Scots pine seedlings were exposed to wet-deposited nickel (Ni) and removal of lichen cover in a dry heath Scots pine forest. Ni deposition affected the colonization of roots by indigenous ectomycorrhizal fungi in contrasting ways in intact and skimmed quadrats. Highest frequencies of tubercle morphotypes of ectomycorrhiza were found in quadrats exposed to 100 mg m−2 year−1 Ni in lichen covered treatment, while in skimmed quadrats these peaked after the treatment with 10 mg Ni m−2 year−1. Removal of the lichen layer increased the value of diversity index (H′) of ectomycorrhizal fungal community, probably due to the increase in the evenness of the morphotype distribution. Lichen removal seemed also to improve the condition of the short roots, as the frequencies of poor and senescent short roots were decreased by the removal.  相似文献

A three-year study was initiated in 1987 to evaluate the impact of O3, acidic precipitation, and soil Mg on ectomycorrhizal colonization of loblolly pine (Pinus taeda L.) seedlings. Thirty-six open-top chambers equipped with a rainfall exclusion/addition system were utilized to administer three levels of O3 (subambient, ambient, or twice ambient) and two precipitation acidity levels (pH 3.8 or 5.2) to seedlings growing in 24-liter plastic pots containing soil having either 35 or 15 mg kg(-1) of exchangeable Mg. Seedlings exposed to the twice ambient O3 treatment exhibited smaller percentages of total ectomycorrhizal short roots at the end of each year of the study, but trends were statistically significant in 1989 only. Changes in number of specific ectomycorrhizal morphotypes in response to O3 were not consistent from year to year. Acidic precipitation treatments had no effect on number or percent of mycorrhizal short roots, and responses of two morphotypes to soil Mg treatments were probably due to differences in the soil environment rather than a result of changes in aboveground processes. Temporal shifts in morphotype frequencies were observed for seedlings in all treatments and indicate that mycorrhizal succession occurred during the study period.  相似文献

Studies were done on the effects of elevated soil concentrations of copper (Cu) and (Ni) on foliar carbohydrates and phenolics in Scots pine (Pinus sylvestris L.). Four year-old seedlings were planted in pots filled with metal-treated mineral forest soil in early June. The experimental design included all combinations of four levels of Cu (0, 25, 40 and 50 mg kg(-1) soil dw) and Ni (0, 5, 15 and 25 mg kg(-1) soil dw). Current year needles were sampled for soluble sugar, starch and phenolics at the end of September. Ni increased sucrose concentration in the needles, indicating disturbances in carbohydrate metabolism. Trees exposed to Ni had higher concentrations of condensed tannins compared with controls. In contrast, concentrations of several other phenolic compounds decreased when seedlings were exposed to high levels of Cu or to a combination of Ni and Cu. The results suggest that concentrations of phenolics in Scots pine needles vary in their responses to Ni and Cu in the forest soil.  相似文献

Six-month-old loblolly pine (Pinus taeda) were grown for 15 weeks in two native soils amended with 0, 30, 60, 120, 240 or 480 mg kg(-1) Pb as PbCl2. Ectomycorrhizae were quantified, by morphotype, as the total number of tips per centimeter, and as the number of tips for each morphotype and for Cenococcum geophilum. Total numbers of non-ectomycorrhizal short roots and necrotic tips were recorded. Total height and biomass exhibited a non-linear response to soil-applied lead. Growth generally was greatest in the controls and higher treatments, and least in the intermediate treatments. In both soils, Pb concentrations in roots increased linearly with increasing levels of soil-applied Pb. Neither foliage nor stems exhibited significant increases in Pb concentrations with increasing levels of Pb. Significant linear decreases in total numbers of ectomycorrhizal tips and significant linear increases in non-ectomycorrhizal short roots and necrotic tips occurred with increasing levels of Pb in the soil. The majority of individual morphotypes decreased with increasing Pb. However, the number of ectomycorrhizal tips formed by C. geophilium increased with increasing soil Pb levels after 15 weeks of treatment. These results indicate that short-term loblolly pine seedling growth is not inhibited by increasing Pb levels. Ectomycorrhizal formation decreased, and alterations in species composition occurred as a result of increasing concentrations of soil-applied Pb. These effects on ectomycorrhizae may cause long-term changes in nutrient and water balances, which could reduce tree vigor.  相似文献

