The causes of disease in cultured shrimp are difficult to ascertain but there is evidence that disease is correlated with environmental factors. Crustaceans are particularly sensitive to insecticides due to their close phylogenetic relationship with insects. The objective of this study was to investigate whether there was an increased susceptibility of the shrimp Litopenaeus vannamei to Vibrio parahaemolyticus, when exposed to methyl parathion. The outline of the study was the following: An LC50 96 h was determined to methyl parathion orally offered to juvenile shrimp. Further experiments were carried out in order to determine a concentration that affected the shrimp (verified by measuring the acethylcholinesterase activity) while producing minimal mortalities. This sublethal concentration was used in a susceptibility experiment where methyl parathion was offered to shrimp which were later injected with V. parahaemolyticus in a dose expected to kill less than 15%. Probit analysis estimated a 96 h median lethal concentration (LC50) of 1.56 microg g(-1). Mortality and AChE activity showed a concentration-response relationship in the exposure treatments. The median inhibitory concentration (IC50) determined was 0.029 microg g(-1). These results suggested that a concentration of 0.1 microg g(-1) was appropriate for the bacteria-pesticide interaction test as it was able to elicit 11.1% mortality after 10 days of exposure, while producing an AChE inhibition of 57.12%. Cumulative mortalities were significantly increased (P<0.01) in the treatment that combined exposure to methyl parathion and V. parahaemolyticus (35.19%) in comparison with methyl parathion or V. parahaemolyticus alone (9.26% and 7.41%, respectively).  相似文献

The white shrimp, Litopenaeus vannamei, a globally important cultured prawn species, is an ideal animal for studying the impairment caused by the effects of heavy metals that are often detected in coastal areas. In this study, L. vannamei was exposed to different concentrations of cadmium (Cd) and zinc (Zn) for up to 28 d. Histopathological alterations in the hepatopancreas were observed in L. vannamei after long-term exposure to Cd and Zn. Hepatopancreatic injury was further confirmed by the inductions of two biochemical markers, hemolymphatic glutamate-oxalacetate transaminase (GOT) and glutamate-pyruvate transaminase (GPT). It was notable that L. vannamei was able to repair its hepatopancreas from the damage caused by Zn, which was evidenced by the results of the histopathological observations, determinations of tissue metal concentrations, and examination of GOT and GPT levels.  相似文献

In this study, a cohort of farmers from the Mateur region in the North of Tunisia, were interviewed and examined for the biochemical effects of pesticides. We studied their haematological profile, lipid parameters, serum markers of nephrotoxicity and hepatotoxicity. We also evaluated the activities of Butyrylcholinesterase (BChE), Acetylcholinesterase (AChE) and thiolactonase-paroxonase (PON). Moreover, lipid peroxidation and activities of antioxidant enzymes superoxide dismutase (SOD) and catalase (CAT) were determined. The duration of pesticide use and the farmers’ age were considered in the analysis. Our results revealed significant differences in some haematological parameters, in liver and kidney functions, in the lipidic status of the pesticide-exposed group. We also reported an increase in the index of incidence of cardiovascular risk in farmer populations. A significant decrease in AChE, BChE and PON levels was found among farmers. Lipid peroxidation, however, increased. The activities of SOD and CAT were remarkably elevated in farmer populations. There was a significant relation between changes in biological markers, the duration of pesticide use and the farmers’ age. This study indicates that a long-term exposure to pesticides may play an important role in the development of vascular diseases via metabolic disorders of lipoproteins, lipid peroxidation and oxidative stress, inhibition of BChE and decrease in thiolactonase-PON levels.  相似文献

The main purpose of the present study was to detect the acute toxicity of Cd and Zn to L. vannamei, and to investigate their effects on gill functions, which have not been carried out in this species before, hoping to obtain some important significance. First of all, we examined the acute toxicity of cadmium (Cd) and zinc (Zn) to L. vannamei and obtained 24-, 48-, 72-, and 96-h medium lethal concentration (LC50) values of 2.58, 1.30, 1.14, and 1.07 mg Cd l−1 and 3.98, 2.14, 1.75, and 1.35 mg Zn l−1, respectively. Furthermore, we also found that exposure of shrimp to Cd or Zn caused an inhibition in oxygen consumption of 91.3% and 75.9% lower than that of the control. However, after separate exposure to Cd or Zn, elevations in ammonium excretion were obtained, which were 153.7% and 144.1%, respectively, higher than the control. It is most likely that elevations in ammonium excretion were related to decreased osmotic pressure of shrimp blood. Finally, the distribution of Cd and Zn within gills was also determined.  相似文献

