1,2-Dichloroethane (1,2-DCA) is one of the most hazardous pollutant of soil and groundwater, and is produced in excess of 5.44 × 109 kg annually. Owing to their toxicity, persistence and potential for bioaccumulation, there is a growing interest in technologies for their removal. Heavy metals are known to be toxic to soil microorganisms at high concentrations and can hinder the biodegradation of organic contaminants. In this study, the inhibitory effect of heavy metals, namely; arsenic, cadmium, mercury and lead, on the aerobic biodegradation of 1,2-DCA by autochthonous microorganisms was evaluated in soil microcosm setting. The presence of heavy metals was observed to have a negative impact on the biodegradation of 1,2-DCA in both soil samples tested, with the toxic effect being more pronounced in loam soil, than in clay soil. Generally, 75 ppm As3+, 840 ppm Hg2+, and 420 ppm Pb2+ resulted in 34.24%, 40.64%, and 45.94% increase in the half live (t½) of 1,2-DCA, respectively, in loam soil, while concentrations above 127.5 ppm Cd2+, 840 ppm Hg2+ and 420 ppm of Pb2+ and less than 75 ppm As3+ was required to cause a >10% increase in the t½ of 1,2-DCA in clay soil. A dose-dependent relationship between degradation rate constant (k1) of 1,2-DCA and metal ion concentrations was observed for all the heavy metals tested, except for Hg2+. This study demonstrated that different heavy metals have different impacts on the degree of 1,2-DCA degradation. Results also suggest that the degree of inhibition is metal specific and is also dependent on several factors including; soil type, pH, moisture content and available nutrients.  相似文献

During steelmaking in electric arc furnaces (EAFs), 10 to 15 kg of dust, consisting of a mixture of zinc, lead and iron oxides and to a lesser degree chrome, nickel and manganese, is generated per ton of steel product. In many countries, the disposal of EAF dust in landfill sites is regarded as an environmental hazard, since toxic metals may be leached into drinking water supplies. This work gives the results of a study of the physical, chemical, mineralogical and toxicological properties of dust produced in Spanish EAFs. EC50 concentrations (Microtox Assay) are determined by studying the incubation time of the bacteria and an exponential ratio is then established between EC50 values and the heavy metal content of the leachates which are obtained (US EPA protocol). The relationship between the pH at which the metals are extracted and their ecotoxicity is also studied. Average EC50 values of 1 250 mg 1-1 are found when the heavy metals are extracted at a pH of 4-4.5.  相似文献

The dissolution of heavy metals from the waste rock is controlled by many factors. Herein, we investigated the release behavior of iron (Fe), chromium (Cr), copper (Cu), and zinc (Zn) from sulfide waste rock under the actions of microorganisms and different environmental factors (solution pH value, particle size of waste rock, temperature, Fe3+ concentration). The release quantity of heavy metals was negatively correlated with pH and particle size and positively correlated with ambient temperature and Fe3+ concentration. Under the experimental conditions of pH value of 3.0, temperature of 35°C, and waste stone particle size of less than 0.075 mm,, the release quantity of Fe, Cr, Cu, and Zn reached 3680, 18.32, 132.20, 26.60 mg·kg?1 after 20 days of leaching, respectively. Rising the temperature to 45 °C, Fe, Cr, Cu, and Zn release quantities increased to 89.30, 5.81, 105.08, and 28.00 mg·kg?1. Six hundred milligrams per liter Fe3+ increased the release of heavy metals considerably (2.63–65.48 folds). The presence of microorganisms can significantly facilitate the release of heavy metals. Compared to the control group, the release quantities of Fe, Cr, Cu, and Zn increased 4.29, 3.17, 1.54, and 2.39 times, respectively. In addition, the waste rock under microbial action was more seriously corroded than that under chemical factors. The release behavior of these four heavy metals was consistent with the interfacial chemical reaction control model, indicating that the reactions mainly occurred on the surface of the waste rock. This study provides an essential reference for the study of heavy metal leaching behavior.

