应用未分化型PC12细胞为体外模型,研究甲基汞对多巴胺能神经元的毒性作用机制.利用MTT法检测甲基汞的细胞毒性,Hoechst33258荧光染色观察细胞形态变化,流式细胞术定量分析细胞周期分布和凋亡比率,并进一步通过比色法评价各项氧化应激指标.结果显示,甲基汞以剂量依赖性方式降低了细胞存活率,干扰细胞周期,造成S期细胞阻滞,细胞出现典型的凋亡性死亡特征.另外,甲基汞暴露导致PC12细胞内O-.2、H2O2和·OH等活性氧含量急剧增加,脂质过氧化程度升高,且SOD、CAT和GSH-Px等抗氧化酶活性降低.因此,甲基汞对多巴胺能神经元具有强烈的毒性,毒性作用机制涉及到干扰细胞周期、诱导细胞凋亡和引发严重的氧化损伤.  相似文献

Enantioselectivity of chiral pollutants is receiving growing concern due to the difference in toxicology and environment fate between enantiomers.In this study,enantiomers of insecticide beta-cypermethrin (beta-CP) were separated on selected chiral column by HPLC,and the toxicity of enantiomers was evaluated using the zebrafish embryo-larval assays.The enantiomers of beta-CP were baseline separated on Chiralcel OD and Chiralpak AD columns and detected by circular dichroism (CD) at 236 nm.Better separation could be achieved at lower temperature (e.g.,20°C) and with lower levels of polar modifiers.Pure enantiomers were obtained on Chiralcel OD.The CD spectra of enantiomers were recorded.By comparing the elution order with a previous similar study,the absolute configuration of beta-CP enantiomers was determined.The individual enantiomers were used in zebrafish embryo test,and the results showed that beta-CP enantioselectively induced yolk sac edema,pericardial edema and crooked body.The 1R-cis-αS and 1R-trans-αS enantiomers showed strong developmental toxicities at concentration of 0.1 mg/L,while the 1S-cis-αR and 1S-trans-αR induced no malformations at higher concentration (e.g.,0.3 mg/L).The results suggest that the enantioselective toxicological effects of beta-CP should be considered when evaluating its ecotoxicological effects.  相似文献

The interaction between zinc and cadmium was investigated in tomato plants (Solanum lycopersicum). Ten-day-old seedlings were treated with 10 μmol/L CDC12 associated to different concentrations of ZnC12 (10, 50, 100, and 150 μmol/L). Zn supply clearly reduced Cd accumulation in leaves and simultaneously increased Zn concentration. Cd induced oxidative stress in leaves as indicated by an increase in thiobarbituric acid-reactive substances (TBARS) level and chlorophyll breakdown. Furthermore, compared with control, Cdtreate plants had significantly higher activities of superoxide dismutase (SOD, EC 1.15.1.1), whereas, catalase (CAT, EC 1.111.1.6),ascorbate peroxidase (APX, EC 1.11.1.11), and glutathione reductase (GR, EC 1.6.4.2) activities were significantly suppressed by Cd addition. Zn supplementation, at low level, restored and enhanced the functional activity of these enzymes (SOD, CAT, APX and GR) as compared to Cd-alone-treated plants. The beneficial effect of adequate Zn level on Cd toxicity was confirmed by a significant decrease in TBARS level and restoration of chlorophyll content. However, when Zn was added at high level in combination with Cd there was an accumulation of oxidative stress, which was higher than that for Cd or excess Zn alone treatments. These results suggested that higher Zn concentrations and Cd are synergistic in their effect on plant growth parameters and oxidative stress.  相似文献

