Electrophoretic identification and genetic analysis of bivalve larvae   总被引:2,自引:0,他引:2  
Taxonomic identification and genetic analysis of larval marine invertebrates have been vexing problems. We describe a polyacrylamide mini-gel electrophoresis technique for resolving proteins from individual larval bivalves (shell length 250 to 350 m) and apply this technique to three species of laboratory-cultured larval oysters [Ostrea edulis L., 1758, Crassostrea gigas (Thunberg, 1793) and c. virginica (Gmelin, 1791)] reared during summer 1989. Electrophoretic patterns of proteins clearly discriminate among the three species and allow genetic analysis of a polymorphic allozyme locus (Pgi) in field-collected larvae and juveniles of C. virginica. This technique provides an economical tool for largescale taxonomic, ecologic, and genetic studies of meroplanktonic stages of various species.  相似文献

The spawning population of European eel (Anguilla anguilla L.) has been considered panmictic on the basis of genetic markers and morphometric studies. This hypothesis was tested by screening glass eel from five locations (Ireland, Italy, Morocco, Sweden and U.K.), belonging to two cohorts at the cytochrome b (cyt b) locus (392 bp) of the mitochondrion and at five nuclear microsatellite loci. Seventeen cyt b haplotypes were detected, of which ten were singletons; the most common haplotype occurred in 47% of all fish. Haplotype number increased significantly with latitude. Phylogeographical structure based on the cytoplasmic marker was weak (FST=0.014) and non-significant. Close similarity was revealed between British and Irish glass eel populations, and weak differentiation among the British/Irish, Atlantic Moroccan, Italian and Swedish Baltic populations, respectively. No hierarchical genetic structure was obvious. Levels of genetic variation detected with five microsatellites were much higher levels than found with allozymes in previous studies (mean number of alleles per locus=11.1; mean expected heterozygosity=0.68). Overall among-population microsatellite variance was low but significant (FST=0.004), and caused by the linked microsatellite loci Aan03 and Aan04. The Hardy-Weinberg-Castle equilibrium and the absence of gametic disequilibria at these loci in the Moroccan population might point to its genetic isolation, although the impact of just two out of five loci is puzzling. Given the weak differentiation typical for marine species and the limitations of our data, the results should be interpreted with caution. However, combined with recent evidence from a related study, the paradigm that the European eel constitutes a panmictic population becomes difficult to maintain.  相似文献

In this study, a proteomic approach was applied for the generation of reference maps and subsequently to detect, quantify and compare the global protein expression between two related species of marine mussels, Mytilus edulis and Mytilus galloprovincialis, growing in their own geographical habitats. A comparative study of the protein profiles generated from analytical two-dimensional electrophoresis gels was performed, and changes in protein expression were analysed quantitatively by computer analysis. An average of 1,278 spots per gel was detected in 16 individuals (8 M. edulis and 8 M. galloprovincialis); however, not all spots were included in the study. Expression of 420 spots was compared, and significant differences in the intensity levels were detected in 37 protein spots (8.8%). Fifteen proteins showed higher expression in M. edulis, and 22 proteins, in M. galloprovincialis. The technique of peptide mass fingerprinting using MALDI-TOF (matrix-assisted laser desorption ionisation/time-of-flight) and/or nanoelectrospray double subfragmentation mass spectrometry enabled the unambiguous identification of 15 of these 37 differentially expressed proteins. Most of the identified proteins can be grouped basically into four broad functional classes: cytoskeletal and myofibrillar proteins, proteins associated with stress response, proteins associated with the storage or production of energy, and proteins related to rearrangement in the synthesis of native structures. These results expand our understanding of the molecular differentiation of the two mussel taxa and serve as a useful base for future ecological, physiological and genetic studies.  相似文献

Four species of adult Carapini (Carapidae) occur on Polynesian coral reefs: Encheliophis gracilis, Carapus boraborensis, C. homei and C. mourlani. Samples collected in Rangiroa and Moorea allowed us to obtain different tenuis (larvae) during their settlement phases or directly inside their hosts. These were separated into four lots on the basis of a combination of pigmentation, meristic, morphological, dental and otolith (sagittae) features. Comparison of these characters with those of the adults allows, for the first time, taxonomic identification of these tenuis-stage larvae.  相似文献

