Altemaria alternata were isolated from wheat seeds collected from villages in Tianshui area of Gansu Province where Kaschin-Beck disease is prevalent. TLC, IR and UV analyses of metabolic extract of A. alternata show that A. alternata produce main secondary metabolites, including alternariol, alternariol methyl ether, and tenuazonic acid.There was a significant decrease in Se-GSH-Px and SOD activities, and an acceleration in lipid peroxidation in rats fed with a diet containing metabolites produced by A. alternata.  相似文献

Armillaria sp. F022, a white rot fungus isolated from tropical rain forest (Samarinda, Indonesia) was used to biodegrade naphthalene in cultured medium. Transformation of naphthalene by Armillaria sp. F022 which is able to use naphthalene, a two ring-polycyclic aromatic hydrocarbon (PAH) as a source of carbon and energy was investigated. The metabolic pathway was elucidated by identifying metabolites, biotransformation studies and monitoring enzyme activities in cell-free extracts. The identification of metabolites suggests that Armillaria sp. F022 initiates its attack on naphthalene by dioxygenation at its C-1 and C-4 positions to give 1,4-naphthoquinone. The intermediate 2-hydroxybenzaldehyde and salicylic acid, and the characteristic of the meta-cleavage of the resulting diol were identified in the long-term incubation. A part from typical metabolites of naphthalene degradation known from mesophiles, benzoic acid was identified as the next intermediate for the naphthalene pathway of this Armillaria sp. F022. Neither phthalic acid, catechol and cis,cis-muconic acid metabolites were detected in culture extracts. Several enzymes (manganese peroxidase, lignin peroxidase, laccase, 1,2-dioxygenase and 2,3-dioxygenase) produced by Armillaria sp. F022 were detected during the incubation.  相似文献

A significant outbreak of fishy odor occurred in a reservoir located in Inner Mongolia, China, in the winter of 2011, and the odor rating, algal density and concentrations of some potential odorous compounds were monitored over a period of two months. The peak odor rating of the fishy odor was 7 according to flavor profile analysis. Among the dominant algal species (two diatom and one chrysophyte species) observed during the survey, the chrysophyte Dinobryon sp. was the most abundant species, with the peak density recorded at 88,520 cells/mL. Seven potential algal metabolites including heptanal, 2,4-heptadienal, 2,4-decadienal, nonanal, 2-octenal, 2,6- nonadienal and hexanal were detected. The principal component analysis result showed that n-hexanal, n-heptanal and 2,4-decadienal, possibly the metabolites of diatoms, and 2,4-heptadienal, possibly the metabolite of Dinobryon sp., might have contributed to the fishy odor episode. This study demonstrated that the fishy odor episode in this reservoir might be caused by the abnormal growth of chrysophytes and diatoms under the ice-cover.  相似文献

To evaluate decolorization and detoxification of Azure B dye by a newly isolated Bacillus sp. MZS10 strain, the cultivation medium and decolorization mechanism of the isolate were investigated. The decolorization was discovered to be dependent on cell density of the isolate and reached 93.55%(0.04 g/L) after 14 hr of cultivation in a 5 L stirred-tank fermenter at 2.0 g/L yeast extract and 6.0 g/L soluble starch and a small amount of mineral salts. The decolorization metabolites were identified with ultra performance liquid chromatography-tandem mass spectroscopy(UPLC-MS). A mechanism for decolorization of Azure B was proposed as follows: the C=N in Azure B was initially reduced to –NH by nicotinamide adenine dinucleotide phosphate(NADPH)-dependent quinone dehydrogenase, and then the –NH further combined with –OH derived from glucose to form a stable and colorless compound through a dehydration reaction. The phytotoxicity was evaluated for both Azure B and its related derivatives produced by Bacillus sp. MZS10 decolorization, indicating that the decolorization metabolites were less toxic than original dye. The decolorization efficiency and mechanism shown by Bacillus sp. MZS10 provided insight on its potential application for the bioremediation of the dye Azure B.  相似文献

