The expected increase in offshore oil exploration and production in the Arctic may lead to crude oil spills along arctic shorelines. To evaluate the potential effectiveness of bioremediation to treat such spills, oil spill bioremediation in arctic sediments was simulated in laboratory microcosms containing beach sediments from Barrow (Alaska), spiked with North Slope Crude, and incubated at varying temperatures and salinities. Biodegradation was measured via respiration rates (CO2 production); volatilization was quantified by gas chromatography/mass spectrophotometry (GC/MS) analysis of hydrocarbons sorbed to activated carbon, and hydrocarbons remaining in the sediment were quantified by GC/flame ionization detector (FID). Higher temperature leads to increased biodegradation by naturally occurring microorganisms, while the release of volatile organic compounds was similar at both temperatures. Increased salinity had a small positive impact on crude oil removal. At higher crude oil dosages, volatilization increased, however CO2 production did not. While only a small percentage of crude oil was completely biodegraded, a larger percentage was volatilized within 6–9 weeks.  相似文献

The improved in vitro bioremediation of Arabian Light crude oil in synthetic sea water by a mixed culture of bacteria is reported. Effects of fish meal, meat meal 80 % proteins and sophorolipids on the biodegradation were evaluated during a three-week incubation using GC/FID and GC/MS analysis of the remaining oil. Fish meal and more particularly meat meal 80 % proteins used at 0,5 mg.ml−1 involve no nitrogen and phosphorous limits when used as sole source of nutrients. Biodegradation processes in the first four days of the incubation supplemented with sophrolipids are accelerated two fold over controls.  相似文献

Given the difficulties caused by low-permeable soils in bioremediation, a new electrokinetic technology is proposed, based on laboratory results with phenanthrene, to afford bioremediation of polycyclic aromatic hydrocarbons (PAH) in clay soils. Microbial activity in a clay soil historically polluted with creosote was promoted using a specially designed electrokinetic cell with a permanent anode-to-cathode flow and controlled pH. The rates of phenanthrene losses during treatment were tenfold higher in soil treated with an electric field than in the control cells without current or microbial activity. Results from experiments with Tenax-assisted desorption and mineralization of 14C-labeled phenanthrene indicated that phenanthrene biodegradation was limited by mass-transfer of the chemical. We suggest that the enhancement effect of the applied electric field on phenanthrene biodegradation resulted from mobilization of the PAH and nutrients dissolved in the soil fluids.  相似文献

Oily drill cuttings and a soil contaminated with weathered crude oils were treated by enhanced biodegradation under tropical conditions in industrial scaled experiments. Oil contaminants were characterized by gas chromatography and mass spectrometry. This allowed for the identification of a mixture of two crude oils in the contaminated soil. After 12 months of bioremediation process, the removal of hydrocarbons reached by biodegradation an extent of 60% although nutrient amendment with elevated concentration of N-urea had highly detrimental effects on the hydrocarbon degrading fungal populations due to the production of toxic concentration of ammonia gas by nitrification. The saturated hydrocarbons were extensively assimilated, though n-alkanes were not completely removed. Aromatic hydrocarbons were less degraded than saturated whereas resin and asphaltene fractions were, surprisingly, partly assimilated. In laboratory conditions, the residual hydrocarbons in the field-treated materials were 15-20% further degraded when metabolic byproducts resulting from biodegradation were diluted or removed.  相似文献

