Photosynthetic assimilation of 14CO2 by the symbiotic green alga Platymonas convolutae Parke et Manton in the marine flatworm Convoluta roscoffensis Graff has been investigated and compared with that in free-living P. subcordiformis and P. tetrathele. All Platymonas species investigated rapidly incorporate 14CO2 into a complex variety of soluble and insoluble assimilates. The rate of dark fixation is considerably lower in P. convolutae. Typical 14C-assimilate patterns are rather uniform in all Platymonas species, but the time courses of 14C-labelling of several compounds are very different. The percentage of 14C-aspartate and 14C-malate is significantly higher in P. convolutae after short-term-photosynthesis, whereas 14C-labelled phosphate esters predominate in the free-living Platymonas species. A comparison of the kinetics of 14C-labelling and of the distribution of 14C-activity between soluble and insoluble fractions suggests that glucose and fructose, not mannitol, as well as several amino acids (especially alanine) move from the algal partner to the tissue of the animal host. The significance of these findings is discussed.  相似文献

Six mediterranean macroalgae were cultivated for more than 2 yr under shade culture conditions, after which light requirements for growth were investigated at 16±2°C. The saturation light levels for growth in the logarithmic phase were related to the bathymetric distribution of the algae on the shore. The eulittoral to supralittoral red alga Bangia atropurpurea was saturated at a photon fluence rate of 71 mol photons m-2 s-1, the upper sublittoral to eulittoral brown algae Scytosiphon lomentaria, Colpomenia peregrina and Kuckuckia spinosa and the sublittoral brown alga Stictyosiphon soriferus at 39 to 71 mol photons m-2 s-1, and the deep-water alga Choristocarpus tenellus at 19 mol photons m-2 s-1. The minimum light requirements for growth of B. atropurpurea and C. tenellus were determined by observing length increase for 56 d under limiting light conditions. The compensation and minimum irradiances required for growth of B. atropurpurea were 0.5 and 1 mol photons m-2 s-1 respectively. The corresponding values for C. tenellus were 0.15 to 0.28 and 0.5 mol photons m-2 s-1 respectively. C. tenellus was the siowest-growing species tested at saturating light conditions, but it grew faster than B. atropurpurea at 1 mol photons m-2 s-1. Both B. atropurpurea and C. tenellus were able to survive 56 d in darkness, but only the latter grew under darkness in the first 14 d.  相似文献

Brown shrimp (Crangon crangon, L.) are subjected to a huge annual temperature range, and certain thermal conditions during winter have been identified to affect the brown shrimp population. Despite that, little is known about its thermal biology with regard to critically low temperatures. In the present study, we determined the critical thermal minima (CTmin) and the critical lethal minima (CLmin) of male and female brown shrimp of different body sizes in laboratory-based experiments. For the CTmin trials, shrimp were acclimated to 4.0, 9.0, and 14.0 °C and exposed to a cooling rate of ?0.2 °C min?1. In the CLmin trials, brown shrimp were exposed to a cooling rate of ?1.0 °C day?1 without prior thermal acclimation. Acclimation temperature significantly affected the temperature tolerance of brown shrimp (p < 0.001). CTmin among the experimental groups just varied slightly, and no clear effect of gender or body size was observed. In the CLmin trials, brown shrimp even tolerated the coldest temperature of ?1.7 °C that could be established in the experimental setup. However, we observed a negative relationship between temperature and reactivity within the range of 7.0 and 1.0 °C that was determined by means of the flicking response. This relationship suddenly broke between 1.0 and 0.0 °C where an abrupt drop in the reactivity of the shrimp became apparent. The results of this study revealed that brown shrimp hold a wider thermal range as originally reported and that it can cope with subzero temperatures. Implications of low-temperature tolerance are discussed in the context of the brown shrimp’s ecology as well as stock assessment.  相似文献

