Four of the most abundant gorgonian species from the southwestern Cape waters, Eunicella papillosa (Esper, 1797), E. alba (Esper, 1797), E. tricoronata Velimirov, 1971 and Lophogorgia flamea (Ellis and Solander, 1786) were analysed for Ca and Mg by atomic absorption spectroscopy (AAS) and ethylenediaminetetraacetate (EDTA) titration. The total mineral content in the peripheral tissues, excluding the axial skeleton, expressed as the sum of CaCO3 and MgCO3 of dry matter was between 65.5 and 83.5%. The mineral content varied in different growth regions and all specimens showed a higher degree of mineralization at the base than at the branch tips. The MgCO3 concentration varied with genus and species and was between 9 and 11 mol %. The variation of the MgCO3 concentration within different growth regions of the same species was small and generally did not exceed 0.8 mol %. From the branch to the stem, CaCO3 and total mineral content was found to increase. The CaCO3:MgCO3 rations in different growth regions of all species indicated that the composition of the mesoskeleton with regard to the relative concentration of CaCO3 and MgCO3 is constant throughout the animal. Mineralogically, the mesoskeleton consists of high magnesian calcite as identified by X-ray diffraction. MgCO3 concentrations determined by the peak shift method and by AAS were in fair agreement. The MgCO3 data in gorgonian samples from the cold Atlantic Ocean and the warmer Indian Ocean show a linear relationship between water temperature and MgCO3 concentration already demonstrated by Chave (1954). Extracts of 39 species of Caribbean gorgonians were tested for antimicrobial activity against 15 strains of marine bacteria. The bacteria consisted of three opportunistic pathogens, Vibrio parahaemolyticus, Leucothrix mucor, and Aerococcus viridans, and 12 strains isolated from either healthy or decayed gorgonians. Overall, only 15% (79 out of 544) of the tests resulted in antibacterial activity with 33% (13 out of 39) of the gorgonians inhibiting only one bacterial strain and 23% (9 out of 39) showing no activity. The extracts of four Pseudopterogorgia species showed relatively high levels of activity, inhibiting 43 to 86% of the bacterial strains. The potency of the active Pseudopterogorgia species was variable, however, and three additional Pseudopterogorgia species were inactive against all bacterial strains. With the exception of one sensitive strain, Vibrio species were resistant to gorgonian metabolites. Our results indicate that organic extracts of most Caribbean gorgonians do not possess potent, broad-spectrum antibacterial activity inhibitory to the growth of opportunistic marine pathogens and bacteria associated with healthy and decayed gorgonian surfaces. These findings suggest that the inhibition of bacterial growth is not the primary ecological function of gorgonian secondary metabolites and that bacteria may not be important selective agents in the evolution of gorgonian secondary chemistry.  相似文献

