Generic Escherichia coli was isolated from surface water and groundwater samples from two dairies in Northern California and tested for susceptibility to antibiotics. Surface samples were collected from flush water, lagoon water, and manure solids, and groundwater samples were collected from monitoring wells. Although E. coli was ubiquitous in surface samples with concentrations ranging from several hundred thousand to over a million colony-forming units per 100 mL of surface water or per gram of surface solids, groundwater under the influence of these high surface microbial loadings had substantially fewer bacteria (3- to 7-log10 reduction). Among 80 isolates of E. coli tested, 34 (42.5 %) were resistant to one or more antibiotics and 22 (27.5 %) were multi-antibiotic resistant (resistant to ≥3 antibiotics), with resistance to tetracycline, cefoxitin, amoxicillin/clavulanic acid, and ampicillin being the most common. E. coli isolates from the calf hutch area exhibited the highest levels of multi-antibiotic resistance, much higher than isolates in surface soil solids from heifer and cow pens, flush alleys, manure storage lagoons, and irrigated fields. Among E. coli isolates from four groundwater samples, only one sample exhibited resistance to ceftriaxone, chloramphenicol, and tetracycline, indicating the potential of groundwater contamination with antibiotic-resistant bacteria from dairy operations.  相似文献

Despite the number of studies describing metal hyper-accumulating plants and their associated bacteria in various regions and countries, there is no information on rhizosphere microbial potential of the Turkish serpentine soils. This study aimed to explore the rhizosphere microbial diversity of Ni-resistant, hyper-accumulating plants grown on Ni-rich soils and their metal tolerance–resistance characteristics. One hundred ninety-one locations were visited to collect soil and plant samples from different serpentine regions of Western Turkey. Following bioavailable and total Ni analysis of collected samples, the seeds of the selected plants with higher Ni content were taken to the growth/germination test in a range of serpentine soils in a growth chamber condition. In order to investigate the rhizosphere microbial diversity, Isatis pinnatiloba and Alyssum dasycarpum which were able to germinate and grow well in the preliminary tests, were introduced to 6-month greenhouse experiment in the range of three serpentine soils with higher bioavailable Ni content. I. pinnatiloba had a better stimulatory effect on the rhizosphere microbial diversity. A total of 22 bacterial isolates were identified from different soil conditions in the end of experiment. Following microbial identification and confirmation tests, 11 isolates were found to be resistant and tolerant to the increasing concentrations of Ni, Pb, Cd and Zn in the range of 50–2,000 mg L − 1, which was considerably higher than those indicated by earlier studies. The strains isolated and identified from the Turkish serpentine soils were the members of genera Arthrobacter, Bacillus, Microbacterium and Staphylococcus.  相似文献

Enterococci bacteria are used to indicate the presence of human and/or animal fecal materials in surface water. In addition to human influences on the quality of surface water, a cattle grazing is a widespread and persistent ecological stressor in the Western United States. Cattle may affect surface water quality directly by depositing nutrients and bacteria, and indirectly by damaging stream banks or removing vegetation cover, which may lead to increased sediment loads. This study used the State of Oregon surface water data to determine the likelihood of animal pathogen presence using enterococci and analyzed the spatial distribution and relationship of biotic (enterococci) and abiotic (nitrogen and phosphorous) surface water constituents to landscape metrics and others (e.g. human use, percent riparian cover, natural covers, grazing, etc.). We used a grazing potential index (GPI) based on proximity to water, land ownership and forage availability. Mean and variability of GPI, forage availability, stream density and length, and landscape metrics were related to enterococci and many forms of nitrogen and phosphorous in standard and logistic regression models. The GPI did not have a significant role in the models, but forage related variables had significant contribution. Urban land use within stream reach was the main driving factor when exceeding the threshold (> or =35 cfu/100 ml), agriculture was the driving force in elevating enterococci in sites where enterococci concentration was <35 cfu/100 ml. Landscape metrics related to amount of agriculture, wetlands and urban all contributed to increasing nutrients in surface water but at different scales. The probability of having sites with concentrations of enterococci above the threshold was much lower in areas of natural land cover and much higher in areas with higher urban land use within 60 m of stream. A 1% increase in natural land cover was associated with a 12% decrease in the predicted odds of having a site exceeding the threshold. Opposite to natural land cover, a one unit change in each of manmade barren and urban land use led to an increase of the likelihood of exceeding the threshold by 73%, and 11%, respectively. Change in urban land use had a higher influence on the likelihood of a site exceeding the threshold than that of natural land cover.  相似文献

