The persistence and fate of chlorpyrifos and its two metabolites, chlorpyrifos-oxon and the 3, 5, 6-trichloro-2-pyridinol (TCP) break-down product were investigated on kale and collard leaves under field conditions. A simultaneous extraction and quantification procedure was developed for chrorpyrifos and its two main metabolites. Residues of chlorpyrifos, chlorpyrifos oxon, and TCP were determined using a gas chromatograph (GC) equipped with an electron capture detector (GC/ECD). Chlorpyrifos metabolites were detectable up to 23 days following application. Residues were confirmed using a GC equipped with a mass selective detector (GC/MSD) in total ion mode. Initial residues of chlorpyrifos were greater on collard (14.5 µg g?1) than kale (8.2 µg g?1) corresponding to half-lives (T1/2) values of 7.4 and 2.2 days, respectively. TCP, the hydrolysis product, was more persistent on collards with an estimated T1/2 of 6.5 days compared to kale (T1/2 of 1.9 days).  相似文献

2株乙酰甲胺磷降解菌的分离鉴定及降解特性研究   总被引:1,自引:0,他引:1  
利用富集及驯化培养方法,从长期生产农药企业的废水处理系统、厂房周边污染的土壤和池水中,分离筛选出2株能够高效降解乙酰甲胺磷的菌株Y3、Y6。在形态特征和生理生化鉴定的基础上,对其16S rDNA序列进行了分析,并重点研究了它们对乙酰甲胺磷及其他农药的降解特性和抗性。结果表明,Y3、Y6分别为寡养单胞菌(Stenotrophomonas)和假单胞菌(Pseudomonas)。Y3、Y6在乙酰甲胺磷浓度分别为500 mg/L和1 000 mg/L,培养温度30℃,初始pH8,接种量为2.5%条件下,一周内可以将80%左右的乙酰甲胺磷矿化为磷酸根。外加葡萄糖及酵母膏对降解效率的研究表明,当酵母膏含量为1 g/L时,降解效果最理想;而外加葡萄糖的量,能相对抑制其对农药的降解。抗性实验显示,Y3、Y6均可在较高浓度的其他有机磷类及氨基甲酸酯类农药的普通培养基中生长,对其他农药的抗性也比较广泛。植物侵染实验显示,Y3、Y6对实验中的豆科、禾本科、十字花科及葫芦科植物不具备致病性,说明Y3、Y6环境安全性较强。  相似文献

The degradation of the herbicide acetochlor, in a neoluvisol and in a calcosol were studied as a function of depth (0-25cm and 25-50cm) and temperature (25 degrees C and 15 degrees C) under controlled laboratory conditions during 58 and 90 days, respectively. The surface and sub-surface soil samples were respectively spiked with 1 and 0.01mgkg(-1) of 14C-acetochlor, the concentrations observed in previous field monitoring. The half-lives (DT50) varied from 1.4 to 14.9 days depending on the soil, temperature and applied concentration. The maximal mineralization (24%) was observed for the surface calcosol at 25 degrees C. The comparison of results obtained for sterilized and non-sterilized soils, the decrease of DT50 with the increase of temperature, the shape of CO2 emissions and the increase of number of aerobic endogenous microflora through the experiment suggested that biological process are dominant in degradation. A particular attention was paid to the formation and dissipation of metabolites ESA (ethanesulphonic acid) and OA (oxanilic acid) during the whole experiment. At 25 degrees C, ESA and OA were observed after three days, but as ESA concentration decreased over time in surface calcosol, it remained constant in surface neoluvisol. A difference in ESA/OA ratio depends on the soil with a predominance of OA in surface neoluvisol and a disappearance of OA in surface calcosol.  相似文献

好氧反硝化苯酚降解菌的分离鉴定及动力学   总被引:1,自引:0,他引:1  
从驯化菌群中分离筛选出一株好氧反硝化苯酚降解细菌,经生理生化反应及16S rDNA测序,鉴定为Diaphorobacter属细菌。在好氧条件下,该菌株以苯酚为唯一碳源和能源,利用NO-3-N作为反硝化电子受体,其生长与反硝化特性研究表明:在接种量5%(体积分数),30℃,180 r/min振荡培养条件下,菌株降酚能力可达1 400 mg/L,同时,能有效去除初始浓度为165 mg/L的硝酸盐氮,60 h其去除率为91.5%,高含量苯酚对菌体生长有一定的抑制作用。应用Haldane方程对其生长过程进行动力学模拟,拟合曲线与实验测定值相关性良好,各参数分别为μmax(最大比增长率)0.324 h-1,Ks(半饱和常数)9.36 mg/L,Ki(抑制常数)146.72 mg/L,通过理论分析及实验验证得,该菌株苯酚降解动力学与其生长动力学表现出相似的趋势。  相似文献

