Eight species of desert vegetation and associated soils were collected from the Nevada National Security Site (N2S2) and analyzed for 238Pu and 239 + 240Pu concentrations. Amongst the plant species sampled were: atmospheric elemental accumulators (moss and lichen), the very slow growing, long-lived creosote bush and the rapidly growing, short-lived cheatgrass brome. The diversity of growth strategies provided insight into the geochemical behavior and bio-availability of Pu at the N2S2. The highest concentrations of Pu were measured in the onion moss (24.27 Bq kg-1 238Pu and 52.78 Bq kg-1 239 + 240Pu) followed by the rimmed navel lichen (8.18 Bq kg-1 and 18.4 Bq kg-1 respectively), pointing to the importance of eolian transport of Pu. Brome and desert globemallow accumulated between 3 and 9 times higher concentrations of Pu than creosote and sage brush species. These results support the importance of species specific elemental accumulation strategies rather than exposure duration as the dominant variable influencing Pu concentrations in these plants. Total vegetation elemental concentrations of Ce, Fe, Al, Sm and others were also analyzed. Strong correlations were observed between Fe and Pu. This supports the conclusion that Pu was accumulated as a consequence of the active accumulation of Fe and other plant required nutrients. Cerium and Pu are considered to be chemical analogs. Strong correlations observed in plants support the conclusion that these elements displayed similar geochemical behavior in the environment as it related to the biochemical uptake process of vegetation. Soils were also sampled in association with vegetation samples. This allowed for the calculation of a concentration ratio (CR). The CR values for Pu in plants were highly influenced by the heterogeneity of Pu distribution among sites. Results from the naturally occurring elements of concern were more evenly distributed between sample sites. This allowed for the development of a pattern of plant species that accumulated Ce, Sm, Fe and Al. The highest accumulators of these elements were onion moss, lichen flowed by brome. The lowest accumulators were creosote bush and fourwing saltbush. This ranked order corresponds to plant accumulations of Pu.  相似文献

An alkaline comet assay and a micronucleus test were carried out on erythrocytes of the European chub, Squalius cephalus L., collected in spring and autumn in 2005 and 2006 at three sampling sites in River Sava, near Zagreb, Croatia. The results of comet assay showed the lowest genotoxic influence at the least polluted site, while higher DNA damage was observed at the polluted sites. Although the basal levels of DNA damage were elevated, a clear gradation of DNA damage was found due to pollution intensity in all sampling periods. The lowest cytogenetic damage as revealed by the micronucleus test (MNT) was observed as well at the least polluted site. High variations in MN frequency were observed between sampling periods, although the number of micronucleated erythrocytes was consistently the highest one at the polluted site. The comet assay as a biomarker of genotoxic effect exhibited higher sensitivity in discriminating the genotoxic capacity of studied polluted sites while the MNT was less sensitive. However, both tests should be used together in biomonitoring studies because they can reveal different aspects of DNA damage; comet assay, the early event of genotoxic exposure, and MNT, its final result as a mutagenic potential.  相似文献

This study investigates the potential of the comet and micronucleus assays of fish DNA as a means of screening the toxicity of aquatic environments. Catla catla and Cirrhinus mrigala collected from the River Chenab in Pakistan were used as a case study for the application of comet and micronucleus techniques. Comet and micronucleus assays were used to compare DNA damage in C. catla and C. mrigala collected from polluted areas of the River Chenab and farmed fish. Atomic absorption spectrophotometry showed an acute level of toxicity from Cd, Cu, Mn, Zn, Pb, Cr, Sn, and Hg in river water. Comet assay showed significant (p < 0.05) DNA damage in C. catla representing 17.33 ± 2.42, 11.53 ± 2.14, and 14.17% DNA in the comet tail, averaged from three sites of the polluted area of the river. Tail moment was observed as 10.06 ± 2.71, 3.11 ± 0.74, and 14.70 ± 1.89, while olive moment was 8.85 ± 1.84, 3.83 ± 0.76, and 7.11 ± 0.73, respectively. Highly significant (p < 0.01) damage was reported in C. mrigala as 37.29 ± 2.51, 34.96 ± 2.53, and 38.80 ± 2.42% DNA in comet tail, tail moment was 23.48 ± 3.90, 19.78 ± 4.26, and 14.30 ± 1.82, and olive moment was 16.22 ± 2.04, 13.83 ± 1.96, and10.99 ± 0.90. Significant (p < 0.05) differences were observed in genotoxicity between farmed and polluted area fish. Micronucleus assay showed a similar picture of significant difference in respect to single and double micronucleus induction: i.e., 23.20 ± 4.19 and 2.80 ± 1.07‰ in C. catla and 44.80 ± 3.73 and 06.20 ± 0.97‰, respectively, in C. mrigala. Nuclear abnormalities were found as 6.00 ± 0.84 and 09.60 ± 1.72/thousand cells, respectively, in both species. The results of this study suggest that these novel fish DNA damage assays can be used as an expedient toxicity screening for aquatic environments.  相似文献