The application of electrical fields and chelating agents is an innovative hybrid technology used for the decontamination of soil polluted by heavy metals. The effects of four center-oriented electrical fields and chelating agents on active fractions of lead and zinc were investigated in this pot experiment. Ethylenediaminetetraacetic acid (EDTA) as a synthetic chelator and cow manure extract (CME) and poultry manure extract (PME) as natural chelators were applied to the pots (2 g kg?1) 30 days after the first irrigation. Two weeks later, four center-oriented electrical fields were applied in each pot (in three levels of 0, 10, and 30 V) for 1 h each day for 14 days. The soil near the cathode and anodes was collected and analyzed as cathodic and anodic soil, respectively. Results indicated that the soluble–exchangeable fraction of lead and zinc were decreased in the cathodic soil, while the carbonate-bound fractions were increased. In the anodic soil, however, the opposite result was observed. EDTA enhanced the soluble–exchangeable form of the metals in both anodic and cathodic soils. Furthermore, the amounts of carbonate-bound heavy metals were increased by the application of CME in both soils. The organic-bound fraction of the metals was increased by the application of natural chelators, while electrical fields had no significant impacts on this fraction.  相似文献

Phytoremediation, an approach that uses plants to remediate contaminated soil through degradation, stabilization or accumulation, may provide an efficient solution to some mercury contamination problems. This paper presents growth chamber experiments that tested the ability of plant species to stabilize mercury in soil. Several indigenous herbaceous species and Salix viminalis were grown in soil collected from a mercury-contaminated site in southern Poland. The uptake and distribution of mercury by these plants were investigated, and the growth and vitality of the plants through a part of one vegetative cycle were assessed. The highest concentrations of mercury were found at the roots, but translocation to the aerial part also occurred. Most of the plant species tested displayed good growth on mercury contaminated soil and sustained a rich microbial population in the rhizosphere. The microbial populations of root-free soil and rhizosphere soil from all species were also examined. An inverse correlation between the number of sulfur amino acid decomposing bacteria and root mercury content was observed. These results indicate the potential for using some species of plants to treat mercury contaminated soil through stabilization rather than extraction. The present investigation proposes a practical cost-effective temporary solution for phytostabilization of soil with moderate mercury contamination as well as the basis for plant selection.  相似文献

This study investigated the effects of supplementing 40 g lauric acid (C12) kg-1 dry matter (DM) in feed on methane emissions from early-lactating dairy cows and the associated effects on methane, nitrous oxide and ammonia release from the manure during storage. Stearic acid (C18), a fatty acid without assumed methane-suppressing potential in the digestive tract of ruminants, was added at 40 g kg-1 DM to a control diet. The complete feed consisted of forage and concentrate in a ratio of 1.5:1 (DM basis). The manure was stored for 14 weeks either as complete slurry or, separately, as urine-rich slurry and farmyard manure representing two common storage systems. Methane release of the cows, as measured in respiratory chambers, was lower with C12 by about 20%, but this was mostly resulting from a reduced feed intake and, partly, from a lower rate of fibre digestion. As milk yield declined less than feed intake, methane emission per kg of milk was significantly lower with C12 (11.4 g) than with C18 (14.0 g). Faeces of C12-fed cows had a higher proportion of undigested fibre and accordingly methane release from their manure was higher compared with the manure obtained from the C18-fed cows. Overall, manure-derived methane accounted for8.2% and 15.4% of total methane after 7 and 14 weeks of storage, respectively. The evolution of methane widely differed between manure types and dietary treatments, with a retarded onset of release in complete slurry particularly in the C12 treatment. Emissions of nitrous oxide were lower in the manures from the C12 treatment. This partially compensated for the higher methane release from the C12 manure with respect to the greenhouse gas potential. The total greenhouse gas potential (cow and manure together) accounted for 8.7 and 10.5 kg equivalents of CO2 cow-1 d-1with C12 and C18, respectively. At unaffected urine-N proportion ammonia and total nitrogen losses from stored manure were lower with C12 than with C18 corresponding to the differences in feed and nitrogen intake. The present results suggest that manure storage significantly contributes to total methane emission from dairy husbandry, and that the identification of effective dietary mitigation strategies has to consider both the digestive tract of the animals and the corresponding manure.  相似文献

