Lead (Pb) accumulation in Sesbania drummondii shoots was enhanced by 654 and 415% in the presence of 100 microM IAA and 100 microM NAA, respectively, compared to control plants (Pb alone). However, when IAA or NAA was added along with EDTA, Pb accumulation further increased in shoots by 1349% and 1252%, respectively. Scanning electron microscopic observations revealed that Pb particles in both leaf and stem of Pb+EDTA+IAA and Pb+EDTA+NAA treated plants were concentrated in the region of vascular bundles. In root tissue, Pb particles were present between epidermis to stele region. Plant growth in both treatments (Pb+100 microM IAA and Pb+100 microM NAA) was comparable to control plants; however, it was significantly inhibited in the treatments containing Pb+EDTA and EDTA at concentrations of 10 microM of IAA or NAA. Moreover, the photosynthetic efficiency and strength of the treated plants were not affected in the presence of IAA or NAA and EDTA.  相似文献

The term "assisted phytoextraction" usually refers to the process of applying a chemical additive to contaminated soil in order to increase the metal uptake by crop plants. In this study three commercially available plant growth regulators (PGRs) based on cytokinins (CKs) were used to boost the assisted phytoextraction of Pb and Zn in contaminated soil collected from a former manufactured gas-plant site. The effects of EDTA treatment in soil and PGR treatment in leaves of Helianthus annuus were investigated in terms of dry weight biomass, Pb and Zn accumulation in the upper parts of the plants, Pb and Zn phytoextraction efficiency and transpiration rate. Metal solubility in soil and its subsequent accumulation in shoots were markedly enhanced by EDTA. The combined effects of EDTA and cytokine resulted in an increase in the Pb and Zn phytoextraction efficiency (up to 890% and 330%, respectively, compared to untreated plants) and up to a 50% increase in foliar transpiration. Our results indicate that exogenous PGRs based on CKs can positively assist the phytoextraction increasing the biomass production, the metal accumulation in shoots and the plant transpiration. The observed increase in biomass could be related to its action in stimulation of cell division and shoot initiation. On the other hand, the increase in metal accumulation in upper parts of plant could be related to both the role of PGRs in the enhancement of plant resistance to stress (as toxic metals) and the increase in transpiration rate, i.e. flux of water-soluble soil components and contaminants by the regulation of stomatal opening.  相似文献

Chelant-enhanced phytoextraction of heavy metals is an emerging technological approach for a non-destructive remediation of contaminated soils. The main objectives of this study were (i) to assess the extraction efficiency of two different synthetic chelating agents (ethylenediaminetetraacetic acid (EDTA) and ethylenediaminedisuccinic acid (EDDS)) for desorbing Pb from two contaminated agricultural soils originating from a mining and smelting district and (ii) to assess the phytoextraction efficiency of maize (Zea mays) and poplar (Populus sp.) after EDTA application. EDTA was more efficient than EDDS in desorbing and complexing Pb from both soils, removing as much as 60% of Pb. Maize exhibited better results than poplar when extracting Pb from the more acidic (pH approximately 4) and more contaminated (up to 1360 mg Pb kg(-1)) agricultural soil originating from the smelting area. On the other hand, poplars proved to be more efficient when grown on the near-neutral (pH approximately 6) and less contaminated (up to 200 mg Pb kg(-1)) agricultural soil originating from the mining area. Furthermore, the addition of EDTA led to a significant increase of Pb content especially in poplar leaves, proving a strong translocation rate within the poplar plants.  相似文献

The uptake and distribution of cadmium in tomato plants (Lycopersicon esculentum, Mill, cv. Tiny Tim) were examined with and without the presence of ethylenediaminetetraacetic acid (EDTA) as chelating agent and 2,4-dinitrophenol (DNP) as metabolic inhibitor. Eight-week-old intact and derooted tomato seedlings were used in hydroculture experiments with cadmium applied as (115)Cd(NO(3))(2) in a range of concentrations. Measurements of the (115)Cd content of roots, stems and leaves were carried out by gamma-ray spectroscopy. The data showed that applications of both EDTA and DNP resulted in reduced total Cd accumulation in the plants, but relatively enhanced Cd transport into the above-ground plant parts. The Cd mobility in the transport channels in the shoots was increased by EDTA in both intact and derooted plants. Application of DNP leads to increased relative Cd import to leaves in derooted plants, but a reduced import into leaves of intact plants. These results suggest that Cd-complexes are formed in root cells before root-to-shoot transport. Furthermore, initial Cd uptake may be associated with adsorption on the negative charges of the cell walls of the root system. The high Cd mobility in shoots, in experiments with intact plants and Cd-EDTA application, indicates the possibility of simultaneous uptake of Cd and EDTA, possibly as a Cd-EDTA complex.  相似文献

