Metal concentrations (Al, Ba, Ca, K, Li, Mg, Na, Se, Sr and Ti) in submerged macrophytes and corresponding water and sediments were studied in 24 eutrophic lakes along the middle and lower reaches of the Yangtze River (China). Results showed that these eutrophic lakes have high metal concentrations in both water and sediments because of human activities. Average concentrations of Al and Na in tissues of submerged macrophytes were very high in sampled eutrophic lakes. By comparison, Ceratophyllum demersum and Najas marina accumulated more metals (e.g. Ba, Ca, K, Mg, Na, Sr and Ti). Strong positive correlations were found between metal concentrations in tissues of submerged macrophytes, probably because of co-accumulation of metals. The concentrations of Li, Mg, Na and Sr in tissues of submerged macrophytes significantly correlated with their corresponding water values, but not sediment values.  相似文献

Elsholtzia splendens is a well-known Cu-tolerant plant; yet, the impact of Cu-contaminated soil on bacterial community in its rhizosphere is not known. We studied the spatial variability of bacteria in the rhizosphere using Cu-contaminated soil with polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) and real-time PCR. In the uncontaminated soil, the content of the dissolved organic carbon (DOC) and bacterial diversity gradually increased in the rhizosphere soil along the root growth direction (from the interface zone to the meristematic zone), while for the Cu-contaminated soil, the highest DOC content and the strongest potential bioavailability of Cu were found in the interface zone, which also had the lowest bacteria diversity. Bacteria diversity was positively correlated with DOC in the uncontaminated soil (p?Firmicutes only existed in the rhizosphere of contaminated soil, while the very small amount (if any) of some species exists such as Deinococcus-Thermus, indicating that the contaminated environment altered the bacterial composition. Moreover, spatial variation of the bacterial community was found among different soil zones. Real-time PCR confirmed the spatial variation via the gene expression of flagellin (fliC) and chemotaxis gene (cheA). The spatial characteristics of cheA expression were consistent with that of DOC and bacterial diversity. In conclusion, we demonstrated that the spatial variation of the bacterial community in the rhizosphere was present, independent of Cu contamination. DOC and Cu toxicity may affect specific gene expressions such as fliC and cheA, resulting in bacterial spatial variation.  相似文献

Experiments were conducted to assess the impact of citric acid (CA) and rhizosphere bacteria on metal uptake in Phragmites australis cultured in a spiked acid mine drainage (AMD) soil. Rhizosphere iron-oxidizing bacteria (Fe(II)OB) enhanced the formation of Fe plaque on roots, which decreased the uptake of Fe and Mn. CA inhibited the growth of Fe(II)OB, decreased the formation of metal plaque, raised the metal mobility in soil, and increased the accumulation of metals in all tissues of the reeds. The higher the CA dosage, the more metals accumulated into reeds. The total amount of metals in reeds increased from 7.8?±?0.5?×?10?6 mol plant?1 (Mn), 1.4?±?0.1?×?10?3 mol plant?1 (Fe), and 1.0?±?0.1?×?10?4 mol plant?1 (Al) in spiked soil without CA to 22.2?±?0.5?×?10?6 mol plant?1 (Mn), 3.5?±?0.06?×?10?3 mol plant?1 (Fe), and 5.0?±?0.2?×?10?4 mol plant?1 (Al) in soil added with 33.616 g C6H8O7·H2O for per kilogram soil. CA could be effective at enhancing the phytoremediation of metals from AMD-contaminated soil.  相似文献