Chemical investigation of the deepwater sponge Macandrewia azorica, collected from the flanks of the Gettysburg and Ormonde Sea Mount, North Atlantic, from a depth of 600 m, has led to the isolation of a new sterol with an unusual side chain (1), along with S-methylergothioneine (2). The structures of compounds 1 and 2 were established by employing spectroscopic techniques (NMR, MS, UV, IR and polarimetry). This is the first report of metabolites of a sponge belonging to the genus Macandrewia.  相似文献

High abundance of algae and eutrophication were observed in mangrove wetlands and these were estimated to be associated with root exudates of some specific mangrove plants to a certain extent.Root exudates form allelopathic effects from mangroves.The main secondary metabolites of Aegiceras corniculatum had been detected to be organic phenolic acids.Gallic acid had been isolated and identified from A.corniculatum.The half-maximal inhibitory concentration of gallic acid on alge Cyclotella caspia was tested as 15.46mg/L.The effects on algal cell morphology were mainly shown as elongated cells,with no apparent cell inclusions,such as oil droplets,chloroplast.At a dose of 2mg/L,gallic acid had a stimulative effect on the specific growth rate of algae on day 3.The contents of malondialdehyde,superoxide dismutase,soluble carbohydrates and chlorophyll a in algal cells showed an overall "low promotion and high suppression".Our results could provide preliminary and valuable reference on the complex influences of mangroves on microecology and microbial communities in the rhizosphere system.  相似文献

The major constituent in the pygidial gland defensive fluid of the carabid beetle Ardistomis schaumii is (R)-(+)-limonene, whereas that of Semiardistomis puncticollis is (S)-(−)-limonene. This was an unanticipated result, since it is not very common to find the opposite enantiomers of the same compound among the secondary metabolites of related species. Moreover, the glandular liquid of A. schaumii contains 1,8-cineole, and that of S. puncticollis has β-pinene, β-phellandrene, sabinene, and p-cymene. Of about 500 carabid species that have been chemically investigated, this is the first report of the presence of such complex mixtures of monoterpenes in their defensive secretions.  相似文献

运用核磁共振一维氢谱研究不同浓度四溴双酚A(TBBPA)急性暴露与亚急性暴露对赤子爱胜蚓(Eisenia fetida)的代谢影响.通过设置不同剂量TBBPA进行急性暴露和亚急性暴露,运用正交偏最小二乘投影分析法(OPLS)比较急性和亚急性暴露下蚯蚓的代谢差异,研究蚯蚓代谢水平与暴露剂量的关系,以找出差异代谢物作为TBBPA毒性效应的代谢标志物.结果表明,急性暴露和亚急性暴露均能很好地区分不同剂量染毒组间的代谢差异,且TBBPA暴露剂量越大,蚯蚓体内代谢与空白组的差异越大.急性暴露和亚急性暴露后,亮氨酸、赖氨酸和甜菜碱等代谢物剂量随TBBPA剂量增加的变化趋势是一致的,亚急性暴露中的变化幅度大于急性暴露.TBBPA暴露剂量为100 mg·kg~(-1)时,赤子爱胜蚓受到毒害作用,体内代谢物变化显著,代谢平衡遭到破坏,差异代谢物对毒性效应的指示作用比较敏感.甜菜碱、亮氨酸和赖氨酸可作为标志性代谢物揭示TBBPA对赤子爱胜蚓在渗透压调节、能量供给以及免疫功能方面的毒性效应,为进一步探索TBBPA毒性机理提供理论基础.  相似文献