The effect of soil ozonation on Pb and Zn extraction with EDTA, bioavailability (Ruby's Physiologically Based Extraction Test, PBET) and mobility (Toxicity Characteristic Leaching Procedure, TCLP) of Pb was studied on contaminated soils taken from 7 different locations in the Mezica Valley, Slovenia. EDTA extraction (40 mmol kg(-1)) removed from 27.4+/-1.5% to 64.8+/-1.5% of Pb, and from 1.9+/-0.2% to 22.4+/-2.0% of Zn from tested soils, and significantly reduced soil Pb bioavailability (PBET) and mobility (TCLP). Pretreatment of tested soils with ozone before EDTA extraction enhanced EDTA extractability of Pb for 11.0 to 28.9%, but had no effect on the extractability of Zn. In most of the soils, ozonation had no statistically significant effect on bioavailability and mobility of Pb, residual after EDTA extraction. Using linear regression analysis we found a significant increase (p<0.01) in EDTA extractability of Pb after soil ozonation in soils with a higher initial Pb content. EDTA extractability of Pb after soil ozonation was also significantly higher for soils with a lower Pb extractability when treated with EDTA alone. We found no correlation between soil organic matter content and the percentage of the Pb fraction bound to soil organic matter (where from 25.6+/-1.3% to 73.2+/-0.6% of Pb reside in tested soils) and Pb extractability with EDTA after soil ozonation.  相似文献

Optimizing process parameters that affect the remediation time and power consumption can improve the treatment efficiency of the electrokinetic remediation as well as determine the cost of a remediation action. Lab-scale electrokinetic remediation of Pb-contaminated soils was investigated for the effect of complexant ethylenediaminetetraacetic acid (EDTA) and acetic acid and approaching anode on the removal efficiency of Pb. When EDTA was added to the catholyte, EDTA dissolved insoluble Pb in soils to form soluble Pb–EDTA complexes, increasing Pb mobility and accordingly removal efficiency. The removal efficiency was enhanced from 47.8 to 61.5 % when the EDTA concentration was increased from 0.1 to 0.2 M, showing that EDTA played an important role in remediation. And the migration rate of Pb was increased to 72.3 % when both EDTA and acetic acid were used in the catholyte. The “approaching anode electrokinetic remediation” process in the presence of both EDTA and acetic acid had a higher Pb-removal efficiency with an average efficiency of 83.8 %. The efficiency of electrokinetic remediation was closely related to Pb speciation. Exchangeable and carbonate-bounded Pb were likely the forms which could be removed. All results indicate that the approaching anode method in the presence of EDTA and acetic acid is an advisable choice for electrokinetic remediation of Pb-contaminated soil.  相似文献

Two heavy metal contaminated calcareous soils from the Mediterranean region of Spain were studied. One soil, from the province of Murcia, was characterised by very high total levels of Pb (1572 mg kg(-1)) and Zn (2602 mg kg(-1)), whilst the second, from Valencia, had elevated concentrations of Cu (72 mg kg(-1)) and Pb (190 mg kg(-1)). The effects of two contrasting organic amendments (fresh manure and mature compost) and the chelate ethylenediaminetetraacetic acid (EDTA) on soil fractionation of Cu, Fe, Mn, Pb and Zn, their uptake by plants and plant growth were determined. For Murcia soil, Brassica juncea (L.) Czern. was grown first, followed by radish (Raphanus sativus L.). For Valencia soil, Beta maritima L. was followed by radish. Bioavailability of metals was expressed in terms of concentrations extractable with 0.1 M CaCl2 or diethylenetriaminepentaacetic acid (DTPA). In the Murcia soil, heavy metal bioavailability was decreased more greatly by manure than by the highly-humified compost. EDTA (2 mmol kg(-1) soil) had only a limited effect on metal uptake by plants. The metal-solubilising effect of EDTA was shorter-lived in the less contaminated, more highly calcareous Valencia soil. When correlation coefficients were calculated for plant tissue and bioavailable metals, the clearest relationships were for Beta maritima and radish.  相似文献

