The family of Lemnaceae,commonly called duckweed,comprise five genera and 37 species of aquaticmonocotyledonous plants that are endemic to lakes,ponds and brackish water bodies on most continents. They are angiospermsthat have the highest biomass production rate and have been used to remediate wastewater from both municipal andindustrial sources. With these attractive features,duckweed is poised to be a novel agricultural platform that can complementcurrent terrestrial crops. In addition,their remarkable ability to adapt to diverse climates and conditions may provide newunderstanding of stress tolerance and genome adaptability mechanisms. However,one of the first challenges facing theseapplications is to establish reliable and rapid methods for identification of closely related strains that may have very differentbehaviour or properties. This review article summarizes the state of the arts for this endeavour and provides the outlook for thisquest in the near future.  相似文献

The greenhouse effect has become increasingly serious globally. Nitrous oxide (N2O) is both the major ozone depleting substance and a potent greenhouse gas having a global warming potential 298 times that of CO2, and the N2O concentration is still increasing at an annual rate of about 0.8 × 10-9. Nitrous oxide reductase (N2OR) can reduce N2O to N2, and until recently, the nosZ gene was the only gene known to be able to encode N2OR. Besides the well-known nosZI, a new lineage of the N2O-reductase (nosZ clade II), which is abundant and widespread in soils, has been identified. In this paper, the main characters of nosZII-containing microbial communities and the related working mechanisms are summarized. In addition to the main differences between nosZI and nosZII, the important environmental factors that regulate the composition, abundance, and expression of nosZII-containing communities are also discussed in this paper. Studies have shown that nosZII communities are distributed among a diverse range of bacterial and archaeal phyla, such as Epsilon-proteobacteria, Bacteroidetes, and Aquificae. Interestingly, most of the nosZII microbes lack a nitrite reductase encoding gene (nirS or nirK) and are therefore unable to denitrify, indicating the importance of these communities as N2O sinks. Soil properties such as texture, pH, C/ N ratio, temperature, and lake physical gradient could regulate nosZII microbe abundance and diversity, and the pH and C/ N ratio may be the most important influencing factors. Studies on the ecological function of nosZII microbes have advanced considerably with the development of molecular biology technology. However, further studies on the community structure of nosZII microbes, the influencing factors of nosZII microbe abundance and diversity, and characteristics of nosZII strains with strong N2O reducing ability are needed. The amount of spent rechargeable lithium batteries (RLBs) is growing rapidly owing to wide application of these batteries in portable electronic devices and electric vehicles, which obliges that spent RLBs should be handled properly. Identification of spent RLBs can supply fundamental information for spent RLBs recycling. This study aimed to determine the differences of physical components and chemical compositions among various spent RLBs. All the samplings of RLBs were rigorously dismantled and measured by an inductive coupled plasma atomic emission spectrometer. The results indicate that the average of total weight of the separator, the anode and the cathode accounted for over 60% of all the RLBs. The weight ratio of valuable metals ranged from 26% to 76%, and approximately 20% of total weight was Cu and Al. Moreover, no significant differences were found among different manufacturers, applications, and electrolyte types. And regarding portable electronic devices, there is also no significant difference in the Co-Li concentration ratios in the leaching liquid of RLBs.  相似文献