Bicarbonate plays a crucial role in limiting the growth of submersed aquatic macrophytes in eutrophic lakes, and high ammonia is often toxic to macrophytes. In order to evaluate the combined effect of HCO3 ? and total ammonia (i.e., the total of NH3 and NH4 +) on submersed macrophytes Vallisneria natans, the growth and physiological response of V. natans in the presence of HCO3 ? and ammonia were studied. The results showed that with the increase of ammonia, morphological parameters of V. natans declined. In contrast, increased HCO3 ? concentration stimulated the growth of V. natans, especially when the NH4 +-N/NO3 ?-N ratio was 1:7. High ammonia concentration induced excess free amino acids (FAA) accumulation and soluble carbohydrates (SC) depletion in plant tissues. However, the elevated HCO3 ? promoted the synthesis of SC and rendered the decrease of FAA/SC ratio. The results also suggested that HCO3 ? could partially alleviate the stress of ammonia, as evidenced by the decrease of FAA/SC ratio and the growth enhancement of V. natans when the ammonia concentration was 0.58 mg?L?1. Given the fact that HCO3 ? is probably the dominant available carbon source in most eutrophic lakes, the ability of V. natans to use HCO3 ? for SC synthesis may explain the alleviating effect of HCO3 ? on V. natans under ammonia stress.  相似文献

Severe eutrophication of surface water has been a major problem of increasing environmental concern worldwide. In the present study, economic plant annual ryegrass (Lolium multiflorum) was grown in floating mats as an economic plant-based treatment system to evaluate its potential after ion implantation for removing nutrients in simulated eutrophic water. The specific weight growth rate of L. multiflorum with ion implantation was significantly greater than that of the control, and the peroxidase, nitrate reductase, and acid phosphatase activities of the irradiated L. multiflorum were found to be greater than those plants without ion implantation. Higher total nitrogen (TN) and total phosphorus (TP) removal efficiencies were obtained for the L. multiflorum irradiated with 25 keV 5.2?×?1016 N+ ions/cm2 and 30 keV 4.16?×?1016 N+ ions/cm2, respectively (p?L. multiflorum itself was directly responsible for 39–49 and 47–58 % of the overall N and P removal in the experiment, respectively. The research results suggested that ion implantation could become a promising approach for increasing phytoremediation efficiency of nutrients from eutrophic water by L. multiflorum.  相似文献

In the present study, potential of Eisenia fetida to recycle the different types of fresh water weeds (macrophytes) used as substrate in different reactors (Azolla pinnata reactor, Trapa natans reactor, Ceratophyllum demersum reactor, free-floating macrophytes mixture reactor, and submerged macrophytes mixture reactor) during 2 months experiment is investigated. E. fetida showed significant variation in number and weight among the reactors and during the different fortnights (P <0.05) with maximum in A. pinnata reactor (number 343.3?±?10.23 %; weight 98.62?±?4.23 % ) and minimum in submerged macrophytes mixture reactor (number 105?±?5.77 %; weight 41.07?±?3.97 % ). ANOVA showed significant variation in cocoon production (F4?=?15.67, P <0.05) and mean body weight (F4?=?13.49, P <0.05) among different reactors whereas growth rate (F3?=?23.62, P <0.05) and relative growth rate (F3?=?4.91, P <0.05) exhibited significant variation during different fortnights. Reactors showed significant variation (P <0.05) in pH, Electrical conductivity (EC), Organic carbon (OC), Organic nitrogen (ON), and C/N ratio during different fortnights with increase in pH, EC, N, and K whereas decrease in OC and C/N ratio. Hierarchical cluster analysis grouped five substrates (weeds) into three clusters—poor vermicompost substrates, moderate vermicompost substrate, and excellent vermicompost substrate. Two principal components (PCs) have been identified by factor analysis with a cumulative variance of 90.43 %. PC1 accounts for 47.17 % of the total variance represents “reproduction factor” and PC2 explaining 43.26 % variance representing “growth factor.” Thus, the nature of macrophyte affects the growth and reproduction pattern of E. fetida among the different reactors, further the addition of A. pinnata in other macrophytes reactors can improve their recycling by E. fetida.  相似文献

