Agricultural crops can be either a source or a sink of ammonia (NH3). Most NH3 exchange models developed so far do not account for the plants nitrogen (N) metabolism and use prescribed compensation points. We present here a leaf-scale simplified NH3 stomatal compensation point model related to the plants N and carbon (C) metabolisms, for C3 plants. Five compartments are considered: xylem, cytoplasm, apoplasm, vacuole and sub-stomatal cavity. The main processes accounted for are the transport of ammonium (NH4+), NH3 and nitrate (NO3) between the different compartments, NH4+ production through photorespiration and NO3 reduction, NH4+ assimilation, chemical and thermodynamic equilibriums in all the compartments, and stomatal transfer of NH3.The simulated compensation point is sensitive to paramaters related to the apoplastic compartment: pH, volume and active transport rate. Determining factors are leaf temperature, stomatal conductance and NH4+ flux to the leaf. Atmospheric NH3 concentration seem to have very little effect on the compensation point in conditions of high N fertilization. Comparison of model outputs to experimental results show that the model underestimates the NH3 compensation point for high N fertilization and that a better parametrisation of sensitive parameters especially active trasport rate of NH4+ may be required.  相似文献

NH 4 + is typically an inhibitor to hydrogen production from organic wastewater by photo-bacteria. In this experiment, biohydrogen generation with wild-type anoxygenic phototrophic bacterium Rhodobacter sphaeroideswas found to be sensitive to NH 4 + due to the significant inhibition of NH 4 + to its nitrogenase. In order to avoid the inhibition of NH 4 + to biohydrogen generation by R. sphaeroides, a glutamine auxotrophic mutant R. sphaeroides AR-3 was obtained by mutagenizing with ethyl methane sulfonate. The AR-3 mutant could generate biohydrogen efficiently in the hydrogen production medium with a higher NH 4 + concentration, because the inhibition of NH 4 + to nitrogenase of AR-3 was released. Under suitable conditions, AR-3 effectively produced biohydrogen from tofu wastewater, which normally contains 50–60 mg/L NH 4 + , with an average generation rate of 14.2 mL/L·h. The ForNBM was applied to the Nashwaak Experimental Watershed Project in central New Brunswick, Canada. The data represented a mixed hardwood site and included information about nutrient leaching, foliage and wood biomass, leaf fall, and ancillary information required for model initialization. Ancillary information included forest cover type, stand density, forest floor depth, soil rooting depth, soil texture, soil substrate type, initial amounts of biomass and N, S, Ca, Mg, and K content in foliage, wood, and roots, and mineral soil nutrient contents in soil solution, on ion-exchange sites, and in the soil.The authors were able to calibrate the model with existing data. The model simulated observed monthly leaching reasonably well. The r2-values of model simulations compared with field observations of monthly leaching of NO3_N, NH4+_N, Ca, Mg, and K were 0.78, 0.7, 0.78, 0.84, and 0.75, respectively. Modeled multi-year cumulative leaching of NO3_N, NH4+_N, Ca, Mg, and K compared with actual values gave r2-values close to one for all cases considered.The results also showed that soil nutrient leaching had increased approximately five-fold for NO3_N, 80% for NH4+_N, 71% for K, 20% for Ca, and 14% for Mg during the 3 years following a stem-only harvest operation applied watershed-wide. However, the model simulation showed that increased nutrient leaching during the 11 years following the stem-only harvest was small compared with the amounts removed in the biomass during the harvest. Increased nutrient leaching following harvesting did not appear to impact site productivity over the long term.  相似文献