The distribution of contaminant radionuclides from the Sellafield nuclear fuel reprocessing plant was used to establish chronologies for three saltmarsh sediment cores from south west Scotland. delta(13)C and (14)C analyses indicated that the cores provided a useful archive record of variations in input of organic matter and carbonate. The results imply that prior to major releases of contaminant (14)C from Sellafield, the (14)C specific activity of organic matter in Irish Sea offshore sediments was about 24 Bq kg(-1) C, while that of the carbonate component was below the limit of detection. These results provide baseline data for modelling the uptake of contaminant (14)C by the Irish Sea sediment system. The study confirmed that small(13)C analyses provide a sensitive means of apportioning the origin of saltmarsh organic matter between C(3) terrigenous plants, C(4) terrigenous plants and suspended particulate marine organic matter. For the <2 mm fraction of sediment, a clear pattern of decreasing marine organic input was observed in response to increasing elevation of the marsh surface as a result of sediment accumulation. Bulk sediment, including detrital vegetation, had a dominant input from terrigenous plants. The combined use of delta(13)C and (14)C data revealed that organic matter in the marine organic component of the <2 mm fraction of contemporary surface sediments of the saltmarshes is dominated by recycled old organic material.  相似文献

The concentrations of the natural radioactivity in water, soil and sediment samples collected from Bendimahi River which originates near the Tendürek Mountain (Van, Turkey), its tributaries and Van Lake (Turkey) were measured. The gross-alpha and gross-beta radioactivity concentrations were investigated in May and August in 2005. Determination of the gross-alpha and gross-beta radioactivity of river water, soil and sediment samples was obtained by using gas-flow proportional counter, PIC-MPC 9604-alpha/beta counter. In water samples, the obtained results show that in May; gross-alpha and gross-beta activity concentrations varied between 0.063-0.782 and 0.021-0.816 Bq l(-1) and in August the values were 0.009-0.037 and 0.081-3.116 Bq l(-1), respectively. The gross-alpha and gross-beta activity concentrations in soil samples ranged from 0.800 to 4.277 Bq g(-1) and 0.951 to 11.773 Bq g(-1) in May and 0.686 to 4.713 Bq g(-1) and 0.073 to 9.524 Bq g(-1) in August, respectively. Concentrations ranging from 0.782 to 4.596 Bq g(-1) and from 0.482 to 10.372 Bq g(-1) in May and from 0.580 to 5.824 Bq g(-1) and from 0.303 to 9.702 Bq g(-1) in August for gross-alpha and gross-beta radioactivity were observed in sediments, respectively.  相似文献

