Summary. The tobacco hornworm, Manduca sexta L. (Lepidoptera: Sphingidae) is a specialist on Solanaceae. This host specificity is induced as the larva feeds on solanaceous foliage, so that solanaceous-reared larvae will refuse to feed on other plants. Experiments were designed to determine the role of dietary constituents on the induction of host specificity and the effects of these on development of M. sexta. Choice assays using leaf discs of cowpea, Vigna sinensis (Fabaceae), were used to monitor the isolation of relevant chemical cues from foliage of potato. An aqueous extract of potato foliage strongly stimulated feeding. This extract was partitioned with n-butanol under alkaline conditions to obtain a highly active butanol extract. Reversed phase flash chromatography with a water-methanol gradient gave an active fraction that was used as a supplement for wheat germ-based artificial diet. Larvae reared on this s-diet became dependent on potato allelochemicals for initiation and continuation of feeding activity. These larvae also developed faster than larvae reared on the control p-diet, but no effect on adult mass was detected. Further flash chromatography of the active fraction under alkaline conditions provided a highly active sub-fraction, and semi-preparative HPLC using gradients of water and acetonitrile resulted in the isolation of a single compound. Bioassays indicated that this compound alone can account for host recognition by solanaceous-reared larvae. The results suggest that the mechanism of induced host specificity in M. sexta involves development of dependence on this compound. Received 21 December 1999; accepted 14 March 2000  相似文献

Summary. We have isolated a caffeoylcyclohexane-1-carboxylic acid derivative, 3-caffeoyl-muco-quinic acid (3-CmQA), as a contact oviposition stimulant for the zebra swallowtail butterfly, Eruytides marcellus (Papilionidae), from the foliage of its primary host plant, Asimina triloba (Annonaceae). This compound alone was as active in stimulating oviposition by females as were the parent ethanolic plant extract and the host plant itself. Other tested isomers of 3-CmQA, including 5-caffeoylquinic acid (5-CQA or trans-chlorogenic acid), were inactive. We found, however, that experienced female butterflies responded strongly to host volatiles, which enhanced landing rates and hence oviposition.? This is the first report of an oviposition stimulant for a swallowtail butterfly of the tribe Graphiini. We found 3-CmQA to be the major caffeoylcyclohexane-1-carboxylic acid isomer in plants of the genus Asimina. These plants lack appreciable amounts of 5-CQA, which has been shown previously to be one of the oviposition stimulants for certain Rutaceae- or Apiaceae-feeding swallowtails of the related tribe Papilionini.? Our findings, along with earlier results from the tribes Troidini and Papilionini, suggest that responses by swallowtails to hydroxycinnamic acid derivatives as oviposition cues date back at least to the ancestor of the subfamily Papilioninae. Summary. Field observations indicated that hornworms select feeding sites non-randomly on tobacco. We tested the hypotheses that differences in feeding site locations of larvae of Manduca sexta L. and Manduca quinquemaculata (Haworth) (Lepidoptera: Sphingidae) on tobacco could be explained by differential nicotine concentrations within plants and leaves, species-specific responses to nicotine, or pressure exerted by natural enemies. Results showed that third-instar larvae of M. sexta fed more proximally and centrally on the leaf, whereas M. quinquemaculata fed more distally. Within-plant selection of leaves did not differ; both species selected leaves in the middle region of the plant. Nicotine concentrations in a high nicotine genotype, NC95, varied within each leaf, increasing 2—3 fold from the basal to apical portion of the leaf, and within each plant, increasing 7—10 fold from the first fully expanded leaf to the twelfth (lowest) leaf. In laboratory bioassays, both Manduca species responded to nicotine as a feeding deterrent. Electrophysiological studies demonstrated that gustatory organs of both species responded to nicotine at concentrations found in tobacco leaves and that M. quinquemaculata generally showed a less vigorous response to nicotine than M. sexta. Field mortality of M. sexta due to parasitism by Cotesia congregata (Say) and to parasitism and predation combined differed among feeding sites; predation alone did not. Results suggest that although nicotine concentration and species specific responses to nicotine play a role in determining feeding site locations, pressure exerted by natural enemies, especially parasitism by C. congregata, is more important. Summary. Ether extract of the abdominal gland of female black chafers Holotrichia loochooana loochooana (Sawada) (Coleoptera: Scarabaeidae) induced a series of pre-mating behaviors that included short-distance orientation, mounting and abdominal bending toward females in the laboratory. When the extract was chromatographed on