Plant biomass. mycorrhizal status and root peroxidase activity were measured in ectomycorrhizal Scots pine (Pinus sylvestris L.) seedlings grown in urban polluted and native, non-polluted forest soils with added ammonium or potassium sulphates simulating N and S deposition of urban areas. Peroxidase activity in the fine roots of seedlings planted in polluted forest soils was higher than in those planted in non-polluted soils and correlated positively with the activities measured in an earlier study in the roots of mature Scots pines growing at the sites from where the soils were collected. Growth of seedlings and mycorrhizal status were not affected by the origin of soil. Exposing the seedlings to winter acclimation conditions for 6 weeks elevated peroxidase activity in the roots. The addition of ammonium or potassium sulphate to non-polluted soils did not induce elevated root peroxidase activity, although at the levels of 0.5 and 1.0 g of ammonium sulphate a slight increasing trend was observed. We suggest, that indirect biotic factors, i.e. changes in the community structure of soil fungi, early stages of recognition, and defence reactions of pine roots against saprophytic and pathogenic fungi may be participating in the elicitation of peroxidase (POD) activity, although the possible role of heavy metals cannot be excluded.  相似文献

Norway spruce seedlings colonized with Hebeloma crustuliniforme were grown in growth pouches. After formation of ectomycorrhizas, the seedlings were exposed to Al or the heavy metals Cd, Cu, Ni, or Zn at various concentrations for 5 weeks to estimate the detection limits of metals with X-ray microanalysis in the cryo-scanning electron microscope. When the lowest metal concentrations (1 mM Al(3+), 0.1 mM Cd(2+), 0.2 mM Cu(2+), 0.5 mM Ni(2+), 2 mM Zn(2+)) were applied, only Al and Zn were detected at low X-ray counts in the ectomycorrhizas. After application of 10-fold higher metal concentrations, distinct metal accumulation patterns were observed. Cd was detected predominantly in the Hartig net, Al and Ni in the Hartig net and in the cell walls of the cortex, and Zn in the Hartig net, the cortical cell walls and the fungal mantle. Cu was not detected at all. By combining X-ray microanalysis with absolute metal concentrations found in the roots, the estimated detection limits of X-ray microanalysis were: Al> or =0.86 mg g(-1), Cd> or =0.26 mg g(-1), Ni> or =1.30 mg g(-1), and Zn> or =0.54 mg g(-1), whereas Cu was not detectable even at root concentrations of 0.47 mg g(-1). Treatments with the highest metal concentrations showed high X-ray counts of metals in cells of the stele but reduced concentrations of the macronutrients K, Mg, and P in roots, indicating a possible disturbance of root and ectomycorrhizal function.  相似文献

The characteristics of UV-absorbing compounds, particularly soluble phenolics, were studied in needles of 63-day-old seed-grown Scots pine (Pinus sylvestris L.) seedlings of two provenances in a UV exclusion field experiment at Pallas-Ounastunturi National Park in Finnish Lapland (68 degrees N, 270 m a.s.l.). The experiment used the following plastic filters in exclosure treatments to manipulate the spectral balance of natural irradiance: (1) 'control' (a polyethene plastic filter); (2) 'UV-B exclusion' (a clear polyester filter); and (3) 'UV-B/UV-A exclusion' (a clear acryl plate). Polyethene transmitted 89% of the ambient levels of total UV (280-400 nm), polyester transmitted 75% of the total UV, but only 0.6% of the UV-B (280-315 nm) component, while acryl plate transmitted 0.2% of UV (280-360 nm). The research also included (4) 'Ambient' plants that were not subjected to any treatment exclosures. After the 58 day UV exclusion, significant (p<0.0001) differences due to treatments were determined for a kaempferol derivative, kaempferol 3-glucoside, and a quercetin derivative, the quantities of which ranged from 0.23 to 0.45, 0.42 to 1.34 and 0.39 to 0.75 micromol g FW(-1), respectively, depending on treatment and provenance. Overall, Scots pine seedlings grown at ambient UV radiation (PAS300, Caldwell's generalized Plant Action Spectrum (PAS) normalized at 300 nm, 72 mW m(-2)) or under a control had significantly (p<0.05) higher quantities of soluble phenolics than seedlings grown under UV-B or UV-B/UV-A exclusion treatments. There were no significant differences in the quantity of soluble phenolics between the two exclosure treatments or between the two Scots pine provenances. The sums of diacylated flavonol glucosides ranging from 3.75 to 4.55 micromol g FW(-1) depending on treatment and provenance, were already present at very low UV-levels under the UV-B/UV-A exclusion treatment. The present study indicated that soluble phenolics, particularly the diacylated flavonol glucosides, may provide an effective preformed protection for young Scots pine seedlings against UV-B and UV-A radiation.  相似文献