This study aimed to test the susceptibility of Escherichia coli strains isolated from the water, bottom sediments and individuals cultivated in shrimp farm ponds, to antibiotics belonging to different families, namely B-Lactams: Imipenem (IPM; 10 μ g), Ampicillin (AMP; 10 μ g), Cephalothin (CEP; 30 μ g), Cefoxitin (FOX; 30 μ g), Ceftriaxone (CRO; 30 μ g); Tetracycline: Tetracycline (TCY; 30 μ g); Aminoglycosides: Gentamicin (GEN; 10 μ g), Amikacin (AMK; 30 μ g); Chloramphenicol: Chloramphenicol (CHO; 30 μ g); Fluoroquinolones: Ciprofloxacin (CIP; 5 μ g); Nitrofurans: Nitrofurantoin (NIT; 300 μ g); Sulfonamides: Trimethoprim-Sulfamethoxazole (SXT; 30 μ g); Quilononas: Nalidixic Acid (NAL; 30 μ g). In the laboratory, the method of dissemination (Test Kirby-Bauer) was performed in order to fulfill the antibiogram tests. The results showed high indices of resistance to Imipenem, Cephalothin and Ampicillin. Chloramphenicol, Nitrofurantoin, Cefoxitin, Ceftiaxone and Ciprofloxacin have displayed the highest index of sensitive strains. The antibiotic resistance index (ARI) and the multiple resistance index (MAR) varied within the ranges of 0.068–0.077 and 0.15–0.39, respectively. More than 90.5% of strains of Escherichia coli showed a variety of resistance profiles to the tested antibiotics. The high indices of resistance may be a consequence of indiscriminate use of antibiotics, but also the transfer of resistance through mobile genetic elements found in shrimp farms.  相似文献

T-2 toxin (T-2), one of the naturally occurring mycotoxins, often accumulates in aquatic animals from contaminated feed. Shrimp (n?=?30 per group) were fed with different concentrations (0, 0.5, 1.5, 4.5 and 13.5?mg kg?1) of T-2 for 20?days. Changes in histopathology, fatty acid and water distribution of shrimp muscle were analyzed. Histopathology of shrimp muscle showed dose-dependent marked degenerative and necrotic changes on exposure to dietary T-2. The T-2 significantly (P?<?0.05) affected the muscle fatty acid composition. ∑SFA, ∑MUFA and ∑PUFA initially decreased and then increased slowly in the high-dosed groups. C16:0, C18:1n-9 and C18:2n-6 were the main saturated fatty acid (SFA), monounsaturated fatty acid (MUFA) and polyunsaturated fatty acid (PUFA), respectively. Also, T-2 significantly affected water distribution in shrimp muscle. High doses of T-2 reduced free water content, resulting in a reduction in the water holding capacity and hence changes to the shrimp muscle quality. Collectively, these results illustrated that T-2 significantly affects the fatty acid and water distribution, and also muscle histopathology, all of which would result in a reduction in the quality and nutritional value of shrimp.  相似文献

Molting in crustaceans is an important endocrine-controlled biological process that plays a critical role in growth and reproduction. Many factors can affect this physiological cycle in crustaceans including environmental stressors and disease agents. For example the pathology of Taura Syndrome Virus (TSV) of shrimp is closely related to molting cycle. Similarly, endosulfan, a commonly used pesticide is a potential endocrine disruptor. This study explores interrelationships between pesticide exposure, virus infection and their interactions with physiology and susceptibility of the shrimp. Litopenaeus vannamei (Pacific white shrimp) were challenged with increasing doses of endosulfan and TSV (TSV-C, a Belize reference strain) to determine the respective median lethal concentrations (LC50s). The 96-h endosulfan LC50 was 5.32 μg L−1, while the 7-d TSV LC50 was 54.74 mg L−1. Subsequently, based on their respective LC50 values, a 20-d interaction experiment with sublethal concentrations of endosulfan (2 μg L−1) and TSV (30 mg L−1) confirmed a significant interaction (p < 0.05, χ2 = 5.29), and thereby the susceptibility of the shrimp. Concurrently, molt-stage of animals, both at the time of exposure and death, was compared with mortality. For animals challenged with TSV, no strong correlation between molt-stage and mortality was observed (p > 0.05). For animals exposed to endosulfan, animals in the postmolt stage were shown to be more susceptible to acute toxicity (p < 0.05). For animals exposed to both TSV and endosulfan, interference of endosulfan-associated stress lead to increasingly higher susceptibility at postmolt (p < 0.05) during the acute phase of the TSV disease cycle.  相似文献