a silica gel column, the activity was eluted with 50% ether in hexane and ether fractions. GC-MS analyses revealed that both the active fractions contained anthranilic acid (2-aminobenzoic acid) as a major compound. When partitioned to basic, acidic and amphoteric fractions, pheromonal activity was observed with the latter two fractions and anthranilic acid was detected in these fractions by HPLC analyses. The amount of anthranilic acid in the female extract was estimated to be ca. 1.3 μg/female. Authentic anthranilic acid induced pre-mating behavior in males of at levels equal to that elicited by the compound in the extract. It also attracted male beetles to the traps in the field. Summary. Nicotine tolerance is well known for Manduca sexta. It also occurs in several other sphingids of the subfamilies Macroglossinae and Sphinginae. Only members of the subfamily Smerinthinae appear to be more susceptible to nicotine intoxication. Phylogenetic trees have been reconstructed from mitochondrial 16S rDNA and nuclear DNA to map nicotine tolerance.?The nicotine binding site of both α-subunits of nicotinic acetylcholine receptors (nAChR) have been amplified and sequenced. No apparent amino acid substitution can be seen in the putative nicotine binding site of the α-subunits of nAChR from nicotine tolerant and nicotine sensitive sphingids. Thus, a simple target-site modification can be ruled out as a cause for nicotine tolerance. This finding agrees with feeding experiments: larvae of M. sexta and other sphingids of the Macroglossinae and Sphinginae not only tolerated nicotine, but also many other alkaloids that affect neuroreceptors other than acetylcholine receptors (nAChR, mAChR).?Only 10 to 20% of nicotine injected into larvae of nicotine-tolerant taxa could be recovered later as free nicotine, nicotine N-oxide or cotinine, i.e., 80 to 90% must have been converted to polar conjugates or degradation products which are not detectable with the methods applied. Usually more than 98% of the recoverable alkaloids were found in the faeces. Excretion reached a maximum 6 h after injection in tolerant taxa. Larvae of Manduca sexta, which were reared on a nicotine-rich diet, showed higher nicotine degradation and faster nicotine elimination than na?ve larvae. Application of the cytochrome P450 inhibitor SKF 525A (proadifen) reduced the formation of nicotine N-oxide and the rate of alkaloid degradation. Thus, an inducible detoxification mechanism, coupled with a rapid and inducible excretion, appear to be a strategy in Sphingidae that helps them to live on host plants rich in otherwise toxic secondary metabolites. Summary. The phorbol ester DHPB has been detected in 5th instars and adults of Pachycoris klugii which feed on Jatropha curcas, a producer of phorbol esters with mollusc-, insect-, and vertebrate toxicity. DHPB from Pachycoris activates protein kinase C (PKC) which appears to be the main molecular target for phorbol esters. Phorbol esters of J. curcas exhibit a wide range of acute toxic effects in vertebrates and insects. It is therefore likely that the sequestration of DHPB, which would explain the aposematic colouration of the bugs, confers chemical protection to P. klugii against vertebrate predators. Summary. It has long been assumed that the North American pipevine swallowtail, Battus philenor (L.) (Papilionidae, Troidini), is protected from natural enemies by aristolochic acids sequestered from its Aristolochia food plants. This study confirmed that populations of B. philenor from Virginia and east Texas sequester these compounds. A comparison of the aristolochic acid profiles of the Virginia butterflies and their A. macrophylla food plants revealed several differences. The aristolochic acid fraction of the foliage was dominated by aristolochic acids I and II, whereas the insects had a much lower proportion of aristolochic acid II and contained, in addition, substantial amounts of aristolochic acids Ia and IVa, which were not detected in the plants. The eggs, larval integument, osmeterial glands, pupal cuticle, and adults (wings and bodies) all contained aristolochic acids. These findings help explain the abundant ecological data indicating that both immature and adult B. philenor are unpalatable and protected from natural enemies. Summary. Juvenile hormones are sesquiterpenoids that regulate developmental processses such as metamorphosis and reproduction in insects. Insect juvenile hormone III (JH III), methyl-10R,11-epoxy-3,7,11-trimethyl-2E,6E-dodecadienoate, has also been identified in two sedge species, Cyperus iria and C. aromaticus (Toong et al. 1988). Potential allelopathic activity of this compound and the structurally related sesquiterpenoid farnesol was investigated using seed germination and seedling growth assays with radish, lettuce and rice. Treatment of seeds with JH III delayed lettuce seed germination and potently inhibited rice shoot growth. Both farnesol and JH III inhibited the growth of C. iria seedlings. The antimicrobial activity of JH III was also tested on a