A greenhouse experiment was conducted to evaluate the potential role of arbuscular mycorrhizal fungi (AMF) in encouraging revegetation of copper (Cu) mine tailings. Two native plant species, Coreopsis drummondii and Pteris vittata, together with a turf grass, Lolium perenne and a leguminous plant Trifolium repens associated with and without AMF Glomus mosseae were grown in Cu mine tailings to assess mycorrhizal effects on plant growth, mineral nutrition and metal uptake. Results indicated that symbiotic associations were successfully established between G. mosseae and all plants tested, and mycorrhizal colonization markedly increased plant dry matter yield except for L. perenne. The beneficial impacts of mycorrhizal colonization on plant growth could be largely explained by both improved P nutrition and decreased shoot Cu, As and Cd concentrations. The experiment provided evidence for the potential use of local plant species in combination with AMF for ecological restoration of metalliferous mine tailings.  相似文献

Seedlings of Sonchus oleraceus L. were transplanted to soil supplied with lead acetate at dosages of 0, 800, 1600 and 3200 mg kg(-1) DW. Measures of chlorophyll content, peroxidase (POD) activity, shoot length, biomass and Pb content in the plant tissues were obtained from the experimental plants. With increasing amounts of Pb in the soil, the chlorophyll content, shoot length and biomass decreased, while POD activity and Pb content in the plant tissues increased. At 3200 mg kg(-1) Pb treatment, Pb content in the plant leaf, stem and root were 65.67, 149.82 and 1113.24 mg kg(-1), respectively. Only at 3200 mg kg(-1) Pb treatment did chlorophyll content, shoot length and biomass significantly increase by 18, 15 and 44%, respectively, while POD decreased by 39% over the control. The potential of applying this species in phytoremediation of Pb contaminated roadside soils and thus restoration of the roadside vegetation are discussed.  相似文献

The effect of inoculation with indigenous naturally occurring microorganisms (an arbuscular mycorrhizal (AM) fungus and rhizosphere bacteria) isolated from a Cd polluted soil was assayed on Trifolium repens growing in soil contaminated with a range of Cd. One of the bacterial isolate showed a marked PGPR effect and was identified as a Brevibacillus sp. Mycorrhizal colonization also enhanced Trifolium growth and N, P, Zn and Ni content and the dually inoculated (AM fungus plus Brevibacillus sp.) plants achieved further growth and nutrition and less Cd concentration, particularly at the highest Cd level. Increasing Cd level in the soil decreased Zn and Pb shoot accumulation. Coinoculation of Brevibacillus sp. and AM fungus increased shoot biomass over single mycorrhizal plants by 18% (at 13.6 mg Cd kg(-1)), 26% (at 33.0 mg Cd kg(-1)) and 35% (at 85.1 mg Cd (kg(1)). In contrast, Cd transfer from soil to plants was substantially reduced and at the highest Cd level Brevibacillus sp. lowered this value by 37.5% in AM plants. Increasing Cd level highly reduced plant mycorrhization and nodulation. Strong positive effect of the bacterium on inocula, are important in plant Cd tolerance and development in Cd polluted soils.  相似文献

采用盆栽实验的方法,研究了不同Cu处理水平(0、100和200 mg/kg)下,接种丛枝菌根真菌(arbuscular my-corrhizal fungi,AMF)Glomus mosseae对旱稻(Oryzal Sativa L.)生长的影响。结果表明,未添加Cu处理下,旱稻菌根侵染率可达69%,随着土壤中Cu添加量增加,旱稻菌根侵染率显著下降(P<0.05)。与未接种处理相比,接种处理显著提高100mg/kg Cu处理下根系生物量以及200 mg/kg Cu处理下地上部分生物量(P<0.05);接种处理显著降低了100 mg/kg Cu处理下旱稻地下部Cu含量,却显著增加了200 mg/kg Cu处理下旱稻地上部分以及地下部分Cu含量(P<0.05)。  相似文献