Phosphorus (P) in agricultural ecosystems is an essential and limited element for plants and microorganisms. However, environmental problems caused by P accumulation as well as by P loss have become more and more serious. Oxygen isotopes of phosphate can trace the sources, migration, and transformation of P in agricultural soils. In order to use the isotopes of phosphate oxygen, appropriate extraction and purification methods for inorganic phosphate from soils are necessary. Here, we combined two different methods to analyze the oxygen isotopic composition of inorganic phosphate (δ18OP) from chemical fertilizers and different fractions (Milli-Q water, 0.5 mol L?1 NaHCO3 (pH = 8.5), 0.1 mol L?1 NaOH and 1 mol L?1 HCl) of agricultural soils from the Beijing area. The δ18OP results of the water extracts and NaHCO3 extracts in most samples were close to the calculated equilibrium value. These phenomena can be explained by rapid P cycling in soils and the influence of chemical fertilizers. The δ18OP value of the water extracts and NaHCO3 extracts in some soil samples below the equilibrium value may be caused by the hydrolysis of organic P fractions mediated by extracellular enzymes. The δ18OP values of the NaOH extracts were above the calculated equilibrium value reflecting the balance state between microbial uptake of phosphate and the release of intracellular phosphate back to the soil. The HCl extracts with the lowest δ18OP values and highest phosphate concentrations indicated that the HCl fraction was affected by microbial activity. Hence, these δ18Op values likely reflected the oxygen isotopic values of the parent materials. The results suggested that phosphate oxygen isotope analyses could be an effective tool in order to trace phosphate sources, transformation processes, and its utilization by microorganisms in agricultural soils.  相似文献

The effects of heavy metals, at sub-lethal concentrations, on activated sludge microbial ecosystem were investigated. Adsorption capacity and rate of copper, chromium, lead and zinc on microbial flocs were much faster than that of organic matters. Metals affected not only the adsorption rate of organic matters but also the COD adsorption capacity of the activated sludge. Effects of heavy metals. on wastewater treatment performance of a sequencing batch reactor were also studied. Metal-laden wastewater at sub-lethal levels affected the performance to different extents, depending on the hydraulic retention time. Metal ions acted as a strong competitor against the organic compounds for active sites on the bioflocs instead of acting as a toxic microbial inhibitor, thus hampering organic adsorption and affecting the COD removal efficiency under shorter HRTs.  相似文献

Soil respiration measurements are an established method to test the abundance, activity and vitality of the soil microorganisms. However, abnormal progressions of soil respiration curves impede a clear interpretation of the data. The aim of this study was to investigate the changes in the microbial structure during the formation of phenomena like double peaks and terraces by analysis of the PLFA composition (phospholipid fatty acid composition). Moreover, 13C labeled glucose was used as substrate; therefore it was possible to measure δ13C values both within the PLFA fraction as well as within the carbon dioxide evolved during respiration. As contaminants trinitrotoluene, cycloheximide, and hexadecane were used. The results showed that the appearance of double peaks was mainly related to the growth of fungi with the marker 18:2δ9,12 due to a toxic effect of trinitrotoluene and cycloheximide. In contrast, the phenomenon of terrace formation was related to the utilization of hexadecane as a carbon source mainly by bacteria.  相似文献

Low pH markedly reduced the growth and photosynthetic activity of an Ankistrodesmus sp. The alga could not grow at pH 3 and only slight growth occurred at pH 4. The alga grew well above pH 4 with maximum growth occurring at pH 6. The fixation of 14CO2 followed a similar pattern with pH. The algal cells were also sensitive to mercury and bisulfite in acidic conditions. Algal growth and photosynthesis were reduced by mercury and bisulfite more at pH 5 than at pH 7. Lead was relatively non-toxic to the algal cells at both pH levels. Bisulfite inhibited the membrane transport of α-amino-14C-isobutyric acid at pH 5 but not at pH 7. The results suggest that algal growth and activity may be reduced in acidic lakes by low pH and that the toxicity of mercury and bisulfite is enhanced in acidic conditions.  相似文献

