Exposure of the tropical arcid blood clam Anadara granosa L., collected from Bombay waters in 1986, to sublethal concentrations of naphthalene (Nap) (5, 10 and 15g Nap ml-1), induced various changes in vital biochemical systems. The condition index (CI) remained largely within the control range, although tissue water content after 96h exposure increased significantly (p<0.001). Glycogen levels appreciably decreased, both in muscles (p<0.001) and digestive glands (p<0.01). AMP activity remained unaltered irrespective of the ambient Nap concentration. ADP levels decreased by more than 20% and ATP levels also decreased significantly (p0.001) by more than 60% with increases in ambient Nap levels from 5 to 15 g ml-1. Lysosomal marker enzymes-arylsulphatase and acid phosphatase-showed various changes. Levels of the former were 10 to 17 times higher than the latter. Total and free arylsulphatase activity in hepatic cells decreased on exposure of clams to 5 and 10g Nap ml-1, but not significantly. However, it was significantly inhibited (p<0.001) towards the upper limits of exposure concentration. The same was also true of acid phosphatase activity. Nicotinamide adenine dinucleotide (NADH2) oxidation of lipochrome showed a dramatic decrease in activity in the range of 37 to 88% over controls with increasing Nap from 5 to 15 g ml-1. Fatty acids also exhibited significant changes in composition with a decrease in polyunsaturated acids and an increase in certain saturated fatty acids. Protein and carbohydrate levels increased significantly (p<0.001) towards upper limits of exposure concentration. Pyruvic acid levels increased significantly by 2 to 6 times over controls on exposure to 5 to 15 g Nap ml-1. Explanation of these changes is attempted in terms of the glycolysis pathway involving pyruvic acid oxidation and oxidative phosphorylation in the electron transport system.  相似文献

Excessive fluoride consumption leads to accelerated red blood cell death and anaemia. Whether that increases the haematological alteration in subjects with haematological disorders (iron deficiency, thalassaemia, and G-6-PD deficiency) is still unclear. The fluoride in serum and urine and haematological parameters of students at Mae Tuen School (fluoride endemic area) were analysed and compared to those of students at Baan Yang Poa and Baan Mai Schools (control areas). Iron deficiency, thalassaemia, and G-6-PD deficiency were also diagnosed in these students. The students at Mae Tuen School had significantly (P < 0.001) higher levels of mean fluoride in the serum and urine than those in control areas. In both control and fluoride endemic areas, students with haematological disorders had significantly lower levels of Hb, Hct, MCV, MCH, and MCHC than those without haematological disorders. Moreover, the lowest levels of Hb, MCH, and MCHC were observed in the students with haematological disorders who live in the fluoride endemic area. Thus, the excessive fluoride consumption increased haematological alteration in subjects with iron deficiency, thalassaemia, and G-6-PD deficiency and that may increase the risk of anaemia in these subjects.  相似文献

Seasonal variations of oxygen consumption rate, haemolymph osmolality and the concentrations of the inorganic ions potassium and sodium in the haemolymph were measured in the littoral amphipod Gammarus oceanicus collected from the Trondheimsfjorden, Norway in 1987. For each season comparisons were made of amphipods acclimated for 1 wk to 0.5, 4.5, 10.0, 15.0 and 20.0°C, in combination with seawater osmolalities of 100, 500 and 1200 mOsm and to the seawater osmolality corresponding to that of the collecting site. The oxygen consumption rate showed a temperature insensitivity when the amphipods were acclimated to low temperatures in winter and high temperatures in summer. Significant differences were found in oxygen consumption between individuals acclimated to various medium osmolalities, possibly indicating higher energy requirements for osmotic and ionic regulation at low seawater osmolalities. Oxygen consumption rate was significantly higher in summer than in other seasons. Haemolymph osmolality and the concentration of the inorganic ions sodium and potassium were not influenced by temperature or season. Determination of haemolymph osmolalities and concentrations of inorganic ions revealed that G. oceanicus is a strong hyper-osmotic and hyper-ionic regulator in dilute seawater. The concentration of potassium in the haemolymph is less influenced by seawater osmolality than haemolymph osmolality and the haemolymph concentration of sodium.  相似文献