The use of a vegetation cover for the management of heavy metal contaminated soils needs prior investigations on the plant species the best sustainable. In this work, behaviors of Trifolium repens and Lolium perenne, growing in a metal-polluted field located near a closed lead smelter, were investigated through Cd, Pb and Zn-plant metal concentrations and their phytotoxicity. In these plant species, metals were preferentially accumulated in roots than in shoots, as follow: Cd>Zn>Pb. Plant exposure to such metals induced oxidative stress in the considered organs as revealed by the variations in malondialdehyde levels and superoxide dismutase activities. These oxidative changes were closely related to metal levels, plant species and organs. Accordingly, L. perenne seemed to be more affected by metal-induced oxidative stress than T. repens. Taken together, these findings allow us to conclude that both the plant species could be suitable for the phytomanagement of metal-polluted soils.  相似文献

A sand culture experiment was established to determine interactions between arbuscular mycorrhizae and heavy metals. Mycorrhizal infection rates, spore densities, maize root and shoot weights, and heavy metal contents in maize were as indexes of responses of arbuscular mycorrhizal fungi (Acaulospora laevis, Glomus caledonium and Glomus manihotis) to heavy metals (Cu and Cd). The mycorrhizal infection rates of G. caledonium were the highest among these three mycorrhizal fungi, but the sporulating ability of G. caledonium was the poorest in the heavy metal treatments. The shoot and root weights of non-mycorrhizal plants were usually greater than those of mycorrhizal plants when the Cu concentrations in solutions are less than 3 mg l(-1) or Cd concentrations less than 1 mg l(-1). When Cd concentrations were 0.5 and 1 mg(-1), the root and shoot weights of plants inoculated with A. laevis were significantly (p < 0.05) lower than those of other treatments. Copper concentrations in shoots of mycorrhizal plants were higher than those of non-mycorrhizal ones at all Cu concentrations in solution, especially at low Cu concentrations. As to A. laevis, Cu concentrations in roots and shoots of the host were higher than those of non-mycorrhizal plants in these treatments. Thus A. laevis was sensitive to Cu and Cd, especially Cd, and G. caledonium was more tolerant to these two heavy metals. It is suggested that G. caledonium might be a promising mycorrhizal fungus for bioremediation of heavy metal contaminated soil.  相似文献

In a compartmented cultivation system, white clover (Trifolium repens Linn.) and ryegrass (Lolium perenne L.), with their roots freely intermingled, or separated by 37 microm nylon mesh or plastic board, were grown together in an arsenic (As) contaminated soil. The influence of AM inoculation on plant growth, As uptake, phosphorus (P) nutrition, and plant competitions were investigated. Results showed that both plant species highly depended on mycorrhizas for surviving the As contamination. Mycorrhizal inoculation substantially improved plant P nutrition, and in contrast markedly decreased root to shoot As translocation and shoot As concentrations. It also showed that mycorrhizas affected the competition between the two co-existing plant species, preferentially benefiting the clover plants in term of nutrient acquisition and biomass production. Based on the present study, the role of AM fungi in plant adaptation to As contamination, and their potential use for ecological restoration of As contaminated soils are discussed.  相似文献

Subterranean clover inoculated or not with the arbuscular mycorrhizal (AM) fungus Glomus intraradices was grown on soil containing six levels of 238U in the range 0-87 mg kg(-1). Increasing U concentration in soil enhanced the U concentration in roots and shoots of both mycorrhizal and nonmycorrhizal plants but had no significant effects on plant dry matter production or root AM colonization. Mycorrhizas increased the shoot dry matter and P concentration in roots and shoots, while in most cases, it decreased the Ca, Mg and K concentrations in plants. The AM fungus influenced U concentration in plants only in the treatment receiving 87 mg U kg(-1) soil. In this case, U concentration in shoots of nonmycorrhizal plants was 1.7 times that of shoots of mycorrhizal plants. These results suggested that mycorrhizal fungi can limit U accumulation by plants exposed to high levels of U in soil.  相似文献

