Two preliminary experiments were performed to explore the use of an intratracheal instillation technique as a cost-effective method of determining the biokinetics of lead (Pb) following respiratory exposure to Pb-contaminated soils. A novel intratracheal instillation procedure was refined and used to deliver a defined dose of Pb-contaminated soil or PbAc to the lower respiratory tract of rats. In the first experiment, rats were sacrificed at numerous time intervals post-dosing, and liver, kidney, blood, and bone tissues were collected for Pb analysis. In the second experiment, rats were dosed with Pb-contaminated soil or PbAc via intratracheal instillation or gastric gavage. All rats were sacrificed 96 h after dose administration and tissues were collected for lead analysis. Data from these experiments indicate Pb is well-absorbed following intratracheal instillation of Pb-contaminated soil, and the intratracheal instillation technique could be used as a cost effective method for exploring the biokinetics of Pb in Pb-contaminated soils following respiratory tract exposure. 相似文献

Furfural is an industrial compound used as a process intermediate and as a solvent; it poses a potential inhalation hazard in occupational settings. This study was carried out to find furfural-induced immunotoxicity in Wistar rat following inhalative exposure. The weights of thymus and lymph node were found decreased, while the weights of the liver and the adrenal gland were significantly increased following furfural exposure. Delayed-type hypersensitivity response was found decreased in furfural vapors-exposed animals when compared to that of control animals. The phagocytic index of peritoneal and alveolar exudates showed significant decrease and was most prominent (90%) in 30 days-exposed groups. The number of anti-rat anti-sheep red blood cell immunoglobulin M plaque forming cells of spleen got decreased in furfural-exposed groups in comparison to control. Taken together, this study indicates that inhalation of furfural induces immunotoxic manifestations that could lead to severe immunological disorders. 相似文献

Among the myriad particles the human respiratory tract is exposed to, a significant number are distinctive in that they include humic substances (HS) and humic-like substances (HULIS) as organic components. HS are heterogeneous, amorphous, organic materials which are ubiquitous occurring in all terrestrial and aqueous environments. HULIS are a complex class of organic, macromolecular compounds initially extracted from atmospheric aerosol particles which share some features with HS including an aromatic, polyacidic nature. As a result of having a variety of oxygen-containing functional groups, both HS and HULIS complex metal cations, especially iron. Following particle uptake by cells resident in the lung, host iron will be sequestered by HS- and HULIS-containing particles initiating pathways of inflammation and subsequent fibrosis. It is proposed that (1) human exposures to HS and HULIS of respirable size (<10 µm diameter) are associated with inflammatory and fibrotic lung disease and (2) following retention of particles which include HS and HULIS, the mechanism of cell and tissue injury involves complexation of host iron. Human inflammatory and fibrotic lung injuries following HS and HULIS exposures may include coal workers’ pneumoconiosis, sarcoidosis, and idiopathic pulmonary fibrosis as well as diseases associated with cigarette smoking and exposures to emission and ambient air pollution particles. 相似文献

2, 4-dichlorophenoxyacetic acid (2,4-D) is a post emergence herbicide. The tests were conducted to study the toxicity of 2,4-D using EC50 value in four dilutions. 2,4-D was applied at concentration of 25, 50, 75, 100, 200 and 300 mg l(-1) in the potato Dextrose agar medium. The effect of this herbicide was evaluated as the colony forming unit (CFU). EC50 value for 10(-3) dilution of soil was 138 +/- 5.944 mg l(-1). Soil physico-chemical parameters and mycofloral properties were also evaluated. Qualitatively 10 genera of fungi were observed in untreated soil, whereas 4 genera were found in 2,4-D administered dose. 相似文献

Adults Swiss mice were administered 5% solution of textile industry wastewater orally for 25 days and haematological parameters like RBC, WBC, Hb, and PCV were studied. Red cell indices like MCV, MCH and MCHC were calculated. Results indicate significant reduction in RBC, Hb and PCV levels. It is inferred that toxic effluents cause metabolic alteration in erythrocytes and reduce their Hb carrying capacity. 相似文献

The present study has been aimed to investigate the protective effect of beta-carotene against radiation-induced oxidative stress in mice tissues using lipid peroxidation and glutathione (GSH) as end points. Fourteen days oral priming administration of beta-carotene (35 mg/kg body weight) followed by an acute dose of gamma radiation (5 Gy) inhibited the augmented level of thiobarbituric acid reactive substance (TBARS) and a statistically significant protection against GSH depletion. Results evaluated from this study clearly indicate the antioxidative property of beta-carotene against gamma radiation, which is suggestive of free radical scavenging and singlet oxygen quenching. 相似文献

