Developing embryos and sexually mature follicles of the male portion of ovotestis proved to be a suitable material as a source of cleaving cells for advanced cytological investigations on the sacoglossan species Oxynoe olivacea Rafinesque, 1819 (Mollusca: Opisthobranchia). O. olivacea has a diploid chromosomal number of 30 made up of 15 pairs of which six are metacentric/submetacentric (M/SM), four subtelocentric (ST) and five on the borderline between SM and ST. Correspondingly, 15 bivalents occur in spermatocytes at Metaphase I. Constitutive heterochromatin is scarce and restricted to small C-bands seen in five pachytene bivalents. The use of combined silver staining and fluorescent in situ hybridization (FISH) with a Paracentrotus lividus (Echinodermata) 4.3 kilobase (kb) rDNA probe (prR14) consisting of sequences from the 3′ end of 18S rDNA to the 3′ end of 26S rDNA, revealed that nucleolus organizer regions (NORs) are situated terminally on one arm of a small metacentric pair. Chromosomes of Perna viridis were characterized by karyotype analysis, C-banding and nucleolus organizer regions (NORs). The diploid chromosome number was confirmed as 30 and the karyotype is composed of ten metacentric and five submetacentric chromosome pairs. Constitutive heterochromatin blocks were found on all chromosomal pairs except chromosomal pair 15, which showed uniform staining throughout the entire chromosomes. Silver staining revealed nucleolus organizer regions on telomeric region of four chromosomal pairs, viz. 1, 3, 7 and 11. This is the first comprehensive study undertaken on chromosomes of Perna viridis.  相似文献

Chromosome preparations from gill tissue of the Australian flat oyster Ostrea angasi Sowerby were studied with conventional Giemsa staining, C-banding and silver staining techniques. A diploid complement of 2n = 20 was observed, consisting of five metacentric, three submetacentric and two subtelocentric pairs. Constitutive heterochromatin was distributed as large centromeric blocks in Chromosome Pairs 3, 6, 8, 9 and 10. The nucleolus organizer regions (NORs) were located terminally on long arms of Chromosome Pairs 9 and 10. This allowed the identification of homologous chromosomes in submetacentric and subtelocentric pairs. Intraspecific variability in NOR pattern as revealed by differences in the number of silver-stained NORs (Ag-NORs) per cell was found to be very common. Comparison of the patterns of karyotype, C-band and Ag-NORs between species of the larviparous oysters for which data have been published demonstrate that the chromosomal structure of the endemic Australian and New Zealand species O. angasi shows little similarity to the Southern Hemisphere oysters O. (Eostrea)puelchana Orbigny and Tiostrea chilensis (Philippi) and the Indo-West Pacific oyster O. denselamellosa, but very high resemblance to the European species O. edulis. A karyotype analysis by several staining techniques was carried out on the great sturgeon, Huso huso (Linnaeus, 1758). The karyotype (2n = 118 ± 2) was composed of 42 pairs of meta-/submetacentric chromosomes and 17 pairs of acrocentrics/microchromosomes. Constitutive heterochromatin was mainly located at the centromeric regions of the acrocentric chromosomes. The biarmed chromosomes showed weak C-bands. Fluorescent staining with GC-specific chromomycin A3 showed clearly recognizable fluorescent regions, whereas a more uniform base composition was revealed by the AT-specific 4,6-diamidino-2 phenylindole. After Ag-staining, nucleolar organizer regions could be observed on the short arms of two medium-sized submetacentrics and on two acrocentrics. Digoxigenated 28S and 5S rDNA probes, prepared from Acipenser naccarii DNA and hybridized to metaphase chromosomes, showed signals on six and two chromosomes, respectively. The telomeric sequence (TTAGGG) n detected by FISH was located at both ends of each chromosome. Results are discussed in relation to karyotype organization and evolution in sturgeons. The karyotypes of the bivalves Ostrea edulis (2 n=20), Crassostrea gigas (2 n=20), Mytilus galloprovincialis (2 n=28) and M. edulis (2 n=28) were obtained from mitotic metaphases of branchial tissue by means of a cellular suspension technique. The occurrence of metacentric, submetacentric and telocentric chromosomes in O. edulis as well as differences in size between Pair 1 and Pair 10 emphasize the cytological interest of this species compared to C. gigas which possesses only metacentric and submetacentric chromosomes. The variation in Pair 2, which is telocentric in M. galloprovincialis and metacencentric in M. edulis, has been observed for the first time and may support taxological separation of these two species. The karyotypes of the mesogastropods Rissoa ventricosa (2 n=32) and Littorina neritoides (2 n=34) were obtained from mitotic metaphases of gonadal tissue by the same technique. Sexual chromosomes were identified in the karyotypes of R. ventricosa. At the beginning of spermatogenesis, the gonad of L. neritoides shows both haploïd and polyploïd meïotic metaphases which correspond to typical (germinal cells) and atypical (nurse cells) forms, respectively.  相似文献