Metabolism of two abundant echinoderm species constituting 99.6% of the epibenthic megafauna in the Santa Catalina Basin, off southern California, USA was measured at 1 300 m during the 1979 “Bathyal Expedition”. Specimens of the ophiuroid Ophiophthalmus normani and the holothurian Scotoplanes globosa, collected by the submersible “Alvin”, were individually placed in respirometers, and measured in situ for O2 consumption and ammonium excretion rates. For O. normani, weight-specific O2 consumption rates decreased with increasing weight and were of comparable magnitude to rates of deep-sea and shallow-water ophiuroids; excretion rates were highly variable. Population O2 consumption and excretion rates for O. normani (estimated from size-frequency distribution, abundance, and rate regression equations) were 1 129.28 μl O2 m-2 h-1 and 27.30 nmol NH 4 + m-2 h-1. Weight-specific O2 consumption and ammonium excretion rates of S. globosa decreased with increasing weight and were of comparable magnitude to rases of shallow-water holothurians. Population O2 constimption and excretion rates of S. globosa were 1.38 μl O2 m-2 h-1 and 4.86 nmol NH 4 + m-2 h-1. Combined population O2 consumption rates for O. normani and S. globosa are of comparable magnitude to that of the sediment community and plankton in the benthic boundary layer (sediment and overlying 50 m water column) of the Santa Catalina Basin.  相似文献

Acute toxicity of ammonia was determined for cultured larval, postlarval, and wild adult lobsters (Homarus americanus) in 1988. Ammonia tolerance was found to increase with ontogenetic development. Based on 96-h LC50 values of 58 mg l–1 NH4 + + NH3 l–1 seawater (0.72 mg NH3 l–1) for Stage I larvae, 87 mg NH4 + + NH3 l–1 (1.7 mg NH3 l–1) for Stage II larvae, 125 mg NH4 + + NH3 l–1 (2.13 mg NH3) for Stage III larvae, 144 mg NH4 + + NH3 l–1 (2.36 mg NH3 l–1) for Stage IV postlarvae, 377 mg NH4 + + NH3 l–1 (5.12 mg NH3 l–1) for adult lobsters at 5°C and 219 mg NH4 + + NH3 l–1 (3.25 mg NH3 l–1) for adult lobsters at 20°C, recommendations for safe levels of total ammonia and un-ionized ammonia were calculated using an application factor of 0.1. Effects of ammonia on osmoregulatory capacity were studied on postlarvae and adults. Ability of postlarvae and adults to hyper-regulate in low-salinity media decreased after exposure to ammonia. In postlarval lobsters, osmoregulatory capacity was significantly affected in ammonia concentrations exceeding 32 mg l–1. Osmoregulatory capacity in adult lobsters (5 and 20°C) was affected at 150 mg l–1. In postlarval lobsters, a minimum exposure time of 12 h was required to impair osmoregulatory capacity. The decrease in hemolymph osmotic pressure was caused by lower hemolymph sodium concentrations. The presence of ammonia in the external medium could markedly affect the Na+/NH4 + transport mechanism by permanently, temporarily, or partially impairing the transport sites for sodium.  相似文献

Microcosms containing different densities of Corophium volutator, ranging from 0 to 6000 ind m-2, were incubated in a flow-through system. Benthic fluxes of CO2, O2, NO3 - and NH4 + were measured regularly. Thirteen days after setup the microcosms were sacrificed and sediment characteristics, pore water NO3 -, NH4 + and exchangeable NH4 + concentrations, and potential nitrification activity were measured. The presence of C. volutator increased overall mineralization processes due to burrow construction and irrigation. The amphipods increased the ratio CO2/O2 fluxes from 0.73 to 0.86 in microcosms inhabited by 0 and 6000 ind m-2, respectively. Burrow ventilation removed NH4 + from the sediment, which was nitrified in the oxic layer and transported NO3 - to the burrow sediment, where denitrification potential was enhanced. Nitrification and total denitrification rates (denitrification of NO3 - coming from the overlying water and of NO3 - generated within the sediment) were calculated and discussed. Bioturbation by C. volutator increased both nitrification and denitrification, but denitrification was stimulated more than nitrification. Denitrification of NO3 - coming from the overlying water was stimulated 1.2- and 1.7-fold in microcosms containing 3000 and 6000 ind m-2 relative to control microcosms. The presence of C. volutator (6000 ind m-2) stimulated nitrogen removal from the system, as dinitrogen, 1.5-fold relative to non-bioturbated microcosms. C. volutator individuals used in our study were collected from Norsminde Fjord, Denmark, in 1990.  相似文献

