The effect of light quality on growth, photosynthesis and carbon metabolism in two species of marine algae,Cyclotella nana (Hustedt) andDunaliella tertiolecta (Butcher), was examined. Relative growth constants forC. nana were 0.37, 0.29 and 0.25 in blue, white and green light, respectively. Corresponding constants were 0.41, 0.31 and 0.29 forD. tertiolecta. Photosynthetic rates in both species were higher in blue light and lower in green light compared with white light of the same intensity. More than 60% of14C assimilated byC. nana orD. tertiolecta grown in blue or green light was incorporated into the ethanol-insoluble fraction, compared with 10 to 30% in this fraction in white light. The relative importance of the various components within this fraction was independent of light quality. Although less14C was assimilated into the ethanol-soluble fraction in blue or green light, there was a relative increase in some amino acids and organic acids in this fraction and a decrease in sugars and sugar phosphates relative to white light of the same intensity. These differences were independent of light intensity, photosynthetic rate and cell density in the cultures.  相似文献

Pocillopora damicornis (Linnaeus) and Montipora verrucosa (Lamarck) were collected from Hawaiian reefs. In two experiments (September 1979-January 1980: ca. 4 mo; August-October 1980; ca. 2 mo), these reef corals were grown under sunlight passed through filters producing light fields of similar quantum flux but different spectral composition. In vitro cultures of symbiotic zooxanthellae (Symbiodinium microadriaticum Freudenthal) from M. verrucosa were cultured under similar conditions for 15 d. Blue or white light promoted more coral skeletal growth than green or red light. In both coral species, blue light increased the total amount of chlorophyll a of the coral-zooxanthellae association. In the perforate species, M. verrucosa, the pigment concentration was elevated by an increase in the density of zooxanthellae, but the pigment concentrations per algal cell remained unchanged; in the non-perforate species, P. damicornis, it appears that pigment concentration was elevated by an increase in pigment per algal cell, and not by an increase in density of zooxanthellae. The sunloving reef-flat coral P. damicornis did not grow as rapidly as the shade-species M. verrucosa at the low quantum flux (about 10% sunlight) provided by the experimental treatments. The in vitro cultures of zooxanthellae from M. verrucosa exhibited growth rates in light of altered spectral quality that correlated with the responses of the host coral species: blue and white light supported significantly greater growth than green light, and red light resulted in the lowest growth rate.Contribution No. 678 of the Hawaii Institute of Marine Biology  相似文献

Pieces of the reef coral Montipora verrucosa (Lam.), collected from Kaneohe Bay, Oahu, Hawaii in 1982, were grown in four low-light treatments (11% sunlight): blue, green, red and the full spectrum of photosynthetically active radiation (PAR); and at high-intensity full PAR (90% sunlight). These acclimated corals were then tested for photosynthetic ability in blue, green, red, and white light. The photosynthetic parameters that were measured were; ligh-saturated photosynthetic rate, the initial slope of the photosynthesis/irradiance curve, the light intensity where these two lines crossed, and dark respiration. While acclimation intensity had a pronounced effect, the results also showed that the color of the acclimation treatment influenced the photosynthetic responses of the corals. Porphyra perforata J. Ag. was collected from a rocky land-fill site near Kitsilano Beach, Vancouver, British Columbia, Canada and was grown for 4 d in media with one of the following forms of inorganic nitrogen: NO 3 - , NH 4 + and NO 3 - plus NH 4 + and for 10 d in nitrogen-free media. Internal nitrogen accumulation (nitrate, ammonium, amino acids and soluble protein), nitrate and ammonium uptake rates, and nitrate reductase activity were measured daily. Short initial periods (10 to 20 min) of rapid ammonium uptake were common in nitrogen-deficient plants. In the case of nitrate uptake, initial uptake rates were low, increasing after 10 to 20 min. Ammonium inhibited nitrate uptake for only the first 10 to 20 min and then nitrate uptake rates were independent of ammonium concentration. Nitrogen starvation for 8 d overcame this initial suppression of nitrate uptake by ammonium. Nitrogen starvation also resulted in a decrease in soluble internal nitrate content and a transient increase in nitrate reductase activity. Little or no decrease was observed in internal ammonium, total amino acids and soluble protein. The cultures grown on nitrate only, maintained high ammonium uptake rates also. The rate of nitrate reduction may have limited the supply of nitrogen available for further assimilation. Internal nitrate concentrations were inversely correlated with nitrate uptake rates. Except for ammonium-grown cultures, internal total amino acids and soluble protein showed no correlation with uptake rates. Both internal pool concentrations and enzyme activities are required to interpret changes in uptake rate during growth.  相似文献