Lake Erhai is the second largest lake of Southwest China and an important drinking water source. The lake is currently defined as the preliminary stage of eutrophic states, but facing a serious threat with transfer into intensive eutrophication. The present study examined the dynamics of Microcystis blooms and toxic Microcystis in Lake Erhai during 2010, based on quantitative real-time PCR method using 16S rRNA gene specific for Microcystis and microcystin systhesis gene (mcy), and chemical analysis on microcystin (MC) concentrations. Total Microcystis cell abundance at 16 sampling sites were shown as an average of 1.7?×?107 cells l?1 (1.3?×?102–3.8?×?109 cells l?1). Microcystin LR (MC-LR) and microcystin RR (MC-RR) were the main variants. The strong southwesterly winds, anticlockwise circular flows and geographical characteristics of lake and phytoplankton community succession impacted the distribution patterns of Chl a and MC in the lake. The concentration of Chl a and MC and abundances of total Microsytis and MC-producing Microsystis (MCM) were shown to be positively correlated with pH, DO and TP, negatively correlated with SD, NO3-N, TN/Chl a and TN/TP, and not correlated with NH4-N, TN, dissolved total nitrogen (DTN) and water temperatures. When TN/TP decrease, Microcystis tended to dominate and MC concentrations tended to increase, suggesting that the “TN/TP rule” can be partially applied to explain the correlation between the cyanobacterial blooms and nutrients N and P only within a certain nutrient level. It is speculated that N and P nutrients and the associated genes (e.g., mcy) may jointly drive MC concentration and toxigenicity of Microcystis in Lake Erhai.  相似文献

In this study, a SYBR green quantitative real-time PCR was developed to quantify and detect the Legionella spp. in various environmental water samples. The water samples were taken from watershed, water treatment plant, and thermal spring area in Taiwan. Legionella was detected in 13.6 % (24/176), and the detection rate for river water, raw drinking water, and thermal spring water was 10, 21.4, and 16.6 %, respectively. Using real-time PCR, concentration of Legionella spp. in detected samples ranged between 9.75?×?104 and 3.47?×?105?cells/L in river water, 6.92?×?104 and 4.29?×?105?cells/L in raw drinking water, and 5.71?×?104 and 2.12?×?106?cells/L for thermal spring water samples. The identified species included Legionella pneumophila (20.8 %), Legionella jordanis (4.2 %), Legionella nautarum (4.2 %), Legionella sp. (4.2 %), and uncultured Legionella sp. (66.6 %). The presence of L. pneumophila in aquatic environments suggested a potential public health threat that must be further examined.  相似文献

This research investigated the role of the pH buffer capacity of sediment on the dechlorination of atrazine using zero valent iron (ZVI). The buffer capacity of the sediment was quantified by batch experiments and estimated to be 5.0 cmol OH? · pH?1. The sediments were spiked with atrazine at 7.25-36.23 mg kg?1 (6.21 × 10?7–3.09 × 10?6 mol atrazine · g?1 sediment) for the batch experiments. The buffer capacity of the sediment maintained the sediment suspension at neutral pH, thereby enabling continuous dechlorination until the buffer capacity of the sediment was depleted. The pseudo-first order dechlorination constants were estimated to be in the range of 1.19 × 10?2?7.04 × 10?2 d?1 for the atrazine-spiked sediments.  相似文献

In August 2012, eight rainwater samples were collected and analyzed for pH and metal ions, viz., iron, copper, and manganese. The pH was within the range 6.84–7.65. The rate of oxidation of dissolved sulfur dioxide was determined using these rainwater samples as reaction medium. Kinetics was defined by the rate law: ?d[S(IV)]/dt = R o = k o[S(IV)]], where k o is the first-order rate constant and R o is the rate of the reaction. The effect of two volatile organic compounds—ethanol and 2-butanol—was examined and found to inhibit the oxidation as defined by the rate law: k obs = k o/(1 + B [Inh]), where k obs is the first-order rate constant in the presence of the inhibitor, [Inh] is the concentration of the inhibitor, and B is the inhibitor parameter—an empirical constant. In the pH range of collected rainwater samples, the values of first-order rate constants ranged from 3.1?×?10?5 to 1.5?×?10?4 s?1 at 25 °C. The values of inhibition parameter were found to be (5.99?±?3.91?×?104) (ethanol) and (3.95?±?2.36)?×?104 (2-butanol) at 25 °C.  相似文献

