Changes in biochemical composition in erythrocyte membrane, erythrocytic osmotic haemolysis, and nitrite and nitrate levels in plasma were analyzed in 12 human volunteers who were exposed regularly to prallethrin, a type I pyrethroid mosquito repellent. The results revealed a decrease in cholesterol (C) and phospholipid (P) moieties in erythrocyte membrane with no consequent change in C:P ratio. Further, a significant decrease in the content of phosphatidyl serine suggested that PS is a sensitive phospholipid species to the pyrethroid action. Significant decrease in membrane lipid peroxidation and enhanced levels of nitrite and nitrate in plasma and erythrocyte indicate that increased generation and availability of nitric oxide might have rendered tolerance to erythrocyte membrane by protecting the cells from haemolysis. Increased NO(2) and NO(3) may be due to increased activity of nitric oxide synthase (NOS) and/or expression of isoforms of NOS. A possible involvement of free radical scavenging and antioxidant effects of nitric oxide might have contributed to the observed decrease in lipid peroxidation in the present study.  相似文献

Extensive use of pharmaceuticals in human and veterinary medicine and aquaculture practices pose a serious threat to aquatic organisms. In the present investigation, Cirrhinus mrigala an Indian major carp was exposed to different concentrations of clofibric acid (CA) and diclofenac (DCF) and certain biochemical and ionoregulatory responses were assessed under short and long term exposures. During short-term (96 h) exposure period, plasma glucose and sodium (Na+) levels were increased at all concentrations (1, 10 and 100 μg L−1) of CA and DCF treated fish. Plasma protein and chloride (Cl) levels were found to be decreased at all concentrations of CA and DCF exposed fish comparatively to control groups. Meanwhile an increase in plasma potassium (K+) level was noted in fish exposed to CA treatments alone and in DCF treatments it was decreased. In long-term exposure (35 d), plasma Na+ and Cl levels were found to be significantly increased at all concentration of CA and DCF. However, a biphasic trend was observed in plasma glucose, protein and K+ levels. In both the treatments, a significant (P < 0.01 and P < 0.05) changes were observed in all parameters measured in fish exposed to different concentrations of CA and DCF. The results of the present investigation indicate that both the drugs caused significant changes in biochemical and ionoregulatory responses of fish at all concentrations. The alterations of these parameters can be useful in monitoring of pharmaceutical residues present in aquatic environment.  相似文献

beta-Sitosterol is a weakly estrogenic phytosterol used extensively in functional foods to lower elevated serum cholesterol concentrations due to its inhibitory action on intestinal cholesterol absorption. It caused previously decreased sex steroid concentrations in fish and lowered sperm counts in rats. In the American mink (Neovison vison), litter size increased slightly due to dietary beta-sitosterol supplement. The aim of the present experiment was to conduct a dose-response study on the effects of beta-sitosterol on the reproduction of the American mink. Juvenile male and female mink (n=480) were exposed to 0, 5, 10 or 50 mg of peroral beta-sitosterol kg(-1)d(-1) for 10 months. After 3 months of exposure in November, 15 males per group were sacrificed and general biochemical variables reflecting overall health were determined. The beta-sitosterol-treated male mink had increased absolute and relative masses of intraabdominal fat and higher blood hemoglobin and serum high-density lipoprotein cholesterol concentrations. In spring, the top-rated male mink were mated with multiple females within each study group and reproductive success was assessed. No differences in the reproductive performance of the males (10-11 per group) or females (47-50 per group) could be detected in the exposed groups and the kits of all groups developed in a similar manner. The results suggest that dietary beta-sitosterol presents no significant risk to mammalian fertility.  相似文献

The mosquito Aedes aegypti is an epidemic vector of several diseases such as dengue fever and yellow fever. Several pesticides are used to control the mosquito population. Because of their frequent use, some mosquitoes have developed resistance. In this study, we used the Y-tube olfactometer to test essential oils of Cymbopogon species and screened specific formulas of components as repellents against Ae. aegypti. At 400 μL, the extracted oil of citronella grass (Cymbopogon nardus) and myrcene produced a low-active response by inhibiting mosquito host-seeking activity. Citronella grass, lemon grass (Cymbopogon citratus), citral and myrcene also produced a low-treatment response to repellents, for more potential to affect host-seeking behavior. Furthermore, the mixture of citral, myrcene, and citronellal oil (C:M:Ci = 6:4:1) greatly affected and inhibited host-seeking behavior (76% active response; 26% treatment response with 40 μL; 42.5%, 18% with 400 μL; and 19%, 23% with 1000 μL). As compared with the result for N,N-diethyl-3-methylbenzamide (DEET; 44%, 22% with 400 μL), adjusting the composition formulas of citronella oil had a synergistic effect, for more effective repellent against Ae. aegypti.  相似文献