Diel migration of Mesodinium rubrum, Eutreptiella sp., Scrippsiella trochoidea, Dinophysis acuminata and Ceratium furca throughout a 24 h cycle is described for a stable, well-stratified estuary (Ría de Vigo, NW Spain). Daily changes in light quantity and in spectral light ratios i.e. red:far-red, blue:red, green:red and blue:green have been analysed. The spectral light ratios changed at twilight and around noon at various depths. Some of the downward migrations were well predicted by Stokes' law, while other migrations were faster and deeper than calculated. The coincidence of these movements with abrupt changes in red:far-red, green:red and blue:green light ratios is discussed. Some species are able to migrate through the pycnocline, whereas others do not seem to be able to do so. Several species are present in maximum numbers at depth at night, while others display upward migration independent of light, suggesting the existence of endogenous rhythms. Upward migration at dusk began with dispersal of populations, with renewed aggregation at the sea surface coincident with an increase in the red:far-red ratio at 6 m and the green:red ratio at 6 and 10 m. Based on direct evidence for the control of flagellar mobility by light quality reported by other authors from laboratory studies, it is suggested that, together with other cues, spectral light ratios of different light qualities modulate vertical phytoplanktonic migration. Invertebrates containing endosymbiotic dinoflagellate algae (zooxanthellae) retain excretory nitrogen, and many are able to take up ammonium from the surrounding seawater. However, the site of assimilation and role of nitrogen recycling between symbiont and host remains unclear. In the present study, ammonium uptake by the symbiotic sea anemone Anemonia viridis (Forskål) was examined by following the pathway of assimilation using 15N-enriched ammonium. Since zooxanthellae became enriched with 15N from ammonium at up to 17 times the rate of the host, they appear to be the primary site of assimilation. In the light, the rate of zooxanthellae enrichment at 20?M was twice that at 10?M, whereas the rate of host enrichment was not significantly affected by ammonium concentration. When anemones were incubated with [15N]ammonium in the dark, after 12?h without light the rate of enrichment was lowered in both zooxanthellae and host. However, while the enrichment of the host was significantly reduced when the light level was lowered from 300 to 150?μmol photons m?2?s?1, zooxanthellae enrichment was unchanged. Low molecular weight material from the zooxanthellae became enriched at 20 times the rate of that from the host, and enrichment was detected in the amino acids glutamate, glutamine, aspartate, alanine, glycine, phenylalanine, threonine, valine, tyrosine, and leucine from zooxanthellae. In the zooxanthellae, amino acids accounted for 65% of the total enrichment of low molecular weight material. Of the amino acids detected in zooxanthellae, over 90% of the enrichment was accounted for by glutamate, glutamine and aspartate. The enrichment of the amide group of glutamine was greater than that of the amine group of glutamate or glutamine, consistent with the glutamine synthetase/glutamine 2-oxoglutarate amidotransferase cycle as the mechanism of ammonium assimilation. To examine the flux of 15N from zooxanthellae to host, anemones were pulse-labelled with [15N]ammonium and then transferred to an unlabelled chase. Over a 2?h period there was no evidence for a flux of nitrogen from zooxanthellae to host. However, during the chase period, the enrichment of low molecular weight material declined and that of high molecular weight material increased in both zooxanthellae and host, indicating that protein was synthesized using 15N from ammonium in both components of the symbiosis. Again by using a pulse-chase system, it was found that glutamate was metabolised most rapidly by zooxanthellae, followed by (in order of decreasing rate of turnover) aspartate, alanine, glycine and valine (no data are available for glutamine). Unlike these amino acids, nitrogen was transferred to the essential amino acids phenylalanine and threonine, increasing their enrichment during the chase period. While recycled nitrogen is clearly important to this symbiosis, the mechanism by which it is cycled remains to be resolved.  相似文献