Water in the Yangtze Estuary is fresh most of the year because of the large discharge of Yangtze River. The Qingcaosha Reservoir built on the Changxing Island in the Yangtze Estuary is an estuarine reservoir for drinking water. Denitrification rate in the top 10 cm sediment of the intertidal marshes and bare mudflat of Yangtze Estuarine islands was measured by the acetylene inhibition method. Annual denitrification rate in the top 10 cm of sediment was 23.1 μmol m?2 h?1 in marshes (ranged from 7.5 to 42.1 μmol m?2 h?1) and 15.1 μmol m?2 h?1 at the mudflat (ranged from 6.6 to 26.5 μmol m?2 h?1). Annual average denitrification rate is higher at mashes than at mudflat, but without a significant difference (p?=?0.084, paired t test.). Taking into account the vegetation and water area of the reservoir, a total 1.42?×?108 g N could be converted into nitrogen gas (N2) annually by the sediment, which is 97.7 % of the dissolved inorganic nitrogen input through precipitation. Denitrification in reservoir sediment can control the bioavailable nitrogen level of the water body. At the Yangtze estuary, denitrification primarily took place in the top 4 cm of sediment, and there was no significant spatial or temporal variation of denitrification during the year at the marshes and mudflat, which led to no single factor determining the denitrification process but the combined effects of the environmental factors, hydrologic condition, and wetland vegetation.  相似文献

Cadmium usually hampers plant growth, but bacterial inoculation may improve stress tolerance in plants to Cd by involving various mechanisms. The objective was to characterize and identify bacteria that improve plant growth under Cd stress and reduce Cd uptake. Cadmium-tolerant bacteria were isolated from rhizosphere soil, which was irrigated with tannery effluent, and six strains were selected as highly tolerant to Cd, showing minimum inhibitory concentration as 500 mg L?1 or 4.45 mmol L?1. These strains were identified by 16S rRNA gene analysis and functional analysis in regard to plant growth promotion characteristics. To determine their effect on cereal growth under Cd stress, seeds were inoculated with these strains individually and grown in soil contaminated with three Cd levels (0, 40 and 80 mg kg?1). Biomass production, relative water content (RWC), electrolyte leakage (ELL) and tissue Cd concentration were measured. Biomass of both cereals was inhibited strongly when exposed to Cd; however, bacterial inoculation significantly reduced the suppressive effect of Cd on cereal growth and physiology. The bacterial isolates belonged to the genera Klebsiella, Stenotrophomonas, Bacillus and Serratia. Maize was more sensitive than wheat to Cd. Klebsiella sp. strain CIK-502 had the most pronounced effects in promoting maize and wheat growth and lowering Cd uptake under Cd stress.  相似文献

The aim of this study was to characterize the features of a Cd-, Pb-, and Zn-resistant endophytic fungus Lasiodiplodia sp. MXSF31 and to investigate the potential of MXSF31 to remove metals from contaminated water and soils. The endophytic fungus was isolated from the stem of Portulaca oleracea growing in metal-contaminated soils. The maximum biosorption capacities of MXSF31 were 3.0?×?103, 1.1?×?104, and 1.3?×?104 mg kg?1 for Cd, Pb, and Zn, respectively. The biosorption processes of Cd, Pb, and Zn by MXSF31 were well characterized with the pseudo-second-order kinetic model. The biosorption isotherm processes of Pb and Zn by the fungus were fitted better with the Langmuir model, while the biosorption processes of Cd was better fitted with the Freundlich model. The biosorption process of MXSF31 was attributed to the functional groups of hydroxyl, amino, carbonyl, and benzene ring on the cell wall. The active biomass of the strain removed more Cd, Pb, and Zn (4.6?×?104, 5.6?×?105, and 7.0?×?104 mg kg?1, respectively) than the dead biomass. The inoculation of MXSF31 increased the biomass of rape (Brassica napus L.), the translocation factor of Cd, and the extraction amount of Cd by rape in the Cd?+?Pb-contaminated soils. The results indicated that the endophytic fungus strain had the potential to remove heavy metals from water and soils contaminated by multiple heavy metals, and plants accumulating multiple metals might harbor diverse fungi suitable for bioremediation of contaminated media.  相似文献

