The diffusive gradients in thin films (DGT) were field deployed alongside the shrimp Litopenaeus vannamei at seven sites with different levels of contamination to assess the potentially bioavailable and toxic fraction of metal contaminants. After 7 days of exposure, several antioxidant biomarkers were quantified in hepatopancreas of exposed shrimps, and tissue levels as well as the total, dissolved, and DGT-labile concentrations of metal contaminants were determined in the pooled site samples. The results showed that the caged shrimps had high tissue contaminant concentrations and significantly inhibited antioxidant responses at the more contaminated sites. DGT-labile metal concentrations provided better spatial resolution of differences in metal contamination when compared with traditional bottle sampling and transplanted shrimp. The total, dissolved, and DGT-labile metal fractions were used to evaluate the potential bioavailability of metal contaminants, comparing with metal accumulation and further linking to antioxidant biomarker responses in tissues of exposed shrimps. Regression analysis showed the significant correlations between DGT-Cu concentrations and tissue-Cu and activities of some biomarker responses in the shrimp hepatopancreas. This indicated that DGT-labile Cu concentrations provided the better prediction of produced biological effects and of the bioavailability than the total or dissolved concentrations. The study supports the use of methods combining transplanted organisms and passive sampling for assessing the chemical and ecotoxicological status of aqueous environments and demonstrates the capability of the DGT technique as a powerful tool for measuring the bioavailability-based water quality in variable coastal environments.  相似文献

Temporal fluctuations in metal (Cd and Cu) concentrations were monitored over four months (May to August) in the liver of juvenile yellow perch (Perca flavescens) sampled from four lakes situated along a metal concentration gradient in northwestern Quebec: Lake Opasatica (reference lake, low metal concentrations), Lake Vaudray (moderate metal concentrations) and lakes Osisko and Dufault (high metal levels). The objectives of this study were to determine if hepatic metal concentrations and metal-handling strategies at the sub-cellular level varied seasonally. Our results showed that Cd and Cu concentrations varied most, in both absolute and relative values, in fish with the highest hepatic metal concentrations, whereas fish sampled from the reference lake did not show any significant variation. To examine the sub-cellular partitioning of these two metals, we used a differential centrifugation technique that allowed the separation of cellular debris, metal detoxified fractions (heat-stable proteins such as metallothionein) and metal sensitive fractions (heat-denaturable proteins (HDP) and organelles). Whereas Cd concentrations in organelle and HDP fractions were maintained at low concentrations in perch from Lakes Opasatica and Vaudray, concentrations in these sensitive fractions were higher and more variable in perch from Lakes Dufault and Osisko, suggesting that there may be some liver dysfunction in these two fish populations. Similarly, Cu concentrations in these sensitive fractions were higher and more variable in perch from the two most Cu-contaminated lakes (Dufault and Osisko) than in perch from the other two lakes, suggesting a breakdown of homeostatic control over this metal. These results suggest not only that metal concentrations vary seasonally, but also that concentrations vary most in fish from contaminated sites. Furthermore, at the sub-cellular level, homeostatic control of metal concentrations in metal-sensitive fractions is difficult to maintain in perch with high hepatic metal concentrations.  相似文献