以3种不同质量浓度的H2O2分别对沉水植物黑藻做3种不同程度的氧化胁迫预处理,再经冷胁迫后测定黑藻顶枝的膜脂过氧化程度(MDA含量)、细胞抗氧化酶(SOD,CAT)活性.结果发现低浓度氧化胁迫预处理(0.06mmol/L的H2O2浸泡4或6 h预处理组)的黑藻经冷胁迫处理后,成活率不降低,其膜脂过氧化程度和细胞抗氧化酶的活性还有提高;而经中、高浓度氧化胁迫预处理(0.60mmol/L的H2O2浸泡6和12 h预处理组或1.00mmol/L的H2O2浸泡4,6和12 h预处理组)后的黑藻在冷胁迫处理后的结果则相反.因此,沉水植物黑藻对氧化胁迫感知并做出反应的机制(氧化应激机制),在其对低温反应和适应过程中起着很重要的调节作用.建议早春时节在进行水生植物恢复过程中可采用适宜浓度的H2O2预处理植株,以提高植株的适应性和保持成活率,进一步提高水生植物恢复的效率和效果.  相似文献

以嗜热四膜虫(Tetrahymena thermophila)作为受试生物,考察了纳米材料氧化石墨烯(GO)对其细胞生长率、乙酰胆碱酯酶(AchE)和氧化应激酶活性、生物膜损伤及细胞凋亡的影响,以探究GO的毒性效应.结果表明,GO浓度高于32mg/L时显著抑制嗜热四膜虫的细胞增殖(P<0.05),细胞存活率低于50%;在0~64mg/L实验范围内,随GO暴露浓度增加,细胞内活性氧自由基(ROS)和超氧化物歧化酶(SOD)水平呈先升后降的趋势,AchE活性受抑;GO抑制位于线粒体内膜的琥珀酸脱氢酶(SDH)活性,促进细胞质中乳酸脱氢酶(LDH)的释放;64mg/L GO导致四膜虫细胞出现明显凋亡现象.以上结果显示,中低浓度GO(0~8mg/L)暴露下,氧化应激机制对细胞毒性起主要贡献作用;高浓度GO(32和64mg/L)作用下,四膜虫凋亡现象的产生可能是GO抑制其生长作用导致的.  相似文献

目前,有关三氟羧草醚(Acifluorfen,AF)的神经毒性未见报道且亟待确定.为探讨AF经口暴露对小鼠学习记忆能力的影响及其可能机制,将30只雄性昆明小鼠随机分成生理盐水对照组、0.13、1.3、13和130 mg·kg~(-1)·d~(-1) AF染毒组共5组,灌胃染毒14 d,进行Morris水迷宫实验,观察脑海马病理切片,并检测脑组织中活性氧(ROS)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、磷酸化cAMP反应元件结合蛋白(pCREB)和脑源性神经营养因子(BDNF)含量.结果显示,与对照组相比,AF染毒剂量分别为13和130 mg·kg~(-1)·d~(-1)时,小鼠行为学上学习记忆能力下降;海马细胞排列松散;出现氧化损伤,其中,ROS含量显著升高(p0.05),GSH含量显著减少(p0.05);神经保护能力减弱,其中,pCREB和BDNF水平显著(p0.05)或极显著(p0.01)下降.结果表明,AF能够导致小鼠学习记忆能力下降,氧化应激和CREB-BDNF通路级联下调可能是AF造成神经毒性的机制之一.  相似文献

Pollution of marine environment has become an issue of major concern in recent years.Serious environmental pollution by heavy metals results from their increasing utilization in industrial processes and because most heavy metals are transported into the marine environment and accumulated without decomposition.The aim of the present study is to investigate the effects on growth,pigments, lipid peroxidation,and some antioxidant enzyme activities of marine microalga Pavlova viridis,in response to elevated concentrations of cobalt(Co)and manganese(Mn),especially with regard to the involvement of antioxidative defences against heavy metal-induced oxidative stress.In response to Co~(2 ),lipid peroxidation was enhanced compared to the control,as an indication of the oxidative damage caused by metal concentration assayed in the microalgal cells but not Mn~(2 ).Exposure of Pavlova viridis to the two metals caused changes in enzyme activities in a different manner,depending on the metal assayed:after Co~(2 )treatments,total superoxide dismutase(SOD)activity was irregular,although it was not significantly affected by Mn~(2 )exposure.Co~(2 )and Mn~(2 )stimulated the activities of catalase(CAT)and glutathione(GSH),whereas,ghitathione peroxidase(GPX)showed a remarkable increase in activity in response to Co~(2 )treatments and decreased gradually with Mn~(2 )concentration,up to 50μmol/L,and then rose very rapidly,reaching to about 38.98% at 200μmol/L Mn~(2 ).These results suggest that an activation of some antioxidant enzymes was enhanced,to counteract the oxidative stress induced by the two metals at higher concentration.  相似文献