Abstract

Information regarding the enantioselective endocrine disruption of chiral herbicides is scarce. This study assessed the disrupting effects of eight typical chiral herbicides on corticosteroids (including glucocorticoids and mineralocorticoids). Enantioselectivity of eight chiral herbicides were evaluated for their agonistic/antagonistic effects on glucocorticoid receptor (GR) and mineralocorticoid receptor (MR) with CHOK1 cell line using reporter gene assay. Their influence on the production of corticosteroids were further investigated in H295R cell line using enzyme-linked immunosorbent assay (ELISA). None of the racemates or enantiomers of eight chiral herbicides exhibited GR or MR agonistic activity at non-cytotoxic concentrations. However, rac-propisochlor and S-imazamox antagonized cortisol-induced transactivation of GR by 21.79% and 38.73% at the concentration of 1.0?×?10?7 M and 1.0?×?10?6 M, respectively, and R-napropamide remarkably attenuated aldosterone-induced MR transactivation by 68.78% at 1.0?×?10?6 M. The secretion of cortisol was significantly restrained after treated with 1.0?×?10?6 M rac-propisochlor and rac-/R-napropamide at the concentration of 1.0?×?10?6 M by 26.49%, 30.10% and 35.27%, respectively, while this glucocorticoid was remarkably induced by 1.0?×?10?5 M rac-diclofop-methyl and its two enantiomers at the concentration of 1.0?×?10?5 M by 75.60%, 100.1% and 68.78%, respectively. Exposure to rac-propisochlor (1.0?×?10?6 M), S-diclofop-methyl (1.0?×?10?5 M) or rac-/S-/R- acetochlor (1.0?×?10?6 M) and rac-/S-/R-lactofen (1.0?×?10?6 M) inhibited the secretion of aldosterone by approximately 40%. Our findings suggested that chiral herbicides disrupted corticosteroid homeostasis in an enantioselective way. The aim of the present work was to establish the kinetics for the degradation of doxycycline in the aquatic environment with a view to arriving at a kinetic model that can be used to predict the persistence of antibiotic with confidence. The degradation of doxycycline in both water and sediment phases of aquatic microcosm experiments, as well as in distilled water control experiments, was studied over a period of 90 days. An initial 21% loss due to adsorption by the sediment was observed in the microcosm experiment soon after charging. Biphasic zero-order linear rates of degradation, attributed to microbial degradation of the free and sediment or colloidal particle-adsorbed antibiotic, were observed for both water phase (2.3 × 10?2 and 4.5 × 10?3 μgg?1 day?1) and sediment phase (7.9 × 10?3 and 1.5 × 10?3 μgg?1 day?1) of the microcosm experiment. The covered distilled water control experiment exhibited a monophasic zero-order linear rate (1.9 × 10?3 μgg?1 day?1) attributed to hydrolysis, while the distilled water experiment exposed to natural light exhibited biphasic liner rates attributed to a combination of hydrolysis and photolysis (2.9 × 10?3 μgg?1 day?1) and to microbial degradation (9.8 × 10?3 μgg?1 day?1). A kinetic model that takes into account hydrolysis, photolysis, microbial degradation as well as sorption/desorption by colloidal and sediment particles is presented to account for the observed zero-order kinetics. The implications of the observed kinetics on the persistence of doxycycline in the aquatic environment are discussed.  相似文献