The effect of bleached kraft pulp mill effluent (BKPME) persistent compounds on phagocyte activities and its organ-specific influence in gill, kidney and liver was studied in European eel (Anguilla anguilla L.). Eels were caged and plunged at 3 different sites-50 m (site 1), 100 m (site 2) and 2,000 m (site 3) away from the end of the closed BKPME discharging channel for 8 and 48 h. Gill, head kidney and peritoneum phagocytes oxidative burst activity (OBA) was measured by the nitroblue tetrazolium reduction assay whereas lipid peroxidation (LPO) in eel gill, kidney and liver was measured by thiobarbituric acid reaction. A significant gill OBA induction was found at 8h on site 2, and on sites 1, 2 and 3 at 48 h exposure. However, in head kidney and peritoneal exudate phagocytes, OBA induction was significant only at sites 2 and 3 after 48 h exposure. In those particular sites, a significant increase in gill, kidney and liver LPO was measured that is assumed to result from OBA induction. Considering OBA and LPO, gill is the most affected tissue compared to kidney and liver. Gill vulnerability towards peroxidative damage was demonstrated at 8h on site 2 and at 48 h on sites 1, 2 and 3, whereas in kidney was observed at sites 2 and 3 only at 48 h. Liver LPO increased at site 2 only after 48 h exposure. Our results demonstrate that the OBA activation pattern in gill and kidney is associated with the induced peroxidative damage extent in those organ, together with water pollution the exposure route, resulting from previous BKPME effluent sediment contamination which may affect the activation pattern of circulating fish phagocytes.  相似文献

Polychlorinated biphenyls (PCBs), hexachlorocyclohexane (HCH) isomers, dichlorodiphenyltrichloroethane (DDT) and its metabolites, other organochlorine pesticides such as hexachlorobenzene (HCB), chlordane compounds (CHLs, including trans-chlordane and cis- and trans-nonachlor) and the heavy metal mercury were quantified in muscle and liver of the European chub (Leuciscus cephalus, Linnaeus, 1758) and in the muscle of the barbel (Barbus cyclolepis, Heckel, 1837) at two sampling sites of the River Nestos, Greece. PCBs in muscle and DDTs in the liver tissues were the predominant organochlorinated contaminants. Among the PCBs, congeners 47 (up to 9.60 ng g(-1) wet wt.), 180 (up to 1.15 ng g(-1) wet wt.) and 190 (up to 1.50 ng g(-1) wet wt.) were the most frequent and abundant. The contamination degree by the sum of PCBs on the fish tissue samples from the River Nestos is lower or similar to PCB levels found in other ecosystems. Among the organochlorine pesticides, essentially only p,p'-DDD, p,p'-DDE and alpha-, beta- and gamma-HCH were found, with the former appearing at mean levels up to 30.71 ng g(-1) wet wt. From a public health standpoint, residue organochlorine pesticide levels from our work are considerably lower than the recommended tolerance limits. Finally, mean values of Hg in chub were significant lower (up to 31.04 ng g(-1) wet wt.) compared to those detected on barbel (up to 169.27 ng g(-1) wet wt.). The concentrations of Hg in fresh water fish from the River Nestos did not exceed WHO and US EPA health guidelines, and were suitable for human consumption.  相似文献

Heavy metal accumulation was studied in urban populations of moles, Talpa europaea L. (Insectivora) at three sites along the "Donaukanal" in Vienna. Kidneys, liver, lung, gonads, spleen, pancreas, femur, stomach + content, heart and skin were analysed for Zn, Cu, Pb and Cd contents. Female moles showed higher Zn concentrations in spleen, gonads and skin, higher Cd concentrations in femur and stomach and lower Cu content in kidneys compared to males. Age classes were determined using teeth section. A significant increase in Cd concentration with age was found in kidneys, liver, heart, lung, skin and femur, with the highest increase for kidney samples. Pb accumulated in bone tissue (femur) with increasing age. Although significant site differences in heavy metal concentrations of soil and vegetation samples existed, these differences were only partly reflected by tissue contents.  相似文献