Current-year seedlings of beech, ash, Norway spruce and Scots pine were exposed during one growing season to different, but moderate, ozone (O(3)) scenarios representative for Switzerland (50, 85, 100% ambient, 50% ambient+30 nl l(-1)) in open-top chambers (OTCs) and to ambient O(3) concentrations in the field. Biomass significantly decreased with increasing O(3) dose in all species except for spruce. Losses of 25.5% (ash), 17.4% (beech), 9.9% (Scots pine) were found per 10 microl l(-1) h accumulated O(3) exposure over a threshold concentration of 40 nl l(-1) during daylight hours (AOT40). Ratios of root/shoot biomass (RSR) also significantly decreased with increasing AOT40 levels in beech and ash, but not in Norway spruce and Scots pine. The data show that the deciduous species beech and ash were more susceptible to O(3) with respect to RSR and biomass than the coniferous species Norway spruce and Scots pine.  相似文献

Experiments were conducted to investigate the effects of single and multiple metal contamination (Cd, Pb, Zn, Sb, Cu) on Scots pine seedlings colonised by ectomycorrhizal (ECM) fungi from natural soil inoculum. Seedlings were grown in either contaminated field soil from the site of a chemical accident, soils amended with five metals contaminating the site, or in soil from an uncontaminated control site. Although contaminated and metal-amended soil significantly inhibited root and shoot growth of the Scots pine seedlings, total root tip density was not affected. Of the five metals tested in amended soils, Cd was the most toxic to ECM Scots pine. Field-contaminated soil had a toxic effect on ECM fungi associated with Scots pine seedlings and caused shifts in ECM species composition on ECM seedlings. When compared to soils amended with only one metal, soils amended with a combination of all five metals tested had lower relative toxicity and less accumulation of Pb, Zn and Sb into seedlings. This would indicate that the toxicity of multiple metal contamination cannot be predicted from the individual toxicity of the metals investigated.  相似文献

Loblolly pine (Pinus taeda) seedlings from three full-sib families were exposed to 0, 50, 100 or 150 ppb ozone (O(3)) (5 h/d, 5 d/week for 6 or 12 weeks). Soil water potential was maintained near pot capacity (-0.03 MPa) or soil was allowed to dry to approximately -1.0 MPa and resaturated. Chlorotic mottling and flecking of needles due to O(3) injury were observed for seedlings from all pine families. Soil water deficit lessened the intensity of O(3) symptoms, possibly due to stomatal closure. Exposure to O(3) and soil water deficit each resulted in less seedling volume growth and dry weight, and changed the nonstructural carbohydrate content of seedlings compared with controls. Increasing O(3) concentrations resulted in a linear reduction in foliar starch content but did and affect hexose or sucrose content. Soil water deficit resulted in less starch and soluble sugar contents in above- and below-ground plant parts compared with controls. Soil water deficit did not affect numbers or percentages of roots that formed ectomycorrhizal tips. A linear dose-response relationship between O(3) and ectomycorrhizae was observed. The number of ectomycorrhizal tips/cm long root and the percentage of feeder roots that formed ectomycorrhizae were lower as O(3) concentration increased. Overall, each stress alone caused less seedling growth and carbohydrate content compared with controls, but only O(3) was responsible for suppression of ectomycorrhizae.  相似文献

This report presents an assessment of chemical variability in natural ecosystems of Wigierski National Park (NE Poland) derived from the calculation of geochemical baselines using a barbell cluster ANOVA design. This method enabled us to obtain statistically valid information with a minimum number of samples collected. Results of summary statistics are presented for elemental concentrations in the soil horizons-O (Ol + Ofh), -A and -B, 1- and 2-year old Pinus sylvestris L. (Scots pine) needles, pine bark and Hypogymnia physodes (L.) Nyl. (lichen) thalli, as well as pH and TOC. The scope of this study also encompassed S and C stable isotope determinations and SEM examinations on Scots pine needles. The variability for S and trace metals in soils and plant bioindicators is primarily governed by parent material lithology and to a lesser extent by anthropogenic factors. This fact enabled us to study concentrations that are close to regional background levels.  相似文献

