The proposed conversion from gasoline powered automobiles to diesel powered vehicels has prompted the Environmental Protection Agency to evaluate the potential health effects associated with exposure to diesel emissions. At present, there is no direct epidemiological link between this exposure and human health. Therefore, a research program was constructed to compare the health effects associated with diesel emissions with those from other emission sources for which epidemiological information was available. The emission sources chosen were cigarette smoke, roofing tar, and coke oven. An additional comparative emission source which was a gasoline catalyst engine. Respirable particles from a variety of combustion sources have the potential of being carcinogenic and mutagenic. The objective of these studies was to determine the relative biological activity of the organic material adsorbed on these particles in both in vitro mutagenesis and in vitro and in vivo bioassays. The organic extracts from the following series of emission sources were quantitatively bioassayed in a matrix of tests for their carcinogenic and mutagenic activity: (1) a light-duty Oldsmobile diesel 350 engine; (2) a heavy-duty Caterpillar diesel engine; (3) a light-duty Nissan engine; (4) a Volkswagen Rabbit diesel engine; (5) cigarette smoke; (6) roofing tar; (7) coke oven; and (8) a gasoline catalyst Mustang. The test matrix consisted of the following bioassay: reverse mutation in Salmonella typhimurium; mitotic recombination in Saccharomyces cerevisiae; DNA damage in Syrian hamster embryo cells (SHE); sister chromatid exchange in CHO cells; gene mutation in L5178Y mouse lymphoma cells, Balb/c 3T3 mouse embryo fibroblasts and CHO cells; viral enhancement of SHE cells; oncogenic transformation in Balb/c 3T3 cells; and skin tumor initiation in SENCAR and C57 black mice. Extracts from emissions of four diesel engines, a gasoline engine and three related environmental samples were tested in four in vitro assay systems designed to detect carcinogenic or mutagenic activity of chemicals. Samples from three of four diesel extracts, the gasoline engine, and all three related samples were positive in an enhancement of viral transformation assay. Two diesel samples, the gasoline engine extract and extract from coke oven emissions were positive for mutation induction in Chinese hamster ovary cells. Only the gasoline engine extract and the coke oven sample were positive in a DNA fragmentation assay using alkaline sucrose gradients. Experiments using chemical transformation of Syrian hamster embryo cells as an assay method have not been completed.  相似文献

The Saccharomyces cerevisiae D3 recombinogenic assay, the assay for forward mutagenesis in L5178Y mouse lymphoma cells, and the sister chromatid exchange (SCE) assay using Chinese hamster ovary cells were used to evaluate the in vitro mutagenic and DNA-damaging effects of eight samples of diesel engine emissions and related environmental emissions. The recombinogenic assay was not sufficiently sensitive for this evaluation, but mutagenicity was detected in the L5178Y mutagenesis assay following exposures of the cells to all of the emission samples, and DNA damage in the SCE assay was induced by most of the emission samples in the presence and absence of metabolic activation. The observation of positive results in the absence of activation indicated that the samples contained substances that were direct-acting mutagens and DNA-damaging agents.  相似文献

Skin tumors can be induced by the sequential application of a subthreshold dose of a carcinogen (initiation phase), followed by repetitive treatment with a noncarcinogenic tumor promoter. There is a very good dose-response relationship between the induction of the number of papillomas per mouse at early times (10 to 20 weeks) by either tumor initiators and promoters and the final carcinoma incidence after a longer latency (20 to 50 weeks) in SENCAR mice. This system not only can be used to determine the tumor initiating and promoting activities of a compound but if the agent is given repeatedly by itself one can also determine if it is a complete carcinogen, i.e., if it has both tumor initiating and promoting activity. In addition, if the agent is given concurrently with a known complete carcinogen or a tumor initiator one can also determine if the agent has cocarcinogenic or cotumor initiating activity or even possibly anticarcinogenic activity. Likewise, if the agent is given concurrently with a known tumor promoter one can determine if the agent has copromoting or antipromoting activity. Using the SENCAR skin carcinogenesis system we have undertaken the determination of the skin carcinogenic, cocarcinogenic, tumor initiating and promoting activities of various diesel emission particle extracts as well as for comparative purposes, standards such as benzo(a)pyrene and 12-0-tetradecanoylphorbol-13-acetate and extracts of emissions from a gasoline engine, roofing tar, coke oven and cigarette smoke condensate. Most of the studies are still in progress but some preliminary data is available on the comparative tumor initiating activities of the various samples at 14 weeks. Caterpillar diesel and cigarette smoke condensate were essentially without activity, whereas the Mustang gasoline catalyst and Olds diesel gave extremely low values although they were slightly above background. Roofing tar, coke oven and Nissan Diesel all gave moderate activity at high doses (1 to 10 mg) but when compared to benzo(a)pyrene were relatively low values.  相似文献