The bioremediation of a long-term contaminated soil through biostimulation and surfactant addition was evaluated. The concentrations of 1,1,1-trichloro-2,2-bis(4-chlorophenyl) ethane (DDT) and its metabolites 1,1-dichloro-2,2-bis(4-chlorophenyl) ethane (DDD) and 1,1-dichloro-2,2-bis(4-chlorophenyl) ethylene (DDE) were monitored during an 8-week remediation process. Physicochemical characterization of the treated soil was performed before and after the bioremediation process. The isolation and identification of predominant microorganisms during the remediation process were also carried out. The efficiency of detoxification was evaluated after each bioremediation protocol. Humidity and pH and the heterotrophic microorganism count were monitored weekly. The DDT concentration was reduced by 79% after 8 weeks via biostimulation with surfactant addition (B + S) and 94.3% via biostimulation alone (B). Likewise, the concentrations of the metabolites DDE and DDD were reduced to levels below the quantification limits. The microorganisms isolated during bioremediation were identified as Bacillus thuringiensis, Flavobacterium sp., Cuprivadius sp., Variovorax soli, Phenylobacterium sp. and Lysobacter sp., among others. Analysis with scanning electron microscopy (SEM) allowed visualization of the colonization patterns of soil particles. The toxicity of the soil before and after bioremediation was evaluated using Vibrio fischeri as a bioluminescent sensor. A decrease in the toxic potential of the soil was verified by the increase of the concentration/effect relationship EC50 to 26.9% and 27.2% for B + S and B, respectively, compared to 0.4% obtained for the soil before treatment and 2.5% by natural attenuation after 8 weeks of treatment.  相似文献

Defensiveness of honeybee colonies of Apis cerana and Apis mellifera (actively balling the wasps but reduction of foraging) against predatory wasps, Vespa velutina, and false wasps was assessed. There were significantly more worker bees in balls of the former than latter. Core temperatures in a ball around a live wasp of A. cerana were significantly higher than those of A. mellifera, and also significantly more when exposed to false wasps. Core temperatures of bee balls exposed to false wasps were significantly lower than those exposed to V. velutina for both A. cerana and for A. mellifera. The lethal thermal limits for V. velutina, A. cerana and A. mellifera were significantly different, so that both species of honeybees have a thermal safety factor in heat-killing such wasp predators. During wasps attacks at the hives measured at 3, 6 and 12 min, the numbers of Apis cerana cerana and Apis cerana indica bees continuing to forage were significantly reduced with increased wasp attack time. Tropical lowland A. c. indica reduced foraging rates significantly more than the highland A. c. cerana bees; but, there was no significant effect on foraging by A. mellifera. The latency to recovery of honeybee foraging was significantly greater the longer the duration of wasp attacks. The results show remarkable thermal fine-tuning in a co-evolving predator–prey relationship.  相似文献

采用小容量全萃取方法,研究了1株铜绿假单胞菌对BDP的降解特性.结果表明,该菌株对DBP具有高效降解能力,当DBP浓度为400 mg/L,投菌量为2 g/L时,t1/2为3.60 d;其降解过程完全符合一级反应动力学.DBP作为该菌生长的唯一碳源时,其降解过程包括快速生物吸附、解析、降解等几个阶段.实验还确认了存在邻苯二甲酸单丁酯和邻苯二甲酸2种中间产物,从而验证了DBP在双加氧水解酶作用下2步水解变为邻苯二甲酸的历程.  相似文献

采用乙醇提取、溶剂萃取法从芦苇中分离出2组能抑制蛋白核小球藻生长的化感物质. 该2组化感物质对蛋白核小球藻的半效应浓度(EC50)分别为0.68mg/L和2.65mg/L. 将前者用碱性氧化铝层析柱分离得到一组抑藻活性更强的化感物质和一组能够促进蛋白核小球藻生长的化感物质: 前者对蛋白核小球藻的EC50为0.50mg/L, 是目前报道的抑藻能力最强的化感物质; 后者在浓度为0.63mg/L时能显著促进蛋白核小球藻的生长. 芦苇化感物质可以用来控制淡水中有害藻类.  相似文献