Protein ubiquitination regulates many aspects of plant development and stress response. The RING-finger type E3 ubiquitin ligase SDIR1 (salt and drought induced ring finger 1) gene plays a key role in plant stress response. In this study, the full-length cDNA and the promoter sequences of CsSDIR1 were isolated from tea plants using the RT-PCR technology, and its bioinformatics characteristics were systematically analyzed. The expression patterns of CsSDIR1 in various tissues and in response to cold, drought, salt, and ABA treatments were also investigated by quantitative real-time RT-PCR (qRT-PCR). The sequence of CsSDIR1 contains a complete open reading frame of 831 bp, coding for a 276-long amino acid protein with a molecular weight of (Mr) 30.085 × 103 and a theoretical isoelectric point of 6.54. CsSDIR1 was predicted to be a hydrophobic protein localized on the intracellular membranes. The analysis of the amino acid sequence characteristics showed that CsSDIR1 contains two putative transmembrane domains at the N-terminus and a C3H2C3 RING-finger domain at the C-terminus; it shares high similarity with other plants' SDIR1, and has the closest relationship to Actinidia sinensis. A cis-acting regulatory elements prediction showed that the CsSDIR1 promoter contains many cis-acting elements, especially drought and salt stress response elements. The qRT-PCR analysis indicated that the CsSDIR1 gene has a high expression level in stems, followed by roots, leaves, and flowers; the expression of the CsSDIR1 gene is up-regulated by ABA, salt, and drought treatments, whereas it is down-regulated in response to cold stress. The acoustic behaviour of Eptesicus serotinus was investigated in the field using a 13.5-m vertical, linear microphone array that allowed for simultaneous recordings at three different heights and for the calculation of flight altitude and distance from the array. Recordings were made at two locations that differed in bat species diversity. E. serotinus hunted on average at an altitude of 10.7 m (±2.7) at one location and 6.8 m (±3.6) at the other location. Search signals were 5–17 ms long depending on flight altitude, and consisted of two to three frequency-modulated harmonics. For bats flying below 8–10 m altitude, signal duration decreased with decreasing flight altitude, whereas signal interval, terminal frequency, peak frequency and frequency range of the first harmonic increased. Above 8–10 m flight altitude, the signal parameters were fairly constant. The –10 dB bandwidth and duty cycle did not change with flight altitude. Source levels were calculated to between 121 and 125 dB peSPL re 20 μPa at 10 cm. For bats flying higher than 9 m, the microphone placed 1.5 m above the ground recorded significantly reduced signal durations and frequency ranges of the first harmonic compared to the same signals recorded with the microphones at heights of 7 or 15 m. We caution the use of ground recordings to fully describe the echolocation signals of high-flying bats. We demonstrate that flight altitude significantly influences the structure of sonar signals from E. serotinus. As a tool for management, query, visualization and analysis of spatially referred information, GIS has been recognized as a method to aid the modeling of diffuse pollution and visualize the results in a spatial context. A common question in integrating diffuse pollution models and GIS is to choose a suitable coupling approach, in which the feature of diffuse pollution models should be taken into account. In this paper, we report on our experience in coupling a distributed diffuse pollution model with a GIS. A prototype of fully integrated system is developed in this paper. This system has high flexibility, extendibility and great data management efficiency. Differences in applicability of loose coupling, tight coupling and fully integrated approaches are addressed. It is concluded that the fully integrated approach can avoid tanglesome data exchange and routine execution and more robust than loose and tight coupling approaches and is suitable for distributed diffuse pollution modes.  相似文献

The water supply network (WSN) system is a critical element of civil infrastructure systems. Its complexity of operation and high number of components mean that all parts of the system cannot be simply assessed. Earthquakes are the most serious natural hazard to a WSN, and seismic risk assessment is essential to identify its vulnerability to different stages of damage and ensure the system safety. In this paper, using a WSN located in the airport area of Tianjin in northern China as a case study, a quantitative vulnerability assessment method was used to assess the damage that the water supply pipelines would suffer in an earthquake, and the finite element software ABAQUS and fuzzy mathematic theory were adopted to construct the assessment method. ABAQUS was applied to simulate the seismic damage to pipe segments and components of the WSN. Membership functions based on fuzzy theory were established to calculate the membership of the components in the system. However, to consider the vulnerability of the whole system, fuzzy cluster analysis was used to distinguish the importance of pipe segments and components. Finally, the vulnerability was quantified by these functions. The proposed methodology aims to assess the performance of WSNs based on pipe vulnerabilities that are simulated and calculated by the model and the mathematical method based on data of damage. In this study, a whole seismic vulnerability assessment method for a WSN was built, and these analyses are expected to provide necessary information for a mitigation plan in an earthquake disaster.  相似文献