Reducing airborne microorganisms may potentially improve the environment in layer breeding houses. The effectiveness of slightly acidic electrolyzed water (SAEW; pH 5.29–6.30) in reducing airborne microorganisms was investigated in a commercial layer house in northern China. The building had a tunnel-ventilation system, with an evaporative cooling. The experimental area was divided into five zones along the length of the house, with zone 1 nearest to an evaporative cooling pad and zone 5 nearest to the fans. The air temperature, relative humidity, dust concentration, and microbial population were measured at the sampling points in the five zones during the study period. The SAEW was sprayed by workers in the whole house. A six-stage air microbial sampler was used to measure airborne microbial population. Results showed that the population of airborne bacteria and fungi were sharply reduced by 0.71 × 105 and 2.82 × 103 colony-forming units (CFU) m?3 after 30 min exposure to SAEW, respectively. Compared with the benzalkonium chloride (BC) solution and povidone-iodine (PVP-I) solution treatments, the population reductions of airborne fungi treated by SAEW were significantly (P < 0.05) more, even though the three disinfectants can decrease both the airborne bacteria and fungi significantly (P < 0.05) 30 min after spraying.
Implications: There are no effective methods for reducing airborne microbial levels in tunnel-ventilated layer breeding houses; additionally, there is limited information available on airborne microorganism distribution. This research investigated the spatial distribution of microbial population, and the effectiveness of spraying slightly acidic electrolyzed water in reducing microbial levels. The research revealed that slightly acidic electrolyzed water spray was a potential method for reducing microbial presence in layer houses. The knowledge gained in this research about the microbial population variations in the building may assist producers in managing the bird housing environment and engineers in designing poultry houses.  相似文献

Abstract

After application, herbicides often reach the soil and affect non-target soil microorganisms, decreasing their population, diversity or affecting metabolic activity. Therefore, laboratory studies were performed to evaluate the effects of diuron, hexazinone and sulfometuron-methyl alone and mixed upon carbon transformation by soil microorganisms in clayey and sandy soils and the effect on bacterial diversity and structure. Control treatment without herbicide application was also performed. Sub-samples from the control and herbicide treatments (10?g – in triplicate) were collected before herbicide application and 7, 14, 28 and 42?days after treatment (DAT), then 1?mL of 14C-glucose solution was applied. The released 14CO2 was trapped in 2?M NaOH solution and the radioactivity was analyzed by liquid scintillation counting (LSC), 12?h after glucose application. The effect of herbicides on bacterial diversity was evaluated by T-RFLP. The experiment was conducted in a complete randomized design. Hexazinone did not affect 14CO2 evolution. Diuron showed a greater 14CO2 evolution in sandy and clayey soil, while sulfometuron-methyl led to an increase in sandy soil, at 42 DAT. A greater evolution of carbon was observed in the treatment with herbicide mixture in sandy soil, compared with the same treatment in clayey soil or control. However, the herbicide mixture application did not affect the soil biological activity measured by the respiration rate induced by substrate. On the other hand, the herbicide mixtures affected the bacterial diversity in both soils, being the strongest effect to diuron and sulfometuron-methyl in clayey soil and hexazinone in sandy soil.  相似文献

Bensulfuron toxicity on soil microbes was evaluated by the methods used in a previous study on cinosulfuron; the effects of the two sulfonylureas were compared. Cinosulfuron and bensulfuron, at the normal field application rate and 100 times higher, had no effect on the total number of bacteria and nitrifiers, or on the respiration activity in the soil, but they did decrease the nitrification activity. In vitro toxicity tests carried out on representative soil microbial strains using bensulfuron at 50 mg l?1 showed some inhibition of three of the 17 bacterial strains and strong inhibition of almost all the 12 fungal strains; cinosulfuron had had no effect on any of these strains in the previous study. It is concluded that, compared with cinosulfuron, bensulfuron is potentially more toxic on soil heterotrophic microorganisms, but only at very high concentrations that are nearly impossible to reach with the usual agricultural use of the herbicides. However, autotrophic nitrifiers were more sensitive to both sulfonylureas than the other microorganisms.  相似文献