Today's large-scale poultry production is often accompanied by high concentrations of airborne microorganisms at working places. However, the microbial communities in those bioaerosols are rarely characterised. In this study, we investigated the bacterial population in bioaerosols from a duck hatchery by both cultivation based and molecular methods and compared the results. Depending on used media, concentrations of airborne culturable bacteria varied between 6 × 10(1) and 7 × 10(6) CFU per m(3) air. The corresponding total cell count of DAPI stained cells was 2 × 10(7) cells per m(3) air. 16S rRNA gene analyses of bacterial isolates and clone libraries revealed a low species richness in hatcheries air, respectively. More than 50% of bacterial isolates were phylogenetically most closely related to bacterial species of the risk group 2 (German TRBA). The sequence composition in clone libraries supported the result of cultivation based approaches, whereby sequences assigned to Staphylococcus, Acinetobacter and Enterococcus are the most common. The high concentration of airborne bacteria which are most closely related to species of potential health risk requires further detailed investigations for these bacterial species.  相似文献

Numerous studies have demonstrated that anaerobic packed-bed reactors (APBRs) containing media such as clay chip, small stone, and porous plastic modules are effective for treating a variety of organic wastes. Most of these studies focused on the treatment capabilities of the processes, yet little work was done about the factors affecting their designs and performance. This paper presents the results of a study on a laboratory scale APBR system examining the significance of media factors such as media roughness, specific surface area, porosity and pore size on treatment performance. The results showed that media roughness and pore size were more important over surface area on the reactor performance. Within the same media porosity, pore size plays a more significant role than surface area in term of COD removal efficiency. The study indicates that the ability of a support medium to retain biomass either as suspended growth entrapped in the void space, or by attachment to the media surface is a significant consideration in an upflow APBR. A testing protocol for measuring methane gas produced by the suspended biomass using a batch serum bottle technique was developed and presented in this study. The method involved measurements of hourly methane production and instantaneous TOC concentration from the suspended biological solids extracted from various reactor heights. The proportion of methane production associated with the suspended biomass was as high as 58% at organic loading rate of 16 g COD/L.day. The results suggest that media pore size and porosity play a significant role in the performance of upflow APBRs.  相似文献

A sampler was needed for a spatial and temporal study of microbial and chemical stratification in a large swine manure lagoon that was known to contain zoonotic bacteria. Conventional samplers were limited to collections of surface water samples near the bank or required a manned boat. A new sampler was developed to allow simultaneous collection of multiple samples at different depths, up to 2.3 m, without a manned boat. The sampler was tethered for stability, used remote control (RC) for sample collection, and accommodated rapid replacement of sterile tubing modules and sample containers. The sampler comprised a PVC pontoon with acrylic deck and watertight enclosures, for a 12 VDC gearmotor, to operate the collection module, and vacuum system, to draw samples into reusable autoclavable tubing and 250-mL bottles. Although designed primarily for water samples, the sampler was easily modified to collect sludge. The sampler held a stable position during deployment, created minimal disturbance in the water column, and was readily cleaned and sanitized for transport. The sampler was field tested initially in a shallow fresh water lake and subsequently in a swine manure treatment lagoon. Analyses of water samples from the lagoon tests showed that chemical and bacterial levels, pH, and EC did not differ between 0.04, 0.47, and 1.0 m depths, but some chemical and bacterial levels differed between winter and spring collections. These results demonstrated the utility of the sampler and suggested that future manure lagoon studies employ fewer or different depths and more sampling dates.  相似文献