taxonomic and ecologically diverse range of fungi. Using the classic cytotoxic disk assay, JH III did not effect growth of the fungal species tested. We believe that JH III may contribute to the aggressive nature of this invasive weed species. Summary. We tested responses to prey chemicals by lizard hatchlings of an oviparous species and neonates of a viviparous species, neither of which had never eaten. Both species responded more strongly to prey chemicals than to odorous and odorless control stimuli presented on cotton swabs. Although only a few species have been examined, all that have been tested have an innate capacity for prey chemical discrimination, suggesting that this innate response to prey chemicals is widespread among lizards that use the lingual-vomeronasal system to locate and identify prey. Innate prey chemical discrimination has the great advantage of permitting lizards lacking prior experience with food to respond appropriately to chemical cues associated with food. Both species discriminated prey chemicals from control substances at age three days, earlier than previously known. Our data hint that Mabuya macularia may be capable of discrimination on its day of birth, but further study is needed to determine the exact onset. A stronger tendency to attack swabs bearing prey chemicals by Scincella lateralis than by M. macularia may be explained by differences in defensiveness near an experimenter or by differences in the importance of visual prey cues for confirmation of chemical cues in the natural habitats of these species. In M. macularia responses to the control stimuli declined over days of testing, suggesting habituation, but responses to prey chemicals did not habituate by the third day of testing, which is interpreted as a possible adaptive response to permit location of food. In the standard method of stimulus presentation, a cotton swab bearing a chemical stimulus is placed anterior to a lizard's snout. We tested a new method in which the swab was placed in continuous contact with the lizard's anterior labial scales. The new method elicited significantly stronger responses from M. macularia. Summary. In earlier investigations on host plant discrimination of leaf beetles glucosinolates were described as feeding stimulants for the Brassicaceae specialist Phaedon cochleariae F. (Coleoptera: Chrysomelidae). However, since these findings could not be confirmed in later studies offering 2-propenylglucosinolate in concentrations corresponding to those detected in host plant leaf material, the identification of feeding stimulants of this leaf beetle species remained unclear. In order to investigate which compounds of the host plant Sinapis alba (Brassicaceae) are involved in feeding stimulation, leaf extracts of different polarities were tested in bioassays with adults of P. cochleariae. Number of feeding beetles and net consumption rates were highest on pea leaves painted with methanol extracts of S. alba, whereas weak feeding responses were also detectable for hexane extracts. In subsequent bioassay-guided fractionations of methanol extracts with semi-preparative high performance liquid chromatography, two distinct fractions, one containing glucosinolates and another containing flavonoids, were found to stimulate beetles to feed to variable degrees. Other collected fractions had zero activity. The combination of both active fractions evoked significantly higher consumption rates and stimulated more beetles to feed than fractions tested individually. At least one compound of each fraction, among these the main glucosinolate of S. alba, 4-hydroxybenzylglucosinolate, act additively. Effects of two different naturally-occurring ratios of glucosinolates and flavonoids on the strength of feeding responses were investigated by use of extracts of two sets of host plants differently exposed to radiation. One set was outdoors-exposed, whereas the second set was kept in the greenhouse. However, the feeding behaviour of P. cochleariae was not affected by the significantly different relative compositions of both compound classes in the host material. In conclusion, mustard leaf beetles need a combination of distinct plant metabolites acting in concert for feeding stimulation, whereby the mere presence of these stimulants, but probably not the ratio of involved compounds, determines their feeding response.  相似文献