以半滑舌鳎肝脏细胞(HTLC)为研究对象,利用噻唑蓝比色法(MTT)、乳酸脱氢酶(LDH)、活性氧自由基(ROS)和超氧化物歧化酶(SOD)等方法分析细胞活性、细胞膜完整程度以及氧化应激反应相关指标,探究海洋环境中放射性核素137Cs、90Sr的生物毒性效应。HTLC细胞活性随137Cs、90Sr活度升高呈先增加后降低的趋势,且在低活度核素作用下,细胞活性增加极其显著(p < 0.01),137Cs对细胞膜无损伤,90Sr则使细胞膜损伤显著(p < 0.05),但二者均能使ROS含量和SOD活性显著性增加(p < 0.05)。高活度核素作用下SOD活性均显著增加(p < 0.05),但137Cs作用时ROS无显著变化,90Sr则使ROS显著增加(p < 0.05)。研究表明,在本实验活度条件下,137Cs和90Sr对海洋鱼类细胞无显著的生物毒性,细胞能够通过自我调节维持正常功能。90Sr能产生氧化应激反应,对膜系统造成损伤,可能影响细胞多种代谢途径,对生物体具有潜在的毒性效应。  相似文献

为研究城市污水处理厂(STPs)尾水是否会引起海洋生物的氧化逆境,将取自青岛市李村河污水处理厂的尾水用海水稀释到不同浓度(0%、1%、5%、10%、20% V/V), 对文蛤(Meretrix meretrix)暴露培养15d, 分析其内脏中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)、谷胱甘肽还原酶(GR)的活性. 结果表明:SOD活性随尾水暴露时间延长基本表现为“抛物线型”的响应特征,体积比10%、20%的尾水暴露6~12d期间,文蛤体内SOD活性均受到诱导,最大增幅为15.88%;CAT和GPx对尾水的主要响应特征是抑制效应,分别在暴露9d、12d时最为明显,最大抑制率分别为64.38%和52.39%;较高浓度(20%)的尾水能够持续抑制GR活性,而其他处理组中,GR对尾水表现为“抛物线型”的响应特征,即:暴露3d或6d时活性增加,而后显著降低.GR活性的最大诱导出现在暴露3d和6d时,分别比对照组上升81.47%和80.91%.研究结果进一步表明STPs不能有效降解有害化学物质以消除尾水的毒性, 文蛤内脏SOD、CAT、GPx和GR活性能够敏感指示尾水诱导的氧化逆境.  相似文献

为了探讨Fas/FasL途径在氟暴露致PC12细胞凋亡中的作用及其机制,采用含20、40、80、160mg/L NaF培养液处理PC12细胞.结果表明,所有剂量NaF处理12、24、36、48h,PC12细胞活性升高;上述不同剂量NaF处理24h后,与对照组比,PC12细胞的活性氧水平、细胞凋亡率、细胞内Fas/FasL信号转导通路Fas和FasL、Caspase8、FADD、Caspase3基因和蛋白表达水平均呈显著上升(P < 0.05),而Bid基因和蛋白表达水平显著下降(P < 0.05),且呈氟暴露剂量依赖性.结果提示Fas/FasL途径在氟暴露致PC12细胞凋亡中起重要作用,其中FADD可能是Fas/FasL凋亡途径中的重要靶分子.  相似文献