Chemical resistance of gorgonian corals against fungal infections   总被引:4,自引:0,他引:4  
The frequency and impact of diseases affecting corals throughout the Caribbean have been increasing but little is known about the factors promoting the emergence and outbreak of disease. A disease caused by a fungal pathogen [Aspergillus sydowii (Thom et Church)] which affects Caribbean sea fan corals provided an opportunity to examine the efficacy of coral crude extracts in disease resistance. Minimum inhibitory concentration (MIC) assays showed that of the 20 common gorgonian species in the Florida Keys, extracts from 15 species had MICs < 15 mg ml−1 against A. sydowii pathogenic to sea fans. Extracts from several species in two gorgonian genera (Pseudoplexaura and Pseudopterogorgia) were among the most active, with MICs < 10 mg ml−1. Gorgonia ventalina L., one of two sea fan species known to be hosts to A. sydowii in the field, had an MIC < 10 mg ml−1, suggesting that complete disease resistance requires more active extracts. For the antifungal compounds to be effective in situ, they must also occur in sufficiently high concentrations in living coral tissue. For example, Pseudopterogorgia americana (Gmelin) had comparatively potent extracts but did not have sufficient concentrations in the tissue to be effective. Conversely, Plexaura homomalla Esper extracts were less potent but occurred in high enough concentrations in the tissue to be effective against A. sydowii. When potency and extract concentration are considered together (i.e. potency × concentration), several other gorgonian corals emerge as likely hosts to A. sydowii. Crude extracts from the most active gorgonian species were also effective against two geographic variants of A. sydowii pathogenic to sea fans, a non-pathogenic terrestrial strain of A. sydowii, and three strains of A. flavus Link known to be human, plant, and insect pathogens (MIC range, 7.5 to > 15 mg ml−1). Although the potency in these assays did not attain a clinically significant level, the potency is comparable to a known antifungal agent, hygromycin B, which had an MIC ≤7.5 mg ml−1 in our assays, highlighting the potential of these gorgonian corals for bioprospecting. The aim of this survey was to test a standardised method to follow the demographic evolution of a dense aggregative ‘forest’ population of the temperate gorgonian Eunicella verrucosa (Octocorallia, Gorgoniidae) using in situ photographic recordings. Distribution, density, growth and demographic evolution of the colonies was compared along two parallel transects. Computer treatment allowed the estimation of the total branch fan surface area, and the individual growth of tagged colonies was determined by measuring the increase in this surface area, using consecutive photographs taken at two-year intervals. To integrate the potential bias of branch overlapping, we proposed a correction factor between the in situ photographic surface area and the surface area of the gorgonian calculated from ex situ photographic surface area with all branches deployed. The surface-frequency distribution of colonies was converted to estimated-age-frequency distribution using an estimated growth curve based on the net growth rate. The technique used revealed significant differences in population structure and the dynamics of gorgonian colonies, as the two transects appeared to be influenced by different environmental conditions. The recruitment also seemed to vary according to year and transect, resulting in different densities. Our work showed clear results in characterising the variations of gorgonian demographic evolution at a small spatial scale; thus, it is assumed that this method could be a sustainable tool for coastal environmental managers.  相似文献

In situ and laboratory obervations of the biology and behaviour of the deep-sea, euryaline brittle star Asteroschema tenue Lyman, from Jamaica, are described. The brittle star is found clinging to the gorgonian Ellisella barbadensis at depths of 168 to 230 m off Jamaica, and is here restricted to this gorgonian. The brittle star, which is usually cryptically coloured, is active principally nocturnally, when it extends arm loops and employs the tube feet for microphagous filter-feeding. Locomotory activity is limited to extension and retraction along the gorgonian, achieved by means of unusual arm-loop patterns. In this paper we describe a cryopreservation protocol followed by the culture of Symbiodinium sp. isolated from the Caribbean gorgonian Pseudopterogorgia elisabethae as a potential renewable source of the dinoflagellate symbiont. Four different freezing protocols were designed: a controlled cooling device designed to cool at 1°C/min, a three-step protocol (−20°C for 2 h, −70°C for 2 h, liquid nitrogen-LN2), a two-step protocol (−70°C for 2 h, LN2), and a one-step protocol (LN2). All cells were stored in LN2 after cryopreservation. The cryoprotective agents (CPA) used were ethanol (EtOH) and methanol (MeOH) at 10 and 20%, and seawater (FSW) was used as a control. Viability measurements using cell counts showed that all cryopreservation protocols were relatively successful, and no trends were observed regarding freezing protocol or CPA used. After 19 weeks in culture the viability of samples which had high biomass was determined by the fluorescent assay CellTiter Blue™. The most viable cultures were those cryopreserved by a two-step protocol using 20% MeOH or 20% EtOH as a CPA. A genetic examination of the DNA of these samples using Symbiodinium-specific PCR primers confirmed that the composition of the culture had not changed. For the first time, we report that Symbiodinium sp. isolated from a gorgonian can be cryopreserved and subsequently cultured successfully. Lory Z. Santiago-Vázquez and Nealie C. The two forms of Eunicella singularis sclerites, spindles and clubs, were examined in two sites in East Mediterranean Sea, Greece. The comparison of the sclerites size was based on the hydrodynamic profile of the sites (the site of Arethoussa was exposed and the Phidonissi site was sheltered), on the sites bathymetry (AR 5–8 m, AR 9–13 m, PH 5–10 m, PH 11–13 m) and on the position occupied by the sclerites on the gorgonian colonies (top, middle and base). With the usage of image system analysis the following microfeatures were measured or calculated: projection, length, width, elongation factor, compactness factor, length × width and length/width. The present study confirms the hypothesis that spindles are involved in the recovering of the gorgonian colonies and clubs contribute to the flexion capacity of the colonies. The spindles of the shallow E. singularis colonies (AR 5–8 m) were significantly bigger (length, width) compared with the others, in order to assure the best recovery of the colonies after their flexion, induced by higher water current velocity in shallow water. The clubs occupy the intern part of the mesoglea and their size (length, width) increased in colonies from the shallow exposed site in order to facilitate the gorgonian flexion. The present study demonstrates the gorgonian adaptation (in a microscopic scale) to the environmental pressure of hydrodynamic conditions.  相似文献