A field study was conducted around two thermal power plants of India to quantify the changes in foliar elemental concentrations due to emission in a low rainfall tropical area. Sulphur dioxide and particulates were at high levels which may cause serious ecological effects. Emission from the power plants has altered the elemental concentrations in the leaves of evergreen and deciduous plants. The foliar total-S and SO4 2–-S levels were significantly higher in all the plants growing at polluted sites. However, the organic-S content was more or less unaltered. In evergreen plants, the SO4 2–-S content increased gradually from summer through winter, whereas in the deciduous plants there was a higher magnitude of increase after onset of new leaves during summer. With the increasing pollution load, foliar Ca2+ and K+ contents increased, whereas N content decreased. The reduction in N content was greater during summer in evergreen plants and during winter in deciduous plants. The plants growing closer to the power plants accumulated more trace elements (Mn, Fe, Cd, Pb and Ni) as compared to those growing at distant sites. Diverse genetic biomarkers have been used to evaluate the effects of pollution by mutagenic agents such as metals and pesticides, as well as a large variety of chemical substances derived from human activities. This work researched the effects that an exposure of 60 days to the insecticide Fipronil (concentrations of 0.05, 0.10 and 0.23 μg/L) can cause in the fish Rhamdia quelen using Comet assay with gills, histopathological analysis of gills and the Piscine Micronucleus test and Nuclear Morphological Alterations. The results for the Comet assay and for gills histopathological injuries showed no difference between the control group and the contaminated groups. In the Piscine Micronucleus test, the smallest concentration of Fipronil (0.05 μg/L) was similar as the control group, while concentrations of 0.10 and 0.23 μg/L caused more damage to the DNA. These results suggested that only the highest concentrations of Fipronil tested cause damage in erythrocytes, but none of these concentrations was sufficient to alter the DNA in the gill cells. R. quelen may be a less sensitive bioindicator than other fish that have been tested. On the other hand, the concentrations used may not have been sufficient to detect alterations in the DNA of R. quelen with the chosen tests. Works like this take on great importance given the enormous quantity of substances that are thrown daily into the environment in an uncontrolled way, without evaluation of the consequences. The application of these tests with other concentrations, tissues and exposure times is suggested for future works.  相似文献

This work aimed to determine the DNA and oxidative damage on mice by mixtures of organic contaminants in wastewater-irrigated soil, in order to assess their usefulness as markers for this kind of pollution. Wastewater-irrigated soil samples in the vicinity of an industrial area in Tangshan, China were collected, and soil irrigated by underground water satisfying drinking water standards was used as control group. Organic pollutants were extracted from the soil using ultrasonic oscillation, and analyzed by gas chromatography–mass spectrometry (GC-MS). Meanwhile, DNA damage on mice was determined by the Comet assay after oral gavage with the extracts, and changes in total superoxide dismutase (T-SOD) activity, glutathione peroxidase glutathione, GSH peroxidase (GSH-PX) activity and malondialdehyde content in serum of mice were investigated. The number of categories and concentrations of organic compounds in the wastewater-irrigated soil is more than those in groundwater-irrigated soil, as identified by the GC-MS. The toxicity test of mice showed that compared with reagent control group, the activities of T-SOD and GSH-PX decreased; the tailing rate of peripheral blood lymphocyte of mice increased and was more than that of the control group. This shows that mammalian toxicity end points can be used to determine the joint toxicity of organic pollutants in soil. When there is no means to identify each and every pollutant in soil, it is feasible to evaluate the combined effects of various pollutants to determine the extent to which the soil is polluted.  相似文献

Many alluvial soils along navigable waterways are affected by disposal of dredged sediments or overbank sedimentation and contain metal concentrations that are elevated compared to baseline levels. Uptake patterns for metals and other elements by several volunteer Salix species growing on these sites were determined during a growing season in field plots and compared with the same species growing on soils with baseline contamination levels. For Cd and Zn, foliar concentrations were clearly higher on dredged sediment landfills. Uptake patterns differed significantly between species. A high uptake of Mn and low uptake of Cu, K and S in S. cinerea was attributed to wetland soil chemistry. Site effects on metal uptake were evaluated in more detail for Salix cinerea and S. alba growing on different sediment-derived sites under field conditions. Foliar Cd concentrations were higher in S. cinerea than in S. alba. This appeared to be a genetic feature not influenced by soil chemical properties, as it was observed both on clean sites and polluted sediment-derived sites. For S. cinerea, soil chemistry was reflected in foliar concentrations, while foliar Cd concentrations and bioavailability were found to be independent of the thickness of the polluted horizon. Dredged sediment landfills and freshwater tidal marshes with comparable Cd soil pollution had significantly different foliar Cd concentrations.  相似文献