Environmental monitoring by means of biomarker parameters assessed in different species is a useful tool. It has the advantage of providing a quantitative response as well as valuable information of ecological relevance on the chronic adverse effects caused by water pollution. The aim of this study was to assess the response of biochemical and physiological parameters of Cnesterodon decemmaculatus, a native teleost, simultaneously caught in two sites of Reconquista river, a highly polluted peri-urban river. This study compared the measured parameters with that of specimens of the same species captured in an unpolluted body water, and correlated the detected changes with the physicochemical profile of the water at each site. A comparison was made of selected parameters of gill, brain and liver and of somatic indices of fish collected from polluted and reference sites. The main parameters whose changes allowed to discriminate between sampling sites were gill (Na(+),K(+))-ATPase, brain acetylcholinesterase (AChE) and liver aminotransaminases activities; tissues' protein content and liver somatic index (LSI) were also sensitive biomarkers in brain and liver, respectively. The results showed that the response of the measured biomarkers allowed for the differentiation of sampling sites according to their water quality and confirmed that Cnesterodon decemmaculatus may be a useful test organism for the biomonitoring of freshwater environments. In addition, the simultaneous measurement of the physicochemical parameters of the water samples showed a good correspondence between the biomarkers responses and the environmental chemical stress conditions.  相似文献

In order to find the causative agent in frequently occuring fish kills in a Belgian river a series of toxicity tests has been conducted in which rudd (Scardinius erythrophtalmus) were exposed to acute lethal and subacute non lethal concentrations of copper, chromium, cadmium and zinc. The concentrations of these metals in gills, opercle, kidney, liver and muscle were measured. Metal levels in gills were the most valuable indicator of acute lethal exposure. This information was compared with levels found in rudd from a surface water storage reservoir and from the river Meuse. Fish collected after fish kills in the river Meuse were analysed. In one case copper could be identified as one of the toxicants concerned by fish tissue analysis. Metal levels in fish tissues can give valuable additional information concerning the cause of kills provided that background information is available about metal levels in water and normal tissue levels.  相似文献

Passive sampling devices (PSDs) sequester the freely dissolved fraction of lipophilic contaminants, mimicking passive chemical uptake and accumulation by biomembranes and lipid tissues. Public Health Assessments that inform the public about health risks from exposure to contaminants through consumption of resident fish are generally based on tissue data, which can be difficult to obtain and requires destructive sampling. The purpose of this study is to apply PSD data in a Public Health Assessment to demonstrate that PSDs can be used as a biological surrogate to evaluate potential human health risks and elucidate spatio-temporal variations in risk. PSDs were used to measure polycyclic aromatic hydrocarbons (PAHs) in the Willamette River; upriver, downriver and within the Portland Harbor Superfund megasite for 3 years during wet and dry seasons. Based on an existing Public Health Assessment for this area, concentrations of PAHs in PSDs were substituted for fish tissue concentrations. PSD measured PAH concentrations captured the magnitude, range and variability of PAH concentrations reported for fish/shellfish from Portland Harbor. Using PSD results in place of fish data revealed an unacceptable risk level for cancer in all seasons but no unacceptable risk for non-cancer endpoints. Estimated cancer risk varied by several orders of magnitude based on season and location. Sites near coal tar contamination demonstrated the highest risk, particularly during the dry season and remediation activities. Incorporating PSD data into Public Health Assessments provides specific spatial and temporal contaminant exposure information that can assist public health professionals in evaluating human health risks.  相似文献

In this study samples of ten species of fish were analyzed for concentrations of organochlorine pesticides, PCBs and heavy metals (Pb, Cd, and Cu). Fish were captured using electric fishing on ten sites along the Dr?me river (Rh?ne-Alpes region). Quantitative determination of the organochlorine and PCBs compounds was performed by gas chromatography-electron-capture detection (GC-ECD). The concentrations of heavy metals were determined by atomic absorption spectrophotometry. Samples contained detectable concentrations of lindane, PCBs, and heavy metals but at concentrations below the maximum residue limit (MRL). Non-parametric statistical analysis was performed to distinguish groups of sites with different levels of contamination. PCBs concentrations increased along the river with four groups of sites significantly different from each other. Cadmium concentrations were below the MRL. Lead contamination showed two groups significantly different and a repartition similar to PCBs. Copper contamination was correlated with the localization of vineyards. We assessed the potential effects of contamination the otter (Lutra lutra). The concentrations of all pollutants analyzed in fish sampled in this study are lower than the threshold values described in literature. The Dr?me river is relatively unpolluted river, and the establishment of otter populations should not be affected by pollution.  相似文献