The effect of cadmium introduced as inorganic salts (chloride, nitrate, sulfate, carbonate, acetate and iodide) or organic chelates (with EDTA, NTA and humic acid) on the physiological behaviour of six species of tropical lamellibranchs — arcid blood clamsAnadara granosa andA. rhombea, venerid clamsMeretrix casta andKatelysia opima, and mytilid musselsPerna viridis andP. indica, collected from various intertidal areas in southern India from 1986 to 1989 — was evaluated. Inorganic salts of Cd are reported to completely ionize upon introduction to full-strength sea water and form predominantly (97%) chloride complexes. Some inorganic Cd salts, therefore, would hardly affect physiological parameters. However, bioaccumulation of Cd was greatest upon exposure to Cd introduced as sulfate, followed by iodide, acetate, chloride, nitrate and carbonate; 96-h LC50's also varied, but in a different order, being highest for chloride (3.5µg ml–1), followed by nitrate, acetate, iodide and sulfate (1.8µg ml–1). Furthermore, Cd levels increased linearly with time of exposure and levels in medium, and were species-specific, being highest in the arcid clams, followed by the venerid clams and mytilid mussels. The presence of other metals such as Zn reduced bioaccumulation of Cd significantly (P < 0.001), but the reverse was not true. Upon exposure to Cd in the presence of Cu, on the other hand, accumulation of both metals was reduced by 15 to 20%. In the presence of organic chelators — EDTA, NTA and reduced glutathione (GSH) — Cd accumulation was significantly inhibited, by 1/2 to 1/3. Humic acid, on the other hand, had no effect. Cd levels in soft tissues were inversely related to ambient salinity in the range of 12 to 32. However, both Zn and Cu significantly inhibited Cd accumulation irrespective of salinity. Cd increased the rate of filtration inAnadara granosa over the first 24 h by 28%, but on prolonged exposure (96 h) that rate decreased significantly, by 50%. Zn alone and in the presence of Cd, on the contrary, initially reduced filtration activity, which subsequently increased to control levels upon continued exposure. Cu alone and in combination with Cd and Zn also significantly (P < 0.001) inhibited filtration relative to controls. This combination also progressively reduced respiratory activity with time, the maximum decrease being 80% after 96 h of exposure. Furthermore, impact of all three metals on both filtration rate and oxygen consumption was initially synergistic, but changed to antagonistic upon prolonged exposure up to 96 h.Deceased. A haemolymph colour index is developed in an attempt to improve the resolution of serum protein data in the characterisation of temporal and spatial changes in the condition and growth of a wild population of Jasus edwardsii. The index can be used as an indicator of nutritional condition if combined with conventional moult staging techniques. Lobsters and haemolymph samples were collected from two high- and two low-growth sites over two fishing seasons. Haemolymph samples were analysed for serum protein and astaxanthin level and categorised according to colour, that is, "pigment stage" (PS). Moult stage data were collected and abdominal and hepatopancreatic tissue analysed for percent dry weight. Haemolymph colour changes from light blue-grey, through beige, to deep orange during the moult cycle. These changes were explained with reference to the major pigment, astaxanthin, which increased from 0.135 mg/l (ǂ.054, n=38) at PS 1 (early intermoult) to 2.670 mg/l (ǂ.599, n=12) at PS 4.5 (late premoult). There were significant increases in percent abdominal and hepatopancreatic dry tissue weight over the moult cycle (Kruskal-Wallis non-parametric ANOVA, P<0.05), especially during intermoult. Serum protein levels increased concomitantly and were significantly correlated with percent dry weight of both tissues (abdomen: r2=0.78, n=871, P<0.001; hepatopancreas: r2=0.64, n=864, P<0.001) There were also significant differences between sites in both PS-specific serum protein and percent dry tissue. Lobster condition differed significantly between sites, probably as a result of temperature-mediated effects on growth rate. The addition of haemolymph pigment to the serum protein index allows the differentiation of lobsters at the beginning, middle and end of intermoult.  相似文献