We undertook a comprehensive study of Latrunculia in New Zealand to determine the relationship between taxonomic, environmental, and chemical variation within the genus. Sponges were collected from five locations around New Zealand: Three Kings Islands, Tutukaka, Wellington, Kaikoura, and Doubtful Sound. Allozyme electrophoresis at nine polymorphic loci indicated that sponges from each geographic location were genetically distinct, and that they displayed genetic differences of the magnitude usually associated with reproductively isolated species (Nei's D between locations =0.375-2.476). Additionally, the comparisons revealed that the green and brown colour morphs of Latrunculia that are sympatric at Three Kings Islands and Kaikoura are distinct from each other, and that there are two genetic groups within the green sponges in Doubtful Sound. On the basis of genetic data we conclude that there are at least eight species of Latrunculia in New Zealand waters, not one to four as had been previously thought. Morphological comparisons of the eight genetic species based on skeletal characters (i.e. skeletal organisation of the choanosome, spicule composition, size, and geometry) indicated that the eight Latrunculia species fell into only two morphological groups that could be easily diagnosed on the basis of discorhabd type. Within these two primary morphological groups, skeletal characteristics among the eight species largely overlap and are not diagnostic. These findings emphasise the limitations of traditional taxonomic methods based solely on skeletal characters for distinguishing species of Latrunculia. However, multivariate analysis (MANOVA and CDA) based on six measured skeletal variables did reveal significant morphological variation among the species (Pillai's Trace=3.28, F=6.90, P<0.0001), supporting the division of the genus into eight species. Comparisons of chemical extracts from Latrunculia also showed that the amounts of five different bioactive compounds (discorhabdins A, B, C, D, and J) varied predictably among the eight species. This finding suggests that discorhabdin variation within Latrunculia, previously thought to be associated with intra-specific environmental variability, is more likely to reflect differences among species rather than phenotypic plasticity. Our results also highlight the importance of thorough taxonomic studies associated with marine natural products research to understand fully the variation in bioactive properties among individuals. The potential processes underlying the unusually high speciation rates in New Zealand Latrunculia that are indicated in our study are discussed.  相似文献

In marine invertebrates multiple modes of development, or poecilogony, may occur in a single species. However, after close examination, many of such putative cases turned out to be sibling species. A case in point may be the cosmopolitan orbiniid polychaete Scoloplos armiger, which inhabits marine shallow sediments. In addition to the well-known direct, holobenthic development from egg cocoons, pelagic larvae have also been described. Our culture experiments revealed a spatially segregated source of the two developmental modes. All females of an intertidal population produced egg cocoons and no pelagic larvae. All but 2 out of 15 females of an adjacent subtidal population produced pelagic larvae and no egg cocoons. Based on these results we performed a molecular genetic analysis (RAPD-PCR) on three intertidal and four subtidal populations in the North Sea. Selected samples from all sites were analysed also by the AFLP method. We found significantly higher genetic diversity within subtidal than within intertidal populations. This is consistent with a wider dispersal by pelagic larvae and a smaller effective population size when development is holobenthic. Total genetic divergence is not related to distance but to the intertidal/subtidal division. We suggest that S. armiger actually represents two sibling species.  相似文献

In aquaculture centers of the northern region of Japan, "Nami-ita" (waved polycarbonate plates), on which the green alga Ulvella lens Crouan frat. (Chaetophoraceae: Chaetophorales) was cultured, are used to promote larval settlement and metamorphosis of the sea urchin species Strongylocentrotus intermedius (A. Agassiz) and S. nudus (A. Agassiz). We investigated chemical inducer(s) for larval settlement and metamorphosis of these sea urchins with extracts of U. lens. Bioassay-guided separation of the methanol extract using a combination of column and thin-layer chromatography led to the isolation of several active compounds, the chemical structures of which were determined by spectral and chemical methods. These active compounds were identified as glycoglycerolipids, all comprising several molecular species: sulfoquinovosyl monoacylglycerols (SQMGs), sulfoquinovosyl diacylglycerols (SQDGs), monogalactosyl monoacylglycerols (MGMGs), monogalactosyl diacylglycerols (MGDGs), digalactosyl monoacylglycerols (DGMGs) and digalactosyl diacylglycerols (DGDGs). Among these glycolipids, SQMGs, MGMGs, MGDGs and DGMGs induced larval metamorphosis of the sea urchin S. intermedius. SQMGs and MGDGs induced larval metamorphosis at a concentration of 5 µg ml-1, whereas SQDGs and DGDGs only induced larval settlement. These glycoglycerolipids are new congeners of chemical inducers to settlement and metamorphosis of planktonic larvae of sea urchins. The findings would provide a better understanding of larval settlement and metamorphosis in sea urchins.  相似文献