The toxicities of 2,4,6-trinitrotoluene (TNT), 1,3,5-trinitrobenzene (TNB), 2,4-dinitrotoluene (2,4-DNT), and 2,6-dinitrotoluene (2,6-DNT) to terrestrial plants alfalfa (Medicago sativa L.), Japanese millet (Echinochloa crusgalli L.), and perennial ryegrass (Lolium perenne L.) were determined in Sassafras sandy loam soil using seedling emergence, fresh shoot, and dry mass measurement endpoints. A 13-week weathering and aging of energetic materials in soils, which included wetting and drying cycles, and exposure to sunlight of individual soil treatments, was incorporated into the study design to better reflect the soil exposure conditions in the field than toxicity determinations in freshly amended soils. Definitive toxicity tests showed that dinitrotoluenes were more phytotoxic for all plant species in freshly amended treatments based on EC20 values for dry shoot ranging from 3 to 24mgkg(-1) compared with values for TNB or TNT ranging from 43 to 62mgkg(-1). Weathering and aging of energetic materials (EMs) in soil significantly decreased the toxicity of TNT, TNB or 2,6-DNT to Japanese millet or ryegrass based on seedling emergence, but significantly increased the toxicity of all four EMs to all three plant species based on shoot growth. Exposure of the three plant species to relatively low concentrations of the four compounds initially stimulated plant growth before the onset of inhibition at greater concentrations (hormesis).  相似文献

A randomised block glasshouse pot experiment compared the growth and Zn uptake of mycorrhizal and nonmycorrhizal white clover plants grown in a sterile soil/sand mixture containing 25 mg Zn kg(-1) to which five application rates of Zn (as ZnSO4) from 0 to 400 mg kg(-1) were made. Two mycorrhizal inocula infected roots from the field and from clover trap cultures, were compared. Mycorrhizal infection (ranging from 33% to 46% of total root length) and Zn application had little effect on plant growth. Increasing Zn application rate led to increased uptake of Zn in roots and shoots (especially roots), but the increases were significantly greater in non-mycorrhizal controls than in mycorrhizal treatments. In contrast, P uptake was higher in mycorrhizal than in non-mycorrhizal plants. Plants that received trap culture inoculum had significantly lower Zn uptake than those that received field inoculum. The results indicate that mycorrhizal infection may have exerted some protective effect against plant Zn accumulation at the range of soil Zn concentrations studied and may have immobilised Zn in or near the roots to some extent. However, this mycorrhizal effect cannot be explained simply by tissue dilution, hyphal sequestration or root immobilisation of Zn.  相似文献

Field trials concerning the establishment of plant cover on a deposit of wastes from the Ammeberg zinc mine in central Sweden were carried out during 1976-1985. Different soil conditioners and manures were applied and plant species cultivars were evaluated with regard to plant biomass, vigour, durability and content of zinc, lead and cadmium. Sewage sludge and topsoil led to better establishment of grasses than did municipal waste, straw and hydraulic seeding. After 2 years, Festuca rubra and Poa pratensis dominated the swards. Other species (Dactylis glomerata, Bromus inermis, Lolium perenne, Phleum nodosum, Festuca pratensis and F. arundinacea) constituted only a minor part of the stand. After 10 years, F. rubra was the most dominant species, while native Agrostis tenuis had invaded 20-50% of the area within the plots. Merlin was the clearly dominant red fescue cultivar. The concentration of zinc in shoots (616 mg kg(-1) dw) was about 10% of that in the soil. Zinc concentration decreased with increasing biomass above ground. It increased with age in Scots pine needles and was very high in birch leaves. Grasses survived longer than legumes in the zinc sand waste. Among the surviving grasses was a group with high (3800 mg kg(-1) dw) and a group with low (320 mg kg(-1) dw) zinc concentrations. The low group included Merlin red fescue and Sobel creeping bent. The cultivar Merlin contained a much lower zinc concentration than the other cultivars of red fescue (375 and 624 mg kg(-1) dw, respectively). A large amount of root biomass was present in plots with dominating Merlin red fescue (1715 g m(-2)), 97% of which was concentrated in the top 10 cm of the soil. The concentration of zinc in the roots was very high (13 000-25 000 mg kg(-1) dw). Nitrate fertilizer, especially ammonium nitrate, and acidic water (pH 4.3) increased zinc leaching.  相似文献