V2O5-WO3/TiO2 catalyst was poisoned by impregnation with NH4Cl, KOH and KCl solution, respectively. The catalysts were characterized by X-ray diffraction (XRD), inductively coupled plasma (ICP), N2 physisorption, Raman, UV-vis, NH3 adsorption, temperature-programmed reduction of hydrogen (H2-TPR), temperature-programmed oxidation of ammonia (NH3-TPO) and selective catalytic reduction of NO x with ammonia (NH3-SCR). The deactivation effects of poisoning agents follow the sequence of KCl>KOH?NH4Cl. The addition of ammonia chloride enlarges the pore size of the titania support, and promotes the formation of highly dispersed V = O vanadyl which improves the oxidation of ammonia and the high-temperature SCR activity. K+ ions are suggested to interact with vanadium and tungsten species chemically, resulting in a poor redox property of catalyst. More importantly, potassium can reduce the Brønsted acidity of catalysts and decrease the stability of Brønsted acid sites significantly. The more severe deactivation of the KCl-treated catalyst can be mainly ascribed to the higher amount of potassium resided on catalyst.  相似文献

Ambient concentrations of urea in the inner Oslofjord, Norway, showed a pronounced yearly cycle in 1980, with values in the range 0.1 to 10.0 μg-at N l-1; this cycle resemble that of ammonia although urea concentrations were usually lower. The uptake of urea by phytoplankton was investigated using 15N. Urea was usually a less important N source than NH 4 + , and accounted for 0 to 53% (mean 19%) of summed NH 4 + +NO 3 - + urea uptake rates from April to October. Absolute as well as relative (specific) uptake rates of urea were higher in the summer (June–August) than at other times. Uptake of urea was inhibited by NH 4 + concentrations higher than 1 to 2 μg-at N l-1. The summed NH 4 + +NO 3 - + urea uptake rate was exponentially related to temperature.  相似文献

Effects of benthic macrofauna (Corophium volutator, Hydrobia sp., Nereis virens) on benthic community metabolism were studied over a 65-d period in microcosms kept in either light/dark cycle (L/D-system) or in continuous darkness (D-system). Sediment and animals were collected in January 1986 in the shallow mesohaline estuary, Norsminde Fjord, Denmark. The primary production in the L/D-system after 10 d acted as a stabilizing agent on the O2 and CO2 flux rates, whereas the D-system showed decreasing O2 and CO2 flux throughout the period. Mean O2 uptake over the experimental period ranged from 0.38 to 1.24 mmol m–2 h–1 and CO2 release varied from 0.80 to 1.63 mmol m–2 h–1 in both systems. The presence of macrofauna stimulated community respiration rates measured in darknes, 1.4 to 3.0 and 0.9 to 2.0 times for O2 and CO2, respectively. In contrast, macrofauna lowered primary production. Gross primary production varied from 1.06 to 2.26 mmol O2 m–2 h–1 and from 1.26 to 2.62 mmol CO2 m–2 h–1. The community respiratory quotient (CRQ, CO2/O2) was generally higher in the begining of the experiment (0–20 d, mean 1.89) than in the period from Days 20 to 65 (mean 1.38). The L/D-system exhibited lower CRQ (ca. 1) than the D-system. The community photosynthetic quotient varied for both net and gross primary production from 0.64 to 1.03, mean 0.81. The heterotrophic D-system revealed a sharp decrease in the sediment content of chlorophyll a as compared to the initial content. In the autotrophic L/D-system, a significant increase in chlorophyll a concentration was observed in cores lacking animals and cores with C. volutator (The latter species died during the experiment). Due to grazing and other macrofauna activities other cores of the L/D-system exhibited no significant change in chlorophyll a concentration. Community primary production was linearly correlated to the chlorophyll a content in the 0 to 0.5 cm layer. Fluxes of DIN (NH4 ++NO2 +NO3 ) did not reveal significant temporal changes during the experiment. Highest rates were found for the cores containing animals, mainly because of an increased NH4 + flux. The release of DIN decreased significantly due to uptake by benthic microalgae in the L/D-system. No effects of the added macrofauna were found on particulate organic carbon (POC), particulate organic nitrogen (PON), total carbon dioxide (TCO2) and NH4 + in the sediment. The ratio between POC and PON was nearly constant (9.69) in all sediment dephts. The relationship between TCO2 and NH4 + was more complex, with ratios below 2 cm depth similar to those for POC/PON, but with low ratios (3.46) at the sediment surface.  相似文献