从太原市某污水处理厂SBR活性污泥中分离纯化得到一株高温(50℃)好氧反硝化菌,命名为XF3。通过生理生化特性鉴定及16S rDNA序列分析,初步鉴定为波茨坦短芽孢杆菌。通过单因子实验考察碳源、C/N、pH及接种量对该菌株的生长情况与反硝化性能的影响。结果表明,菌株XF3最适碳源为琥珀酸钠,最佳C/N为12∶1,最佳pH为7,最适接种量10%(体积分数)。同时该菌株具有良好的异养硝化能力,48 h可以将73 mg/L氨氮几乎全部降解。  相似文献

为了得到一株具有降解微囊藻毒素-RR(MC-RR)特性的产芽孢菌株,采用加热富集芽孢菌的方法,从太湖分离到一株MC-RR降解菌CM1,该菌对MC-RR具有强烈的降解特性。通过形态学特征、生理生化特征及16S rDNA序列分析鉴定该菌株属于耐硼赖氨酸芽孢杆菌(Lysinibacillus boronitolerans)。通过研究温度和pH值对菌株CM1降解MC-RR能力的影响,发现菌株CM1在60 h将MC-RR由12.77μg/mL降解到1.67μg/mL,降解率达86.90%,最适降解温度为37℃,最适pH值为7.0。CM1菌株的胞外物质和胞内物质均能降解MC-RR,但胞内物质具有更强烈的降解特性,12 h可以将7.27μg/mL的MC-RR完全降解。为丰富MC-RR降解菌纯菌种研究以及在去除水体中MC-RR应用研究方面提供了理论基础。  相似文献

一株DBP高效降解菌的分离、鉴定与降解性能   总被引:1,自引:0,他引:1  
从镇江某垃圾站污染土壤中分离出1株能够以邻苯二甲酸二丁酯为唯一碳源和能源生长的细菌高效降解菌TM。经形态观察、生化鉴定、16S rRNA序列及系统发育分析,鉴定该菌株为变形假单胞菌(Pseudomonas plecoglossicida)。采用正交实验和单因素对照实验对这株菌株的降解条件进行优化,确定其最适生长条件为:温度30℃,pH=7.0。在最适降解条件下,其在72 h内对400 mg/L DBP降解率达到88.56%,为邻苯二甲酸二丁酯的高效降解菌。底物广谱性实验表明,该菌株邻苯二甲酸二辛脂(DOP)、邻苯二甲酸(2-乙基已基)酯(DEHP)都具有良好的降解能力,表明其具备良好的底物广谱性,说明该菌株在处理邻苯二甲酸酯类化合物的污染治理中有独特的应用潜力。  相似文献

鉴定了从黑龙江扎龙湿地土壤中筛选出的一株抗铅镉的菌株,研究了其生物学特性和部分生理生化指标。利用16SrDNA序列分析鉴定其菌属,并且研究Pb2+、Cd2+、Pb2+/Cd2+、温度、pH、盐以及抗生素对菌株生长的影响。经鉴定,该菌株为阴沟肠杆菌属(Enterobacter cloacae)。该菌株对Pb2+的去除率和吸附率分别达到了70.34%和44.39%,对Cd2+的去除率和吸附率分别达到了40.54%和25.14%。该菌株最适生长温度为25℃,最适生长pH为7.0左右。此菌株对抗生素亚胺培南最敏感。随着盐、Pb2+、Cd2+、Pb2+/Cd2+混合浓度的升高,该菌株生长受到抑制。  相似文献