The combined effects of exposure to copper and temperature were investigated in adult specimens and germlings of the canopy-forming brown alga Fucus serratus. A matrix of four temperatures, 6, 12, 17 and 22 °C, and three concentrations of copper, 0, 100 and 1,000 nM total copper were used. Measured endpoints were growth rate, chlorophyll fluorescence parameters and for germlings also survival. The growth rate of adult specimens of F. serratus changed with increasing temperature. Growth tended to be negatively affected by high concentrations of copper when exposed to heat (22 °C) though not significantly so. The photosynthetic performance (i.e., chlorophyll fluorescence parameters: F v/F m, maximum electron transport rate (ETRmax) and maximum non-photosynthetic quenching (NPQmax) of adults was largely unaffected by both copper and temperature. Germling survival, growth rate and chlorophyll fluorescence parameters were affected by the combination of copper concentration and temperature. Increasing temperature led to reduced survival, increased rhizoid growth and higher F v/F m and ETRmax, whereas high copper concentration had a negative effect on the latter three endpoints. The negative effect of high copper concentration was amplified by high temperature. We conclude that juveniles of F. serratus are more susceptible to environmental stressors than adult specimens and recommend therefore including early life stages when assessing the risk of exposure to toxic compounds. Considering the response of adult specimens only may lead to false conclusions regarding the ecological impact of environmental stress.  相似文献

The short- and long-term fixation of 14CO2 by Phaeodactylum tricornutum was studied using methods of fractionation that allowed examination of all the products labelled with 14C. There was no doubt that the major pathway of CO2 fixation was into 3-phosphoglycerate, but there was also significant incorporation by -carboxylation by means of phosphoenol-pyruvate carboxylase and transamination into aspartate. At short time intervals (10 sec), 90% of the radioactive products found were accounted for by 3-phosphoglycerate and aspartate. The lipids associated with the photosynthetic apparatus contained a high proportion of the 14C fixed, which at 60 sec was located mainly in the carbohydrate portion of the lipids. At 30 and 300 min, the chlorophylls, carotenoids and the long-chain fatty acids were heavily labelled with 14C. The monogalacto-diglycerides, the digalacto-diglycerides and the sulpholipids each had a characteristic long-chain fatty acid composition. The cell proteins and a reserve polysaccharide were also labelled with 14C at short time intervals and increased their radioactivity in a linear fashion up to the longest period studied, 300 min. The activity of the enzymes ribulose diphosphate carboxylase and phosphoenolpyruvate carboxylase was sufficient to account for the pattern of fixation found.  相似文献

The mass mortality by disease of a localized population of sea urchins, Strongylocencrotus franciscanus, on the seaward side of a kelp forest was followed by the rapid seaward expansion of 4 species of brown algae, Macrocystis pyrifera, Laminaria dentigera, Pterygophora california, and to a lesser extent, Nereocystis leutkeana. One other brown alga, Cystoseira osmundacea, failed to become established in the newly available area. Competition among M. pyrifera, L. dentigera, P. californica, and N. Leutkeana apparently was severe, and within 1 year after the demise of the sea urchins, M. pyrifera formed a dense, nearly monospecific stand. Experimental removal of M. pyrifera demonstrated that the canopy of these plants limited light penetration to levels below that necessary for the growth and survival of other brown and red algae.  相似文献

Scutellidium lamellipes and Ampithoe tea live in patches formed by the brown alga Pelvetia fastigiata. The distributions and abundance of both crustaceans were examined on P. fastigiata obtained during 1975 and 1976 from 7 collecting and 6 experimental sites in the mid-intertidal zone at La Jolla, California (USA). At each collecting site, the abundance of each crustacean generally increases with increasing plant size. Following defaunation and transplantation of algae, abundances of both crustaceans on individual plants appear to be governed by equilibria determined by in situ reproduction, immigration and emigration. Immigration rates of both animal species decrease significantly with increasing distance from areas occupied by P. fastigiata. During short-term experiments, neither crustacean was able to establish colonies on defaunated plants placed across a 30 m P. fastigiata-free zone and both species would have to reproduce to persitst on plants transplanted to that site. More frequent immigration and reproduction (and, for S. lamellipes at least, decreasing emigration rates with increasing patch size) would increase abundances and decrease probabilities of local extinction on larger and/or less distant plants. However, even within sites with isolated plants, the smallest plants often are not inhabited by either crustacean. The densitics of both species differ with plant size and distribution. Densities of S. lamellipes are greatest within a large aggregation of P. fastigiata, whereas those of A. tea are greatest on moderate-sized isolated plants at mid-tide levels.  相似文献