The seasonal variation in biogenic fluxes of NH4 +, PO4 3? and SiO2 calculated from the nutrient excretion rates of dominant bivalves (Ruditapes philippinarum and Arcuatula [=Musculista] senhousia), and pore-water nutrient (NH4 +, PO4 3?, SiO2 and NO3 ? + NO2 ?), sedimentary acid-volatile sulfide (AVS) and benthic chlorophyll-a (Chl-a) concentrations was assessed on an intertidal sandflat in the Seto Inland Sea (Japan) from summer 1994 to autumn 1995. In spite of the large variability between sampling dates and stations, significant correlations between biogenic nutrient fluxes and pore-water nutrient concentrations were found, suggesting a seasonal linkage between bivalve-mediated biological processes and chemical features of sediments. This linkage was stronger in surface (0–0.5 cm) than subsurface (0.5–2 cm) sediments, consistent with the autoecological characteristics of R. philippinarum and A. senhousia inhabiting the uppermost sediment layer. Significant temporal variation in pore-water NO3 ? + NO2 ?, sedimentary AVS and benthic Chl-a concentrations was also found, which was related to both occasional extreme events (e.g., dystrophy) and alternating periods of production and decomposition. This study may serve twofold in (1) contributing to unravel the ecological structure and functioning of natural tidal flats, and the scale of seasonal variability in biotic and sedimentary parameters and (2) providing useful information for assessing the effectiveness of the physico-chemical and biological structure of artificial tidal flats which are growing in number and extension worldwide.  相似文献

To determine the impact of photosynthesis and transpiration on nitrogen removal in wetlands, an artificial wetland planted with reeds was constructed to treat highly concentrated domestic wastewater. Under different meteorological and hydraulic conditions, the daily changes of photosynthesis and transpiration of reeds, as well as nitrogen removal efficiency were measured. It was found that net photosynthesis rate per unit leaf area was maintained on a high level (average 19.0 μmol CO2/(m2·s)) from 10:00 to 14:00 in July 2004 and reached a peak of 21.1 μmol CO2/(m2·s) when Photon Flux Density was high during the day. Meanwhile, TN and NH4 +-N removal efficiency rose to 79.6% and 89.6%, respectively—the maximum values observed in the test. Correlation coefficient analysis demonstrated a positive correlation among photon flux density, net photosynthetic rate, transpiration rate, and TN and NH4 +-N removal efficiency. In contrast, there was a negative correlation between stomatal conductance and TN and NH4 +-N removal efficiency. Results suggest that the photosynthesis and transpiration of wetland plants have a great impact on nitrogen removal efficiency of wetlands, which can be enhanced by an increase in the photosynthesis and transpiration rate. In addition, the efficiency of water usage by reeds and nitrogen removal efficiency could be affected by the water level in wetlands; a higher level boosts nitrogen removal efficiency.  相似文献