Paramuricea clavata (Risso, 1826) and Eunicella singularis (Esper, 1794) are the most representative gorgonian species in hard bottoms sublittoral communities in the Western Mediterranean Sea. Reproductive cycles of two populations of both species were studied in two distinct locations approximately 600 km apart (Medes Islands and Cape of Palos), in order to assess interpopulation variability on a relevant geographic scale. Seasonal variation of lipid concentration levels in the gorgonian tissue was used as a tool to quantify energy storage by each studied population in order to explain possible interpopulation differences in gonadal output. Sex ratio in Medes Islands populations of both species was 1:1, while in Cape of Palos sex ratio was significantly male biased (1:7) in P. clavata, and female biased (1.7:1) in E. singularis populations. Spawning timing occurred in all cases coinciding with a marked increase in sea-water temperature in spring, and after the most successful feeding season, but comparing localities there was a clear temporal shift in the time of gametes release, appearing well linked to the shift in sea-water temperature rising in spring in both sites at the depth where populations are placed. Therefore, in this study the temperature appears as the main synchronizing factor of gonadal development within these populations. Significant differences in gonadal volume per polyp were found in both species owing mainly to differences in the number of gonads per polyp between populations, with Cape of Palos populations displaying higher values in both studied species, suggesting that the exposition to different local conditions may be reverted in a different gonadal output. But the observed patterns in lipid concentrations levels in gorgonians disable us to conclude that lipid concentration levels explain the observed differences in gonadal output found in this study.  相似文献

Little is known concerning the fine-scale diversity, population structure, and biogeography for Symbiodinium spp. populations inhabiting particular invertebrate species, including the gorgonian corals, which are prevalent members of reef communities in the Gulf of Mexico, the Caribbean, and the western Atlantic. This study examined the Symbiodinium sp. clade B symbionts hosted by the Caribbean gorgonian Pseudopterogorgia elisabethae (Bayer). A total of 575 colonies of P. elisabethae were sampled in 1995 and 1998–2000 from 12 populations lying along an ~450 km transect in the Bahamas and their Symbiodinium sp. clade B symbionts genotyped at two polymorphic dinucleotide microsatellite loci. Twenty-three unique, two-locus genotypes were identified in association with these P. elisabethae colonies. Most colonies hosted only a single Symbiodinium sp. clade B genotype; however, in some instances ( n=25), two genotypes were harbored simultaneously. For 10 of the 12 populations, 66–100% of the P. elisabethae colonies hosted the same symbiont genotype. Added to this, in 9 of the 12 populations, a Symbiodinium sp. clade B genotype was either unique to a population or found infrequently in other populations. This distribution of Symbiodinium sp. clade B genotypes resulted in statistically significant ( P<0.05 or <0.001) differentiation in 62 of 66 pairwise comparisons of P. elisabethae populations. Tests of linkage disequilibrium suggested that a combination of clonal propagation of the haploid phase and recombination is responsible for maintaining these distinct Symbiodinium sp. clade B populations.  相似文献