Plots of Arabidopsis thaliana L. (Heynh.) plants were grown in two hydroponic systems, one using a solid rooting support media (vermiculite) and the other a flowing solution culture. Both were tested with various concentrations of bromacil (5-bromo-3-sec-butyl-6-methyluracil), dichlobenil (2,6-dichlorobenzonitrile), copper sulfate (CuSO4·5H2O) and 3-aminotriazole. The effects on biomass production were compared. Plants treated with bromacil at concentrations up to 0.05 mg l–1 were similarly reduced in growth in both culture methods. However, the toxic effects of dichlobenil, copper sulfate and 3-aminotriazole were different for both culture methods. Dichlobenil decreased plant growth in the solution culture at lower concentrations than in the vermiculite system. All aspects of growth were suppressed in the solution culture while the vegetative weight was unaffected in the vermiculite culture. Plants treated with 20 mg l–1 copper sulfate in solution culture yielded mean seed weight of 1.2 g/plot compared to 3.1 g/plot when treated with the same concentration in the double-plot system. Plants treated with 0.5 mg l–1 3-aminotriazole in solution culture produced no seed, while plants with the same nominal concentration in the double-pot method produced a mean seed weight of 6.5 g/plot. Solution culture testing was generally more sensitive and since roots were bathed directly in a solution of known pH, nutrient level and chemical concentration, the test conditions were more accurately defined.  相似文献

Nanotechnology is currently at the forefront of scientific research and technological developments that have resulted in the manufacture of novel consumer products and numerous industrial applications using engineered nanomaterials (ENMs). With the increasing number of applications and uses of ENMs comes an increasing likelihood of nanoscale materials posing potential risks to the environment and engineered technical systems such as wastewater treatment plants (WWTPs). Recent scientific data suggests that ENMs that are useful in, for example, medical applications due to their novel physicochemical properties, may also cause adverse effects to the bacterial populations used in wastewater treatment systems. In this review, the toxicological effects of titanium nanoparticles (nTiO(2)), zinc oxide (nZnO), carbon nanotubes (CNTs), fullerenes (C(60)) and silver nanoparticles (AgNPs) to bacteria were examined. The results suggest that the potential ENMs risks to bacteria are non-uniform (need to be assessed case-by-case), and are dependent on numerous factors (e.g. size, pH, surface area, natural organic matter). Currently available data are therefore insufficient for evaluating the risks that ENMs pose in WWTPs. To fill these knowledge gaps, we recommend scenario specific studies aimed at improving our understanding on: (i) estimated volumes of ENMs in effluents, (ii) the antibacterial sensitivity of cultures within WWTPs towards selected ENMs, and (iii) processes improving the stability of ENMs in solutions. Two factors that merit consideration for elucidating the potential risks systematically are the toxicity mechanisms of ENMs to bacteria, and the influencing factors based on inherent physicochemical properties and environmental factors. Furthermore, the complexity of behaviour and fate of ENMs in real WWTPs requires case studies for assessing the ENMs risks to bacteria in vivo. The current laboratory results derived using simplified exposure media do not reflect actual environmental conditions.  相似文献

A cellulose acetate membrane, positioned in a passive sampler system, was coated with the perfluorinated polymer Nafion. Aqueous metal transport through the membrane and the Nafion film was investigated. The Nafion-coated membrane prevents the transport of metals bound to natural organic material to the receiving phase in the passive sampler, while copper associated with small synthetic ligands pass through the Nafion film. Increasing thickness of the Nafion film decreases the metal uptake rate to the receiving phase in the passive sampler and increases the response time. A decreased response time for the passive sampler would provide a better estimation of rapid changes in concentration, particularly for dynamic aquatic systems such as urban runoff. Limited biofouling protection with the Nafion coating was demonstrated by growth inhibition of natural periphyton communities on glass surfaces.  相似文献