Since the ammonia in the effluent of the traditional water purification process could not meet the supply demand, the advanced treatment of a high concentration of NH4 +-N micro-polluted source water by biological activated carbon filter (BACF) was tested. The filter was operated in the downflow manner and the results showed that the removing rate of NH4 +-N was related to the influent concentration of NH4 +-N. Its removing rate could be higher than 95% when influent concentration was under 1.0 mg/L. It could also decrease with the increasing influent concentration when the NH4 +-N concentration was in the range from 1.5 to 4.9 mg/L and the dissolved oxygen (DO) in the influent was under 10 mg/L, and the minimum removing rate could be 30%. The key factor of restricting nitrification in BACF was the influent DO. When the influent NH4 +-N concentration was high, the DO in water was almost depleted entirely by the nitrifying and hetetrophic bacteria in the depth of 0.4 m filter and the filter layer was divided into aerobic and anoxic zones. The nitrification and degradation of organic matters existed in the aerobic zone, while the denitrification occurred in the anoxic zone. Due to the limited carbon source, the denitrification could not be carried out properly, which led to the accumulation of the denitrification intermediates such as NO2 ?. In addition to the denitrification bacteria, the nitrification and the heterotrophic bacteria existed in the anoxic zone.  相似文献

Suspension-feeding bivalves increase the quantity and quality of sedimenting organic matter through the production of faeces and pseudofaeces that are remineralised in coastal sediments and thus increase sediment oxygen demand and nutrient regeneration. Bivalves are intensively cultivated worldwide; however, no bivalve biodeposit decay rates are available to parameterise models describing the environmental effects of bivalve culture. We examined sediment biogeochemical changes as bivalve biodeposits age by incubating coastal sediments to which we added fresh mussel (Perna canaliculus) biodeposits and measured O2 and nutrient fluxes as well as sediment characteristics over an 11-day period. Biodeposits elevated organic matter, chlorophyll a, phaeophytin a, organic carbon and nitrogen concentrations in the surface sediments. Sediment oxygen consumption (SOC) increased significantly (P=0.016) by ∼1.5 times to 1,010 μmol m−2 h−1 immediately after biodeposit addition and remained elevated compared to control cores without additions for the incubation period. This increase is in the range of observed in situ oxygen demand enhancements under mussel farms. To calculate a decay rate for biodeposits in sediments we fitted a first-order G model to the observed increase in SOC. The significant model fit (P=0.001, r 2=0.72) generated a decay rate of 0.16 day−1 (P=0.033, SE=0.05) that corresponds to a half-life time of 4.3 day. This decay rate is 1–2 orders of magnitude higher than published decay rates of coastal sediments without organic enrichment but similar to rates of decaying zooplankton faecal pellets. NH4+ release increased rapidly on the day of biodeposit addition (P=0.013) and reached a maximum of 144 μmol m−2 h−1 after 5 days which was 3.6 times higher compared to control cores. During this period NH4+ release was significantly (P<0.001 to P=0.043) higher in the cores with biodeposit additions than in control cores.  相似文献

Based on a series of short-term incubations involving the marine diatom Chaetoceros simplex (Bbsm), precultured in NH 4 + -, NO 3 - -and urea-limited continuous cultures at several dilution rates, we found that both the short-term specific rate of 14CO2 uptake and the amount of CO2 fixed after 8- and 16-min incubations were unaffected by enrichment with NH 4 + , urea, or NO 3 - when NH 4 + or urea were the preconditioning forms of N, but were slightly suppressed when the cells were first grown on NO 3 - . Similar enrichments in the dark, however, led to significant CO2 uptake under all conditions of NH 4 + enrichment and to similarly enhanced CO2 uptake, but only at high growth rates, when urea was the source of enrichment nitrogen. Our light results are contrary to some contemporary findings, but there does seem to be agreement that photosynthetic rates of rapidly growing phytoplankton will not be affected by exposure to pulses of nitrogen. Enhanced dark uptake, in contrast, appears to be characteristic of phytoplankton under all degrees of N limitation, and, as such, may be useful as an “all or nothing” index of the nitrogen status of natural waters. There is some indication that the index may be useful in determining both the form of and the degree of N limitation as well.  相似文献