In a pot experiment, the potential use of 10 plant species, including six dicotyledon species and four monocotyledon species, was investigated for the EDTA-enhanced phytoextraction of Pb from contaminated soil. Mung bean and buckwheat had a higher sensitivity to the EDTA treatment in soils. In the 2.5 and 5.0 mmol kg(-1) EDTA treatments, the Pb concentrations in the shoots of the six dicotyledon species ranged from 1,000 to 3,000 mg kg(-1) of dry matter, which were higher than those of the monocotyledon species. The highest amount of phytoextracted Pb (2.9 mg Pb pot(-1)) was achieved in sunflowers, due to the high concentration of Pb in their shoots and large biomass, followed by corns (1.8 mg Pb pot(-1)) and peas (1.1 mg Pb pot(-1)). The leaching behavior of heavy metals as a result of applying EDTA to the surface of the soil was also investigated using short soil-leaching columns (9.0-cm diameter, 20-cm height) by the percolation of artificial rainfall. About 3.5%, 15.8%, 13.7% and 20.6% of soil Pb, Cu, Zn and Cd, respectively, were leached from the soil columns after the application of 5.0 mmol kg(-1) of EDTA. The growth of sunflowers in the soil columns had little effect on the amount of metals that were leached out. This was probably due to the shallowness of the layer of soil, the short time-span of the uptake of metals by the plant and the plant's simple root systems.  相似文献

Significant hyperaccumulation of Zn, Cd and Pb in field samples of Thlaspi praecox Wulf. collected from a heavy metal polluted area in Slovenia was found, with maximal shoot concentrations of 14,590 mg kg(-1) Zn, 5960 mg kg(-1) Cd and 3500 mg kg(-1) Pb. Shoot/root ratios of 9.6 for Zn and 5.6 for Cd show that the metals were preferentially transported to the shoots. Shoot bioaccumulation factors exceeded total soil Cd levels 75-fold and total soil Zn levels 20-fold, further supporting the hyperaccumulation of Cd and Zn. Eighty percent of Pb was retained in roots, thus indicating exclusion as a tolerance strategy for Pb. Low level colonisation with arbuscular mycorrhizal fungi (AMF) of a Paris type was observed at the polluted site, whereas at the non-polluted site Arum type colonisation was more common. To our knowledge this is the first report of Cd hyperaccumulation and AMF colonisation in metal hyperaccumulating T. praecox.  相似文献

Six plant species in the family Gramineae were used to investigate the relationship between Cs uptake, nutrient regime and plant growth strategy sensu Grime (1979: Plant Growth Strategies and Vegetation Processes, John Wiley). The roots of 66 day old Elymus repens (L.) Gould., Bromus sterilis L., Agrostis stolonifera L., Anthoxanthum odoratum L., Festuca ovina L. and Nardus stricta L. plants grown in acid-washed sand at high and low nutrient levels were exposed to a 96 h pulse of stable Cs at 0.05 mM, 0.15 mM, 0.3 mM, 1.0 mM and 3.0 mM concentrations. Different nutrient regimes induced large differences in dry wt in E. repens, B. sterilis and A. stolonifera plants but only small differences in N. stricta and F. ovina plants. At high nutrient concentrations, A. stolonifera, A. odoratum, F. ovina and N. stricta shoots showed significantly greater increases in internal Cs concentration with rising external Cs concentrations than did E. repens and B. sterilis shoots. The relationship between increases in shoot and external Cs concentrations was statistically indistinguishable between species in plants grown at the low nutrient concentration. These patterns of Cs uptake ensured that with long-term high K concentrations the more competitive plants (E. repens and B. sterilis) accumulated higher concentrations of Cs from low external concentrations than did non-competitive plants or competitive plants grown at low nutrient levels. It is suggested that the relationship between plant growth strategy sensu Grime (1979) and Cs accumulation patterns may help to explain the different concentrations to which species accumulate radiocaesium from the soil.  相似文献