Variation at five microsatellite loci (Sal1, Sal2, Sal3, Sal4 and Sal5) was examined in approximately 1300 Pacific ocean perch (Sebastes alutus) sampled from 14 coastal sites in British Columbia, Canada. Mean observed heterozygosities by locus ranged from 71% to 88%, and by sample ranged from 75% to 84%. Theta values ranged from 0 to 0.04 over the five loci, and averaged 0.015. Among Pacific ocean perch samples, Š ranged from 0.001 to 0.056. Canonical discriminate analysis of multilocus genotypes and neighbour-joining analysis of pairwise genetic distances between samples both indicated the presence of three populations, one off the west coast of Vancouver Island (the Vancouver Island population) and two co-existing populations in Queen Charlotte Sound, Dixon Entrance and along the west coast of the Queen Charlotte Islands (the eastern and western QCI populations). Pacific ocean perch of the eastern and western QCI populations were caught in close proximity to each other, but individual samples showed little evidence of admixture. Fall and spring samples collected within geographic areas were genetically similar, indicating seasonally stable population structure. Restricted gene flow between the Vancouver Island and the two more northerly populations may result from limited adult dispersal and larval retention within the California Current and Alaska Gyre, respectively, but the presence of two populations within Queen Charlotte Sound cannot be explained entirely by larval retention hypotheses. The presence of two Pacific ocean perch populations in central British Columbia has implications for fisheries management.  相似文献

The ahermatypic scleractinian Balanophyllia elegans has served as a model of limited larval dispersal in an aclonal species. However, other species from the same family (Dendrophylliidae) produce larvae asexually, and closely connected polyps of B. elegans, potentially the result of asexual reproduction, are commonly observed in the field. Here, we use a combination of laboratory experiments, controlled crosses, and genetic surveys of field-collected individuals to demonstrate (1) marker allozymes are inherited in accordance with Mendel's laws, (2) polyps that are connected in the field are not genetically identical, and (3) laboratory manipulation of a single polyp can produce a second polyp on the aboral side of the original, but not adjacent to the original. In combination, these results suggest that the larvae of B. elegans result from mictic sexual reproduction, and that connected polyps result not from asexual budding but rather from the fusion of genetically distinct individuals.  相似文献

Accurate species identification of decapod crustacean larvae is required to understand their population distributions, life cycle dynamics and interactions with their habitats. Analysis of plankton samples using morphological taxonomic methods and microscopy is time-consuming, requires highly skilled and trained operatives and may often be inaccurate. As complementary tools to classical identification methods, recent work has focused on the development of molecular approaches and shows their feasibility for species-specific identification. This study has developed real-time PCR assays utilising species-specific Taqman® probes designed in the cytochrome oxidase I (COI) gene of Liocarcinus depurator, Necora puber, Carcinus maenas and Cancer pagurus. Our study then employed the probe and primers designed for L. depurator to obtain accurate identification and relative abundance estimates of L. depurator larvae in plankton samples collected between March 2005 and October 2006. Ranges of larval abundances were derived from a standard curve created from plankton samples spiked with a known number of larvae reared in the laboratory. Inhibition of the PCR reaction was shown to be an important factor and our results suggested that 0.1 ng of DNA as template provided accurate identification and avoided inhibition. Real-time PCR was shown to provide accurate species identification on unsorted plankton samples and could be suitable for the estimation of larval abundances in the plankton, although more work must be done to improve the accuracy of those estimations.  相似文献