采用盆栽实验的方法,研究了百菌清对旱稻 (Oryza sativa L.)生长和氧化胁迫的影响,以及接种丛枝菌根真菌(arbuscular mycorrhizal fungi, AMF)对百菌清污染下旱稻生长的影响和对旱稻氧化胁迫的影响.结果表明,百菌清处理对旱稻的生长有明显的抑制作用,施加百菌清后可以使旱稻地上部的生物量由2.5 g·pot-1下降到1.0 g·pot-1以下;地下部分的生物量在施加百菌清处理后由0.9 g·pot-1下降到0.3 g·pot-1以下.百菌清处理下接种AMF能显著提高旱稻生物量,接种菌根真菌能使50 mg·kg-1百菌清处理下旱稻的生物量增加1倍以上.百菌清处理能显著降低旱稻对磷(P)营养元素的吸收,在50 mg·kg-1百菌清处理下旱稻地上部分P的吸收量由3 200 μg·pot-1下降到860 μg·pot-1,接种AMF能使旱稻地上部P的吸收量增加到1 900 μg·pot-1.百菌清处理还可以引起旱稻体内的氧化胁迫,改变旱稻体内抗氧化酶系统的活性,而接种菌根真菌能降低百菌清对旱稻产生的氧化胁迫,从而减轻百菌清污染对旱稻的毒性作用.总之,百菌清土壤污染能够引起旱稻体内的氧化胁迫、降低旱稻P的吸收量而影响植物生长,接种菌根真菌能显著改善旱稻的生长和降低百菌清对旱稻的影响.  相似文献

Prior to a prospective application of amniotic fluid (AF) cell filtration to early amniocentesis, we tested the technique on a surplus from mid-trimester samples. By using the same sample size of 5 ml in experiments with a filter and in routine diagnostic procedures (control), we evaluated an optimal filter system. The prolonged culture time of filtered cells and the reduced number of clones are most probably due to mechanical stress (filtration pressure), whereas loss of the cells by adhesion to the filter system, and an AF-free culture medium (growth factors) are suggested to be less important. The AF cells are very sensitive to mechanical stress. Slow filtration (⩽3 ml AF/min) through filters with a high porosity and the largest possible pore size should be preferred. A mixed cellulose ester filter membrane with a pore size of 5·0 μm proved to be the most efficient, allowing harvest of the filtered cells after only a slight prolongation of the culture time (+2·4 days) compared with unfiltered aliquots. A filter set with a bypass connected by three-way taps allows cell filtration during either aspiration or reinjection of the AF. Cell filtration after amniocentesis and consecutive reverse flushing of the membrane with the appropriate amount of culture medium proved to be the best with regard to easy handling and reducing the risk of bacterial contamination.  相似文献

研究了聚苯乙烯微塑料(PS,0.5μm,10~100mg/L)在120h内对三角褐指藻(Phaeodactylum tricornutum)的生长及其释放三卤代甲烷的影响.结果表明,PS显著抑制三角褐指藻的生长.高浓度的PS (100mg/L)条件下,三角褐指藻的细胞生长,叶绿素a含量及光合效率的最大抑制率分别达31.75%,10.38%和8.82%.PS诱导微藻细胞内产生氧化应激,增加细胞内活性氧的含量.培养前期(0~48h),PS引起微藻细胞发生氧化应激,促进三角褐指藻释放3种三卤代甲烷;培养后期(72~120h),PS抑制细胞的生理状态及3种三卤代甲烷的释放.这些结果表明微塑料在一定浓度下会影响藻类细胞生长代谢,三卤甲烷的生产释放是一种通过细胞代谢来抵抗氧化应激的保护机制.  相似文献