This paper reports the results of cytogenetic analyses carried out on Porichthys plectrodon using conventional Giemsa staining, C-banding and silver staining techniques. A diploid chromosome count of 2n=44 was observed, consisting of 8 metacentric, 10 submetacentric, 6 subtelocentric and 20 acrocentric chromosomes. Differences in length made it possible to identify homologous chromosomes within the metacentric group. Constitutive heterochromatin was distributed as large pericentromeric blocks in pairs 1 and 2, while the rest of the chromosomes were marked in centromeric regions, some more conspicuously than others. A karyological study ofTrachurus trachurus andT. mediterraneus (Perciformes: Carangidae) was conducted by standard, fluorochrome staining (CMA3, mithramycin, quinacrine mustard, DAPI), C-, Ag-NOR, and Alu-I banding methods. The karyotypes of both species consisted of 2n = 48 chromosomes, but of different FN:T. trachurus possessed a chromosome complement of 2 metacentric and 46 acrocentric elements, fundamental number (FN) = 50 andT. mediterraneus, a chromosome complement of 4 metacentric, 4 submetacentric, 14 subtelocentric and 26 acrocentric chromosomes, FN = 70. In neither of the two taxa investigated were heteromorphic sex chromosomes observed. The nucleolar organizer region was interstitially located on the long arm of the Ist pair of chromosomes in both species, intermediate inT. mediterraneus and subterminal inT. trachurus. Constitutive heterochromatin was found in nearly all centromeric and telomeric regions inT. trachurus; inT. mediterraneus it formed less intense telomeric and centromeric bands and thin interstitial bands on eight chromosome pairs. In addition, the C-positive material reacted differently to the digestion with endonuclease Alu-l in the two species. The results are discussed and compared with karyological data known for other species of Carangidae.  相似文献

A karyotypic analysis was carried out in turbot (Scophthalmus maximus) using conventional banding techniques, fluorochromes and restriction endonucleasebanding. The standard karyotype of turbot is 2n=44 chromosomes, with 48 chromosome arms. Nucleolar organizer regions, which were localized in the short arm of Chromosome Pair No. 3, showed C-and chromomycin A3-positive staining. C-bands were mainly located at the centromeres, but were also present in some interstitial and telomeric locations. No differences were revealed among constitutive heterochromatin bands after treatment of fixed chromosomes with fluorochrome staining and restriction endonucleases. Digestion with DdeI restriction enzyme produced a pattern of interstitial banding which suggested some degree of differentiation along the chromosome arms in turbot. Neither variation in chromosome number of arm number nor polymorphism in nucleolar organizer regions was revealed in the turbot analyzed.  相似文献