The distribution of Mytilus edulis Linnaeus, M. galloprovincialis Lamarck, and their hybrids was examined in mussel populations in southwest England in 1996 and 1998. This is a region where both parental taxa and populations containing large numbers of hybrids co-occur yet a fine-scale mapping of the hybrid populations has not been conducted. In this study the geographic distribution of hybrid populations was determined in southwest England over 360 km of coast from Tintagel Castle in north Cornwall to Beer in south Devon. Sample localities were spaced from 5 to 20 km apart. Genotypes for individual mussels were determined using PCR to amplify a size polymorphism at the Glu-5' locus that is completely differentiated between M. edulis and M. galloprovincialis. Hybrid populations, characterized by high frequencies of individuals with heterozygous genotypes and a pattern of decreasing frequency of M. edulis alleles with increasing shell length, were continuously distributed along 180 km of open coast in southwest England. This "hybrid patch" was bordered at one end by geographically extensive populations of pure M. edulis and at the other end by nearly pure populations of M. galloprovincialis. Strong natural selection in hybrid populations results in a decline in the frequency of M. edulis alleles with increasing size. Wave-exposure has previously been implicated as the agent producing this pattern of selection, but in the present study the relationship between allele frequency and body size was not correlated with variation in wave shock intensity among localities within the hybrid zone. The transition between hybrid populations and those containing pure populations of M. edulis or M. galloprovincialis is abrupt which suggests that coastal circulation patterns may provide strong barriers to larval dispersal which accounts for the position and maintenance of the hybrid zone.  相似文献

Information concerning the way juvenile crabs choose their diet from a variety of prey types can be useful for a better understanding of community dynamics, as well as for the adequate management of natural resources. Prey size and species selection by juvenile Carcinus maenas (15-35 mm carapace width, CW) and Cancer pagurus (20-40 mm CW) feeding on four bivalves of contrasting shell morphology were investigated. When offered a wide size range of Mytilus edulis, Ostrea edulis, Crassostrea gigas, and Cerastoderma edule presented individually, crabs generally showed evidence of size-selective predation. Cancer pagurus selected larger mussels relative to the size of their chelae (relative prey size, RPS) than did Carcinus maenas of similar and even larger carapace width. However, the RPS of selected O. edulis and Cerastoderma edule were similar for all crabs, suggesting that certain prey features constitute effective barriers even to the powerful chelae of Cancer pagurus. When offered a wide size range of mussels and oysters simultaneously, all crabs consistently selected mussels. When offered O. edulis and Crassostrea gigas, crabs consumed both these oyster species in similar numbers. Carcinus maenas consumed similar numbers of mussels and cockles; Cancer pagurus, however, showed no preference for either prey in the smaller size classes but selected more mussels than cockles as prey increased in size. Although previous studies report that adult Carcinus maenas select prey species according to their profitability (amount of food ingested per unit of handling time, milligrams per second), consumption rates of the size classes of prey selected by juvenile shore crabs did not always parallel prey value. Although variations in crab strength can account for many of the differences between the foraging strategy of juvenile and adult C. maenas, our results suggest that juvenile crabs are less species selective than adults as a result of the restrictions imposed on small individuals that have limited access to larger prey.  相似文献

Patterns of genetic diversity of the marine deep-water redfish, Sebastes mentella Travin, from 21 sampling locations (n=46 per sample, from 1995 to 1998) were examined throughout its range in the North Atlantic. Eight microsatellite loci were used in order to: (1) define the genetic structure and consider the possible influence of dispersal, geographic distance, oceanic currents and historical factors; and (2) relate the structure to present management units and practices. Three divergent population units (mean Š=0.012) associated with distinct geographic areas were defined: Eastern (Norway and Barents Sea), Panoceanic and Western (Gulf of St. Lawrence and offshore Newfoundland). The most important observation was the lack of genetic differences (mean Š=-0.0004) and lack of genetic isolation by geographic distance (r=0.318, P=0.112) among samples within the Panoceanic zone, from the Faroe Islands to the Grand Banks (6,000 km). A combination of vicariance, historical introgression with a closely related species, S. fasciatus, and contemporary patterns of oceanic circulation are likely to have shaped the observed population structure. These results only partially support current management units, and call for more integrated practices for regulating the exploitation of S. mentella throughout its extensive range.  相似文献

