Studies investigating disease resistance in marine plants have indicated that secondary metabolites may have important defensive functions against harmful marine microorganisms. The goal of this study was to systematically screen extracts from marine plants for antimicrobial effects against marine pathogens and saprophytes. Lipophilic and hydrophilic extracts from species of 49 marine algae and 3 seagrasses collected in the tropical Atlantic were screened for antimicrobial activity against five ecologically relevant marine microorganisms from three separate kingdoms. These assay microbes consisted of the pathogenic fungus Lindra thalassiae, the saprophytic fungus Dendryphiella salina, the saprophytic stramenopiles, Halophytophthora spinosa and Schizochytrium aggregatum, and the pathogenic bacterium Pseudoaltermonas bacteriolytica. Overall, 90% of all species surveyed yielded extracts that were active against one or more, and 77% yielded extracts that were active against two or more assay microorganisms. Broad-spectrum activity against three or four assay microorganisms was observed in the extracts from 48 and 27% of all species, respectively. The green algae Halimeda copiosa and Penicillus capitatus (Chlorophyta) were the only species to yield extracts active against all assay microorganisms. Among all assay microorganisms, both fungi were the most resistant to the extracts tested, with less than 21% of all extracts inhibiting the growth of either L. thalassiae or D. salina. In contrast, over half of all lipophylic extracts were active against the stramenopiles H. spinosa and S. aggregatum, and the bacterium P. bacteriolytica. Growth sensitivity to hydrophilic extracts varied considerably between individual assay microorganisms. While 48% of all hydrophilic extracts were active against H. spinosa, 27% were active against P. bacteriolytica, and only 14% were active against S. aggregatum. Overall, more lipophilic extracts inhibited microbial growth than hydrophilic extracts. The variability observed in the antimicrobial effects of individual extracts against each assay microorganism reflects the importance of choosing appropriate test microbes in assays from which ecologically relevant information is sought. Results from this survey demonstrate that antimicrobial activities are prevalent among extracts from marine algae and seagrasses, suggesting that antimicrobial chemical defenses are widespread among marine plants.  相似文献

Aqueous and flavonoid-enriched extract as well as essential oil (EO) obtained from leaves of Pistacia lentiscus were assessed for antibacterial and antimutagenic activities. Antibacterial activity of different extracts and EO were evaluated against six bacterial strains. A marked inhibitory effect was observed against Salmonella typhimurium, whereas lower activity was observed against Staphylococcus aureus, Pseudomonas aeruginosa, and Salmonella enteritidis. EO showed significant inhibitory effects against Salmonella typhimurium, Salmonella enteritidis and Staphylococcus aureus. The antimutagenic activity of the different extracts against Aflatoxin B1 (AFB1) and sodium azide was demonstrated with the Salmonella typhymurium assay. The number of revertants per plate decreased significantly when the plant extracts were added to the assay system using Salmonella typhimurium TA100, TA98 and TA1535.  相似文献

The use of microorganisms as biological control agents (BCAs) has become an effective alternative to chemical means of controlling plant pathogens. The antagonistic and inhibitory activity of 71 Bacillus spp. strains, which were isolated from different Mexican sites, were tested against several phytopathogen fungi, Fusarium oxysporum, Fusarium equiseti, Fusarium avenaceum, Bipolaris spp. and Alternaria spp. From the antagonism study, the strain ELI149 showed a marked inhibition of growth against all tested fungi; therefore crude metabolites from this strain were extracted using ethyl acetate and amberlite resin and probed against the same fungi as well as strains of Mucor sp., Penicillium spp. and Paecilomyces spp. The results indicated that amberlite was more suitable for extraction of secondary metabolites with antifungal activity. Finally, observation of cell damage in the tested pathogenic fungi showed marked morphological changes on reproductive structures in all tested fungi indicating that antibiosis was the mechanism of the antagonistic effect. These results suggest that metabolites from the Bacillus strains have a wide spectrum of antibiotic activities, which can be used as biocontrol agents for controlling fungal plant diseases of agricultural importance.  相似文献