Rainbow pink (Dianthus chinensis), a potential phytoextraction plant, can accumulate high concentrations of Cd from contaminated soils. Vetiver grass (Vetiver zizanioides) has strong and long root tissues and is a potential phytostabilization plant since it can tolerate and grow well in soils contaminated with multiple heavy metals. Soil was moderately artificially contaminated by cadmium (20 mg/kg), zinc (500 mg/kg), and lead (1000 mg/kg) in pot experiments. Three concentrations of Na2-EDTA solution (0, 5, and 10 mmol/kg soil) were added to the contaminated soils to study the influence of EDTA solution on phytoextraction by rainbow pink or phytostabilization by vetiver grass. The results showed that the concentrations of Cd, Zn, and Pb in a soil solution of rainbow pink significantly increased following the addition of EDTA (p < 0.05). The concentrations of Cd and Pb in the shoots of rainbow pink also significantly increased after EDTA solution was applied (p < 0.05), but the increase for Zn was insignificant. EDTA treatment significantly increased the total uptake of Pb in the shoot, over that obtained with the control treatment (p < 0.001), but it did not significantly increase the total uptake of Cd and Zn. The concentrations of Zn and Pb in the shoots of rainbow pink are significantly correlated with those in the soil solution, but no relationship exists with concentrations in vetiver grass. The toxicity of highly contaminating metals did not affect the growth of vetiver grass, which was found to grow very well in this study. Results of this study indicate that rainbow pink can be considered to be a potential phytoextraction plant for removing Cd or Zn from metal-contaminated soils, and that vetiver grass can be regarded as a potential phytostabilization plant that can be grown in a site contaminated with multiple heavy metals.  相似文献

A better understanding of metal uptake and translocation by aquatic plants can be used to enhance the performance of constructed wetland systems for stormwater treatment. Specifically, this study examines whether the uptake of Zn, Cu, Cd, and Pb by Potamogeton natans is via the leaves, stems, or roots, and whether there is translocation from organs of uptake to other plant parts. Competition between the metals at uptake and at the level of the cell wall-bound part of the metals accumulated in stem and leaf tissue was also examined. The results show that Zn, Cu, Cd, and Pb were taken up by the leaves, stems, and roots, with the highest accumulation found in the roots. At the elevated metal concentrations common in stormwater the uptake of Cu, but not of Zn, Cd, or Pb, by the roots was somewhat limited at uptake due to competition with other metals. Between 24% and 59% of the metal content was bound to the cell walls of the plant. Except in the case of Pb, the cell wall-bound fraction was generally smaller in stems than in leaves. No translocation of the metals to other parts of the plant was found, except for Cd which was translocated from leaf to stem and vice versa. Dispersion of metals from sediment to water through P. natans is therefore unlikely.  相似文献

Metal-contaminated soils in the vicinity of industrial sites become of ever-increasing concern. Diagnostic criteria and ecological technologies for soil remediation should be calibrated for various soil conditions; actually, our knowledge of calcareous soil is poor. Silty soils near smelters at Evin (Pas de Calais, France) have been contaminated by non-ferrous metal fallout and regularly limed using foams. Therefore, the mobility, bioavailability, and potential phytotoxicity of Cd, Pb and Zn, were investigated using single soil extractions (i.e. water, 0.1 n Ca(NO(3))(2), and EDTA pH 7), and vegetation experiments, in parallel with a biological test based on (iso)-enzymes in leaves and roots, before and following soil treatment with chemical agents, i.e. Thomas basic slags (TBS), hydrous manganese oxide (HMO), steel shots (ST) and beringite. No visible toxicity symptoms developed on the above-ground parts of ryegrass, tobacco and bean plants grown in potted soil under controlled environmental conditions. Cd, Zn and Pb uptake resulted in high concentrations in the above-ground plant parts, but the enzyme capacities in leaves and roots, and the peroxidase pattern indicated that these metal concentrations were not phytotoxic for beans as test plants. The addition of chemical agents to the soil did not increase biomass production, but treatment with either HMO, ST or beringite markedly decreased the mobility of Cd, Zn and Pb. These agents were proven to be effective in mitigating the Cd uptake by plants. HMO and ST decreased either Pb or Zn uptake by ryegrass. TBS was effective in lowering Pb uptake by the same species. Beringite decreased Cd uptake by beans. If fallout could be restricted, the metal content of food crops in this area should be lowered by soil treatment. However, the differences in Cd uptake between plant species were not suppressed, regardless of the type of agents applied to the soil.  相似文献