Background, Aims and Scope It is well known that the composition of petroleum or some of its processing products changes in the environment mostly under the influence of microorganisms. A series of experiments was conducted in order to define the optimum conditions for an efficient biodegradation of petroleum pollutant, or bioremediation of different segments of the environment. The aim of these investigations was to show to what extent the hydrocarbons of a petroleum pollutant are degraded by microbial cultures which were isolated as dominant microorganisms from a surface water of a wastewater canal of an oil refinery and a nitrogen plant. Biodegradation experiments were conducted on one paraffinic, and one naphthenic type of petroleum during a three month period under aerobic conditions, varying the following parameters: Inorganic (Kp) or an organic medium (Bh) with or without exposition to light. Methods Microorganisms were analyzed in a surface water sample from a canal (Pančevo, Serbia), into which wastewater from an oil refinery and a nitrogen plant is released. The consortia of microorganisms were isolated from the water sample (most abundant species: Phormidium foveolarum - filamentous Cyanobacteria, blue-green algae and Achanthes minutissima, diatoms, algae). The simulation experiments of biodegradation were conducted with the biomass suspension and crude oils Sirakovo (Sir, paraffinic type) and Velebit (Ve, naphthenic type). After a three month period, organic substance was extracted by means of chloroform. In the extracts, the content of saturated hydrocarbons, aromatic hydrocarbons, alcohols and fatty acids was determined (the group composition). n-Alkanes and isoprenoid aliphatic alkanes, pristane and phytane, in the aliphatic fractions, were analyzed using gas chromatography (GC). Total isoprenoid aliphatic alkanes and polycyclic alkanes of sterane and triterpane types were analyzed by GC-MS. Results and discussion. Paraffinic type petroleums have a significant loss of saturated hydrocarbons. For naphthenic type petroleum, such a trend has not been observed. The most intensive degradation of n-alkanes and isoprenoid aliphatic alkanes (in paraffinic oil) and isoprenoids (in naphthenic oil) was observed using the inorganic medium Kp in the light; the microbial conversion is somewhat lower with Kp in the dark; with organic medium Bh in the light the degradation is of low intensity; with the same medium in the dark the degradation is hardly to be seen. Steranes and triterpanes were not affected by microbial degradation under the conditions used in our experiments. Obviously, the petroleum biodegradation was restricted to the acyclic aliphatics (n-alkanes and isoprenoids). Conclusion Phormidium foveolarum (filamentous Cyanobacteria - blue-green algae) and Achanthes minutissima (diatoms, algae), microbial cultures isolated as dominant algae from a surface water in a wastewater canal of an oil refinery and a nitrogen plant, have degradable effects dominantly involving petroleum hydocarbons. Petroleum microbiological degradation is more intensive when inorganic medium (in the light) is applied. Having in mind that the inorganic pollutants have been released into the canal as well, this medium reflects more the natural environmental conditions. Polycyclic alkanes of sterane and triterpane type, in spite of the fact that these compounds could be degraded, have remained unchanged regarding abundance and distribution. Since this is the case even for naphthenic type petroleum (which is depleted in n-alkanes), it can be concluded that the biodegradation of petroleum type pollutants, under natural conditions, will be restrained to the n-alkane and isoprenoid degradation. Recommendation and Outlook Performed experiments and simulations of petroleum microbiological degradation may serve for the prediction of the fate of petroleum type pollutants, as well as for definition of conditions for bioremediation of some environmental segments.  相似文献

When hydrocarbon-contaminated soil is subjected to bioremediation technology, hydrocarbon depletion is typically marked by an initially rapid reduction rate. This rate decreases over time and frequently a residual concentration remains in the soil. This kinetic has been attributed primarily to the enrichment of more recalcitrant fractions, as well as to the lack of resting hydrocarbon bioavailability. Thus, at the end of the bioremediation process, a part of the residual hydrocarbon soil concentration represents the non-bioavailable fraction, which is difficult to degrade by microbial populations and which poses a minor hazard. Therefore, determination of the bioavailable fraction in a bioremediation project represents both an estimation of the maximum level of achievable biodegradation, as well as an additional indication of the environmental health hazard. In the present study, aged creosote-contaminated soil was subjected to biostimulation processes, and the bioavailable fraction for several target polycyclic aromatic hydrocarbons (PAHs) was calculated using a mild extraction with cyclodextrines. The amount of PAH extracted corresponded to the desorbing fraction and can be regarded as the bioavailable fraction. The non-desorbing fraction data obtained from this procedure were compared to the remaining PAH concentrations following bioremediation treatment of soil microcosms. These results permitted the establishment of a theoretical biodegradation limit based on the desorbing fraction. In addition, neither accumulation of intermediate metabolites, nor the formation of bound-residues or reduced acute toxicity was observed.  相似文献