Red clover plants inoculated with Glomus mosseae were grown in a sterile pasture soil containing 50 mg Zn kg(-1) in 'Plexiglas' (acrylic) containers with nylon net partitions (30 microm mesh) designed to separate the soil into a central root zone and two outer zones for hyphal growth with no root penetration. Two porous plastic soil moisture samplers were installed in each pot, one in the root compartment and the other in one of the hyphal compartments. The soil in the outer compartments was amended with one of the four application rates of Zn (as ZnSO4) ranging from 0 to 1000 mg kg(-1). Non-mycorrhizal controls were included, and there were five replicates of each treatment in a randomised block in a glasshouse. Uninoculated plants received supplementary P to avoid yield limitation due to low soil P status. Plants grew in the central compartment for nine weeks. Soil moisture samples were collected 4, 24 and 62 days after sowing to monitor changes in the Zn concentration and pH of the soil solution. At harvest, the mean mycorrhizal infection rate of inoculated plants ranged from 29% to 34% of total root length and was little affected by Zn application. Root and shoot yields were not affected by mycorrhizal infection. Plant Zn concentration and uptake were lower in mycorrhizal plants than non-mycorrhizal controls, and this effect was more pronounced with increasing Zn application rate to the soil. Soil solution Zn concentrations were lower and pH values were higher in mycorrhizal treatments than non-mycorrhizal controls and the mycorrhiza effect was more pronounced at higher Zn application rates. The protective effect of mycorrhiza against plant Zn uptake may have been associated with changes in Zn solubility mediated by changes in the soil solution pH, or by immobilisation of Zn in the extraradical mycelium.  相似文献

A modified glass bead compartment cultivation system is described in which glass beads continue to be used in the hyphal compartment but are replaced by coarse river sand in the compartments for host plant roots and mycorrhizal hyphae. Arbuscular mycorrhizal (AM) associations were established using two host plant species, maize (Zea mays L.) and red clover (Trifolium pratense L.) and two AM fungi, Glomus mosseae and G. versiforme. When the standard and modified cultivation systems were compared, the new method yielded much more fungal tissue in the hyphal compartment. Using G. versiforme as the fungal symbiont, up to 30 mg of fungal dry matter (DM) was recovered from the hyphal compartment of mycorrhizal maize and about 6 mg from red clover. Multi-element analysis was conducted on samples of host plant roots and shoots and on harvested fungal biomass. Concentrations of P, Cu and Zn were much higher in the fungal biomass than in the roots or shoots of the host plants but fungal concentrations of K, Ca, Mg, Fe and Mn were similar to or lower than those in the plants. There were also significant differences in nutrient concentrations between the two AM fungi and these may be related to differences in their proportions of extraradical mycelium to spores. The high affinity of the fungal mycelium for Zn was very striking and is discussed in relation to the potential use of arbuscular mycorrhiza in the phytoremediation of Zn-polluted soils.  相似文献

In a pot experiment, red clover (Trifolium pratense) was grown in sterilized Zn-amended low available P soil (0, 50 or 400 mg Zn kg(-1)) with or without 100 mg kg(-1) added P and with or without inoculation with the arbuscular mycorrhizal (AM) fungus G. mosseae. When the plants were harvested after 40 days, AM colonization of the roots was still at an early stage, with only 14-38% of total root length colonized on average. AM colonization was highest in low-P soil, and was lowest in soil amended with 400 mg Zn kg(-1). Shoot yields were highest in AM plants with added P, but root yields were unaffected by AM inoculation. Shoot and root yields were higher with 100 mg added P kg(-1) soil, but lower with 400 mg Zn kg(-1) than 50 mg Zn kg(-1) or controls unamended with Zn. Shoot and root P concentrations were seldom higher in AM plants, but shoot P offtakes were higher in AM plants with added P. Concentrations of Zn and Cu were much higher in the roots than in the shoots. Shoot and root Zn and shoot Cu were lower, but root Cu was higher, in AM plants. Soil residual pH after plant growth was higher in AM treatments, and residual total Zn was also higher, indicating lower Zn uptake by AM plants. Soil solution pH was higher in AM treatments, and soil solution Zn was lower in the presence of mycorrhiza. The results are discussed in terms of AM protection of the plants against excessive shoot Zn uptake.  相似文献