Micronucleus assay combined with Giemsa and DAPI staining was performed on blood samples of subjects occupationally exposed to ultrasound. Lymphocytes were cultivated in vitro for 72 h. At 44h cytochalasin-B was added in cultures. Frequencies of micronuclei in exposed subjects statistically significant increased compared to control. The frequency of micronucleated cells and micronuclei in exposed subjects shows interindividual variability. Using DAPI staining we observed signal-positive and signal-negative micronuclei. Percentage of signal-positive micronuclei varies between 0 and 66.7% and signal-negative micronuclei between 33.3% and 100%. This study indicate harmful effects of ultrasound on human genome, but further investigations are necessary.  相似文献

Residues of polybrominated diphenylethers (PBDEs), extensively applied as flame retardants, are widely spread in the aquatic environment and biota. The present study investigates effects of the environmentally relevant lower brominated diphenylethers in two fish species in vivo under controlled laboratory conditions. Euryhaline flounder (Platichthys flesus) and freshwater zebrafish (Danio rerio) were exposed to a range of concentrations of a commercial pentabromodiphenylether mixture, DE-71. Chemical analysis of exposed fish showed a pattern of PBDE congeners that was very similar to that in wild fish. The resulting range included environmentally relevant, as well as higher levels. Animals were investigated histopathologically with emphasis on endocrine and reproductive organs. In zebrafish, hatching of embryos and larval development were assessed. Biochemical parameters were investigated in flounder as markers for suggested dioxin-like activity (ethoxyresorufin-O-deethylase=EROD), and activation of endogenous estrogen synthesis (gonad aromatase activity). Thyroid hormones were analyzed in plasma in both species. Benchmark analysis using internal PBDE concentrations showed a mild dose-dependent decrease of hepatic EROD and ovarian aromatase activities, and plasma thyroxin levels in flounder, and an increase of plasma thyroid hormone levels in zebrafish. These trends did not result in statistically significant differences from control fish, and major histopathological changes were not observed. Reproduction in zebrafish appeared to be the most sensitive parameter with statistically significantly reduced larval survival and non-significant indications for decreased egg production at internal levels that were more than 55 times the highest environmental recordings. The present results indicate limited risk for endocrine or reproductive effects of current environmental PBDE contamination in fish.  相似文献

Antibiotics are frequently used in agricultural systems to promote livestock health and to control bacterial contaminants. Given the upsurge of the resistant fecal indicator bacteria (FIB) in the surface waters, a novel statistical method namely, microbial risk assessment (MRA) was performed, to evaluate the probability of infection by resistant FIB on populations exposed to recreational waters. Diarrheagenic Escherichia coli, except E. coli O157:H7, were selected for their prevalence in aquatic ecosystem. A comparative study between a typical E. coli pathway and a case scenario aggravated by antibiotic use has been performed via Crystal Ball® software in an effort to analyze a set of available inputs provided by the US institutions including E. coli concentrations in US Great Lakes through using random sampling and probability distributions. Results from forecasting a possible worst-case scenario dose-response, accounted for an approximate 50% chance for 20% of the exposed human populations to be infected by recreational water in the U.S. However, in a typical scenario, there is a 50% chance of infection for only 1% of the exposed human populations. The uncertain variable, E. coli concentration accounted for approximately 92.1% in a typical scenario as the major contributing factor of the dose-response model. Resistant FIB in recreational waters that are exacerbated by a low dose of antibiotic pollutants would increase the adverse health effects in exposed human populations by 10 fold.  相似文献

In the context of two Environmental Health Surveillance Programs, launched in response to public and scientific concern in relation to waste incinerators located near Lisbon and in Madeira Island, two human biomonitoring projects have been started in Portugal, focussed in dioxins and dioxin-like compounds in human milk. Results from the undertaken studies have already provided data on the extent and pattern of dioxin body burden of both studied groups as well as a preliminary temporal trend of dioxin levels for the population residing near Lisbon incinerator. The present paper investigates difference between exposed and non-exposed subjects under study and, from a preventive perspective, possible covariates of the dioxin levels in human milk. Emissions from both incinerators appear to be well controlled as there is no increase of human body burden of dioxins as measured in human milk of individuals living near these facilities. Concerning other determinants of dioxin levels, results suggest confirmation of previously found significant age-dependent trend towards higher levels of dioxins in aged subjects. On the contrary, association between mother's levels of dioxins and parity lost significance. Apart from the issue of incineration, the general conclusion for the general population is that living in Lisbon as compared to Madeira results in higher milk dioxin levels and possible health risks. The profile of the single congeners for PCDD/Fs in human milk from Madeira and Lisbon shows similar contributions for 12378-PCDD, 23478-PCDF, 123678-HCDD and 2378-TCDD, that account altogether for about 84% of the total identified dioxin body burden in the studied groups.  相似文献