The effects of long-term enhanced UV-B radiation on growth and secondary compounds of two conifer species were studied in an outdoor experiment. Scots pine (Pinus sylvestris) seedlings were exposed for two growing seasons and Norway spruce (Picea abies) seedlings for three growing seasons to supplemental UV-B radiation, corresponding to a 30% increase in ambient UV-B radiation. The experiment also included appropriate controls for ambient and increased UV-A radiation. Enhanced UV-B did not affect the growth of the conifer seedlings. In addition, neither the concentrations of terpenes and phenolics in the needles nor the concentrations of terpenes in the wood were affected. However, in the UV-A control treatment the concentrations of diterpenes in the wood of Scots pine decreased significantly compared to the ambient control. Apparently, a small increase in UV-B radiation has no significant effects on the secondary compounds and growth of Scots pine and Norway spruce seedlings.  相似文献

Empirical critical loads (CL) for N deposition were determined from changes in epiphytic lichen communities, elevated NO(3)(-) leaching in streamwater, and reduced fine root biomass in ponderosa pine (Pinus ponderosa Dougl. ex Laws.) at sites with varying N deposition. The CL for lichen community impacts of 3.1kg ha(-1) year(-1) is expected to protect all components of the forest ecosystem from the adverse effects of N deposition. Much of the western Sierra Nevada is above the lichen-based CL, showing significant changes in lichen indicator groups. The empirical N deposition threshold and that simulated by the DayCent model for enhanced NO(3)(-)leaching were 17kg N ha(-1) year(-1). DayCent estimated that elevated NO(3)(-) leaching in the San Bernardino Mountains began in the late 1950s. Critical values for litter C:N (34.1), ponderosa pine foliar N (1.1%), and N concentrations (1.0%) in the lichen Letharia vulpina ((L.) Hue) are indicative of CL exceedance.  相似文献

An open top chamber experiment was carried out in the summer of 2003 to examine the effect of nitrous acid (HONO) gas on the physiological status of Scots pine saplings (Pinus sylvestris). Four-year-old pine trees were exposed to two different levels of HONO gas (at ca. 2.5 ppb and 5.0 ppb) and a control (filtered air) from early evening to early morning (18:00-6:00), in duplicate open top chambers. Significant decreases in the ratios of chlorophylls a to b, an increase in the carbon to nitrogen (C/N) ratio, and a reduction of maximum yield of PS II (F(v)/F(m)) in pine needles were also observed after the 2 months' fumigation. Cation contents of pine needles were also decreased by the fumigation with HONO gas. The results could be explained by the harmful effects of OH radicals, generated from photolysis of HONO gas, and/or aqueous phase HONO (NO(2)(-)/HONO), on the photosynthetic capacity of pine needles.  相似文献

The effects of prolonged simulated acid rain on the biochemistry of Scots pine needles were studied in Finnish Lapland. Pine trees were exposed by spraying the foliage and soil with either clean water or simulated acid rain (SAR; both sulphuric and nitric acids) over the period 1985-1991. The concentrations of carbohydrates (starch, glucose, fructose, sucrose) in one-year-old pine needles were not affected by SAR-treatments. The SAR-treatments did not have significant effects on protein bound amino acids, which was true also for most of the free amino acids. However, the citrulline concentration was over three-fold greater in the foliage of pines exposed to SAR of pH 3 compared to irrigated controls. The concentrations of total phenolics, individual low molecular weight phenolics and soluble proanthocyanidins were not affected by the treatments, but insoluble proanthocyanidins had increased in acid-treated trees. Some of the studied biochemical compounds showed significant differences between two sub-areas (similar treatments) only 120 m apart.  相似文献

Soil metal pollution can trigger evolutionary adaptation in soil-borne organisms. An in vitro screening test showed cadmium adaptation in populations of Suillus luteus (L.: Fr.) Roussel, an ectomycorrhizal fungus of pine trees. Cadmium stress was subsequently investigated in Scots pine (Pinus sylvestris L.) seedlings inoculated with a Cd-tolerant S. luteus, isolated from a heavy metal contaminated site, and compared to plants inoculated with a Cd-sensitive isolate from a non-polluted area. A dose-response experiment with mycorrhizal pines showed better plant protection by a Cd-adapted fungus: more fungal biomass and a higher nutrient uptake at high Cd exposure. In addition, less Cd was transferred to aboveground plant parts. Because of the key role of the ectomycorrhizal symbiosis for tree fitness, the evolution of Cd tolerance in an ectomycorrhizal partner such as S. luteus can be of major importance for the establishment of pine forests on Cd-contaminated soils.  相似文献