从好氧活性污泥中分离得到一株能以1,1-二氯乙烯(1,1-DCE)作为惟一碳源和能源生长的革兰氏阴性菌株D-B,经鉴定属于产碱杆菌属(Alcaligenessp.)。当维持菌株D-B浓度一定时,1,1-DCE的去除率随着1,1-DCE浓度的增大先增加后降低,且降解过程主要发生在加入1,1-DCE后的3~5 h内。当1,1-DCE的初始浓度为300μg/L时去除率达到最大值85.32%。菌株D-B对1,1-DCE的降解符合Monod方程,饱和常数Ks=21.96 mg/L,1,1-DCE的最大比基质降解速率Vmax=50.76 mg/(L.h)。  相似文献

从农药厂污泥中分离到一株能降解甲氰菊酯的光合细菌PSB07-19.鉴定结果表明,PSB07-19为沼泽红假单胞菌(Rhodopseudomonas palustris),其最适生长温度为30℃,最适pH为6.5.实验结果表明,该菌以共代谢方式降解甲氰菊酯,对甲氰菊酯的最大耐受质量浓度为600 mg/L,培养15 d对400 mg/L甲氰菊酯的降解率达45.51%,降解最适pH、温度分别为7.5、30℃.  相似文献

In this study, a bacterial strain, CH-1, capable of degrading 3-bromocarbazole (3-BCZ) was isolated from a polluted soil. Based on its physio-biochemical characteristics and 16S rRNA genes, strain CH-1 was identified as a Stenotrophomonas sp. Strain CH-1 was able to degrade 70% of 50 mg/L 3-BCZ within 8 d at pH 7.0 and 30°C in mineral salt medium (MSM). During the process, the main intermediate metabolite was identified as (2E, 4Z)-6-(2-amino-5-bromophenyl)-2-hydroxy-6-oxhexa-2, 4-dienoic by gas (2E, 4Z)-6-(2-amino-5-bromophenyl)-2-hydroxy-6-oxhexa-2,4-dienoic via gas chromatograph-mass spectrometry (GC-MS) analysis. The metabolite disappeared after 14 d, suggesting that the metabolite can also be degraded by strain CH-1. 3-BCZ is a new persistent organic pollutant. This is the first report of the biodegradation of 3-BCZ. The results indicated that strain CH-1 may be a promising bacterial candidate for the bioremediation of environments polluted with polyhalogenated carbazoles (PHCs).  相似文献

从纺织厂的工业废水排污口取样,分离筛选出了一株嗜热的高效偶氮染料脱色菌PDR2。通过形态学、生理生化特征以及16S rDNA基因序列分析等方法对该菌株进行鉴定,发现该菌为厌氧芽孢杆菌,与Anoxybacillus sp(GQ471935)同源性最高。此外,分别考察了菌株PDR2在不同碳源、氮源、pH以及培养温度等条件下对偶氮染料直接黑38脱色性能的影响,在此基础上进一步研究了菌株PDR2对不同浓度直接黑38染料的耐受性。结果表明,菌株PDR2在55℃、pH 6.0且以葡萄糖为碳源,牛肉膏和蛋白胨为混合氮源时其脱色效果最佳,且菌株对偶氮染料直接黑38有着较强的耐受能力,在染料浓度低于0.4 g·L-1时其脱色效果良好。在最佳条件下静置培养24 h时,菌株PDR2对0.1 g·L-1直接黑38染料的脱色率高达92.34%,展出了良好的应用潜力。  相似文献

以腐植酸(HA) 溶液为吸附剂、从受多环芳烃污染的土壤中分离出来的降解菌制成为生物修复剂,以多环芳烃(PAHs)萘、菲、芘、荧蒽、苯并蒽、苯并芘为土壤污染物,对PAHs污染土壤进行修复实验。目的是筛选与分离吸附于HA的PAHs降解菌,研究HA与降解菌的协同效应对PAHs的降解效率的影响。用经过HA吸附的PAHs富集分离培养出1株高效降解菌株, 命名为Tzyx3,鉴定其为解脂耶氏酵母菌(Yarrowia lipolytica)。15 d后,土壤中萘、菲、芘、荧蒽、苯并蒽、苯并芘的降解率分别为90.7%、91.0%、74.7%、86.9%、84.7%和74.7%,表明Tzyx3和HA在PAHs污染土壤中存在协作关系,Tzyx3能够直接利用HA对土壤中的多环芳烃进行降解。  相似文献