Effects of remediation technologies on polar compounds of crude oil in contaminated soils have not been well understood when compared to hydrocarbons. In this study, ultrahigh resolution Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) was used to characterize the changes in NSO polar compounds of crude oil and residual oil after long-term natural attenuation, biostimulation and subsequent ozonation following biostimulation of contaminated soils. N1 and O1 species, which were abundant in the crude oil, were selectively biodegraded, and species with higher double bond equivalent values and smaller carbon numbers appeared to be more resistant to microbial alteration. O2-O6 species were enriched by biodegradation and contained a large number of compounds with a high degree of unsaturation. Ozone could react with a variety of polar compounds in residual oil after biodegradation and showed high reactivity with polar species containing aromatic or multi-aliphatic rings, including the residual N1 and O1 species, naphthenic acids and unsaturated O3-O6 compounds. Fatty acids and O3-O8 species dominated by saturated alkyl compounds were resistant to ozonation or the primarily incomplete ozonation products. Principal component analysis of identified peaks in the FT-ICR MS spectra provided a comprehensive overview of the complex samples at the molecular level and the results were consistent with the detailed analysis. Taken together, these results showed the high complexity of polar compounds in residual oils after biodegradation or ozonation in contaminated soil and would contribute to a better understanding of bioremediation and ozonation processes.  相似文献

GOAL, SCOPE AND BACKGROUND: Pentachlorophenol (PCP) is the second highest volume pesticide used in the United States. It is a mutagenic compound whose exposure poses significant health effects, One of the most desirable, environmentally friendly treatment methods is bioremediation. For soil-based contamination, the effectiveness of bioremediation will also be affected by the presence of an active indigenous population, sorption of the contaminant onto the soil, and environmental parameters. METHODS: Two pure strains and their mixed culture were used to evaluate PCP biodegradation in two different field soils, Columbia (CO) and New Mexico (NM). Biostimulation of the indigenous microbes was evaluated by adding nutrients. The efficiency of adding bacteria strains (bioaugmentation) for degrading PCP was determined with Arthrobacter sp., Flavobacterium sp. and a 50:50 mixture of the two bacteria strains. RESULTS: In CO soil, only 24%, 12% and 25% of the initial PCP concentration were degraded by Flavobacterium sp., Arthrobacter sp. and mixed culture, respectively. Arthrobacter sp. was used in NM soil with two initial concentrations and achieved degradation efficiencies of 57% and 61% for 361 and 95 mg kg- concentrations, respectively. Discussion. Analysis via statistical methods showed that the bacteria had different efficiencies on PCP degradation in each soil. 2 CONCLUSIONS: All bacteria catalyzed a higher PCP degradation when present in NM soil. Second, Flavobacterium sp. degraded more PCP than Arthrobacter sp. in CO soil. The mixed culture achieved the highest degradation efficiency regardless of the initial concentration or soil origin. RECOMMENDATIONS AND PERSPECTIVES: The effect of the soil properties, such as the soil organic matter (SOM) on PCP biodegradation should be investigated. Future work can also investigate the effect of aging time on biodegradation.  相似文献

以混合柴油为靶污染物,通过对比实验研究了油污染物在模拟水环境中的降解效果。研究表明,模拟自然条件下混合柴油污染物总体降解较慢,油质去除率低;生物强化降解条件下,向混合柴油污染水样中添加驯化培养的微生物混合菌群,生物降解速率明显提高,油质去除率达到98%以上。研究还发现,各污染水样中油的降解速率与降解效果随柴油的配比而不同,混合柴油样本中生物柴油的比例越高,样本的降解率越高,表明生物柴油作为碳源有效改善了水中有机营养配比,促进了柴油的去除效果。进一步分析表明,混合柴油在水中的降解过程符合一级反应动力学,生物强化降解条件下,生物柴油比例越高,混合柴油降解速率越快,除油微生物以菌胶团、球菌和丝状菌为主。  相似文献

Mass-transfer models and biodegradation models were developed for three theoretical physical states of polycyclic aromatic hydrocarbons (PAHs) in soil. These mechanistic models were used to calculate the treatment periods necessary for complete removal of the PAH pollutants from the soil under batch conditions. Results indicate that the bioremediation of PAHs in such systems is mainly mass-transfer limited. The potential for bioremediation as a treatment technique for PAH contaminated soils is therefore mainly determined by the mass-transfer dynamics of PAHs. Under mass-transfer limited conditions simplified mathematical models, based on the assumption of a zero dissolved PAH concentrations, can be used to predict the period of time needed for complete bioremediation.  相似文献