Goal, Scope and Background This paper is a part of the research work on ‘Integrated treatment of industrial wastes towards prevention of regional water resources contamination — INTREAT’ the project. It addresses the environmental pollution problems associated with solid and liquid waste/effluents produced by sulfide ore mining and metallurgical activities in the Copper Mining and Smelting Complex Bor (RTB-BOR), Serbia. However, since the minimum solubility for the different metals usually found in the polluted water occurs at different pH values and the hydroxide precipitates are amphoteric in nature, selective removal of mixed metals could be achieved as the multiple stage precipitation. For this reason, acid mine water had to be treated in multiple stages in a continuous precipitation system-cascade line reactor. Materials and Methods All experiments were performed using synthetic metal-bearing effluent with chemical a composition similar to the effluent from open pit, Copper Mining and Smelting Complex Bor (RTB-BOR). That effluent is characterized by low pH (1.78) due to the content of sulfuric acid and heavy metals, such as Cu, Fe, Ni, Mn, Zn with concentrations of 76.680, 26.130, 0.113, 11.490, 1.020 mg/dm3, respectively. The cascade line reactor is equipped with the following components: for feeding of effluents, for injection of the precipitation agent, for pH measurements and control, and for removal of the process gases. The precipitation agent was 1M NaOH. In each of the three reactors, a changing of pH and temperature was observed. In order to verify efficiency of heavy metals removal, chemical analyses of samples taken at different pH was done using AES-ICP. Results Consumption of NaOH in reactors was 370 cm3, 40 cm3 and 80 cm3, respectively. Total time of the experiment was 4 h including feeding of the first reactor. The time necessary to achieve the defined pH value was 25 min for the first reactor and 13 min for both second and third reactors. Taking into account the complete process in the cascade line reactor, the difference between maximum and minimum temperature was as low as 6°C. The quantity of solid residue in reactors respectively was 0.62 g, 2.05 g and 3.91 g. In the case of copper, minimum achieved concentration was 0.62 mg/dm3 at pH = 10.4. At pH = 4.50 content of iron has rapidly decreased to < 0.1 mg/dm3 and maintained constant at all higher pH values. That means that precipitation has already ended at pH=4.5 and maximum efficiency of iron removal was 99.53%. The concentration of manganese was minimum at pH value of 11.0. Minimum obtained concentration of Zn was 2.18 mg/dm3 at a pH value of 11. If pH value is higher than 11, Zn can be re-dissolved. The maximum efficiency of Ni removal reached 76.30% at a pH value of 10.4. Discussion Obtained results show that efficiency of copper, iron and manganese removal is very satisfactory (higher than 90%). The obtained efficiency of Zn and Ni removal is lower (72.30% and 76.31%, respectively). The treated effluent met discharge water standard according to The Council Directive 76/464/EEC on pollution caused by certain dangerous substances into the aquatic environment of the Community. Maximum changing of temperature during the whole process was 6°C. Conclusion This technology, which was based on inducing chemical precipitation of heavy metals is viable for selective removal of heavy metals from metal-bearing effluents in three reactor systems in a cascade line. Recommendations and Perspectives The worldwide increasing concern for the environment and guidelines regarding effluent discharge make their treatment necessary for safe discharge in water receivers. In the case where the effluents contain valuable metals, there is also an additional economic interest to recover these metals and to recycle them as secondary raw materials in different production routes. Surface sediment from large and eutrophic Lake Chaohu was investigated to determine the occurrence, spatial distribution, sources, and risks of polychlorinated biphenyls (PCBs) and heavy metals in one of the five biggest freshwater lakes in China. Total concentration of PCBs (Σ34PCBs) in Lake Chaohu was 672 pg g?1 dry weight (dw), with a range of 7 to 3999 pg g?1 dw, which was lower than other water bodies worldwide. The majority of heavy metals were detected at all sampling locations, except for Sr, B, and In. Concentrations of Al, Fe, Ca, Mn, Sr, Co, Zn, Cd, Pb, and Hg were similar to that reported for other lakes globally. Concentrations of K, Mg, Na, Li, Ga, and Ag were greater than the average, whereas those of Cr, Ni, and Cu were lower. Cluster analysis (CA) and positive matrix factorization (PMF) yielded accordant results for the source apportionment of PCBs. The technical PCBs and microbial degradation accounted for 34.2 % and 65.8 % of total PCBs using PMF, and PMF revealed that natural and anthropogenic sources of heavy metals accounted for 38.1 % and 61.8 %, respectively. CA indicated that some toxic heavy metals (e.g., Cd, In, Tl, and Hg) were associated with Ca–Na–Mg minerals rather than Fe–Mn minerals. The uncorrelated results between organic matter revealed by pyrolysis technology and heavy metals might be caused by the existence of competitive adsorption between organic matter and minerals. PCBs and heavy metals were coupling discharge without organochlorine pesticides (OCPs), but with polycyclic aromatic hydrocarbons (PAHs) and polybrominated diphenyl ethers (PBDEs). No sediment sample exceeded the toxic threshold for dioxin-like PCBs (dl-PCBs) set at 20 pg toxicity equivalency quantity (TEQ)?g?1, (max dl-PCBs, 10.9 pg TEQ g?1). However, concentrations of Ag, Cd, and Hg were at levels of environmental concern. The sediment in the drinking water source area (DWSA) was threatened by heavy metals from other areas, and some fundamental solutions were proposed to protect the DWSA.  相似文献