The main purpose of this study was to ascertain whether occupational exposure to asphalt fumes, under normal working conditions, is associated with any hepatotoxic, nephrotoxic, or hematotoxic response. This was a cross-sectional study in which 210 subjects (80 exposed and 130 reference subjects) were investigated. Using standard methods, atmospheric concentrations of total particulate and benzene-soluble fractions of asphalt fumes were measured. Additionally, urine and blood samples were taken from subjects for complete blood count, white blood cell differential test, urinalysis, and routine biochemical tests of kidney and liver function. Both groups were similar as far as their demographic variables and smoking habits were concerned. The levels of exposure to total particulate and benzene-soluble fractions were estimated to be 0.9?±?0.2 and 0.32?±?0.09 mg m?3, respectively (mean?±?SD), both of which being lower than the existing threshold limit values. Statistically, significant associations between exposure to asphalt fumes and changes in most liver and kidney function tests and complete blood count parameters provided circumstantial evidence in favor of the notion that exposure to sub-threshold limit value levels of total particulate and benzene-soluble fractions is associated with early liver and kidney dysfunction as well as hematological disorders. 相似文献

Ocean acidification, or the lowering of seawater pH, is caused by sequestration of atmospheric CO2 into the oceans. This study investigated the effects of present-day pH 8.0, predicted ocean surface pH for the years 2100
and 2300 (pH 7.7 and pH 7.3, respectively) and an extreme pH (pH 7.0) on fertilisation and embryogenesis in the Antarctic
nemertean worm Parborlasia corrugatus and sea urchin Sterechinus neumayeri. Fertilisation success was not affected by pH in P. corrugatus across a range of sperm concentrations. Fertilisation success in S. neumayeri declined significantly in pH 7.0 and 7.3 seawater, but only at a low sperm concentration. Seawater pH had no effect on the
rate of egg cleavage in S. neumayeri, or the proportion of abnormal embryos 1-day post-fertilisation. P. corrugatus embryogenesis was also relatively robust to pH changes, with a significant effect detected only when the seawater pH was
decreased to 7.0. While fertilisation and early cell division were relatively robust, later development through to the gastrula
was sensitive to pH. In S. neumayeri, an effect of pH on development was evident by the gastrula stage, while there were significantly more abnormal P. corrugatus embryos in pH 7.0 up to the blastula stage, and in pH 7.0 and pH 7.3 at the coeloblastula stage. Our results are similar
to the observations on other marine invertebrate species where fertilisation and early embryonic development are generally
robust to lowered seawater pH, while the older coeloblastula and gastrula stages are more responsive. We also found no evidence
to suggest that Antarctic species are more adversely affected by lower seawater pH compared with the findings for non-Antarctic
counterparts. We conclude that in the two species we examined, near-future decreases in pH (decreases of ≈0.3–0.5 pH units)
may not have a significant effect on fertilisation and early embryogenesis, while predicted longer term decreases (decreases
of ≈0.7–0.77 pH units) could reduce fertilisation success in S. neumayeri if sperm concentrations are low and may increase abnormalities in P. corrugatus during later embryogenesis. 相似文献

Phenoxyherbicides induce peroxisome proliferation, as do other structurally related or unrelated chemicals which were found to be nongenotoxic carcinogens in rodents. The widespread use of 2,4-dichlorophenoxyacetic acid (2,4-D) raised concerns over the potential to produce cancer, yet results from epidemiological and experimental studies do not provide definitive evidence that 2,4-D is carcinogenic. The objective of the present study was to verify whether 2,4-D might interfere with cell growth similar to other peroxisome proliferators using cultured avian hepatocytes. Primary hepatocyte cultures from 18-day-old chick embryos were exposed to 2,4-D for up to 72 h followed by an assessment of replicative DNA synthesis and apoptosis. Further, peroxisome proliferation was evaluated. Results showed that 2,4-D stimulated DNA synthesis and suppressed spontaneous apoptosis, while the effects on peroxisome proliferation were less evident. 相似文献

By means of scanning and transmission electron microscopy, the influence of the red tide species Chattonella antiqua was examined with respect to the surface ultrastructures of chloride cells of the yellowtail Seriola quinqueradiata. Conspicuous ultrastructural alterations occurred on the apical surface of these cells. The majority of chloride cells in the control gills showed an apical surface with numerous cellular extensions, while more than half the chloride cells affected by red tide organisms exhibited an apical surface with fewer and smaller extensions, a wrinkled apical surface, or a protruded apical surface. These ultrastructural alterations of chloride cell surface may be due to the partial disturbance of salinity by C. antiqua, and reflect the changes of the ion-transport function in yellowtail gills exposed to red tide water. 相似文献