The sperm kinetics and fertilisation literature in marine invertebrates is heavily biased toward free-spawning species. Nonetheless, many species (e.g. cephalopods) transfer and/or fertilise gametes in confined external spaces or internally, creating very different selective pressures on sperm storage, sperm longevity and hence sperm competition. Here we report the results of an investigation into the effects of sperm age, water temperature and sperm concentration on sperm motility in the giant cuttlefish (Sepia apama). Significant positive correlations were found between percent motility and sperm concentration, and between sperm motile speed and sperm concentration. Mean percent motility of cuttlefish sperm suspension was still 9% eight hours after being released from the spermatophore and diluted into filtered seawater at 12°C (ambient field temperature during the spawning season). Sperm resuspended from spermatangia taken from (mated) females in the field were motile for up to 100 hours. When spermatophores were stored at 4°C motility was still observed in resuspended sperm after two months. Our results show that spermatangia and spermatophores can retain and release live sperm for long periods. The observed longevity of sperm in S. apama greatly increases the potential for sperm competition in this species.  相似文献

The study deals with the toxicological impact of cadmium nanoparticles (Cd NPs) on Bacillus subtilis as a model Gram-positive bacterium. Cadmium oxide (CdO) NPs (~22 nm) and cadmium sulfide (CdS) NPs (~3 nm) were used in this study. Both the NPs were found to inhibit the cell viability of B. subtilis when added to the culture at mid-log phase, the viable cell number declined with increasing concentration of Cd NPs. At mid-log phase, 15 mg L?1 CdO NPs inhibited growth by ~50%, whereas at 30 mg L?1 growth completely ceased. Under the same conditions, CdS NPs inhibited growth by ~50% at a concentration of 8 mg L?1, and at 20 mg L?1 growth was completely retarded. The cells changed their morphological features to a filamentous form with increasing Cd NPs exposure time, leading to associated with clumping. NPs treated cells when stained with 4′, 6-diamino-2-phenylindole, showed filamentous multinucleated bead structure, suggesting irregularities in cell division. Increasing intracellular oxidative stress due to Cd NPs exposure might be one of the reasons for the cell morphological changes and toxicity in B. subtilis.  相似文献

Axial skeletons of two closely related, sympatric species of gorgonians were examined for details of gross morphology, internal organization and fine structure. Banded fibers with a periodicity of 430 Å and oriented circum-axially to the longitudinal axis of the skeleton (i.e., lying on the plane perpendicular to the axis), were revealed by electron microscopy in the axial skeleton of Muricea californica; they indicate the presence of a collagen-like protein in the gorgoin of this species. No circum-axial fibers were found in the axial skeleton of M. fruticosa. Differences in fine structure are related to gross morphological differences in the skeletons. A sequence of events for the addition of new material to the axial skeleton (growth) is described based on histological observation. Iodine-125 is used as a tracer for the deposition of new skeletal material, and is suggested as a new method for studying gorgonian growth rates.  相似文献

This paper presents the results of a preliminary study on the uptake of 45Ca in the mesoskeleton of the gorgonian Eunicella papillosa (Octocorallia). Hourly seawater aliquots over 12 h periods displayed considerable fluctuations of activity with time, reflecting a disappearance and reappearance of tracer during the experiment. Analysis of total coenenchyme mass showed an initial rapid but fluctuating uptake of tracer and a progressive decrease with time. Distinct differences in the calcification rate of specific growth regions of the coral were detected, the rate being higher in branch tips than in lower branch regions. Chase experiments on colonies previously incubated for 24 h revealed that up to 45% of the tracer taken up during labelling were returned to the seawater. A comparison of the tracer content of the coenenchyme and the calcite spicules revealed that up to 70% of the calcium taken up remained in the coral tissue. Quantification of isotopic exchange phenomena with dead corals and isolated spicules emphasized the importance of the live coral tissue as a barrier and regulator of Ca uptake. The results are discussed in the light of the limitations of radioisotope techniques for the determination of calcification rates, and an attempt to compare rates with data in the literature is made.  相似文献