The aim of this study was to characterize the labile part of dissolved organic carbon (DOC) present in groundwater by identification of natural organic carbon substrates and to assess their microbial utilization during aeration of the groundwater. The studied chlorophenol (CP) contaminated groundwater contained 60-2650 micromoll(-1) of DOC of which up to 98.0% were CPs; 1.7% were low-molecular weight organic acids and 0.2% were dissolved free amino acids. Traces of following natural organic carbon substrates were identified: L-alanine, L-isoleucine, L-leucine, L-serine, L-threonine, L-tyrosine, L-valine, L-aspartic, acetic, citric, formic, lactic, malic and oxalic acid. Dissolved oxygen concentration inside the CP-plume was lower (mean 25 micromoll(-1)) than outside of the plume (mean 102 micromoll(-1)). Over a monitoring period of four years the concentrations of CPs, Fe(II) and NH4+ were higher inside than outside of the CP-plume. Oxygen availability within the CP-plume limits in situ biological oxidation of CPs, DOC, NH4+ and Fe(II). The microbial enzymatic hydrolysis rates of 4-methylumbelliferyl and 7-amino-4-methylcoumarin-linked substrates varied from 0.01 to 52 micromoll(-1)h(-1) and was slightly higher inside than outside the plume. Microbial uptake rates of 14C-acetate, 14C-glucose and 14C-leucine were on average 28, 4 and 4 pmoll(-1)h(-1) outside and 17, 25 and 8 pmoll(-1)h(-1) inside the plume, respectively. The indigenous microorganisms were shown able of hydrolysis of dissolved organic matter, uptake and utilization of natural organic carbon substrates. Therefore, the labile part of DOC serves as a pool of secondary substrates beside the CP-contaminants in the groundwater and possibly help in sustaining the growth of CP-degrading bacteria.  相似文献

Heavy metal distribution in medicinal plants is gaining importance not only as an alternative medicine, but also for possible concern due to effects of metal toxicity. The present study has been focused on emphasizing the heavy metal status and bioaccumulation factors of V, Mn, Fe, Co, Cu, Zn, Se (essential metals) and Cr, Ni, Cd, As and Pb (potentially toxic metals) in medicinal plants grown under two different environmental conditions e.g., near to Khetri copper mine and those in fertile soils of Haridwar, both in India, using Instrumental Neutron Activation Analysis (relative method) and Atomic Absorption Spectrometry. The copper levels in the medicinal plants from Khetri were found to be 3-4 folds higher (31.6–76.5 mg kg?1) than those from Haridwar samples (7.40–15.3 mg kg?1), which is correlated with very high copper levels (763 mg kg?1) in Khetri soil. Among various heavy metals, Cr (2.60–5.92 mg kg?1), Cd (1.47–2.97 mg kg?1) and Pb (3.97–6.63 mg kg?1) are also higher in concentration in the medicinal plants from Khetri. The essential metals like Mn (36.4–69.3 mg kg?1), Fe (192–601 mg kg?1), Zn (24.9–49.9 mg kg?1) and Se (0.13–0.91 mg kg?1) and potentially toxic metals like Ni (3.09–9.01 mg kg?1) and As (0.41–2.09 mg kg?1) did not show much variations in concentration in the medicinal plants from both Khetri and Haridwar. The medicinal plants from Khetri, e.g., Ocimum sanctum, Cassia fistula, Withania somnifera and Azadirachta Indica were found rich in Ca and Mg contents while Aloe barbadensis showed moderately high Ca and Mg. Higher levels of Ca-Mg were found to correlate with Zn (except Azadirachta Indica). The bioaccumulation factors (BAFS) of the heavy metals were estimated to understand the soil-to-plant transfer pattern of the heavy metals. Significantly lower BAF values of Cu and Cr were found in the medicinal plants from Khetri, indicating majority fraction of these metals are precipitated and were immobilized species unsuitable for plant uptake. Overall, Withania somnifera (Ashwagandha) showed very high metal bioaccumulation.  相似文献