Malaria is a serious global health issue, with around 200 million cases per year. As such, great effort has been put into the mass distribution of bed nets as a means of prophylaxis within Africa. Distributed mosquito nets are intended to be used for malaria protection, yet increasing evidence suggests that fishing is a primary use for these nets, providing fresh concerns for already stressed coastal ecosystems. While research documents the scale of mosquito net fisheries globally, no quantitative analysis of their landings exists. The effects of these fisheries on the wider ecosystem assemblages have not previously been examined. In this study, we present the first detailed analysis of the sustainability of these fisheries by examining the diversity, age class, trophic structure and magnitude of biomass removal. Dragnet landings, one of two gear types in which mosquito nets can be utilised, were recorded across ten sites in northern Mozambique where the use of Mosquito nets for fishing is common. Our results indicate a substantial removal of juveniles from coastal seagrass meadows, many of which are commercially important in the region or play important ecological roles. We conclude that the use of mosquito nets for fishing may contribute to food insecurity, greater poverty and the loss of ecosystem functioning.

To test the hypothesis that Bti was specific to mosquito larvae, the granular form of Bti was tested on commonly found Hong Kong species from four naturally occuring aquatic insect orders and one species of decapod crustacean (Neocardina serra). Because data on Bti impacts on species in tropical and subtropical countries is relatively rare, the present study was conducted in Hong Kong's New Territories. Using static acute toxicity bioassays, all mosquito larvae exposed to Bti at the recommended dosage were dead except for some dark pigmented fourth instar individuals that had stopped feeding prior to emergence and as a result, did not ingest the Bti toxin. The only non-target species killed by the Bti were some chironomid species. In 1998 Bti inoculated pools along the Tai Tan River in the New Territories of Hong Kong had significantly fewer chironomids and mosquito larvae than control pools. By January, 1999, these same Bti inoculated pools still had no mosquito larvae in them, however, chironomids were no longer significantly rarer when compared to the control ponds. Thus, it would appear, that during the 1998-99 study in which Bti was added to the study sites at weekly intervals, Bti resistant chironomid species replaced Bti sensitive species in the Bti inoculated pools.  相似文献

Perfluorooctane sulfonate (PFOS) (C(8)F(17)SO(3)) and perfluorooctanoic acid (PFOA) (C(8)HF(15)O(2)) are synthetic chemicals widely used in industrial applications for their hydrophobic and oleophobic properties. They are persistent, bioaccumulative, and toxic to mammalian species. Their widespread distribution on earth and contamination of human serum raised concerns about long-term side effects. They are suspected to be carcinogenic through a nongenotoxic mode of action, a mechanism supported by recent findings that PFOS induced cell transformation but no genotoxicity in Syrian hamster embryo (SHE) cells. In the present study, we evaluated carcinogenic potential of PFOA using the cell transformation assay on SHE cells. The chemical was applied alone or in combination with a nontransformant concentration of benzo[a]pyrene (BaP, 0.4?μM) in order to detect PFOA ability to act as tumor initiator or tumor promoter. The results showed that PFOA tested alone in the range 3.7?×?10(-5) to 300?μM did not induce SHE cell transformation frequency in a 7-day treatment. On the other side, the combination BaP/PFOA induced cell transformation at all PFOA concentrations tested, which revealed synergistic effects. No genotoxicity of PFOA on SHE cells was detected using the comet assay after 5 and 24?h of exposure. No significant increase in DNA breakage was found in BaP-initiated cells exposed to PFOA in a 7-day treatment. The whole results showed that PFOA acts as a tumor promoter and a nongenotoxic carcinogen. Cell transformation in initiated cells was observed at concentrations equivalent to the ones found in human serum of nonoccupationally and occupationally exposed populations. An involvement of PFOA in increased incidence of cancer recorded in occupationally exposed population cannot be ruled out.  相似文献