为了探究新型增塑剂邻苯二甲酸二异壬酯(DINP)对小鼠过敏性皮肤炎症的影响,采用皮肤染毒的暴露方式,56只SPF级雄性Balb/C小鼠随机平均分为7组处理40d.用0、1.4、14和140mg/kg DINP对小鼠进行皮肤染毒,并用抗氧化剂褪黑素设置2组来探究氧化应激的介导作用,以及1组生理盐水对照组.染毒结束后取耳组织测定耳肿指数和双耳重量差,并制作耳组织病理学切片,最后检测氧化应激指标.实验结果显示, 14和140mg/kg DINP皮肤暴露组的皮肤炎症有明显加剧(P<0.01),氧化应激指标也有明显变化(P <0.01).这表明,一定浓度的DINP,经较长时间的暴露,对小鼠过敏性皮肤炎症具有明显的佐剂作用.并且,随着DINP浓度升高,小鼠机体氧化应激过度活化,造成机体组织或细胞的氧化损伤,并加剧了炎症反应.  相似文献

Chiral pesticides account for 30% of pesticides. Pesticides are inevitably leached into the groundwater by runoff. At the watershed level, the distribution characteristics of enantiomers in sediments collected from the river network of an agricultural area near the middle and lower reaches of the Yangtze River were tested, and their potential correlations with the physicochemical properties and microbial communities of the sediments were analyzed. The sediment pollution was serious at sites 8 and 9, with their pollution source possibly being agricultural or industrial sewage. Moreover, there were higher cumulative contents of pesticide residues at sites 4, 8, and 9. Specifically, Cycloxaprid was the most detected chiral pesticide in the study area, followed by Dinotefuran and Diclofop-methyl. Additionally, Ethiprole and Difenoconazole had strong enantioselectivity in the study area. Interestingly, the enantiomers of some chiral pesticides, such as Tebuconazole, had completely different distributions at different sites. Pearson correlation analysis showed that sediment catalase and microbial biomass carbon were important factors for enantioselectivity of chiral pesticides. The effect of sediment physicochemical properties on enantioselective distribution was achieved by influencing the microorganisms in the sediment. Furthermore, the enantioselective distribution of Tebuconazole was closely related to the genus Arenimonas. Overall, the enantioselective distribution of most of the chiral pesticides was positively correlated with the prokaryotic microbial community. This study provides empirical support for agricultural non-point source pollution caused by chiral pesticides, and also lays a research foundation for exploring the factors that affect the fate of chiral pesticides in the environment.  相似文献

手性农药分子的不同对映体在环境中的降解速率和对于生物的毒性可能存在很大的差异,因此拆分并使用对靶标生物毒性较高或对非靶标生物毒性较低的单一或浓缩的对映体配方可以有效地降低杀虫剂的环境风险.本文用手性柱拆分了一种广泛使用的手性农药,氟虫腈的两种对映体,研究了这两种对映体和消旋体对靶标生物小菜蛾和非靶标生物斑马鱼的急性毒性.结果表明氟虫腈对受试靶标和非靶标生物的毒性均无手性选择性,说明无法通过对氟虫腈进行手性拆分并使用单一对映体配方来降低氟虫腈的环境风险.这一结论间接支持了农业部新近出台的禁止氟虫腈用于防治农田害虫的政策.  相似文献

米氏凯伦藻(Karenia mikimotoi)是我国近岸海域常见的有毒有害藻华肇事种,能产生多种毒素,其发生藻华时会使大量鱼类或贝类死亡。本研究从细胞生物学、氧化和免疫酶活性角度研究米氏凯伦藻对海洋青鳉鱼(Oryzias melastigma)的急性毒性效应(72 h),以期揭示米氏凯伦藻对鱼类的致死原因。组织切片结果表明,高浓度的米氏凯伦藻[(1.37~1.52)×107 cells/L]可以导致海洋青鳉鱼鳃丝断裂,成团状与相邻鳃丝融合,鳃丝上皮细胞的细胞核发生偏移,并使肝细胞出现空泡化现象。高浓度的米氏凯伦藻能够抑制海洋青鳉鱼的鳃和肝组织的SOD酶活性和CAT酶活性,引起海洋青鳉鱼脂质过氧化,导致丙二醛(MDA)含量升高,对组织造成损伤。本研究结果表明,米氏凯伦藻可能通过氧化应激反应使海洋青鳉鱼的鳃和肝组织受损,为深入阐明米氏凯伦藻的毒性机制提供参考。  相似文献