The dissolved nitrogen pool in aquatic systems is comprised of many different nitrogen forms, both inorganic and organic. Interaction among these nitrogen forms at the level of uptake and enzyme activity is, with the exception of NH4+ and NO3, not completely understood. Nitrate reductase (NR) and urease (UA) activities in the marine diatom Thalassiosira weissflogii (Grunow) Fryxell et Hasle were measured in NO3, NH4+, and urea-sufficient cultures before and after challenge additions of NH4+, NO3, and urea in a factorial design. NR and UA were constitutively expressed during growth on NO3, NH4+, and urea. Growth on NH4+ or urea resulted in NR activities that were <10% of the activity observed in the NO3-grown culture, while growth on NO3 resulted in UA values that were ~35% of the activities during growth on either NH4+ or urea. The addition of NH4+ or urea to NO3-grown cultures resulted in an immediate decrease in cellular NO3 uptake rate, which was not mirrored by an immediate repression of in vitro NR activity; however, the diel peak in NR was suppressed in these challenge experiments. The addition of NO3 or NH4+ to urea-grown cultures resulted in non-significant decreases in the urea uptake rate. UA was not impacted by NO3 addition, but NH4+ addition significantly decreased UA throughout the experiment. These studies demonstrate that the uptake and assimilation of NO3 and urea may not be subject to the same internal feedback mechanism when challenged with other nitrogen substrates.Communicated by J.P. We examined the impact of exposing natural populations of marine bacteria (from seawater collected near Woods Hole, Massachusetts, USA) to multiple nitrogen and carbon sources in a series of batch growth experiments conducted from 1989 through 1990. The substrate C:N ratio (C:Ns) was varied from 1.5:1 to 10:1 either with equal amounts of NH 4 + and different amino acids or an amino acid mixture, all supplemented with glucose to maintain the C:Ns ratio equal to that of the respective amino acid, or with combinations of glucose and NH 4 + alone. A common feature of the experiments involving amino acids was the concurrent uptake of NH 4 + and amino acids that persisted as long as a readily assimilable carbon source (glucose in our case) was taken up. There was no net regeneration of NH 4 + , even though catabolism of amino acids occurred. Regeneration of NH 4 + was evident only after glucose was completely utilized, which usually occurred at the end of exponential growth. The contribution of15NH 4 + to total nitrogen uptake by the end of exponential growth varied from ~60 to 80% when individual amino acids were present and down to ~24% when the amino acid mixture was added. These estimates are conservative because we did not account for possible isotope dilution effects resulting from amino acid catabolism. When NH 4 + and glucose were the sole nitrogen and carbon sources, there was a stoichiometric balance between glucose and NH 4 + uptake over a wide range of C:Ns ratios, leading to a constant bacterial biomass C:N ratio (C:NB) of ~4.5:1. As a result NH 4 + usage varied from 50% when the C:Ns ratio was 3.6:1, to 100% when the C:Ns ratio was 10:1. Gross growth efficiency varied from ~60% when NH 4 + plus glucose were added alone or with the amino acid mixture, to 47% when the individual amino acids were used in place of the mixture. It is thus evident that actively growing bacteria will act as sinks for nitrogen when a carbon source that can be assimilated easily is available to balance NH 4 + uptake, even when amino acids are available and are being co-metabolized.  相似文献

Batch experiments were conducted to study the short-term biological effects of rare earth ions (La3+, Ce3+) and their mixture on the nitrogen removal in a sequencing batch reactor (SBR). The data showed that higher NH4 +-N removal rate, total inorganic nitrogen removal efficiency, and denitrification efficiency were achieved at lower concentrations of rare earth elements (REEs) (<1 mg/L). In the first hour of the aeration stage of SBR, the presence of REEs increased the total inorganic nitrogen removal efficiency and NH4 +-N removal efficiency by 15.7% and 10%–15%, respectively. When the concentrations of REEs were higher than 1 mg/L, the total inorganic nitrogen removal efficiency decreased, and nitrate was found to accumulate in the effluent. When the concentrations of REEs was up to 50.0 mg/L, the total inorganic nitrogen removal efficiency was less than 30% of the control efficiency with a high level of nitrate. Lower concentrations of REEs were found to accelerate the nitrogen conversion and removal in SBR.  相似文献