The knowledge of enteric bacteria survival kinetic is very important for environmental scientists. Enteric bacteria andspecifically the fecal indicator bacteria are typically used to measure the sanitary quality of water for recreational, industrial, agricultural and water supply purposes. They are released into the environment with feces, and are then exposedto a variety of environmental conditions that eventually causetheir death. In general, it is believed that the fecal indicatorcannot grow in natural environments, since they are adapted to live in the gastrointestinal tract. Studies have shown that fecalindicator bacteria survive from a few hours up to several daysin surface water, but may survive for days or months in lake-sediments, where they may be protected from sunlight andpredators. We assume that pathogens similar to the fecal indicator bacteria die at the same rate as fecal indicator bacteria. Therefore, if we find relatively high numbers of fecalindicator bacteria in an environment, we assume that there is anincreased likelihood of pathogens being present as well. The kinetic of enteric bacteria survival in natural waters is affected by a large number of factors. One of them is the temperature. The aim of this contribution was the experimentalresearch of the survival kinetic of enteric bacteria applying a simple mathematical formula, which describes the survival kineticpredicting the decay phase at various temperatures. We aspire that the results will lead both to the solution of many engineering problems and to future research.  相似文献

The occurrence of pharmaceutically active chemicals (PACs) in the natural aquatic environment is recognized as an emerging issue due to the potential adverse effects these compounds pose to aquatic life and humans. This study presents the monitoring of two major categories of PACs in surface water: steroid hormones and antibiotics. Surface water samples were collected in the fall season from 21 locations in suburban (4), agricultural (5) and mixed (12) use suburban and agricultural areas. The water samples collected were analyzed using GC/MS for aqueous concentration of eleven steroid hormones: six natural (17alpha-estradiol, 17beta-estradiol, estrone, estriol, 17alpha-dihydroequilin, progesterone) and five synthetic (gestodene, norgestrel, levonorgestrel, medrogestone, trimegestone). In addition, 12 antibiotics (oxytetracycline, chlorotetracycline, tetracycline, sulfamethoxazole, sulfamethazine, trimethoprim, lincomycin, norfloxacin, ofloxacin, roxithromycin, erythromycin, tylosin tartrate) were analyzed using LC/MS. Steroid hormones detected in surface water were: 17alpha-estradiol, 17beta-estradiol, 17alpha-dihydroequilin, estriol, estrone, progesterone and trimegestone. Estrone had the highest detection frequency of >90% with concentrations ranging from 0.6 to 2.6 ng/l. The second most frequently detected estrogen was estriol (>80%) with concentrations ranging from 0.8 to 19 ng/l. The detection frequency varied at different sampling locations. No antibiotics were detected in the 21 streams sampled. This study aims to give a better understanding on the presence, fate and transport of PACs derived from humans and animals.  相似文献

The presence of humic acid (HA) in water poses environmental problems because it enhances the transport of several contaminants. A series of column experiments was conducted toward studying HA transport in different porous media under various pH, ionic strength, and flow rate conditions. The results showed that decreasing pH and increasing ionic strength increased adsorption and therefore delayed the transport of HA in porous media. However, increasing flow rate accelerated the transport of HA in porous media. The effects of pH, ionic strength, and flow rate varied with the solid matrix and were more evident in sands of smaller average particle diameter than in those with larger ones. These factors must be considered when predicting the environmental fate of HA. The results also suggested that HA adsorption was an important process controlling HA transport and should be considered in studies of HA behavior in porous media.  相似文献

The mobility of arsenic in Cambisol under the influence of added barnyard manure and application of a phosphate solution to the soil was described on the basis of column experiments. A soil sample containing 126 mg/kg total As and 3.72 mg/kg specifically-sorbed As was extracted using demineralized water (DIW) or a 28 μmol/l phosphate solution in a column containing untreated soil and in a column with added barnyard fertilizer. The pH, Eh, alkalinity, main components, including DOC, and selected trace elements (Al, Mn, Pb, Cu, Zn, Cd, Cr, Co, Ni, Ba and As) were determined in the extracts. Hydrodynamic tests of the flow and transport of the substances in the columns were carried out. The addition of barnyard manure was manifested in elevated concentrations of the main inorganic components, DOC and Al and a decrease in the Eh value and Mn concentration. Application of a phosphate solution was manifested in a decrease in the NO3 and SO4 concentrations, probably as a result of the effect of increased biological fixation. The As concentration in extracts varied from 8.8 to 15.5 μg/l and was not dependent on the composition of the extracting solution or the addition of barnyard fertilizer.  相似文献