Particulate extracts from six different environmental emission sources were assayed for genotoxic activity in mouse BALB/c 3T3 clone A31-1 cells. All compounds were tested simultaneously for both transforming and mutagenic (induction of ouabain-resistance) potential with and without exogenous metabolic activation in the form of a 9000 × g postmitochondrial hepatic supernatant fraction from Aroclor-1254 induced Fischer 344 rats. Dichloromethane particulate extracts from the exhaust of two light duty diesel engines (Oldsmobile and Nissan), one heavy duty diesel engine (Caterpillar) and one late model gasoline engine (Mustang II) were assayed in an identical manner to particulate extracts from the emissions of a roofing tar pot and a coke oven. No clear dose-dependent responses were observed, but several of the samples showed significant transforming and mutagenic activity. A qualitative ranking system showed the activity of these particulate extracts for either mutagenesis or transformation was: coke oven = Mustang II gasoline engine > Nissan diesel engine > roofing tar. Particulate extracts from the Oldsmobile diesel engine and the Caterpillar diesel engine showed essentially no activity.  相似文献

Mutagenic activity in diesel emission particulate extracts was detected by the Salmonella typhimurium/microsome assay. Direct-acting mutagens as well as promutagens requiring metabolic activation were detected. The extracts were fractionated into acidic, basic, and neutral fractions, and the neutral fraction was chromatographed into seven subfractions. Differences in the mutagenic potency of these fractions and subfractions were determined by the Salmonella assay. Fractions containing as yet unidentified compounds, but not polynuclear aromatic hydrocarbons, were found to make a major contribution to mutagenic activity of the extracts.  相似文献

Female Swiss mice were exposed 8 h/day to diesel exhaust for 1, 3, and 7 weeks. Urine was collected overnight for 4 days prior to sacrifice while the mice continued to be exposed for eight hours during the day. The presence of mutagens was determined by the Ames Salmonella test. One hour prior to sacrifice each mouse received 1 mg/kg colcemide. After sacrifice, the marrow from each femur was obtained. The marrow from one femur was used to prepare slides for metaphase analysis and the other for micronuclei assay. Other mice received IP 50 mg/kg cyclophosphamide 24 h prior to sacrifice or 1 μmole/kg benzo(a)pyrene in each of four daily doses prior to sacrifice and served as positive controls. The Ames Salmonella assay of the unconcentrated urine after 1, 3, and 7 weeks and concentrated urine after 7 weeks exposure to diesel exhaust did not significantly vary from clean air controls. In the micronucleus test, and metaphase analysis, cyclophosphamide produced a strong positive response and the 7 week diesel exposure was not different from clean air controls.  相似文献

The inertization of sewage sludge in ceramic matrices to be used in structural or red ceramic material for buildings has proved to be a good case of reuse of waste material. However, its practical application has not yet been fully implemented in real-case scenarios, and environmental concern seems to be the main hurdle to overcome for its definitive massive approval by the building industry. In this contribution, air emissions related to the sintering of ceramic bricks made of mixtures of clay with some percentage of sewage sludge have been analyzed (in terms of gases, suspended particles and odors). Tests conducted during this work have shown higher VOC emissions in samples with some percentage of sludge in their composition (still under the regulated emission limit values), and some of them (mercaptans) are associated with odor nuisances. Besides, limit emissions values were exceeded by three inorganic pollutants (suspended particles, NOx and HCl). Measurements in an industrial scenario test showed a high variability in air pollution emissions, suggesting the need of in situ testing for definitive implementation. With the experience collected in this and several previous works in the area of emissions related to the production of added sewage sludge ceramic, some guidelines and recommendations are given to minimize the environmental impact of ceramic production plants implementing this particular waste revalorization process. Guidelines cover different aspects: workplace implementation of gas cleaning equipment; geographical context and local wind pattern analysis; monitoring of emission and immission levels; and information policy through social control participation procedures for reporting of nuisance episodes.  相似文献