SCOPE AND BACKGROUND: Contamination of soils, aquifers and groundwater by nonaqueous phase liquid (NAPL) pollutants constitutes a major environmental issue of concern, worldwide. The residual (biodegradation-resistant) hydrophobic fuel hydrocarbons entrapped in the soil porous matrix, possess a particular bioremediation challenge due to their becoming virtually immobile, nor desorbable, or water dispersible. Consequently, they are not available as substrates to the micro-organism-based biodegradation. MATERIALS AND METHODS: Our research involves the development of economically feasible, surfactant/surfactant-nutrient mix (SSNM)-enhanced bioremediation methodologies for sustainable, in situ bioremediation of fuel-contaminated aquifers. This requires, methodologically, (a) the optimization, via in vitro 'flow' (columns) lab experiments and screening processes, of an effective mixture for the intended SSNM-enhanced bioremediation; and (b) the study of the combined effect of the optimized SSNM on the solubilization/mobilization and biodegradation of NAPL (fuel) in in vitro site/aquifer-simulated bioremediation. RESULTS AND DISCUSSION: The essence of our findings: (1) kerosene's maximum enhanced mobilization - f = 3.6, compared with that of deionized water, was achieved with an SSNM having the composition of linear alkylbenzene sulfonate (LABS): coco-amphodiacetate (containing N): surfactant-nutrient X (containing both N and P) = 0.15: 0.15: 0.05 g/L, respectively; (2) 62-64% of the initial amount of kerosene in the initially saturated soil matrix, 'packed' in a column, has been eluted from it during approximately 30 days, compared with 68% of kerosene biodegradation in 'vessel' settings, in 21 days. CONCLUSIONS: (1) The indigenous microorganisms present in th vadose zones of fuel-contaminated sandy soil aquifers are potentially capable of unassisted removal of approximately 80% of the initially contained fuel (kerosene), during a period of about 42 days; (2) the major effects of the SSNM addition are (a) enhanced mobilization of the bulky NAPL; and (b) enhanced desorbtion/ solubilization/dispersion of the entrapped NAPL which, in turn, facilitate their enhanced biodegradation. RECOMMENDATIONS AND PERSPECTIVE: Our findings suggest that pre-optimized, biodegradable SSNM is essential for surfactants-based bioremediation of NAPL-contaminated aquifers, in order to make this in-situ methodology both technologically and economically feasible.  相似文献

Different aspects of bacterial degradation of organic contaminants in soil, and how to improve the efficiency and reproducibility is discussed in this review. Although bioremediation in principle includes the use of any type of organism in improving the condition of a contaminated site, most commonly bacteria are the degraders and other organisms, such as soil animals or plant roots, play a role in dissemination of bacteria and, indirectly, plasmids between bacteria, and in providing nutrients and co-substrates for the bacteria active in the degradation process. There are a number of different procedures that have been tested more-or-less successfully in attempts to improve reliability, cost efficiency and speed of bioremediation. The methods range from minimal intervention, such as mere monitoring of intrinsic bioremediation, through in situ introduction of nutrients and/or bacterial inocula or improvement of physico-chemical conditions, all the way to excavation followed by on site or ex situ composting in its different varieties. In the past the rule has been that more intervention (leading to higher costs) has been more reliable, but novel ideas are continuously tried out, both as a means to come up with new truly functional applications and also as a line of studies in basic soil microbial ecology. Both approaches generate valuable information needed when predicting outcome of remediation activities, evaluating environmental risks, deciding on cleaning-up approaches, etc. The emphasis of this review is to discuss some of the novel methods for which the value has not been clearly shown, but that in our view merit continued studies and efforts to make them work, separately or in combination.  相似文献

In recent years, microbial degradation and bioremediation approaches of polychlorinated biphenyls (PCBs) have been studied extensively considering their toxicity, carcinogenicity and persistency potential in the environment. In this direction, different catabolic enzymes have been identified and reported for biodegradation of different PCB congeners along with optimization of biological processes. A genome analysis of PCB-degrading bacteria has led in an improved understanding of their metabolic potential and adaptation to stressful conditions. However, many stones in this area are left unturned. For example, the role and diversity of uncultivable microbes in PCB degradation are still not fully understood. Improved knowledge and understanding on this front will open up new avenues for improved bioremediation technologies which will bring economic, environmental and societal benefits. This article highlights on recent advances in bioremediation of PCBs in soil. It is demonstrated that bioremediation is the most effective and innovative technology which includes biostimulation, bioaugmentation, phytoremediation and rhizoremediation and acts as a model solution for pollution abatement. More recently, transgenic plants and genetically modified microorganisms have proved to be revolutionary in the bioremediation of PCBs. Additionally, other important aspects such as pretreatment using chemical/physical agents for enhanced biodegradation are also addressed. Efforts have been made to identify challenges, research gaps and necessary approaches which in future, can be harnessed for successful use of bioremediation under field conditions. Emphases have been given on the quality/efficiency of bioremediation technology and its related cost which determines its ultimate acceptability.  相似文献