Auto-pollution is the by-product of our mechanized mobility, which adversely affects both plant and human life. However, plants growing in the urban locations provide a great respite to us from the brunt of auto-pollution by absorbing the pollutants at their foliar surface. Foliar surface configuration and biochemical changes in two selected plant species, namely Ficus religiosa L. and Thevetia nerifolia L., growing at IT crossing (highly polluted sites), Picup bhawan crossing (moderately polluted site) and Kukrail Forest Picnic Spot (Low polluted site) were investigated. It was observed that auto-exhaust pollution showed marked alterations in photosynthetic pigments, protein and cysteine contents and also in leaf area and foliar surface architecture of plants growing at HP site as compared to LP site. The changes in the foliar configuration reveal that these plants can be used as biomarkers of auto-pollution.  相似文献

应用小鼠淋巴细胞彗星试验对长治市5个点位地下水水质作遗传毒性研究。结果表明,各水样有机浓集物均可对小鼠淋巴细胞DNA产生不同程度的遗传损伤,随着剂量的增加,Olive尾矩与阴性对照组(0.72±0.09)相比均有显著性差异(P0.05,P0.01)。在试验浓度范围内,以彗星尾部DNA含量、彗星尾长、尾矩和Olive尾矩所指示的DNA损伤程度随剂量增大而逐渐增加,存在显著的剂量-效应关系,其相关系数0.815。  相似文献

This research was conducted to search and identify spontaneously growing heavy metal-tolerant plant species that are potentially useful for phytoremediation in contaminated sediment. Five sites were selected for collection of plants growing on polluted shore (river bank) sediment of the Xiang River, China. The concentrations of Zn, Pb, Cu and Cd in plants, sediments, and grasshoppers were determined using flame atomic absorption spectrophotometer (AAS700, Perkin-Elmer, USA). Considering translocation factor and bioaccumulation factor, Rumex crispus (Polygonaceae), Rumex dentatus (Polygonaceae), and Lagopsis supina (Labiatae) could be potentially useful for phytostabilization of metals. R. crispus can be considered potentially useful for phytoextraction of Cd. In light of the biomagnification factors, grasshoppers are deconcentrators for Pb and Cd, microconcentrators for Zn and macroconcentrators for Cu to the plants, respectively. To the best of our knowledge, the present study is the first report on Zn, Pb, Cu and Cd accumulation in R. crispus and L. supina, providing a pioneer contribution to the very small volume of data available on the potential use of native plant species from contaminated sediments in phytostabilization and phytoremediation technologies.  相似文献

This study was conducted in the urban environment of Varanasi, India, to evaluate the plant responses to urban air pollution. Twenty sites were selected in four different zones of the city. At each site, seven woody perennials of same age classes were selected. Out of the four zones (I, II, III and IV), zone IV was used as a reference (control) zone as it received the minimum pollution input. Plant species growing in polluted and control areas were compared with respect to foliar dust load, per cent leaf area injury, leaf area, specific leaf weight and chlorophyll, ascorbic acid, SO 4 2– S and total N concentration in the leaves. Results indicated that the air pollution level in Varanasi causes leaf damage, reduces leaf area, specific leaf weight and chlorophyll, ascorbic acid and total N concentrations in the leaves. Sulphur concentration in leaves increased with increasing level of SO2 in the ambient air. The magnitude of such changes was maximum at the zone receiving maximum pollution load. Carissa carandas was found to be the most sensitive species and Bougainvillea spectabilis, the least. The study shows that the urban air pollution level in Varanasi is detrimental for the growth of plants involved in this study.  相似文献

Contamination by 2,4,6-trinitrotoluene (TNT) is a global environmental problem at sites of former explosive production, handling, or storage, and could have deleterious consequences for human and ecological health. We investigated its sublethal effects to Eisenia fetida, using two nonspecific biomarkers. In coelomocytes of earthworms exposed 24, 48, or 72 h, we evaluated DNA damage (comet assay) and neutral red retention time (NRRT), using the filter paper contact test. Both percentage of damage (D%) and calculated damage index showed significant DNA damage at almost all concentrations, at all time points assayed. Along exposure time, two different patterns were observed. At the lower TNT concentrations (0.25?C0.5 ??g/cm2) an increased DNA migration at 48 h, with a decrease close to initial levels after 72 h exposure, was observed. This decrease could be attributed to activation of the DNA repair system. At higher concentrations (1.0?C2.0 ??g/cm2), the high DNA damage observed remained constant during the 72 h exposure, suggesting that the rate of DNA repair was not enough to compensate such damage. Analysis of NRRT results showed a significant interaction between time and treatment. After 48 h, a significant decrease was observed at 4.0 ??g/cm2. After 72 h, NRRT presented a concentration-dependent decrease, significantly different with respect to control at 0.5, 1.0, 2.0, and 4.0 ??g/cm2. The two assayed methods, performed on the same sample, showed clear responses to sublethal TNT exposure in E. fetida, providing sensitive unspecific biomarkers of cell injury and DNA damage.  相似文献