The activities of extracellular enzymes are important in understanding decomposition of soil organic matter in wetlands subjecting to drying. The activity of soil extracellular enzymes (β-glucosidase, N-acetylglucosaminidase, and phosphatase), and related physicochemical parameters were monitored in constructed freshwater wetlands during a one-month drying manipulation. Drying increased redox potential and decreased soil water content significantly (P<0.05). Higher content of soil organic matter (P<0.05) and higher concentrations of inorganic N (nitrate, P<0.01; and ammonia, P<0.001) were also observed significantly under drying condition. Soil hydrolase enzyme activities were stimulated significantly (β-glucosidase, P<0.05; N-acetylglucosaminidase, P<0.01; and phosphatase, P<0.001), and a two-phase pattern of enzyme activities was revealed under drying condition. The increase of soil enzyme activities under drying condition was significantly related to soil redox potential (P<0.001). Drying strongly affected soil enzyme activities only when soil water content remained above an optimal level for enzymatic catalysis (higher than 23% w/w), corresponding to redox potentials below 250–300 mV. Our results suggest that, under drying condition, potential enzyme activities may be regulated by redox potential, in respect to soil moisture, and consequently alter nutrient availability in wetlands.

The effect of sublethal concentration (29 mg/l) of cadmium on haematological parameters of Channa punctatus after the exposure periods of 7, 15 and 30 days were studied. In treated groups of cadmium exposed fish have been found to significantly decreased in total erythrocyte count, haemoglobin content, packed cell volume, mean corpuscular haemoglobin concentration and oxygen carrying capacity of blood, and at the same time the white blood cell count, mean cell volume and mean corpuscular haemoglobin were found to significantly increased. Both increasing and decreasing of the above mentioned haematological characteristics of blood cells of cadmium intoxicant are mainly with time dependent effects. With regard to the pathological characteristics of blood cells of cadmium intoxicant fish shows fragility and rupture of erythrocytic membrane and leads to haemolysis, which are very severe on 30 day of exposure group.  相似文献

The oxygen-binding properties of haemolymph from laboratory-reared Artemia franciscana were investigated in vitro. Adult female brine shrimp without eggs were acclimated to seven different combinations of salinity and oxygen. The oxygen affinity (P50) of unprocessed haemolymph rises with acclimation oxygen partial pressure (PO2) up to normoxic values, after which no influence of oxygen occurs. The increase in P50 with acclimation PO2 can be explained by the change in proportion of each of three different haemoglobins in the haemolymph. Salinity acclimation has no effect on haemolymph P50. The effects of the different major salts [NaNO3, NaCl, Ca(NO3)2, Mg(NO3)2], pH, and the metal-binding ligand EGTA on the oxygen-binding properties of buffered haemolymph (of shrimp acclimated to 10 salinity) were also studied. Little or no effect of these salts could be found. A small Bohr effect (pH 6.5 to 9.1, =-0.11) was noted. Addition of EGTA caused a significant decrease of the oxygen affinity at concentrations up to 50 mmoll-1.  相似文献