Summary. Research on insect migration has justifiably emphasized females – the so-called “oogenesis-flight syndrome”– since it is the females that place the eggs into new habitats. The large and small milkweed bugs, Oncopeltus fasciatus and Lygaeus kalmii, respectively, have featured prominently in studies of insect migration and sequestration of host plant toxins for chemical defense. Here we report that males of these species, and males of another well-studied lygaeine (Neacoryphus bicrucis), produce pheromones in glands usually considered to serve only a defensive role in Heteroptera (the metathoracic scent glands), and that these pheromones are exploited by a tachinid parasitoid as a host-finding kairomone. The pheromones are mixtures of C6 and C8 saturated and unsaturated esters reminiscent of lepidopteran pheromones, and the key compound of the O. fasciatus pheromone has now been correctly identified as (E)-2,7-octadienyl acetate. It is proposed that the concept of the oogenesis-flight syndrome for these kinds of insects should accommodate the role of males in the migration process. The hypothesis is presented that male-produced pheromones play a significant role in guiding colonization of new habitats in many heteropteran species. In addition, data are presented suggesting that there is a trade-off between the amount of pheromone produced by colonizing males and the host breadth of the species. Summary The chemical stimulation of oviposition byPieris rapae on cabbage was investigated by leaf washing and extraction. Isolation of the stimulant by various chromatographic techniques was monitored by a bioassay using Sieva bean as a surrogate host plant. Cold water, chloroform, or chloroform followed by cold water washes failed to release the stimulant from leaf surfaces. Boiling water or chloroform followed by methanol was required. The most active stimulatory compound was identified as 3-indolylmethyl glucosinolate (glucobrassicin). Other glucosinolates were identified as sinigrin, which was only slightly active, and glucoiberin, which was completely inactive as a stimulant. The significance of the selective response ofP. rapae andP. brassicae to different glucosinolates and the implications of the binding of polar allelochemicals to leaf surfaces is discussed with respect to host utilization and perception mechanisms of pierids.  相似文献

Summary. Schistocerca gregaria, the desert locust, has been shown to regulate its dietary intake with respect to specific macronutrients in synthetic foods. This study examined the nutrients in the leaves of two starch mutants of Arabidopsis thaliana, and then compared the feeding behaviour of locusts on the two starch mutants. The high-starch mutant had c. 25 times more starch than the no-starch mutant. Newly molted 5th stadium locusts were preconditioned for 3 days on one of the mutants, and then observed for 90 min while exposed to the same or the alternative mutant. Locusts pretreated with the no-starch mutant fed longer during the first meal on high-starch mutants, spent more time feeding, and had the smaller latency to begin a meal when compared to the locusts pretreated on the high-starch mutant. Summary. Field collected exocrine defensive secretions of nine neotropical Platyphora species were analyzed for the presence of plant acquired pyrrolizidine alkaloids (PAs) and pentacyclic triterpene saponins. All species secrete saponins. In addition, five species feeding on Tournefortia (Boraginaceae), Koanophyllon (Asteraceae, tribe Eupatorieae) and Prestonia (Apocynaceae) were shown to sequester PAs of the lycopsamine type, which are characteristic for species of the three plant families. The PA sequestering species commonly store intermedine, lycopsamine and their O3′-acetyl or propionyl esters as well as O7- and O9-hydroxyisovaleryl esters of retronecine. The latter as well as the O3′-acyl esters were not found in the beetles’ host plants, suggesting the ability of the beetles to esterify plant derived retronecine and intermedine or its stereoisomers. Despite the conformity of the beetles’ PA patterns, considerable inconsistencies exist regarding the PA patterns of the respective host plants. One host plant was devoid of PAs, while another contained only simple necines. Since the previous history of the field collected beetles was unknown this discrepancy remains obscure. In contrast to the Palearctic chrysomeline leaf beetles, e.g. some Oreina species which ingest and store PAs as their non-toxic N-oxides, Platyphora leaf beetles absorb and store PAs as the toxic free base (tertiary PA), but apparently avoid to accumulate PAs in the haemolymph. This suggests that Chrysolina and Platyphora leaf beetles developed different lines of adaptations in their parallel evolution of PA mediated chemical defense. Summary. Extracts from the sex pheromone gland of Ostrinia latipennis (Lepidoptera: Crambidae) were analyzed by gas chromatography-electroantennographic detection (GC-EAD) and GC-mass spectrometry. Only an EAD-active compound was detected in the extract, and it was identified as (E)-11-tetradecenol (E11-14:OH). In a wind-tunnel bioassay, E11-14:OH elicited a series of mate finding behaviors from males, although it was far less active than virgin females and crude extract of the pheromone gland. The attractiveness of E11-14:OH to O. latipennis males was confirmed by field trapping experiments. Based on these findings, we concluded that E11-14:OH, which is novel to the genus Ostrinia, is a major component of the sex pheromone in O. latipennis. The significance of the use of alcohol in place of the usual acetates in Ostrinia is discussed in relation to the pheromone biosynthesis system. Summary. The sugar