Heterotrophic nitrogen-fixation (acetylene reduction) was measured during decomposition (under dark conditions) of Rhizophora mangle L. and Avicennia germinans (L.) Stearn leaf litter. Nitrogen-fixation rates in leaf litter increased following 24 d incubation, then decreased after ≃44 d for both species. Maximum rates of 66.2 and 64.6 nmol C2H4 g−1 dry wt h−1 were reached by R. mangle and A. germinans leaf litter, respectively. Higher fixation rates of leaf litter were associated with an increase in water content and sediment particles on leaf surfaces of both species. Rates of nitrogen fixation by diazotrophs attached to sediment particles were not significantly different from zero. With additions of d-glucose, ethylene production rates increased by factors of 625-, 34- and 7-fold for sediment, R. mangle and A.␣germinans leaf litter, respectively, compared to rates prior to enrichment. These organically enhanced rates of nitrogen fixation on leaves could be accounted for by increased activity associated with attached sediment particles and not the leaf material. Total phenolics [reported as tannic acid equivalent (TAE) units] decreased nitrogen-fixation rates when added to d-glucose-enriched sediment at >20 mg TAE l−1. Phenolic compounds could explain the initial lag in rates of nitrogen fixation during leaf-litter decomposition of R. mangle (initial content of 110.8 mg TAE g−1 dry wt), but not of A. germinans (initial content of 23.4 mg TAE g−1 dry wt). The higher phenolic content and reportedly lower carbon substrate of R. mangle did not result in species-specific differences in either the magnitude or temporal pattern of nitrogen fixation compared to A. germinans leaf litter. We conclude that the availability of organic substrates leached from the leaf litter along with colonization by the heterotrophic diazotrophs (as indicated by sediment accumulation) controls nitrogen-fixation rates in a similar manner in the leaf litter of both species. A method is presented in which the 15N at.% of urea is determined with high precision on liquid samples containing as little as 10 nmol of urea. The method involves removal of interference from NH4 + initially present in the sample by cation exchange. Urea in the sample is subsequently hydrolyzed to NH4 + by the enzyme urease. Liberated NH4 + is separated from the alkaline sample by diffusion as NH3 through a helium gas phase where it is finally oxidized to N2 by reaction with hypobromite iodine. The isotopically labeled N2 thus formed is mixed with the N2 initially present in the sample, and the 15N at.% of urea is calculated from the relative amounts of 14N15N and 15N15N by means of the isotope pairing principle. The presented method for 15N-urea analysis proved to be precise (SE < 0.4 at.%, n = 5), when applied on both marine and freshwater samples with an 15N enrichment >1 at.%, and interference was found only from volatile methyl amines. In this paper, the effect of pH on biological degradation of Microcystis aeruginosa by alga-lysing bacteria in laboratory-scale sequencing batch biofilm reactors (SBBRs) was investigated. After 10 d filming with waste activated sludge, the biological film could be formed, and the bioreactors in which laid polyolefin resin filler were used to treat algal culture. By comparing the removal efficiency of chlorophyll a at different aerobic time, the optimum time was determined as 5 h. Under pH 6.5, 7.5, and 8.5 conditions, the removal rates of Microcystis aeruginosa were respectively 75.9%, 83.6%, and 78.3% (in term of chlorophyll a), and that of Chemical Oxygen Demand (CODMn) were 30.6%, 35.8%, and 33.5%. While the removal efficiencies of ammonia nitrogen (NH 4 + - N) were all 100%. It was observed that the sequence of the removal efficiencies of algae, NH 4 + - N and organic matter were pH 7.5>pH 8.5>pH 6.5. The results showed that the dominant alga-lysing bacteria in the SBBRs was strain HM-01, which was identified as Bacillus sp. by Polymerase Chain Reaction (PCR) amplification of the 16S rRNA gene, Basic Local Alignment Search Tool (BLAST) analysis, and comparison with sequences in the GenBank nucleotide database. The algicidal activated substance which HM-01 strain excreted could withstand high temperature and pressure, also had better hydrophily and stronger polarity.  相似文献