The concentration of glutamine in Tridacna gigas haemolymph increased >35-fold following exposure to sea water supplemented with ammonium (20 μM), but no increase was observed with nitrate (20 μM). Lack of a diel cycle, no decrease in haemolymph glucose levels, the expression patterns of glutamine synthetase in zooxanthellae and host, and the lack of glutamine release in response to nitrate supplementation all support the proposition that the increase in haemolymph glutamine is a product of the host and not the zooxanthellae. Unlike ammonium, nitrate accumulates rapidly in the haemolymph. It has no effect on the concentration of glutamine in the haemolymph, but there is an increase in arginine, histidine and lysine in the haemolymph, suggesting the release of these essential amino acids from zooxanthellae. Glutamine synthetase (GS) activity decreased markedly in the gill and less so in the mantle over a period of 6 d exposure to elevated ammonium (20 μM). In contrast, GS activity in zooxan- thellae doubled. The response of zooxanthellae in situ was confirmed by incubating freshly isolated zooxanthellae for 4 d in ammonium, which resulted in a ten-fold increase in GS activity. Comparison of the in situ response of zooxanthellae with that obtained in vitro indicates that the symbionts are likely to be exposed to ammonium concentrations lower than that found in the haemolymph. Changes in vital physiological activities — shell movements and burrowing behaviour, uptake and depuration, filtration and respiration — on exposure to sublethal concentrations of naphthalene (Nap) were investigated in the tropical arcid blood clam Anadara granosa L. from the Bombay coast in 1986. On exposure to ambient concentrations exceeding 5g Nap ml-1, shell valves opened widely within the first hour of exposure. The compressed muscular foot was stretched vertically upwards with copious secretion of mucus, and did not exhibit any evidence of burrowing behaviour. Those exposed to 5g Nap ml-1 regained their normal physiological activities on transfer to stressor-free medium, whereas those exposed to higher levels became intoxicated and narcotized. Bioaccumulation of Nap was dependent upon environmental concentration, increasing exponentially with time over a 9h exposure period. Further exposure, up to 96h, however, did not increase tissue levels substantially. About 65% of the total body burden of Nap was depurated within 3h of transfer to Nap-free medium. Rates of filtration and oxygen consumption were significantly reduced (ca 70%, p<0.001) compared to control clams. Percent inhibition in these physiological activities was dependent upon tissue and ambient concentrations of Nap. On transfer to pollutant-free medium, clams exhibited remarkable recovery. Rates of both filtration and oxygen consumption were gradually increased and restored to normal levels, as observed in controls. However, clams exposed to upper limits lost their ability to burrow back into the sedimentary bed, leaving them susceptable to predators. Furthermore, induction of anaerobiosis and disruption of osmotic balance on exposure to Nap together with aerial exposure at low-water periods and salinity changes, acted synergistically and proved detrimental. The rates of growth and mortality observed in the natural population of blood clams, harvested from the coastal waters off Bombay, have been explained in terms of the impact of petroleum and allied waste products released from petrochemical industries.  相似文献

Specimens of the West African blood clam Anadara senilis were exposed to both sinusoidal and abrupt salinity regimes. Measurements showed that haemolymph osmolality and concentrations of Na, K+, Ca2+ and Mg2+ followed the concentrations of the external medium up to the time of shell valve closure. Shell valves closed when the seawater concentration had fallen to about 15.4% S and reopened at a similar salinity. The closure mechanism was effective in preventing excessive haemolymph dilution.  相似文献

Mytilus edulis L. were exposed to abrupt (3015 and 1530) and fluctuating (sinusoidal 12 h cycles of 301530) changes in salinity, and the changes in the total osmoconcentration of the haemolymph were recorded. The response of nitrogen metabolism to the altered extracellular osmotic concentrations was investigated in terms of the concentrations of the total NPS (ninhydrin-positive substances) pool and the individual amino acids of the tissues, the concentration of the amino acids of the haemolymph, and the rates of excretion of ammonia and amino acids by whole individuals. The haemolymph became isosmotic with the seawater with abrupt changes in salinity, but with fluctuating salinity was slightly hyperosmotic as the salinity decreased and then slightly hypo-osmotic as the salinity increased. This resulted in a reduction in the extent of the extracellular osmotic change compared to the change in fluctuating salinity to which it was exposed. Total NPS of the tissues decreased with an abrupt decrease in salinity and increased with an abrupt increase in salinity, but a seasonal dependence of the response was indicated. The short-term response of tissue NPS to fluctuating salinity was equivocal, but with long-term exposure the concentration declined. Ammonia and amino acid excretion increased with both an abrupt decrease in salinity and fluctuating salinity and decreased with an abrupt increase in salinity. Haemolymph amino acids increased with an abrupt decrease in salinity. The increased rates of nitrogen excretion accounted for the reductions in the NPS concentrations of the tissues except in the early stages of fluctuating salinity. Taurine, aspartate, threonine, serine, glycine and arginine declined with an abrupt decrease in salinity while alanine and glutamate increased slightly. With an abrupt increase in salinity, alanine and ammonia accumulated in the tissues and then declined while the other amino acids increased slowly over a longer time-course. Similar individual amino acid responses were seen with long-term exposure to fluctuating salinity, except for taurine which did not decrease in concentration. On the basis of the changes in tissue amino acids and ammonia, it is suggested that the alanine dehydrogenase reaction is the primary nitrogen-fixing reaction in marine bivalves such as M. edulis.  相似文献