A cytogenetic analysis by several staining techniques was carried out on specimens of Seriola dumerili from the Tyrrhenian coast of Sicily. The karyotype was found to be composed of 48 chromosomes in all the specimens examined. All chromosomes are subtelo- and acrocentrics, except for one pair of small-sized submetacentric chromosomes. After Ag-staining, nucleolus organizer regions (NORs) can be observed on the short arms of a medium-sized subtelocentric chromosome pair, and after chromomycin A3-staining a bright fluorescent signal is evident at the same sites. The 18S rDNA mapping by fluorescence in situ hybridization confirms this unique NOR location. The constitutive heterochromatin is mainly restricted to centromeres and the fluorochrome-staining by chromomycin A3 and 4′,6-diamidino-2-phenylindole revealed a uniform base composition along the chromosomal arms; therefore stock/population-specific marker chromosomes cannot be identified. Three different karyomorphs with 2n=46, 2n=44 and 2n=42 for Capros aper (L., 1758) (Zeiformes) collected from the Gulf of Lion, near Banyuls-sur-Mer, France, in July 1990 were determined. Karyomorphs were characterized by the same arm number [Fundamental number (FN)=50], suggesting that chromosome variations are due to Robertsonian fusions. In somatic metaphase spreads stained with silver nitrate, nucleolus organizer regions (NORs) consistently occupied a terminal position on the short arms of two small submetacentric chromosomes. Ca. 30% of silver-stained metaphases in each specimen showed NOR chromosomes associated in pairs by their nucleolus organizer regions.  相似文献

Low dispersal and sexual selection are characteristic of the coastal polychaeteNereis acuminata Ehlers 1868 [also known asNereis arenaceodentata Moore 1903 andNereis (Neanthes) caudata Delle Chiaje 1841]. We assessed levels of premating isolation between populations of this polychaete. Four North American populations were used, two from the Atlantic and two from the Pacific. Worms from all sites (1) were collected in 1987 and 1988 from the same habitat type, (2) were morphologically similar and keyed out asN. acuminata, and (3) reproduced monogamously and exhibited male parental care, an extremely rare reproductive mode in marine invertebrates. There was no evidence from 10-min or 36-h trials of premating isolation between the two Pacific populations. Incomplete premating isolation was found between the two Atlantic populations. High aggression and non-pairing occurred in some 10-min trials between males and females. However, in 36-h trials males and females from the two Atlantic populations always paired to mate. Complete premating isolation was found between Atlantic and Pacific populations. During 10-min trials, males and females from different oceans often attacked and then avoided each other, and they never paired to mate. Nor did they pair to mate in longer, 36-h trials. One Pacific and one Atlantic population were compared for tolerance to cold temperature. Pacific individuals were less able to tolerate cold water than Atlantic individuals. Two Atlantic populations studied had karyotypes with 11 pairs of small acrocentric chromosomes (2n=22), while the two Pacific populations had nine pairs of large metacentric or submetacentric chromosomes (2n=18). Such extreme dissimilarity in karyotype was not expected considering the similarity in morphology, habitat, and reproductive mode. Results suggest strongly that the Atlantic and Pacific populations have been allopatric for a long time, and are different species.  相似文献