The toxicity of copper and zinc to the estuarine copepod Acartia tonsa and to the two diatom food species Thalassiosira pseudonana and T. weissflogii was measured in nitrilotriacetate-trace metal ion buffer systems at 25 S. Overall, A. tonsa appeared to be more sensitive to cupric and zinc ion activity than either of the diatoms; however, its sensitivity varied among the different life stages examined. Adult survival was not affected within the zinc ion activity range 10-11 to 10-8 M and cupric ion activity range 10-13 to 10-11 M over a 96-h period, but a cupric ion activity of 10-10 M caused total mortality of adults within 72 h. Egg-laying rate was most sensitive to zinc, and was reduced at zinc ion activities 10-10 M. Naupliar survival after 96 h was reduced by zinc ion activities 10-8 M and by cupric ion activities 10-11 M, and was reduced to zero at a zinc ion activity of 10-7 M and at a cupric ion activity of 10-10.5 M. In an interspecies comparison of 96-h adult survival, Centropages typicus was more sensitive to copper and zinc than A. tonsa and the survival of Labidocera aestiva was dependent on the ratio of cupric to zinc ion activity. A comparison of our results with estimates of zinc and cupric ion activities in estuaries suggests that ionic activities of these metals are high enough in some polluted estuaries to affect the survival and reproduction of copepods.  相似文献

Lead-contaminated mine tailings were bioremediated using microbial/phyto remediation. The optimum lead accumulation and tolerance capacity of the plant–microbe partnership were investigated, and their mechanisms were evaluated further under varied levels of lead contamination through a flowerpot experiment in a greenhouse. Enzymes activities revealed that bioremediation has improved fertility and metabolism of tailing soil. The removal efficiency of lead was in the order of microbial/phytoremediation?>?phytoremediation. Solanum nigrum L. was not shown to be a hyperaccumulator for lead. Mucor circinelloides significantly enhanced the growth response and lead accumulation in plants more than Mortierella and Trichoderma asperellum. Moreover, Mortierella was discovered to have good metal tolerance capacity under high Pb concentrations (1200 and 1600?mg?kg?1). The results for lead bioavailability showed that phytostabilisation serves as a major repair pathway for S. nigrum L. Effective fractions were immobilised for decreased bioavailability by T. asperellum and M. circinelloides. On the contrary, an increased amount of lead was mobilised for increased bioavailability by Mortierella. This study provides new insights into the feasibility of using S. nigrum L. and the aforementioned indigenous fungus strains for large-scale bioremediation of mine tailings.  相似文献

This paper reports a study of the distribution of organo-chlorine pesticides (DDT and HCH) between rice plants and the soil system by spraying before the heading stage at four different dosage levels – control, normal dosage (15 kg ha–1 of 6% HCH and 7.5 kg ha–1 of 25% DDT), double dosage and four times dosage. Soil and plant samples were taken respectively at the 1st h, 3rd, 10th, 20th, and 40th day after spraying and at the harvest time. The results indicate that less than 5% of HCH and 15% of DDT were absorbed by the surface of rice leaves for normal dosage. Most of both pesticides moved into the soil in solution after spraying. Compared with DDT, HCH was degraded and run off more easily. HCH residues in the surface soil layer (1–3 cm) were already below 6.4 g kg–1 at the mature stage, lower than Chinese Environmental Quality Standard for Agricultural Soils: HCH <0.05 mg kg–1. However DDT residues in the surface soil layer remained 172 g kg–1, higher than the national standard: DDT <0.05 mg kg–1. According to the test f OCP residues in rice seeds, it can be concluded that the OCP sprayed onto the surface of rice leaves can move into rice plants and accumulate in the seeds at the mature stage. HCH residues in rice seeds of the double and four times dosage treatments, and DDT residues in all treatments, exceeded the Chinese National Food Standard (HCH <0.10 mg kg–1, DDT <0.20 mg kg–1).  相似文献