Vertical zonation of Nerita plicata Linnaeus, N. undata Linnaeus, and N. textilis Dillwyn, inhabiting the rocky cliffs of Nyali, Kenya, was studied through several spring-neap tidal cycles from November to December (northeastern monsoon) 1981 and May to July (southeastern monsoon) 1982. For the first time, a monsoon-related cycle of migration in Nerita plicata and N. undata, superimposed on the shorter term spring-neap cycle is demonstrated. Both species rest predominantly in the upper eulittoral zone and the littoral fringe. N. plicata and N. undata rest at significantly higher levels during the southeastern monsoon (SEM) than during the northeastern monsoon (NEM) (P<0.001). On and around neap-tide days N. plicata, N. undata, and N. textilis occupy significantly distinct mean resting positions (P<0.001) in both seasons; N. undata being the highest in the SEM, followed by N. plicata, with N. textilis always in the lowest level. In the NEM, N. plicata and N. undata sometimes change order. On and around springtide days in both seasons, the upper two species, N. plicata and N. undata, rest close together with no significant difference in their mean resting positions, but with both significantly higher than N. textilis (P<0.001). Both the spring-neap and the monsoon migrations of N. plicata and N. undata are interpreted as either direct or indirect responses to wave action.  相似文献

The oxygen-binding properties of haemocyanins (Hc) from three species of gammaridean amphipods, Gammarus locusta (L.) (subtidal), Echinogammarus pirloti (Sexton and Spooner), (intertidal, marine) and E. marinus (Leach) (intertidal, estuarine), one species of hyalid amphipod Hyale nilsonni Rathke (high intertidal, marine) and the talitrid amphipod Orchestia gammarellus (Pallas) (semi-terrestrial) have been studied. All the species were collected from the Firth of Clyde, Scotland, during the spring of 1992. The oxygen-carrying capacity of haemolymph from each species was low, although variable, and was correlated with the low concentration of Hc present. The Hc oxygen-affinity of native gammarid haemolymph was relatively high [partial pressure of oxygen required for half-saturation, P50=4 to 5 torr (0.53 to 0.67 kPa)] at their respective in vivo pH values. At equivalent pH, however, Hc from G. locusta displayed a lower O2-affinity than either Echinogammarus species. Gammarid Hcs had a large Bohr effect ( log P50/ pH=-1.16 to-1.47). Resuspended Hc isolated from whole H. nilsonni showed similar O2-binding properties to those of the gammaridean amphipods [P50=6.3 torr (1.44 kPa) at pH=8.0; log P50/pH=-1.20]. Comparable data for haemolymph from O. gammarellus showed that the Hc had a lower affinity for O2 [P50=14.1 torr (1.87 kPa) at in vivo pH] and exhibited a more moderate Bohr effect ( log P50/ pH=-0.79). To eliminate the possibility that these differences were due to the different haemolymph constituents, each of the Hcs were pelleted and resuspended in physiological saline. The differences noted above persisted, demonstrating that they were due to inherent O2-binding properties of the Hc molecules themselves. An increase in L-lactate resulted in an increase in Hc oxygen-affinity for both Echinogammarus species but not for O. gammarellus. This study has confirmed that there is a clear difference between Hcs from aquatic and semi-terrestrial amphipod genera. The results lend further support to the hypothesis that the move on to land by amphipod crustaceans is accompanied by a decrease in Hc oxygen-affinity, a decrease in the Bohr effect and a decrease in effector (in this case L-lactate) sensitivity.  相似文献

The lysosomal marker enzymes, arylsulfatase and acid phosphatase, in a tropical burrowing arcid clam Anadara granosa L. have been found to exhibit seasonal variations. The activity of both enzymes decreased with increase in ambient temperature and fell with increase in salinity. Lysosomal latency for these enzymes, however, was not significantly affected by environmental parameters, including salinity, temperature, nutritional status, breeding season, etc. The physico-chemical characteristics of the habitat were found to induce reversible changes in the lysosomal latency and enzyme activity. Exposure to elevated temperature (T=7° to 8°C), towards the upper limit of its physiological tolerance, was found to labilize the lysosomes. The biochemical methods optimized for the demonstration of lysosomal stability under exposure to environmental stressors could also be extended to quantify the impact of various pollutants, including heavy metals, radionuclides, etc. at the subcellular level.  相似文献