As low oxygen and high ultraviolet (UV) exposure might significantly affect the microbial existence in plateau, it could lead to a specialized microbial community. To determine the abundance and distribution of ammonia-oxidizing archaea (AOA) in agricultural soil of plateau, seven soil samples were collected respectively from farmlands in Tibet and Yunnan cultivating the wheat, highland-barley, and colza, which are located at altitudes of 3200-3800 m above sea level. Quantitative PCR (q-PCR) and clone library targeting on amoA gene were used to quantify the abundances of AOA and ammonia-oxidizing bacteria (AOB), and characterize the community structures of AOA in the samples. The number of AOA cells (9.34 × 10^7-2.32× 10^8 g^-1 soil) was 3.86-21.84 times greater than that of AOB cells (6.91 × 10^6-1.24 × 10^8 g^-1 soil) in most of the samples, except a soil sample cultivating highland- barley with an AOA/AOB ratio of 0.90. Based Kendall's correlation coefficient, no remarkable correlation between AOA abundance and the environmental factor was observed. Additionally, the diversities of AOA community were affected by total nitrogen and organic matter concentration in soils, suggesting that AOA was probably sensitive to several environmental factors, and could adjust its community structure to adapt to the environmental variation while maintaining its abundance.  相似文献

Cadmium (Cd2+) pollution in an aquatic environment can negatively affect certain reproductive parameters of aquatic animals. β-N-acetyl-D-glucosaminidase (NAGase) is considered to play an important role in the fertilization process. The aim of the present study was to investigate the effect of Cd2+ on the activity of NAGase purified f rom the testis of Nile tilapia, toward contributing new knowledge on the breadth of negative effects of Cd2+ for Nile tilapia production. The kinetic method of substrate reaction was used for this assessment, and an inhibitory model was established to study the kinetics of NAGase under inhibition by Cd2+. The results showed that Cd2+ could reversibly inhibit the enzymatic activity of NAGase, and the half-maximal inhibitory concentration was estimated to be 40.95 mmol/L. Cd2+ was found to be a competitive inhibitor of NAGase, and the inhibitory constant was determined to be 17.13 mmol/L. The microscopic rate constants of inactivation were also determined. Together, these findings demonstrate that Cd2+ is a reversible inhibitor that can competitively inhibit NAGase. These results may provide a theoretical foundation for further studies on the reproduction of tilapia. © 2018 Science Press. All rights reserved.  相似文献

Use of 16S -23S intergenic transcribed spacer (ITS) variability, as a relatively new method, is becoming an important supplement to the molecular methods based on 16S rRNA for which has a fairly constant size and is not divergent enough to give good separation in close relationships. This paper summarizes the structures and characteristics of ITS regions that are extremely variable in copy number, length and sequence per genome. The ITS region can be amplified easily taking advantage of conserved nucleotide stretches at the 5′of the 16S and 3′of the 23S gene, and the amplicon can contain different amounts of the 16S rDNA by choosing primers at different conserved areas within this gene. These primers are listed and discussed for perfecting the methodology of ITS. Furthermore, some recent progresses on the taxonomy, identification and community analysis of bacteria by means of ITS in epidemiology, ecology and artificial environment are reviewed, as well, the virtues and limitations of that method are discussed. Problems treating sewage in rural areas in China have attracted increasing attention in recent years. However, only 6% of rural areas had spray drainage and wastewater treatment systems by the end of 2010. It is necessary to assess the technologies currently used so that sewage treatment in rural areas can be developed appropriately, to provide reliable and affordable waste- water treatment, in these areas. We evaluated the sewage treatment technologies currently used in rural areas by conducting case studies on 62 facilities that had each been operating for at least one year. Our study was carried out between 2009 and 2011, and the aim was to assess the situation during that period and assess any problems involved in decentralized sewage treatment in villages. We found that decentralized sewage treatment is the most popular wastewater treatment method in villages, and that the most common primary treatment technology used in rural areas is the septic tank. Our investigation highlights the need to establish standards for assessing effluent quality, including a range of target pollutants. Our results also show that effluents should be reused to meet the local environmental requirements in different areas, especially in ecologically sensitive areas.  相似文献