The invertebrates living on specimens of the brown alga Pelvetia fastigiata, growing in the intertidal zone at La Jolla, California (USA) in November 1975 were enumerated. Within 7 collecting sites, larger plants generally shelter more animal species and individuals than smaller plants. The number of species on a given plant can be described as an equilibrium number; numbers of species and individuals can become similar on defaunated and transplanted algae of comparable sizes placed at the same experimental site. Such equilibria are site-specific because the colonization, immigration and loss rates of animals differ among the sites studied. Relationships between plant size and the number of animal species and individuals on P. fastigiata also differ among the collecting sites. The between-site differences are related to tidal level, to habitat diversity and to habitat patch-size. Small isolated plants without epiphytes shelter few species. The faunas of larger isolated plants at mid-tide levels generally include many thallus-dwelling, tubiculous, vagrant and epiphyte-dwelling species, but few such species commonly inhabit plants within aggregations of P. fastigiata. Within aggregations, the plants host relatively few epiphytes and thus lack habitat diversity, and the net emigration rates of many animals including epiphyte-dwellers are relatively high. Plants within aggregations, however, usually shelter more animal indivuduals than isolated plants. Thus, faunal diversity is reduced, not increased within the largest patches of P. fastigiata.  相似文献

Egg and faecal pellet production and egg hatching success of the calanoid copepod Calanus finmarchicus were monitored over a period of 14?days (14?C28 April, 2008) while fed water from 4 differently treated mesocosms and ambient water. Two of the mesocosms used were inoculated with the polyunsaturated aldehyde (PUA)-producing diatom Skeletonema marinoi, while 2 received only nutrient additions with or without silica. The mesocosms developed blooms of S. marinoi, mixed diatoms or the haptophyte Phaeocystis pouchetii, respectively. Faecal pellet production of C. finmarchicus increased with increasing food availability. Egg production increased with time in all mesocosms to a maximum single female production of 232 eggs female?1?day?1 (average of 90 eggs female?1?day?1) and followed the development of ciliates and P. pouchetii, but was not affected by the observed high (up to 15?nmol?L?1) PUA production potential of the phytoplankton. The hatching success of the eggs produced on the mesocosm diets was high (78?C96%) and was not affected by either aldehydes in the maternal diet or exposure to the dissolved aldehydes in the water.  相似文献

Natural marine phytoplankton populations from the German Wadden Sea and unialgal cultures of the haptophycean Phaeocystis pouchetii were tested in 1989 under controlled UV-B stress conditions. Assimilation of 15N-nitrate in phytoplankton consisting mainly of P. pouchetii, or in pure cultures of this alga, was found to be very sensitive to enhanced UV-B dosage in comparison 15N-ammonia uptake. In contrast, in phytoplankton samples containing Ceratium spp., Coscinodiscus sp., Noctiluca sp. or others, rate of 15NO3 - uptake was higher and only slightly affected by UV-B irradiance compared to the P. pouchetii sample. UV-B inhibitory effect on uptake of inorganic nitrogen by P. pouchetii was more pronounced under strong white-light conditions and after a UV-B pre-illumination period of several hours than under low white light. Pools of glutamine and alanine decreased after UV-B exposure. Results are discussed with reference to the damaging effects of white light and UV-B on nitrogen metabolism.  相似文献