Methane-oxidizing bacteria (methanotrophs) in the soil are a unique group of methylotrophic bacteria that utilize methane (CH4) as their sole source of carbon and energy which limit the flux of methane to the atmosphere from soils and consume atmospheric methane. A field experiment was conducted to determine the effect of nitrogen application rates and the nitrification inhibitor dicyandiamide (DCD) on the abundance of methanotrophs and on methane flux in a grazed pasture soil. Nitrogen (N) was applied at four different rates, with urea applied at 50 and 100 kg N ha?1 and animal urine at 300 and 600 kg N ha?1. DCD was applied at 10 kg ha?1. The results showed that both the DNA and selected mRNA copy numbers of the methanotroph pmoA gene were not affected by the application of urea, urine or DCD. The methanotroph DNA and mRNA pmoA gene copy numbers were low in this soil, below 7.13?×?103 g?1 soil and 3.75?×?103 μg?1 RNA, respectively. Daily CH4 flux varied slightly among different treatments during the experimental period, ranging from ?12.89 g CH4 ha?1 day?1 to ?0.83 g CH4 ha?1 day?1, but no significant treatment effect was found. This study suggests that the application of urea fertilizer, animal urine returns and the use of the nitrification inhibitor DCD do not significantly affect soil methanotroph abundance or daily CH4 fluxes in grazed grassland soils.  相似文献

Understanding the effects of sediment contaminants is pivotal to reducing their impact in aquatic environments. Outdoor mesocosms enable us to decipher the effects of these contaminants in environmentally realistic scenarios, providing a valuable link between laboratory and field experiments. However, because of their scale, mesocosm experiments are often complex to set up and manage. The creation of environmentally realistic conditions, particularly when using artificially contaminated sediment, is one issue. Here, we describe changes in geochemistry over 1.5 years of a sediment spiked with four different concentrations of copper, within a large freshwater mesocosm facility. The spiking procedure included proportional amendments with garden lime to counteract the decreases in pH caused by the copper additions. The majority of copper within the spiked mesocosm sediments partitioned to the particulate phase with low microgram per liter concentrations measured in the pore waters and overlying waters. The minimum partition coefficient following equilibration between pore waters and sediments was 1.5?×?104 L/kg, which is well within the range observed for field-contaminated sediments (1?×?104 to 1?×?106 L/kg). Recommendations are made for the in situ spiking of sediments with metals in large outdoor mesocosms. These include selecting an appropriate sediment type, adjusting the pH, allowing sufficient equilibration time, and regular mixing and monitoring of metal partitioning throughout the experimental period.  相似文献