The possibility that disruptive sexual selection alone can cause sympatric speciation is currently a subject of much debate. The initial difficulty for new and rare ornament phenotypes to invade a population, and the stabilisation of the resulting polymorphism in trait and preference make this hypothesis problematic. Recent theoretical work indicates that the invasion is facilitated if males with the new phenotype have an initial advantage in male-male competition. We studied a pair of sympatric incipient species of cichlids from Lake Victoria, in which the red (Pundamilia nyererei) and blue males (P. pundamilia) vigorously defend territories. Other studies suggested that red phenotypes may have repeatedly invaded blue populations in independent episodes of speciation. We hypothesised that red coloration confers an advantage in male-male competition, assisting red phenotypes to invade. To test this hypothesis, we staged contests between red and blue males from a population where the phenotypes are interbreeding morphs or incipient species. We staged contests under both white and green light condition. Green light effectively masks the difference between red and blue coloration. Red males dominated blue males under white light, but their competitive advantage was significantly diminished under green light. Contests were shorter when colour differences were visible. Experience of blue males with red males did not affect the outcome of a contest. The advantage of red over blue in combats may assist the red phenotype to invade blue populations. The apparently stable co-existence of red and blue incipient species in many populations of Lake Victoria cichlids is discussed.Communicated by C. St. Mary  相似文献

Gracilaria verrucosa (Hudson) Papenfuss exposed to nutrient enriched media (0.1 mM PO4; 1.0 mM NH 4 + ) by pulse feeding 2 h every third day for a period of 5 wk at 20°C and 25–30 salinity showed significantly higher rates of photosynthesis regardless of photon flux density correlated with increased pigment levels. Algae in nonenriched media showed significantly higher levels of soluble carbohydrates and decreased levels of phycoerythrin and chlorophyll a. Photosynthetic and respiratory responses to temperature 15°, 25°, 30°C and salinity (15, 25, 30 S) combinations indicate broad tolerances by both nutrient enriched and non-nutrient enriched algae. Photosynthetic and respiratory rates were highest at the high temperatures. Pulse-fed algae had significantly higher photosynthetic rates than non-nutrient enriched plants at all temperature and salinity combinations. Non-nutrient enriched algae had significantly higher respiratory rates than nutrient enriched algae at only 30°C and 15. The respiratory rates of both nutrient enriched and non-nutrient algae decreased under combinations of higher temperatures and salinities. G. verrucosa, grown without nutrients, has lower tolerances to environmental stresses.  相似文献

The induction of reproductive activity by blue light in female gametophytes of the brown alga Lamnaria saccharina has been investigated and related to other effects of red and blue light on these plants. Although germination of zoo-spores is slightly delayed in red light, female gametophytes grow at similar rates in blue and red light of the same quantum irradiance up to an age of 8 to 10 days, when plants in blue light begin to form eggs. The percentage of plants forming eggs is proportional to the total quanta of blue wavelengths received up to a saturating value of about 400 μE·cm-2; a 50% response is induced by 200 μE·cm-2. The action spectrum for the induction of fertility has a main peak at 430 to 450 nm, with two subsidiary peaks in the blue and one in the near-ultraviolet light, but the action spectrum for photosynthesis is quite different, with peaks in the red, green and blue regions. These results indicate that egg formation in female gametophytes occurs as a specific morphogenetic response to blue light, which is independent of photosynthesis and growth, and trial experiments with male gametophytes of L. saccharina and gametophytes of other species of Laminariales indicate that these plants react similarly.  相似文献