The diffusive gradients in thin films (DGT) were field deployed alongside the shrimp Litopenaeus vannamei at seven sites with different levels of contamination to assess the potentially bioavailable and toxic fraction of metal contaminants. After 7 days of exposure, several antioxidant biomarkers were quantified in hepatopancreas of exposed shrimps, and tissue levels as well as the total, dissolved, and DGT-labile concentrations of metal contaminants were determined in the pooled site samples. The results showed that the caged shrimps had high tissue contaminant concentrations and significantly inhibited antioxidant responses at the more contaminated sites. DGT-labile metal concentrations provided better spatial resolution of differences in metal contamination when compared with traditional bottle sampling and transplanted shrimp. The total, dissolved, and DGT-labile metal fractions were used to evaluate the potential bioavailability of metal contaminants, comparing with metal accumulation and further linking to antioxidant biomarker responses in tissues of exposed shrimps. Regression analysis showed the significant correlations between DGT-Cu concentrations and tissue-Cu and activities of some biomarker responses in the shrimp hepatopancreas. This indicated that DGT-labile Cu concentrations provided the better prediction of produced biological effects and of the bioavailability than the total or dissolved concentrations. The study supports the use of methods combining transplanted organisms and passive sampling for assessing the chemical and ecotoxicological status of aqueous environments and demonstrates the capability of the DGT technique as a powerful tool for measuring the bioavailability-based water quality in variable coastal environments.  相似文献

Trace-metal concentrations (Fe, Mn, Ni, Cu, Co, Cd, Cr, and Zn) were measured in estuarine postlarvae, juveniles, and marine adults of the shrimp Penaeus vannamei collected along the Pacific coast of Mexico. With the exception of Fe, the trace-metal concentrations fell within the ranges of similar organisms collected elsewhere. Size-dependent relationships were observed only for Ni, Fe, and Zn and varied depending on the element considered. Small individuals had higher concentrations of Fe and Ni than larger individuals. For Zn, the opposite tendency occurred. This may reflect different metabolic requirements of young and old shrimps.  相似文献

Histopathological alterations in gill, liver and kidney of common carp, Cyprinus carpio, intoxicated with sub-lethal concentrations of chlorpyrifos (O,O,-diethyl-O-3,5,6-trichloro-2-pyridyl phosphorothioate) pesticide (1 and 100 μg/L) for a period of 14 days were analyzed under light microscope. Gill exhibited hyperplasia and hypertrophy of gill epithelium, blood congestion, dilation of marginal channel, epithelial lifting, lamellar fusion, lamellar disorganization, lamellar aneurysm, rupture of the lamellar epithelium, rupture of pillar cells and necrosis. Alterations in hepatocytes were more pronounced, including nuclear and cellular hypertrophy, cellular atrophy, irregular contour of cells and nucleus, cytoplasmic vacuolation, cytoplasmic and nuclear degeneration, cellular rupture, pyknotic nucleus, necrosis and melanomacrophages aggregations. Histopathological lesions in kidney were cellular and nuclear hypertrophy, narrowing of tubular lumen, cytoplasmic vacuolation, hyaline droplet degeneration, nuclear degeneration, occlusion of tubular lumen, tubular regeneration, dilation of glomerular capillaries, degeneration of glomerulus and hemorrhage in Bowman's space. The most significant conclusion drawn from this study was that with the increased concentration and duration the toxicosis of chlorpyrifos would be enhanced as shown through the analysis of mean assessment value (MAV) and degree of tissue changes (DTC) also.  相似文献

Chronic low-level exposure to synthetic pesticides is implicated in many health conditions that result from the induction of oxidative stress, including cytogenetic damage. The objective of this study was to assess the risk of genotoxicity using micronuclei (MN) formation in lymphocytes and to determine changes in blood antioxidants superoxide dismutase (SOD) in erythrocytes (E) and glutathione (GSH) in E and plasma (PL) in farm workers for six months during a growing season. Blood and urine samples were collected once a month for six months (June to November 2003) from farm workers (n = 15) and urban unexposed controls (n = 10). Lymphocytes from blood were separated by density gradient centrifugation using Histopaque and cultured using the standard technique. There was no significant difference in the cytokinesis blocked proliferation index (CBPI) of lymphocytes between the farm workers and the control group, but there was a 76% increase in average MN frequency in lymphocytes of the farm worker group (P 相似文献