Nitrogen contamination of surface water is a worldwide environmental problem with intensive agricul- ture and high population densities. We assessed the spatial and seasonal variation in concentrations of total nitrogen and different nitrogen species present in surface-water in Beijing, China. Also, chemical (NO3-N/C1-) and isotopic (615Nnitrate) indicators were used to identify nitrate sources. The results showed that, during 2009 and 2010, nitrate nitrogen concentrations ranged from 0.7 to 7.6 mg· L^-1, ammonium nitrogen from 0. I to 3.4 mg· L^-1, and total nitrogen from 2.4 to 17.0mg· L^-1. Inorganic nitrogen accounted for between 60 and 100% of total nitrogen at the ten monitoring sites. Nitrate nitrogen, ammonium nitrogen, and total nitrogen concentrations at the 2 downstream monitoring sites in south-eastern Beijing were significantly higher than those at the other eight upstream monitoring sites (P 〈 0.01). Examination of seasonal variation showed that there was a significant inverse relationship between nitrate nitrogen concentrations and precipitation, and that nitrate nitrogen concentrations peaked in the dry seasons. The information given by the 15Nnitrate values and nitrate nitrogen concentrations, combined with the NO3-N/C1- ratio distribution, showed that domestic sewage was the major source of nitrate in Beijing. Methods to control and reduce sewage pollution are urgently needed to help manage surface water quality in Beijing.  相似文献

Currently, environmental protection and resources conservation continue to be challenges faced by solid-waste managers in China. These challenges are being further compounded by rapid socioeconomic devel- opment and population growth associated with increased waste generation rates and decreased waste disposal capacities. In response to these challenges, an interval joint-probabilistic mixed-integer programming (IJMP) method is developed for supporting long-term planning of waste management activities in the city of Tianjin, which is one of the largest municipalities in the northern part of China. In the IJMP, joint probabilistic constraints are introduced into an interval-parameter mixed-integer programming framework, such that uncertainties presented in terms of interval values and random variables can be reflected. Moreover, a number of violation levels for the waste-management-capacity constraints are examined, which can facilitate in-depth analyses of tradeoffs among economic objective and system-failure risk. The results indicate that reasonable solutions have been generated. They are valuable for supporting the adjustment of the city's existing waste-management practices and the long- term planning of the city's waste-management facilities.  相似文献

Ethanol production from lignocellulosic waste has attracted considerable attention because of its feasi- bility and the generation of valuable products. Previous studies have shown that pretreatment and hydrolysis are key processes for lignocellulose conversion. Hydrothermal process is a promising technique because of its efficiency to break down the lignocellulosic structures and produce fermentable hexoses. Most studies in this field have therefore focused on understanding these processes or optimizing the parameters, but commonly reported low yields of fermentable hexoses. The inability to produce high yields of fermentable hexoses is mainly attributed to inadequate information on the conversion mechanisms of lignocellulose, particularly the reaction rules of dissolu- tion, which is a limiting step in the entire conversion process. This paper critically reviewed the progress done in the research and development of the hydrothermal dissolution and hydrolysis of lignocellulose. Principles, processes, and related studies on separate dissolution and asynchronous hydrolysis of lignin, hemieellulose, and cellulose are presented. Potential research prospects are also suggested.  相似文献

Thermal pretreatment can be applied to sludge anaerobic digestion or dewatering. To analyze the variation in humic substances during thermal sludge treatment, sludge humic and fulvic acids were extracted before and after 30-min thermal treatment at 180℃, and then their contents, molecular weight distributions, elementary compositions, and spectral characteristics were compared. The results showed that the total contents of humic and fulvic acids in the sludge almost remained constant during thermal treatment, but 35% ofhumic and fulvic acids were dissolved from the sludge solids. Moreover, both humic and fulvic acids were partly decomposed and 32% of humic acids were converted to fulvic acids. The median value of the molecular weights of humic acids decreased from 81 to 41 kDa and that of fulvic acids decreased from 15 to 2 kDa. Besides the reduction in molecular size, the chemical structures of humic and fulvic acids also exhibited a slight change, i.e. some oxygen functional groups disappeared and aromatic structures increased after thermal sludge treatment.  相似文献