In a pot experiment, the potential use of 10 plant species, including six dicotyledon species and four monocotyledon species, was investigated for the EDTA-enhanced phytoextraction of Pb from contaminated soil. Mung bean and buckwheat had a higher sensitivity to the EDTA treatment in soils. In the 2.5 and 5.0 mmol kg(-1) EDTA treatments, the Pb concentrations in the shoots of the six dicotyledon species ranged from 1,000 to 3,000 mg kg(-1) of dry matter, which were higher than those of the monocotyledon species. The highest amount of phytoextracted Pb (2.9 mg Pb pot(-1)) was achieved in sunflowers, due to the high concentration of Pb in their shoots and large biomass, followed by corns (1.8 mg Pb pot(-1)) and peas (1.1 mg Pb pot(-1)). The leaching behavior of heavy metals as a result of applying EDTA to the surface of the soil was also investigated using short soil-leaching columns (9.0-cm diameter, 20-cm height) by the percolation of artificial rainfall. About 3.5%, 15.8%, 13.7% and 20.6% of soil Pb, Cu, Zn and Cd, respectively, were leached from the soil columns after the application of 5.0 mmol kg(-1) of EDTA. The growth of sunflowers in the soil columns had little effect on the amount of metals that were leached out. This was probably due to the shallowness of the layer of soil, the short time-span of the uptake of metals by the plant and the plant's simple root systems.  相似文献

Chemically enhanced phytoextraction has been proposed as an effective approach to removing heavy metals from contaminated soil through the use of high biomass plants. Using pot experiments, the effects of the application of EDTA, EDDS and citric acid on the uptake of Cu, Pb, Zn and Cd by corn (Zea mays L. cv. Nongda 108) and bean (Phaseolus vulgaris L. white bean) plants were studied. The results showed that EDDS was more effective than EDTA at increasing the concentration of Cu in corn and beans. The application of 5 mmol kg-1 soil EDDS to soil significantly increased concentrations of Cu in shoots, with maximum levels of 2060 and 5130 mg kg-1 DW in corn and beans, respectively, which were 45- and 135-fold higher than that in the corresponding control plants to which chelate had not been applied. Concentrations of Zn in shoots were also higher in the plants treated with EDDS than in those treated with EDTA. For Pb and Cd, EDDS was less effective than EDTA. The maximum Cu phytoextraction was found with the EDDS treatment. The application of EDTA and EDDS also significantly increased the shoot-to-root ratios of the concentrations of Cu, Pb, Zn and Cd in both plant species. The results of metal extraction with chelates showed that EDDS was more efficient at solubilizing Cu and Zn than EDTA, and that EDTA was better at solubilizing Pb and Cd than EDDS.  相似文献

Ethylenediamene tetraacetic acid (EDTA) has been used to mobilize soil lead (Pb) and enhance plant uptake for phytoremediation. Chelant bound Pb is considered less toxic compared to free Pb ions and hence might induce less stress on plants. Characterization of possible Pb complexes with phytochelatins (PCn, metal-binding peptides) and EDTA in plant tissues will enhance our understanding of Pb tolerance mechanisms. In a previous study, we showed that vetiver grass (Vetiveria zizanioides L.) can accumulate up to 19,800 and 3350 mg Pb kg−1 dry weight in root and shoot tissues, respectively; in a hydroponics set-up. Following the basic incubation study, a greenhouse experiment was conducted to elucidate the efficiency of vetiver grass (with or without EDTA) in remediating Pb-contaminated soils from actual residential sites where Pb-based paints were used. The levels of total thiols, PCn, and catalase (an antioxidant enzyme) were measured in vetiver root and shoot following chelant-assisted phytostabilization. In the presence of 15 mM kg −1 EDTA, vetiver accumulated 4460 and 480 mg Pb kg−1 dry root and shoot tissue, respectively; that are 15- and 24-fold higher compared to those in untreated controls. Despite higher Pb concentrations in the plant tissues, the amount of total thiols and catalase activity in EDTA treated vetiver tissues was comparable to chelant unamended controls, indicating lowered Pb toxicity by chelation with EDTA. The identification of glutathione (referred as PC1) (m/z 308.2), along with chelated complexes like Pb-EDTA (m/z 498.8) and PC1-Pb-EDTA (m/z 805.3) in vetiver root tissue using electrospray tandem mass spectrometry (ES-MS) highlights the possible role of such species towards Pb tolerance in vetiver grass.  相似文献