A total of 20 soil samples were collected from mining and non-mining areas of coal field regions of Dhanbad, India. These samples were grown on different types of selective media and a total of 141 isolates, each having plant growth-promoting (PGP) characteristics were obtained. The isolates were further tested for PGP traits (phosphate solubilisation, production of indole acetic acid, hydrogen cyanide, ammonia and siderophores) and a biochemical analysis was done to determine the prevalent species of the region. All the strains selected showed at least two PGP characteristics, which indicated that they can be used as plant growth promoters. Bacillus and Pseudomonas species were prevalent in both mining and non-mining areas, whereas Rhizobium, Azospirillum and Azotobacter were found mostly in the non-mining area. A total of 48 Bacillus, 29 Pseudomonas, 4 Rhizobium, 11 Azospirillum and 10 Azotobacter species were obtained from the two selected regions. The soil samples from the mining region were poor in nutrient content and thus relatively lower numbers of PGP isolates were attained.  相似文献

Summary. Hyperaccumulation of metals in the shoot system of plants is uncommon, yet taxonomically and geographically widespread. It may have a variety of functions, including defense against herbivores. This study investigated the effects of hyperaccumulation on metal concentrations across trophic levels. We collected plant material, soil, and invertebrates from Portuguese serpentine outcrops whose vegetation is dominated by the nickel hyperaccumulator Alyssum pintodasilvae. Samples were analyzed for nickel, chromium, and cobalt. Grasshoppers, spiders, and other invertebrates collected from sites where A. pintodasilvae was common had significantly elevated concentrations of nickel, compared to nearby sites where this hyperaccumulator was not found. Chromium and cobalt, occurring in high concentrations in the serpentine soil but not accumulated by A. pintodasilvae, were not elevated in the invertebrates. Therefore, it appears likely that a flux of nickel to herbivore and carnivore trophic levels is specifically facilitated by the presence of plants that hyperaccumulate this metal. The results may be relevant to the development of phytoremediation and phytomining technologies, which use plants to extract metals from the soil. Reveived 22 August 2002; accepted 2 April 2003. The contamination of soils by toxic and/or hazardous organic pollutants, especially with crude oil, is a widespread problem. This study was conducted in a petroleum-contaminated area in a Tehran oil refinery to find petroleum-resistant plants and their rhizospheral fungal strains with bioremediation potency. The plants growing in the oil-polluted area were collected and determined taxonomically. Root samples of the plant species were collected from a polluted area and fungal strains determined by laboratory methods and taxonomical keys. The growth ability of the isolated fungal strains was studied in media containing 1%–15% crude oil. Results showed that seven plant species were of the highest density in the contaminated area: Alhagi persarum, Hordeum marinum, Peganum harmala, Phragmites australis, Prosopis farcta, Salsola kali, and Senecio glaucus. The root-associated fungi were isolated and showed that the fungal variation in the oil-polluted area is higher than that in a non-polluted area. The growth assay of isolated fungal strains showed that all studied fungal strains were able to form colonies at the applied concentrations but Alternaria sp. and Rhizopus sp. were the most resistant ones. Some plants were resistant to oil pollution, which also had positive effects on the fungal strains.  相似文献