The red algaGelidium sesquipedale (Clem.) Born. et Thur. has been cultured in chemostats to assess the effects of light quality and photon-fluence rate (PFR) on growth, photosynthesis and biochemical composition. Plants under blue and red light (BL and RL) showed higher growth rates than under white light (WL) of the same PFR (40 mol m–2 s–1). The light-saturated rate of photosynthesis was higher for algae grown under BL and RL than for algae grown under WL. When algae were transferred to WL of moderate PFR (100 mol m–2 s–1), the light-saturated rate of photosynthesis decreased, being higher in previously RL-grown algae than in previously BL- and WL-grown algae. The initial slope of photosynthesis-irradiance (PI) curves () was affected by PFR but not by light quality. Pigment content was little affected by light quality. Light-quality treatments also affected the biochemical composition of the alga; previous exposure to various light treatments activate or repress several metabolical pathways that are fully expressed in the subsequent phase of WL of moderate PFR. Thus, phycobiliproteins and soluble proteins increased for previously BL- and RL-grown algae, whereas insoluble carbohydrate concentration was reduced, indicating a change of the C-partitioning between carbon compounds and organic nitrogen compounds. Inorganic nitrogen metabolism was also affected by light: under WL of moderate PFR, NO3 was totally depleted from sea water, and maximal values of NO3 uptake were recorded. In addition, neither NO2 nor NH4 + was released. However, when algae were transferred to a low PFR, there was a drastic reduction of NO3 uptake under WL, which only partially recovered over time. It was accompanied by the release of NO2 , but not NH4 +, to the culture medium. Under BL and RL, however, there was a transient enhancement of NO3 uptake that was followed by a net release of NO2 and NH4 . Growth rates were not correlated with PFR. This could be due to the the dynamics of internal carbon mobilization and accumulation in the algae. When algae were exposed to a moderate PFR of WL, carbon requirements for growth were satisfied by photosynthesis. Thus, there was a net accumulation of carbon in the tissue. In contrast, when algae were exposed to low PFRs of either WL, BL or RL, observed growth rates could not be maintained by photosynthesis and carbon was mobilized.  相似文献

The marine pseudomonad bacterium PL1 contains an intracellular pool of free amino acids which consist mainly of glutamate with small amounts of glutamine and aspartate when grown in a nutrient medium containing 0.2 M NaCl. When the NaCl concentration of the growth medium is increased to 0.8 M, proline becomes a major component of the intracellular pool together with glutamate—at this molarity and under suitable nutrient conditions these amino acids comprise 20% of total bacterial amino acid nitrogen. When grown in a nutrient growth medium containing a constant level of NaCl, the intracellular pool size can vary by a factor of 4 depending on the concentration of carbon and nitrogen in the medium. Experiments show that the amino acid pool can act as a nitrogen reserve but has little function as a carbon reserve. At high NaCl concentrations there is a marked dependence for growth on the presence of sufficient potassium in the medium. However, no correlation between K+ and glutamate concentration in either nitrogen or K+-limited cultures has been found. None of the enzymes associated with glutamate biosynthesis was influenced by NaCl levels between 0.2 and 0.5 M. Neither Na+ or K+ stimulated the activity of these enzymes when tested in vitro.  相似文献

Growth and photosynthesis in the blade of Saccharina sporophytes are strongly stimulated by blue light. However, little is known about the effect of blue light on the early development and longitudinal photosynthesis profile of Saccharina sporophytes. In this study, S. japonica sporelings were cultured under blue or red light for 8 weeks. Blue light affected longitudinal, tangential cell divisions and tissue differentiation early in sporophyte development. The number of latitudinal cells in the blade under blue light was over fivefold that under red light. In addition, the number of cell layers was higher in the growing point than in the blade under blue light, whereas sporelings grown under red light contained only a single cell layer. Under blue light, the photosynthetic capacities of the growing region, blade and stipe were similar, and the maximum relative electron transport rate was even lower in the growing point than in the blade. The longitudinal photosynthesis profile suggested that blue light stimulated the enzymes participating in light-independent carbon fixation in the growing point and accordingly was less dependent on high light irradiances. Collectively, the results indicated that blue light promotes the early development of S. japonica sporophytes, which was attributed to both photomorphogenetic responses and photosynthetic reactions.  相似文献