The first objective of this study was to provide data of arsenic (As) levels in Peninsular Malaysia based on soil samples and accumulation of As in Centella asiatica collected from 12 sampling sites in Peninsular Malaysia. The second objective was to assess the accumulation of As in transplanted C. asiatica between control and semi-polluted or polluted sites. Four sites were selected which were UPM (clean site), Balakong (semi-polluted site), Seri Kembangan (semi-polluted site) and Juru (polluted site). The As concentrations of plant and soil samples were determined by Instrumental Neutron Activation Analysis. The As levels ranged from 9.38 to 57.05 μg/g dw in soils, 0.21 to 4.33 μg/g dw in leaves, 0.18 to 1.83 μg/g dw in stems and 1.32–20.76 μg/g dw in roots. All sampling sites had As levels exceeding the CCME guideline (12 μg/g dw) except for Kelantan, P. Pauh, and Senawang with P. Klang having the highest As in soil (57.05 μg/g dw). In C. asiatica, As accumulation was highest in roots followed by leaves and stems. When the As level in soils were higher, the uptake of As in plants would also be increased. After the transplantation of plants to semi-polluted and polluted sites for 3 weeks, all concentration factors were greater than 50 % of the initial As level. The elimination factor was around 39 % when the plants were transplanted back to the clean sites for 3 weeks. The findings of the present study indicated that the leaves, stems and roots of C. asiatica are ideal biomonitors of As contamination. The present data results the most comprehensive data obtained on As levels in Malaysia.  相似文献

The mutagenic activity of XAD-2 adsorbates and water extracts recovered from nine locations of the Kumho River was tested on S. typhimurium TA98 strain to identify the source of the mutagenicity. A sampling site, receiving effluents from the textile industrial complex located in Daegu City, showed extraordinarily high mutagenic activity, especially in the presence of S9 mixture, at all sampling time in both XAD-2 adsorbates and dichloromethane extracts. This indicated the existence of the frame-shift mutagens in the Kumho River, same type of mutagens detected in previous studies by other researchers in the Nakdong River into which the Kumho River discharges. The fractionation study showed that the mutagenic chemicals in the river water are mid-polar. Furthermore, mean tail length obtained by single cell gel electrophoresis assay (Comet assay) showed consistent dose-dependent DNA damage, indicating that the chemicals in the river water not only act as frame-shift mutagens but also break human lymphocytes DNA strain. Chemical identification of the mutagens should be required.  相似文献

The level of pollution with various mercury species (organomercury, water- and acid-soluble mercury, mercury bound to humic matter and to sulphides) of the floodplain soils and sediments from middle part of the Warta River has been assessed using self-organizing maps (SOM). Chemometric evaluation allowed identification of moderately (median 173-187 ng g(-1), range 54-375 ng g(-1) in soil and 130 ng g(-1), range 47-310 ng g(-1) in sediment) and heavily polluted samples (662 ng g(-1), range 426-884 ng g(-1)). Heavily polluted were located mainly below and in the area of the Poznań city. Statistical comparison of mercury species distribution in floodplain soils of the Warta River shows different patterns for moderately and heavily polluted samples. In heavily polluted soils the contribution of mobile mercury (sum of organomercury species, water- and acid soluble species) is lower (4.2%) than in moderately polluted soils (6.1%). Higher contribution of mobile mercury was observed in sediments of the Warta River (12%). In case of moderately polluted samples, statistical differences in the contribution of mercury species are relatively low and thus the environmental risk from mercury deposited in aquatic system of the Warta River is relatively low. However, higher water levels and heavy floods may incite remobilisation of some organomercuries (2.2-2.9 ng g(-1) in soil and 10 ng g(-1) in sediment) and acid-soluble species of mercury (2.6-2.9 ng g(-1) in soil and 0.5 ng g(-1) in sediment).  相似文献