High levels of fecal indicator bacteria (FIB) in surface waters is a common problem in urban areas that often leads to impairment of beneficial uses such as swimming. Once impaired, common management and regulatory solutions include development of total maximum daily loads (TMDLs) and other water quality management plans. A critical element of these plans is establishment of a "reference" level of exceedances against which to assess management goals and TMDL compliance. The goal of this study was to provide information on indicator bacteria contributions from natural streams in undeveloped catchments throughout southern California during dry weather, non-storm conditions. To help establish a regional reference data set, bacteria levels [i.e. Escherichia coli (E. coli), enterococci and total coliforms] were measured from 15 unimpaired streams in 11 southern California watersheds weekly for one full year. Concentrations measured from reference areas were typically between one to two orders of magnitude lower than levels found in developed watersheds. Nearly 82% of the time, samples did not exceed daily and monthly bacterial indicator thresholds. E. coli had the lowest daily percent exceedance (1.5%). A total of 13.7% of enterococci exceeded daily thresholds. Indicator bacteria levels fluctuated seasonally with an average of 79% of both enterococci and total coliforms exceedances occurring during summer months (June to August). Temperature, at all sites, explained about one-half the variation in total coliforms density suggesting that stream temperatures regulated bacterial populations. Accounting for natural background levels will allow for management targets that are more reflective of the contributions from natural sources.  相似文献

Titanium nanomaterials are likely to sink into sediments in substantial quantities due to their wide use in a number of applications for decades. To assess the potential environmental consequences, a better understanding is required of the occurrence and sources of titanium (Ti) nanomaterials in sediments. In this research, we provide the first report of the Ti concentrations and the morphology and composition of Ti-based solids in surface sediments from Xiamen Bay, China. Results indicated that the anthropogenic Ti concentrations in the surface sediments from Xiamen Bay reached approximately 2.74 g kg(-1). Ti nanomaterials could be found in sediments with elevated Ti concentrations, which were often aggregated to a few hundred nanometers (<300 nm) and were composed of several spherical particles, less than 50 nm in size, that were made solely of TiO(x). However, Ti particles (approx. 300-700 nm) could be also found in sediments with lower Ti concentrations, which were presumably components of the natural clay mineral kaolinite. Ti nanomaterials could be easily distributed in sediments associated with elevated levels of organic matter and preferentially attach to those sediments with elevated fine fractions. As a sentinel, or tracer, for other nanomaterials, the field-scale investigation of Ti nanomaterials would contribute to increasing our knowledge on the behavior of engineered nanomaterials in an aquatic environment.  相似文献

Bacteria play a significant role in water contamination. Chemicals are mostly used for the treatment of bacteriologically contaminated water. The use of bacterial interactions is a new approach to limit the pathogens' growth. Detection of antimicrobial substances produced by lactic acid bacteria against the waterborne pathogens is the objective of this work. Microbiological and biochemical methods were used to identify lactic acid bacteria having an antimicrobial activity. Evaluation of antimicrobial activity with growth kinetic measurements was performed. Four isolates of lactic acid bacteria obtained from whey and curd were identified. The predominant species belonging to the Lactobacillus genera are: Lactobacillus rhamnosus, Lactobacillus sakei, Lactobacillus paracasei, and Lactobacillus paraplantarum. The present study revealed that the Lactobacillus consortium is able to inhibit Staphylococcus aureus's growth along with Escherichia coli and Vibrio species. In mixed culture, after 24?h, the Lactobacillus consortium reduces the growth of S. aureus by 2.03 log; moreover, the growth of the latter bacteria totally ceased after 72?h of incubation. The protein produced by the Lactobacillus consortium was responsible for arresting the growth of S. aureus.  相似文献