A promising bacterial strain for biodegrading microcystin-LR (MC-LR) as the sole carbon and nitrogen source was successfully isolated from Lake Dianchi, China. The strain was identified as Sphingopyxis sp. USTB-05, which was the first isolated MCs-biodegrading Sphingopyxis sp. in China. The average biodegradation rate of MC-LR by Sphingopyxis sp. USTB-05 was 28.8 mg·L−1 per day, which was apparently higher than those of other bacteria reported so far. The optimal temperature and pH for both strain USTB-05 growth and MC-LR biodegradation were 30°C and 7.0, respectively. The release of MC-LR from the cyanobacterial cells collected from Lake Guishui and the biodegradation of MC-LR by both strain and cell-free extract (CE) were investigated. The results indicated that MC-LR with the initial concentration of 4.0 mg·L−1 in water was biodegraded by Sphingopyxis sp. USTB-05 within 4 d, while MC-LR with the initial concentration of 28.8 mg·L−1 could be completely removed in 3 h by CE of Sphingopyxis sp. USTB-05 containing 350 mg·L−1 protein. During enzymatic biodegradation of MC-LR, two intermediate metabolites and a dead-end product were observed on an HPLC chromatogram. Moreover, the similar scanning profiles of MC-LR and its metabolic products indicate that the Adda side-chain of MC-LR was kept intact in all products.  相似文献

提出并研究了真菌和细菌协同强化原位修复石油污染土壤.从中原油田石油污染土壤中筛选出刺孢小克银汉霉菌(Cunninghamella echinulata)和阴沟肠杆菌(Enterobacter cloacae),在泥浆体系中考察了Cun.echinulata和E.cloacae的生长及其对石油烃的降解过程.结果表明,混合培养体系中细菌和真菌的数量最高分别为其纯培养体系的3倍和20倍,细胞衰亡则被明显推迟;混合培养体系中石油烃的去除率高于真菌和细菌纯培养体系石油烃去除率的总和;采用多次接种新鲜菌种的培养方式能够强化混合培养体系对石油烃的降解.在石油烃污染土壤中接入由Cun.echinulata和E.Cloacae所组成的真菌-细菌微生物制剂实施原位修复,结果表明,Cun.echinulata和E.Cloacae的生长及其对石油烃的降解不受土著微生物的抑制,最适工艺条件为:水和木屑含量分别为25%和6%(质量分数),Cun.echinulata和E.cloacae接种量分别为2.5×104和2.5 × 107CFU/g.接种上述真菌-细菌40 d后石油烃去除率可达约65%,而土著微生物对石油烃去除率为16.0%.  相似文献

Hybridization between Aegilops geniculata, A. biuncialis and bread wheat Triticum aestivum has been evaluated during two seasons under simulated field conditions to estimate the field hybridization rate under Central Spain conditions. The mean frequencies of hybridization between A. biuncialis and A. geniculata with wheat were 0.34% and 0.31%, respectively. Data from 10 hybrid plants for each combination showed that hybrids can be partially fertile by backcrossing with wheat parent, with percent averages of 3.17 grains/spikelets for A. biuncialis × wheat hybrids and 2.87 grains/spikelets for A. geniculata × wheat hybrids. Self-pollination, although at very low rates, was also possible in hybrids. The potential risks associated with natural hybridization in the context of transgenic wheat cultivation are discussed.  相似文献

对二甲苯作为一种重要的苯系物化工原料,生产使用量非常大.进入人体的对二甲苯主要在肝脏中代谢,生成对甲基苄醇和对甲基苯甲酸.目前,关于对二甲苯及其代谢物的肝脏毒性数据仍不够充分.本研究以人肝癌细胞(HepG2)作为实验模型,在细胞层面评价了对二甲苯及其代谢产物(对甲基苄醇和对甲基苯甲酸)的毒性效应,着重探讨了化合物对细胞活力、细胞膜完整性、细胞凋亡的影响.研究结果显示,对二甲苯及其代谢产物对HepG2细胞的急性毒性大小顺序为:对甲基苄醇对甲基苯甲酸对二甲苯.显然,对二甲苯在机体内发生代谢转化后具有毒性增加的趋势.另外,对二甲苯及其代谢产物暴露能够引起细胞膜破损,导致细胞坏死,并诱导一定程度的细胞凋亡现象.该研究发现为对二甲苯作为常见溶剂的生产使用提供了安全性评价的重要科学数据.  相似文献