Brassica species are very effective in remediation of heavy metal contaminated sites. Lead (Pb) as a toxic pollutant causes number of morphological and biochemical variations in the plants. Synthetic chelator such as ethylenediaminetetraacetic acid (EDTA) improves the capability of plants to uptake heavy metals from polluted soil. In this regard, the role of EDTA in phytoextraction of lead, the seedlings of Brassica napus L. were grown hydroponically. Lead levels (50 and 100 μM) were supplied alone or together with 2.5 mM EDTA in the nutrient culture. After 7 weeks of stress, plants indicated that toxicity of Pb caused negative effects on plants and significantly reduced growth, biomass, chlorophyll content, gas exchange characteristics, and antioxidant enzymes activities such as superoxide dismutase (SOD), guaiacol peroxidase (POD), ascorbate peroxidase (APX), and catalase (CAT). Exposure to Pb induced the malondialdehyde (MDA), and hydrogen peroxide (H2O2) generation in both shoots and roots. The addition of EDTA alone or in combination with Pb significantly improved the plant growth, biomass, gas exchange characteristics, chlorophyll content, and antioxidant enzymes activities. EDTA also caused substantial improvement in Pb accumulation in Brassica plants. It can be deduced that application of EDTA significantly lessened the adverse effects of lead toxicity. Additionally, B. napus L. exhibited greater degree of tolerance against Pb toxicity and it also accumulated significant concentration of Pb from media.  相似文献

Chemically enhanced phytoextraction is achieved by the application of chelates to soils. Using pot experiments, the effect of the combined application of EDTA and EDDS on the uptake of Cu, Pb, Zn and Cd by Zea mays L. was studied. Among the tested application ratios of 1:1, 1:2, and 2:1 (EDTA/EDDS), 2:1 of EDTA:EDDS was the most efficient ratio for increasing the concentrations of Cu, Pb, Zn and Cd in the shoots. The combined application of 3.33 mmol kg(-1) soil of EDTA+1.67 mmol kg(-1) soil of EDDS produced 650 mg kg(-1) of Pb in the shoots, which was 2.4 and 5.9 times the concentration of Pb in the shoots treated with 5 mmol kg(-1) of EDTA and EDDS alone, respectively. The total phytoextraction of Pb reached 1710 microg kg(-1) soil, which was 2.1 and 6.1 times the total Pb from 5 mmol kg(-1) EDTA and EDDS alone, respectively. The combined application of EDTA and EDDS also significantly increased the translocation of Pb from the roots to the shoots. The mechanism of enhancing the phytoextraction of Pb by the combined application of EDTA+EDDS did not involve a change in the pH of the soil. The increase in the phytoextraction of Pb by the shoots of Z. mays L. was more pronounced than the increase of Pb in the soil solution with the combined application of EDTA and EDDS. It was thought that the major role of EDDS might be to increase the uptake and translocation of Pb from the roots to the shoots of plants.  相似文献