Phenanthrene is commonly present together with heavy metals at many contaminated sites. This study investigated the influence of coexisting lead (Pb2+) or cadmium (Cd2+) on phenanthrene adsorption on soils. Batch experiments were conducted under different geochemical conditions including pH, mineral structure, organic matter content, and varying amounts of heavy metals. The results showed that the presence of heavy metals in solution at a fixed pH of 5.8 ± 0.1 enhanced phenanthrene adsorption, the extent of which was closely related to the concentrations and the electro-negativity of the metals. The enhancement on phenanthrene adsorption was positively correlated to the amount of adsorbed metals. Although Cd2+ is a softer Lewis acid, Pb2+ displayed a more significant effect as it was adsorbed to a greater extent on the soil surfaces. Thus, density of cation accumulation appears to be more influential than metal softness in enhancing phenanthrene adsorption. Moreover, with a portion of organic matter removed by heating at 550 °C, there was a stronger enhancement of phenanthrene adsorption by coexisting Pb2+, indicating an increasingly dominant mechanisms associated with Pb2+ at a lower organic matter content. Similar enhancement phenomenon was observed on bentonite and kaolinite, probably resulting from the cation-π bonding between the adsorbed soft metal cations and the aromatic ring of phenanthrene in solution. The desorption experiments further suggested that the bonding of phenanthrene adsorption was strengthened in the presence of Pb2+ and that a larger proportion of adsorbed phenanthrene remained on the soils (residual fraction) even after sequential methanol extractions. Further spectroscopic analyses and surface characterization are required to provide direct evidence of the formation and relative significance of cation-π bond for phenanthrene adsorption.  相似文献

The main purpose of the present study was to detect the acute toxicity of cadmium (Cd) in F. paulensis and to investigate its effect on oxygen consumption and ammonium excretion different salinities. First, we examined the acute toxicity of Cd in F. paulensis at 24, 48, 72, and 96-h lethal concentration (LC50). Cd was significantly more toxic at 5 salinity than at 20 and 36. The oxygen consumption and ammonium excretion were estimated through experiments performed on each of the twelve possible combinations of three salinities (36, 20 and 5), at temperature 20 °C. Cd showed a reduction in oxygen consumption at 5 salinity, the results show that the oxygen consumption decreases with respect to the Cd concentration. At the highest Cd concentration employed (2 mg L−1), the salinity 5 and the temperature at 20 °C, oxygen consumption decreases 53.7% in relation to the control. In addition, after separate exposure to Cd, elevation in ammonium excretion was obtained, wish were 72%, 65% and 95% higher than the control, respectively. The results show that Cd is more toxic to F. paulensis at lower salinities.  相似文献

EC50s for cadmium, copper, lead and zinc were determined for juvenile production of Folsomia candida at pH6.0, 5.0 and 4.5 in a standard laboratory test system. In contrast to most previous studies where metal toxicity was increased at low pHs, in our experiments there was no clear relationship between soil acidity and EC50-reproduction in this species. The EC50-reproduction values (μg g−1) for cadmium and zinc were similar at all three pHs (pH6.0: Cd 590, Zn 900; pH5.0: Cd 780, Zn 600; pH4.5: Cd 480, Zn 590). In contaminated field sites adjacent to primary zinc smelters, zinc is invariably present in soils at concentrations of at least 50 times that of cadmium Thus deleterious effects of mixtures of these metals on populations of Collembola in such sites can be attributed to zinc rather than cadmium.  相似文献