Three pot experiments were conducted to investigate the role of arbuscular mycorrhiza (AM) in Zn uptake by red clover. Plants inoculated with Glomus mosseae and uninoculated controls were grown in a sterile calcareous soil in 'Plexiglas' (Acrylic) containers with two nylon net partitions (30 microm mesh) to separate the central root zone from the two outer hyphal zones. The effects of mycorrhiza on plant growth and Zn uptake changed dramatically with increasing Zn addition level (range 0-1200 mg kg(-1)) in the root zone. With Zn addition levels <300 mg kg(-1), added Zn did not affect plant yield and above the critical level plant yield gradually decreased but was always higher for mycorrhizal than for controls. Below the critical Zn application rate (50 mg kg(-1)), Zn uptake was enhanced while above this level Zn translocation to the shoots decreased. At all Zn addition levels, mycorrhizal colonization increased Zn absorption and accumulation in the roots, and this may help to explain the alleviation of Zn toxicity at high Zn application rates. As expected, AM colonization enhanced P nutrition and hence yield at all added Zn levels studied. Efforts were made to obtain direct evidence for hyphal contribution to Zn uptake by applying both Zn to the hyphal growth zone and additional P to the root zone to avoid the 'growth dilution effect'. The data demonstrate that mycorrhizal hyphae could absorb Zn directly from the soil and then transfer it to the plant roots. The hyphal contribution to Zn uptake by the host plant reached its maximum value at the Zn addition level of 50 mg kg(-1), in which Zn uptake via the extramatrical hyphae comprised 22% of total uptake, thus confirming the critical Zn application level found previously.  相似文献

The acute toxicities of lead (Pb) and copper (Cu) to important crop plants Sorghum bicolor, Cucumis sativus, Triticum aestivum, and Zea mays were compared. The EC50 values (the concentration of metals in the soil that reduces the growth of shoots and roots by 50%) were derived using the Trimmed Spearman-Karber method. The EC50s-shoot (root) in mg Pb kg-1 dry soil and mg Cu kg-1 dry soil were in the range of 519 to >1280 (285-445), and 48-232 (<40-110), respectively. Those concentrations are likely to occur in some abandoned mine areas in Korea. The figures indicate that Cu is more toxic than Pb to the plants in this study, and that root growth is more sensitive to the toxicity endpoint than shoot growth in Cu- or Pb-amended soils. On the other hand, seed germination is insensitive to both Pb and Cu toxicities. The Pb- and Cu-sensitive plants were also identified. Among the plants tested, T. aestivum and S. bicolor were most sensitive to Pb and Cu, respectively. Z. mays was most resistant to both Pb and Cu. The combined effects of Pb and Cu depend on the plant species, and no general phenomenon was observed. Bioaccumulations of Pb and Cu were observed in all test species, and they are concentration-dependent. These differences in the toxicities of Pb and Cu in plant species should be taken into account in biomonitoring and ecological risk assessment.  相似文献

The influence of light on phytotoxicity of increased concentration (2, 5, 10 mg/l) of intact fluoranthene (FLT) and photomodified fluoranthene (phFLT) diluted in experimental solutions was investigated. The germination rate of lettuce (Lactuca sativa L.), onion (Allium cepa L.) and tomato (Lycopersicum esculentum L.) seeds and some parameters of seedlings primary growth of these plant species were used as laboratory indicators of phytotoxicity. Among them a length of root and shoot, their dry weight and a content of photosynthetic pigments in shoot were measured. The results demonstrated that the higher concentration (5 and 10 mg/l) of FLT and especially of phFLT significantly inhibited the germination rate of seeds and the length of root and shoot seedlings of all plant species. Decreased production of biomass expressed by dry weight of root and shoot was found in lettuce seedlings under the inhibitory effect of FLT and phFLT. An increased concentration of FLT and phFLT did not exhibit an unambiguous effect on the content of photosynthetic pigments in shoot of experimental plants. Only the highest concentration (10 mg/l) of FLT significantly increased content of chlorophylls a and b in lettuce, onion and tomato plants and content of carotenoids in lettuce and onion. Light intensified a significant inhibitory effect of phFLT in the most testified parameters of germination and seedling growth.  相似文献