To investigate the potential use of Lentinus edodes (L. edodes) residue for Cd2+ adsorption, poly alcohol Na alginate (PVA) was applied to immobilize it. The parameters including contact time, pH, adsorbent dosages, and coexisting metal ions were studied. The suitable pH for immobilized L. edodes was 4?C7 wider than that for raw L. edodes (pH 6?C7). In the presence of Pb2+ concentration varying from 0 to 30 mg·L?1, the Cd2+ adsorption ratios declined by 6.71% and 47.45% for immobilized and raw L. edodes, respectively. While, with the coexisting ion Cu2+ concentration varied from 0 to 30 mg·L?1, the Cd2+ adsorption ratios declined by 12.97% and 50.56% for immobilized and raw L. edodes, respectively. The Cd2+ adsorption isotherms in single-metal and dual-metal solutions were analyzed by using Langmuir, Freundlich, and Dubinin-Radushkevich models. The Cd2+ adsorption capacities (q m) in single-metal solution were 6.448 mg·L?1 and 2.832 mg·L?1 for immobilized and raw L. edodes, respectively. The q m of immobilized L. edodes were 1.850 mg Cd·g?1 in Cd2+ + Pb2+ solution and 3.961 mg Cd·g?1 in Cd2+ + Cu2+ solution, respectively. The Cd2+ adsorption processes subjected to both adsorbents follow pseudo-second-order model. Mechanism study showed the functional group of L. edodes was -OH, -NH, -CO, and PVA played an important role in metal adsorbing. Mining wastewater treatment test showed that PVA-SA-immobilized L. edodes was effective in mixed pollutant treatment even for wastewater containing metal ions in very low concentration.  相似文献

The filtration rates of Mytilus edilis (=galloprovincialis; 40 mm) were determined in relation to food concentration and temperature, using pure suspensions of the unicellular alga Platymonas suecica in concentrations ranging from 3x105 cells/l to 1.5x108 cells/l. The rate of filtration (ml/h/mussel) generally decreased as cell concentrations increased, and dropped to low values when concentrations above 5x107 cells/l were supplied. The amount of water swept clear varied continuously, and noticeable differences in the filtration activity of M. edulis were observed over short time intervals (5 min). Fluctuations of filtered volumes per unit time were greater with lower than with higher concentrations of algae. The influence of temperature on filtration activity was highest between 5°–15°C and 25°–30°C. A temperature increase from 15° to 25°C resulted in only a slight increase in filtration rate. At 5° and 30°C, filtration dropped to very low values, namely 350 and 100 ml/h, respectively. The temperature coefficients for the filtration rates of M. edulis were determined as: Q10 (5° to 15°C)=4.96; Q10 (10° to 20°C)=1.22. The amount of algae cells ingested per mussel per hour is directly related to food concentration. The maximum number of cells filtered/mussel/h in an algal suspension of 70x106 cells/l was 21.5x105 cells/h. Cell concentrations of up to 40x106 cells/l were swept clear without producing pseudofaeces. The critical cell density for M. edulis was reached at algal concentrations of 70 to 80x106 cells/l. Above these concentrations no normal filtration activity was observed.  相似文献

The brown alga Fucus ceranoides L. was studied in several estuaries on the Isle of Man (Great Britain) in the summers of 1989 and 1990. The objective were to determine the success of natural fertilization in a dioecious organism with external fertilization and to contribute to our understanding of processes such as polyspermy blocks and propagule settlement under natural conditions. Gamete release occurred on a semilunar cycle near full and new moons; at such times, settlement densities of 500 zygotes cm-2 d-1 were common. Gamete release was largely restricted to daytime high tides. Fertilization success was high: about 95 to 100% of all eggs released were fertilized, and 1 to 9% of these zygotes were polyspermic. The incidence of polyspermy increased towards the upper limit of F. ceranoides in the estuary of the river Neb; this may be significant in the context of the Na+-dependent block against polyspermy. Levels of polyspermy were not related to fertilization success (R2=0.06). Rates of pronuclear migration and karyogamy determined in the laboratory were used as an internal clock to estimate when eggs were fertilized in the field. Most eggs were fertilized 30 to 120 min after plants were immersed by the incoming tide, which corresponded to the period 60 min before to 30 min after high tide. The high success of fertilization suggests that polyspermy blocks are important in nature, and, in combination with the high settlement of zygotes, shows that population size in F. ceranoides is determined by post-settlement mortality, not by propagule availability.  相似文献