The skeletons of 3 gorgonian and 2 antipatharian corals were subjected to quantitative chemical analysis. Protein values ranged from 70.4 to 93.6%; ash from 3.0 to 19.4%; lipid from 0 to 8.4%; carbohydrate from 1.24 to 3.94% and halogen from 4.2 to 24.9% of the dry skeletal weight. Hydroxyproline and phenolic values were 0 to 3.0% and 11.6 to 25.0% of the protein, respectively. Lipid, present in 2 gorgonian species and 1 antipatharian, significantly decreased with age in all three cases. Glucose and galactose accounted for over 90%, and sialic acids for an additional 1 to 10%, of the carbohydrate in the gorgonian skeletons studied; the glucose content of the gorgonian skeleton decreased with age. The antipatharian skeletons possessed no glucose or galactose, but contained significant levels of amino sugars; the presence of chitin is confirmed. In the gorgonians, bromine and iodine, the predominant halogens, increased with skeletal age and were present in nearly equal amounts. Small amounts of bromine were found in the antipatharian skeletons, but very large amounts of iodine were found in older parts of the skeleton. The basal regions of both antipatharian species contained >23% by weight of iodine, the highest content of iodine yet recorded for any organism.  相似文献

Gorgonian species may contribute to the three-dimensional seascape in soft bottom-gravel areas, but the information on their biology and ecology is very scarce. The biometry and secondary production of the Mediterranean soft bottom-gravel passive suspension feeder Leptogorgia sarmentosa (Cnidaria: Octocorallia) was studied using photographic monitoring of the primary branches from May 1998 to September 2000. The primary branches observed had a high density of polyps (2.2 ± 0.2 SD polyps mm−1) and a high organic matter content (63.2 ± 9.1 SD %). During the two-year sampling period, there was a net negative growth in 90% of the gorgonian population. The mean loss during the 27-month period was −2.9 ± 0.9 SD cm per branch (5.7 mg C branch−1). However, considering only the initial and final diameters and maximum height in the 27 months elapsed time, the gorgonians showed positive growth, which meant that photographic sampling of single branches was a more appropriate method for gorgonian secondary production monitoring. A water mass anomaly detected in 1999 in the north-western Mediterranean Sea may have been the cause of the net negative growth in L. sarmentosa in the studied area. Partial mortality due to different factors, such as strong currents, predation, disease, etc., could be a common strategy in sessile colonial benthic populations that would facilitate their maintenance even during very stressful circumstances.  相似文献

Medicinal herbs are steadily increasing in complementary use for chronic and alterative therapies. The health risks associated with herbal supplements have therefore been identified as a top research priority. Tecoma stans (Tronadora), a Bignoniaceae plant, is a herbal drug traditionally consumed as tea in South America for the control of diabetes. It contains the alkaloids of tecomine and tecostanine which were shown as functional compounds responsible for hypoglycemic activity. However, the side effects of aqueous extracts of this herbal tea have not been reported. In this study, studies sought to evaluate the cytotoxicity of water extracts from T. stans in human hepatoblastoma (HepG2). Toxic effects of T. stans were concentration- and time-dependent in the presence and absence of fetal bovine serum (FBS). Cells were incubated for up to 72-h with varying concentrations of herbal extracts (60–100%), cytotoxicity was determined spectrophotometrically by MTT and reported in terms of % cell viability. For IC50 assay (24 h exposure), cytotoxicity was found at concentration of 60–100%. Evaluation of the effects of T. stans and Brickellia cavanillesi (Asteraceae) in combination revealed that extracts of both herbal extracts' nontoxic levels of T. stans significantly reduced cell viability in a time-dependent manner. Further studies are needed regarding the identification, toxicity, and molecular mechanism of action of active compounds, as well as their cytotoxicity to other cell lines following exposure to both single and combined herbal extracts of these plants with anti-diabetic properties.  相似文献

Effects of oceanic and terrestrial sediment suspensions on the behavioral traits activity, colony expansion, and mucus formation of the gorgonian octocoral Subergorgia suberosa (Pallas, 1766) were studied in the laboratory. Increased sediment concentration significantly changed the logits of activity indices (as colony expansion) and was correlated with decreased activity. For both sources of sediments, the activity index decreased as the concentration of sediment increased. The level of decrease was more significance for terrestrial sediments. The activity index showed no clear time pattern for the control group, but a decrease over time occurred in both sediment treatment groups. Activity change was linearly correlated with exposure time and concentration of terrestrial and ocean sediments. The activity indices of polyps were negatively correlated with the formation of mucus sheets (correlation = −0.536, p = 0.048). For both sources of sediments, the frequency of mucus sheet formation (MSF) increased with sediment concentration. Increases in terrestrial sediment loads resulted in more striking effects. The probability for MSF under different sediment groups at different concentrations over time was modeled by regressions. Frequency of polyp activity is suggested as a suitable biomarker to monitor the effects of sedimentation stress.  相似文献