In this study the possible toxic and biochemical effects of one intraperitoneal dose of 5 or 50 mg kg(-1) of 3,3',4,4'-tetrachlorobiphenyl (CB-77) or 50 or 200 mg kg(-1) of Clophen A50 (CloA50) on 28-day-old eider ducklings (Somateria mollissima) were investigated. After ten days, no significant differences could be observed in any of the toxic and biochemical parameters studied, apart from ethoxyresorufin (EROD) activity, when comparing group average values of the dosed and control animals. However, significant correlations were observed at day 10 after exposure between the individual internal PCB concentration and body weight gain and beak length growth (negative correlations in the CloA50 groups); relative liver weight and cytochrome P4501A activity (positive correlations in CB-77 and CloA50 groups); plasma thyroid-hormone and hepatic retinoid levels (negative correlations in CB-77 groups); and plasma retinol levels and the ratio plasma retin/hepatic retinyl palmitate (positive correlations in CB-77 groups only). Animal activity was significantly reduced in the group that received 50 mg CB-77 kg(-1). These observations indicate that eider ducks are a sensitive species to PCB toxicity and may be at risk for development of adverse health effects in relatively highly contaminated areas such as the Waddenzee.  相似文献

In the present study the acute toxicity of arsenic trioxide in fingerlings of Catla catla an Indian major carp was evaluated with renewal bioassay method. The median lethal concentration of arsenic trioxide to the fish C. catla for 96 h was found to be 20.41 ppm (with 95% confidence limits). From this a non-lethal dose of (2.041 ppm; 1/10th of LC 50 96 value) was selected and fingerlings were exposed to 35 d and hematological, biochemical and ionoregulatory responses were studied at days 7, 14, 21, 28 and 35. Arsenic trioxide produced a significant increase in hemoglobin, hematocrit, WBC count, plasma GPT levels and reduction in RBC count, plasma sodium, chloride, potassium, glucose, protein, GOT, LDH levels as compared to the control group. Gill Na+/K+-ATPase activity was influenced by arsenic trioxide exposure. A biphasic response was noted in the value of MCH and MCV. However the MCHC level was not altered in arsenic trioxide treated fish throughout the study period. Results of the present investigation suggest that arsenic trioxide affects the hematological, biochemical and ionoregulatory parameters of fish and alterations of these parameters can be useful in environmental biomonitoring of arsenic contamination.  相似文献

Ozone is an ubiquitous air pollutant that affects both human health and vegetation. There is concern about the number of hours human populations in nonattainment areas in the United States are exposed to levels of 03 at which effects have been observed. As improvement in air quality is achieved, it is possible that 03 control strategies may produce distributions of 1-h 03 concentrations that result in different diurnal profiles that produce greater potential exposures to 03 at known effects levels for multiple hours of the day. These concerns have prompted new analysis of aerometric data. In this analysis, the change in the seasonally averaged diurnal pattern was investigated as changes in 03 levels occurred. For the data used in this analysis, 25 of the 36 sites that changed compliance status across years showed no statistically significant change in the shape of the average diurnal profile (averaged by 03 season). For 71 percent (10 out of 14) of the sites in southern California and Dallas-Fort Worth, Texas, that showed improvement in O3 levels (i.e., reductions in the number of exceedances over the years), but still remained in nonattainment, a statistically significant change in the shape of the seasonally averaged diurnal profile occurred. Based on the results obtained in this study, the evaluation of diurnal patterns may be useful for identifying the influence of changes in emission levels versus meteorological variation on attainment status. Using data from the southern California and Dallas-Fort Worth sites, which showed improvements in 03 levels, changes were observed in the seasonally averaged diurnal profiles. On the other hand, for the sites moving between attainment and nonattainment status, such a change in shape was generally not observed and it was possible that meteorology played a more important role than changes in emission levels relative to attainment status.  相似文献

In researching health effects of air pollution, pollutant levels from fixed-site monitors are commonly assigned to the subjects. However, these concentrations may not reflect the exposure these individuals actually experience. A previous study of ozone (O3) exposure and lung function among shoe-cleaners working in central Mexico City used fixed-site measurements from a monitoring station near the outdoor work sites as surrogates for personal exposure. The present study assesses the degree to which these estimates represented individual exposures. In 1996, personal O3 exposures of 39 shoe-cleaners working outdoors were measured using an active integrated personal sampler. Using mixed models, we assessed the relationship between measured personal O3 exposure and ambient O3 measurements from the fixed-site monitoring station. Ambient concentrations were approximately 50 parts per billion higher, on average, than personal exposures. The association between personal and ambient O3 was highly significant (mixed model slope p < 0.0001). The personal/ambient ratio was not constant, so use of the outdoor monitor would not be appropriate to rank O3 exposure and evaluate health effects between workers. However, the strong within-worker longitudinal association validates previous findings associating day-to-day changes in fixed-site O3 levels with adverse health effects among these shoe-cleaners and suggests fixed-site O3 monitors may adequately estimate exposure for other repeated-measure health studies of outdoor workers.  相似文献