This study assesses the applicability of a sentinel fish species as a biomonitor of metal pollution. Gudgeon (Gobio gobio) were collected from 14 sites in metal polluted lowland rivers in Flanders, including a clear Cd and Zn pollution gradient. Fish tissues were analysed for Cd, Cr, Cu, Ni, Pb and Zn. Metal levels in organs were related to levels in water and sediment. At some sites Cd and Zn levels were up to 50 times higher compared to reference values and literature. Significant relationships between Cd and Zn in fish tissue and environmental levels were found, especially in the pollution gradient where up to 70% of the observed variation could be described. Condition factor differed significantly among the sampling sites but could not be directly related to the environmental metal levels. Nevertheless, in the pollution gradient, threshold tissue concentrations could be defined above which the condition factor was always low.  相似文献

Priority organic pollutants were investigated in sediments and fish collected along the Ebro river basin (NE Spain) to evaluate their occurrence, transport and bioavailability. Sediments were collected in 18 sites and two species of fish were captured in nine sites according to the availability in each area. The sampling sites covered industrial, urban and agricultural areas. Four methods were used to detect 20 organochlorine compounds (OCs), 8 polycyclic aromatic hydrocarbons, 3 organotin compounds, 2 alkylphenols and 40 polybrominated diphenyl ethers (PBDEs) from purified extracts. The contamination pattern was site specific and no downstream increase in concentration of pollutants was observed but rather a generalized low level diffuse pollution. Target compounds were detected in sediments at 0.01 to 2331 microg/kg dry weight, and only OCs and PBDEs were accumulated in benthopelagic fish. Toxicological assessment was performed according to predicted environmental levels and revealed sites where adverse effects could occur.  相似文献

The contamination of water by metal compounds is a worldwide environmental problem. This study was undertaken to evaluate the impact of short-term cadmium exposure on metabolic patterns of the freshwater fish Oreochromis niloticus. The fish were exposed to 320, 640, 1,280 and 2,560 microg/l sublethal concentrations of Cd++ (CdCl2) in water for 7 days. The specific activities of the enzymes phosphofructo kinase (PFK-E.C.2.7.1.11.), lactate dehydrogenase (LDH-E.C.1.1.1.27.) and creatine kinase (CKE.C.2.7.3.2.) were decreased in white muscle after cadmium treatments, indicating decreases in the capacity of glycolysis in this tissue. Cadmium exposure induced increased glucose concentration in white muscle of fish. On the other hand, cadmium exposure at sublethal concentrations increased phosphofructo kinase and LDH in red muscle of fish. Cadmium significantly decreased total protein concentrations in liver and white muscle regardless of tissue glycogen levels. The data suggest that cadmium acts as a stressor, leading to metabolic alterations similar to those observed in starvation.  相似文献

Histological and Immunohistochemical studies were carried out to document the possible impact of heavy metal contamination in different tissues of Chanos chanos. Heavy metals such as Cu, Pb, Zn, Cd, Mn and Fe were predominant in water, sediment and biota of Kaattuppalli Island and varied significantly between two different sites. Histological changes such as swelling of muscle fiber and break down of muscle bundles were noted in the muscle. Similarly gill filament cell proliferation, increase in intercellular spaces and primary and secondary lamellar epithelium were evident in gills. The hepatocytes showed damage of central vein and rupture of irregular hepatic plate with more number of vacuoles in the fish collected from polluted site compared to that from the less polluted site. The impact of pollution was also assessed in different tissues by immunohistochemistry using primary antibody (mouse monoclonal HSP70 antibody 1:2000) and secondary antibody (HRP conjugated antibody) for expression of stress protein. Immunostaining analysis showed expression of HSP70 with high intensity in the tissues of fish collected from polluted site compared to less polluted sites. Further, HSP70 positive cells were analyzed from six locations per fish tissue section. One-way analysis of variance (ANOVA) followed by least significant difference (LSD) was used to check if the expression was significant. Results indicate that the values are statistically significant at the two different sampling sites (P<0.05).  相似文献