The Norway lobster, Nephrops norvegicus, is moderately tolerant of sulphide [the lethal time to 50% mortality (LT50) was 22.5 h when exposed to 500 M sulphide] but, whenever possible, it attempts to avoid the presence of sulphide in its immediate vicinity. Any sulphide entering the animal is oxidized to thiosulphate, which accumulates in the haemolymph and in the tissues. During exposure to low concentrations of sulphide, the rate of oxygen consumption is maintained or even enhanced even though the lobsters become quiescent. The apparent increase in oxygen consumption is probably due to the oxidation of sulphide to thiosulphate. At higher concentrations, oxygen consumption decreases, perhaps because of the inhibitory effect of sulphide on electron transport, and N. norvegicus resorts to anaerobic metabolism as indicated by the accumulation of lactate in the haemolymph and in the tissues.Communicated by L. The effect of ionizing radiation on the iron-linked protein (haemoglobin) of the marine lamellibranchs Anadara granosa (Linn.) and Cardita antiquata (Lam.) from Bombay waters, India is discussed. Purified haemoglobin solutions were exposed to a 60Cobalt source delivering a dose of about 4,600 rad/min, at the sample irradiation point. Radiation damage or degradation was measured spectrophometrically by studying changes in the absorption spectra following irradiation in the presence and absence of oxygen. Exposure to ionizing radiation in general caused a decrease in absorption of both haemoglobins, irrespective of location, viz extra-versus intracellular, at Soret (412 m), (540 to 42 m) and (574 to 76 m) peaks, and an increase in absorption at 510 and 630 m. Upon exposure to a higher dose, O2Hb of C. antiquata showed an increase in absorption at the protein peak (280 m); exposure to a lower dose, however, resulted in decreased absorption. Furthermore, the changes following irradiation were dependent upon the initial state of the pigment. Oxyhaemoglobin, when exposed to radiation, oxidized to hemiglobin, and hemiglobin reduced to oxyhaemoglobin. The extracellular haemoglobin of high molecular weight (3x106) of the false cockle C. antiquata was found to be extremely radio-resistant, whereas intracellular haemoglobin of low molecular weight (74,000) of the arcid clam A. granosa was highly radiosensitive, since it could not be exposed to doses exceeding 18,000 r.  相似文献

A study was conducted to determine the effects of long-term exposure to carbofuran on erythrocyte and liver acetylcholinesterase (AChE) activity in Cyprinus carpio, and assess which tissue was more sensitive to pesticide exposure examining this enzyme. Fish were allowed to accilimatize in toxicant-free water for 24 days after 60-day exposure. AChE activity was determined with a spectrophotometer using acetylthiocholine iodide as a substrate in erythrocytes and liver. Enzymic activity of erythrocytes and liver decreased over time. A higher degree of enzyme inhibition was observed in erythrocyte compared to liver. The degree of enzyme inhibition was positively correlated with exposure time. When exposed fish were transferred to clean water, recovery was greater in erythrocytes than liver. Erythrocyte AChE activity recovered after 18 days, while it required 21 days in liver. The findings of this study indicate that erythrocyte AChE is more sensitive to carbofuran than liver. The greater sensitivity of erythrocyte AChE suggests that it may be more useful as a biomarker for monitoring status of pollution in freshwater bodies.  相似文献

Hemolymph osmolality and tissue water of laboratory-reared Penaeus chinensis Osbeck juveniles (0.83 to 1.86 g) were investigated, after they had been transferred individually from 10, 20, 30 and 40 ppt to 10, 20, 30 and 40 ppt for 0.25, 0.5, 1, 2, 5 and 10 d, respectively. Hemolymph osmolality and tissue water of shrimp were stablilized within 5 d after they had been subjected to a sudden change in salinity from each salinity level. Hemolymph osmolality had a positively linear relationship with medium osmolality. Tissue water decreased with increased medium osmolality, and decreased with increased hemolymph osmolality. The mean (SD) isosmotic point was 703 (8) mOsm kg–1 which is equivalent to 24.2 (1.0) ppt. P. chinensis juveniles exhibited hyperosmotic regulation in salinities below isosmotic value, and hypoosmotic regulation in those above. The shrimp originally adapted to high salinity levels (30 and 40 ppt) showed less fluctuation of tissue water than those adapted to low salinity levels (10 and 20 ppt) when they were subjected to a sudden change in salinity.  相似文献