Bacterial strains with poly-3-hydroxybutyrate (PHB) degradation potential were isolated from waste yard soil samples of selected industrial sites in Uttarakhand, India, and two microbial consortia were developed, i.e. Consortium I comprises Pseudomonas sp. strain Rb10, Pseudomonas sp. strain Rb11 and Bacillus sp. strain Rb18, and Consortium II is composed of Lysinibacillus sp. strain Rb1, Pseudomonas sp. strain Rb13 and Pseudomonas sp. strain Rb19. The current study involved enrichment selection via liquid and semi-solid media, followed by isolation and screening of bacterial strains using PHB pellets and films. Furthermore, the identification and characterisation was done by triphasic approach. The utilisation of PHB by the characterised strains was confirmed by Fourier transform infrared spectroscopy and scanning electron microscopy. Moreover, the minimum inhibitory concentration of solubilised PHB was found to be 2.5?mg/mL, which was detected through ‘clear zone assay’. Further, the selection and biocompatibility testing of potential isolates were performed for the formation of bacterial consortia. Thus, the present work would provide direct and standardised protocol for screening and selection of potential microbiomes for biodegradation of polymers by overcoming the negative effect of organic solvents. Moreover, indigenously developed consortia would be evaluated for their in situ biodegradation potential against various bioplastic films.  相似文献

Plants growing in waterlogged environments are subjected to low oxygen levels around submerged tissues. While internal oxygen transport has been postulated as an important factor governing flooding tolerance, respiration rates and abilities to take up oxygen under hypoxic conditions have been largely ignored in plant studies. In this study, physiological characteristics related to internal oxygen transport, respiration, and oxygen affinity were studied in low intertidal marsh species (Spartina alterniflora and S. anglica) and middle to high intertidal species (S. densiflora, S. patens, S. foliosa, a S. alterniflora × S. foliosa hybrid, S. spartinae, and Distichlis spicata). These marsh plants were compared to the inland species S. pectinata and the crop species rice (Oryza sativa), corn (Zea mays), and oat (Avena sativa). Plants were grown in a greenhouse under simulated estuarine conditions. The low marsh species S. anglica was found to transport oxygen internally at rates up to 2.2 μmol O2 g fresh root weight−1 h−1. In contrast, marsh species from higher zones and crop species were found to transport significantly less oxygen internally, although rice plants were able to transport 1.4 μmol g−1 h−1. Under hypoxic conditions, low marsh species were better able to remove dissolved oxygen from the medium compared to higher marsh species and crops. The oxygen concentration at which respiration rates declined due to limited oxygen (P crit) was significantly lower in low marsh species compared to inland and crop species; P crit ranged from <4 μM O2 in the low marsh species S. anglica up to 20 μM in the inland species corn. Flooding-sensitive crop species had significantly higher aerobic respiration rates compared to flooding-tolerant species in this study. Crop species took up 3.6–6.7 μmol O2 g−1 h−1 while all but one marsh species took up <3.5 μmol O2 g−1 h−1. We conclude that oxygen transport, aerobic demand, and oxygen affinity all play important and interrelated roles in flood tolerance and salt marsh zonation.  相似文献

