表面疏水性的明显增大,且低浓度鼠李糖脂的这种能力明显优于高浓度鼠李糖脂,蜡状芽孢杆菌DQ01和芽孢杆菌DQ02分别在加入0.4 mmol/L和0.2 mmol/L鼠李糖脂时疏水性最大. 尤其是芽孢杆菌DQ02在对数生长后期BATH(bacterial adherence to hydrocarbon)达到44%,远大于未加鼠李糖脂时的BATH. 但在葡萄糖培养基中鼠李糖脂并没有明显影响菌种的生物量和BATH. 另外,加入鼠李糖脂后, 2株菌的培养基接触角比未加鼠李糖脂的培养基接触角均减小了约一半. 未加入鼠李糖脂的菌体细胞表面比较光滑,且菌体之间独立生长,而加入鼠李糖脂后影响了降解菌的外表形态,菌体细胞表面略显粗糙且相互粘连,有助于细胞与疏水性有机物的接触.  相似文献

以全氟辛烷磺酸(PFOS)为目标去除物,选取牛血清蛋白(BSA)为典型蛋白质类有机物,考察了BSA及其浓度,以及BSA与无机离子共存时,离子强度、离子种类对聚酰胺纳滤膜去除水中PFOS的影响.研究发现,原液中存在BSA时,PFOS的去除率有显著提高,而且BSA浓度越高,PFOS的去除率越高;当BSA与无机离子共存时,离子强度越大,PFOS去除率越高.这可能是因为BSA不但会吸附一部分PFOS,还会造成膜污染,BSA浓度越大,膜污染越严重,膜的筛分能力越强,且膜面与PFOS之间的静电排斥力越大,从而提高了PFOS的截留率.而无机离子的存在减小了BSA分子之间及与膜面之间的静电排斥力,使BSA污染层更加厚实,进一步增强了膜的筛分能力.此外,Ca2+提高PFOS去除率的能力优于Na+.  相似文献

A Pt/WO3/TiO2 catalyst for propane oxidation was prepared by a stepwise wet impregnation method, and was aged at 800℃ for 5 hr. Compared to the sulfate-derived titania supported catalyst, the introduction of tungsten oxide as stable Bronsted acid sites led to the formation of more metallic platinum active sites at the Pt/WO3 interface. The dissociation of surface intermediates for propane oxidation was promoted on the WO3-modified catalyst. This, as well as the inhibition effects of tungsten oxide on the sintering of anatase and the phase transformation to rutile, resulting in a high activity and thermal stability for the Pt/WO3/TiO2 catalyst.  相似文献

采用盆栽试验模拟Cd污染土壤,研究施用烟秆炭(0、10、20 g·kg~(-1))对土壤pH、有机质含量、土壤Cd形态分布及烤烟Cd含量和Cd累积量的影响.结果表明:随着生育期的延长,土壤pH和有机质含量均呈现升高的趋势.在同一Cd污染水平下,土壤pH和有机质含量均随生物炭施用量的增加而升高.随生物炭施用量的增加,土壤有效态Cd和可交换态Cd含量显著降低,土壤碳酸盐结合态Cd、铁锰氧化物结合态Cd、有机结合态Cd和残渣态Cd含量升高.移栽后30 d,处理G2T2和G2T1的土壤有效态Cd含量的降幅分别达到45.59%和24.56%.外源添加Cd的土壤中,土壤pH和有机质含量均与土壤有效态Cd和可交换态Cd含量呈显著负相关关系(p0.01),与土壤碳酸盐结合态Cd、铁锰氧化物结合态Cd和有机结合态Cd含量呈显著正相关关系(p0.01).在Cd污染土壤上,施用生物炭能明显降低烟株各部位Cd含量,烟株各部位干物质积累量随生物炭施用量的增加显著升高,烟株各部位Cd累积量和总Cd累积量随生物炭施用量的增加呈降低的趋势.由此说明,在Cd污染的土壤上施用生物炭能够使土壤中可被植物吸收利用的有效态Cd转化为潜在的无效态Cd,从而达到修复土壤并减轻作物受Cd毒害的目的.  相似文献