Dispersants are important tools in oil spill response. Taking advantage of the energy in even small waves, they disperse floating oil slicks into tiny droplets (<70 μm) that entrain in the water column and drift apart so that they do not re-agglomerate to re-form a floating slick. The dramatically increased surface area allows microbial access to much more of the oil, and diffusion and dilution lead to oil concentrations where natural background levels of biologically available oxygen, nitrogen, and phosphorus are sufficient for microbial growth and oil consumption. Dispersants are only used on substantial spills in relatively deep water (usually >10 m), conditions that are impossible to replicate in the laboratory. To date, laboratory experiments aimed at following the biodegradation of dispersed oil usually show only minimal stimulation of the rate of biodegradation, but principally because the oil in these experiments disperses fairly effectively without dispersant. What is needed is a test protocol that allows comparison between an untreated slick that remains on the water surface during the entire biodegradation study and dispersant-treated oil that remains in the water column as small dispersed oil droplets. We show here that when this is accomplished, the rate of biodegradation is dramatically stimulated by an effective dispersant, Corexit 9500®. Further development of this approach might result in a useful tool for comparing the full benefits of different dispersants.  相似文献

This study investigates the fungal biodegradation of fluorene, a polycyclic aromatic hydrocarbon, in liquid medium and soil slurry. Fungal strains and cyclodextrins were used in order to degrade fluorene and optimize fluorene bioavailability and degradation in soil slurries. After a procedure of selection in solid and liquid media, maltosyl-cyclodextrin, a branched cyclodextrin was chosen. 47 fungal strains isolated from a contaminated site were tested for biodegradation. Results showed the greater efficiency of "adapted" fungi isolated from contaminated soil vs reference strains belonging to the collection of the laboratory. These assays allowed us to select the most efficient strain, Absidia cylindrospora, which was used in a bioaugmentation process. Bioaugmentation tests were performed in an artificially contaminated non-sterile soil. In the presence of A. cylindrospora, more than 90% of the fluorene was degraded within 288 h, while 576 h were necessary in the absence of fungal bioremediation. It also appeared that biodegradation was enhanced by amendment with previously selected maltosyl-cyclodextrin. The results of this study indicate that A. cylindrospora and maltosyl-cyclodextrin could be used successfully in fluorene bioremediation systems.  相似文献

In situ bioremediation is a safe and cost-effective technology for the cleanup of contaminated sites, but its remediation rate is usually very slow. This study attempted to accelerate the process of bioremediation by employing non-uniform electrokinetic transport processes to mix organic pollutants and degrading bacteria in soils under in situ conditions (namely, in situ bioelectrokinetic remediation) by use of an electrode matrix and a rotational operation mode. A bench-scale non-uniform electrokinetic system with periodic polarity-reversal was developed for this purpose, and tested by using a sandy loam spiked with phenol as a model organic pollutant. The results demonstrated that non-uniform electrokinetic processes could enhance the in situ biodegradation of phenol in the soil, the efficiency of which depended upon the operational mode of the electric field. Compared with the unidirectional operation and the bidirectional operation, the rotational operation could effectively stimulate the biodegradation of phenol in the soil if adopting appropriate time intervals of polarity-reversal and electrode matrixes. A reversal interval of 3.0 h and a square-shaped electrode matrix with four electrode couples appeared appropriate for the in situ biodegradation of phenol, at which a maximum phenol removal of 58% was achieved in 10d and the bioremediation rate was increased about five times as compared to that with no electric field applied. The results also showed that adopting a small polarity-reversal interval and an appropriate electrode array could produce a high and uniform removal of phenol from the soil. It is believed that in situ bioelectrokinetic remediation holds the potential for field application.  相似文献

通过在堆肥中加入经过驯化的降解菌这种土壤有机污染生物修复技术 ,对堆肥中多环芳烃的浓度变化进行监测 ,从而了解降解菌对堆肥中多环芳烃的降解作用。实验结果表明 ,降解菌的加入能明显地提高多环芳烃的降解率 ,本次实验中 ,菲、芴的去除率提高了 2 5 %左右 ,芘的去除率提高了约 4 5 %。  相似文献