We used random amplified polymorphic DNA (RAPDs) to examine small-scale spatial genetic structure in the red alga Delisea pulchra (Greville) Montagne at two locations near Sydney, Australia. We examined genetic structure among plants at four spatial scales ranging from 2 km apart down to <50 cm apart between locations, among sites within locations, among quadrats within sites, and among plants within quadrats. Haploid stages of D. pulchra were absent from the populations studied, suggesting that they are maintained through asexual reproduction of diploid plants. Consistent with this, we found that 19 RAPD phenotypes scored in this study had multiple individuals, indicating the presence of clones in these populations. However, there were no RAPD phenotypes common to two locations separated by only 2 km. Analysis of molecular variance revealed that strong genetic differences occurred between plants from these two locations, with 46.3% of the total genetic variation occurring at this scale, most probably reflecting limited gene flow. Within each location, <25% of the genetic variation was attributable to differences among sites or quadrats, indicating gene flow at those smaller scales. Most of the variation within each location occurred at the smallest spatial scale, among plants within 0.25 m2 quadrats. Nonetheless, some pairwise genetic distances (φST) between sites or quadrats within locations were large, indicating some genetic divergence on smaller scales. Genetic distance was independent of spatial distance within both locations, suggesting that fine-scale differences within locations were most probably caused by variation in fine-scale patterns of water movement or fine-scale natural selection. We assessed the impact of one potential selective agent, grazing sea urchins, on the fine-scale genetic structure of D. pulchra. There was no evidence that grazing by sea urchins affected the genetic structure of D. pulchra. In combination with demographic data, our results indicated that local populations of D. pulchra within locations were relatively open and that fine-scale genetic structure was probably constrained by gene flow. At the larger scale however, strong genetic differentiation indicated little gene flow between locations and restricted dispersal of spores. This study evaluated the tolerance and accumulation potential in the mangrove Kandelia obovata under moderate and high levels of external Zn. A greenhouse experiment was conducted to investigate the effects of a range of external Zn concentrations (0–400 mg L?1) on the growth of this species by counting the lateral root number and measuring the root length, leaf area, and total dry biomass. We also determined the Zn accumulation in plant tissues. K. obovata survived with external Zn concentrations of up to 400 mg L?1, although the excess metal resulted in a biomass reduction of 34%. A significant increase in antioxidant enzyme activities occurred in roots of plants under high-level Zn stress, suggesting that K. obovata seedlings could tolerate up to 200 mg L?1 zinc treatment.  相似文献

Two microalgal strains (Chlorella sorokiniana, A&B,) grown optimally at pH 9 (A) and 7 (B) were isolated from a municipal wastewater treatment plant. These strains were used to check their efficacy for nutrients and organic carbon removal capacities from the tertiary wastewater. The strains were characterised using 18S rDNA sequencing method and both the strains closely related to Chlorella sorokiniana. Different inoculum doses (IDs) of both the strains were tried to check their efficacy on wastewater treatment. Due to the increase in the IDs, hydraulic retention time decreased from 6 d to 1?d. On an average, 12–100% of total kjeldahl nitrogen, 53–96% NO3–N, and 59–92% PO4–P, were removed. Despite the same growth environment, strain A with the highest ID (700?mg/L) showed the best performance in terms of overall organic carbon removal, nutrient removal, and value-added products recovery. Strain A showed an increase in carbohydrate and protein content by 42% and 13%, respectively. Both the strains showed luxury phosphorus uptake and found suitable for advanced wastewater treatment.  相似文献

Summary A crude cuticular extract from 3450 virgin 9–13 day old female fruit flies(Drosophila virilis), was subjected to chromatography accompanied by bioassay for sex pheromone activity. After three chromatographic steps, fractions containing active monoenes and dienes were obtained. Chemical analysis by infrared absorption, gas liquid chromatography and gas chromatography/mass spectrometry of the active fraction indicated that active monoenes were comprised chiefly of (Z)-11-pentacosene (abbreviated (Z)-11-C25:1), (Z)-13-C27:1, (Z)-13- and (Z)-14-C29:1. Synthetic monoenes were made, and only (Z)-11-C25:1 elicited good courtship behaviour in maleD. virilis. Therefore it was concluded that (Z)-11-C25:1 was a major sex pheromone. A total of 16.2±1.32 µg of cuticular hydrocarbons was isolated from 10 day old females, including 5.9±0.56 µg of (Z)-11-C25:1. An additive effect was suggested from the higher observed courtship response when using a mixture of active dienes with the active monoene.  相似文献