Organisms exposed to suboptimal environments incur a cost of dealing with stress in terms of metabolic resources. The total amount of energy available for maintenance, growth and reproduction, based on the biochemical analysis of the energy budget, may provide a sensitive measure of stress in an organism. While the concept is clear, linking cellular or biochemical responses to the individual and population or community level remains difficult. The aim of this study was to validate, under field conditions, using cellular energy budgets [i.e. changes in glycogen-, lipid- and protein-content and mitochondrial electron transport system (ETS)] as an ecologically relevant measurement of stress by comparing these responses to physiological and organismal endpoints. Therefore, a 28-day in situ bioassay with zebra mussels (Dreissena polymorpha) was performed in an effluent-dominated stream. Five locations were selected along the pollution gradient and compared with a nearby (reference) site. Cellular Energy Allocation (CEA) served as a biomarker of cellular energetics, while Scope for Growth (SFG) indicated effects on a physiological level and Tissue Condition Index and wet tissue weight/dry tissue weight ratio were used as endpoints of organismal effects. Results indicated that energy budgets at a cellular level of biological organization provided the fastest and most sensitive response and energy budgets are a relevant currency to extrapolate cellular effects to higher levels of biological organization within the exposed mussels.  相似文献

We evaluated the impact of the 1999 Belgian dioxin incident on the blood plasma polychlorinated dibenzo-p-dioxin (PCDD) and polychlorinated dibenzofuran (PCDF) levels among 232 Belgian blood donors (74% men, mean age 47 years). The Red Cross made plasma samples from before the incident of these donors available. A second plasma sample was collected during the second half of 2000. The sum of the 17 PCDD/F congeners was significantly lower in 2000 compared to 1998 (417 pg/g fat versus 445 pg/g fat, respectively). This could be completely attributed to the significant decrease of OctaCDD (301 pg/g fat in 2000 versus 277 pg/g fat in 1998). Moreover a slight but significant decrease was observed for 2,3,7,8-TetraCDF and for 1,2,3,4,6,7,8-HeptaCDF. 1,2,3,7,8-PentaCDF and 2,3,4,7,8-PentaCDF however showed a slight but significant increase (respective levels in 1998 were 0.004 and 14.5 pg/g fat compared to 0.006 and 17.9 pg/g fat in 2000). Given their significantly higher presence in incident related food samples this increase can be attributed to the food contamination episode. However, the total toxicity remained unchanged (22.9 in 1998 versus 23.1 pg WHO-TEQ/g fat, p>0.05). Moreover the observed congener profiles and the total PCDD/F levels were similar to those of other European non-occupationally exposed populations. In conclusion, the 1999 PCB/dioxin incident was traceable in the plasma profiles (rise of the two specific PCDF congeners), but comparison of the results for both years indicates that the changes were too small to cause an adverse public health effect.  相似文献

During the past several years, chlorine residuals and chlorinated organic compounds in drinking waters and aquatic environments have become a significant topic of study for scientists concerned about the quality of life in aquatic ecosystems as well as general public health. The effects of direct toxicity and/or carcinogenicity to human and aquatic life are the focal points for this concern.The effects of chloramines and chlorinated organic compounds present in the water distribution system after chlorination treatments are reviewed. Also discussed are the effects of chlorinated discharges from municipal secondary treatment plants and power plants on human health and aquatic life. Reactive-nitrogen (Nr) has a wide variety of beneficial and detrimental effects on human health. The most important of the beneficial effects are increasing global and regional food supplies and increased nutritional quality of available foods. However, lack of adequate dietary intake of amino acids and proteins is a serious cause of malnutrition when food supplies are inadequate because of poverty, drought, floods, wars, and displacements of people as refugees. There is sufficient, though limited, quantitative data indicating that increased circulation of Nr in the environment is responsible for significant human health effects via other exposure pathways. Nr can lead to harmful health effects from airborne occupational exposures and population-wide indoor and outdoor air pollution exposures to nitrogen dioxide and ozone. Nr can also affect health via water pollution problems, including methemoglobinemia from contaminated ground water, eutrophication causing fish kills and algal blooms that can be toxic to humans, and via global warming. The environmental pollutants stemming from reactive nitrogen are ubiquitous, making it difficult to identify the extent to which Nr exerts a specific health effect. As all populations are susceptible, continued interdisciplinary investigations are needed to determine the extent and nature of the beneficial and harmful effects on human health of nitrogen-related pollutants and their derivatives.  相似文献