 A cytogenetic analysis was carried out on specimens of Epinephelus marginatus (Lowe, 1834) from three localities in the Mediterranean Sea, to deepen knowledge of the chromosome complement of the species and identify any possible population-specific cytogenetic markers. All specimens had a 2n = 48 acrocentric karyotype with two Ag-, chromomycin A3- and C-positive NORs (nucleolar organizer regions) in the subcentromeric region of the smallest chromosome pair. The constitutive heterochromatin was distributed centromerically. Except for NORs, neither eu- nor heterochromatin show fluorescence after fluorochrome staining, i.e. there is no localized increase of AT- or GC-rich DNA. In all populations, the (TTAGGG) n telomeric sequences are restricted to the telomeres, and the 18S and the 5S rDNA clusters are located on different chromosome pairs. In specimens from Sardinia, additional signals after C-banding, Ag-staining and FISH (fluorescence in situ hybridization) with 18S rDNA can be observed in the telomeric region of one or two large-sized chromosomes, classified as No. 2. This suggests that additional and variable NORs could be detected in the species in addition to those on the genus-specific, NOR-bearing Chromosome Pair 24. Populations of Mercenaria mercenaria (L.) from South Carolina, USA, and M. campechiensis (Gmelin) from the Gulf of Mexico, Florida, USA, were sampled in 1987. The two species differed at all of seven enzyme loci tested, as well as in the thickness of shell ridges and nacre color. The difference in lunule shape was not great, although differences in relative shell width, shell weight, and lunule size make morphometric discrimination between the species possible. Shell ridges, nacre color, and multivariate morphometrics in a sample of clams collected from the Indian River Lagoon on the Atlantic coast of Florida in 1985 do not assort independently. Individuals with thick ridges, white nacre, and/or campechiensis-like morphometrics have significantly different allele frequencies at most enzyme loci from individuals with thin ridges, purple nacre, and/or mercenaria-like morphometrics. The deviations are in the direction predicted from the analysis of the allopatric populations of M. mercenaria and M. campechiensis. The study of the male and female karyotypes of the polychaete Dinophilus gyrociliatus was performed with the aim of ascertaining the diploid number of chromosomes and confirming the existence of karyological differences between the two sexes. Our results evidenced that the female karyotype of D. gyrociliatus consists of 2n=24 chromosomes, while the male karyotype has 2n=23 chromosomes. X chromosomes are subtelocentric and much bigger than the autosomes. The size of the D. gyrociliatus chromosomes, which in the female embryos vary in length from little more than 2.8 wm to less than 0.7 wm, are relatively small by comparison with those of other polychaetes. The existence of maternal and environmental factors able to influence the sex ratio of D. gyrociliatus and chromosomal differences between the sexes confirms that: (1) sex determination is chromosomal and syngamic of the XX-X0 type and (2) control of the sex ratio is progamic and depends on genetic and environmental factors.  相似文献

Shell growth rate is an important component of fitness in bivalve molluscs. Using the parameter computed from the von Bertalanffy growth equation, we quantitatively compared rates of annual shell grwoth among the hard clams Mercenaria mercenaria, M. campechiensis, and their hybrids sampled from a variety of habitats in the Indian River lagoon, Florida, USA, a zone of species overlap and natural hybridization. Our results indicate that the classical paradigm describing hard clam growth, in which growth rate is fastest in M. campechiensis, intermediate in hybrids, and slowest in M. mercenaria is not supported in the Indian River lagoon. Instead, M. campechiensis has a growth advantage in deep-water habitats in the northern section of our study area. In the central and southern sections of our study area, hybrids have a growth advantage over M. mercenaria in shallow-water habitats, but M. mercenaria has a growth advantage over hybrids in deep-water habitats. In all other sampled habitats, either growth rate among genotype classes is equal, or M. mercenaria has a growth advantage. This complex relationship between genotype and habitat-specific growth provides a mechanism for selection to act on hard clams in the Indian River.  相似文献

The quahog Mercenaria mercenaria has been introduced repeatedly to the Pacific coast of North America, but only one population is known to have become established. In the 1970s, the population of M. mercenaria at Colorado Lagoon, in Los Angeles County, California (33o46′16″N, 118o08′05″W), was estimated at more than 300,000 individuals. To determine the current status of this non-indigenous species (NIS), in 2009, we sampled 57 intertidal and 20 shallow subtidal plots, identifying and quantifying collected bivalves. No quahogs were found among the 2,490 living bivalves in our plots, though two were found intertidally outside of our plots. The M. mercenaria population has thus collapsed since 1980, but the native community has not recovered. Six of the fourteen living bivalve species we encountered were NIS; three are new records for the location, including the clam Venerupis philippinarum, which made up 87.6% of collected individuals. Though M. mercenaria is likely on its way to extinction on the US Pacific coast, the bivalve assemblage at this location remains heavily dominated by NIS.  相似文献