Few studies have carried out soil washing experiments using pot experiments to simulate in situ soil washing operations, particularly for alkaline soils. This study explored the effects of multiple washing operations using pot experiments on the removal efficiencies of potentially toxic metals (PTM) from alkaline farmland soil and the reuse strategy of washed soil for safe agricultural production. The results showed that the removal efficiencies of Cd, Pb, Cu, and Zn after seven washings with a mixed chelator (EDTA, GLDA, and citric acid) were 41.1%, 47.1%, 14.7%, and 26.5%, respectively, which was close to the results of the EDTA treatment. For the alkaline soil studied, the second washing with the mixed chelators most effectively removed PTM owing to the activation of them after the first washing operation. The mixed chelator more effectively increased the proportion of stable fraction of PTM and maintained soil nutrients (e.g., nitrogen content) than EDTA, indicating little disturbance of alkaline soil quality after washing with the mixed chelator. After the amendment of the washed soil, there was no visible difference in the biomass weight of crops from the soils washed with different agents, indicating that the inhibitory effect of both washing agents on plant growth was effectively alleviated. The Cd and Pb contents in Z. mays were below the threshold of Hygienical Standard for Feeds of China (GB 13078–2017) (1 and 30 mg·kg?1). Moreover, after three cropping operations, the available concentrations of PTM in the soil washed with the mixed chelator were lower than those in the soil washed with EDTA, indicating the value and potential of agricultural reuse of alkaline farmland soil washed with the mixed chelator.

The aim of the present work was to study the response of two sunflower genotypes (cultivated sunflower Helianthus annuus cv. 1114 and newly developed genotype H. annuus?×?Helianthus argophyllus) to Pb medium-term stress and the role of exogenously applied EDTA in alleviating Pb toxicity in hydroponics. Plant growth, morpho-anatomical characteristics of the leaf tissues, electrolyte leakage, total antioxidant activity, free radical scavenging capacity, total flavonoid content, and superoxide dismutase isoenzyme profile were studied by conventional methods. Differential responses of both genotypes to Pb supplied in the nutrient solution were recorded. Pb treatment induced a decrease in the relative growth rate, disturbance of plasma membrane integrity, and changes in the morpho-anatomical characteristics of the leaf tissues and in the antioxidant capacity, which were more pronounced in the cultivated sunflower H. annuus cv. 1114. The new genotype demonstrated higher tolerance to Pb when compared with the cultivar. This was mainly due to increased photosynthetically active area, maintenance of plasma membrane integrity, permanently high total antioxidant activity, and free radical scavenging capacity as well as total flavonoid content. The addition of EDTA into the nutrient solution led to limitation of the negative impact of Pb ions on the above parameters in both genotypes. This could be related to the reduced content of Pb in the roots, stems, and leaves, suggesting that the presence of EDTA limited the uptake of Pb. The comparative analysis of the responses to Pb treatment showed that the deleterious effect of Pb was more pronounced in the cultivated sunflower H. annuus cv. 1114. The new genotype H. annuus?×?H. argophyllus was more productive and demonstrated higher tolerance to Pb medium-term stress, which could indicate that it may possess certain mechanisms to tolerate high Pb concentrations. This character could be inherited from the wild parent used in the interspecific hybridization. The ability of EDTA to prevent Pb absorption by the plants could underly the mechanism of limiting of the negative impact of Pb ions. Hence, EDTA cannot be used to enhance Pb absorption from nutrient solution by sunflower plants for phytoremediation purposes.  相似文献

The phytoextraction process was conducted under laboratory conditions with the use of garden cress plants (Lepidium sativum). The experiment was carried out in a model soil, which was characterized before conducting the process. Inorganic forms of mercury (HgCl(2), HgSO(4), Hg(NO(3))(2)) were used for contamination of the soil. The phytoextraction process was conducted after EDTA application to the soil and after urease application. Also the influence of simultaneous addition of ethylenediaminetetraacetic acid (EDTA) and urease into the soil on phytoextraction process was measured. In all variants of phytoextraction process the total mercury concentrations in roots, stems and leaves of garden cress were determined. The result showed that garden cress accumulated mercury from soil. The overall maximum concentration of mercury in its compounds was found in roots of the plant. In all cases, before addition of urease and EDTA, the translocation process and distribution of mercury in the plant tissues were limited. The addition of urease caused an increase of enzyme activity in the soil and at the same time caused an increase of mercury concentration in plant tissues. Application of EDTA increased solubility of mercury and caused an increase of metal accumulation by plants. After simultaneous addition of EDTA and urease into the soil garden cress accumulated about 20% of total mercury concentration in the soil. Most of mercury compounds were accumulated in leaves and stems of the plants (46.0-56.9% of total mercury concentration in the plant tissues).  相似文献