In view of considerable natural background radioactivity reported from southwest coast of India, the current study documents bioaccumulation of 210Po in two dominant coastal sand dune perennial mat-forming wild legumes, Canavalia cathartica and C. maritima. Root, stem, leaf, mature beans, dry seeds and rhizosphere sand were analyzed for 210Po concentration. 210Po activity decreased in the order of leaves > roots > seeds > stems > beans. The highest 210Po activity was recorded in rhizosphere sand samples (5.78-5.88 Bq kg(-1)) followed by the leaf samples (3.27-3.07 Bq kg(-1)), while it was lowest in mature beans (0.13-0.20 Bq kg(-1)). 210Po activities or tissue moisture between plant species were not significantly different (p > 0.05; t-test). But 210Po activity vs. moisture differed significantly in all tissues (p = 0.0001), vegetative tissues (root, stem and leaf) (p = 0.0016), seeds (p = 0.0393) and proteins in seeds (p = 2.355 x 10(-6)) indicating the importance of moisture and protein in 210Po accumulation. Although 210Po has affinity for proteins, it did not concentrate too much in seed proteins of Canavalia. Concentration of 210Po in mature beans is at safe levels as fisher folk only consume tender pods occasionally. 210Po activity in Canavalia is compared with other plant materials. As the landraces of C. cathartica and C. maritima are distributed throughout pantropical coastal areas, the current study emphasizes on considering them as bioindicators to monitor 210Po in coastal sand dune biomes and in turn the health of coastal population.  相似文献

The analytical techniques used for the determination of butyltin compounds in biological tissues are generally based on a succession of steps (e.g., extraction, derivatization, separation, detection) which are all prone to systematic errors. An interlaboratory programme performed at the EU level and based on a stepwise approach has enabled one to identify and remove most of these errors and to improve considerably the state of the art so that certification of a mussel reference material has been made possible. This paper describes the preparation of this mussel reference material containing monobutyltin (MBT), dibutyltin (DBT) and tributyltin (TBT), the homogeneity and stability studies and the analytical work performed for the certification of the contents of MBT [Sn(C4H9)3+], DBT [Sn(C4H9)2(2+)] and TBT [Sn(C4H9)3+]. The results obtained by a group of expert EU laboratories are discussed along with the methods used to certify the mass fractions (based on dry mass) of monobutyltin (1.50 +/- 0.28 mg kg-1 as MBT cations), dibutyltin (1.54 +/- 0.12 mg kg-1 as DBT cations) and tributyltin (2.20 +/- 0.19 mg kg-1 as TBT cations). The paper also describes an attempt to certify the content of triphenyltin in this material which, although reasonable agreement was found among the participants' results, failed owing to the instability of this compound. This new material will be of great support to ensure the quality control of butyltin determination in shellfish, as carried out in environmental monitoring programmes.  相似文献

Plants can be used as both passive biomonitors and biomitigators in urban and industrial environments to indicate the environmental quality and to ameliorate pollution level in a locality. Many studies reveal that plants are negatively affected by the ambient levels of air pollutants. The present study was conducted to evaluate the impact of air pollution on comparative basis with reference to changes in photosynthetic pigments, plant height, leaves, as well as, biochemical parameters of plants of different sites around Udaipur city receiving varying levels of pollution load. The investigated tree species Dalbergia sissoo Roxb. (Family: Fabaceae) exhibited a reduction in various physiological and biochemical growth parameters that correspond with air pollution levels at different sites. The tree species growing in polluted and control areas were compared with respect to foliar dust load, leaf area, and chlorophyll and total carbohydrate and total protein concentration in the leaves. Our studies suggest that D. sissoo Roxb. can successfully be grown in an area for monitoring air pollution, where it is mild and droughts are common. It will prove as an ideal tree species to control pollution effectively beside acting as a shade tree and being a source of food for birds and animals. By plantation of D. sissoo Roxb., mitigative measure at the polluted sites to control generation of particulate matter and the air quality required can be ensured. Our results also confirm that industrial and vehicular air pollution level in Udaipur city is shifting beyond limits.  相似文献