Sea scallops Placopecten magellanicus, which had been trawl-collected in late November, 1982 off the Rhode Island, USA coast, were exposed to 20 g of Cd or Cu per liter for a period of seven weeks in a flowing seawater system. Metal analyses of kidneys from both groups indicated uptake of both metals, although the tissue concentrations of Cd declined markedly in the Cu-treated scallops. The ultrastructural appearance of tubule cells of kidneys from Cd-exposed scallops was indistinguishable from controls. In contrast, tubule cells from scallops exposed to Cu showed marked cellular degeneration with loss of concretions. These ultrastructural changes were associated with significant reductions in renal isocitrate dehydrogenase activity in the Cu-treated scallops. Elemental analyses conducted on the kidney concretions and on the cytosolic metal-binding proteins of Cd-exposed scallops showed a 6-to 7-fold increase in Cd content of both these metalsequestering compartments, with concomitant changes in Zn, Mn, and Cu content. Loss of the concretions from Cu-treated scallops precluded analysis of this compartment, but Cu sequestration within cytosolic metal-binding proteins was associated with marked reductions of Zn and Cd from these proteins, suggesting disruption of this cellular mechanism for control of divalent metal cations. These findings support the hypothesis that toxic metal perturbation of normal homeostatic mechanisms that control divalent metal cation bioavailability is important factor in mediating cell injury from these agents. 相似文献

Weight specific oxygen consumption and heart rates were investigated in blue swimming crab, Portunus pelagicus to indicate the stress in response to increasing salinities (50, 75, 100 and 125%SW) and increasing copper concentration (0, 0.75, 1.5, 3 and 6 ppm copper) at temperature 25 degrees C. Oxygen consumption (micromol.g(-1).h(-1) was measured using closed system respirometer (Strathkelvin Instruments oxygen meter model 781). Heart rates (beat.min(-1)) were recorded using impedance techniques. It has been found that oxygen consumption and heart rate increased with decreasing salinities and increasing copper concentration. The statistic showed weight specific oxygen consumption of P. pelagicus after 96 hr exposure to 6 ppm copper in all salinities were significantly different (P < 0.05) from 0 ppm copper. Mean heart rate of P. pelagicus in 50%SW after 96 hr exposure to 6 ppm copper were significantly different (P < 0.05) from 0 ppm copper. 相似文献

Environmental hypoxia and the release of reduced manganese (Mn) are often combined stress factors in marine sediments. Previous investigations have shown that hypoxia induces variable responses in the concentration of the respiratory pigment haemocyanin (Hc) of crustaceans. A recent study demonstrated that the magnitude and direction of the changes was dependent on the initial level of Hc in individuals of the Norway lobster Nephrops norvegicus. The changes also took place within 24 h rather than days as suggested by previous studies. In this present study we investigated changes of Hc in individual N. norvegicus in uncontaminated sea water after exposure to and release from realistic hypoxic stress (PO 2=6.0 kPa). Furthermore we investigated how concomitant exposure to realistic concentrations of manganese (20 mg l -1) modified the responses we observed. We confirmed a between-individual variation in response and a change in Hc towards an optimum level after hypoxia, but also showed that changes that took place post-hypoxic exposure occurred over a timescale similar to that of the hypoxic response itself. Manganese exposure resulted in no significant changes in Hc whereas Mn exposure combined with hypoxia resulted in a significant decrease (15%) in all individuals independent of initial Hc. N. norvegicus was thus unable to compensate for the effects of hypoxia by synthesis of Hc after exposure to naturally occurring Mn concentrations. 相似文献

Summary. The effect of rearing larvae of Trichoplusia ni on
individual feeding deterrents or on binary mixtures of deterrents
on their subsequent gustatory sensitivity was measured
in paired choice leaf disc bioassays. Our working hypothesis
was that mixtures of antifeedants (pure allelochemicals)
would mitigate decreased feeding deterrent response following
prolonged exposure in this generalist herbivore. Neonate
larvae were reared on cabbage leaves treated with individual
feeding deterrents (digitoxin, thymol, toosendanin or xanthotoxin),
or with binary mixtures of these until the third instar.
Feeding deterrent responses to each antifeedant or mixture
was then determined in a leaf disc choice bioassay. All of the
mixtures produced additive deterrence when presented to
naïve larvae. Larvae reared on individual antifeedants
showed a significantly decreased feeding deterrent response
(except to digitoxin), whereas larvae reared on binary mixtures
of antifeedants did not show a decreased feeding deterrent
response to any of them. Such mixtures were synergistic
in terms of their feeding deterrence to experienced larvae.
Our experiment supports the hypothesis (Jermy 1986) that
mixtures of deterrents can prevent decreased feeding deterrent
response following prolonged exposure, and provides one
explanation for the multiplicity of chemical defenses found