The mutagenic potential of aqueous, total oligomers flavonoids (TOF), ethyl acetate and methanol extracts from tubers of Cyperus rotundus L. was assessed using Ames Salmonella tester strains TA98 and TA100, and SOS chromotest strain Escherichia coli PQ37 with and without metabolic activation (S9). None of the different extracts produced a mutagenic effect. Likewise, the antimutagenicity of the same extracts was tested using the “Ames test” and the “SOS chromotest”. Our results showed that C. rotundus extracts possess antimutagenic effects against S. typhimurium TA98 and TA100 strains towards the mutagen aflatoxin B1 (AFB1), similar to E. coli PQ37 strain against AFB1 and Nifuroxazide mutagens using the SOS chromotest assay. A free radical scavenging test was used in order to explore the antioxidant capacity of the extracts obtained from the tubers of C. rotundus. TOF, ethyl acetate and methanol extracts showed an important free radical scavenging activity towards the 1,1-diphenyl-2-picrylhydrazine (DPPH) free radical. These extracts showed an IC50 value of respectively 5, 20 and 65?µg?mL?1. The beneficial effects of TOF, ethyl acetate and methanol extracts of C. rotundus were assessed by antioxidant and antimutagenic activities.  相似文献

Flubendiamide, a comparatively new insecticide, is used against lepidopteran insect pests and presumed safe for non-target Drosophila melanogaster (D. melanogaster). In this study, treatment concentrations (0.5, 1, 2 5, 10, and 20 μg/ml) of flubendiamide, lower than agricultural concentrations (rice 50μg/ml or cotton 100 μg/ml), were able to inhibit acetylcholinesterase activity in third instar larvae of D. melanogaster indicating a neurotoxic potential. In addition, larvae exposed to flubendiamide also manifested increased amounts of stress protein hsp70. The larvae expressing such stress response when allowed to emerge as adults displayed severe eye structure deformities found by scanning electron microscopy. These findings indicate a toxic potential for flubendiamide in D. melanogaster.  相似文献

Summary. Previous studies indicated the presence of antennally-active compounds in extracts of eggs laid by female cabbage root flies, Delia radicum, that stimulated oviposition by conspecific females. We confirmed that previously laid D. radicum eggs stimulated oviposition by other D. radicum females, in a dose-dependent manner. Methanol extracts of conspecific eggs stimulated oviposition by females D. radicum, whereas egg extracts of D. antiqua and Psila rosae had no effect. Electrophysiological recordings from the tarsal sensilla of D. radicum females indicated that neurones of the C5 sensillum responded to the egg extracts from both D. radicum and D. antiqua, but not P. rosae. Chemical analysis revealed that the extract of eggs from D. radicum contained the thia-triaza-fluorene compound, 1,2-dihydro-3-thia-4,10,10b-triaza-cyclopenta[.a.]fluorene-1-carboxylic acid (CIF-1), an oviposition stimulant found previously only in cruciferous plants. The potency of free-living and animal-associated marine bacteria to produce antimicrobial substances has been studied in 491 strains isolated from northern and southern parts of the Pacific Ocean. A total of 26% (126 out of 491) of the strains examined produced antimicrobial compounds against 11 test bacterial strains (TBS) including the fish pathogens Aeromonas hydrophila and Vibrio anquillarum. Antimicrobial substances (AS) produced by marine bacteria were especially active against Staphylococcus epidermidis, Proteus vulgaris, Enterococcus faecalis, and Candida albicans. Twelve strains, isolated from different sources, were chosen as promising candidates, producing a number of AS. Production of AS varied within 24 to 72 h, increasing in a culture medium based on natural sea water with Br-ions, and after attachment to polymeric surfaces. In order to study the influence of adsorption, selected strains with a high potential for antimicrobial production were cultivated on polymeric surfaces with different hydrophobicities and chemical functionalities. These parameters of the surface hydrophobicity (measured by means of water contact angles) and chemical functionality of the surfaces were manipulated using the photo- and thermochemistry of a polymeric system (diazo-naphto-quinone/novolak) commonly used as a photoresistant material in semiconducto-manufacturing. The highest antimicrobial activities occurred on hydrophilic surfaces (standard exposed photoresistant films), whereas the number of attached cells was greater on hydrophobic surfaces, characterized as unexposed resistant films. These results suggest that the chemical nature of induced hydrophilicity may also be a major factor in controlling antimicrobial activity of adsorbed bacteria.