Clonal reproduction, a common life history strategy among sessile marine invertebrates, can lead to high local abundances of one to a few genotypes in a population. Analysis of the clonal structure of such populations can provide insight into the ecological and evolutionary history of the population, but requires markers that can identify individual genets. Forensic and demographic studies have demonstrated that DNA fingerprinting can provide markers that are unique for an individual genotype. We have generated DNA fingerprints for over 70 colonies of the clonal gorgonian, Plexaura A (Plexaura sp. A) collected from June 1990 through July 1991 in the San Blas Islands, Panama. DNA fingerprints within a singic individual were identical and fingerprinting resolved multiple genotypes within and among reefs. On one reef in the San Blas Islands, Panama, 59% of the colonies sampled were of one genotype and this genotype was not found on any other sampled reefs. A previous study using tissue grafts identified 13 putative clones on these reefs, while DNA fingerprints of the same colonies differentiated 17 genotypes. The present study demonstrates the utility of DNA fingerprinting for distinguishing clones and for identifying clonal structure of marine invertebrate populations.  相似文献

以3种氨基糖苷类(AG)抗生素:硫酸安普霉素(APR)、双氢链霉素(DIH)和硫酸链霉素(STS)为研究对象,以生态系统中2类重要的水生生物分解者如青海弧菌(Vibrio qinghaiensis sp.-Q67,Q67)和生产者蛋白核小球藻(Chlorella pyrenoidosa,CP)为受试生物,运用均匀设计射线法设计抗生素三元混合物体系,共5条具有不同浓度配比的射线,应用已建立的分别基于Q67和CP的时间毒性微板分析法系统测试抗生素及其三元混合物射线对Q67和CP在不同暴露时间的毒性。对于Q67和CP,暴露时间分别为0.25、2、4、8、12 h和12、24、48、72、96 h。应用浓度加和(CA)模型分析混合物在不同暴露时间的毒性相互作用。结果表明:APR、DIH和STS及其5条混合物射线对2种指示生物的毒性均具有明显的时间依赖性,且Q67对AG抗生素及其混合物射线的响应比CP的灵敏;以半数效应浓度的负对数p EC50值为毒性大小指标,3种抗生素对2种指示生物的毒性大小顺序随暴露时间的变化而变化,3种AG抗生素对Q67和CP分别在12 h和96 h的毒性大小顺序均为STSDIHAPR;5条具有不同浓度配比的混合物射线对Q67在不同暴露时间的毒性均呈加和作用,但对CP的毒性既有加和作用也有拮抗作用,且拮抗作用随暴露时间和组分浓度配比的变化而变化,表明AG抗生素毒性的联合毒性作用与暴露生物、暴露时间以及混合物组分的浓度配比等有关。  相似文献

Our previous study suggested that toxicokinetic parameters of fluoride were significantly changed on the 30th day as compared with 1st day, after repeated oral administration of sodium fluoride alone for 30 days in goats. The purpose of this study was to investigate whether aluminum sulfate has ameliorative effect on the toxicokinetics of fluoride. For this, sodium fluoride (20 mg kg?1 b. wt.) along with aluminum sulfate (150 mg kg?1 b. wt.) was administered in goats for 30 days to investigate its effect on toxicokinetics of fluoride on days 1 and 30. The fluoride levels against time were adequately described by one compartment open model. The peak plasma level of fluoride was observed at 2 h. The toxicokinetics data revealed a nonsignificant decrease in β (elimination rate constant) and C max (the maximum plasma concentration) and increase in t 1/2 K a (absorption half-lives), t 1/2 β (elimination half-lives), area under the plasma (AUC) concentration versus time curve and mean residence time (MRT) was observed for the last dose from their respective first dose values. On the basis of changes in the toxicokinetic parameters, it was concluded that repeated oral administration has cumulative effect on plasma fluoride level and concurrent oral administration of aluminum sulfate has ameliorative action on the toxicokinetics of fluoride in goats.  相似文献