Concentrations of antimony in invertebrates and small mammals from grasslands in the vicinity of an antimony smelter were significantly elevated compared to a control site. Higher concentrations of antimony were recorded in liver, lung and kidney tissue of herbivorous and insectivorous mammals from the contaminated sites. However, there is little evidence of bioaccumulation of antimony in food chains represented by the soil-vegetation-invertebrate-insectivore pathway of the grasslands, and little indication of significant accumulation by herbivorous mammals despite marked contamination of their diet.  相似文献

Small mammals were trapped, tagged and recaptured in 0.45 ha plots at six hazardous industrial waste disposal sites to determine if populations, body mass and age structures were different from paired control site plots. Low numbers of six species of small mammals were captured on industrial waste sites or control sites. Only populations of hispid cotton rats at industrial waste sites and control sites were large enough for comparisons. Overall population numbers, age structure, and body mass of adult male and female cotton rats were similar at industrial waste sites and control sites. Populations of small mammals (particularly hispid cotton rats) may not suffice as indicators of environments with hazardous industrial waste contamination.  相似文献

Milkfish (Chanos chanos) from four fish-culture ponds adjacent to different metal recovery facilities along the Er-Jen River area, Tainan, Taiwan, were sampled to determine their metal contents. Chemical analysis showed that fish tissue contained different concentrations of Cu: 0.71-6.37 micrograms/g, Pb: ND (not detectable) approximately 41.04 micrograms/g, Cd: ND approximately 0.41 microgram/g, Al: 6.75-64.11 micrograms/g, Ni: 0.062-0.504 microgram/g and Zn: 16.11-41.86 micrograms/g. The average concentrations of Cu, Al, Zn, Cd and Pb in fish samples from some of the ponds were significantly higher than those from the reference pond. In addition, there were variations in metal concentrations of fish collected from different ponds. Pond D had the highest mean values of Cu, Cd and Zn, and Pond B of Al and Pb. Further investigations are needed to determine the source of metal contamination in the fish.  相似文献

From June 1993 to October 1994, studies have been carried out on the effects of mercury in the Oder River and pike tissue contamination (muscle, kidney, liver). The mean mercury contents in the sediment range from 0.03 to 1.1 mg/kg dry weight. In the pike muscle, between 0.22 and 0.85 mg/kg, on a wet weight basis, were found. The measured mercury concentrations were analysed in relation to the number of macrophage centres of the liver, spleen and kidney of the pike. Positive correlations between mercury and MC response (0.54 ≤ r ≤ 0.79, p < 0.05) were found in all of these organs. The suitability of the macrophage-centre-response as a possible bioindicator for mercury pollution is discussed in the literature. In our study, the response of MCs was found to be suitable as a biomarker for the impairment of fish health.  相似文献