Throughfall was collected in a Scots pine forest exposed to about 14 microg m(-3) of both SO2 and NO2, and in a control forest with 1 microg m(-3) SO2 and < 1 microg m(-3) NO2. Precipitation was collected in a nearby open field. Collection was performed on an event basis during the whole vegetation period. Exposure was made by an open-air release system during the vegetation period, except during rain and at night. Additional sulfate deposition in the exposed forest (compared to control forest) was nearly equal to dry deposition of sulfur dioxide, as estimated with a stomatal conductance model adapted for the particular forest. It is thus concluded that essentially all of the dry deposited sulfur dioxide is eventually extracted and appears in throughfall-including the fraction that has been deposited through stomata. Attempts to relate net throughfall deposition to dry deposition of sulfate in the control forest were inconclusive, since a minor (10%) uncertainty in the water balance had a major influence on calculated deposition velocity for particulate sulfate. Nitrate throughfall deposition is about half of the open field wet deposition, both for the exposed and control forest. Thus, a long-term exposure with about 14 microg m(-3) NO2 decreased nitrate throughfall deposition.  相似文献

A series of batch experiments was conducted to compare the ability of 11 microalgal species of the same cell density in removing nickel (Ni) and zinc (Zn) from synthetic wastewater. These included Chlorella vulgaris (commercially available), Chlorella sorokiniana and Scenedesmus quadricauda (isolates from polluted water of Wuhan, China), and eight different isolates from Hong Kong. The Wuhan isolate of Scenedesmus removed most Ni, probably due to its large biomass. Nickel concentration was reduced from an initial 30 to 0.9 mg/l after 5 min (97% Ni removal), and further declined to 0.4 mg/l after 90 min of treatment. In wastewater containing 30 mg/l Ni and 30 mg/l Zn, more than 98%, Ni and Zn were removed simultaneously at the end of 5 min treatment, indicating that the presence of Zn in wastewater did not affect Ni removal by this Scenedesmus isolate. The second most effective species for Ni removal was an isolate, tentatively identified as Chlorella miniata, Ni concentration was reduced to 10 mg/l after 90 min, and was only slightly interfered by the presence of Zn. In terms of metal removal per unit biomass or unit surface area of algal cells, C. miniata was the best species in removing Ni and Zn. At the other extreme, one Hong Kong isolate (Synechocystis sp.) did not remove any Ni and only achieved 40% Zn removal. Performance of the other isolates was comparable with the commercial C. vulgaris, less than 50% Ni was removed after 5 h of treatment and Ni removal was significantly reduced by the presence of Zn. All algae tested were found to be viable, showing these 11 species could tolerate a mixture of 30 mg/l Ni and 30 mg/l Zn in wastewater.  相似文献

Effects after long-term fumigation and simulated acidic rain (pH 4.0) treatment in open-top chambers (OTC) during 5 vegetation periods on ectomycorrhizal (EM) frequency, fine root structure and distribution of short roots were recorded for Norway spruce. Simultaneous application of ozone and SO(2) resulted in a significant decrease (P< or =0.01) in the proportion of EM root tips and an increase in the numbers of necrotic short roots. The observed differences were partly due to more than additive effects of ozone and SO(2). On the basis of EM and non-mycorrhizal (NM) short roots in the irrigated controls (OTC-V, pH 5.6, Ah-horizon: EM + NM/cm=0.05) the acid treatments resulted in drastic effects (P< or =0.05) on the branching density and the proportion of EM root tips and NM short roots. Relative numbers of short roots infected by indigenous EM fungi with trees in the fumigation treatments at pH 4.0 were similar: 1.5-1.9 EM tips/cm and differed noticeably from the control: 5.2 EM tips/cm. Again, increasing numbers of necrotic and NM short roots were noted. Parallel measurements by the ingrowth-core technique confirm the argument, that EM formation seems to be a reliable indicator of pollution-induced stress.  相似文献

Total monoterpene levels of the order of 100 μg m −3 were assessed in the summer night air of a typical young planted forest of Scots pine (Pinus silvestris) in Sweden. These concentrations are much higher than those reported in previous studies of ambient monoterpenes. Predominant species were α-pinene and 3-carene. Camphene, β-pinene, myrcene, β-phellandrene and limonene were other prominent components determined. The monoterpenes were shown to originate almost exclusively from Scots pine and the proportions between them to be characteristic of this conifer species.  相似文献