In this continuing study on photosynthesis of the marine red alga Gracilaria conferta, it was found that ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) in crude extracts had a K m (CO2) of 85 M. Since seawater contains only ca. 10 M CO2, it appears that this alga must possess a CO2 concetrating system in order to supply sufficient CO2 to the vicinity of the enzyme. Because this species is a C3 plant (and thus lacks the C4 system for concentrating CO2), but can utilize HCO3 - as an exogenous carbon source, we examined whether HCO3 - uptake could be the initial step of such a CO2 concetrating system. The surface pH of G. conferta thalli was 9.4 during photosynthesis. At this pH, estimated maximal uncatalyzed HCO3 - dehydration (CO2 formation) within the unstirred layer was too slow to account for measured phostosynthetic rates, even in the presence of an external carbonic anhydrase inhibitor. This observation, and the marked pH increase in the unstirred layer following the onset of light, suggests that a HCO3 - transport system (probably coupled to transmembrane H+/OH- fluxes) operates at the plasmalemma level. The involvement of surface-bound carbonic anhydrase in such a system remains, however, obscure. The apparent need of marine macroalgae such as G. conferta for CO2 concentrating mechanisms is discussed with regard to their low affinity of Rubisco to CO2 and the low rate of CO2 supply in water. The close similarity between rates of Rubisco carboxylation and measured photosynthesis further suggests that the carboxylase activity, rather than inorganic carbon transport and intercoversion events, could be an internal limiting factor for photosynthetic rates of G. conferta.  相似文献

Because iron is not available generally in oxygenated sea water, it may be a limiting factor in marine primary production. This hypothesis was tested in the context of Davies Reef, Latitude 18°50′S (one of the coral reefs in the central region of the Great Barrier Reef system). Samples were collected for study in the period August, 1980 to March, 1981. Sea water around the reef contained ≦2x10-6 M Fe, surface sediments from the reef contained 66±26 (1 SD) ppm total Fe, and interstitial water near the surface contained ≧5x10-7 M Fe. Thus, Fe constituted a trace component of the reef environment, but limited Fe should be available to algae associated with the sediments. Specific biochemical analyses to test the Fe status of benthic photosynthetic organisms were carried out with a common blue-green alga, Phormidium sp., and a ubiquitous symbiotic dinoflagellate, Gymnodinium microadriaticum (zooxanthellae). The blue-green alga contained the electron transport protein, flavodoxin, which is found only in Fe-deficient organisms. Supporting evidence for Fe stress in this organism included chlorosis in the presence of plentiful biliprotein, and very low extractable photosynthetic cytochrome, c-553. The latter observations were shown to be the result of Fe deficiency in laboratory cultures of a blue-green alga, Synechococcus sp. These cultures showed that production of flavodoxin is not a universal response of algae to Fe stress, but that lowered cellular concentrations of Fe-containing proteins involved in photosynthesis probably is universal. The zooxanthellae from a soft coral, Sinularia sp., had three-fold lower total Fe and ferredoxin (an electron transport protein), than the same alga from a clam, Tridacna maxima. Thus, some algae in symbiotic associations may also suffer Fe-deficiency. It was concluded that the degree and extent of Fe-stress in primary producers on a coral reef may influence growth rates, biomass, and distribution of species.  相似文献

When the marine opisthobranchiate slug Hermaea bifida Mont. is incubated in a H14CO 3 - -seawater medium in the light, a considerable net gain of 14C-assimilates is observed. Electron microscopic control provided evidence that this 14C-fixation is due to photosynthesis by chloroplasts (=rhodoplasts) endosymbiotic in the cells of the digestive gland of the slug. After thin-layer chromatographic analysis various 14C-labelled photosynthates could be traced. The assimilate pattern of the rhodoplasts is compared with that of the marine red alga Griffithsia flosculosa (Ellis) Batt., from which the plastids are acquired by feeding. The nutritional relationship of this endosymbiosis is discussed, with emphasis on the occurrence of functional chloroplast endosymbioses among the eolidiform species of the Sacoglossa.  相似文献