The development of the escape response of walleye pollock (Theragra chalcogramma) larvae from attacks by macrozooplanktonic and small-fish predators was quantified in laboratory experiments. Behavior was recorded using video cameras with silhouette illumination from infrared-emitting diodes and by visual observation. Laboratory-reared larvae of 1, 3, 8, 10, 12, 18, 22, 27, 42 days post-hatching, ranging in size from 4 mm to 10 mm total length, were used in the experiments. Even the youngest larvae were observed to exhibit a fast startle response. The percentage of successful larval escapes from the different predators increased as the larvae developed. Euphausiids (Thysanoessa raschii) and amphipods (Calliopiella pratti) often touched larvae but the larvae were usually able to escape and no successful captures of larvae over 22 days old were observed. Although successful escape from initial attacks by three-spine sticklebacks (Gasterosteus aculeatus) increased ontogenetically, sticklebacks were able to consume most larvae, even of the oldest age group, by repeated attacks. Day-old larvae had the lowest percent of escapes after encounters with jellyfish (Sarsia sp.), but the percentage of escapes increased dramatically for 3-day-old larvae. Escape speeds after an attack also increased with age, and tended to be higher after stickleback attacks and lower after jellyfish attacks. This study revealed that the escape response of larval pollock to attack by predators improves rapidly with development during the early larval stage.  相似文献

The ribbed limpet, Lottia digitalis, is found high in rocky intertidal habitat throughout its geographic range. In order to identify likely natural settlement locations for larvae of this species, laboratory-reared larvae were settled onto substrata collected from within and near an adult L. digitalis habitat. Of larvae exposed to rock chiseled from within high-intertidal adult habitat, 31.0% and 23.3% underwent metamorphosis during two separate experiments. Similarly, an unidentified filamentous green alga that was isolated from this rock induced metamorphosis in 26.6% and 8.7% of larvae during additional experiments. In contrast, larvae did not metamorphose upon bare rocks or upon rocks encrusted with a crustose corraline alga (CCA) that were collected from lower intertidal zones, nor did they metamorphose upon the macroalgae Ulva sp., Enteromorpha contorta, Alaria marginata, or Polysiphonia sp. The presence of mucus from adult conspecifics during these experiments did not enhance metamorphosis onto rock taken from adult habitat, but it did induce metamorphosis in an average of 13.5% and 7.0% of larvae introduced to the mid-intertidal bare rock and CCA substrata, respectively. Finally, 38.0% and 34.4% of larvae from two experiments underwent metamorphosis when exposed to the high-intertidal barnacle Pollicipes polymerus. These results suggest that larval settlement, rather than differential post-settlement mortality and/or migration, drives recruitment of L. digitalis in high-intertidal habitats.  相似文献

The genetic population structure of red grouper, Epinephelus morio (Valenciennes), and scamp, Mycteroperca phenax Jordan and Swain, from the southeastern U.S. Atlantic coast and the Gulf of Mexico was examined using nuclear microsatellite DNA markers in order to test the null hypothesis of panmixia throughout this range. Physical and biological data indicate that relatively isolated populations of these fish exist. Genetic variation was assessed at four microsatellite loci in red grouper and six loci in scamp. The fish were collected on different dates between 1991 and 2001. The microsatellite loci were highly polymorphic, with an average expected heterozygosity of 0.75 in red grouper and 0.68 in scamp. Heterozygote deficiencies (significant deviations from Hardy–Weinberg equilibrium, HWE) were found at two of four loci in all red grouper samples except the eastern Gulf of Mexico, and for all red grouper combined. In contrast, all loci conformed to HWE in the separate scamp samples. Minimal genetic differences distinguished southeastern U.S. Atlantic or Mexican red grouper from other localities, and no indication of genetic differentiation was observed in scamp. This large-scale genetic homogeneity may be attributed to ongoing gene flow and/or historical contact between present-day populations. For management purposes, genetic homogeneity does not necessarily imply a single stock. Because larval dispersal may be sufficient to homogenize gene frequencies but not to replenish depleted stocks, other data must be considered in the management of these species.Electronic Supplementary Material Supplementary material is available in the online version of this article at .Communicated by J.P. The principles of energetics were used to examine the energetic requirements of leptocephali. Respiration and excretion rates and daily growth rates combined with proximate composition were used to examine the allocation of energy into each of the three main components of energetics: metabolism, excretion and growth. The daily energetic requirements for leptocephali, referred to as type 2 larvae based upon their unique developmental strategy, were compared to the requirements of non-leptocephalus larvae, known as type 1. Leptocephalus daily energetic requirements were also compared to the energy available from the leptocephalus' proposed food sources. The four species of eel larvae selected were all from the order Anguilliformes: Paraconger caudilimbatus (Poey), Ariosoma balearicum (Delaroche), Gymnothorax saxicola Jordan and Davis, and Ophichthus gomesii (Castelnau). The allocation of energy to each of the components of energetics as well as the total energetic requirements for the leptocephali proved to be very different from those of type 1 larvae. Metabolism received the majority, 60-92%, of the energy required per day. Growth and excretion were allocated 4-39% and <1-21%, respectively, of the total energy needed per day. Leptocephali required <50% of the energy needed by type 1 larvae of equal dry mass. The unique growth strategy used by leptocephali allows them to increase rapidly in size while allocating the majority of their energy, not to growth as in most larval fish, but to metabolism.  相似文献