Heavy metal (Cd, Cr, Cu, Fe, Pb, Zn) concentrations in the muscle, gill and liver of six fish species (Sparus auratus, Atherina hepsetus, Mugil cephalus, Trigla cuculus, Sardina pilchardus and Scomberesox saurus) from the northeast Mediterranean Sea were measured and the relationships between fish size (length and weight) and metal concentrations in the tissues were investigated by linear regression analysis. Metal concentrations (as microg/g d.w.) were highest in the liver, except for iron in the gill of Scomberesox sauris and lowest in the muscle of all the fish species. Highest concentrations of Cd (4.50), Cr (17.1) and Pb (41.2) were measured in liver tissues of T. cuculus, Sardina pilchardus and A. hepsetus, respectively. The liver of M. cephalus showed strikingly high Cu concentrations (202.8). The gill of Scomberesox saurus was the only tissue that showed highest (885.5) iron concentrations. Results of linear regression analysis showed that, except in a few cases, significant relationships between metal concentrations and fish size were negative. Highly significant (P<0.001) negative relationships were found between fish length and Cr concentrations in the liver of A. hepsetus and M. cephalus, and Cr concentrations in the gill of T. cuculus. Cr and Pb concentrations in the liver and Cu concentrations in all the tissues of Scomberesox saurus also showed very significant (P<0.001) negative relationships. The biochemical response to chronic heavy metal exposure was studied in tissues of bank voles Clethrionomys glareolus. Animals were collected from three sites located 4, 8 and 30km from a zinc-lead smelter, the area's main source of metal contamination. Concentrations of Cd, Pb, Zn and Fe were measured in the liver, kidneys and gonads to assess the level of metal intoxication. In response to intoxication, organisms activate detoxification mechanisms which can protect animals from metals' toxicity. Glutathione plays an important role in toxic substance detoxification. Total glutathione (tGSH) and glutathione disulfide (GSSG) were measured in the tissues. Also, the activity of glutathione reductase (GR), glutathione peroxidase (GPX), and glutathione-S-transferase (GST) was measured in the studied tissues. Results indicate that levels of all studied parameters were tissue and site-dependent. Evidence indicates that the most sensitive parameter of metal toxicity for animals living in a chronically contaminated environment is the GSH/GSSG ratio. In our study, the GSH/GSSG ratio was decreased in the liver of animals with high Cd levels. However, the relationship between Pb and the GSH/GSSG ratio was positive in the gonads. Cadmium and lead negatively influenced GPX activity in the liver; this was probably connected with inhibition of the Se-dependent fraction. The relationship between iron and GR activity in the kidney was also negative, but other correlations for iron both in liver and kidney were not significant. Positive correlations between Zn levels and GST and GR activity were found in the gonads of bank voles.  相似文献

We investigated dioxin concentrations in freshwater fish in Japan by standardizing species to detect subtle decreasing trends of dioxin concentrations in the future with the reinforcement of regulations. The fish studied were crucian carp (Carassius auratus (gibelio) langsdorfii), an omnivorous species. Fish and sediments were collected from 14 rivers and lakes located in remote areas, agricultural areas, and small and large cities throughout Japan. The total toxic equivalent (TEQ) dioxin concentrations at the three remote sites were about 20% of the concentrations at the other 11 sites, which all had similar concentrations. The average concentrations in fish collected from these 11 sites were 0.69pgTEQg(-1) wet wt. (95% CI 0.52-0.85) or 57.3pgTEQg(-1) fat (95% CI 47.9-66.7). There were notable differences in congener profiles of polychlorinated p-dibenzodioxins (PCDDs) and polychlorinated dibenzofurans (PCDFs) between sampling areas. A chemical mass balance method was used to estimate the proportions of dioxin contributions from different sources. The sampling areas were classified into four groups on the basis of the predominant source of dioxin contamination: a combustion group, a chlornitrofen herbicide group, a chlornitrofen and pentachlorphenol herbicide group, and a remote group. The congener profiles of dioxin like polychlorinated biphenyls (DL-PCBs) were almost the same in all areas and close to those in Kanechlor, which indicates that pollution from PCB products has spread throughout Japan. From samples collected during the spawning season, it was determined that about 20% of the dioxins in adult females were transferred to the eggs, resulting in higher dioxin concentrations in adult males than in females. Biota-sediment accumulation factors (BSAFs) of 2,3,7,8-substituted PCDDs/DFs for crucian carp were larger than those of non-2,3,7,8-substituted congeners, and BASFs decreased with increasing number of chlorines. The BSAFs of DL-PCBs were 10 times greater than those of PCDDs/DFs, and BSAFs of mono-ortho type DL-PCBs were higher than those of non-ortho types.  相似文献