We show the potentiality of coupling together different compound-specific isotopic analyses in a laboratory experiment, where 13C-depleted leaf litter was incubated on a 13C-enriched soil. The aim of our study was to identify the soil compounds where the C derived from three different litter species is retained. Three 13C-depleted leaf litter (Liquidambar styraciflua L., Cercis canadensis L. and Pinus taeda L., δ13CvsPDB ≈ ?43‰), differing in their degradability, were incubated on a C4 soil (δ13CvsPDB ≈ ?18‰) under laboratory-controlled conditions for 8 months. At harvest, compound-specific isotope analyses were performed on different classes of soil compounds [i.e. phospholipids fatty acids (PLFAs), n-alkanes and soil pyrolysis products]. Linoleic acid (PLFA 18:2ω6,9) was found to be very depleted in 13C (δ13CvsPDB ≈ from ?38 to ?42‰) compared to all other PLFAs (δ13CvsPDB ≈ from ?14 to ?35‰). Because of this, fungi were identified as the first among microbes to use the litter as source of C. Among n-alkanes, long-chain (C27–C31) n-alkanes were the only to have a depleted δ13C. This is an indication that not all of the C derived from litter in the soil was transformed by microbes. The depletion in 13C was also found in different classes of pyrolysis products, suggesting that the litter-derived C is incorporated in less or more chemically stable compounds, even only after 8 months decomposition.  相似文献

Phosphate depleted Pyrocystis noctiluca (Murray) Schuett 1895 has at least one phosphomonoesterase (EC 3:1:3:1) which is triphasic between 0.1 and 222 mol P. The enzyme has a broad temperature range with maximum activity at 50 °C and a Q10 of 1.4 to 1.5. A break in the Arrhenius plot at 35 °C implies the enzyme is membrane-bound. Cytological staining of whole cells and cell fractionation studies (showing 26 times higher specific activity in the particulate compared with the cytoplasmic fraction) suggest the enzyme is plasmalemma-bound. The enzyme has an absolute metal requirement which would be satisfied by Mg++ but not Mn++, Zn++, Fe++, or Co++ at seawater concentrations. Alkaline phosphatase is a stable enzyme whose activity is not altered by inhibitors of protein synthesis. Orthophosphate inhibition of enzyme activity was largely eliminated in the presence of these inhibitors. Apparently, a protein induced by PO4 3-, rather than PO4 3- itself, inhibits alkaline phosphatase. Cell-free alkaline phosphatase can hydrolyze a variety of phosphate esters and linear polymers of inorganic phosphorus as well as disolved organic phosphorus from tropical oceanic waters. These same hydrolysable organic and inorganic phosphorus compounds support the axenic culture growth of P. noctiluca, suggesting that naturally occurring hydrolysable organic phosphorus compounds may also support the growth of this alga.  相似文献

Laboratory-cultured larvae of the black chiton Katharina tunicata (Wood), collected from central California in May and June of 1979 and 1980, settled preferentially on the encrusting coralline alga Lithothamnium sp. Metamorphosis, which involves the loss of the prototrochal ciliary band, occurred within a period of 2.5 h after larvae began crawling upon the alga. In response to gamma-aminobutyric acid (GABA) at a concentration of 10-6 M, the lecithotrophic trochophore larvae ceased swimming and settled rapidly. Developmental metamorphosis did not follow settlement in the absence of encrusting coralline alga. However, when both GABA and Lithothamnium sp. were present, larval metamorphosis occurred within 2 h of settlement. Significantly different rates of settlement exist for sibling larvae of different ages; 12 d-old larvae responded more rapily and in greater numbers to treatment with GABA than 10 d-old larvae. Differences in settling rates are attributed to the onset of metamorphic competence.  相似文献

The South African abalone Haliotis midae is a commercially valuable species, but its numbers are seriously depleted due to illegal fishing. Overfishing not only affects the targeted species, but also potentially influences the integrity and functioning of the ecosystem through associated changes in community composition. We assessed the herbivorous effects of H. midae on the benthos through a 6-month field experiment involving exclusion/inclusion plots. This showed that benthic community composition remained largely unchanged whether H. midae was present or absent and specifically revealed no evidence that H. midae diminishes algal biomass, probably because in the region where we worked, it feeds mainly by trapping drift kelp rather than grazing on attached algae. The absence of responses to the removal of abalone was additionally evident in comparison with indices of abundance and diversity, functional groups and individual species. In the absence of abalone, there were subtle increases in the green alga Codium stephensiae and the crustose brown alga Zeacarpa leiomorpha and decreases in the four species of foliose macroalgae, but none of these effects were significant. The presence of H. midae thus had no negative grazing effects on any elements of the epilithic community composition. It does, however, have other influences, including occupation of space and provision of habitat in the form of its shell, which supports a community significantly different from the surrounding rocks.  相似文献