重金属污染会影响微生物组成和群落结构,其与微生物的相互关系一直是重金属污染生态学研究的热点.河南省新乡市是中国著名的电池生产城市,因电池生产废水污灌造成严重的土壤重金属污染.目前有关新乡市长期灌溉电池废水造成的重金属污染对微生物群落组成和代谢功能的影响鲜见报道.采集了新乡市某长期生产电池厂旁3个重金属污染位点,重金属含量测定表明Cd和Pb超过相关标准,分别超标34~66倍和1.5~2.32倍.采用高通量测序分析了细菌和真菌群落组成,结果表明细菌由芽孢杆菌属、节杆菌属、鞘氨醇单胞菌属和类诺卡氏菌属等优势属,真菌由油壶菌属、小不整球壳属、Gibellulopsis和被孢霉属等优势属组成,表明经过长期的重金属胁迫,新乡长期重金属污染土壤富含重金属抗性细菌和真菌.环境因子和微生物群落相关性分析表明,总量Cu、DTPA提取态Cu和水溶态Pb显著影响细菌群落组成,总量Cd、Ni、Pb、Zn、DTPA提取态Cu和水溶态Pb显著影响真菌群落组成.目前,关于重金属污染对微生物群落代谢影响的研究较少,采用非靶向代谢组学分析表明,不同位点间代谢产物具有差异.通路富集分析表明,这些差异代谢物涉及代谢过程、环境信息处理和翻译等遗传信息加工等,在微生物缓解重金属导致的胁迫和环境适应中可能起到作用.  相似文献

Soil contamination with tetrabromobisphenol A(TBBPA) has caused great concerns;however, the presence of heavy metals and soil organic matter on the biodegradation of TBBPA is still unclear. We isolated Pseudomonas sp. strain CDT, a TBBPA-degrading bacterium, from activated sludge and incubated it with ~(14)C-labeled TBBPA for 87 days in the absence and presence of Cu~(2+)and humic acids(HA). TBBPA was degraded to organic-solvent extractable(59.4% ± 2.2%) and non-extractable(25.1% ± 1.3%) metabolites,mineralized to CO_2(4.8% ± 0.8%), and assimilated into cells(10.6% ± 0.9%) at the end of incubation. When Cu~(2+)was present, the transformation of extractable metabolites into non-extractable metabolites and mineralization were inhibited, possibly due to the toxicity of Cu~(2+)to cells. HA significantly inhibited both dissipation and mineralization of TBBPA and altered the fate of TBBPA in the culture by formation of HA-bound residues that amounted to 22.1% ± 3.7% of the transformed TBBPA. The inhibition from HA was attributed to adsorption of TBBPA and formation of bound residues with HA via reaction of reactive metabolites with HA molecules, which decreased bioavailability of TBBPA and metabolites in the culture. When Cu~(2+)and HA were both present, Cu~(2+)significantly promoted the HA inhibition on TBBPA dissipation but not on metabolite degradation. The results provide insights into individual and interactive effects of Cu~(2+)and soil organic matter on the biotransformation of TBBPA and indicate that soil organic matter plays an essential role in determining the fate of organic pollutants in soil and mitigating heavy metal toxicity.  相似文献

吲哚是一种典型的氮杂环芳烃污染物,在焦化废水和畜牧废水中大量存在.本研究从近海泥沙中分离纯化得到一株高效吲哚降解菌DCX,经16S rRNA基因序列比对分析,鉴定其为普罗维登斯菌属(Providencia sp.).该菌株能够以吲哚为唯一碳源,在28 h内将100 mg·L~(-1)吲哚完全降解.液相色谱/飞行时间-质谱联用(LC/TOF-MS)的结果表明,靛蓝、靛红、靛红酸及邻氨基苯甲酸是菌株DCX降解吲哚过程中的中间产物.此外,本研究中发现,外加营养物质可以促进菌株DCX降解吲哚,特别是加入酵母浸粉后,反应体系中会产生大量靛蓝.利用表面响应法确定菌株DCX转化吲哚合成靛蓝的最优条件为:吲哚207.49 mg·L~(-1),酵母浸粉2.9 g·L-1,接菌量4.23%(V/V),并且在最优条件下,靛蓝产率达到最高(9.90%),比初始条件提高了4.38倍.  相似文献