A novel photocatalytic reactor for wastewater treatment was designed and constructed. The main part of the reactor was an aluminum tube in which 12 stainless steel circular baffles and four quartz tube were placed inside of the reactor like shell and tube heat exchangers. Four UV–C lamps were housed within the space of the quartz tubes. Surface of the baffles was coated with TiO2. A simple method was employed for TiO2 immobilization, while the characterization of the supported photocatalyst was based on the results obtained through performing some common analytical methods such as X-ray diffraction (XRD), scanning electron microscope (SEM), and BET. Phenol was selected as a model pollutant. A solution of a known initial concentration (20, 60, and 100 ppmv) was introduced to the reactor. The reactor also has a recycle flow to make turbulent flow inside of the reactor. The selected recycle flow rate was 7?×?10?5 m3.s?1, while the flow rate of feed was 2.53?×?10?7, 7.56?×?10?7, and 1.26?×?10?6 m3.s?1, respectively. To evaluate performance of the reactor, response surface methodology was employed. A four-factor three-level Box–Behnken design was developed to evaluate the reactor performance for degradation of phenol. Effects of phenol inlet concentration (20–100 ppmv), pH (3–9), liquid flow rate (2.53?×?10?7?1.26?×?10?6 m3.s?1), and TiO2 loading (8.8–17.6 g.m?2) were analyzed with this method. The adjusted R 2 value (0.9936) was in close agreement with that of corresponding R 2 value (0.9961). The maximum predicted degradation of phenol was 75.50 % at the optimum processing conditions (initial phenol concentration of 20 ppmv, pH?~?6.41, and flow rate of 2.53?×?10?7 m3.s?1 and catalyst loading of 17.6 g.m?2). Experimental degradation of phenol determined at the optimum conditions was 73.7 %. XRD patterns and SEM images at the optimum conditions revealed that crystal size is approximately 25 nm and TiO2 nanoparticles with visible agglomerates distribute densely and uniformly over the surface of stainless steel substrate. BET specific surface area of immobilized TiO2 was 47.2 and 45.8 m2 g?1 before and after the experiments, respectively. A number of human health effects have been associated with exposure to metal removal fluids (MRFs). Multiple lines of research suggest that a newly identified organism, Mycobacterium immunogenum (MI), appears to have an etiologic role in hypersensitivity pneumonitis (HP) in case of MRFs exposed workers. However, our knowledge of this organism, other possible causative agents (e.g., Pseudomonads), and the microbial ecology of MRFs in general, is limited. In this study, culture-based methods and small subunit ribosomal RNA gene clone library approach were used to characterize microbial communities in MRF bulk fluid and associated biofilm samples collected from fluid systems in an automobile engine plant. PCR amplification data using universal primers indicate that all samples had bacterial and fungal contaminated. Five among 15 samples formed colonies on the Mycobacteria agar 7H9 suggesting the likely presence of Mycobacteria in these five samples. This observation was confirmed with PCR amplification of 16S rRNA gene fragment using Mycobacteria specific primers. Two additional samples, Biofilm-1 and Biofilm-3, were positive in PCR amplification for Mycobacteria, yet no colonies formed on the 7H9 cultivation agar plates. Real-time PCR was used to quantify the abundance of M. immunogenum in these samples, and the data showed that the copies of M. immunogenum 16S rRNA gene in the samples ranges from 4.33?×?104 copy/ml to 4.61?×?107 copy/ml. Clone library analysis revealed that Paecilomyces sp. and Acremonium sp. and Acremonium-like were dominant fungi in MRF samples. Various bacterial species from the major phylum of proteobacteria were found and Pseudomonas is the dominant bacterial genus in these samples. Mycobacteria (more specifically MI) were found in all biofilm samples, including biofilms collected from inside the MRF systems and from adjacent environmental surfaces, suggesting that biofilms may play an important role in microbial ecology in MRFs. Biofilms may provide a shield or sheltered microenvironment for the growth and/or colonization of Mycobacteria in MRFs.  相似文献

Purpose

Combinatorial bio/chemical approach was applied to investigate dioxin-like contamination of soil and sediment at the petrochemical and organochlorine plant in Pancevo, Serbia, after the destruction of manufacturing facilities that occurred in the spring of 1999 and subsequent remediation actions.

Materials and methods

Soil samples were analyzed for indicator polychlorinated biphenyls (PCBs) by gas chromatography/electron capture detection (GC/ECD). Prioritized soil sample and sediment samples from the waste water channel were analyzed for polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) by high-resolution gas chromatography/high-resolution mass spectrometry (HRGC/HRMS). Microethoxyresorufin o-deethylase (Micro-EROD) and H4IIE?Cluciferase bioassays were used for monitoring of dioxin-like compounds (DLC) and for better characterization of dioxin-like activity of soil samples.

Results

Bioanalytical results indicated high dioxin-like activity in one localized soil sample, while the chemical analysis confirmed the presence of large quantities of DLC: 3.0?×?105 ng/g d.w. of seven-key PCBs, 8.2 ng/g d.w. of PCDD/Fs, and 3.0?×?105 ng/g d.w. of planar and mono-ortho PCBs. In the sediment, contaminant concentrations were in the range 2?C8 ng/g d.w. of PCDD/Fs and 9?C20 ng/g d.w. of PCBs.

Conclusions

This study demonstrates the utility of combined application of bioassays and instrumental analysis, especially for developing and transition country which do not have capacity of the expensive instrumental analysis. The results indicate the high contamination of soil in the area of petrochemical plant, and PCDD/Fs contamination of the sediment from the waste water channel originating from the ethylene dichloride production.  相似文献