alcohol dulcitol is a strong feeding stimulant for larvae of the small ermine moth Yponomeuta cagnagellus. In this paper we tested the hypothesis that dulcitol also acts as an oviposition stimulant for this species. We found that the sugar-alcohol dulcitol was present on the surface of the host Euonymus europaeus. We also showed that (as yet unidentified compounds) can be systemically transferred (i.e. by uptake and transport via the vascular system) from E. europaeus, to the non-host Crataegus monogyna and stimulate oviposition. However, no evidence was found that this stimulatory activity was due to dulcitol. Systemic enrichment of C. monogyna with dulcitol did not induce oviposition on this plant. Neither was the application of pure dulcitol on artificial twigs effective. In addition it was shown that when dulcitol was removed from host plant extracts, oviposition stimulatory activity was retained in the fraction without dulcitol. Synergism between dulcitol and other stimulants could not be demonstrated, however, high concentrations of dulcitol in combination with the main stimulant(s) showed a trend towards enhanced oviposition. It is concluded that the sugar alcohol dulcitol can only play a limited role in adult host acceptance behaviour. The hypothesis that a host shift in the genus Yponomeuta from Celastraceae to Rosaceae might have been facilitated by the presence of small amounts of dulcitol in Rosaceae therefore needs to be restricted to larval feeding behaviour. Summary. Workers of the amblyoponine species Mystrium rogeri employ trail communication during recruitment to food sources and new nest sites. The trail pheromone originates from a hitherto unknown sternal gland located in the 7th abdominal sternite. The recruiting ant deposits the gland secretions by a special gaster-dragging behavior. The recruitment behavior can be complemented by a rapid vertical body shaking performed by some recruiting ants inside the nest. M. rogeri workers possess a large pygidial gland, the secretion of which elicits a repellent response in other ant species. Received 25 May 1998; accepted 15 June 1998.  相似文献

Summary. Benzoic acid is a known stimulant of olfactory receptor cells in trichoid sensilla on the antennae of adult females of Bombyx mori. Exposure of freshly excised antennae, from B. mori females and males, to vapours of [14C]-benzoic acid revealed that the adsorbed acid is rapidly metabolized (15% conversion in 10 s), suggesting the presence of a highly active enzymatic system. The major product could be identified as N-benzoylserine by use of [2H5]-benzoic acid, and mass spectrometry linked to a gas chromatograph (GC-MS) or a liquid chromatograph (LC-MS), for analysis of the metabolites. The conjugation of benzoic acid with an amino acid apparently represents a novel mode for the rapid deactivation of odorant molecules. Received 26 March 2001.  相似文献

Summary. Young larvae of Adalia bipunctata search an area more intensively when exposed to the odour from other larvae feeding on aphids than when exposed only to the odour of aphids. In an olfactometer young larvae were significantly attracted either to the odour of crushed aphids or larvae feeding on aphids, but not to that of aphids, larvae, larvae plus aphids or larvae feeding on an artificial diet. That is, the change in searching behaviour appears in response to a volatile released by aphids when attacked. The odour released by crushed aphids is made up entirely of aphid alarm pheromone, β-farnesene. It is likely that the adaptive significance of this response is that it increases the ability of larvae to locate larvae that have already caught prey. By sharing the aphid kill of another larva it is likely that a first instar ladybird larva greatly increases its probability of surviving to the next instar. It is suggested that this social feeding is facilitated by egg clustering, which also may additionally account for why aphidophagous ladybirds lay their eggs in clusters. Summary. The Na+, K+-ATPase of the Monarch butterfly (Danaus plexippus) is insensitive to the inhibition by cardiac glycosides due to an amino acid replacement: histidine instead of asparagine at position 122 of the α-subunit representing the ouabain binding site. By PCR amplification of the DNA sequence of this site, a PCR product of 270 bp was obtained from DNA extracted from Danainae species (Danaus plexippus, D. chrysippus, D. gillipus, D. philene, D. genutia, Tirumala hamata, Euploea spp., Parantica weiskei, P. melusine), Sphingidae (Daphnis nerii) and mimics of milkweed butterflies (Hypolimnas missipus, Limenitis archippus and L. arthemis, Nymphalidae). Analysis of the nucleotide sequences revealed that the single point mutation in the ouabain binding domain (AAC-Asn for CAC-His) was present only in Danaus plexippus, but not in the other species investigated. Since these milkweed butterflies also store cardenolides, other structural modifications of the Na+, K+-ATPase may have occurred or other strategies of cardenolide tolerance have been developed.