不同肥力红壤有机无机复合状况及氮素形态分布的研究结果表明,土壤肥力水平提高,有机无机复合量增大.复合量与土壤有机质、全氮、碱解氮、速效磷和CEC呈显著或极显著正相关,与速效钾、缓效钾的相关性不显著;复合度与上述肥力性状的相关性均不显著.因此,可以用复合量来评价肥力状况.红壤酸解性氮占全氮77%-86%;在酸解性氮中,各组分氮所占比例为氨基酸态氮>酸解氨态氮>酸解未鉴别态氮>氨基糖态氮.氨基酸态氮占酸解性氮的比率为高肥力红壤大于低肥力红壤,且与土壤有机质、全氮、碱解氮、速效磷里显著正相关,也可作为衡量土壤肥力状况的指标之一.  相似文献

Nutrient enrichment of seagrass beds in a rhode island coastal lagoon   总被引:7,自引:0,他引:7  
Seagrass and algal beds showed a variety of reponses when the water column was treated with low level additions of ammonium, nitrate and phosphate. The nutrients were added separately to 3 uniform seagrass beds of a temperature coastal lagoon during 1979 and 1980. (1) Ammonium caused the production of dense mats of free-floating green algae Enteromorpha plumosa and Ulva lactuca. It also stimulated growth in both the leaf and root-rhizome fractions of Zostera marina. This growth response in Z. marina was greater in the area where current reached 12 cm · s-1 than in the area with little or no current. The concentration of nitrogen in the tissue did not change. In contrast, where current was lacking, Z. marina growth increase with ammonium was small, but the concentration of nitrogen in the tissue doubled over that in control plots. The growth of Ruppia maritima was inversely related to the growth of green algae in the same plots. The red alga Gracilaria tikvahiae did not grow better in ammonium, but its tissue reddened. (2) Nitrate additions enhanced the growth of the green seaweeds Enteromorpha spp. and U. lactuca, but not Z. marina or R. maritima. G. tikvahiae, when fertilized in isolation from other plants, showed a marginal response to this nutrient, and the tissue always reddened. (3) Phosphate enhanced growth in Z. marina and R. maritima exposed to moderate current. G. tikvahiae growing alone showed a small growth response to phosphate. The phosphate made no difference in the growth of the green seaweeds. (4) None of the nutrient supplements noticeably altered the species composition of either epiphytic or planktonic algae associated with the beds, although we did detect small increases in their numbers. The rapid and dense growth of green algae in nitrogen-enriched water probably limited growth of adjacent seagrasses and red algae. Because these seaweeds did not use the phosphate, it became available to other plant components. The overall floral response to nutrient addition in seagrass communities depends, therefore, upon the particular nutrient supplied, the ability of alternate species in the area to compete for that nutrient and the velocity of current in the specific area.  相似文献

At three stations in Bahamas waters, in 1989, 15 to 30% of all the dinoflagellates >20µm diameter observed in near-surface waters fluoresced green under blue excitation light, 55 to 66% fluoresced red, and the remainder did not fluoresce at all. The abundance of these green-fluorescing dinoflagellates ranged from ca 5 to 10 cells l–1 at the study sites. Under UV excitation, however, the dinoflagellates had a blue to blue-green appearance. Almost all the blue-green fluorescing dinoflagellates appeared to be heterotrophic, except for one species,Phalacroma rapa Stein, which also contained red-fluorescing (under blue light) chlorophylla. The emission spectra from all species examined were of three basic types. Type 1 typically had two fluorescence emission peaks (ca 440 and ca 510 nm). Type 2 spectra possessed one sharp peak at 495 nm. Spectra belonging to Type 3 had a broad peak around 470 to 480 nm. The green fluorescence thus is likely caused by different substances in individual species. The attempt to reconstitute observed spectra with nicotinamide adenine dinucleotide (NADH) and riboflavin 5-phosphate (FMN) solutions was unsuccessful.  相似文献