This investigation was undertaken to determine the impact of the insecticides Dursban 480 EC (with organophosphate compound chlorpyrifos as the active ingredient) and Talstar 10 EC (with pyrethroid bifenthrin as the active ingredient) on the respiration activity and microbial diversity in a sandy loam luvisol soil. The insecticides were applied in two doses: the maximum recommended dose for field application (15 mg kg?1 for Dursban 480 EC and 6 mg kg?1 for Talstar 10 EC) and a 100-fold higher dose for extrapolation of their effect. Bacterial and fungal genetic diversity was analysed in soil samples using PCR DGGE and the functional diversity (catabolic potential) was studied using BIOLOG EcoPlates at 1, 3, 7, 14, 28, 56 and 112 days after insecticide application. Five bacterial groups (α, β, γ proteobacteria, firmibacteria and actinomycetes) and five groups of fungi or fungus-like microorganisms (Ascomycota, Basidiomycota, Chytridiomycota, Oomycota and Zygomycota) were analysed using specific primer sets. This approach provides high resolution of the analysis covering majority of microorganisms in the soil. Only the high-dose Dursban 480 EC significantly changed the community of microorganisms. We observed its negative effect on α- and γ-proteobacteria, as the number of OTUs (operational taxonomic units) decreased until the end of incubation. In the β-proteobacteria group, initial increase of OTUs was followed by strong decrease. Diversity in the firmibacteria, actinomycetes and Zygomycota groups was minimally disturbed by the insecticide application. Dursban 480 EC, however, both positively and negatively affected certain species. Among negatively affected species Sphingomonas, Flavobacterium or Penicillium were detected, but Achromobacter, Luteibacter or Aspergillus were supported by applied insecticide. The analysis of BIOLOG plates using AWCD values indicated a significant increase in metabolic potential of microorganisms in the soil after the high-dose Dursban application. Analysis of respiration demonstrated high microbial activity after insecticide treatments; thus, microbial degradation was relatively fast. The half-life of the active insecticide compounds were estimated within the range of 25 to 27 days for Talstar and 6 to 11 days for Dursban and higher doses stimulated degradation. The recommended dose levels of both insecticides can be considered as safe for microbial community in the soil.  相似文献

Lead (II) has been as one of the most toxic heavy metals because it is associated with many health hazards. Therefore, people are increasingly interested in discovering new methods for effectively and economically scavenging lead (II) from the aquatic system. Recent studies demonstrate biosorption is a promising technology for the treatment of pollutant streams. To apply these techniques, suitable adsorbents with high efficiency and low cost are demanded. The waste biomass of Bacillus gibsonii S-2 biosorbent was used as low-cost biosorbent to remove metallic cations lead (II) from aqueous solution. To optimize the maximum removal efficiency, the effect of pH and temperature on the adsorption process was studied. The isotherm models, kinetic models and thermodynamic parameters were analysed to describe the adsorptive behaviour of B. gibsonii S-2 biosorbent. The mechanisms of lead (II) biosorption were also analysed by FTIR and EDX. The results showed that the optimum pH values for the biosorption at three different temperatures, i.e. 20, 30 and 40 °C, were determined as 4. The equilibrium data were well fitted to Langmuir model, with the maximum lead (II) uptake capacities of 333.3 mg?g?1. The kinetics for lead (II) biosorption followed the pseudo-second-order kinetic equation. The thermodynamic data showed that the biosorption process were endothermic (?G?<?0), spontaneous (?H?>?0) and irreversible (?S?>?0). The mechanism of lead (II) biosorption by the waste biomass of B. gibsonii S-2 biosorbent could be a combination of ion exchange and complexation with the functional groups present on the biosorbent surface. The application of the waste biomass of B. gibsonii S-2 for lead (II) adsorption, characterized with higher lead (II) sorption capacity and lower cost, may find potential application in industrial wastewater treatment.  相似文献