The susceptibility of 2-week-old individuals of the freshwater shrimp, Paratya compressa improvisa, to five kinds of insecticide and five kinds of herbicide was examined in comparison with that of two species of Cladocera, Daphnia magna and Moina macrocopa. The shrimp was especially sensitive to two organophosphorus insecticides. The 48-h LC50 values for fenitrothion and fenthion to the shrimp were 1.15 and 1.04 microg litre(-1) (mean value, n=2), in contrast with 37.8 and 35.3 microg litre(-1) in the case of M. macrocopa, and more than 50 microg litre(-1) with D. magna. The shrimp also showed the higher susceptibility to other insecticides, diazinon, carbaryl (NAC) and BPMC, apart from D. magna to diazinon and NAC. The shrimp also showed higher susceptibility to herbicides. The 48-h LC50 values of CNP, benthiocarb, oxadiazon, butachlor, and symetryne to the shrimp were two to eight times lower than those of two species of Cladocera, except for the LC50 value of oxadiazon to M. macrocopa, which was very slightly higher. However, the shrimp showed a somewhat lower susceptibility to heavy metals than the two species of Cladocera, especially to copper, and to cadmium and zinc in comparison with D. magna. A bioassay using the shrimp with river water, collected from the river adjacent to the paddy field, showed clearly the high mortality of the shrimp following the aerial spraying with pesticides.  相似文献

This study investigated the effects of increased temperature and salinity, two potential impacts of global climate change, on the toxicity of two common pesticides to the estuarine grass shrimp, Palaemonetes pugio. Larval and adult grass shrimp were exposed to the fungicide chlorothalonil and the insecticide Scourge® under standard toxicity test conditions, a 10°C increase in temperature, a 10 ppt increase in salinity, and a combined increased temperature and salinity exposure. Toxicity of the fungicide chlorothalonil increased with temperature and salinity. Toxicity of the insecticide Scourge® also increased with temperature; while increased salinity reduced Scourge® toxicity, but only in adult shrimp. These findings suggest that changes in temperature and salinity may alter the toxicity of certain pesticides, and that the nature of the effect will depend on both the organism's life stage and the chemical contaminant. Standard toxicity bioassays may not be predictive of actual pesticide toxicity under variable environmental conditions, and testing under a wider range of exposure conditions could improve the accuracy of chemical risk assessments.  相似文献

This study aims to identify levels of several organochlorine and organophosphorus compounds in shrimp-raising areas of coastal El Salvador, to assess potential impacts on shrimp growth and survival that hamper the sustainability of aquaculture in the region. The paper reports the current levels of γ-HCH, 4,4'-DDT, 4,4'-DDE, 4,4'-DDD, endrin, dieldrin, heptachlor, parathion, methyl parathion, and etoprophos in soils (depth 20 cm), sediments (depth 5 cm), shrimp (Penaeus sp.), and water of three rearing ponds and also in the sediment (depth 5 cm) and water surrounding those ponds in Jiquilisco Bay. Sampling was carried out during the dry (January-March) and rainy (June-August) seasons of 2008. The presence of pesticides in the samples of water, shrimp, and sediment at shrimp ponds was not detected in either season; however, in soil samples (depth 20 cm) taken from these ponds, heptachlor, endrin, dieldrin, 4,4'-DDD, and 4,4'-DDT were identified at concentrations below the method limit of quantification (LOQ), and 4,4'-DDE was found in a concentration falling in the range from 3.85 to 19.61 ng/g. In samples of water taken at the bay water intakes to the rearing ponds, we observed dieldrin concentrations in the range between 0.085 ng/mL and 0.182 ng/mL during the dry season. In the samples of sediments taken in the surrounding areas of shrimp ponds, we found-for both seasons-that in 60 % of the samples, 4,4'-DDE was present in concentrations ranging from 3.75 ng/g to 30.97 ng/g. Additionally, in the rainy season, we observed heptachlor in sediment at concentrations below the method quantification limit. It was concluded that organochlorine compounds from pesticides are still present in Jiquilisco Bay, trapped in deep sediment, even though they have been banned since the 1980s. These were not detected in shrimp tissue, surface water, and shallow sediment in rearing ponds, and hence, we do not believe their presence has any major impact on shrimp production in sampled areas.  相似文献