Rates of calcium carbonate removal from shell pieces of Argopecten irradians (Say) and Mercenaria mercenaria (L.) were dependent on the type of etching fluid used and not shell origin. Etching was uniform over the entire shell surface, but surface morphology differed with etching fluids. Peak radioactivity was in early eluant fractions of shells etched immediately after radioactive labeling, and in later fractions when individuals were placed in isotope-free seawater after labeling. The etching technique can measure growth during the labeling period and subsequently by estimating the amount of calcium in fractions prior to the radioactive peak. Geometry of shell layers influenced the pattern of radioactivity seen in fractions. Peak location varied inside and outside the pallial line of individual M. mercenaria. A significant portion of the inorganic carbon used in shell formation by M. mercenaria was derived from metabolic CO2.  相似文献

This study continues a comparative cytogenetic analysis of the fish family Mugilidae, reporting the karyotype characterization of the leaping mullet, Liza saliens, by C-banding, Ag- and fluorochrome-staining, and completing the fluorescence in situ hybridization (FISH)-mapping of the 18S and 5S rRNA genes (rDNA) to the chromosomes of the six Mediterranean mullets, namely L. saliens, L. ramada, L. aurata, Mugil cephalus, Chelon labrosus and Oedalechilus labeo. In all species, except M. cephalus, the 5S rDNA sites were localized on a medium-sized acrocentric chromosome pair, which was considered homeologous in all of them. In L. saliens, an additional 5S rDNA site was detected in a location close to the one shown by major ribosomal genes in M. cephalus, i.e. the subtelomeric region of chromosome pair 1. The 5S rDNA site in M. cephalus is located on the smallest chromosome pair of the complement, which, on the other hand, though on a different position, bears 18S rDNA in all the species of Liza and Chelon examined. The heterochromatin composition and the major and minor ribosomal gene locations suggest that the karyotype of L. saliens (subgenus Protomugil) can be considered intermediate between the karyotype of the more primitive M. cephalus and those of the other Liza (subgenus Liza) species and of the representatives of the more derived genera Chelon and Oedalechilus.  相似文献

The metabolism of 14C-labelled lactate was investigated in the horseshoe crab Limulus polyphemus and the stone crab Menippe mercenaria. When a bolus of (14C-U)-D-lactate was injected into L. polyphemus, there was substantial release pf 14CO2 into the medium. In the case of M. mercenaria, 14CO2 release was also observed after injection of (14C-U)-L-lactate into experimental individuals. Analysis of the distribution of radioactivity in whole body extracts of both species revealed easily detectable amounts of radioactivity in the glycogen fraction, although the bulk of the radioactivity was in the cation, anion/neutral and CO2 fractions. To investigate the metabolism of lactate further, 14C-labelled lactate was injected into large individuals of L. polyphemus and M. mercenaria, and the distribution of radioactivity was determined in the hemolymph, muscle and hepatopancreas. Utilization of (14C-U)-D-lactate by L. polyphemus resulted in the accumulation of significant amounts of labelled glucose in all three body compartments as well as the production of labelled glycogen in the telson levator muscle and hepatopancreas. Utilization of (14C-U)-L-lactate resulted in a similar pattern of glucose and glycogen labelling in the hemolymph, cheliped muscle and hepatopancreas of M. mercenaria. These studies demonstrate that both L. polyphemus and M. mercenaria have the capacity for glyco- and gluconeogenesis using lactate as the substrate.  相似文献

Two species of bivalve molluscs, Mercenaria mercenaria Linné and Crassostrea virginica Gmelin, were maintained in an identical laboratory environment and fed the same diet for 1 month prior to measuring the quantity of ammonia which they excreted per gram dry weight. Data derived from juveniles and adults of these species were fitted to a log-log equation. M. mercenaria excreted more ammonia per gram body weight than C. virginica, and considerably more scatter is evident in the fit of the data from M. mercenaria to the log-log relationship. Experiments showed that neither temperature fluctuation during the experiments nor decomposition of organic nitrogen in the test water account for the scatter of points.