The nutritional history of corals is known to affect metabolic processes such as inorganic nutrient uptake and photosynthesis, but little is known about how it affects assimilation efficiency of ingested prey items or the partitioning of prey nitrogen between the host and symbiont. The temperate scleractinian coral Oculina arbuscula and its tropical congener Oculina diffusa were acclimated to three nutritional regimes (fed twice weekly, starved, starved with an inorganic nutrient supplement), then fed Artemia nauplii labeled with the stable isotope tracer 15N. Fed corals of both species had the lowest assimilation efficiencies (36–51% for O. arbuscula, 38–57% for O. diffusa), but were not statistically different from the other nutritional regimes. Fed and starved corals also had similar NH4+ excretion rates. This is inconsistent with decreased nitrogen excretion and reduced amino acid catabolism predicted by both the nitrogen recycling and conservation paradigms. In coral host tissue, ~90% of the ingested 15N was in the TCA-insoluble (protein and nucleic acids) and ethanol-soluble (amino acids/low molecular weight compounds) within 4 h of feeding. The TCA-insoluble pool was also the dominant repository of the label in zooxanthellae of both species (40–53% in O. arbuscula, 50–60% in O. diffusa). However, nutritional history had no effect on the distribution of prey 15N within the biochemical pools of the host or the zooxanthellae for either species. This result is consistent with the nitrogen conservation hypothesis, as preferential carbon metabolism would minimize the effects of starvation on nitrogen-containing biochemical pools.Communicated by P.W. Inorganic nitrogen metabolism in blue light was studied for the green algaUlva rigida C. Agardh collected in the south of Spain (Punta Carnero, Algeciras) in the winter of 1987. NH4 + has been reported to inhibit NO3 - uptake; however,U. rigida showed a net NO3 - uptake even when the NH4 + concentration of the external medium was three or four times greater than the concentration of NO3 -. NO3 - uptake rates were similar in both darkness and in blue light of various photon fluence rates (PFR) ranging from 17 to 160 mol m-2 s-1. Since NO3 - uptake is an active mechanism involving the consumption of ATP, respiratory metabolism can provide enough ATP to maintain the energetic requirement of NO3 - transport even in darkness. In contrast, NO3 - reduction inU. rigida was highly dependent on the net photosynthetic rate. After 7 h in blue light, intracellular NO3 - concentrations ([NO3 -] i ) were higher in specimens exposed to intensities below the light compensation point (LCP) than in those incubated at a PFR above the LCP. When PFR is below the light compensation point, NO3 - reduction is low, probably because all the NADH produced by the cells is oxidized in the respiratory chain in order to produce ATP to maintain a steady NO3 - transport rate. The total nitrogen (TN) and carbon (TC) contents decreased from darkness to 33 mol m-2 s-1 in blue light. In this range, catabolic processes prevailed over anabolic ones. In contrast, increases in TN and TC contents were observed above the light compensation point. The C : N ratio increased with light intensity, reaching a stable value of 17 at 78 mol m-2 s-1 in blue light. Intracellular NO3 - concentration and NO3 - reduction appear to be directly controlled by light intensity. This external control of [NO3 -]i and the small capacity ofU. rigida to retain incorporated NO3 -, NO2 - and NH4 + ions may explain its nitrophilic character.  相似文献