Crucibulum quiriquinae (Lesson, 1830) is the only species of Crucibulum currently recognized in northern Chile. Recent analysis of three Crucibulum populations obtained in northern Chile demonstrates the existence of morphological, genetic, and reproductive differences among populations. Two populations present in Bahía Tongoy (30°15'S), one inhabiting the shells of the snail Turritella cingulata and the other shells of the pectinid Argopecten purpuratus, showed morphological differences. However, both had planktonic larval development and show low genetic divergence (D=0.002). A third population from Bahía La Herradura (29°58'S), which also inhabits the shells of T. cingulata, did not show morphological differences compared with its counterpart from Bahía Tongoy. However the Bahía La Herradura population had intracapsular development and metamorphosis, and a larger genetic distance (D=0.06) from both Tongoy populations. The results of the reproductive and genetic analyses strongly suggested that the two Tongoy populations, although showing morphological differences, are biologically the same species, Crucibulum quiriquinae, whereas the La Herradura population is a new species.  相似文献

In a semiterrestrial and estuarine tropical crab, Armases angustipes Dana (Grapsoidea: Sesarmidae), changes in biomass (measured as dry mass, W; carbon, C; nitrogen, N; and hydrogen, H; per individual) and relative elemental composition (C, N, H, in percent of W; C:N mass ratio) were studied during development from an early egg stage through hatching, the complete larval phase, metamorphosis and the first juvenile crab stage (CI). In the megalopa and CI, growth was measured also within the moulting cycle, and biomass and elemental composition were determined in cast exuviae. From an early egg stage to the freshly hatched larva, A. angustipes lost about 20% of W, 29% of C, 5% of N and 32% of H. Proportionally higher losses in C than in N were reflected also in a significantly decreasing C:N mass ratio (from 5.02 to 3.74). These results indicate that lipids mobilised from yolk reserves represented the principal metabolic substrate for embryonic energy production, while proteins were catabolised at a much lower rate. The present data of growth and exuviation are compared with previously published data from a congener, A. miersii Rathbun, which has an abbreviated and facultatively lecithotrophic mode of larval development (with three instead of four zoeal stages; stages I and II in principle independent of food). When growth is measured as an increase in the final (premoult) biomass of successive developmental stages, both species show an exponential pattern. Within the moulting cycles of the megalopa and the first juvenile, both species show parabola-shaped growth curves, with a rapid biomass increase in postmoult and intermoult stages, and losses in the premoult phase. Thus, the two Armases species show, in general, similar patterns of larval and early juvenile growth. However, the initial size of eggs and larvae is about four times larger in A. miersii, and its biomass remains higher throughout the period of larval and early juvenile development. A. angustipes is able to partially make up for this difference, as it has an additional zoeal stage, and its megalopa and CI stages show higher relative biomass increments (in percent of initial values). Due to this compensatory growth pattern, A. angustipes reaches in its CI stage about half the biomass of a juvenile A. miersii. When exuvial losses of megalopae and juveniles are compared between these two species, A. miersii shows higher biomass losses per individual (corresponding with its larger size), but lower relative losses (C, N, H, in percent of late premoult body mass or in percent of previously achieved growth increments). Differences in larval and early juvenile growth and in the exuvial losses of megalopae and juveniles of these two congeners are discussed in relation to their differential ecology, life history and reproductive strategy.  相似文献