Integration has become an important element of natural resource management over recent decades, and managers are now required to consider the social, economic, ecological, and bio-physical effects of alternative management interventions. The problem of managing across many issues is sometimes tackled by putting together a number of individual models. For this approach to work effectively, attention must be paid to both the technical details of the interactions between system components and also to the processes through which these integrated models are developed. This paper explores technical and social aspects of the development of integrated models for environmental management, and discusses two cases within which open modelling and interface prototyping processes were undertaken. The results of this exploration suggest that, for models to be used and accepted widely in integrated environmental management, developers must undertake a process that involves stakeholders and potential users, that exploits the current knowledge, and that illustrates the influence of uncertainty in the technical knowledge. This requires strict attention to the social and technical process of modelling, as well as additional skills in group facilitation and shared vision exploration, so that user expectations can be developed and met in such a way that the best information available is used by decision makers.  相似文献

This study examines the relationship between national-level militarism and consumption-based carbon dioxide emissions. We analyze panel data from 1990 to 2010 for 81 nations to determine whether the magnitude of the effects of (1) military expenditures as percent of total gross domestic product and (2) military personnel as percent of total labor force on carbon emissions change over time. Results of two-way fixed effects models highlight the temporal stability of the environmental impacts of both national-level military characteristics. The findings also reveal that the effect of military expenditures on emissions is larger in the more developed OECD nations than in the developing non-OECD nations. Overall, the results support the treadmill of destruction perspective, which suggests that the nations’ militaries are an important social institution to consider in sustainability science research on the human drivers of global environmental change.  相似文献

Corporate Environmental Responsibility (CER) is the subsystem of Corporate Social Responsibility (CSR), which continues to increase prominence in the global economy. Nowadays, CER becomes more important in corporate sustainable development. Based on Carroll’s pyramid mode of CSR, this article explores the systematic feature of CER. From a systematic perspective, it sorts out its elements, structure, function, and principles of CER. And the four levels of economic, legal, ethical, and charitable structure are emphasized. The objective of this paper is to discuss the main barriers for modelling and integrating the environmental performances in the automotive concept design. Incorporating environmental assessment in the early design phase of a vehicle component is known as an important challenge that car makers need to face in order to develop more sustainable design solutions; in this regard, the Life Cycle Assessment is the most widespread methodology for the environmental assessment and comparison of alternatives. The present work illustrates the combination of such methodology with the traditional design procedure at two different levels of the component design phase, material choice and concept design. In particular, the potential benefits originated by a lightweight solution for the automotive component Throttle Body are evaluated by considering environmental and technical implications at the same level. The case study shows that a multi-disciplinary approach for design effectively allows the integration of the environmental issue in the company’s established procedures. However, interpretation of results is still a challenging aspect due to the inevitable contradicting elements which should not discourage to develop comprehensive sustainability assessment within the early design stage.  相似文献

In the analysis of how environmental regulation affects the comparative advantage of trade, existing literature ignores industry’s inherent heterogeneity, which draws remarkably different conclusions. In view of this, the paper analyzed the mechanism of environmental regulation on the export quality of different industries from the perspective of factor input structure heterogeneity. Based on the panel data of China’s manufacturing industry, the paper used the system generalized method of moments method to examine the heterogeneity influence of environmental regulation on manufacturing export quality. The study found that, first, environmental regulation affected the export quality upgrade of the manufacturing sector through offset effect and compensation effect, and the direction of the impact would mainly depend on the industry’s factor input structure. Second, for industries with larger fixed-asset investment (FAI) ratio in the factor input structure, the current environmental regulation policy was not conducive to the export quality upgrading of the industries. However, there was a significant U-shaped dynamic relationship between them. As environmental regulations became stricter, when regulatory stringency went beyond the inflection point, the policy would promote the upgrading of export quality. But for industries with smaller proportion of FAI, environmental regulation exerted a favorable impact on the export quality upgrade, following a J-shaped marginal growth curve. Third, for industries with different factor input structure, their export quality had been effectively upgraded as expected by factors like human capital investment, independent R&D, technology introduction, and foreign direct investment; but raising per capita capital stock and expanding enterprise size did not produce significant direct impact on export quality upgrade. These conclusions remained robust after using different measurement methods and replacing with other variables. Therefore, this paper suggests that governments should take industry heterogeneity into consideration and formulate differentiated hierarchical environmental policies. Besides, they should strengthen the enforcement of the current environmental regulation policies. By doing so, enterprises are forced to improve their technology and product quality so that they can better cope with rising compliance costs, eliminate backward industries, and resolve excess capacity. In this way, the economic structure would be transformed and upgraded from the supply side.  相似文献