The uterotrophic assay has been commonly used to test environmental estrogens in vivo, however, it is often not sensitive enough sometimes. An alternative way is to evaluate estrogenicity through biomarker genes. MicroRNA (miRNA) is a class of regulatory gene, which has been shown to be a good biomarker for many diseases and toxicological effects in recent years, and some evidences showed that estrogen induced response was partially mediated by miRNAs. In this study, two types of microarrays were used to test the 17[3-estradiol (E2) induced miRNA expression profile at different time points in the immature mouse uterus. Statistical analysis showed the aldehyde slide based array had less variation than the amino slide based array, and 11 dysregulated miRNAs were screened out for significant fold change. Real-time PCR was performed to further confirm that 4 out of 7 selected miRNAs, namely miR-451, miR-155, miR-335- 5p, and miR-365, are E2 regulated miRNAs in the uterus. The function of the predicted targets of these miRNAs is involved in cell grow control, which is consistent with the main E2 function in the uterus. MiR-451 had similar strong responses to E2 in the uterus of both immature and overiectomized mice, and could be a potential biomarker for estrogenicity in the uterus.  相似文献

Furfural is a toxic metabolic inhibitor that is created during the conversion of lignocellulose to produce fuel, which can retard fermentation and increase production costs. thus, it is important for lignocellulosic conversion that the ability of the strain to resist furfural stress be improved. A cellulose-degrading bacterium BREC-11 with tolerance to furfural was isolated from the intestinal tract of Omphisa fuscidentalis hampson larvae via the addition of furfural in the medium. Based on analyses of morphological observations, physiological and biochemical characterizations, and 16S rDNA sequences, strain BREC-11 was shown to represent a member of the genus Bacillus and was named B. siamensis BREC-11. to study the tolerance concentration of strain BREC-11, a wide range of furfural formaldehyde concentrations were tested and strain BREC-11 was shown to grow in the mineral medium containing furfural up to 3.5 g/L. Cellulase activity of strain BREC-11 was determined at the tolerable concentration of 3.5 g/L furfural after incubation at 30 ℃ and 150 r/min for 2 days. Results indicated that filter paper enzyme, CMC-Na enzyme, and β-glucosidase activity was 0.1 U/mL, 0.21 U/mL, and 0.07 U/mL, respectively. BREC-11 is a cellulose-degrading bacterium with resistance to furfural, which has potential application in future bio-refinery processes. © 2018 Science Press. All rights reserved.  相似文献

The relationship between the chemical con- taminants and soil microbial toxicity of waste foundry sand (WFS) was investigated. Five different types of WFS from typical ferrous, aluminum, and steel foundries in China were examined for total metals, leachable metals, and organic contaminants. The soil microbial toxicity of each WFS was evaluated by measuring the dehydrogenase activity (DHA) of a blended soil and WFS mixture and then comparing it to that of unblended soil. The results show that the five WFSs had very different compositions of metal and organic contaminants and thus exhibited very different levels of soil microbial inhibition when blended with soil. For a given WFS blended with soil in the range of 10wt.%-50wt.% WFS, the DHA decreased almost linearly with increased blending ratio. Furthermore, for a given blending ratio, the WFSs with higher concentrations of metal and organic contaminants exhibited greater microbial toxicity. Correlation analysis shows that the relationship between ecotoxicity and metal and organic contaminants of WFSs can be described by an empirical logarithmic linear model. This model may be used to control WFS blending ratios in soil-related applications based on chemical analysis results to prevent significant inhibition of soil microbial activity.  相似文献