In-vitro inhibitions of brain AChE in brook trout, Salvelinus fontinalis (Mitchill), by aminocarb (4-dimethylamino-m-tolyl N-methylcarbamate) and its toxic metabolites, MAA (4-methylamino-m-tolyl N-methylcarbamate), AA(4-amino-m-tolyl N-methylcarbamate), MFA (4-methylformamido-m-tolyl N-methylcarbamate) and FA (4-formamido-m-tolyl N-methylcarbamate) were investigated. The molar concentrations of inhibitors causing 50% inhibition (I50s) were AA (3.62 X 10(-6] less than MAA (7.92 X 10(-6] less than aminocarb (1.01 X 10(-5] less than MFA (4.29 X 10(-5] less than FA (7.11 X 10(-5]. After exposure of fish to various concentrations of aminocarb (25, 250 and 2500 ppb) and MAA (25, 250, 500 and 2500 ppb) at 9 degrees C in dechlorinated tap water for 96 h, inhibitions of brain AChE ranged from 13 to 77%. Mortality occurred only in fish exposed to 500 ppb (22%) and 2500 ppb (100%) of MAA. Enzyme activities recovered to the control levels 12 to 96 h after the fish had been transferred to clean water for clearing; in survivors of the 500-ppb MAA exposure, however, AChE activities decreased again thereafter.  相似文献

石油污染土壤生物修复菌Z1a-B的分离鉴定与调控效应研究   总被引:2,自引:2,他引:2  
从山东东营胜利油田附近的石油污染土壤中分离筛选得到一株高效石油降解菌Z1a-B,通过菌落形态及显微镜个体形态观察对其初步鉴定到属,并采用气相色谱/质谱(GC/MS)法分析了Z1a-B的石油降解性能,采用投加石油降解菌、调节土壤N、P含量和优化环境因素等措施,进行了为期60d的石油污染土壤室外自然堆制生物修复实验。结果表明,Z1a-B为链霉菌属白孢类群,其摇瓶培养的石油降解率为66.4%;Z1a-B有着很宽的烷烃降解谱;N、P最佳的添加量组合为KNO32.50g/kg、K2HPO40.35g/kg,即N/P(质量比)为5.57:1.00,此时的石油降解率达63.5%,土壤脱氢酶活性达最高值,为2.99μL/g;石油降解的最佳环境条件为:将石油质量分数为3.3%的100g土样调节pH至8.5后,装入容积为300mL的锥形瓶中灭菌,再接种孢子密度为2.7×108个/mL的菌剂5.5mL,于28℃下进行生物降解,在此条件下的石油降解率可达76.5%;土壤脱氢酶活性的测定结果可以作为检验石油污染土壤生物修复效果的重要指示指标之一;室外自然堆制生物修复实验中,添加菌剂、锯末、秸秆以及N、P后,石油降解率可达69.9%,总体来说,室外自然堆制生物修复是一种投资少、见效快、治理效果较好的石油污染土壤治理方法。  相似文献

一株高效异养硝化-好氧反硝化菌的分离鉴定及脱氮性能   总被引:6,自引:0,他引:6  
从经驯化的污泥中筛选出一株异养硝化-好氧反硝化细菌,编号为TN-05,通过形态学特征观察,生理生化特征试验和核酸序列分析鉴定其为门多萨假单胞菌(pseudomonasmendocina)。同时对其进行脱氮性能研究,结果表明,TN-05具有较好的异养硝化能力,菌株在培养至48h时对总氮和氨氮去除率均能达95%以上。通过反硝化能力验证实验发现,菌株对NO3-N和N0f—N也分别具有较好的去除效率。将菌株应用于人工合成废水中,发现对废水中氨氮优先利用并能在24h时使去除率接近100%,对硝态氮和亚硝态氮也具有一定的去除效率。因此,菌株TN-05是一株同时具备异养硝化和好氧反硝化能力的高效菌株。  相似文献

The main metabolites formed from Iprodione and Procymidone during the composting process have been isolated and identified by HPLC-DAD-MSD. After addition of the fungicides to the composting pile, we monitored the reaction of the two analytes and the formation of their degradation products for eight months. We verified the nature of the metabolites by comparison with those hypothesised in the literature and by comparison with the behaviour of an abiotic process in aqueous acetonitrile pH 6 and at 35 degrees C. After taking into account the different kinetic behaviours of the fungicides on degradation in compost and hydro-organic solution, breakdown pathways are proposed for biodegradation.  相似文献