 The effects of cadmium exposure and dietary status on cadmium accumulation, fatty acid (FA) content and profiles were investigated in two colour forms of the shore crab Carcinus maenas. Groups of shore crabs were either starved or fed with blue mussels, Mytilus edulis, during a 40 d exposure period to 2 or 6 μM Cd2+ (as CdCl2). Starved green individuals accumulated more cadmium in haemolymph and hepatopancreas than did red crabs and green crabs fed during the experiments. In the red colour form, no difference in cadmium accumulation was observed between starved and fed individuals. In both colour forms, hepatopancreas contained more FA than gills and muscle. The FAs often present in the largest amounts in the tissues were 16:0, 16:1ω7, 18:1ω7, 18:1ω9, 20:4ω6, 20:5ω3 and 22:6ω3. However, saturated (SAFA) and mono-unsaturated fatty acids (MUFAs) were dominant in hepatopancreas, whereas poly-unsaturated fatty acids (PUFAs) were dominant in gills and muscles. At the beginning of the experiment, the total FA content in the hepatopancreas was 111.6 mg g−1 (dry weight) for red crabs and 78.4 mg g−1 for green shore crabs. During the experiment, however, the FA content decreased in red crabs. This decrease was more pronounced for starved individuals than for fed individuals. Also, the decrease in FA content was more pronounced in crabs exposed to 6 μM cadmium compared to crabs exposed to 2 μM or crabs not exposed to cadmium. No change in FA content was observed in green shore crabs, irrespective of diet and cadmium exposure. For both colour forms, no change in FA content was observed for gills and muscle. In red crabs, a decrease was observed for all FAs in the hepatopancreas. This decrease, however, was more pronounced for SAFAs and MUFAs than for PUFAs, indicating that the metabolism of FAs during starvation and cadmium exposure is selective. The experiments indicate that green colour forms of shore crabs are more tolerant of natural stress such as starvation and anthropogenic stress, e.g. cadmium exposure, than are red colour forms of shore crabs. An adult giant kelp plant (Macrocystis pyrifera), moved from an inshore kelp forest to an offshore, low-nitrogen environment near Santa Catalina Island, California (USA), maintained growth for 2 wk on internal nitrogen reserves. Frond elongation rates decreased significantly during the third week, and plant growth rate (wet wt) dropped from an initial inshore rate of 3.6 to 0.9% d-1. During this 3 wk period, nitrogen contents and free amino acid concentrations decreased, while mannitol and dry contents increased in frond tissues. After depletion of internal nitrogen reserves, the nitrogen content of lamina and stipe tissues averaged 1.1 and 0.7% dry wt, respectively. The experimental plant was exposed to higher ambient nitrogen concentrations during the last 2 wk. Rates of frond elongation and plant growth increased, but nitrogen content and amino acids in frond tissues remained low. Of the total nitrogen contained in frond tissue of the plant before transplantation, 58% was used to support growth in the absence of significant external nitrogen supply. Amino acids constituted a small proportion of these internal nitrogen reserves. Net movement of nitrogen occurred within large fronds, but not between different frond size classes. The nitrogen content of holdfast tissue remained relatively constant at 2.4% dry wt and accounted for 18 to 29% of the total nitrogen. Holdfast nitrogen was not used to support growth of nitrogen-depleted fronds. In comparison to Laminaria longicruris, which is adapted to long seasonal periods of low nitrogen availability, M. pyrifera has small nitrogen-storage capacity. However, internal reserves of M. pyrifera appear adequate to make nitrogen starvation uncommon in southern California kelp forests.  相似文献

Platymonas subcordiformis (UTEX 171) was cultured axenically for 4 d in constant light in a nitrate-containing medium and harvested in the log-phase of cell division. Cells were resuspended in artificial sea water without nutrients and either kept in constant light or placed in constant darkness. High-performance liquid chromatography was used to measure the free amino acid pools of the cells and to determine rates of net entry of each of a mixture of 18 amino acids at daily intervals for 5 d. Free amino acid pools decreased both in light and darkness in the absence of a nutrient sypply. The influx of amino acids in cells maintained in the light increased selectively. Comparison of the rate of entry of 14C-labeled glycine and net disappearance of glycine from the medium indicated extrusion of non-volatile labeled carbon that did not interact with reagents specific for amine groups. Light was required for synthesis of additional transporter protein which was apparently responsible for increased influx in cells maintained in the light. This response was blocked in the presence of cycloheximide. Cells maintained in the dark for prolonged periods retained the capacity to respond to light by synthesis of new transporter protein. Analysis of incorporation of amino acids into macromolecules indicated that both the overall rate and the pattern of amino acid incorporation were modified in the light. Analysis of the kinetics of glycine entry at a series of temperatures indicated that the concentration of glycine at which entry is half the maximum rate is approximately 2.7 M at the cell surface.  相似文献
